令人惊讶地发现,在培养过程中,接种在此特定结构化表面上的生物细胞沿着平行的微米尺寸和纳米尺寸的凹槽或岭定向生长,从而,由于由此引发的机械转导,从接种的单个细胞发育成神经元细胞,并最终稳定可靠地发育成具有清晰神经元结构和神经功能的神经元组织。在本发明的培养基质的结构化表面上的分化效率非常高。
这些积极效果可以进一步提高。在一个优选的实施方案中,所述基体的生物相容性聚合物或生物聚合物至少在结构化表面的区域中的刚度(机械强度)等于生物细胞的相应机械性能(刚度),优选不大于该刚度,特别优选小于该刚度。与先前的设想相反,已经表明,相对柔性且刚度较小的结构化表面显著提高了机械转导的效率以及所产生的神经元细胞和组织的质量。优选地,通过选择生物相容性聚合物或生物聚合物和/或其组成或制备方式来确定所述基质的结构化表面的刚度。众所周知,可以通过选择组成和/或交联方式来调节此类聚合物的机械性能。合适的聚合物材料如下所述。优选的是凝胶状聚合物基质。
令人惊讶地,发明人发现,使用本发明的基质的微米结构和纳米结构的表面的平行凹槽或沟槽或岭的非常特定的尺寸,可以实现所获得的分化神经元细胞和组织的特别高的分化效率和的质量。令人惊讶地,在这种情况下发现,低周期性微米结构与较高周期性纳米结构的高度周期性交替使得分化特别成功。即,纳米结构的多个周期被嵌入在微米结构的一个周期中。所述微米结构优选地由深度或高度为0.7至1.5μm且宽度为1至2μm的凹槽或沟槽或岭形成。所述微米结构的各个凹槽或沟槽或岭彼此之间的中心距离(周期)为3至12μm。已经表明,结构的尺寸在这里非常重要。在一特定实施例中,将微米结构的凹槽形成为沟槽。在一替代实施例中,将微米结构的凹槽制成相应尺寸的岭(阴模)。
嵌入上述微米结构中的纳米结构优选地由深度或高度为15至35μm且宽度为450至550μm的凹槽或沟槽或岭形成。纳米结构的各个凹槽或沟槽或岭彼此之间的中心距离(周期,即节距)为500至550μm。已经表明,结构的尺寸在这里非常重要。在一特定的实施方案中,将纳米结构的凹槽形成为沟槽。在一替代实施例中,将纳米结构的凹槽制成相应尺寸的岭(阴模)。

在第二方面,本发明提供了一种制备这种神经元诱导培养基质的新颖方法,其中使用基体的生物相容性聚合物或生物聚合物的微米结构化方法,特别是使用机械加工方法,在基质的基体上产生结构化表面。根据本发明,优选形成周期性微米结构和嵌入其中的周期性纳米结构,特别是在单个微结构化工艺步骤中一起形成。
本发明的微结构化方法是一种使用相对被加工材料运动的机械加工刀具的机械加工方法,优选地选自刨削、冲压和铣削。所述机械加工方法优选是一种使用固定的机械加工刀具和运动的待加工材料的方法,尤其是车削。所述微结构化方法尤其优选为用车刀进行车削。
在一个优选的实施例中,该机械加工用切削刀具进行,特别是刀片、车刀或铣刀,所述切削刀具的刀刃的尖端半径(rE)小于10μm,并且优选地由单晶金刚石构成。令人惊讶地发现,通过使用具有这种刀刃的这种切削刀具,可以以特别简单和可靠的方式获得本文所述的特定结构。纳米结构可以优选在单个工艺步骤中与微米结构一起生产。纳米结构直接取决于切削刀具的进给量(每转或每冲程)和尖端半径(rE)。
所述机械加工刀具的刀刃的尖端半径(rE)优选为5μm或小于5μm,2μm或优选小于2μm,或1μm或优选小于1μm,或500nm或小于500nm。在一个变型中所述机械加工工具是尖的,在另一个变型中,所述机械加工工具是刻面的。优选避免后缘切削。优选的是MDC切削刀具,其顶端角为1×30°,刀具半径(rE)为2μm或1μm。进给量为500到550nm,对应于要生产的纳米结构的周期(节距)。
