Abstract
本发明公开了一种布鲁氏菌液体气溶胶肺递送感染小鼠模型的制备方法。本发明方法包括如下步骤:通过液体气溶胶的方式将布鲁氏菌A19递送至动物的肺部,得到动物模型。本发明通过布鲁氏菌A19液体气溶胶肺递送感染BALB/c小鼠,成功建立小鼠布病模型。A19引起BALB/c小鼠慢性感染,感染的小鼠出现脾肺载菌多,持续时间长,脾肿大明显等毒理反应;同时激发了细胞免疫为主的免疫反应,感染过程经历了Th1向Th2的转变。本发明对于初步认识布鲁氏菌气溶胶感染小鼠引起的免疫毒理反应,为进一步研究其感染机制和开展疫苗研发奠定基础。

背景技术
布鲁氏菌(Brucella)是一种可以导致人兽共患传染病-布鲁氏菌病(简称布病)的革兰氏阴性胞内寄生菌[PAPPAS G,AKRITIDIS N,BOSILKOVSKI M,et al.Brucellosis[J].New England Journal of Medicine,2005,352(22):2325.],根据宿主的不同分为10个种,其中以牛、羊、猪种感染发病最常见[AHMED W,ZHENG K,LIU Z F.Establishment ofChronic Infection:Brucella's Stealth Strategy[J].Frontiers in Cellular&Infection Microbiology,2016,6(83)],在我国牛种感染率高[满腾飞,王大力,崔步云,等2009年伞国布鲁氏菌病监测数据分析[J].疾病监测,2010,25(12):944-946]。其感染途径多样:呼吸道、消化道和皮肤接触,其中以气溶胶形式经呼吸道感染是最常见的感染途径[李金岭.布鲁氏菌病的流行特点、症状及危害[J].兽医导刊,2010,(3):32-32]。
布鲁氏菌感染动物能导致流产、不育,给畜牧业造成严重的经济损失;感染人能引起发热、盗汗、疲劳,关节炎,心内膜炎,脑炎,肝脾脓肿等全身性疾病[PAPPAS G,AKRITIDISN,BOSILKOVSKI M,et al.Brucellosis[J].New England Journal of Medicine,2005,352(22):2325.KUZMAN I,et al.Human brucelosis;an overview oftreatment;proceedings of the Croatian Congress on Infectious Diseaseszagreb,F,2006[C].]。目前认为,免疫预防是防止布病最有效的方法[丁家波,毛开荣,程君生,等布氏杆菌病疫苗的应用和研究现状[J].微生物学报,2006,46(5):856-859.],疫苗的使用显著降低了动物布病的发生,但很多疫苗有干扰血清学诊断和使动物致病的弊端[叶俊贤,冯宇,陈瑞爱,等.布鲁氏菌S19疫苗应用及研究进展[J].中国兽药杂志,2017,51(5):62-67.],因此克服这些弊端,开展研发安全有效的疫苗,是人们努力的方向,而对布鲁氏菌感染致病后发展规律的认识是新型疫苗研发的前提。
研究感染性疾病的致病机制,寻找有效的防控措施,需要建立理想的动物模型,静脉接种被认为是制备小鼠感染模型最常用的途径[SZABO E K,MACCALLUM D M.Thecontribution of mouse models to our understanding of systemic candidiasis[J].Fems Microbiology Letters,2011,320(1):1–8.]。气溶胶感染是布鲁氏菌较为常见的感染途径,不同感染途径对机体的影响存在差异,不同来源的同一株菌毒副作用存在很大差异,因此建立布鲁氏菌感染模型,确定菌株的毒力、感染途径和剂量是关键。
