Abstract
本发明提供了FGF‑2纳米抗体作为蛋白质和/或多肽保护剂的应用,所述的FGF‑2纳米抗体为申请人筛选得到的纳米抗体,其氨基酸序列如中国专利申请CN201810573403.3的SEQ ID NO:31所示。本发明发现所述的纳米抗体可有效地保持处于较宽温度范围、较宽pH范围或机械力等状态下的蛋白质或多肽(如FGF‑2)的稳定,能够显著提高蛋白质和/或多肽在多种化学或物理环境因素中的稳定性;并且不会对FGF‑2生物学活性产生负面影响。同时,本发明还发现所述的纳米抗体与其他蛋白保护剂联用时,能进一步增强其对蛋白质和/或多肽的保护作用,降低外源物质的不良影响,对蛋白质和/或多肽的生产具有重要意义。
Description
技术领域
本发明属于生物医药领域,特别涉及FGF-2纳米抗体在蛋白质和/或多肽保护剂的应用。
背景技术
传统抗体分子由两条相同的重链(H链)和两条相同的轻链(L链)组成,这一基本结构在哺乳动物中都非常保守。1993年,研究人员首次报道了骆驼血中存在着天然缺失轻链的奇特抗体,它只有重链,缺少轻链,即重链抗体(heavy-chain antibody,HCAb)。通过基因工程的办法克隆其可变区即可得到抗原抗体结合的最小抗体片段,即单域抗体(variabledomain of heavy chain of heavy-chain antibody,VHH),亦称为“纳米抗体”。纳米抗体分子量很小,约为传统抗体分子量的1/10,仅有15KD,远远小于Fab段(60KD)和普通抗体(150KD),纳米抗体分子量小这一特性为其广泛应用奠定了基础。
纳米抗体由于其特殊的结构和性质,具有许多独特的优势:(1)首先其分子量比较小,结构简单,所以纳米抗体的制备相对更加容易,重组纳米抗体能在大肠杆菌、酵母等微生物中大量表达,易于大规模生产。(2)纳米抗体具有高的水溶性。(3)纳米抗体能够耐受高温且具有高度的构象稳定性,此外,纳米抗体还具有可逆的去折叠能力,例如,90℃高温条件下长时间处理传统单克隆抗体和纳米抗体,发现纳米抗体仍然保持较高的抗原结合活性,而传统抗体都丧失活性。此外在蛋白酶和极度pH值条件下(如胃液和内脏中),正常抗体会失效或分解,而纳米抗体仍具有高度的稳定性。(4)纳米抗体具有高的抗原结合性,由于纳米抗体能够识别独特的抗原表位,比普通抗体具有更广泛的抗原结合能力,且其与抗原的结合能力更强。(3)纳米抗体分子量小,没有Fc段,不会引起CMC反应,所以对人体的免疫原性较弱,与人有较好的生物相容性。(6)纳米抗体具有很好的组织穿透能力,可以有效的穿透血脑屏障,这一特性为脑部新药研发提供了新的方向。
由于有诸多优点,所以纳米抗体在医疗领域具有广泛的应用价值,(1)纳米抗体可以用于对肿瘤进行治疗,纳米抗体与小分子药物偶联可以达到更好的治疗效果,例如纳米抗体与β-内酰胺酶相连接,能够选择性的激活抗癌前体药物,有效的与结肠癌细胞(LS174T)上的癌胚抗原相结合,在原位杀伤肿瘤,且无毒副作用。此外,纳米抗体也可以用于癌症的诊断。(2)纳米抗体在体内可以迅速扩散并达到体内组织中毒素聚集的地方,具有很好的中和毒素的作用,可以用于抗毒素治疗方面。(3)纳米抗体可以穿透血脑屏障,在中枢神经系统疾病治疗中也发挥重要作用。(4)此外,纳米抗体还可以应用与消化系统疾病的治疗。(5)由于纳米抗体与抗原高亲和力、高稳定性,目前纳米抗体也广泛应用于疾病的诊断方面,例如纳米抗体应用于禽流感的检测,牙龈卟啉单胞菌、布鲁菌的鉴别方面。
