Abstract
本发明公开了DJ‑1功能丧失的细胞模型的构建方法与应用。该细胞模型的构建方法包括如下步骤:降低多能干细胞的DJ‑1蛋白活性和/或表达量、抑制多能干细胞的DJ‑1基因表达、或、敲除多能干细胞的DJ‑1基因,得到DJ‑1功能丧失的多能干细胞;诱导所述DJ‑1功能丧失的多能干细胞为神经干细胞或间充质干细胞,得到细胞模型;多能干细胞为胚胎干细胞或诱导多能干细胞。采用本发明构建的细胞模型可以制备DJ‑1突变导致的帕金森病的细胞模型。本发明为帕金森病的发病机制进行疾病治疗奠定基础,具有重要的应用前景。
Description
技术领域
本发明属于生物技术领域,具体涉及DJ-1功能丧失的细胞模型的构建方法与应用,尤其涉及DJ-1功能丧失的人胚胎干细胞模型、DJ-1功能丧失的神经干细胞模型和DJ-1功能丧失的间充质干细胞模型的构建方法与应用。
背景技术
衰老的自由基学说认为,在衰老过程中,细胞会不断产生活性氧自由基(reactiveoxygen species,ROS),而这种自由基可以引起DNA损伤从而导致突变,也可以使细胞中的多种物质发生氧化,损伤生物膜。干细胞(如神经干细胞(Neural stem cell,NSC)、间充质干细胞(Mesenchymal stem cells,MSC))对细胞内和细胞外的氧化压力高度敏感,过度的氧化压力会导致干细胞的衰老,导致个体衰老以及衰老相关疾病。因此,研究氧化压力对干细胞内稳态的调控具有重要的生物学意义。
成体干细胞是存在于机体内,具有自我更新和一定分化潜能的细胞总称,对于机体内环境稳态维持、补充损耗的多种终末分化细胞有重要作用,成体干细胞的稳态失衡、衰退及耗竭与机体衰老关系密切。神经干细胞以对称分裂和不对称分裂两种方式增殖,在特定条件下诱导分化为神经元、星形胶质细胞和少突胶质细胞。神经干细胞具有很强的可塑性,不仅可以分化为各种类型的成熟神经细胞,还可以在特定环境下分化产生不同组组织的细胞。例如神经干细胞可实现跨胚层转化,分化为骨骼肌细胞和造血样细胞,具有重要的实用价值。间充质干细胞是成体干细胞的重要成员,最初来自发育早期的中胚层和外胚层,具有自我复制能力以及向脂肪、肌肉、心肌、软骨、成骨等多种细胞分化的能力,对于机体内炎症反应的调控也具有重要意义,间充质干细胞的异常或加速耗竭是引起骨质疏松、肌肉萎缩及某些早衰症的重要诱因。在应用方面,基于分离、体外分化、培养人类间充质干细胞的体系较为成熟,及间充质干细胞具有重要的功能、作用等优点,间充质干细胞已经成为细胞治疗领域重要的工具细胞类型。研究表明,体外培养的间充质干细胞明显地表现出复制性衰老的表型。综上,在人类成体干细胞(如神经干细胞、间充质干细胞)中研究氧化还原状态的维持具有重要的作用。
DJ-1基因又名PARK7(Parkinsonism associated deglycase)基因,位于人1号染色体(1p36.12-1p36.33),共7个外显子。DJ-1基因编码DJ-1蛋白,DJ-1蛋白由189个氨基酸组成,分子量为21KDa。DJ-1的突变导致其功能的失活,造成常染色体隐性遗传性帕金森病。在氧化压力条件下,DJ-1能够通过自身氧化或者调控抗氧化相关蛋白(如NRF2、SOD1)清除氧化压力。
发明内容
本发明的目的是提供一种可以用来制备帕金森病细胞模型的细胞模型。
本发明首先保护一种构建细胞模型的方法,可为a1)或a2)或a3):
a1)降低干细胞的DJ-1蛋白活性和/或表达量,得到细胞模型;
a2)抑制干细胞的DJ-1基因表达,得到细胞模型;
a3)敲除干细胞的DJ-1基因,得到细胞模型。
上述方法中,所述干细胞可为多能干细胞、神经干细胞或间充质干细胞。
上述方法中,干细胞为多能干细胞时,所述细胞模型的制备方法包括如下步骤:降低多能干细胞的DJ-1蛋白活性和/或表达量、抑制多能干细胞的DJ-1基因表达、或、敲除多能干细胞的DJ-1基因,得到DJ-1功能丧失的多能干细胞;诱导所述DJ-1功能丧失的多能干细胞为神经干细胞或间充质干细胞,得到细胞模型(即神经干细胞细胞模型或间充质干细胞细胞模型)。
所述诱导DJ-1功能丧失的多能干细胞为神经干细胞的方法可包括如下步骤:取所述DJ-1功能丧失的多能干细胞,先用神经干细胞诱导培养基培养,再用神经干细胞基础诱导培养基传代,得到的细胞即为神经干细胞细胞模型。
进一步的,所述诱导DJ-1功能丧失的多能干细胞为神经干细胞的方法可包括如下步骤:取所述DJ-1功能丧失的多能干细胞,先用神经干细胞诱导培养基(具体可为神经干细胞诱导培养基1)培养1-3天(如1-2天、2-3天、1天、2天或3天),之后用神经干细胞诱导培养基(具体可为神经干细胞诱导培养基2)培养4-6天(如4-5天、5-6天、4天、5天或6天),再用神经干细胞基础诱导培养基传代3次(每4-5天传代一次),得到的细胞为DJ-1功能丧失的神经干细胞,即神经干细胞细胞模型。
进一步的,所述诱导DJ-1功能丧失的多能干细胞为间充质干细胞的方法可包括如下步骤:将所述DJ-1功能丧失的多能干细胞进行拟胚体分化,CD73、CD90和CD105均为阳性的细胞为DJ-1功能丧失的间充质干细胞,即间充质干细胞细胞模型。
上述任一所述多能干细胞可为胚胎干细胞或诱导多能干细胞。
进一步的,所述胚胎干细胞可为人胚胎干细胞。所述诱导多能干细胞可为人诱导多能干细胞。
更进一步的,所述人胚胎干细胞可为商业化人胚胎干细胞,如人胚胎干细胞H9。
上文中,拟胚体分化之后、筛选“CD73、CD90和CD105均为阳性的细胞”之前,还包括用间充质干细胞培养基培养,获得纤维状细胞的步骤。所述培养时间可为1-3周(如1-2周、2-3周、1周、2周或3周)。
上述任一所述“降低干细胞的DJ-1蛋白活性和/或表达量”可通过如下任一种方式:抑制干细胞中DJ-1蛋白的合成、抑制干细胞中DJ-1蛋白的功能、促进干细胞中DJ-1蛋白的降解。
进一步的,所述“降低干细胞的DJ-1蛋白活性和/或表达量”可通过向干细胞中加入DJ-1蛋白抑制剂来实现。所述DJ-1蛋白抑制剂可为抑制DJ-1蛋白合成或抑制DJ-1蛋白功能或促进DJ-1蛋白降解的蛋白质、多肽或小分子化合物。
上述任一所述“抑制干细胞的DJ-1基因表达”或上述任一所述“敲除干细胞的DJ-1基因”可通过如下任一种方式:RNA干扰、基因沉默、基因敲除、基因突变。
进一步的,所述“降低干细胞的DJ-1蛋白活性和/或表达量”、所述“抑制干细胞的DJ-1基因表达”或所述“敲除干细胞的DJ-1基因”是通过基因编辑干细胞中DJ-1基因实现的。
更进一步的,所述“基因编辑干细胞中DJ-1基因”为向干细胞中DJ-1基因第2外显子插入单碱基,从而DJ-1蛋白的活性和/或表达量降低、DJ-1基因的表达受到抑制或DJ-1基因敲除。所述基因编辑是通过CRISPR/Cas9方法来实现的。
