非甾体抗炎药(下文中称NSAID)处方的主要适应症为不同性质和 位置的炎性过程、疼痛和发热。NSAID是一种最广泛使用的药物组。 例如,NSAID被开处方给约20%的患有各种内脏器官疾病的患者 [Guidelines for Pre-clinical Drug Research.Part one/edited by A.N.Mironov.—M.:Grif and K Publ.,2012.p.944]。

迄今为止已知的NSAID(即,非选择性环氧合酶抑制剂)的缺点是 其显著的胃毒性[Handa,O.The impact of non-steroidal anti-inflammatory drugs on the smallintestinal epithelium/O. Handa,Y.Naito,A.Fukui,T.Omatsu,T.Yoshikawa//J.Clin.Biochem.Nutr.-2014.-V.54,N.1.-P.2-6.]。COX-2的选择性抑 制剂胃毒性更小,但是已知其对于心血管系统有不良影响[Singh,B.K. 从申请人的观点来看,基于技术水平的分析,开发抗炎药物的最 有希望的方向是寻找非选择性NSAID。同时,如上所述,开发者面对 减少副作用(主要的是胃毒性)的任务。
保护非选择性COX抑制剂的羧基是减少该组NSAID的毒性的主要 方式之一。酯保护最通常用于该目的[Liu,W.Synthesis and biological evaluation of curcuminderivatives containing NSAIDs for their anti-inflammatory activity[Text]/W.Liu,Y. 包含萘普生的三种片段的原型型式(prototype version)是一种 有效的抗炎化合物。特别地,其对于体内亚急性(福尔马林)水肿模型 具有显著的抗炎作用。
然而,对于体内急性(角叉菜胶)水肿模型,该原型不是足够有效 的。重要的是注意到该原型并非同时具有所述技术方案固有的一组性 质,即高的抗炎(对于体内急性(角叉菜胶)水肿模型)、镇痛和解热活 性与低毒性(包括胃毒性)的组合。为此,本申请人并不考虑将该原型 作为比较药物,反而将现代的非甾体抗炎药物萘普生及其类似物用于 该目的(表3)。
所要求组合物的所要求技术方案与所述原型中描述的不同,在于 存在另外的萘普生片段,以本申请人的实验证实的观点来看,其引起 出现药理学显著的积极作用。因此,在所要求的分子中在生理条件下 分裂的共价酯键和连接萘普生和吡哆醇片段的非共价离子键的组合, 导致出现对于专业人员而言不明显的协同作用。这些作用表现为抗炎、 镇痛和解热活性升高,以及胃毒性和急性毒性显著降低。特别地,针 对降低胃毒性和急性毒性的背景下所要求的式I的化合物确保了快速 产生对于体内急性和亚急性水肿两者模型的显著抗炎作用,由此本申 请人可以解决包括萘普生的原型和许多其它NSAID(后者也是所要求 技术方案的原型)的特征性的表面上不可逾越的问题。
所要求技术方案的目的是产生基于萘普生的药物,其具有高的抗 炎、镇痛和解热活性与低毒性(包括胃毒性)的组合,显著地扩大了明 确提出的目的的已知方式的药物库。
本发明的技术成果是一种基于吡哆醇和萘普生的新的非甾体抗炎 药,与已知的NSAID(包括原型)相比,其在显著较低毒性(包括胃毒性) 的背景下,显示出高的抗炎、镇痛和解热活性。

NMR光谱记录在Bruker AVANCE-400装置上。测定相对于氘化溶 剂的残余质子(1H和13C)信号的化学位移。使用Stanford Research Systems MPA-100OptiMelt以1℃/min的加热速率测定产品的熔化 温度。使用UV分光光度计Evolution 300(Thermo scientific)研究 式I化合物溶液的光谱特性。在使用石英基质管100° Z的自动偏光计 ADP440+(B&S)(英国)上测定比旋度。使用TripleTOF 5600,AB Sciex (德国)质谱仪,通过离子-涡轮喷雾(TIS)—在氮分子的碰撞能量等于 10eV下从甲醇溶液中记录高分辨率质谱(HRMS)。

