发明背景
白细胞介素-6 (IL-6)为由广泛范围的抗原呈递细胞(包括B细胞和T细胞)产生的炎性反应的主要介质之一。IL-6由活化的单核细胞或巨噬细胞、内皮细胞、成纤维细胞、活化的T细胞以及一系列不是免疫细胞的细胞产生。与许多其他细胞因子一起,其参与与免疫反应、血管生成、炎症、骨代谢相关的过程。IL-6的主要作用与其作为辅因子参与B淋巴细胞的分化、其成熟和转化成分泌免疫球蛋白的浆细胞相关。除此之外,IL-6促进活化的免疫细胞上的IL-6受体表达,并诱导Т细胞产生IL-2。该细胞因子刺激T淋巴细胞的增殖和造血反应。就产物的细胞来源多样性和生物效应的目标而言,白细胞介素-6为参与实现免疫和炎性反应的最活跃细胞因子之一。已表明,IL-6的促炎和抗炎作用之间的失衡导致各种自身免疫性疾病、慢性炎症和骨质疏松症、牛皮癣,而其过度产生导致不同形式的癌症。

受体IL-6 (IL-6R, IL6R, CD126)为一种分子复合物,活化时启动细胞反应级联反应,导致参与进一步炎性应答反应的蛋白质的主动合成。该受体在IL-6与具有80 kDa质量的受体α单元(其防止信号转导)和两个具有130 kDa质量的gp130分子(其在细胞内转导信号)结合期间被激活(SimonA. 关于抗体多肽序列的术语“同源的”应解释为相对于多肽序列呈现出至少70%,优选地80%,更优选地90%和最优选地95%序列同一性的抗体。与核酸序列相关的术语应解释为相对于核酸序列呈现出至少85%,优选地90%,更优选地95%和最优选地97%序列同一性的核苷酸序列。
术语“双特异性抗体”或“多特异性抗体”包括能够选择性结合两个或更多个表位的抗体。双特异性抗体,例如可包含两个不同抗原结合部分,其中所述抗原结合部分特异性结合不同分子(例如抗原)或相同分子(例如相同抗原)上的不同表位。如果双特异性抗体能够选择性结合两个不同表位(第一表位和第二表位),则第一抗原结合部分对第一表位的亲和力将一般地比第一抗原结合部分对第二表位的亲和力低至少1-2,或3或4个数量级,反之亦然。由双特异性抗体识别的表位可为相同或不同的靶标(例如在相同或不同蛋白质上)。例如,双特异性抗体可通过组合识别相同抗原上的不同表位的重链来制备。例如,编码识别不同表位的可变重链序列的核酸序列可与编码各种重链恒定区的核酸序列融合,并且这种序列可在表达免疫球蛋白轻链的细胞中表达。典型的双特异性抗体包含两条重链(每条包含3个重链CDR,随后是(从N-末端到C-末端)CH1结构域、铰链区、CH2结构域和CH3结构域)及免疫球蛋白轻链,其不具有抗原结合特异性但能够与每条重链组合,或者能够与每条重链组合并结合受抗原结合重链区限制的一个或多个表位,或者能够与每条重链组合并促进1或2条重链与1或2个表位的结合。
2. 目的
本发明的目的为产生具有与人白细胞介素-6受体特异性结合的能力的备选抗体或其片段,其将用作治疗或诊断由白细胞介素-6介导的疾病或减轻由白细胞介素-6介导的症状的医药产品。
3. 本发明的抗体及其片段
本发明提供一种抗体或其抗原结合片段,其中由于其包括与SEQ ID NO: 3的序列至少75%同源的氨基酸序列而确保与人白细胞介素-6 (IL-6)受体结合的能力。
在一个实施方案中,本发明涉及包含SEQ ID NO: 3的氨基酸序列的抗体或其片段。

有效量至少表示最小剂量,但小于确保患者的治疗或预防效果必要的活性剂的毒性剂量。另一方面,本发明抗体的有效量表示在哺乳动物(优选地为人)中降低IL-6生物活性的量。
本发明抗体的给予途径可为口服、胃肠外、吸入或局部。优选地,本发明的抗体可包含在胃肠外给予可接受的药用组合物中。术语“胃肠外”包括静脉内、肌内、皮下、直肠、阴道或腹膜内给予。静脉内、腹膜内或皮下注射为优选的给予途径。这种注射可接受的药用载体为现有技术熟知的。

这些建议的抗体量很大程度上取决于治疗专家的决定。预期效果为选择适当剂量和方案的关键因素。本文考虑的因素包括待治疗的某种疾病、待接受治疗的某种哺乳动物、某位患者的临床状况、障碍原因、抗体给予部位、具体抗体类型、给予途径、给予方案和医学领域熟知的其他因素。
可将抗体或其片段冷冻或冻干,并在应用之前在合适的无菌载体中重构。冻干和重构可导致抗体活性的一些丧失。剂量可能需要调整以补偿这种丧失。通常,对于药用组合物,pH优选在6-8之间。
9. 制品
在本发明的另一个实施方案中提供包含可用于治疗或预防上述障碍或病症的物质的制品。