所述CRISPR/Cas9中靶向DJ-1基因的gRNA的序列可如SEQ ID NO:1所示。
所述“向干细胞中DJ-1基因第2外显子插入单碱基”具体可为向干细胞中第2外显子第107个核苷酸和第108个核苷酸之间纯合插入单碱基A。
所述采用CRISPR/Cas9方法基因编辑干细胞中DJ-1基因进一步可包括如下步骤:将表达所述gRNA表达载体和表达Cas9的载体导入所述干细胞中,实现DJ-1蛋白的活性和/或表达量降低、DJ-1基因的表达受到抑制或DJ-1基因敲除。
所述gRNA表达载体具体可为DJ-1-gRNA-mCherry表达载体。所述DJ-1-gRNA-mCherry表达载体的构建方法为:(1)采用DJ-1gRNA-F:5’-TTTCTTGGCTTTATATATCTTGTGGAAAGGACGAAACACCGAGATGTCATGAGGCGAGCT-3’和DJ-1gRNA-R:5’-GACTAGCCTTATTTTAACTTGCTATTTCTAGCTCTAAAACAGCTCGCCTCATGACATCTC-3’进行退火扩增,回收核酸片段;(2)将gRNA-mCherry质粒(Addgene)用AflII酶切,回收约6kb的载体骨架;(3)将核酸片段和载体骨架进行连接,得到DJ-1-gRNA-mCherry表达载体。所述表达Cas9的载体具体可为Cas9表达载体(Addgene公司的产品,货号为#87109)。所述gRNA表达载体和所述表达Cas9的载体的质量配比为可1:2。
所述培养具体可为:将成功转入gRNA表达载体和表达Cas9的载体的人胚胎干细胞(H9)转接到含有预先用丝裂霉素灭活处理过的小鼠胚胎成纤维细胞的饲养层体系中,用人多能干细胞培养基(即CDF12培养基)培养1-3周(如1-2周、2-3周、1周、2周或3周),获得所述DJ-1功能丧失的人胚胎干细胞。
进一步所述将gRNA表达载体和表达Cas9的载体导入干细胞中后还包括鉴定阳性克隆的步骤,具体为采用PCR技术初步筛选基因组水平编辑成功的人胚胎干细胞克隆,并将其扩大培养。
采用上述任一所述方法制备的多能干细胞细胞模型或神经干细胞细胞模型即可以制备帕金森病细胞模型。
本发明还保护任一所述方法构建的细胞模型。其中,多能干细胞细胞模型或神经干细胞细胞模型为进一步制备帕金森病细胞模型。
本发明还保护一种用于构建细胞模型的产品,可包括用于降低DJ-1蛋白活性和/或表达量的物质甲、或、用于抑制DJ-1基因表达的物质乙、或、用于敲除DJ-1基因的物质丙。
上述产品中,所述物质甲、所述物质乙或所述物质丙可包括靶向DJ-1基因的gRNA或表达靶向DJ-1基因的gRNA的载体。
所述靶向DJ-1基因的gRNA可为靶向DJ-1基因的第2外显子的gRNA。
所述靶向DJ-1基因的gRNA的序列可如SEQ ID NO:1所示。
所述表达靶向DJ-1基因的gRNA的载体具体可为所述DJ-1-gRNA-mCherry表达载体。
所述物质甲、所述物质乙或所述物质丙具体可为靶向DJ-1基因的gRNA或表达靶向DJ-1基因的gRNA的载体。

上述应用中,DJ-1蛋白可以结合在编码CHCHD2蛋白的基因的启动子区域,从而达到抑制CHCHD2蛋白的表达的目的。所述CHCHD2蛋白为另一个帕金森病相关的蛋白,CHCHD2基因的突变导致常染色体显性遗传病帕金森病的发生。
上述应用中,所述药物可为天然有机物和/或小分子化合物和/或毒性分子。

上述任一所述间充质干细胞(MSC)培养基可为:含10%(体积百分含量)胎牛血清、0.1mM非必需氨基酸、1%(1g/100mL)青霉素/链霉素和10ng/mL人FGF2的α-MEM培养基。
上文中,采用细胞模型(如本发明构建的多能干细胞细胞模型或神经干细胞细胞模型)制备帕金森病细胞模型的方法为本领域通用的方法,主要是进行多巴胺能神经元分化。
采用本发明构建的细胞模型可以制备DJ-1突变导致的帕金森病的细胞模型。本发明为帕金森病的发病机制进行疾病治疗奠定基础,具有重要的应用前景。

实施例1、DJ-1功能丧失的人胚胎干细胞系(即DJ-1-/-hESC细胞系)的获得和鉴定
本实施例涉及在人胚胎干细胞中靶向编辑人类DJ-1基因(基因组序列为GenBank:NC_000001.11的第7961654-7985505位),使DJ-1蛋白(GeneID:11315)功能丧失,得到DJ-1功能丧失的人胚胎干细胞。
本发明的发明人首先设计并通过分子克隆方法获得靶向人类基因组中DJ-1基因第2外显子的gRNA序列,然后将该gRNA序列插入gRNA-mCherry质粒(Addgene公司产品,商品名和货号分别为gRNACloning Vector,Plasmid#41824)的AflII的酶切识别位点之间,得到DJ-1-gRNA-mCherry表达载体。将DJ-1-gRNA-mCherry表达载体和Cas9表达载体共同电转化人胚胎干细胞,获得DJ-1基因第2外显子第107个核苷酸和第108个核苷酸之间纯合插入单碱基A的人胚胎干细胞。其中,电转化人胚胎干细胞后,还包括使用流式细胞仪分选出GFP与mCherry双阳性的细胞的步骤(即使用流式细胞仪进行克隆筛选);在克隆筛选后还包括对挑选的克隆用基因组PCR的方法对其基因编辑的情况进行确认的步骤。具体方法如下:
一、DJ-1-/-hESC细胞系的获得
1、DJ-1-gRNA-mCherry表达载体的构建
(1)根据NCBI提供DJ-1基因组数据查找分析,设计靶向DJ-1基因第2外显子的gRNA序列。设计的靶序列为:5’-GAGATGTCATGAGGCGAGCT-3’(SEQ ID NO:1)。
(2)采用DJ-1gRNA-F:
5’-TTTCTTGGCTTTATATATCTTGTGGAAAGGACGAAACACCGAGATGTCATGAGGCGAGCT-3’(下划线为靶序列)和DJ-1gRNA-R:
5’-GACTAGCCTTATTTTAACTTGCTATTTCTAGCTCTAAAACAGCTCGCCTCATGACATCTC-3’(下划线为靶序列)进行退火扩增,之后用PCR回收试剂盒(QIAquick公司产品,货号:28106)回收gRNA序列。
扩增体系为50μL,由5×GC buffer(Phusion聚合酶自带)10μL、dNTP(浓度为2.5mM)4μL、DJ-1gRNA-F(浓度为10μM)2μL、DJ-1gRNA-R(浓度为10μM)2μL、Phusion聚合酶(NEB公司产品,货号:M0530L)0.5μL和ddH2O 31.5μL组成。
PCR反应条件如下:98℃10min;0.1℃/s降至25℃,每降10℃停1min;25℃1min;72℃10min;16℃∞。
3)将gRNA-mCherry质粒用AflII酶切,回收后的载体片段(约6kb)与步骤(2)回收的gRNA序列进行连接,得到DJ-1-gRNA-mCherry表达载体。DJ-1-gRNA-mCherry表达载体含有SEQ ID NO:1所示的DNA片段。
