Abstract
本发明提供了一种液态植物乳杆菌饲料添加剂及其制备方法,植物乳杆菌发酵液和稳定剂按照一定的比例混合而成。本发明的液态植物乳杆菌饲料添加剂的稳定性好,可以很好的应用于养殖动物中。相比较植物乳杆菌固体饲料添加剂,生产成本降低,节省生产时间。同时降低了养殖户的养殖成本。

技术领域
本发明属于农业生物技术领域,涉及一种液态植物乳杆菌饲料添加剂及其制备方法。
背景技术
中国每年抗生素的使用量有16.2万吨,其中使用在动物上的占52%,约有8.4万吨;使用在人上的占比48%。如果饲料中的药物浓度达到300ppm,这些药物可以生产2.80亿吨饲料,这些饲料足以养活我国所有的食品动物。大量抗生素的使用带来了很多副作用,人类病原菌的耐药性与动物使用促生长抗生素直接相关。耐药菌通过食物链从动物传递到人类,每年约2万人死于超级细菌感染。抗生素的长期使用会破坏肠道菌群平衡,破坏肠道屏障,进而影响动物免疫力,导致疫病难于控制。2016年我国又开启了抗生素禁用的历程,2017年4月30日之后禁止硫酸粘杆菌素添加在饲料中作为生长促进剂。
植物乳杆菌可以抑制肠道中的病原菌,提高机体免疫力、抗氧化能力等,是潜在的一种替代抗生素产品,具有无毒副作用、无污染、无耐药的特点。
发明内容
本发明提供一种液态植物乳杆菌饲料添加剂及其制备方法,作为一种新型替代抗生素产品,具有无毒副作用、无污染、无耐药的特点。
本发明解决上述问题所采用的技术方案是,提供一种液态植物乳杆菌饲料添加剂及其制备方法,主要由以下重量组分制成:植物乳杆菌发酵液96%~98%、稳定剂2%~4%。
所述的植物乳杆菌发酵液的制备方法,包括以下步骤:
1)制备种子液:采用植物乳杆菌优化培养基对植物乳杆菌在33~37℃静置培养16~20h,制得植物乳杆菌种子液。
2)制备发酵液:向经过121℃灭菌20min的植物乳杆菌优化培养基中以质量百分数3%~10%的比例加入种子液,在温度33~37℃、pH值为6.2~6.8、压力0.02~0.05MPa,培养14-18h,植物乳杆菌活菌数≥2x1010cfu/mL,制得所述的植物乳杆菌发酵液。
3)按照比例将稳定剂加入到2)中的植物乳杆菌发酵液中,混匀,即得液态植物乳杆菌饲料添加剂。
所述优化培养基包括以下质量百分数的组分:蔗糖1.5%-3%、酵母浸粉1%-2.5%、磷酸氢二钾0.01%-0.03%、硫酸镁0.005-0.008%、余量为无菌水。
所述稳定剂每克中包括以下组分:葡萄糖氧化酶0.18~0.32g、低聚异麦芽糖0.1~0.15g、低聚果糖0.1~0.15g、海藻糖0.15~0.25g、其余余量为甘油。
益生元类物质可以被植物乳杆菌的迅速吸收,为植物乳杆菌提供养分,延长植物乳杆菌的活力和寿命,有效的提高液态植物乳杆菌的稳定性。而葡萄糖氧化酶的加入,可以分解葡萄糖,从而抑制其他细菌的生长,进而保持液态植物乳杆菌的稳定性。
本发明的有益效果为:
本发明制备方法简单可行,符合工业化生产要求。提高了液体植物乳杆菌的稳定性,可以很好的应用于养殖动物中。相比较植物乳杆菌固体饲料添加剂,生产成本降低,节省生产时间。同时降低了养殖户的养殖成本。
具体实施方式
下面结合实施案例对本发明作更进一步的说明,但本发明的实施方式不限于此,下述实施例中所使用的实验方法如无特殊说明,均为常规方法。
实施例1
取植物乳杆菌发酵液和稳定剂98重量份和2重量份,混匀,制得液体饲料添加剂。
液态植物乳杆菌饲料添加剂制备
1)制备种子液:采用植物乳杆菌优化培养基对植物乳杆菌在33~37℃静置培养16~20h,制得植物乳杆菌种子液;
2)制备发酵液:向经过121℃灭菌20min的植物乳杆菌优化培养基中以7wt%加入种子液,在温度35℃、pH值为6.5、压力0.02MPa,培养16~18h,植物乳杆菌活菌数为2.6x1010cfu/mL,制得所述的植物乳杆菌发酵液;
3)按照2%比例将每克中包括以下组分:葡萄糖氧化酶0.18g、低聚异麦芽糖0.10g、低聚果糖0.1g、海藻糖0.15g、其余余量为甘油的稳定剂加入到2)中的植物乳杆菌发酵液中,混匀,即得液态植物乳杆菌饲料添加剂。
使用的优化培养基为:蔗糖1.5%、酵母浸粉1%、磷酸氢二钾0.01%、硫酸镁0.005%、余量为无菌水。
本实施案例中提供的液体植物乳杆菌饲料添加剂初始活菌数为2.58x1010cfu/mL,室温存放60天后,活菌数≥1x1010cfu/mL。
实施例2液态植物乳杆菌饲料添加剂制备
取植物乳杆菌发酵液和稳定剂96重量份和4重量份,混匀,制得液体饲料添加剂。
制备发酵液:采用植物乳杆菌优化培养基对植物乳杆菌在33~37℃静置培养16~20h,制得植物乳杆菌种子液;向经过121℃灭菌20min的植物乳杆菌优化培养基中以7wt%加入种子液,在温度35℃、pH值为6.5、压力0.02MPa,培养16~18h,植物乳杆菌活菌数为2.6x1010cfu/mL,制得所述的植物乳杆菌发酵液;按照96:4将每克中包括以下组分:葡萄糖氧化酶0.32g、低聚异麦芽糖0.15g、低聚果糖0.15g、海藻糖0.25g、其余余量为甘油的稳定剂加入到2)中的植物乳杆菌发酵液中,混匀,即得液态植物乳杆菌饲料添加剂。
使用的优化培养基包括以下质量百分数的组分:蔗糖3%、酵母浸粉2.5%、磷酸氢二钾0.03%、硫酸镁0.008%、余量为无菌水。
本实施案例中提供的液体植物乳杆菌饲料添加剂初始活菌数为2.54x1010cfu/mL,室温存放60天后,活菌数≥1x1010cfu/mL。
实施例3:效果例在提高动物抗氧化能力方面中的应用
为验证本发明的应用效果,本发明的饲料添加剂在1日龄AA肉鸡中进行试验。将鸡分为2组,分别为对照组和试验组,每组60只,其中试验组为饲喂本发明的实施例2饲料添加剂组,按照饮水量的0.1%(g/w)使用,使用周期为1~28d。养殖方式为笼养,自由采食和饮水。舍内光照、温度、湿度严格按照常规饲养管理要求进行,按照常规免疫程序对雏鸡进行新城疫和法氏囊免疫,定期对食槽和饮水器进行消毒。试验周期为28天,分别在肉鸡14、28日龄从每组中随机选取10只鸡,颈动脉采血,3000r/min离心10min分离血清,使用T-AOC、MDA试剂盒检测含量。