在一个优选的变型中,通过该方法将该机械加工的微米结构直接施加到基质的聚合物基体上,以便直接获得根据本发明构造的神经元诱导表面。在替代的和优选的变型中,首先以这种方式首先在模具上形成将该表面结构,优选作为阴模,然后通过该表面结构化的模具由生物相容性聚合物或生物聚合物在基质的基体上产生实际的神经元诱导表面。优选的是,将所述基体的聚合物,优选地仍为液体形式,浇铸在表面结构化的模具上,也就是说,至少浇铸在微结构化之后在模具上形成的结构化表面上。在替代的和优选的变型中,用该模具对所述基体的聚合物进行压印,使得结构化表面在基体上形成。在每种情况下可选地使聚合物本身在该模具的结构化表面上固化或干燥,然后如此表面结构化的基质以固化和/或干燥的形式从模具上分离,尤其是剥离。在一个优选的实施方案中,在使用基质的另一步骤中,使固化的或特别是干燥的聚合物基体与液体,特别是培养基接触,从而通过溶胀和水合作用产生所需的最终形状和机械刚度。所述基体优选以干燥形式包装和存储,并且直到将其用于细胞培养时才进行溶胀和水合作用。该表面结构化的模具可重复使用以生产本发明的其他神经元诱导培养基质。通过这种压印或浇铸方法可以容易地大量生产培养基质。另外,简单的结构化方法允许提供表面积较大的神经元诱导培养基质,从而也可以生产大面积的神经元细胞聚集体和神经元组织。
在一种特殊的实施方式中,以此方式将表面结构化的模具设计为压印模或压印辊。本发明的结构化表面在液体或刚固化的聚合物凝胶上的压印与印刷技术中使用的相似。也可以以此方式产生用于生产神经元细胞或组织的大量和大面积的培养基质。
所述生物相容性聚合物应具有足够的亲水性以允许细胞粘附。优选地,所述生物相容性聚合物或生物聚合物选自包含胶原蛋白、琼脂糖、其衍生物及其半合成修饰物的生物材料,例如间位丙烯酸酯化明胶(GelMA)。优选在呈凝胶状之前先以液体形式存在的聚合物。特别优选冻干的胶原蛋白。或者优选的是间丙烯酸酯化的明胶(GelMA)。
特别优选的是具有神经转导形貌的基质的制备方法,其中将液体聚合物,特别是冻干的胶原蛋白溶液,浇铸到具有本发明的结构化表面的模具上,在那里固化并干燥。干燥时间取决于体积、表面和凝胶类型。干燥优选通过干燥器进行。在可以使用的基质基材中,本发明的模制的神经诱导形貌易于在可用的基质底物中保留数周。以这种方式,可以容易地制备大量基质以形成神经元细胞或组织,并可以储存使用。当接种细胞悬浮液时,干燥的凝胶基质溶胀,然后显示其最初压印的神经诱导表面结构。
特别地,诸如硅酮弹性体(RTV)或聚氨酯等弹性材料适合作为浇铸或压印模具。或者,可以使用低弹性或非弹性的柔性膜或刚性材料。优选提供金属作为用于浇铸或压印模具的材料。这些优选地选自有色金属,特别是铜、含铜合金或含镍合金。可替代地,优选提供塑料作为浇铸或压印模具的材料。
特别优选一种压印模具,该压印模具是金属或塑料圆柱体,其通过与本文所述的切削刀具的旋转而旋转,从而具有微米和纳米结构。在该优选实施例中,用于产生表面结构化基质的模制工艺是薄膜压印工艺。
在本发明的另一方面,本文所述的神经诱导基质用于将干细胞机械转导分化成神经元组织。
最后,在另一方面,本发明提供了一种由分离的生物细胞,特别是干细胞、多能细胞或可塑性细胞,生产神经元细胞或组织的新方法。该方法至少包括以下步骤:使分离的生物细胞与本发明的神经元诱导基质接触,然后在该基质的结构化表面上培养生物细胞,从而将这些细胞机械转导分化为神经元细胞和/或神经元组织,从而获得分化的神经元细胞和/或神经元组织。