蛋白质具有一级、二级、三级及四级结构,往往蛋白质发挥生物学活性需要形成正确的空间构象,一些蛋白质由于特殊的氨基酸组成或者分子量较大,稳定性较差,保存困难,甚至有些蛋白冷冻干燥之后,复溶困难,如何保持蛋白质的稳定性是蛋白质在应用中需要克服的难题之一。针对溶液剂型的蛋白质和多肽类药物,通过添加无毒可溶的外源物也是一种保护蛋白质的方法,增强蛋白质稳定性的方法主要有以下几种:(1)可以通过优化蛋白溶液中的成分,添加糖类和醇类物质等保护剂,如甘油、蔗糖等;在溶液中会与蛋白质产生排斥作用,使蛋白质构象保持更紧密的状态,但浓度过高或过低都会使作用改变,一般在0.3~1M用量为宜。比如:分别在不同浓度条件下,蔗糖可以稳定核酸酶,也可以抑制白介素二聚体的形成和盐酸胍诱导条件下干扰素的聚合。(2)此外还可以对蛋白进行化学修饰,例如PEG修饰等方法;(3)氨基酸和蛋白多肽。在不影响目的蛋白的情况下,向溶剂中加入某些氨基酸或外源蛋白质可以起到稳定蛋白药物的作用。例如,人血清白蛋白作为蛋白大分子添加物,一般用做疫苗冻干粉的保护剂。(4)也可以通过优化蛋白质制剂的剂型,例如利用冷冻干燥技术使蛋白质以固态的形式保存,降低了分子在介质中的热运动以及分子间的相互作用,提高了蛋白的稳定性。还可以通过脂质体包裹等方式来提高蛋白质的稳定性。
成纤维细胞生长因子-2(fibroblast growth factor-2,FGF-2)最初由研究人员从牛脑组织和牛脑垂体中分离纯化获得,它对Balb/c 3T3成纤维细胞有促分裂增殖作用,且等电点呈碱性(pH=9.6),故也称其为碱性成纤维细胞生长因子。在病理条件下,FGF-2在黑色素瘤、乳腺癌、肺癌、膀胱癌等肿瘤组织中都存在高表达,它与肿瘤的发生发展、肿瘤转移、血管生成及预后有着密切的关系。
在正常情况下,FGF-2在机体内主要分布于垂体、脑、神经组织、视网膜、肾上腺和胎盘等部位,它对血管生成、损伤组织修复、创伤愈合、胚胎发育与分化等过程起着重要的调节作用。FGF-2也广泛应用与创伤修复方面,目前也有含有FGF-2的凝胶药物用于治疗烧伤、烫伤方面。但是,由于FGF-2是具有生物活性的大分子,极易因多种环境因素而影响其稳定性;同时,FGF-2自身容易发生聚合,对保持其药物活性的稳定产生了很大影响。如何提高FGF-2作为蛋白药物的稳定性,降低潜在变质风险,有效地延长FGF-2类药物的保存时间,对开发多种剂型的药物具有重要意义。
发明内容
本发明的首要目的在于克服现有技术的缺点与不足,提供FGF-2纳米抗体作为蛋白质和/或多肽保护剂的应用。
本发明的另一目的在于提供一种含有FGF-2纳米抗体的蛋白质和/或多肽组合物。
本发明的又一目的在于提供一种提高蛋白质和/或多肽的稳定性的方法。
本发明的目的通过下述技术方案实现:
FGF-2纳米抗体作为蛋白质和/或多肽保护剂的应用,所述的FGF-2纳米抗体为申请人筛选得到的纳米抗体,其氨基酸序列如中国专利申请CN201810573403.3的SEQ ID NO:31所示。
一种含有所述的FGF-2纳米抗体的蛋白质和/或多肽组合物,所述的FGF-2纳米抗体为所述的蛋白质和/或多肽的保护剂。
所述的蛋白质优选为成纤维细胞生长因子-2(FGF-2);所述的FGF-2纳米抗体可以作为FGF-2的保护剂和/或增效剂。
所述的蛋白质和/或多肽组合物优选为含有蛋白质药物或多肽药物的蛋白质和/或多肽组合物。
所述的蛋白质和/或多肽组合物优选为液态的蛋白质和/或多肽组合物,或者是液体冻干后储藏,但使用前需要复溶为液体的蛋白质和/或多肽组合物。
所述的蛋白质和/或多肽组合物还可以包括其他蛋白保护剂。
所述的其他蛋白保护剂优选为蔗糖、甘油、右旋糖酐40、肝素钠、海藻糖、甘露醇中的至少一种。
所述的其他蛋白保护剂的终浓度优选为0.03mol/L以上。
一种提高蛋白质和/或多肽稳定性的方法,在所述的蛋白质和/或多肽中添加所述的FGF-2纳米抗体。
所述的FGF-2纳米抗体的终浓度优选为1nM~70nM;进一步优选为4nM~64nM。