同行龄数据表不同小写字母表示差异显著(P<0.05),相同小写字母或无字母标示差异不显著(P>0.05)。
由实施案例2的对比实验可以看出,本发明的饲料添加剂可以显著提高肉鸡的抗氧化能力,进而改善肉鸡的免疫功能,提高生产性能。

1.一种液态植物乳杆菌饲料添加剂,其特征在于,各组分重量百分比为植物乳杆菌发酵液96%~98%、稳定剂2%~4%。
2.一种液态植物乳杆菌饲料添加剂的制备方法,其特征在于,包含以下步骤:
1)制备种子液:采用植物乳杆菌优化培养基对植物乳杆菌在33~37℃静置培养16~20h,制得植物乳杆菌种子液;
2)制备发酵液:向经过121℃灭菌20min的植物乳杆菌优化培养基中以质量百分数3%~10%的比例加入种子液,在温度33~37℃、pH值为6.2~6.8、压力0.02~0.05MPa,培养14~18h,植物乳杆菌活菌数≥2x1010cfu/mL,制得所述的植物乳杆菌发酵液。
3)按照比例将稳定剂加入到2)中的植物乳杆菌发酵液中,混匀,即得液态植物乳杆菌饲料添加剂。
3.根据权利要求2所述的一种液态植物乳杆菌饲料添加剂的制备方法,其特征在于,所述优化培养基包括以下质量百分数的组分:蔗糖1.5%~3%、酵母浸粉1%~2.5%、磷酸氢二钾0.01%~0.03%、硫酸镁0.005~0.008%、余量为无菌水。