优选地,嵌入该基质中的神经元细胞或组织可再次使用。在另一个实施方案中,用已知的方法将所述神经元细胞或神经元组织与基质分离。可选地,将神经元细胞与基质分离,然后分散成悬浮液。神经元细胞的悬浮液可以进一步培养。
本发明的培养基质还有利地允许从成体组织分化出少量的可塑性细胞。优选来自脂肪组织、骨髓和皮肤的细胞。还优选从脐带血获得的细胞。可分化为神经元细胞的分离的生物细胞优选为干细胞,优选为间充质干细胞(MSC);以此方式获得分化的MSC,尤其是自体MSC。例如,间充质干细胞可取自自体供体的脂肪组织并分离。此类细胞在治疗中的用途已获批准,并且在伦理上完全可以接受。直接用他自己的脂肪组织细胞治疗患者。
这些获得的神经元细胞或组织可以直接用于患者的治疗和预防中,尤其是用作患者特异性细胞,即用作医疗产品。
所获得的神经元细胞或组织还可以用作特别是患者特异性细胞,也可以用于药物开发,特别是还可以用于患者特异性药物的开发,以及用于患者的治疗和预防。

图2为本发明的神经转导培养基质的另一个变型的不同放大倍数的横截面示意图。在基质的基体(10)的一侧上形成结构化表面(20)。该结构化表面的特征在于具有平行的微米尺寸的凹槽(23)的周期性微米结构(22)和具有平行于微米结构(22)的平行纳米尺寸的凹槽(27)的周期性纳米结构(26),周期性纳米结构(26)嵌入微米结构(22)的凹槽(23)中。
图3A和3B为本发明的表面结构化的培养基质的电子显微照片的平面图(标有比例尺)。图3A示出了具有根据图1的示意图(不按比例)的结构的培养基质。
具体实施方式
干细胞机械转导分化为神经元细胞或神经元组织的整个过程包括:(a)生产具有本发明的形貌的阴图的母模;(b)将该形貌转移至可模制的材料;(c)可选地从母模产生进一步的印模;和(d)在模制的结构化材料上培养干细胞。
为了生产表面结构化的母模,将铜或镍(或相应合金)制成的圆柱滚子夹紧在车床中,并通过车刀以150min-1的转速用MDC刀刃进行车削,该MDC刀刃的顶端角为1×30°,顶端半径为1μm。引导机械加工刀具,以便在圆柱体上切割出宽度为1.2μm,间距为11.2μm的岭(形貌1)。替代地,设置进给量,以使得在圆柱体上产生宽度为1.24μm且间距为3.8μm的岭(形貌2)。所述岭形成微米结构。
对于机械加工,选择每转(或刀具行程)0.5mm的进给量。在金属圆柱体上的微米结构的岭之间,形成具有彼此间隔500nm的32nm沟槽(形貌1)或深度为19.15nm且彼此间隔40nm的沟槽(形貌2)的纳米结构。
以这种方式生产的表面结构化的母模作为压印辊被夹持在用于压印胶原薄膜的设备中。由新鲜倒入的冻干胶原蛋白制成的基体用作胶原蛋白薄膜。辊的沟槽结构被压入胶原蛋白薄膜的表面。随后,通过在干燥器中脱水来干燥具有结构化表面的胶原蛋白薄膜。
在替代方法中,通过用相应的机械加工刀具进行刨削来生产相应结构化的扁平金属板。该金属板用作模板。将新鲜制备的冻干胶原蛋白液体溶液浇铸在其上,并使其硬化。随后,将产生的胶原蛋白薄膜在干燥器中干燥约四天。薄膜收缩了百分之几。由于收缩,很容易将其从模板中移除。
在另一替代方法中,通过箔压印或通过金属母模浇铸来生产由硅酮弹性体制成的相应结构化的扁平浇铸模具。然后将该模具用作模板。
