在基因突变形式中,单核苷酸多态性(SNPs)是最常见的基因变异形式,与多种遗传性疾病、癌症和药物治疗等密切相关,因此,SNPs作为新一代生物标志物用于遗传病和癌症的预防、诊断和治疗得到了广泛的关注。现阶段常用SNPs基因分型检测方法包括等位基因特异性酶技术、等位基因特异性结合荧光技术等。等位基因特异性酶技术是一种基于引物特异性杂交或延伸和酶侵入性切割的方法,这种检测方法虽然在室温下也能提供较为灵敏和准确的检查,但通常需要复杂的步骤,测定的程序繁琐,检测中酶活性不稳定以及修饰引物价格昂贵等缺点。等位基因特异性结合荧光技术,如分子信标、二元探针、碱基区分荧光探针和立足介导的链置换反应,提供了一种新型的简单、直接和无酶参与的鉴定特定核酸序列的技术。然而,大多数这种类型的方法在室温下灵敏度低或特异性差,并且有效地鉴别SNPs通常需要使用专门的设备,训练有素的人员,精确的环境温度控制和严格的实验条件。因此,开发一种简单,快速、无标记、易于携带、能在室温条件下对SNPs进行灵敏度高和选择性强的基因分型和定量操作的生物传感器,以简化检测过程,降低检测费用,满足现场实际应用中的需求是极为重要的。
肽核酸(PNA)是DNA的非天然类似物,其中整个脱氧核糖磷酸二酯骨架已被电中性聚酰胺骨架取代,并且肽核酸优于标准核酸寡聚体的特性如下:肽核酸可以在低盐浓度下通过Watson-Crick碱基配对特异性地与带负电荷的DNA或RNA杂交;肽核酸和DNA或RNA复合物具有更高的热稳定性;肽核酸具有抵抗核酸酶和蛋白酶的降解,完全匹配的DNA/肽核酸双链体中的肽核酸可以有效地保护单链DNA(ssDNA)免受特异性核酸酶的切割;肽核酸还可以通过非共价或共价相互作用吸附在纳米材料上,构建灵敏的生物传感器,并且其优异的稳定性和特异性结合核酸的能力使PNA成为普通环境中诊断工具的出色候选者。
近年来以DNA为模板制备的银纳米团簇(DNA-AgNCs)具有高量子产率,强光稳定性和小尺寸等优势。模板DNA序列的变化,影响着合成出银纳米团簇的尺寸、电子和光学性质和组装行为,并且有时甚至赋予其新的功能,使得合成出的银纳米簇荧光发射光谱随之变化。其中,以dC12(即12个C的DNA模板)为模板合成的银纳米簇可以与巯基产生荧光响应,使得AgNCs的荧光大幅度增强。可能的两种解释如下:第一可能是AgNCs和硫醇化合物之间通过Ag-S键的形成使配体到金属中心产生电荷转移(LMNCT),以及硫醇化合物中的其他富电子基团(-COOH,-NH2)的影响,均有可能增强荧光信号;另一种可能的解释是由于添加硫醇化合物改变了dC12模板的微环境,使其形成更紧凑的dC12结构,为AgNCs提供更好地保护,从而最大限度地减少溶液对AgNCs的荧光猝灭,导致荧光增强。本发明在巯基肽核酸存在下,由于呈电中性的单链肽核酸吸附到AgNCs表面,以及巯基基团与AgNCs的荧光响应机制,双重作用诱导AgNCs的荧光强度显著增强。相反,巯基肽核酸与DNA双链则不会导致AgNCs的荧光增强效应。因此,巯基功能化肽核酸与AgNCs之间这种特殊的相互作用,具有无标记、低成本且灵敏度高的优点,该材料有望应用于化学生物传感和生物成像等领域,例如检测特定的基因突变,如TP53基因。
TP53基因是迄今发现的与人类肿瘤相关性最高的基因、是防止正常细胞变成癌细胞的一个重要的抑癌基因,因此,TP53基因被称作“基因卫士”,其编码的TP53蛋白是一类重要的细胞周期因子和肿瘤抑制因子,在阻止肿瘤细胞分裂、修复DNA损伤、老化细胞、诱导细胞死亡等方面发挥着重要的作用。然而,突变的TP53基因所表达的蛋白质,在结构与功能上存在一定缺陷,失去了原有的抑制肿瘤细胞生长的生物学特性,并促进其持续大量地恶性转化,从而导致恶性肿瘤的产生。研究表明,在各种形式的人类癌症中,TP53基因是最常见的突变基因。据统计数据显示,75%的癌症患者都存在TP53基因突变。TP53的SNPs与近一半以上的癌症发生有关,包括肺癌、胃癌、肝癌、膀胱癌、食管癌、乳腺癌、宫颈癌等多种癌症。因此,高选择性和灵敏度的TP53基因的SNPs的分型和定量,对于多种遗传相关风险评估、疾病诊断、药物开发具有重大的临床指导和实际应用价值。