该制品包括具有含有抗体的药用组合物以及标签的容器,并且可能还有药品说明书。合适的容器包括例如小瓶、安瓿、注射器和分析管。容器可由多种材料制成,比如玻璃或聚合物材料。容器包含有效治疗IL-6介导的疾病或障碍的本发明组合物,并可具有无菌入口(例如容器可为静脉内溶液袋或具有皮下注射针可刺穿的瓶塞的小瓶)。组合物中的活性剂为本发明的抗IL-6R抗体。位于容器上的标签或附于其上的药品说明书表明组合物用于治疗期望的疾病。制品可进一步包括具有药学上可接受的缓冲液(比如磷酸盐缓冲盐水、林格氏溶液和右旋糖溶液)的第二容器。其可进一步包括从商业和消费者角度期望的其他材料,包括其他缓冲剂、稀释剂、过滤器、针头、注射器和药品说明书。

本发明的实施方案
用于获得和测试本发明抗体的方法的一般描述
在第一阶段,选择与可溶形式的人白细胞介素-6受体(sIL-6R)即其α-亚基特异性结合的多肽。为此目的,使用基于人免疫球蛋白的重链和轻链氨基酸序列设计的噬菌体文库。为了建立噬菌体文库,从超过1000个供体的血液分离和纯化RNA分子,其中供体免疫球蛋白的重链和轻链(分子)基因通过在几个阶段中逆转录和PCR来合成。来自重链和轻链结合区的基因的各种组合以适当的取向插入噬菌体载体中,并用于产生杂交噬菌体文库(图4)。
对人sIL-6R蛋白选择获得的噬菌体文库,所述人sIL-6R蛋白在瞬时СНО-Т细胞系统中培养,并在IMAC BIORAD吸附剂上从细胞培养液纯化。
产生sIL-6R的细胞通过将人白细胞介素-6受体的α亚基基因再克隆至用于瞬时培养pEE的载体中,随后进行СНО-Т转染而获得。
将生产者细胞在温育箱中培养7-10天。
蛋白质以单一步骤纯化,靶蛋白通过蛋白质印迹使用特异性抗体(来自人IL-6sRDuoSet ELISA Development Kit目录号:DY227 R&D System的人IL-6sR检测抗体PartNo.840244)验证。
获得的噬菌体文库以两个或更多个阶段进行选择。选择与固定于塑料上的sIL-6R特异性结合的噬菌体。结果,获得了大量与sIL-6R结合的噬菌体,从中分离出噬菌体载体。从这些载体扩增与蛋白质特异性结合的多肽基因并在用于在大肠杆菌原核细胞中合成蛋白质分子的表达载体中再克隆。在转化用质粒获得的细胞和合成单个多肽之后,我们筛选并选择与IL-6sR特异性结合并阻断配体与受体结合的分子。将所获得蛋白质的亲和常数相互比较,并插入pEE表达载体的最成功基因中,用于表达已经全尺寸的抗体。在中国仓鼠卵巢(CHO)的悬浮细胞培养物中培养抗体并将其纯化之后,评估其亲和力并且各种细胞功能试验确保白细胞介素-6信号被阻断。作为所实施研究和分析的结果,选择了一种在细胞试验中与sIL-6R特异性结合并阻断IL-6活性的抗体分子。其重链和轻链的核苷酸序列在psX载体中再克隆,借此我们获得了稳定产生抗体的细胞系。然后,用获得的细胞系培养选择的抗体,对其选择了纯化方案和储存条件。我们设计并开始了所获得医药产品的临床前研究。
实施例1. 在哺乳动物的悬浮细胞培养物中产生重组抗原和抗体
根据方案,在从中国仓鼠卵巢细胞获得的瞬时细胞系(品系CHO-K1)的细胞中产生抗体和抗原。根据制造商的使用说明书,使用Life Technologies Corporation的用于悬浮培养的无血清培养基,在轨道振荡器上的烧瓶中实施悬浮培养。对于瞬时表达,用线性聚乙烯亚胺(“Polysciences”的PEI “MAX”)转染细胞。DNA/PEI比率为1:3-1:10。转染之后7-10天,将培养基以2000 g离心20分钟,并通过0.22 μm孔径的过滤器过滤。通过亲和HPLC从培养液分离靶蛋白。
实施例2. 从哺乳动物的悬浮细胞培养物纯化抗原和抗体
在蛋白质C-末端包含6个His氨基酸的sil-6R重组蛋白使用Profinity IMAC Ni-荷电树脂(Bio-Rad)从培养基分离和纯化。纯化之前,在培养基中加入NiCl2直至其浓度为1mM。然后在培养基中加入1/200-1/500的Profinity IMAC Ni-荷电吸附剂体积,并在室温下于振荡器中混合1小时。然后将吸附剂转移至体积为5或10 ml的ThermoscientificPolypropylene柱上,用5倍柱体积的含有5 mM咪唑和0.3 M氯化钠的10 mM рН8磷酸盐缓冲液冲洗以洗涤非特异性结合的组分。使用0.3 M咪唑, рН 8 0.3 М NaCl洗脱结合的抗原。然后通过使用Snake Skin Dialysis Tubing技术进行透析将蛋白质转移至PBS (pH7.4)中,过滤(0.22 μm)并转移至管中。将管储存于-70℃下。用SDS凝胶电泳评价所获得溶液的纯度,并使用特异性抗IL-6R抗体和缀合的山羊抗人抗体抗体,通过蛋白质印迹技术鉴定(参见图1和2)。