连接反应体系(20μL)如下:
连接反应条件如下:50℃,30min。
2、DJ-1-/-hESC细胞系的获得
(1)培养H9细胞系,培养方法为a或b:
a、将H9细胞系接种至预先培养了经过丝裂霉素(Selleck公司产品,货号:S8146)灭活的小鼠胚胎成纤维细胞(Invitrogen公司产品,货号:S1520-100)的培养板中,使用人类胚胎干细胞培养基(即CDF12培养基)与小鼠胚胎成纤维细胞共同培养;
b、将H9细胞系接种至预先用细胞外基质(qualified-Matrigel,BD Biosciences公司产品,货号:354277)包被的培养板中,使用mTeSR培养基(StemCell Technologies公司产品)培养。
(2)将处于对数期增殖的H9细胞,PBS清洗一次,TrypLE Express(Invitrogen公司,货号为12604021)消化5-10min后,轻轻吹打成单细胞(即获得H9单细胞)。
(3)将步骤(2)得到的5×106个H9单细胞、14μg Cas9表达载体(Addgene公司的产品,货号为#87109)和7μg DJ-1-gRNA-mCherry表达载体混合,然后使用opti-MEM(lifetechnology)定容至100μL,用4D电转仪(Lonza公司,货号:4D-NucleofectorTM System)进行电转。
4)将电转后的细胞加入Matrigel包被的培养板中,培养24h后,用荧光显微镜观察绿色荧光和红色荧光。如果双阳性比例大于1%(即同时发出绿色荧光和红色荧光的细胞比例大于1%),可进行下一步。
5)电转48h后,采用TrypLE消化收集细胞,然后用流式细胞仪分选GFP和mCherry双阳性细胞。将分选得到的双阳性细胞置于预先铺好经过丝裂霉素灭活处理的MEF的六孔板的孔中,用人多能干细胞培养基(即CDF12培养基)培养,放回培养箱中。
6)持续用人多能干细胞培养基(即CDF12培养基)培养约14天,即可看到由单细胞形成的小克隆,用PCR技术扩增出包含靶向序列的位点,再进行DNA测序分析。PCR技术使用的引物为DJ-1-F:5’-TGTGAAATGCAGGAGACCCACGTA-3’和DJ-1-R:5’-TGAGCGACACAGAGAGACACCATC-3’。
鉴定结果见图1中a。结果表明,最终成功获得DJ-1基因第2外显子第107个核苷酸和第108个核苷酸之间纯合插入单碱基A的基因编辑克隆,该克隆即DJ-1-/-hESC细胞系。
二、DJ-1-/-hESC细胞系的鉴定
通过Western blotting、细胞免疫荧光等手段鉴定产生的克隆是否被正确打靶。
1、Western blotting鉴定DJ-1蛋白
提取供试细胞(H9细胞系或DJ-1-/-hESC细胞系)的总蛋白,采用Western blotting检测细胞表达的蛋白质。一抗为DJ-1N端抗体(anti-DJ-1,兔源单克隆,Abcam公司产品,货号:ab76008),二抗为HRP标记的羊抗兔抗体(Santa cruz公司产品,货号:sc-2005)。一抗为DJ-1C端抗体(anti-DJ-1,羊源多克隆,Abcam公司产品,货号:ab4150),二抗为HRP标记的兔抗羊抗体(Santa cruz公司产品,货号:sc-2005)。以β-actin为内参,一抗为鼠源抗β-actin抗体(Santa cruz公司产品,货号:sc-8432),二抗为HRP标记的羊抗鼠抗体(Santa cruz公司产品,货号:sc-2005)。
检测结果见图1中b(上图为DJ-1N端的检测,下图为DJ-1C端的检测)。结果表明,DJ-1-/-hESC细胞系用DJ-1N端抗体和DJ-1C端抗体均无法检测到DJ-1蛋白的表达,H9细胞系用DJ-1N端抗体和DJ-1C端抗体均可检测到DJ-1蛋白的表达(分子量约为21kDa)。
2、免疫荧光鉴定DJ-1蛋白
采用免疫荧光技术检测供试细胞(H9细胞系或DJ-1-/-hESC细胞系)中DJ-1蛋白的表达情况。具体步骤依次如下:
(1)将培养于盖玻片上的供试细胞(H9细胞系或DJ-1-/-hESC细胞系)用4%(v/v)多聚甲醛室温固定30min,然后PBS缓冲液漂洗3次,每次5min。
(2)完成步骤(1)后,用含0.4%(v/v)Triton X-100的PBS缓冲液室温孵育30min,PBS缓冲液漂洗1次(约5min)。
(3)完成步骤(2)后,用10%(v/v)驴血清(Jackson ImmunoResearchLaboratories,Inc.公司产品,货号:017-000-121)室温封闭1h;然后换用添加抗人源DJ-1抗体(Abcam公司的产品,货号为ab76008)的封闭液4℃孵育过夜;最后用PBS缓冲液漂洗3次,每次5min。
(4)完成步骤(3)后,加入二抗,室温孵育1h;然后用PBS缓冲液漂洗3次,每次5min。
(5)完成步骤(4)后,用工作浓度为2μg/mL的Hoechst 33342(Life technology公司产品,货号:H3570)室温孵育15min,最后封片和观察。
检测结果见图1中c(第一列为DJ-1蛋白,第二列为DNA)。结果表明,H9细胞系中DJ-1蛋白主要定位于细胞质,细胞核中也有部分定位;DJ-1-/-hESC细胞系中则检测不到DJ-1蛋白的表达。
3、免疫荧光检测干细胞特性
采用免疫荧光技术检测供试细胞(H9细胞系或DJ-1-/-hESC细胞系)中干细胞标记物NANOG、OCT4和SOX2的表达情况。具体步骤依次如下:
(1)同步骤2中(1)。
(2)同步骤2中(2)。
(3)完成步骤(2)后,用10%(v/v)驴血清(Jackson ImmunoResearchLaboratories,Inc.公司产品,货号:017-000-121)室温封闭1h;然后换用添加一抗(抗人鼠源OCT4抗体(Santa Cruz公司的产品,货号为sc-5279)、抗人羊源SOX2抗体(Santa Cruz公司的产品,货号为sc-17320)或抗人兔源NANOG抗体(Abcam公司的产品,货号为ab21624)的封闭液4℃孵育过夜;最后用PBS缓冲液漂洗3次,每次5min。
(4)同步骤2中(4)。
(5)同步骤2中(5)。
检测结果见图1中d(第一列为细胞形态图,第二列为NANOG,第三列为OCT4,第四列为SOX2,第五列为DNA)。结果表明,与H9细胞系的细胞类似,DJ-1-/-hESC细胞系的细胞也能够表达NANOG、OCT4和SOX2。由此可见,DJ-1-/-hESC细胞系对干细胞干性基因的表达没有显著影响。
4、通过畸胎瘤实验检测细胞的多能干性