发明内容
本发明的目的是提供一种布鲁氏菌液体气溶胶肺递送感染小鼠模型的制备方法。
第一方面,本发明要求保护一种制备布鲁氏菌液体气溶胶肺递送感染动物模型的方法。
本发明所提供的制备布鲁氏菌液体气溶胶肺递送感染动物模型的方法,具体可包括如下步骤:通过液体气溶胶的方式将布鲁氏菌A19递送至动物的肺部,得到动物模型。
进一步地,所述动物可为小鼠,如BALB/c小鼠,具体如BALB/c雌性小鼠。
进一步地,所述布鲁氏菌A19的递送量可为108CFU/体重为0.018-0.02kg的所述动物。如每只小鼠108CFU(每只小鼠体重0.018-0.02kg)。
进一步地,所述“通过液体气溶胶的方式将布鲁氏菌A19递送至动物的肺部”可为将所述布鲁氏菌A19通过液体气溶胶肺递送装置递送至所述动物的肺部。
在本发明的一个实施例中,具体为借助喉镜,使用手持式液体气溶胶肺递送装置将所述布鲁氏菌A19菌液递送至所述动物的肺部。
更进一步地,所述布鲁氏菌A19菌液具体为将所述布鲁氏菌A19悬浮在含有0.05%(体积百分含量)泊洛沙姆的生理盐水中得到的菌液。
其中,0.05%(体积百分含量)泊洛沙姆是为了增加菌的分散性,防止细菌结块,以这个比例的浓度配比对菌的分散性最好。

其中,所述布鲁氏菌A19可以菌液形式存在。所述菌液具体可为将布鲁氏菌A19悬浮在含有0.05%(体积百分含量)泊洛沙姆(如泊洛沙姆188)的生理盐水中得到的菌液。更加具体地,所述布鲁氏菌A19菌液中所述布鲁氏菌A19的含量可为5×109~4.5×1010CFU/ml(如5×109CFU/ml)。
所述液体气溶胶肺递送装置可为手持式液体气溶胶肺递送装置。
第五方面,本发明要求保护所述试剂盒在制备布鲁氏菌液体气溶胶肺递送感染动物模型中的应用。
其中,所述动物可为小鼠,如BALB/c小鼠,具体如BALB/c雌性小鼠。
本发明以BALB/c小鼠为模式实验动物,构建布鲁氏菌液体气溶胶肺递送感染模型,以脾肿大、脾肺载菌量、血清抗体和细胞因子的消长变化等指标来评价气溶胶感染小鼠布病的发生发展规律。结果显示:本发明通过手持式液体气溶胶发生装置,使布鲁氏菌A19液体气溶胶化,肺递送感染BALB/c小鼠,成功建立气溶胶感染小鼠布病模型。A19引起BALB/c小鼠慢性感染,感染的小鼠出现脾肺载菌多,持续时间长,脾肿大明显等毒理反应;同时激发了较强的细胞免疫和体液免疫反应,以细胞免疫为主,感染过程经历了Th1向Th2的转变。本发明中,布鲁氏菌通过气溶胶途径易感性被用作生物战剂,本发明制备气溶胶途径的毒理感染模型,对布鲁氏菌气溶胶感染机体的毒理反应及选择有效的对抗措施提供参考依据,对气溶胶免疫能否对同种途径的感染起到预防保护作用提供研究依据,同时为进一步研究其感染机制和开展疫苗研发奠定基础。

具体实施方式
下述实施例中所使用的实验方法如无特殊说明,均为常规方法。
下述实施例中所用的材料、试剂等,如无特殊说明,均可从商业途径得到。
实施例1、布鲁氏菌液体气溶胶肺递送感染动物模型的制备及鉴定
一、实验材料
1、实验动物与菌株来源
6~8周龄无特定病原体(SPF级)BALB/c雌性小鼠,18~20g(北京维通利华实验动物技术有限公司,许可证号SCXK(京)2016-0011)。牛种布鲁氏菌A19为新疆天牧公司产品。
2、主要试剂与仪器
BBLTM Brucella Agar、BBLTM Brucella Broth、TSA Tryptic Soy Agar和TSBTryptic Soy Broth培养基,美国BD公司;Brucella selective supplement,英国OXOID公司;ELISA试剂盒,深圳达科为生物技术有限公司;细胞因子检测试剂盒,美国e Bioscience公司。小动物喉镜,北京慧荣和科技有限公司;手持式液体气溶胶肺递送装置,北京慧荣和科技有限公司。