所要求的技术方案的具体实施的实施例
根据下述方案,使用市售可获得的底物、试剂和溶剂,以两个步 骤进行包含吡哆醇(维生素B6)和萘普生的片段的所要求式I的化合物 的获得。
实施例1.获得(2S,2'S)-(5-((S)-2-(6-甲氧基萘-2-基)丙酰氧 基)-6-甲基吡啶-3,4-二基)双(亚甲基)双(2-(6-甲氧基萘-2-基)丙酸 酯)(化合物IV)
向圆底烧瓶中装入在5l丙酮中的28.35g的盐酸吡哆醇II、 95.23g的氧萘丙酸III、50.52g的4-N,N-二甲基氨基吡啶和123.73 g的二环己基碳二亚胺。搅拌反应混合物直到形成二环己基脲沉淀才 停止,然后过滤沉淀,用200ml的丙酮洗涤,并且蒸发混合渗滤液。 用在硅胶上的柱色谱(洗脱液乙酸乙酯-石油醚1:1)纯化产物。获得白 色结晶物质形式的产物IV(产率82.0g,74%)。更详细地,该化合 物的制备及其性质描述在专利RU2513098的发明中。
实施例2.3-((S)-2-(6-甲氧基萘-2-基)丙酰氧基-4,5-双 (((S)-2-(6-甲氧基萘-2-基)丙酰氧基)甲基)-2-甲基吡啶鎓 (S)-2-(6-甲氧基萘-2-基)丙酸盐(化合物I)的制备
将82.0g的化合物IV放入圆底烧瓶中,加入1L的丙酮,并搅 拌该混合物10分钟。向得到的溶液中加入18.22g的(S)-2-(6-甲氧 基-2-萘基)丙酸(氧萘丙酸)III,并搅拌直到沉淀完全溶解。在旋转蒸 发器(90rpm,残余压力10mbar,温度40℃)上,在真空中除去丙酮。收集沉淀,并在旋转蒸发器(90rpm,残余压力10mbar,温度80℃) 上,在真空下干燥。所要求的化合物I的产率为100.1g(99.9%)。
熔点T为145℃。[α]24 D=+22.8°(c=3.01,CH2Cl2)。在乙腈和亚甲基氯中,在范围200至450nm的波长中,测量式I化合物溶液的吸收光谱,在此情况下,其具有的最大吸收位于262、272、317和332nm。最大位点偏差为±2nm。溶液浓度为40μg/ml。在1H NMR 光谱(400MHz,CDCl3,δ,ppm,J/Hz)中观察到下述信号:1.33(br d,3H,CH3,3J=3.7);1.40(d,3H,CH3,3J=7.1);1.50(d,6H, 2CH3,3J=7.1);1.90(br s,3H,CH3);3.47(br s,1H,);3.65 (k,1H,СН,3J=7.1);3.74-3.87(m,13H);4.50(br s,1N); 4.80(br s,1N);4.95和5.04(AB,2H,CH2,2J=-12.8); 6.95-7.66(m,20N),8.19(s,1H,CH)。在13C NMR光谱(100MHz, CDCl3,δ,ppm)中观察到下述信号:17.96;18.42;18.52;18.92; 45.02;45.34;45.38;55.40;57.13;61.65;105.66;105.73; 119.09;119.13;119.18;119.38;125.96;126.09;126.16;126.20; 126.38;126.57;127.30;127.36;127.49;128.96;129.01;129.39; 129.64;133.78;133.81;133.86;134.05;134.13;135.18;135.21; 135.42;136.16;144.78;147.16;152.75;157.74;157.79;157.96; 171.95;174.02;179.30。
目标物质I为化合物IV与萘普生的盐。在这方面,在反相HPLC 条件下(表1),在色谱图中看到两个对应于化合物IV和萘普生的峰, 其质谱对应于理论数据和文献数据(分别为806.3324和229.0870)。
式I化合物的急性毒性的研究
根据固定剂量方法,在两种性别的Wistar大鼠(每组6只动物) 中进行试验。胃内给药的初始剂量为5000mg/kg。使用的溶剂为0.5% 的吐温80溶液(吐温80-聚氧乙烯,脱水山梨醇和油酸的衍生物,一 种市售可获得的聚合物),其是通过将0.5g的土温80溶于100ml的蒸馏水中制备的。
为了制备5000mg/kg的剂量,将25g的试验化合物用天平Vibra (Shinko Denshi,Japan,cat.№AF225DRCE)称重于聚苯乙烯皿中, 转移到精确等级A的100ml容量瓶中,用0.5%的吐温80水溶液调节 至100ml,并使用分散器SilentCrusher(Heidolph,Germany,P/N 595-06000-00-3)搅拌制剂悬浮液至均匀稠度。
向禁食(不少于8小时的时间)且自由获取水的动物进行引入。基 于在即将引入物质之前记录的体重,分别计算每只动物的给药体积。 在引入之后一小时重新开始获取食物。在给药30分钟之后分别观察动 物,然后每小时观察至少一次,共4小时,接着每日一天一次进行观 察,共14天。在即将给药所述制剂之前记录体重,以计算给药体积, 然后每两天记录体重一次。如果在研究期间动物死亡,则尽可能精确 地确定并记录死亡时间。尽快将动物称重并解剖。将死亡的动物称重, 处以安乐死并解剖。通过吸入二氧化碳将动物处以安乐死。用IBM SPSS 统计学软件,使用概率分析计算中毒剂量LD10、LD16、LD50和LD84。
当胃内给药5000mg/kg的剂量时,观察到对动物活动性的微小抑 制。在20-30分钟内,该症状消失。在1.5-2小时之后,动物开始进 食和水。三天后,1只大鼠(雌性)死去。在整个研究中再没有任何致 死例。
在整个实验期间,实验动物中生命活动的所有主要指标响应正常, 与对照动物没有不同。小鼠具有良好的食欲,皮毛光亮,可见的粘膜 颜色是浅粉色,在观察期期间,行为符合这种动物行为,没有观察到 任何异常。在试验开始之后2天和4天,大鼠的体重分别增加(1-4)%和 (2-6)%,其约相当于对照组大鼠的体重增加。在研究的第8天,动物 体重增加(2-8)%。在试验结束时,体重增加(5-13)%。急性毒性参数列 在表2中。