TuJ1蛋白为外胚层标记物。FOXA2为内胚层标记物。α-SMA为中胚层标记物。
(1)将供试细胞(H9细胞系或DJ-1-/-hESC细胞系)接种至无饲养层的Matrigel包被的培养板上培养,待细胞长满,消化计数。
(2)完成步骤(1)后,取5×106个供试细胞,用混合液(由1体积份Matrigel和4体积份mTesR混合而成)重悬,得到重悬液。重悬液中,供试细胞的浓度为1×104细胞/μL。
(3)完成步骤(2)后,取免疫缺陷小鼠(NOD/SCID,北京维通利华实验动物技术有限公司的产品),皮下注射500μL重悬液,然后常规饲养6周,注射部分即长成畸胎瘤。脱颈处死小鼠,然后取出畸胎瘤。
(4)完成步骤(3)后,畸胎瘤用4%多聚甲醛固定2天,用30%(30g/100mL)蔗糖溶液充分脱水,然后使用Compound O.C.T包埋剂进行包埋,最后在冰冻切片机上切片(厚度为10μm),得到切片组织。将切片组织晾干。
(5)完成步骤(4)后,将切片组织,用4%(v/v)多聚甲醛室温固定30min,然后PBS缓冲液漂洗3次,每次5min。
(6)完成步骤(5)后,用含0.4%(v/v)Triton X-100的PBS缓冲液室温孵育30min,PBS缓冲液漂洗1次(约5min)。
(7)完成步骤(6)后,用10%(v/v)驴血清(Jackson ImmunoResearchLaboratories,Inc.公司产品,货号:017-000-121)室温封闭1h;然后换用添加一抗(抗人兔源TuJ1抗体(Sigma公司的产品,货号为T2200)、抗人兔源FOXA2抗体(Cell SignallingTechnology公司的产品,货号为2158S)或抗人兔源SMA抗体(Abcam公司的产品,货号为32575)的封闭液4℃孵育过夜;最后用PBS缓冲液漂洗3次,每次5min。
(8)完成步骤(7)后,加入二抗,室温孵育1h;然后用PBS缓冲液漂洗3次,每次5min。
(9)完成步骤(8)后,用工作浓度为2μg/mL的Hoechst 33342(Life technology公司产品,货号:H3570)室温孵育5min,最后封片和观察。
检测结果见图1中e(第一列为TuJ1,第二列为SMA,第三列为FOXA2)。结果表明,H9细胞系和DJ-1-/-hESC细胞系均能在体内形成畸胎瘤,可以分化成外胚层、中胚层和内胚层三个胚层。由此可见,DJ-1-/-hESC细胞系保持了良好的多能干性。
5、核型分析检测细胞染色体的稳定性
(1)取DJ-1-/-hESC细胞系,用秋水仙素处理40min。
(2)完成步骤(1)后,收集细胞,先用0.075M KCl低渗液在37℃条件下处理30min,再用含甲醇和冰醋酸的的固定液(甲醇和冰醋酸的体积比为3:1)固定两次。
(3)完成步骤(2)后,按适当密度将细胞滴到载玻片上,马上在90℃水浴锅上用水汽蒸10s左右,放入70℃烘箱烤3h;然后胰酶消化25-45s,生理盐水漂洗后,在37℃条件下用Gimsa染液染色5-10min,自来水晾干,显微镜观察。
检测结果见图1中f。结果表明,DJ-1-/-hESC细胞系具有正常的核型。由此可见,DJ-1-/-hESC细胞系保持了染色体的稳定性。
实施例2、DJ-1-/-hESC细胞系体外定向分化制备神经干细胞(即DJ-1-/-hNSC)和神经元(DJ-1-/-神经元)及其鉴定和表型分析
一、诱导分化形成神经干细胞
1、将H9细胞系接种至用丝裂霉素处理过的小鼠胚胎成纤维细胞上,用人多能干细胞培养基(即CDF12培养基)培养至其密度达到20%左右,得到1次培养后的细胞。
2、完成步骤1后,将1次培养后的细胞在神经干细胞诱导培养基1继续培养2天(每天换液),得到2次培养后的细胞。
3、完成步骤2后,将2次培养后的细胞在神经干细胞诱导培养基2继续培养5天(每天换液),得到3次培养后的细胞。
4、完成步骤3后,将3次培养后的细胞以单细胞转到Matrigel包被的培养板中用神经干细胞基础诱导培养基进行培养,每4-5天传代一次,连续传至第三代,即得到H9的神经干细胞,将其命名为DJ-1+/+hNSC。
按照上述步骤,将H9细胞系替换为DJ-1-/-hESC细胞系,其它步骤均不变,得到DJ-1-/-hNSC。
二、神经干细胞的鉴定
1、Western blotting鉴定DJ-1蛋白
按照实施例1步骤二中1的方法,将供试细胞替换为DJ-1+/+hNSC或DJ-1-/-hNSC,其它步骤均不变。
检测结果见图2中a(左图为DJ-1N端的检测,右图为DJ-1C端的检测)。结果表明,DJ-1-/-hNSC用DJ-1N端抗体和DJ-1C端抗体均无法检测到DJ-1蛋白的表达,DJ-1+/+hNSC用DJ-1N端抗体和DJ-1C端抗体均可检测到DJ-1蛋白的表达(分子量约为21kDa)。
2、免疫荧光鉴定DJ-1蛋白
按照实施例1步骤二中2的方法,将供试细胞替换为DJ-1+/+hNSC或DJ-1-/-hNSC,其它步骤均不变。
检测结果见图2中b(第一列为DJ-1蛋白,第二列为线粒体蛋白,第三列为DJ-1与线粒体蛋白合并)。结果表明,DJ-1+/+hNSC中DJ-1蛋白主要定位于细胞质,在线粒体和细胞核中也有部分定位;DJ-1-/-hNSC中则检测不到DJ-1蛋白的表达。
3、免疫荧光检测神经干细胞特性
按照实施例1步骤二中3的方法,将供试细胞替换为DJ-1+/+hNSC或DJ-1-/-hNSC,步骤(3)替换为步骤(3A),其它步骤均不变,检测供试细胞中神经干细胞标记物Nestin、PAX6和SOX2的表达情况。
步骤(3A)为:完成步骤(2)后,用10%(v/v)驴血清(Jackson ImmunoResearchLaboratories,Inc.公司产品,货号:017-000-121)室温封闭1h;然后换用添加一抗(抗人Nestin抗体(BD公司,货号560422,稀释比例1:200)、抗人PAX6抗体(Biolegend公司,货号901301,稀释比例1:200)或抗人SOX2抗体(Santa Cruz,货号为sc-17320,稀释比例1:100)的封闭液4℃孵育过夜;最后用PBS缓冲液漂洗3次,每次5min。
检测结果见图2中c(第一列为PAX6,第二列为Nestin,第三列为SOX2,第四列为DNA)。结果表明,DJ-1+/+hNSC和DJ-1-/-hNSC均可表达神经干细胞的特异性标记物NANOG、OCT4和SOX2。
4、神经干细胞向神经元进行分化
神经干细胞的一个特性就是向神经元进行分化。检测DJ-1-/-hNSC是否具有进一步分化成神经元的能力,具体步骤依次如下:
1、将供试细胞(DJ-1+/+hNSC或DJ-1-/-hNSC)接种至matrigel处理的六孔板上(密度为1.2×105/孔),使用神经干细胞培养基(具体可为神经干细胞基础诱导培养基)培养1-3天以形成克隆。