使用1 ml HiTrap rProteinA FF柱(GE Healthcare)纯化所研究的抗IL-6R抗体。使澄清的培养液通过1 ml HiTrap rProtein A FF (GE Healthcare),其已用磷酸盐缓冲盐水(PBS, pH 7.4)平衡。然后用5倍柱体积的PBS洗涤柱以去除非特异性结合的组分。使用0.1 M甘氨酸缓冲液(рН 3)洗脱结合的蛋白质。收集主要的洗脱蛋白质峰,并用1 M的Tris缓冲液(pH8)中和其pH。所有阶段均以1 ml/分钟的流速实施。然后通过使用Snake SkinDialysis Tubing技术进行透析将蛋白质转移至PBS (pH7.4)中,过滤(0.22 μm),转移至管中,并储存于-70℃下。用SDS凝胶电泳评价所获得蛋白质的纯度(参见图7和8)。
重组IL6-H6-EPEA配体(在C-末端具有6个组氨酸的“肽尾”和EPEA-标签的白细胞介素-6)在GE Healthcare C16/20柱上用5 ml Capture Select C标签亲和基质进行纯化。所有阶段均以5 ml/分钟的流速实施。澄清的培养基通过柱进行处理,所述柱已用磷酸盐缓冲盐水(PBS, pH 7.4)平衡。然后用PBS洗涤柱,直至色谱系统分光光度计信号达到稳定水平以去除非特异性结合的组分。使用20 mM Tris 2M MgCl2 (pH 7.1)洗脱结合的蛋白。收集主要洗脱的蛋白质峰,并通过使用Snake Skin Dialysis Tubing技术进行透析转移至PBS(pH7.4)中。然后将其过滤(0.22 μm)并转移至管中,且储存于-70℃下。用SDS凝胶电泳评价所获得溶液的纯度(参见图3)。
实施例3. 人噬菌体抗体的Fab文库的选择
将具有与sIL-6R特异性结合的Fab片段的噬菌体从组合噬菌体Fab展示文库MeganLibTM (BIOCAD)分离,所述文库包含超过1011个单个克隆并根据修改的方案(JBiolChem. 1999 Jun 25; 274(26): 18218-30)基于人免疫球蛋白基因的可变结构域合成(图4)。为了产生该文库,使用超过一千个供体的淋巴细胞。在上述条件下基于重组人sIL-6R-H6F实施选择(NatBiotechnol. 1996 Mar;14(3):309-14; J Mol Biol.1991 Dec 5;222(3): 581-97)。为了通过淘选实施选择,将人的sIL-6R-H6或sIL-6R-Fc在Immune管EIA/RIA高结合(Greineer bio-one)的表面上于4℃下在50 mM碳酸盐缓冲液(pH 9.5)中吸附过夜。管用PBST (PBS pH 7.4和0.1% Tween 20 v/V)洗涤几次,并然后用脱脂乳溶液(PBSTpH 7.4中的0.5% m/V)封闭管表面的空白蛋白结合位点。将脱脂乳溶液在管中温育1小时。然后用PBST溶液洗涤管。然后,将MeganLibTM噬菌体文库在2-4 ml含有0.5%脱脂乳的PBST溶液中稀释直至噬菌体的最终浓度为1013/ml。将制备的文库加入到其表面结合有抗原的管中,并在搅拌下温育1小时。通过用PBST将管进行连续洗涤去除未结合的噬菌体。洗涤量从第一轮至第三轮分别增加:20-30-40次。保持结合的噬菌体颗粒用0.1 M Gly-HCl溶液(pH2.2)从塑料管中洗脱15分钟,同时搅拌。然后将溶液转移至洁净塑料管中并用1M Tris-HCl(pH 8) 5:1中和。用获得的噬菌体感染大肠杆菌TG1菌株细菌,扩增噬菌体并用于下一个选择循环。
实施例4. 噬菌体扩增
选择之后,使用大肠杆菌TG1菌株培养噬菌体。通过用噬菌体培养物感染宿主菌株随后生长12-15小时来实施扩增。选择之后,将噬菌体溶液与大肠杆菌TG1生长的细胞培养物(OD600=0.3-0.4)混合,并在37℃下温育1.5小时。然后将细胞以3000-4000 rpm离心10-15分钟,重悬于1 ml 2TY培养基中,并在含有用于选择的抗生素(氨苄青霉素)的培养皿中弄碎。使菌落在30℃恒温器中生长过夜。12-15小时之后,计数菌落数并用5-10 ml 2TY培养基从培养皿洗去。为此目的,将100 mcl细胞悬液加入到20 ml含有抗生素(氨苄青霉素)的2TY培养基中,并于37℃下在振荡器上生长直至OD600为0.35-0.5。将К07辅助噬菌体加入到细胞悬液中至最终浓度为1010个粒子/ml (1 mcl К07辅助噬菌体/10 ml细胞悬液),并在37℃下温育1.5小时,同时轻微搅拌。然后,向细胞培养物中加入相同体积的含有单剂量氨苄青霉素(100 µg/ml)和双剂量卡那霉素(60 µg/ml)和双剂量IPTG (0.2 mM)的培养基。将烧瓶装载至振荡器中,并将噬菌体在30℃下培养3-5小时。将细胞培养物以10000 rpm离心20-30分钟,并将上清液收集在管中。此后,将包含20%聚乙二醇和2.5 M氯化钠的1/6溶液体积加入到上清液中并剧烈搅拌。使溶液在冰中温育至少3小时。然后将溶液以8000 g离心10分钟,在1 ml TBS缓冲液中稀释形成的包含噬菌体的沉积物。