所述的蛋白质和/或多肽优选为液态下的蛋白质和/或多肽,或者是液体冻干后储藏,但使用前需要复溶为液体的蛋白质和/或多肽。
所述的蛋白质和/或多肽可以为处于pH为5~11、温度为4℃~60℃或相当于不超过转速1000r/min的震荡外力作用等至少一种情况下的蛋白质和/或多肽。
进一步的,所述的蛋白质和/或多肽为处于所述的温度为4℃~40℃的蛋白质和/或多肽。
本发明相对于现有技术具有如下的优点及效果:
1.本发明发现具有FGF-2高亲和力的纳米抗体Nb4可有效地保持处于较宽温度范围(如4℃~40℃)、较宽pH范围(pH=5~11)或机械力(如高速搅拌)等状态下的蛋白质或多肽(如FGF-2)的稳定,能够显著提高蛋白质和/或多肽在多种化学或物理环境因素中的稳定性。
2.本发明的纳米抗体Nb4在对FGF-2的稳定性提供有效保护的同时,不会对FGF-2生物学活性产生负面影响。
3.本发明的纳米抗体Nb4与其他蛋白保护剂联用时,能进一步增强其对蛋白质和/或多肽的保护作用,降低外源物质的不良影响。
4.本发明的纳米抗体Nb4及提高蛋白质和/或多肽稳定性的方法可为蛋白质和/或多肽的运输和保存提供新的思路,对蛋白质和/或多肽的生产具有重要意义。

具体实施方式
下面结合实施例及附图对本发明作进一步详细的描述,但本发明的实施方式不限于此。
以下实施例中的纳米抗体Nb4与Nb6为发明人团队筛选得到的抗重组人碱性成纤维细胞生长因子纳米抗体,其氨基酸序列分别如中国专利申请CN201810573403.3的SEQ IDNO:31、SEQ ID NO:33所示。
参照中国专利申请CN201810573403.3“一种抗重组人碱性成纤维细胞生长因子纳米抗体及其应用”实施例1的方法或本领域常规技术获得所述的纳米抗体Nb4或Nb6,进行以下实施例的研究。
实施例1纳米抗体Nb4与FGF-2亲和力测定
酶联免疫吸附实验检测Nb4与FGF-2(购自北京义翘神州科技有限公司,货号10014-HNAE)亲和力,具体实验步骤是用ELISA板每孔加入100μL、4μg/mL FGF-2,4℃包被过夜。PBS-T洗板3次,每次5min,分别加入不同浓度的Nb4(以2μg/mL为起始浓度,1:4倍比稀释10个梯度),室温孵育2h。PBS-T洗板3次,每次5min,每孔加入100μL Anti-Flag-HRP抗体(购自SIGMA公司,货号A8592),室温孵育1h,PBS-T洗板3次,每次5min。分别在450nm和630nm处测出吸光值,通过origin8.0分析结果。
Nb4与FGF-2亲和力曲线如图1所示,通过前述的酶联免疫吸附实验检测FGF-2与纳米抗体Nb4的亲和力,EC50为3.6ng/mL,说明Nb4与FGF-2有极高的亲和力。
实施例2纳米抗体Nb4对FGF-2稳定性的影响
由于FGF-2蛋白的稳定性受pH、温度的影响比较大,因此,本实施例主要研究在不同pH、不同温度条件下,FGF-2纳米抗体Nb4对FGF-2的稳定性的影响。实验分为3组:

按照表1.的分组,每个组别下均设实验组和对照组,对实验组FGF-2与Nb4混合物(终浓度均为200μg/mL)和对照组样品终浓度为200μg/mL的FGF-2分别进行经不同pH、不同温度和不同处理时间的实验研究。处理完成后,western blot检测FGF-2的蛋白水平。
具体实验步骤如下:
(1)处理之后的样品加入5×蛋白上样缓冲液,100℃加热6分钟,12%SDS-PAGE电泳。(2)蛋白电泳结束后,转膜夹上依次放上海绵垫、滤纸、NC膜、蛋白胶、滤纸和海绵垫,蛋白胶靠近转膜槽阴极一端,250mA恒流转膜2小时。(3)转膜结束后,NC膜用5%脱脂牛奶封闭1小时,加入Anti-Flag-HRP抗体(购自SIGMA公司,货号A8592),4℃孵育过夜;再用PBST洗膜3次,每次5分钟;ECL发光试剂盒(购自赛默飞,货号32209)显影,记录并分析实验结果。western blot分析FGF-2蛋白水平。