2、完成步骤1后,使用Advanced DMEM/F12培养基洗一遍,然后用神经元分化培养基培养2天。
3、完成步骤2后,加入laminin(Sigma公司,货号为2020)并使laminin在体系中的浓度为20μg/mL,过夜孵育;然后弃培养基,加入神经元分化培养基培养1天。
4、完成步骤3后,弃培养基,加入神经元分化培养基培养2-3周(期间隔天换一次液),此时绝大多数细胞具有完好、长的树突/轴突(即分化为神经元)。
5、完成步骤4后,将神经元细胞用4%(v/v)多聚甲醛室温固定30min,然后PBS缓冲液漂洗3次,每次5min。
6、完成步骤5后,用含0.4%(v/v)Triton X-100的PBS缓冲液室温孵育30min,PBS缓冲液漂洗1次(约5min)。
7、完成步骤6后,用10%(v/v)驴血清(Jackson ImmunoResearch Laboratories,Inc.公司产品,货号:017-000-121)室温封闭1h;然后换用添加一抗MAP2(Sigma,货号为M4403,稀释比例1:200)或anti-β-tubulin III(TUJ1,Sigma,货号为T2200,稀释比例1:500))的封闭液4℃孵育过夜;最后用PBS缓冲液漂洗3次,每次5min。
8、完成步骤7后,加入二抗,室温孵育1h;然后用PBS缓冲液漂洗3次,每次5min。
9、完成步骤8后,用工作浓度为2μg/mL的Hoechst 33342(Life technology公司产品,货号:H3570)室温孵育5min,最后封片和观察。
检测结果见图2中d(第一列为分化形成的神经元形态图,第二列为MAP2,第三列为TuJ1,第四列为DNA)。结果表明,DJ-1+/+神经元或DJ-1-/-神经元均表达神经元的特异性标志物MAP2和TuJ1。由此可见,DJ-1蛋白并不影响神经干细胞向神经元进行分化。
三、DJ-1-/-hNSC的表型分析
1、DJ-1-/-hNSC细胞中活性氧的测定
利用CM-H2DCFDA探针(Molecular Probe公司)检测DJ-1+/+hNSC和DJ-1-/-hNSC中有害代谢物活性氧(ROS)的水平。具体步骤如下:
(1)用TrypLE消化供试细胞(DJ-1+/+hNSC或DJ-1-/-hNSC),计数。
(2)完成步骤(1)后,将1×105个供试细胞溶于100μL PBS缓冲液,然后加入CM-H2DCFDA探针,得到体系。该体系中,CM-H2DCFDA探针的浓度为1μM。
(3)完成步骤(2)后,将所述体系置于细胞培养箱,37℃避光孵育15min。
(4)完成步骤(3)后,用过量PBS缓冲液清洗,然后用300μL PBS缓冲液重悬,使用流式细胞仪(BD公司)进行检测分析,检测通道为FITC。
检测结果见图2中e。结果表明,DJ-1+/+hNSC和DJ-1-/-hNSC中ROS水平无显著差异。这与DJ-1在肿瘤细胞以及小鼠的原代细胞作为的抗氧化蛋白的作用是不同的,DJ-1蛋白对于神经干细胞的氧化压力稳态维持可能是非必须的。
2、DJ-1-/-hNSC在氧化压力诱导物、DNA损伤诱导物和蛋白酶体抑制剂刺激下的细胞活力检测
氧化压力诱导物为PX12、Paraquat(即百草枯)、CCCP或TTFA。
DNA损伤诱导物为Zeocin(即博来霉素)、MMC或CPT。
蛋白酶体抑制剂为MG132。
采用MTS实验检测氧化压力诱导物、DNA损伤诱导物或蛋白酶体抑制剂刺激处理下供试细胞(DJ-1+/+hNSC或DJ-1-/-hNSC)的凋亡水平。具体步骤如下:
(1)取Matrigel处理的96孔板,每孔加入100μL含20000个供试细胞(DJ-1+/+hNSC或DJ-1-/-hNSC)的NSC培养基,37℃培养24h。
(2)完成步骤(1)后,弃培养基,每孔加入100μL含有药物(氧化压力诱导物、DNA损伤诱导物或蛋白酶体抑制剂)的NSC培养基,37℃培养24h。各个药物在体系中的浓度见图2中f和g。
(3)完成步骤(2)后,弃培养基,PBS缓冲液洗一遍,然后每孔加入100μL含10%MTS染料的NSC培养基,37℃培养2-4h。
按照上述步骤,将步骤(2)中含有药物的NSC培养基替换为NSC培养基,其它步骤不变。作为对照组。
取Matrigel处理的96孔板,每孔加入100μL含10%MTS染料的NSC培养基,37℃培养2-4h。作为空白组。
(4)完成步骤(3)后,使用Synergy H1全功能微孔板检测仪检测在490nm处的吸光度值,然后按照下述公式计算供试细胞的相对活力:细胞活力=(实验组吸光度值-空白组吸光度值)/(对照组吸光度值-空白组吸光度值)
对结果进行统计检验。使用GraphPad软件将每个实验组同空白组进行双尾t检验。“ns”表示无显著差异。
检测结果见图2中f和g。结果表明,与DJ-1+/+hNSC相比,DJ-1-/-hNSC对于上述药物刺激并不敏感,进一步表明DJ-1蛋白对于神经干细胞的稳态维持并不是必须的。
实施例3、DJ-1-/-hESC细胞系体外定向分化制备间充质干细胞(即DJ-1-/-hMSC)及其表型分析
一、DJ-1-/-hESC细胞系体外定向分化制备DJ-1-/-hMSC
1、将H9细胞系进行拟胚体(EB)分化,分化3天,得到拟胚体。
2、将拟胚体接种于基质胶(Matrigel)(Corning公司)包被的6孔板中,用间充质干细胞培养基进行培养,继续培养2周至纤维状细胞出现。再经过一次传代后,用流式细胞术分选其中的CD73、CD90和CD105均为阳性的细胞类群(CD73、CD90和CD105均为间充质干细胞的细胞表面标志物,见图3中a),即获得H9的间充质干细胞,将其命名为DJ-1+/+hMSC。
按照上述步骤,将H9细胞系替换为DJ-1-/-hESC细胞系,其它步骤均不变,得到DJ-1-/-hMSC。流式细胞术分选见图3中a。
二、DJ-1-/-hMSC的表型分析
1、Western blotting鉴定DJ-1蛋白
按照实施例1步骤二中1的方法,将供试细胞替换为DJ-1+/+hMSC或DJ-1-/-hMSC,其它步骤均不变。
检测结果见图3中b(左图为DJ-1N端的检测,右图为DJ-1C端的检测)。结果表明,DJ-1-/-hMSC用DJ-1N端抗体和DJ-1C端抗体均无法检测到DJ-1蛋白的表达,DJ-1+/+hMSC用DJ-1N端抗体和DJ-1C端抗体均可检测到DJ-1蛋白的表达(分子量约为21kDa)。
2、免疫荧光鉴定DJ-1蛋白
按照实施例1步骤二中2的方法,将供试细胞替换为DJ-1+/+hMSC或DJ-1-/-hMSC,其它步骤均不变。
检测结果见图3中c(第一列为DJ-1蛋白,第二列为线粒体蛋白,第三列为DJ-1与线粒体蛋白合并)。结果表明,DJ-1+/+hMSC中DJ-1蛋白主要定位于细胞质,在线粒体和细胞核中也有部分定位;DJ-1-/-hMSC中则检测不到DJ-1蛋白的表达。