实施例5. 在表达质粒中再克隆具有各种可变区的Fab片段的基因
第三轮选择之后,使得到的M13噬菌体的噬菌体DNA (噬菌粒)与大肠杆菌TG1细胞分离。扩增具有各种可变区的Fab片段的基因,并使用PCR和末端特异性引物再克隆至pll4质粒中。基因通过位点NheI、NotI再克隆。通过4个随机菌落的测序检查插入物。
实施例6. 在具有蛋白质表达的大肠杆菌BL21Gold细胞中生长Fab片段至生长细胞培养基中
使基于所获得基因的Fab片段在大肠杆菌BL21Gold细胞中合成。将第2和第3轮选择之后具有从噬菌粒再克隆到其中的Fab片段基因的pLL4表达载体转化(根据标准方案电穿孔)至大肠杆菌BL21Gold表达菌株中。细胞在2TY培养基中生长至含有选择抗生素(卡那霉素30 μg/ml、氨苄青霉素100 μg/ml和葡萄糖0.2%)的琼脂化培养基(培养皿)上。使细胞以使细胞滴度适合于选择单个克隆的方式生长,菌落在30℃下于恒温器中生长14-16小时。将某些菌落重新注入到96孔板(U形),每孔含100 mcl 2TY培养基(卡那霉素30 μg/ml、氨苄青霉素100 μg/ml和葡萄糖0.4%),并在室温下于振荡器上以800 rpm生长16-18小时。使细胞培养物通过96通道复制器筛出到无菌96孔板(V形)上,其中含有100 mcl 2TY培养基(卡那霉素30 μg/ml、氨苄青霉素100 μg/ml和葡萄糖0.1%、TX100 0.01%和IPTG 0.5 mM),并在室温下于振荡器上以800 rpm生长过夜。将板以3500 rpm离心20分钟,并将用于ELISA的具有Fab片段的培养基收集在洁净96孔板(U形)中,将板储存于-70℃下。
实施例7. 与人IL-6sR-H6特异性结合的Fab的筛选
ELISA用于选择与人IL-6sR-H6特异性结合的Fab片段。复制的Fab Actemra(托珠单抗) [US20110245473 seq.id 15和US20110245473 seq.id 16]用作阳性对照。为了筛选特异性结合,使用96孔ELISA板(NuncImmunoMaxisorp)。在孔表面实施IL-6sR-H6抗原结合,为此,向每孔加入50 μl结合碳酸盐缓冲液中的IL-6sR-H6溶液(0.5 μg/ml)。将板覆盖并在4℃下温育过夜。使用基于机器人系统Genetix Qpix2xt (Molecular Device)和TecanFreedom EVO 200 (Tecan)的高通量自动化平台,按照标准ELISA方案实施所有后续步骤。第二天,板孔用PBST洗涤5次。将每孔200 μl封闭缓冲液(PBST中的0.5%脱脂乳)加入板中以防止非特异性结合。将板在室温下于振荡器上温育1小时。用PBST洗涤之后,向每孔加入50μl含有受试Fab的受试细胞上清液和封闭缓冲液的1:1混合物。将含有Fab溶液的板在室温下于振荡器上温育1小时。1小时之后,板孔用PBST洗涤5次。然后将与过氧化物酶缀合的山羊二抗抗人Fab (Pierce ThermoScientific) (1:5000)在PBST中的溶液加入板孔中(50mcl/孔)。将板在旋转振荡器中振荡(室温下50分钟),并然后如上所述用PBST洗涤5次。通过加入在乙酸盐缓冲液рН 5.5和Н2О2 0.02%中的ТМВ底物溶液(50 mcl/孔)获得比色信号,温育直至获得饱和信号(平均3-5分钟),通过添加10%硫酸溶液(30 μl/孔)终止反应。用Tecan-Sunrise读板器(Tecan)在450 nm处测量信号。与抗原结合的Fab数目与显示的信号成比例。因此,选择其信号超过背景信号超过5倍的克隆。然后以竞争性ELISA检查所选择的Fab以检测阻断IL6与其受体之间相互作用的拮抗性Fab。将具有最大信号的大肠杆菌BL21Gold细胞悬液转移至洁净板中,并在15%甘油中在-70℃下孵育。
实施例8. 用Fab片段阻断IL6配体与IL-6R受体的相互作用的竞争性ELISA
