Abstract
本发明公开了一种螺旋藻的养殖方法,所述方法包括以下步骤,(1)、清洗养殖池,往养殖池内加入石灰水进行消毒;(2)、将制备好的螺旋藻培养液使用紫外杀菌装置对培养液进行杀菌处理,使杀菌过后培养液中微生物含量少于70cfu/毫升;然后加入到养殖池内;(3)、往养殖池内加入碳酸氢铵和过磷酸钙,其中碳酸氢铵和过磷酸钙的质量比为4.8:1.3;(4)、制备无机硒盐溶液;(5)、将螺旋藻的藻种洒入养殖池中,接种完成并自动搅拌均匀,定期清除杂藻和灰尘;养殖期间采用30‑1000W、时长1‑5分钟、频率15‑30KHz的超声波振动刺激。本发明不影响螺旋藻生长、又能够使得螺旋藻在培育过程中富含硒。

本发明涉及生物技术领域,具体涉及一种螺旋藻的养殖方法。
背景技术
螺旋藻属于蓝藻门,颤藻科的一种光能自养型生物,细胞内没有真正的细胞核,又称蓝细菌。经研究表明,螺旋藻中蛋白质含量高,氨基酸组成十分合理,其中8种必须氨基酸的含量接近FAO推荐的标准,是迄今为止科学家发现的最优秀的营养最全面、最均衡的纯天然蛋白质食品源。由于螺旋藻的细胞壁是由多糖组成,易于消化吸收,生物利用率达68%,同时螺旋藻中还含有丰富的维生素和多种微量元素,具有增强免疫、抗病毒、降低血糖、降胆固醇等功效。但螺旋藻的天然资源非常有限,所以国内外普遍通过人工养殖螺旋藻。
硒是联合国卫生组织所确定的人体所必需的微量元素,是人和动物体内谷胱甘肽过氧化物酶(glutathione per-oxidase,GSHPX)的辅助因子,GSHPX广泛存在于生命机体的肝、肾、心、肺等,硒代替半胱氨酸为该酶提供其活性中心的必需基团,GSHPX利用谷胱甘肽使有毒性的过氧化物还原为无害的羟基化合物,使过氧化物分解,清除活性自由基部位,保护细胞膜结构和功能,修复分子损伤。因此,硒元素具有抗氧化、抗癌、抗衰老、增强机体免疫力等多种生物学功能,传统的螺旋藻食品是在螺旋藻的后期加工中加入一些功能性原料复配(如富硒酵母提取物)来增强保健作用,这样就多了一道加工工序,而且也不易于加工。
发明内容
本发明所要解决的问题是:提供一种螺旋藻的养殖方法,不影响螺旋藻生长、又能够使得螺旋藻在培育过程中富含硒。
本发明为解决上述问题所提供的技术方案为:一种螺旋藻的养殖方法,所述方法包括以下步骤,
(1)、清洗养殖池,往养殖池内加入石灰水进行消毒;
(2)、将制备好的螺旋藻培养液使用紫外杀菌装置对培养液进行杀菌处理,使杀菌过后培养液中微生物含量少于70cfu/毫升;然后加入到养殖池内;
(3)、往养殖池内加入碳酸氢铵和过磷酸钙,其中碳酸氢铵和过磷酸钙的质量比为4.8:1.3;
(4)、制备无机硒盐溶液;称量水和亚硒酸钠,使亚硒酸钠充分溶解于水,得到亚硒酸钠稀释液,将亚硒酸钠稀释液加入到养殖池内;
(5)、将螺旋藻的藻种洒入养殖池中,接种完成并自动搅拌均匀,定期清除杂藻和灰尘;养殖期间采用30-1000W、时长1-5分钟、频率15-30KHz的超声波振动刺激。
优选的,所述步骤(2)中的螺旋藻培养液为0.5-1g/L的硝酸钠、0.05-0.25g/L的氯化钙、5-6g/L的小苏打、0-0.01g/L的食盐、0.7-0.85g/L的纯碱、0.6-0.7g/L的硝酸钠、0.04-0.05g/L的硫酸镁、0.46-0.53g/L的氯化钾、0.13-0.17g/L的磷酸、0.012-0.014g/L的硫酸亚铁和0.02-0.06g/L的尿素。
优选的,所述步骤(4)中将亚硒酸钠粉末放入水中进行溶解,亚硒酸钠与水的重量比为80:1,将亚硒酸钠和水进行均匀混合。
与现有技术相比,本发明的优点是:本发明在培养的过程中加入无机硒盐溶液,螺旋藻在生长的过程中将无机硒盐溶液生物集成成有机硒多糖成分,使得通过本发明培育成的螺旋藻富含有机硒,有机硒体系中的有机硒多糖是基本无毒的有机硒产品,即使过量食用也不会对人体自身造成损害;通过加入碳酸氢铵肥料和过磷酸钙肥料作为辅助培育营养源,提升了培养液的营养的均衡性;培养过程中能够对杂藻进行有效的控制,同时培养成本低、可以大规模培养。