二、实验方法
1、布鲁氏菌A19的菌液制备
取20μl布鲁氏菌A19甘油菌接种于20ml的Brucella Broth中,于37℃摇床200rpm振荡培养18h左右至对数中期(OD600≈1.0),20倍稀释转接于20ml的Brucella Broth中,继续于摇床振荡培养10h左右至对数中期(OD600≈1.0),离心集菌,用含0.05%(体积百分含量)泊洛沙姆188(Poloxamer 188(Pluronic F68)|泊咯沙姆188(普朗尼克F68)sigma公司,CAS号:9003-11-6)的生理盐水洗菌2次后再次重悬,调OD600使得菌浓度约为5×109CFU/ml用于后期小鼠感染模型建立。另取少量菌液进行梯度稀释后涂板计数,计算实际菌浓度,使得建模所用菌液的实际菌浓度在5×109~4.5×1010CFU/ml范围内(本实验菌液实际浓度为5×109CFU/ml)。
2、感染模型建立
将制备好的菌液,经肺递送和滴鼻接种小鼠。实验设肺递送组和滴鼻组,并分别针对不同途径设阴性对照组(接种等体积的重悬液,即含体积百分含量为0.05%的泊洛沙姆188的生理盐水)。即实验分组如下:【A19肺递送组(Z1):肺递送接种布鲁氏菌A19。空白对照肺递送组(Z2):肺递送接种菌体重悬液。A19滴鼻组(Z3):滴鼻接种布鲁氏菌A19;空白对照滴鼻组(Z4):滴鼻接种菌体重悬液】。30只/组。肺递送组和滴鼻组,均需腹腔注射1%戊巴比妥160μl~180μl/只麻醉,3分钟后小鼠夹捏背部皮肤无反应,说明小鼠麻醉状态良好,可用于后续实验。滴鼻组:用20μl量程的移液枪,调整至20μl,吸取菌液(使用前再次混匀)左手抓住小鼠背部,翻转,使小鼠腹侧朝上,头高位倾斜,直接缓慢滴入小鼠鼻孔内,滴速不宜过快,以免引起小鼠呛咳反应或窒息而死。肺递送组:借住镊子,夹住小鼠舌头,向口外牵拉,打开喉镜,顺小鼠口咽方向探伸,可见到随呼吸一张一合的会厌,在会厌张开时,用备好的手持式气溶胶肺递送装置深入咽喉到达气管,并快速推进一个卡扣(50μl,把步骤1中的的菌液稀释2.5倍,使得每只小鼠的菌株递送量为108CFU)量程,小鼠接种完毕,均头高位放置,以免菌液通过小鼠气管倒流,保种接种剂量的准确。接种后3h左右小鼠苏醒,未见死亡。实验过程中给予小鼠充足的食物和水,每天12h光照。注:实验过程中,滴鼻组和肺递送组两组菌液是同时制备的,把调好的菌液(浓度为5×109CFU/ml)一分为二,一份10ml直接用于滴鼻,每只小鼠20μl;一份4ml菌液加入6ml 0.05%(体积百分含量)泊洛沙姆188的生理盐水,即用含0.05%(体积百分含量)泊洛沙姆188的生理盐水稀释2.5倍,用于肺递送,每只小鼠递送50μl。
3、临床体征的观察和体重的测定
分别于第0周(接种前)、接种后第2、4、6、8周观察并记录小鼠病征(耸毛、呼吸困难、强迫性腹部呼吸、被触摸或外部刺激后反应迟钝);测定体重。
4、布鲁氏菌A19菌株在小鼠肺脏、脾脏内的定居力检测
无菌分离脾脏和肺脏,称重,匀浆,5倍梯度稀释成不同浓度,每个稀释度设3个重复,10μl/孔涂布于方板TSA培养基,37℃恒温倒置培养3~4天,计数。
5、小鼠血清抗体IgG、IgM、IgA、IgG2a和IgG1检测
分别于第0周(接种前)、接种后第2、4、6、8周,每组取6只小鼠,摘眼球取血,将血液于4℃静置过夜,3000rpm离心5min,收集血清,-20℃保存备用。按照ELISA试剂盒说明书操作方法,测定感染后小鼠血清抗体滴度。
6、小鼠血清、肺匀浆细胞因子检测