结果表明,(1)处理时间会影响FGF-2的降解,相同处理温度条件下,FGF-2放置时间越长,蛋白降解越多(图2(A)和图2(B))。(2)溶液的pH对FGF-2的稳定性影响很大,pH越高,FGF-2稳定性越差,容易形成多聚体;FGF-2溶液的pH=11的条件下,FGF-2降解明显加快,4℃处理28天和25℃处理13天时,FGF-2几乎完全降解;但是加入纳米抗体Nb4的处理组,在pH=11的条件下能够很好的提高FGF-2的稳定性(图2(B)和图2(C))。
实施例3不同温度条件下,纳米抗体对FGF-2稳定性的影响
通过ELISA方法检测处理之后的FGF-2的浓度,具体实验步骤是参考中国专利申请CN201810574975.3“抗人bFGF纳米抗体检测bFGF的方法及试剂盒”中实施例3所构建的基于抗人bFGF纳米抗体的双夹心ELISA法。
首先将FGF-2稀释成一系列浓度(400ng/mL、300ng/mL、200ng/mL、100ng/mL、80ng/mL、40ng/mL、20ng/mL、10ng/mL),绘制ELISA方法测定FGF-2浓度的标准曲线,结果如图3所示,FGF-2浓度在10ng/mL至400ng/mL浓度范围内样品浓度与吸光值呈线性关系。
所述的双夹心ELISA法检测FGF-2的浓度的具体步骤如下:
(1)包板:用包被液将Nb6蛋白配制成终浓度为5μg/mL的溶液,在96孔酶标板中,每孔加入100μL,放置在4℃环境下过夜。
(2)洗板:将0.5%PBST加入96孔酶标板中,每孔300μL,静置3min后弃掉液体。
(3)封闭:配制5%脱脂牛奶用于封闭酶标板,每孔300μL,在37℃孵育2h。
(4)洗板:将0.5%PBST加入96孔酶标板中,每孔300μL,静置3min后弃掉液体。
(5)加样:通过前述绘制的标准曲线,以不加样品的1×PBS为阴性对照,并加入经不同条件处理的样品,每孔100μL,每个浓度设三个复孔,在37℃孵育1h。
(6)洗板:将0.5%PBST加入96孔酶标板中,每孔300μL,静置3min后弃掉液体,重复该步骤5次。
(7)加入Anti-bFGF抗体(购自Abcam公司,货号ab8880):用0.5%BSA稀释Anti-bFGF,稀释比例为1:2000,混匀后加入96孔酶标板中,每孔100μL,在37℃孵育1h。
(8)洗板:将0.5%PBST加入96孔酶标板中,每孔300μL,静置3min后弃掉液体,重复该步骤3次。
(9)加入Anti-rabbit-HRP(购自Abcam公司,货号ab6721):用0.5%BSA稀释Anti-rabbit-HRP,稀释比例为1:5000,混匀后加入96孔酶标板中,每孔100μL,在37℃孵育1h。
(10)洗板:将0.5%PBST加入96孔酶标板中,每孔300μL,静置3min后弃掉液体,重复该步骤3次。
(11)显色:用TMB显色液以每孔100μL加入96孔酶标板,在37℃孵育10min。
(12)终止:用2.29%硫酸终止反应,以每孔100μL加入96孔酶标板,分别在酶标仪450nm和630nm波长处测吸光值。
设置如下组别进行实验研究:
(1)FGF-2+Nb4组:FGF-2蛋白溶液(终浓度为200μg/mL)中加入终浓度为200μg/mL的纳米抗体Nb4后混匀;
(2)FGF-2组:终浓度为200μg/mL的FGF-2蛋白溶液;
以上样品均以1×PBS作为样品溶剂。