发明内容
本发明的一方面提供了一种巯基功能化肽核酸增强银纳米簇荧光信号检测单核酸多态性的方法,所述方法包括以下步骤:
1)获得N端巯基基团修饰的目标肽核酸探针;
2)获得dC12为模板合成的银纳米簇探针;
3)将所述目标肽核酸探针和银纳米簇探针混合得到第一混合溶液;将所述目标肽核酸探针与待测DNA片段进行杂交,然后与银纳米簇探针混合得到第二混合溶液;
4)在相同条件下测定荧光光谱,分别获取第一混合溶液的第一光谱信息和第二混合溶液的第二光谱信息;对比第一光谱信息和第二光谱信息,鉴别碱基错配。
在优选的实施方式中,当待测DNA片段含碱基突变时,所述目标肽核酸探针与所述DNA片段形成弱稳定性双链;
当待测DNA片段不含碱基突变时,所述目标肽核酸探针与所述DNA片段形成强稳定性双链。
在优选的实施方式中,所述步骤1)包括:
通过固相多肽合成技术制备目标肽核酸寡聚体,并在所述目标肽核酸的末端进行巯基基团的修饰。
在优选的实施方式中,所述步骤2)中包括:
使dC12模板中的两个胞嘧啶碱基C与银溶液中的Ag+反应形成C-Ag+-C复合物,然后使所述复合物在强还原剂溶液中形成AgNCs。
在优选的实施方式中,其特征在于,所述银溶液为硝酸银溶液;所述强还原剂为硼氢化钠。
在优选的实施方式中,在所述步骤3)中,所述目标肽核酸探针与待测DNA片段杂交时,所述目标肽核酸与待测DNA的摩尔比为1:1。
在优选的实施方式中,在所述步骤3)中,在第一混合溶液和第二混合液中,所述银纳米簇探针的浓度为1.5~2.5μM;
在第二混合溶液中,目标肽核酸探针与待测DNA片段杂交得到的双链浓度为0.05~1μM。
在优选的实施方式中,在所述步骤4)中,所述第一光谱信息为荧光信号,所述第二光谱信息为荧光信号。
在可选的实施方式中,在所述步骤3)中,目标肽核酸探针与DNA杂交的退火温度为90℃,加热时间为3min,然后在1小时内缓慢冷却至室温;杂交结束后加入AgNCs后,在室温下避光反应15min。
本发明的方法中,肽核酸与DNA混合溶液于90℃加热3min后缓慢冷却至室温即可实现碱基互补配对,通常情况下室温缓慢冷却30min至1h。
在优选的实施方式中,所述方法还包括,将所述目标肽核酸探针与不同浓度的完全互补配对的DNA片段标准品进行杂交,建立标准曲线来测试所述方法灵敏度的步骤;
优选地,所述不同浓度的目标DNA浓度分别为:0.05,0.1,0.2,0.4,0.6,0.8和1.0μM,肽核酸探针浓度为1μM。
所述含目标DNA片段标准品可以购买或根据需要自行合成。
含目标突变基因的DNA片段标准品,所述目标肽核酸序列如SEQ ID NO.1所示,银纳米簇探针的模板DNA序列如SEQ ID NO.2所示,不含突变的待测DNA片段的基因序列如SEQID NO.3所示,Mutant C突变序列如SEQ ID NO.4所示,Mutant A突变序列如SEQ ID NO.5所示,Mutant T突变序列如SEQ ID NO.6所示。所述序列如表1所示。

a在目标DNA和PNA探针序列中,突变碱基的位置在方框中突出显示。