竞争性ELISA用于检查先前选择的Fab片段与人IL-6R特异性结合的拮抗能力。作为拮抗剂的阳性对照,我们使用抗IL-6R Fab片段。通过向孔中加入50 mcl在碳酸盐缓冲液中的蛋白质溶液(1 µg/ml)使配体IL-6-EPEA固定于ELISA板(Maxisorp)上。将溶液在4℃下于板中温育过夜。所有后续阶段均按照标准ELISA技术,并进行一些变化,使用基于GenetixQ-pix2xt机器人系统(MolecularDevice)和TecanFreedom EVO 200 (Tecan)的高性能自动化平台实施。为了阻断非特异性结合,加入封闭缓冲液(200 mcl/孔,PBST中的0.5%脱脂乳)。将板在室温下于振荡器上温育1小时。
与此平行地,将包含所研究的抗IL-6R的Fab片段的每个50 mcl受试细胞上清液与50 mcl的IL-6sR-H6F受体溶液(0.4 μg/ ml,在PBST中的1%脱脂乳中)在非结合96孔板中混合。将其在37℃下于振荡器上以500 rpm温育1小时。
在从含固定化配体IL6的板上洗去封闭缓冲液之后,将抗IL-6R的Fab片段和IL-6sR受体的上述反应混合物转移至这些板中。将板再次在室温下于振荡器上温育45分钟,并然后板用PBST缓冲液洗涤5次。加入PBST中的抗FLAG小鼠抗体(FLAG肽为IL-6sR-H6F蛋白上的C-末端肽) (50 μl/孔)并在相同条件下温育45分钟。板用PBST缓冲液洗涤5次,然后加入PBST中的Pierce的山羊抗小鼠过氧化物酶缀合的抗体产物(稀释度为1:5000)。如前所述,使板在室温下于振荡器上温育45分钟,并然后用PBST缓冲液洗涤5次。用ТМВ溶液(50mcl/孔)获得比色信号直至饱和(平均3-5分钟),通过加入10%硫酸溶液(30 mcl /孔)停止反应。使用Tecan-Sunrise读板器(Tecan)在450 nm波长下测量彩色信号。二级缀合抗体结合的程度与彩色信号成比例。
选择在受控的抗IL-6R的Fab片段水平下表明信号阻断的克隆并在进一步分析期间使用。根据标准方案在Applied Biosystems 3130 Genetic Analyzer (AppliedBiosystems)上对该阶段之后所选克隆的可变结构域的基因进行测序和分析。
实施例9. 抗IL-6R Fab的比较性koff (kdis)筛选
将与IL-6R受体特异性结合并阻断受体与其配体相互作用的Fab片段通过对受体的亲和力进行相互比较。使用Octet Red 96和抗FABCH1生物传感器(Pall-ForteBio)实施抗IL-6R候选物的比较性koff (kdis)筛选(图6)。使生物传感器在包含10 mM PBS pH 7.2-7.4、0.1% Tween-20和0.1% BSA的运行缓冲液中再水合。在所研究的包含抗IL-6R Fab片段的大肠杆菌细胞生长培养基样品中加入1/10体积的10X运行缓冲液并混合。然后将抗FABCH1生物传感器在4℃下浸入包含Fab片段的溶液中12小时。12小时之后,将具有表面固定的Fab片段的传感器转移至具有运行缓冲液的孔中,此时设定基线(60秒)。然后将传感器转移至具有分析物溶液(IL-6sR-H6F, 30 µg/ml)的孔中以实现抗原/Fab-片段复合物缔合(300秒)。之后,使传感器返回至具有工作缓冲液的孔中进行进一步解离(300秒)。使所用的传感器在每次测试之后经受再生:将其3次置于再生缓冲液(Gly-HCl, рH 1.7)中,并然后用于进一步的实验。使用1:1相互作用模型,根据标准程序,使用OctetDataAnalysis (版本7.0)分析获得的曲线。
抗IL-6R候选物的koff筛选结果如表1所示。显示了所有Fab片段与人IL-6R的特异性结合,再克隆了具有最小值的候选物以获得全长抗体。

实施例10. 通过从Fab片段再克隆可变结构域构建全长IgG1抗体
通过从先前用于培养Fab片段的表达质粒再克隆可变结构域基因来构建全长IgG1抗体。Clontech的In-Fusion® HD EcoDry™ CloningKit用于再克隆。用特异性引物通过PCR获得可变结构域基因插入物,通过用DpnI限制酶处理去除PRC中的基质质粒。pEE质粒载体通过轻链的SalI和BsiWI限制酶和重链的SalI/NheI限制酶的限定而线性化。将基因插入物和线性化载体在10 mcl水中混合,并转移至In-Fusion系统的单独条带状小瓶中。通过吸液搅拌。使带状小瓶在37℃下温育15分钟,然后在50℃下温育15分钟,然后将其转移至冰中。将具有所获得构建体的部分反应体积用于细胞转化。从获得的克隆提取质粒,并通过序列检查插入物。
实施例11. 用抗IL-6R抗体对DS-1细胞增殖的细胞抑制试验