3、DJ-1-/-hMSC细胞中活性氧的测定
按照实施例2步骤三中1的方法,将供试细胞替换为DJ-1+/+hMSC或DJ-1-/-hMSC,其它步骤均不变。
检测结果见图3中d。结果表明,DJ-1+/+hMSC和DJ-1-/-hMSC中ROS水平无显著差异。由此可见,DJ-1蛋白对于间充质干细胞的氧化压力稳态维持可能是非必须的。
4、DJ-1-/-hMSC在氧化压力诱导物和DNA损伤诱导物刺激下的细胞活力检测
按照实施例2步骤三中2的方法,将供试细胞替换为DJ-1+/+hMSC或DJ-1-/-hMSC,其它步骤均不变。其中,氧化压力诱导物为PX12、H2O2(即过氧化氢)或TTFA,DNA损伤诱导物为Zeocin(即博来霉素)、MMC、4NQO或CPT。各个药物在体系中的浓度见图3中e和f。
检测结果见图3中e和f。结果表明,与DJ-1+/+hMSC相比,DJ-1-/-hMSC对于上述药物刺激并不敏感,进一步表明DJ-1蛋白对于间充质干细胞的稳态维持并不是必须的。
5、DJ-1-/-hMSC细胞小鼠体内留存能力测定
为了验证DJ-1+/+hMSC和DJ-1-/-hMSC的体内生存能力,首先用表达荧光素酶Luciferase的病毒载体分别感染DJ-1+/+hMSC和DJ-1-/-hMSC细胞,感染3-5天后,将两种细胞分别消化为单细胞状态,随后分别注射入裸鼠(北京维通利华实验动物技术有限公司的产品)左、右胫骨前肌,两种细胞的注射量均相同(注射量为1×106个细胞/只)。注射第0天、第1天、第3天或第5天后利用小动物活体成像系统(Xenogen IVIS spectrum,PE公司产品)检测小鼠左、右胫骨前肌中luciferase活性,从而反映hMSC的体内留存能力。具体操作方法如下:
(1)选取生长状态良好、细胞密度为60~80%的供试细胞(DJ-1+/+hMSC或DJ-1-/-hMSC),感染表达荧光素酶Luciferase的病毒载体;
(2)感染后3-5天,待细胞长满,记为第0天,TrypLE消化为单细胞;
(3)细胞计数,每1×106个细胞以100μL PBS缓冲液重悬;
(4)取等量细胞,与Luciferase底物(D-Luciferin Firefly,potassium salt,GOLDBIO公司产品)混合,使用酶标仪测定荧光强度与细胞数的关系;
(5)取100μL细胞悬液注射入小鼠胫骨前肌(左腿注射DJ-1+/+hMSC,右腿注射DJ-1-/-hMSC);
(6)每日观察小鼠状态;
(7)注射第0天、第1天、第3天或第5天后,取出小鼠,腹腔注射Luciferase底物,麻醉后用小动物活体成像系统进行分析。统计4组生物学重复的荧光强度。
结果表明,DJ-1-/-hMSC(右腿)在胫骨前肌中的luciferase活性较DJ-1+/+hMSC(左腿)在胫骨前肌中无明显差异。由此可见,DJ-1蛋白并不影响hMSC在体内的存留能力。
实施例4、DJ-1蛋白通过结合在CHCHD2基因的启动子区域抑制DJ-1蛋白的表达。
一、DJ-1-/-hNSC和DJ-1-/-hMSC中抗氧化蛋白CHCHD2代偿性上调
1、对供试细胞(DJ-1+/+hNSC、DJ-1-/-hNSC、DJ-1+/+hMSC或DJ-1-/-hMSC)进行转录组分析。
结果表明,抗氧化蛋白CHCHD2在DJ-1-/-hNSC和DJ-1-/-hMSC中显著上调(图4中a)。
2、Western blotting鉴定CHCHD2蛋白
提取供试细胞(DJ-1+/+hNSC、DJ-1-/-hNSC、DJ-1+/+hMSC、DJ-1-/-hMSC、DJ-1+/+神经元或DJ-1-/-神经元)的总蛋白,采用Western blotting检测CHCHD2蛋白和DJ-1蛋白。
检测CHCHD2蛋白的一抗为兔抗人CHCHD2抗体(Proteintech公司的产品,货号为19424-1-AP)。
检测DJ-1蛋白的兔抗人DJ-1抗体。
以β-actin为内参,一抗为鼠源抗β-actin抗体(Santa cruz公司产品,货号:sc-8432)。
检测结果见图4中b。结果表明,CHCHD2蛋白在DJ-1-/-hNSC、DJ-1-/-hMSC和DJ-1-/-神经元中均显著上调。
3、免疫荧光鉴定CHCHD2蛋白
按照实施例1步骤二中2的方法,将供试细胞替换为DJ-1+/+hNSC、DJ-1-/-hNSC、DJ-1+/+hMSC、DJ-1-/-hMSC,将抗人源DJ-1抗体替换为兔抗人CHCHD2抗体,其它步骤均不变。
部分检测结果见图4中c。结果表明,CHCHD2蛋白在DJ-1-/-hNSC、DJ-1-/-hMSC和DJ-1-/-神经元中均显著上调。
上述结果表明,DJ-1-/-hNSC和DJ-1-/-hMSC中抗氧化蛋白CHCHD2代偿性上调。
二、DJ-1蛋白通过结合在CHCHD2基因的启动子区域抑制DJ-1蛋白的表达
为了研究CHCHD2基因的转录,CHCHD2蛋白水平上调是否受DJ-1蛋白的调控,以DJ-1+/+hMSC和DJ-1-/-hMSC为供试细胞进行双荧光素报告酶实验。具体步骤如下:
1、pGL3-CHCHD2-promoter表达载体的构建
(1)以H9细胞系的基因组DNA为模板,采用CHCHD2-Promoter-F:5’-ccgggtaccatgcacccatcacctgagcaa-3’(下划线为限制性内切酶KpnI的识别位点)和CHCHD2-Promoter-R:5’-cggctcgagctgaacttggcgccaggatct-3’(下划线为限制性内切酶XhoI的识别位点)组成的引物对进行PCR扩增,用PCR回收试剂盒回收PCR扩增产物。
(2)取所述PCR扩增产物,用限制性内切酶KpnI和XhoI酶切,回收约1500bp的酶切片段。
(3)取pGL3-basic载体(Promega),用限制性内切酶KpnI和XhoI酶切,回收约4.8kb的载体骨架。
(4)将酶切片段和载体骨架进行连接,得到pGL3-CHCHD2-promoter表达载体。
将pGL3-CHCHD2-promoter表达载体进行测序。根据测序结果,对pGL3-CHCHD2-promoter表达载体进行结构描述如下:pGL3-CHCHD2-promoter表达载体为将pGL3-basic载体中限制性内切酶KpnI和XhoI之间的DNA小片段替换为SEQ ID NO:2所示的DNA片段,得到的重组质粒。
2、瞬时转染
(1)取6孔板,每孔加入2×105个供试细胞(DJ-1+/+hMSC或DJ-1-/-hMSC)和间充质干细胞培养基,培养3天,直至细胞密度达到60%-70%。