匀浆后的肺组织3000rpm离心5min,收集血清,-20℃保存备用。按照细胞因子检测试剂盒说明书,测定感染前和感染后2、4、6、8周,血清、肺匀浆液细胞因子(IFN-γ和TNF-α)浓度的变化。
7、数据统计分析
应用SAS统计软件分析数据。阴性对照组与实验组之间的比较采用t检验,实验组之间的比较采用单因素方差分析,P<0.05,表示差异有统计学意义。
三、实验结果
1、小鼠临床表现、体重和脾重测定
与空白对照组Z2、Z4相比,Z1、Z3组小鼠状态均良好,无耸毛、呼吸困难、强迫性腹部呼吸、反应迟钝等表现。图1中A所示:Z2、Z3和Z4小鼠体重持续平稳上升,Z1小鼠体重在感染6周后有持续下降趋势,检测时间内组间差异均无统计学意义。图1中B所示:在第2周,Z1脾脏质量达到峰值,约0.4g,与Z2比较,P<0.05,有统计学意义;Z3脾重有明显升高,约0.2g,与Z4比较,P<0.05,有统计学意义;Z1明显高于Z3,P<0.05,有统计学意义。在第4周,与第2周比较,Z1脾重下降,约0.2g,仍显著高于Z2,P<0.05,有统计学意义;Z3脾重达到峰值,约0.3g,与Z4相比,P<0.05,有统计学意义;Z3大于Z1,P<0.05,有统计学意义。在第6周,Z1和Z3均持续迅速下降,组间差异无统计学意义;Z1大于Z2,Z3大于Z4,P<0.05,有统计学意义。第8周,组间差异均无统计学意义。上述实验结果表明与滴鼻途径相比,肺递送感染组出现较早较明显的脾肿大。
2、脾脏和肺脏载菌量检测
如图2所示,在0周,脾肺载菌量未检出,在第2、4、6、8周,Z1与Z2相比,Z3与Z4相比,差异均有统计学意义(P<0.05)。Z1和Z3均在第2周达到峰值107CFU/g,之后有持续下降的趋势,在第4周,Z3脾肺载菌量显著高于Z1(P<0.05),有统计学意义,第8周,Z1脾载菌量显著高于Z3(P<0.05),有统计学意义,第2周和第6周两组脏器载菌量差异不显著(P>0.05)。
3、血清抗体水平的检测
如图3所示,与空白对照组相比,Z1和Z3血清抗体均在感染后2到8周内维持较高水平,Z1和Z3之间血清抗体水平差异无统计学意义(P>0.05)。本研究对血清中IgG2a/IgG1的比值进行了分析,感染后2周Z3血清IgG2a/IgG1值高于Z1组(P<0.005),有统计学意义,第8周Z1组高于Z3组(P<0.05),有统计学意义,两组IgG2a/IgG1值在感染后2到8周内均大于1,呈下降趋势。
4、细胞因子的检测
如图4所示,与空白对照组相比,Z1和Z3血清和肺匀浆均产生了高浓度细胞因子IFN-γ,血清在第2周达到峰值,肺匀浆在第4周达到峰值,Z1峰值均高于Z3(P<0.05),有统计学意义。血清TNF-α在第2达到峰值,Z1高于Z3(P>0.05),没有统计学意义,Z1在4周后出现上升趋势,Z3持续下降;肺匀浆则在第6周达到峰值,Z1显著高于Z3(P<0.05),阴性对照组Z2和Z4细胞因子浓度低于下限值。
5、小结
脏器载菌和脾肿大是检测布鲁氏菌毒力的重要指标。本发明中,两种感染途径(肺递送和滴鼻)对应的小鼠肺脏载菌量(CFU/g)的峰值之间无显著差异。两种途径感染小鼠脾脏载菌量(CFU/spleen)的峰值接近,但由于肺递送组脾肿大明显,单位质量脾脏载菌量(CFU/g)小于滴鼻组。肺递送组比滴鼻组出现较早较明显的脾脏肿大,细胞因子浓度也显著高于滴鼻组,说明肺递送气溶胶感染途径对小鼠的毒理作用更加显著,是一种有效的感染途径。