DS-1细胞生长取决于细胞外的外部IL6配体的存在,并且阻断其与细胞表面上的受体的结合导致其细胞生长抑制。使DS-1细胞培养物在补充有10%灭活胎牛血清、1 mM丙酮酸钠和7.5 ng/ml IL6的RPMI-1640培养基上生长。实验前一天,将细胞以PBS从IL6残留物洗涤两次,并在不添加IL6的生长培养基中以1000个细胞/孔的比率接种至96孔板中。第二天,在加入5 ng/ml IL6的完全生长培养基中制备一系列抗体稀释溶液,从20 μg/ml开始,间隔为3。然后,向细胞加入相同体积的所制备抗体溶液,孔中IL6的最终浓度为2.5 ng/ml,第一点的抗体为10 μg/ ml。将细胞在37℃和5% СО2下温育3天。然后,在FluoroskanAscent FL 2.5上用AlamarBlue活体染料测量活的增殖细胞的数量(图9)。根据研究结果,选择一种最大限度抑制DS-1细胞增殖的抗IL-6R抗体,因此,其与白细胞介素-6的膜受体结合并阻断配体与受体的结合。
实施例12. HEK-BlueTM IL6细胞培养物中的STAT3抑制作用分析
在补充有10%灭活胎牛血清的DMEM培养基(细胞生长培养基)中制备浓度为1*106克隆/ml的HEK-Blue IL6细胞悬液。将细胞以100 mcl/孔的比率接种至96孔板中(5*104克隆/孔)。
在细胞生长培养基中从200 μg/ml制备一系列抗体稀释溶液,间隔3个10点,最后一个点为对照,无抗体。以浓度为4 ng/ml在细胞生长培养基中制备人IL6溶液。然后,向细胞加入50 mcl的稀释抗体,并在CO2温育箱中温育45分钟。向具有抗体的细胞加入50 mcl的IL6溶液,并使细胞与抗体和IL6一起在CO2温育箱中温育过夜。
第二天制备QUANTI-Blue TM检测培养基:使1包干燥培养基溶于50 ml纯净水中,在37℃水浴上加热10分钟,并通过0.22 μm过滤器过滤。
向具有180 mcl检测培养基的每孔加入20 mcl细胞培养液,并置于CO2温育箱中2-3小时。在分光光度计上以630 nm的波长测量吸光度水平(图10)。
实施例13. BCD89抗体与人白细胞介素-6受体及其直系同源物的结合亲和常数的测定
用ELISA检查所选抗-IL6R与食蟹猴、豚鼠、狗和小鼠的白细胞介素-6受体的相互作用(图11,12)。用Octet Red 96 (ForteBio)获得BCD89抗体对人、食蟹猴、豚鼠、狗和小鼠的IL-6sR α亚基的结合亲和常数。根据制造商关于制备和固定AR2G传感器的说明书,使用标准方案将BCD89非特异性地固定于第二代氨基反应性传感器(AR2G)的表面上。使用包含0.1% Tween-20和0.1% BSA的PBS作为工作缓冲液在30℃下实施分析。使用浓度为126 nM-2nM,间隔为2的运行缓冲液实施人、食蟹猴、豚鼠、狗、兔和小鼠的IL-6R的滴定。
在减去参考信号之后,使用Octet数据分析软件(版本7.0)按照标准程序并使用1:1相互作用模型分析结合曲线。结果如表2所示。
可以看出,BCD89以高亲和力与人的重组IL-6R和食蟹猴的IL-6sR结合(参见图13、图14)。此外,候选物与豚鼠IL-6R相互作用,其常数比与人低3个数量级(参见图16)。未显示与小鼠受体的相互作用(参见图15)。

实施例14. 热应力条件下BCD89聚集稳定性的测定
通过以10 kDa/0.5 ml的离心过滤器AmiconUltra (Millipore)超滤,将所研究的样品浓缩至5 mg/ml。通过UV分光光度法以280 nm波长测定蛋白质含量。将每个获得的样品按150 mcl分成几个部分并转移至单独的管中:将每种化合物的一只管放入冰箱中储存于+4℃下,其他管放入管恒温器中并在50℃下恒温持续设定的时间。
一旦加热,将管从恒温器中取出,冷却至室温,通过以13000 g离心溶液10分钟澄清,上清液用UV检测器递送进行凝胶过滤。在加热前后分析色谱图上的全尺寸蛋白质峰,在Agilent USA 1100系列M色谱仪上实施色谱分析。使用柱Tosoh TSK-Gel G3000SWXL 7.8mm ID X 30 cm和前柱Tosoh TSK-Gel GuardSWXL 6 mm ID X 4 cm, 7 mcm。流速为0.7ml/分钟,样品体积为10 mcl,样品浓度为5 mg/ml。检测器波长为220和280 nm,洗脱时间为25分钟(图17)。
通过内部归一化方法实施计算。通过下式计算单体(Х)的百分比含量:

在计算期间,流动相色谱图和规定的缓冲溶液中存在的峰被忽略。