当pH=9时,FGF-2+Nb4组和FGF-2组分别经过4℃、25℃、37℃和40℃处理28天,通过前述ELISA方法测定溶液中的FGF-2蛋白浓度,结果如表2所示,结果表明:蛋白活性随着温度升高而降低,FGF-2+Nb4组的FGF-2蛋白浓度均高于FGF-2组,两组浓度差值也随温度上升而变大,说明FGF-2蛋白溶液中加入Nb4能够显著提高其稳定性。

实施例4不同pH条件下,纳米抗体Nb4能够显著提高FGF-2的稳定性
本实施例中FGF-2+Nb4组与FGF-2组的设置同实施例3。
pH值对蛋白质稳定性有极大影响,不合适的pH会导致蛋白质聚集、沉淀,以及蛋白活性的丧失。我们比较了FGF-2+Nb4组和FGF-2组在不同pH条件(pH=5、pH=7、pH=9、pH=11)下,37℃保存7天后,通过western blot检测FGF-2蛋白水平,根据western blot的检测结果,用Image J软件进行灰度分析,如图4所示,结果表明,在pH值碱性条件下,FGF-2的稳定性显著下降;在pH=11条件下,FGF-2完全检测不到;但是加入纳米抗体Nb4之后,在碱性条件下,能够显著提高FGF-2的稳定性(图4)。
实施例5纳米抗体Nb4在不同搅拌速率条件下对FGF-2的稳定性的影响
本实施例中FGF-2+Nb4组与FGF-2组的设置同实施例3。
蛋白质稳定性会受到蛋白分子间运动强度的影响,例如温度会加速分子间的运动,从而影响蛋白质的稳定性,此外,蛋白在溶液状态下,溶液的搅拌速度也会影响蛋白质的稳定性。FGF-2+Nb4组与FGF-2组分别在25℃,pH=7的条件下,搅拌速度分别为500r/min和1000r/min处理6h和24h之后,通过实施例3的ELISA方法测定溶液中FGF-2蛋白浓度,以及western blot检测FGF-2蛋白水平。
结果如表3和图5所示,从处理时间上看,FGF-2+Nb4组相对于FGF-2组在6h的搅拌震荡下,没有显著性差异,而在处理24h后,FGF-2溶液中加入Nb4能显著提高FGF-2的稳定性。

实施例6纳米抗体Nb4联合不同蛋白稳定剂对FGF-2稳定性的影响
向溶液剂型的蛋白药物中添加蛋白保护剂是维持蛋白药物稳定的重要方式之一,常见的蛋白保护剂有糖类、醇类物质等,比如:蔗糖、海藻糖、甘露醇、甘油;右旋糖酐40常用于疫苗佐剂。
按实施例3设置FGF-2+Nb4组与FGF-2组,分别加入不同类型和浓度的蛋白保护剂,pH=9、40℃条件下处理7天,通过实施例3的ELISA方法测定溶液中FGF-2的蛋白浓度。

结果如表4和图6所示,表明FGF-2蛋白溶液中添加蔗糖、甘油、右旋糖酐40、肝素钠对FGF-2稳定性有一定的保护作用,同时,纳米抗体Nb4与这些蛋白保护剂联合使用后,对FGF-2稳定性的保护作用有了进一步显著的提高。单独使用海藻糖、甘露醇对FGF-2的保护作用不明显,较高浓度的海藻糖甚至对FGF-2的稳定不利。但加入本发明的纳米抗体Nb4后,对FGF-2稳定性的保护作用有大幅度的提高;令人意外地,纳米抗体Nb4甚至可以逆转较高浓度的海藻糖对FGF-2稳定性的不利影响,并在一定程度上增强了纳米抗体Nb4自身对FGF-2的保护作用。
实施例7纳米抗体Nb4对FGF-2活性的影响研究
FGF-2中分别加入终浓度为1nM、4nM、16nM、64nM的Nb4,以及只有FGF-2不加纳米抗体Nb4作为阴性对照组,样品分别在4℃、pH=11条件下处理7天时间,细胞划痕实验检测FGF-2对细胞的促增殖效果。