(2)、将制备好的螺旋藻培养液使用紫外杀菌装置对培养液进行杀菌处理,使杀菌过后培养液中微生物含量少于70cfu/毫升;然后加入到养殖池内,其中,螺旋藻培养液为0.5g/L的硝酸钠、0.05g/L的氯化钙、5g/L的小苏打、0.7g/L的纯碱、0.6g/L的硝酸钠、0.04g/L的硫酸镁、0.46g/L的氯化钾、0.13g/L的磷酸、0.012g/L的硫酸亚铁和0.02g/L的尿素;
(3)、往养殖池内加入碳酸氢铵和过磷酸钙,其中碳酸氢铵和过磷酸钙的质量比为4.8:1.3;
(4)、制备无机硒盐溶液;称量水和亚硒酸钠,亚硒酸钠与水的重量比为80:1,使亚硒酸钠充分溶解于水,得到亚硒酸钠稀释液,将亚硒酸钠稀释液加入到养殖池内;
(5)、将螺旋藻的藻种洒入养殖池中,接种完成并自动搅拌均匀,定期清除杂藻和灰尘;养殖期间采用30W、时长1分钟、频率15KHz的超声波振动刺激。
实施例2
一种螺旋藻的养殖方法,所述方法包括以下步骤,
(1)、清洗养殖池,往养殖池内加入石灰水进行消毒;
(2)、将制备好的螺旋藻培养液使用紫外杀菌装置对培养液进行杀菌处理,使杀菌过后培养液中微生物含量少于70cfu/毫升;然后加入到养殖池内,其中,螺旋藻培养液为0.8g/L的硝酸钠、0.1g/L的氯化钙、5.5g/L的小苏打、0.005g/L的食盐、0.75g/L的纯碱、0.65g/L的硝酸钠、0.045g/L的硫酸镁、0.5g/L的氯化钾、0.15g/L的磷酸、0.013g/L的硫酸亚铁和0.04g/L的尿素;
(3)、往养殖池内加入碳酸氢铵和过磷酸钙,其中碳酸氢铵和过磷酸钙的质量比为4.8:1.3;
(4)、制备无机硒盐溶液;称量水和亚硒酸钠,亚硒酸钠与水的重量比为80:1,使亚硒酸钠充分溶解于水,得到亚硒酸钠稀释液,将亚硒酸钠稀释液加入到养殖池内;
(5)、将螺旋藻的藻种洒入养殖池中,接种完成并自动搅拌均匀,定期清除杂藻和灰尘;养殖期间采用500W、时长3分钟、频率20KHz的超声波振动刺激。
实施例3
一种螺旋藻的养殖方法,所述方法包括以下步骤,
(1)、清洗养殖池,往养殖池内加入石灰水进行消毒;
(2)、将制备好的螺旋藻培养液使用紫外杀菌装置对培养液进行杀菌处理,使杀菌过后培养液中微生物含量少于70cfu/毫升;然后加入到养殖池内,其中,螺旋藻培养液为1g/L的硝酸钠、0.25g/L的氯化钙、6g/L的小苏打、0.01g/L的食盐、0.85g/L的纯碱、0.7g/L的硝酸钠、0.05g/L的硫酸镁、0.53g/L的氯化钾、0.17g/L的磷酸、0.014g/L的硫酸亚铁和0.06g/L的尿素;
(3)、往养殖池内加入碳酸氢铵和过磷酸钙,其中碳酸氢铵和过磷酸钙的质量比为4.8:1.3;
(4)、制备无机硒盐溶液;称量水和亚硒酸钠,亚硒酸钠与水的重量比为80:1,使亚硒酸钠充分溶解于水,得到亚硒酸钠稀释液,将亚硒酸钠稀释液加入到养殖池内;
(5)、将螺旋藻的藻种洒入养殖池中,接种完成并自动搅拌均匀,定期清除杂藻和灰尘;养殖期间采用1000W、时长5分钟、频率30KHz的超声波振动刺激。

Claims (3)
1.一种螺旋藻的养殖方法,其特征在于:所述方法包括以下步骤,
(1)、清洗养殖池,往养殖池内加入石灰水进行消毒;
(2)、将制备好的螺旋藻培养液使用紫外杀菌装置对培养液进行杀菌处理,使杀菌过后培养液中微生物含量少于70cfu/毫升;然后加入到养殖池内;
(3)、往养殖池内加入碳酸氢铵和过磷酸钙,其中碳酸氢铵和过磷酸钙的质量比为4.8:1.3;
(4)、制备无机硒盐溶液;称量水和亚硒酸钠,使亚硒酸钠充分溶解于水,得到亚硒酸钠稀释液,将亚硒酸钠稀释液加入到养殖池内;
(5)、将螺旋藻的藻种洒入养殖池中,接种完成并自动搅拌均匀,定期清除杂藻和灰尘;养殖期间采用30-1000W、时长1-5分钟、频率15-30KHz的超声波振动刺激。
2.根据权利要求1所述的一种螺旋藻的养殖方法,其特征在于:所述步骤(2)中的螺旋藻培养液为0.5-1g/L的硝酸钠、0.05-0.25g/L的氯化钙、5-6g/L的小苏打、0-0.01g/L的食盐、0.7-0.85g/L的纯碱、0.6-0.7g/L的硝酸钠、0.04-0.05g/L的硫酸镁、0.46-0.53g/L的氯化钾、0.13-0.17g/L的磷酸、0.012-0.014g/L的硫酸亚铁和0.02-0.06g/L的尿素。
3.根据权利要求1所述的一种螺旋藻的养殖方法,其特征在于:所述步骤(4)中将亚硒酸钠粉末放入水中进行溶解,亚硒酸钠与水的重量比为80:1,将亚硒酸钠和水进行均匀混合。