实施例15. BCD-089产物使用胶原蛋白诱导的关节炎灵长类动物模型的抗炎活性评价
使用胶原蛋白诱导的关节炎灵长类动物模型在雄性食蟹猴(Macaca fascicularis)实施研究。参与实验的动物总数共计20头,每组包括4只猴。实验期间使用4种剂量水平的产物:1.0 mg/kg、4.0 mg/kg、10.0 mg/kg、20.0 mg/kg,对照组动物给予安慰剂。在用胶原蛋白初步致敏之后开始给予产物和安慰剂。实验期间,测量所有组的动物以计算关节表面和炎症面积百分比(PIA),在研究结束时获得掌指和跖趾关节来评价破坏性变化的严重程度。将动物分成5组。组的名称如表3所示。

对于关节炎诱导,将牛II型胶原蛋白(Sigma)的乳液3次给予动物。
第一次给予胶原蛋白。每只实验动物给予的胶原蛋白总量为2 mg。为此,使2 mg胶原蛋白溶于0.7 ml 0.1 M乙酸中。向该溶液加入0.7 ml弗氏不完全佐剂。
在第一次给予胶原蛋白之后将动物保持28天。
第二次给予胶原蛋白。每只实验动物给予的胶原蛋白总量为3 mg。为此,使3 mg胶原蛋白溶于1.0 ml 0.1 M乙酸中。向该溶液加入1.0 ml弗氏完全佐剂。
在第二次给予胶原蛋白之后将动物保持21天。
第三次给予胶原蛋白。每只实验动物给予的胶原蛋白总量为3 mg。为此,使3 mg胶原蛋白溶于1.0 ml 0.1 M乙酸中。向该溶液加入1.0 ml弗氏不完全佐剂。
在以下时间点用卡钳实施关节大小的测量:
-第一次给予胶原蛋白之前
-第二次给予胶原蛋白时
-第二次给予之后紧接给予胶原蛋白之前每周,持续7周。
在测量过程期间估计了关节纵横轴的量,除拇指以外,对所有掌指和跖趾关节实施该程序。面积计算通过下式实施:
JA = 纵轴值×横轴值×3.14×0.25。
每只动物的16个关节的数据用于计算炎症面积百分比(PIA)的值。计算通过下式实施:

实施例17. 在食蟹猴中多次皮下给予1个月接着2周无给予期后的毒性研究

实验期间评价以下参数:
-临床检查结果;
-动物体重(给予之前和另外每周);
-体温(给予之前和然后每周直至实验终止);
-基于通过Poly-Spectrum心电图评估的心脏生物电活动的心血管系统效应,评估在给予之前和然后在实验的第3、5、7周实施;
-尿液分析(给予之前和实验的第3、5、7周);
-以下参数的全血分析:红细胞数、白细胞数、血红蛋白浓度、淋巴细胞数、单核细胞数、嗜中性粒细胞数、嗜酸性粒细胞数、嗜碱性粒细胞数、血小板数(给予之前和然后从实验的第1周开始一周一次);
-以下参数的凝血系统效应评估:活化部分凝血活酶时间、纤维蛋白原浓度、凝血酶原时间-在产物给予之前,然后在实验的第3、5、7周实施;
-以下参数的血清生化分析:钠、钾、肌酸酐、尿素、碱性磷酸酶、乳酸脱氢酶、总胆红素、总蛋白质、葡萄糖、甘油三酯、天冬氨酸氨基转移酶、丙氨酸氨基转移酶、总胆固醇(给予之前和实验的第3、5、7周);
-在给予期结束时,对最大剂量的卫星组动物实施安乐死,随后对其进行病理形态学检查;在研究结束时对最大剂量组和对照组的动物同样进行;
-作为毒性研究的一部分,还评估了产物的局部刺激作用,并因此选择位于注射区域附近的软组织并进行组织学检查。
实施例18. 在食蟹猴中在4周内多次皮下给予BCD-089产物后的免疫原性研究
在相关动物-食蟹猴上在多次皮下给予1个月接着2周恢复期的情况下实施免疫原性的检查。在实验中使用3个剂量水平。实验组的方案如下表所示。

基于结合抗体水平实施免疫原性的评价,为此,在产物给予之前和实验的第3、5、7周,收集血液样品,随后进行血清分离。
实施例19. 1个月内在食蟹猴中多次皮下给予BCD-089产物后的药代动力学研究
在相关动物物种-食蟹猴中实施多次皮下给予1个月接着2周恢复期后的药代动力学研究。在实验中使用3个剂量水平。实验组的方案如表7所示。

为了评价灵长类动物血清中的产物水平变化,在实验开始之前和实验的第1、2、8、9、15、16、22、23、29、36和43天收集血液样品。
实施例20. 多次皮下给予1个月接着2周恢复期后的BCD-089产物免疫毒性研究
在相关动物物种-食蟹猴中实施多次皮下给予1个月接着2周恢复期后的免疫毒性研究。在实验中使用3个剂量水平。实验组的方案如表8所示。

实验期间评价以下参数:
-淋巴细胞亚群组成,其在制剂给予之前和然后在实验的第2、4、6周进行评估;
-在给予之前和实验的第2、4、6周评估免疫球蛋白类别的比率;
-在给予之前和实验的第2、4、6周评估对吞噬作用的影响。
实施例21. 获得药用组合物