本发明的方法中,巯基肽核酸-DNA双链与银纳米簇探针(AgNCs)混匀室温反应15min,由于完全互补的巯基肽核酸与目标DNA杂交形成稳定的双链,使肽核酸与AgNCs之间无相互作用,AgNCs的荧光强度不变;当目标DNA存在单碱基突变时,DNA与巯基肽核酸形成的双链稳定性弱,肽核酸与AgNCs之间非共价吸附作用增强,导致AgNCs的荧光显著增强,从而实现了SNPs的快速检测。当步骤4)中的比对中第二光谱信息和第一光谱信息吻合时,则说明待测目标DNA中不含SNPs,否则说明待测目标DNA中含有SNPs。
优选地,所述第一光谱信息为荧光信号,所述第二光谱信息为荧光信号。
波长646nm处的荧光强度直接反应AgNCs荧光强度变化,而本专利中AgNCs荧光强度变化程度是由SNPs造成的,单碱基突变的DNA强度高,完全匹配的目标DNA链强度低,这个强度大小的对比可作为SNPs区分度的衡量。
优选地,巯基功能化肽核酸探针的合成过程如下:利用固相多肽合成技术进行肽核酸的合成。将目标肽的第一个单体的羧基以共价键的形式与固体载体(MBHA树脂)的氨基相连,固定到树脂上。再以这一单体的氨基为合成起点,使其与相邻单体的羧基发生酰化反应,形成肽键。然后让包含有这两个单体的树脂肽的氨基与下一个单体的羧基反应,不断重复这一过程,直至目标肽核酸形成为止,其N端以氨基结束。取2.926mL 3-巯基丙酸,加入30mL二甲基甲酰胺(DMF),磁力搅拌混匀。随后同时加入9.8g 1-(3-二甲氨基丙基)-3-乙基碳二亚胺(EDC)和5.865g N-N-羟基琥珀酰亚胺(NHS),在氮气保护条件下搅拌30min,然后密封搅拌过夜,即得到3-巯基丙酸活性酯(HSCH2CH2CO-NHS),将溶液置于4℃避光静置12h。取600μL加入到上述N端以氨基结尾的肽核酸中,震荡反应过夜,即得到的N端巯基修饰的目标肽核酸寡聚体。将目标肽核酸从树脂上裂解、纯化、冻干、定量,最终得到巯基肽核酸探针;
优选地,DNA(dC12)为模板的银纳米簇(AgNCs)的制备如下:在磷酸盐缓冲溶液中(2mM,pH7.4)加入dC12和新制硝酸银(AgNO3)溶液,然后置于冰水中反应15min。在30s内快速加入新鲜配制的硼氢化钠(NaBH4)溶液进行还原,震荡2分钟,并在室温下避光保存3h。最后,将溶液在4℃下保存过夜,即得到dC12为模板合成的AgNCs。dC12,AgNO3和NaBH4的最终浓度分别为10、60和60μM(摩尔比为1:6:6)。
本发明还提供了一种目标肽核酸探针、一种银纳米簇探针。
本发明的又一方面提供了所述目标肽核酸探针、银纳米簇探针以及上述方法在化学生物传感和生物成像等领域的应用。
本申请能产生的有益效果包括:
1)本申请中合成的N端修饰巯基的目标肽核酸探针的方法,具有路线简单,合成方便,产物稳定性高,易纯化等优点。
2)本申请提供的巯基功能化肽核酸增强银纳米簇荧光信号检测单核酸多态性的方法,具有无标记、低成本且灵敏度高和选择性强的特点。本方法可以通过简单地改变特异性肽核酸探针的序列来检测任何种类的基因突变。
3)本申请建立的巯基功能化肽核酸银纳米簇探针,具有合成简单,荧光稳定,响应快的优点,可作为生物传感平台,广泛应用于DNA、蛋白质、酶、金属离子等的分析和定量。

具体实施方式
下面结合实施例详述本申请,但本申请并不局限于这些实施例。
如无特别说明,本申请实施例中原料均通过商业途径购买,其中肽核酸探针通过固相肽合成技术制备,DNA序列材料购自上海杰李生物技术有限公司,所有核酸序列均通过紫外-可见吸收光谱仪进行精确定量,所有的玻璃仪器都用王水浸泡24h,并用双蒸水清洗,晾干备用。实验中使用的水均为18.2MΩ的Milli-Q超纯水。MBHA树脂购自上海希施生物科技有限公司。
实施例1