(2)制备A液和B液。A液由125μL opti-MEM(Invitrogen)、0.4μg Renilla质粒(Promega)、3.6μg pGL3-CHCHD2-promoter表达载体和6μL P3000(Invitrogen)组成。B液由125μL opti-MEM和3.75μL Lipo3000(Invitrogen)组成。
(3)将A液逐滴加入B液中,混匀,室温静置20min,得到混合液。
(4)取完成步骤(1)的6孔板,每孔逐滴加入混合液,37℃、CO2培养48h。期间,加入混合液的第10-12h,换液为新鲜的MSC培养基。
(5)完成步骤(4)后,离心,收集沉淀;然后用PBS清洗一次,TrypLE Express消化5-10min后,轻轻吹打,即获得单细胞溶液。
3、双荧光报告系统检测
(1)取步骤2获得的单细胞溶液,加入200μL 1×ULB(双蒸水稀释5×ULB(维格拉斯)获得),室温裂解10min,得到细胞裂解液。
(2)分别取Fassay Buffer I、Fassay Substrate I和Rassay Buffer II,室温融化并混匀;按照20:1比例用Fassay Buffer I稀释Fassay Substrate I,得到FassayReagent I;按照50:1比例用Rassay Buffer II稀释Fassay Reagent I,得到RassayReagent II。
(3)取6孔板,每孔加入20μL细胞裂解液和90μL Fassay Reagent I,轻轻敲击3-5次(目的为混匀),然后采用Synergy H1全功能微孔板检测仪(BioTek)检测发光值,记录发光值为Firefly luciferase的发光单位。
(4)完成步骤(3)后,每孔加入Rassay Reagent II 90μL,轻轻敲击3-5次(目的为混匀),然后采用Synergy H1全功能微孔板检测仪检测发光值,记录发光值为Renillaluciferase的发光单位。
(5)计算相对荧光值。
相对荧光值=Firefly luciferase发光值/Renilla luciferase发光值
检测结果见图4中d。结果表明,与DJ-1+/+hMSC相比,DJ-1-/-hMSC的相对荧光值显著提高。由此可见,DJ-1蛋白通过结合在CHCHD2基因的启动子区域,反式调控CHCHD2,抑制CHCHD2蛋白的表达。
三、ChIP-qPCR分析检测DJ-1对CHCHD2的转录调控
RIPA Buffer:含0.5mM EGTA、140mM NaCl、1%(v/v)Triton X-100、0.1%(v/v)SDS、1mM EDTA和10mM pH7.5的Tris-HCl缓冲液的水溶液。
RIPA ChIP Buffer:向10mL RIPA Buffer加入1片蛋白酶抑制剂(Roche公司的产品,货号为4693159001)获得。
TE Buffer:含5mM EDTA、50mM NaCl和20mM pH8.0的Tris-HCl缓冲液的水溶液。
Elution Buffer:含5mM EDTA、50mM NaCl和20mM pH7.5的Tris-HCl缓冲液的水溶液。
Complete Elution Buffer:含5mM EDTA、50mM NaCl、1%(v/v)SDS、200mg/mlproteinase K和20mM pH7.5的Tris-HCl缓冲液的水溶液。
Lysis Buffer:含1%(v/v)SDS、10mM EDTA、pH8.0的Tris-HCl缓冲液的水溶液。
EGTA为Sigma公司的产品,货号为03777。NaCl为Sigma公司的产品,货号为V900058。Tris为AMRESCO公司的产品,货号为0497。Triton X-100为Sigma公司的产品,货号为T9284。SDS为AMRESCO公司的产品,货号为0227。EDTA为Sigma公司的产品,货号为EDS-500G。proteinase K为NEB公司,货号为P8107S。
为了进一步确认双荧光素报告酶系统的实验结果,通过ChIP-qPCR检测DJ-1对CHCHD2的调控作用。具体步骤如下:
1、含有ANTIBODY-BEADS的PCR管的制备
(1)向PCR管(规格为0.2mL)中加入60μL Dynabeads Protein A(Thermo FisherScientific公司的产品,货号为10002D),然后置于冰上的磁力架上3min,弃上清。
(2)完成步骤(1)后,向所述PCR管中加入180μL RIPA buffer,涡旋,然后置于冰上的磁力架上3min,弃上清。
(3)完成步骤(2)后,向所述PCR管中加入180μL RIPA buffer,涡旋,然后置于冰上的磁力架上3min,弃上清。
(4)完成步骤(3)后,向所述PCR管中加入65μL RIPA buffer,涡旋,然后置于冰上,得到Dynabeads Protein A悬浊液。
(5)分别取PCR管(规格为0.2mL),每管加入90μL RIPA buffer,然后置于冰上,每管分别加入10μL Dynabeads Protein A悬浊液和2.5μg DJ-1抗体(Abcam公司的产品,货号为ab4150),混匀;之后分别将PCR管粘在旋转仪上,4℃、20rpm旋转2h,得到含有ANTIBODY-BEADS的PCR管。
2、孵育和洗脱
(1)取5×106个供试细胞(DJ-1+/+hNSC、DJ-1-/-hNSC、DJ-1+/+hMSC或DJ-1-/-hMSC),TrypLE Express消化5-10min,然后用1%的甲醛交联细胞14min,不停颠倒混匀,使用125mM甘氨酸终止5min。之后,4℃、2000rpm离心8min,弃上清,沉淀使用PBS缓冲液洗涤1次。
(2)完成步骤(1)后,取所述供试细胞,加入130μL Lysis Buffer,冰上裂解5min,然后涡旋,离心,收集沉淀并超声破碎(covaris chromatin-8程序,超声时间为11min),得到超声破碎液。
(3)完成步骤(2)后,取所述超声破碎液,加入400μL预冷的RIPAChIP buffer,涡旋混匀,4℃、12000rpm离心10min,收集沉淀1和上清1。
(4)完成步骤(3)后,取所述沉淀1,加入410μL预冷的RIPA ChIP buffer,涡旋混匀,4℃、12000rpm离心10min,收集上清2。
(5)完成步骤(4)后,将上清1和上清2合并,得到上清液。每个样品分别取出40μL上清液,作为每个样品对应的Input。
(6)完成步骤(5)后,将步骤1得到的PCR管的ANTIBODY-BEADS转移到1.5mL的EP管中,然后将EP管置于磁力架上3min,弃上清;然后每个EP管加入400μL步骤(5)获得的上清液,涡旋混匀;之后分别将EP管置于旋转仪上,4℃、20rpm旋转过夜。