细胞划痕实验步骤如下:(1)BALB/C-3T3细胞(-CCL-163TM,购自ATCC)密度为2.5×105个/mL,每孔2mL加入六孔细胞培养板中,37℃、5%CO2细胞培养箱中培养24h。(3)待细胞密度达到60%时,用大小均一的力将200μL枪头在6孔培养板中划井字样横线,保持划痕宽度一致。(4)吸去培养基,并加入用PBS轻洗细胞一次,再吸去PBS,从而吸去飘起的细胞。(5)用基础培养基(0.02%FBS+DMEM/F12+10μg/mL人转铁蛋白+200μg/mL BSA)分别配制不同浓度FGF-2和Nb4样品,即:14.7ng/mL FGF-2+1nM Nb4、14.7ng/mL FGF-2+4nM Nb4、14.7ng/mL FGF-2+16nM Nb4、14.7ng/mL FGF-2+64nM Nb4,每个样品两个复孔,每孔加2mL,设置不加FGF-2和Nb4的基础培养基为阴性对照,并以14.7ng/mL FGF-2组为阳性对照。(6)分别在加样后0h、24h用显微镜拍照,记录同一划痕处细胞的生长状况。(7)用Image J计算划痕面积,计算细胞的迁移率。细胞迁移率(%)=[给药前划痕面积-给药后划痕面积]/给药前划痕面积×100%。
结果如图7所示,阳性对照组和FGF-2+1nM Nb4组的迁移率显著高于阴性对照,FGF-2+4nM Nb4、FGF-2+16nM Nb4以及FGF-2+64nM Nb4组相比阴性对照有极显著差异,说明FGF-2可以有效促BALB/C-3T3细胞的增殖,Nb4的存在不会抑制FGF-2的活性,甚至在添加量加大时有利于FGF-2发挥其活性功能。FGF-2+1nM Nb4组与阳性对照组的迁移率无明显差异,从FGF-2+4nM Nb4、FGF-2+16nM Nb4组开始,迁移率达到最高,说明随着Nb4剂量的增加,增强了FGF-2的稳定性,提高了FGF-2的活性,Nb4在浓度4nM~64nM时,细胞的增殖迁移能力尤为显著。
上述实施例为本发明较佳的实施方式,但本发明的实施方式并不受上述实施例的限制,其他的任何未背离本发明的精神实质与原理下所作的改变、修饰、替代、组合、简化,均应为等效的置换方式,都包含在本发明的保护范围之内。

3.一种含有权利要求1所述的FGF-2纳米抗体的蛋白质和/或多肽组合物,其特征在于:
所述的FGF-2纳米抗体为所述的蛋白质和/或多肽的保护剂。
4.根据权利要求3所述的含有权利要求1所述的FGF-2纳米抗体的蛋白质和/或多肽组合物,其特征在于:
所述的蛋白质为成纤维细胞生长因子-2。
5.根据权利要求4所述的含有权利要求1所述的FGF-2纳米抗体的蛋白质和/或多肽组合物,其特征在于:
所述的FGF-2纳米抗体作为FGF-2的保护剂和/或增效剂。
6.根据权利要求3所述的含有权利要求1所述的FGF-2纳米抗体的蛋白质和/或多肽组合物,其特征在于:
所述的蛋白质和/或多肽组合物还可以包括其他蛋白保护剂;
所述的蛋白质和/或多肽组合物为液态的蛋白质和/或多肽组合物,或者是液体冻干后储藏,但使用前需要复溶为液体的蛋白质和/或多肽组合物。
7.根据权利要求6所述的含有权利要求1所述的FGF-2纳米抗体的蛋白质和/或多肽组合物,其特征在于:
所述的其他蛋白保护剂为蔗糖、甘油、右旋糖酐40、肝素钠、海藻糖、甘露醇中的至少一种;
所述的其他蛋白保护剂的终浓度为0.03mol/L以上。
8.一种提高蛋白质和/或多肽稳定性的方法,其特征在于:
在所述的蛋白质和/或多肽中添加权利要求1所述的FGF-2纳米抗体。
9.根据权利要求8所述的提高蛋白质和/或多肽稳定性的方法,其特征在于:
所述的FGF-2纳米抗体的终浓度为1nM~70nM;
所述的蛋白质和/或多肽为液态下的蛋白质和/或多肽,或者是液体冻干后储藏,但使用前需要复溶为液体的蛋白质和/或多肽;
所述的蛋白质和/或多肽为处于pH为5~11、温度为4℃~60℃或相当于不超过转速1000r/min的震荡外力作用至少一种情况下的蛋白质和/或多肽。
10.根据权利要求8所述的提高蛋白质和/或多肽稳定性的方法,其特征在于:
所述的FGF-2纳米抗体的终浓度为4nM~64nM;
所述的蛋白质和/或多肽为处于温度为4℃~40℃的蛋白质和/或多肽。