1.巯基功能化肽核酸的合成:利用固相多肽合成技术进行肽核酸的合成。将目标肽的第一个单体的羧基以共价键的形式与固体载体(MBHA树脂)的氨基相连,固定到树脂上。再以这一单体的氨基为合成起点,使其与相邻单体的羧基发生酰化反应,形成肽键。然后让包含有这两个单体的树脂肽的氨基与下一个单体的羧基反应,不断重复这一过程,直至目标肽核酸形成为止,其N端以氨基结束。取2.926mL 3-巯基丙酸,加入30mL二甲基甲酰胺(DMF),磁力搅拌混匀。随后同时加入9.8g 1-(3-二甲氨基丙基)-3-乙基碳二亚胺(EDC)和5.865g N-N-羟基琥珀酰亚胺(NHS),在氮气保护条件下搅拌30min,然后密封搅拌过夜,即得到3-巯基丙酸活性酯(HSCH2CH2CO-NHS),将溶液置于4℃避光静置12h。取600μL加入到上述N端以氨基结尾的肽核酸中,震荡反应过夜,即得到的N端巯基修饰的目标肽核酸寡聚体。将目标肽核酸从树脂上裂解、纯化、冻干、定量,最终得到巯基肽核酸探针;
2.以DNA(dC12)为模板的银纳米簇(AgNCs)的制备:在磷酸盐缓冲溶液中(2mM,pH7.4)加入dC12和新制硝酸银(AgNO3)溶液,然后置于冰水中反应15min。在30s内快速加入新鲜配制的硼氢化钠(NaBH4)溶液进行还原,震荡2min,并在室温下避光保存3h。最后,将溶液在4℃下保存过夜,即得到dC12为模板合成的AgNCs。dC12,AgNO3和NaBH4的最终浓度分别为10μM、60μM和60μM(摩尔比为1:6:6)。
3.TP53基因的不同单碱基突变类型的荧光光谱分析:野生型DNA(DNAWT)序列为本发明采用了中间位置的突变,其中G改变为C-C(G→C),C-A(G→A)和C-T(G→T)。首先分别将巯基肽核酸(1μM)与不同突变类型的目标DNA在磷酸盐缓冲液中混合,总体积为20μL;将混合物加热至90℃保持3min,然后在1h内缓慢冷却至室温;最后,向溶液中加入浓度为2μM的AgNCs,在室温下避光反应15min;最后在在LS55荧光光谱仪(PerkinElmer)上进行荧光测试,获得荧光光谱。
4.TP53基因的单碱基突变的检测限;使用具有不同浓度的完全互补配对的目标DNA(0.05,0.1,0.2,0.4,0.6,0.8和1.0μM)加入巯基肽核酸(1μM)和AgNCs(2μM)中评估该生物传感器检测单碱基错配的灵敏度。
图1为本实施例中使用荧光激发/发射光谱测定合成出的AgNCs的光学特征,其中曲线由下至上分别表示AgNCs+PNA-SH和AgNCs。AgNCs最大荧光激发/发射峰分别为585和646nm。将终浓度为1μΜ的PNA-SH加入到2μM的AgNCs溶液中时。荧光强度大幅度增强,与纯浓度为2μM的AgNCs溶液相比,荧光强度约为其3倍,该现象可能是由于配体-金属-金属电荷转移(LMMCT)或配体-金属电荷转移(LMCT)从S原子到Ag原子以及通过金属中心三重态的辐射弛豫。
图2为本实施例中只有完全匹配的野生型DNA(C→G)才能导致荧光信号不变,而在3个不同错配碱基DNA(C-C(G→C),C-A(G→A)和C-T(G→T))中,观察到银纳米簇明显的荧光信号增强,且显示出相似的强度,这表明在检测单个不匹配碱基时,可忽略不匹配的碱基类型变化。
图3至图4为本实施例中银纳米簇荧光强度随目标DNA的浓度增大而降低,原因是加入目标DNA与巯基肽核酸形成肽核酸/DNA双链体。在图3中,曲线从下至上依次表示浓度:0.05μM、0.1μM、0.2μM、0.4μM、0.6μM、0.8μM和1.0μM。巯基肽核酸单链可导致银纳米簇溶液荧光强度大幅度提升,完全匹配的目标DNA的加入将通过形成肽核酸/DNA双链体的量来降低单链PNA-SH的量。PNA/DNA双链体含量越多,PNA-SH单链越少,银纳米簇溶液的荧光强度越低。在646nm处的荧光强度和目标DNA浓度之间存在线性相关性(R2=0.988)。这些数据显示使用巯基肽核酸结合银纳米簇构建的生物传感器对SNP的分型和定量分析具有高灵敏度和良好的线性关系。