(7)完成步骤(6)后,重复如下步骤2次:分别取所述EP管,瞬时离心1s,置于磁力架上3min,弃上清;然后每管加入500μL预冷的RIPA buffer,轻柔的颠倒混匀,置于旋转仪上,4℃、20rpm旋转4min。
(8)完成步骤(7)后,分别取所述EP管,瞬时离心1s,置于磁力架上3min,弃上清;然后每管加入500μL TE buffer,置于旋转仪上,4℃、20rpm旋转4min。
(9)完成步骤(8)后,分别取所述EP管,瞬时离心,然后置于冰上,混匀磁珠后转移至EP管中,磁力架上3min,弃上清。
3、供试细胞DNA溶液的获得
(1)取EP管,加入40μL input、260μL Elution Buffer和3μL proteinase K(NEB公司,货号为P8107S),涡旋混匀。
(2)完成步骤(1)后,分别取完成步骤2的EP管,每管加入150μL Complete ElutionBuffer,混匀然后转移至新的EP管中。
(3)完成步骤(2)后,将EP管置于金属浴,68℃、1300rpm孵育2h。
(4)完成步骤(3)后,取所述EP管,短暂离心3s,置于磁力架上3min,收集上清1。
(5)完成步骤(4)后,取所述EP管,加入150μL Elution Buffer,混匀,置于金属浴,68℃、1300rpm孵育5min。
(6)完成步骤(5)后,取所述EP管,短暂离心,置于磁力架上3min,收集上清2。
(7)完成步骤(6)后,将上清1和上清2混匀,得到上清液。
(8)取EP管,加入步骤(7)得到的上清液(约300μL)和200μL Elution Buffer,混匀;然后加入500μL酚-氯仿-异戊醇溶液(酚:氯仿:异戊醇=25:24:1),涡旋充分混匀(目的为提取DNA),然后15000g离心5min,收集上层液相。
(9)完成步骤(8)后,向上层液相中加入等体积的氯仿-异戊醇溶液(氯仿:异戊醇=24:1)(目的为除去苯酚),然后15000g离心5min,收集上清液至新的EP管中。
(10)完成步骤(9)后,向步骤(9)收集的上清液中加入44μL pH5.2、3M醋酸钠溶液和1μL糖原溶液(浓度为20mg/mL),涡旋混匀,短暂离心,然后加入-20℃的乙醇1mL,颠倒轻弹20次以上,-80℃沉淀1h。
(11)完成步骤(10)后,取所述EP管,4℃、13200rpm离心15min,弃上清;然后加入-20℃的70%(v/v)乙醇水溶液1mL洗涤,4℃、13200rpm离心15min,弃上清;之后将所述EP管置于37℃金属浴开盖至少烘干1h,加入20μL H2O,得到供试细胞DNA溶液。
4、实时定量PCR分析
以步骤3中得到的供试细胞DNA溶液为模板,采用SYBR qPCR mix(东洋纺公司的产品,货号为QPS-201C)实时定量PCR检测DJ-1蛋白在CHCHD2的启动子区域的富集情况。以GAPDH的启动子区域的序列作为阴性对照。CHCHD2的启动子区域选择的位置是转录起始位点上游的526bp-724bp,使用的引物序列为:正向:5’-aacacatggcttgcttcc-3’,反向:5’-atggagatactgcaccaa-3’。GAPDH的启动子区域选择的位置是转录起始位点上游的139bp–296bp,使用的引物序列为:正向:5’-cacagtccagtcctgggaac-3’,反向:5’-tagtagccgggccctacttt-3’。
检测结果见图4中e。结果表明,在hNSC中,与DJ-1-/-hNSC相比,CHCHD2在DJ-1+/+hNSC中的富集程度显著上调。在hMSC中,与DJ-1-/-相比,CHCHD2在DJ-1+/+hMSC中的富集程度显著上调。DJ-1在CHCHD2启动子富集的序列为:5’–aacacatggcttgcttccccacctcttcaagggctccgttcaaatgactcccccccgccccgcctaacctctaatttccagtctccttcctctcttttcatgtcaccaccacctgatttacgtaggcgtttgtgtctcccctccttgaaacttaacttcgtgaaggtagaatcttatctgatttggtgcagtatctccat-3’。由此可见,DJ-1蛋白通过结合在CHCHD2基因的启动子区域,反式抑制CHCHD2的转录和蛋白的表达。

Claims (10)
1.一种构建细胞模型的方法,为a1)或a2)或a3):
a1)降低干细胞的DJ-1蛋白活性和/或表达量,得到细胞模型;
a2)抑制干细胞的DJ-1基因表达,得到细胞模型;
a3)敲除干细胞的DJ-1基因,得到细胞模型;
所述降低干细胞的DJ-1蛋白活性和/或表达量、所述抑制干细胞的DJ-1基因表达或所述敲除干细胞的DJ-1基因是通过基因编辑干细胞中DJ-1基因实现的;
所述基因编辑干细胞中DJ-1基因为向干细胞中DJ-1基因第2外显子插入单碱基,从而DJ-1蛋白的活性和/或表达量降低、DJ-1基因的表达受到抑制或DJ-1基因敲除;
所述基因编辑是通过CRISPR/Cas9方法来实现的;
所述CRISPR/Cas9中靶向DJ-1基因的gRNA的序列如SEQ ID NO:1所示。
2.如权利要求1所述的方法,其特征在于:所述干细胞为多能干细胞、神经干细胞或间充质干细胞。
3.如权利要求2所述的方法,其特征在于:所述干细胞为多能干细胞时,所述细胞模型的制备方法包括如下步骤:降低多能干细胞的DJ-1蛋白活性和/或表达量、抑制多能干细胞的DJ-1基因表达或敲除多能干细胞的DJ-1基因,得到DJ-1功能丧失的多能干细胞;诱导所述DJ-1功能丧失的多能干细胞为神经干细胞或间充质干细胞,得到所述细胞模型。
4.如权利要求2所述的方法,其特征在于:所述多能干细胞为胚胎干细胞或诱导多能干细胞。
5.采用权利要求1至4任一所述方法构建的细胞模型。
6.一种用于构建细胞模型的产品,包括用于降低DJ-1蛋白活性和/或表达量的物质甲、或、用于抑制DJ-1基因表达的物质乙、或、用于敲除DJ-1基因的物质丙;
所述物质甲、所述物质乙或所述物质丙包括靶向DJ-1基因的gRNA或表达靶向DJ-1基因的gRNA的载体;
所述靶向DJ-1基因的gRNA的序列如SEQ ID NO:1所示。
7.采用权利要求1至4任一所述方法构建的多能干细胞细胞模型或神经干细胞细胞模型的应用,为Y1)-Y3)中的至少一种:
Y1)制备帕金森病细胞模型;
Y2)研究帕金森病;
Y3)研究帕金森病发病机制。
8.如权利要求7所述的应用,其特征在于:所述帕金森病为常染色体隐性遗传性帕金森病。
9.CHCHD2蛋白作为药物靶点在制备预防和/或治疗帕金森病的产品中的应用。
10.如权利要求9所述的应用,其特征在于:所述帕金森病为常染色体隐性遗传性帕金森病。