将BCD089抗体转移至适当的缓冲液中直至浓度达到180 mg/ml,过滤所得溶液(灭菌过滤)并吸入注射器中。
Claims (25)
1.一种具有与人白细胞介素-6(IL-6)受体结合的能力并包含结合结构域的抗体或其抗原结合片段,所述抗体或其抗原结合片段包含重链和轻链,所述重链包含氨基酸序列如SEQ ID NO:1所示的CDR1、氨基酸序列如SEQ ID NO:2所示的CDR2、氨基酸序列如SEQ IDNO:3所示的CDR3;所述轻链包含氨基酸序列如SEQ ID NO:4所示的CDR1、氨基酸序列如SEQID NO:5所示的CDR2、氨基酸序列如SEQ ID NO:6所示的CDR3。
2.权利要求1的抗体或其片段,其特征在于结合结构域包含含有SEQ ID NO:7的氨基酸序列的重链可变结构域。
3.权利要求1的抗体或其片段,其特征在于其包含具有SEQ ID NO:9的序列的重链。
4.权利要求1的抗体或其片段,其特征在于其包含具有SEQ ID NO:10的序列的轻链。
5.权利要求1的抗体或其片段,其特征在于其含有:
-重链包含SEQ ID NO:9的序列,和
-轻链序列包含SEQ ID NO:10的序列。
6.权利要求1的抗体或其片段,其特征在于其涉及以下人类同种型之一:IgG1、IgG2、IgG3、IgG4。
7.权利要求1的抗体或其片段,其特征在于IgG1同种型的Fc恒定区包含E233P、L234A、L235A、E236P、L237V和/或L238A突变。
8.权利要求1的抗体或其片段,其特征在于IgG1同种型的Fc恒定区包含增加动物或人药代动力学参数t1/2β(小时)或Cmax(μg/ml)的值的任何突变。
9.权利要求1的抗体或其片段,其特征在于IgG1同种型的Fc恒定区包含增加动物或人药代动力学参数t1/2β(小时)或Cmax(μg/ml)的值的M255Y、S257T和/或T259E突变。
10.权利要求1的抗体或其片段,其特征在于其具有以下性质中的至少一种:
a)具有聚集稳定性,使得在浓度高于10mg/ml和储存温度为T=4℃下超过6个月时,聚集体含量增加不超过在溶液中初始含量的5%;
b)具有聚集稳定性,使得在浓度高于10mg/ml和温度增加至37℃超过2周时,聚集体含量增加不超过在溶液中初始含量的5%;
c)具有聚集稳定性,使得在浓度高于10mg/ml和温度增加至50℃超过24小时时,聚集体含量增加不超过在溶液中初始含量的5%;
d)当与人IL-6受体结合时,解离常数KD不大于10-9M;
e)当与人IL-6受体结合时,动力学缔合常数kon为至少105 1/Ms;
f)当与人IL-6受体结合时,动力学解离常数dis不大于10-4 1/s;
g)对白细胞介素-6依赖性DS1细胞的培养物显示抗增殖活性,其中额定值IC50不超过10-8M;
h)对白细胞介素-6依赖性细胞的培养物显示STAT-3信号传导的阻断,其中额定值IC50不超过10-8nM。
11.一种双特异性抗体,其包含权利要求1或从属于权利要求1的任何一项权利要求的抗体的抗原结合片段。
12.一种分离的核酸分子,其编码权利要求1-10中任何一项的抗体或其抗原结合片段。
13.一种表达载体,其包含一种或多种权利要求12的分离的核酸分子。
14.一种细胞系,其包含在细胞中的权利要求13的表达载体或权利要求12的分离的核酸分子。
15.一种用于制备权利要求1或从属于权利要求1的任何一项权利要求的抗体或其抗原结合片段的方法,所述方法包括:在足以产生所述抗体或其抗原结合片段的条件下,在培养基中培养权利要求14的细胞系,并且随后分离和纯化获得的抗体或其抗原结合片段。
16.一种药用组合物,其包含与一种或多种药学上可接受的赋形剂、稀释剂或载体组合的权利要求1-10中任何一项的抗体或其抗原结合片段。
17.权利要求16的药用组合物,其用于治疗与白细胞介素-6(IL-6)的作用相关的疾病或病症或者用于去除或缓解与IL-6的不期望的作用相关的症状。
18.权利要求16的药用组合物,其特征在于其为一种用于胃肠外给予的溶液。
19.权利要求16的药用组合物,其特征在于其为一种冻干粉。
20.权利要求17的药用组合物,其特征在于其意欲用于治疗和/或诊断选自以下疾病或障碍的疾病:皮炎、关节病、自身免疫性肝炎、脱髓鞘病、动脉炎、脱发。

25.权利要求1-10中任何一项的抗体或其片段用于制备药物的用途,其特征在于所述药物意欲用于治疗和/或诊断选自以下疾病或障碍的疾病:成人急性呼吸窘迫综合征、与溃疡性结肠炎相关的牛皮癣性关节病、炎症后间质性肺病、感染后间质性肺病、经典自身免疫性或狼疮样肝炎、晶状体源性葡萄膜炎、幼年类风湿性关节炎。