将冻干胶原蛋白的液体溶液浇铸在模具上并使其在那里硬化。随后,将产生的胶原蛋白薄膜在干燥器中干燥约四天。薄膜收缩了百分之几。由于收缩,很容易将其从模板中移除。或者,由于其弹性,将浇铸模具从固化或干燥的胶原蛋白凝胶中移除。
已经表明,压印的或浇铸的胶原蛋白薄膜可以在干燥后被存储数周。在当前情况下,在四周的储存时间之后将压印的薄膜润湿,从而形成包括本发明的结构化表面的原始形状。
新鲜印制或再水化的胶原蛋白薄膜用作培养基质。培养基质的表面结构在图3中示出。图3A和3B示出了用具有形貌1的模具压印的表面结构化的明胶基质(甲基丙烯酸酯化明胶)的SEM图像。以如本文以胶原蛋白基质为例所述的方式的相似方式制备表面结构化的明胶基质。图3A显示了用镍母模模制的明胶基质。图3B显示了用PDMS母模模制的明胶基质。
浇铸或压印的胶原蛋白薄膜和压纹的明胶基质都可以容易地以干燥状态保存至少三周,从而在溶胀凝胶时添加水或培养基后,可以获得最初压印的表面结构。
为了产生神经元细胞,将悬浮的源自成体组织的间充质干细胞接种到压印的明胶基质或胶原蛋白薄膜上,并培养四周。
培养的细胞沿着培养基质表面的微结构形成延伸和平行排列的形态。细胞迅速失去其原始的成纤维细胞样形状,几天后就可识别出典型的神经元细胞形态。这些细胞会形成很长的树突或轴突延伸,这决定了它们作为神经细胞的良好功能。
培养四周后,用胶原蛋白酶消化胶原蛋白基质中残留的胶原蛋白,从而可以温和地分离细胞而不会破坏其获得的形态。活力测试表明,胶原蛋白酶消化后,这些细胞的活力没有任何限制。在测定中,在基因和蛋白质水平上在细胞中检测到神经元标记。
Claims (5)
1.一种用于培养生物细胞并使之分化为神经元细胞或神经元组织的基质的制备方法,其特征在于,所述基质由具有结构化表面(20)的由生物相容性聚合物或生物聚合物组成的基体(10)组成,所述基质包括:
-由平行的微米尺寸的凹槽(23)组成的周期性微米结构(22),和
-由平行的纳米尺寸的凹槽(27)构成的周期性纳米结构(26),其平行于微米结构(22)延伸,并且分别嵌入微米结构(22)的凹槽(23)中,所述制备方法包括以下步骤:
-通过机械加工在模具上形成结构化表面,所述机械加工是利用机械加工刀具执行的,所述机械加工刀具的刀刃由单晶金刚石制成,尖端半径为10μm或更小,并且周期性微米结构(22)和嵌入其中的周期性纳米结构(26)形成阴模,以及
-在由生物相容性聚合物或生物聚合物组成的基体(10)上形成结构化表面(20):
-将基体(10)的聚合物浇铸在模具上,或
-用模具压印基体(10)的聚合物,和
-将模具上的基体(10)的聚合物干燥,并从模具上分离出干燥的基体(10),
从而获得具有结构化表面(20)的干燥的基体(10)作为基质。
2.根据权利要求1所述的方法,其特征在于,所述聚合物选自胶原蛋白、明胶和琼脂糖。
3.根据权利要求1或2所述的方法,其特征在于,所述聚合物在所述结构化表面具有的刚度等于或不大于在此处培养的生物细胞的刚度。
4.根据权利要求1所述的方法,其特征在于,所述微米结构(22)由深度为0.7至1.5μm,宽度为1至2μm,彼此之间的中心距离为3至12μm的凹槽(23)形成,并且所述纳米结构(26)由深度为15至35nm,宽度为450至550nm,彼此之间的中心距离为500至550nm的凹槽(27)形成。
5.根据权利要求1所述的方法,其特征在于,用表面结构压印模具(30)压印辊对所述基体(10)的聚合物进行压印,使得结构化表面(20)在基体(10)上形成。