两种途径感染后,小鼠均未表现出典型病征,在感染的时间点内体重差异没有统计学意义,但肺递送组在6周后有明显下降趋势,但无统计学差异,这可能是因为布病本身是一种消耗性疾病,本实验小鼠感染8周后脾脏菌仍大量存在,未被清除的菌在体内增殖,持续感染引起的。脾肺载菌量在感染后2周持续下降,说明获得性免疫反应发挥了重要作用。其中Th1为主的免疫反应以释放细胞因子和炎症介质发挥杀伤胞内菌的作用,Th2通过抗体的中和作用减少细菌对组织细胞的黏附和侵袭,对清除胞外菌发挥作用,同时Th2细胞释放的细胞因子抑制Th1细胞分泌炎症因子,减轻免疫炎症损伤,同时诱导机体免疫反应失能,Th1、Th2平衡对于抗感染和减轻炎症创伤起重要作用。布鲁氏菌是胞内寄生菌,细胞免疫(Th1)发挥关键作用,体液免疫(Th2)也是必不可少的。本发明检测了抗体滴度及细胞因子浓度的动态变化,结果表明:与空白对照组比较,感染后不同时间点小鼠血清中IgG、IgM、IgA均有不同程度的升高,并在8周内未见明显下降,说明以108CFU布鲁氏菌A19感染BALB/c小鼠后,机体产生较强的体液免疫反应。感染后2到8周,IgG2a/IgG1≥1,IgG2a/IgG1值呈逐渐下降趋势,提示本发明使用布鲁氏菌A19感染小鼠,感染初期细胞免疫占优势,感染过程中向体液免疫偏移。胞内菌因其胞内寄生的特点,体液免疫的保护作用有限,抗感染作用以细胞免疫为主。细胞免疫反应依赖细胞因子的参与,IFN-γ能强化和促进Th1免疫应答,使TNF-α分泌增加,TNF-α引起发热,体重减轻,组织坏死等典型的中毒症状和炎症反应。本发明结果表明布鲁氏菌A19感染早期机体分泌细胞因子IFN-γ、TNF-α,二者均在第2周达到峰值,之后持续下降,变化趋势与肺、脾脏载菌量变化趋势基本一致,这提示胞内布鲁氏菌的清除可能与介导细胞毒和免疫杀伤作用的细胞因子有关,随着体内菌的清除,这两种细胞因子显著下降。两种途径比较,肺递送组细胞因子IFN-γ和TNF-α均显著高于滴鼻组,说明肺递送诱发的细胞免疫可能更强,也因此导致脾肿大更明显。肺匀浆细胞因子IFN-γ、TNF-α均在第4周或第6周达到峰值,肺递送组均高于滴鼻组,可能是由于肺递送途径直接将菌递送至肺脏,局部淋巴细胞发挥作用,滴鼻途径经过了鼻腔粘膜免疫和排异,感染肺脏后,引发局部淋巴细胞释放细胞因子略少。随着炎症因子的下降,免疫反应向Th2的转变,一方面减轻免疫炎症损伤,一方面有利于胞内布鲁氏菌的增殖,导致感染慢性化,这与人布病的发展规律一致。抗原的持续刺激诱导T细胞无能,免疫耐受,这可能是导致胞内菌在体内持续感染的重要原因之一。
Claims (10)
1.一种制备布鲁氏菌液体气溶胶肺递送感染动物模型的方法,包括如下步骤:通过液体气溶胶的方式将布鲁氏菌A19递送至动物的肺部,得到动物模型。
2.根据权利要求1所述的方法,其特征在于:所述动物为小鼠。
3.根据权利要求1或2所述的方法,其特征在于:所述布鲁氏菌A19的递送剂量为108CFU/体重为0.018-0.02kg的所述动物。
4.根据权利要求1-3中任一所述的方法,其特征在于:所述通过液体气溶胶的方式将布鲁氏菌A19递送至动物的肺部为将所述布鲁氏菌A19的菌液通过液体气溶胶肺递送装置递送至所述动物的肺部。
5.根据权利要求4所述的方法,其特征在于:所述布鲁氏菌A19的菌液为将布鲁氏菌A19悬浮在含有体积百分含量为0.05%的泊洛沙姆的生理盐水中得到的菌液。
6.根据权利要求5所述的方法,其特征在于:所述泊洛沙姆为泊洛沙姆188。
7.权利要求1-6中任一所述方法制备的动物模型在筛选布鲁氏菌病的治疗药物或疫苗中的应用。
8.权利要求1-6中任一所述方法制备的动物模型在研究布鲁氏菌病的发病机制和/或研究布鲁氏菌病的发生发展规律中的应用。
9.一种用于制备布鲁氏菌液体气溶胶肺递送感染动物模型的试剂盒,含有布鲁氏菌A19和液体气溶胶肺递送装置。
10.权利要求9所述的试剂盒在制备布鲁氏菌液体气溶胶肺递送感染动物模型中的应用。