以上所述,仅是本申请的几个实施例,并非对本申请做任何形式的限制,虽然本申请以较佳实施例揭示如上,然而并非用以限制本申请,任何熟悉本专业的技术人员,在不脱离本申请技术方案的范围内,利用上述揭示的技术内容做出些许的变动或修饰均等同于等效实施案例,均属于技术方案范围内。

Claims (10)
1.一种巯基功能化肽核酸增强银纳米簇荧光信号检测单核酸多态性的方法,其特征在于,所述方法包括以下步骤:
1)获得N端巯基基团修饰的目标肽核酸探针;
2)获得dC12为模板合成的银纳米簇探针;
3)将所述目标肽核酸探针和银纳米簇探针混合得到第一混合溶液;将所述目标肽核酸探针与待测DNA片段进行杂交,然后与银纳米簇探针混合得到第二混合溶液;
4)在相同条件下测定荧光光谱,分别获取第一混合溶液的第一光谱信息和第二混合溶液的第二光谱信息;对比第一光谱信息和第二光谱信息,鉴别碱基错配。
2.根据权利要求1所述的巯基功能化肽核酸增强银纳米簇荧光信号检测单核酸多态性的方法,其特征在于,
当待测DNA片段含碱基突变时,所述目标肽核酸探针与所述DNA片段形成弱稳定性双链;
当待测DNA片段不含碱基突变时,所述目标肽核酸探针与所述DNA片段形成强稳定性双链。
3.根据权利要求1所述的巯基功能化肽核酸增强银纳米簇荧光信号检测单核酸多态性的方法,其特征在于,所述步骤1)包括:
通过固相多肽合成技术制备目标肽核酸寡聚体,并在所述目标肽核酸的末端进行巯基基团的修饰。
4.根据权利要求1所述的巯基功能化肽核酸增强银纳米簇荧光信号检测单核酸多态性的方法,其特征在于,所述步骤2)中包括:
使dC12模板中的两个胞嘧啶碱基C与银溶液中的Ag+反应形成C-Ag+-C复合物,然后使所述复合物在强还原剂溶液中形成AgNCs。
5.根据权利要求4所述的巯基功能化肽核酸增强银纳米簇荧光信号检测单核酸多态性的方法,其特征在于,所述银溶液为硝酸银溶液;
所述强还原剂为硼氢化钠;
优选地,所述步骤3)中,所述目标肽核酸探针与待测DNA片段杂交时,所述目标肽核酸与待测DNA的摩尔比为1:1。
优选地,所述步骤3)中,
在第一混合溶液和第二混合液中,所述银纳米簇探针的浓度为1.5~2.5μM;
在第二混合溶液中,目标肽核酸探针与待测DNA片段杂交得到的双链浓度为0.05~1μM。
优选地,所述步骤4)中,所述第一光谱信息为荧光信号,所述第二光谱信息为荧光信号。
6.根据权利要求1所述的巯基功能化肽核酸增强银纳米簇荧光信号检测单核酸多态性的方法,其特征在于,所述方法还包括,将所述目标肽核酸探针与不同浓度的完全互补配对的DNA片段标准品进行杂交,建立标准曲线来测试所述方法灵敏度的步骤;
优选地,所述不同浓度的目标DNA浓度分别为:0.05,0.1,0.2,0.4,0.6,0.8和1.0μM,肽核酸探针浓度为1μM。
7.根据权利要求1所述的巯基功能化肽核酸增强银纳米簇荧光信号检测单核酸多态性的方法,其特征在于,所述目标肽核酸序列如SEQ ID NO.1所示,银纳米簇探针的模板DNA序列如SEQ ID NO.2所示,不含突变的待测DNA片段的基因序列如SEQ ID NO.3所示,Mutant C突变序列如SEQ ID NO.4所示,Mutant A突变序列如SEQ ID NO.5所示,Mutant T突变序列如SEQ ID NO.6所示。
8.一种目标肽核酸探针,其特征在于,所述目标肽核酸探针通过权利要求3所述的方法制备。
9.一种银纳米簇探针,其特征在于,所述目标银纳米簇探针通过权利要求4所述的方法制备。
10.根据权利要求8所述的目标肽核酸探针、根据权利要求9所述的银纳米簇探针、权利要求1至7中任一项所述的巯基功能化肽核酸增强银纳米簇荧光信号检测单核酸多态性的方法在化学生物传感和生物成像领域的应用。