胃内给药式I化合物的急性毒性
在实验结束时,进行对照动物和实验动物的安乐死和病理形态解 剖。在大鼠尸体解剖期间没有观察到任何变化。动物尸体具有合适的 构成(correct constitution),一般肥胖或高于一般肥胖。自然孔 (openings):嘴巴闭合,舌头在嘴中,嘴唇粘膜和齿龈是浅粉色、光 滑且发亮。鼻孔-粘膜为浅粉色、干、没有流出物,渗透性良好。耳壳 没有变化;外耳道干净。肛门闭合,粘膜是浅粉色。毛发保持良好, 毛发亮。皮肤有弹性,皮下纤维表现良好,具有浅黄色颜色且有弹性。 肌肉微红,发育良好,肌腱和韧带是白色、弹性且耐用。骨骼和关节 的结构没有破坏。胸腔和腹腔器官的位置在解剖学上是正确的。胸腔 和腹腔中没有任何体液。咽和食道的通畅性没有被破坏。心脏体积没 有变化。心脏内腔包含少量未结块血液,心内膜光滑且发亮。肺为浅 粉色至红色,颜色均匀,没有溶胀体征,分叶征(lobulation)表现良 好。脾没有增大,边缘尖锐,形状为椭圆形,具有弹性密实度,颜色 为红褐色。肝没有增大,边缘尖锐,形状没有变化,密实度稠密,颜 色为红褐色。胃包含均匀密实度的灰色食块。胃粘膜为浅灰色。肠的 薄部分和厚部分的粘膜具有浅粉色或浅灰色颜色。肾脏为豆形,颜色 为深褐色,前肾体中存在中等量的脂肪,包膜容易分开,皮层和脑区之间的边界清楚。膀胱是空的或充满浅黄色颜色的尿液,粘膜具有浅 粉色颜色。生殖器没有异常。雄性的睾丸具有弹性密实度,位于阴囊 腔中,具有椭圆形。雌性具有正常的卵巢和子宫。脑没有水肿,脑物 质是弹性的,没有出血。
因此,进行的研究表明在胃内给药之后所要求的式I的化合物属 于第4危险类型,即低危险物质(GOST 12.1.007-76“Harmful substances.Classification and generalsafety requirements"), 并且根据LD50致死量方面的其安全性超过了迄今为止已知的大多数 NSAID(表3)。例如,所要求的化合物的毒性比萘普生低8倍。

式I化合物的胃毒性
在胃毒性研究中,采用6-7周龄的雌性和雄性Wistar大鼠,体重 180-220g。每组动物数量为10。
在研究之前,向禁食16小时的大鼠胃内给药式I的化合物一次。 在给药2000mg/kg剂量(0.5%的吐温80水溶液)的试验物质悬浮液之 后三小时,将动物处以安乐死;切除它们的胃,沿着胃小弯切开,并 在生理溶液中洗涤以除去内含物。
以4-点量表进行胃毒性的评价:
0—没有损伤;
0.5—充血;
1—单个较小损伤(1或2点出血);
2—多发性损伤(糜烂,点状出血);
3—显著且多发性粘膜损伤(糜烂,出血);
4—覆盖整个粘膜表面的总体损伤(大出血,糜烂,穿孔)。
根据评价结果,测定UD50—引起相当于2点的胃毒性(产生溃疡) 作用的试验物质剂量。
为了制备2000mg/kg的剂量,将32.0g的式I化合物用天平 Vibra(Shinko Denshi,Japan,cat.№AF225DRCE)称量于聚苯乙烯 皿中,转移到精确等级A的400ml容量瓶中,用0.5%的吐温80水溶 液调节至400ml,并使用分散器SilentCrusher(Heidolph,Germany,P/N 595-06000-00-3)搅拌制剂悬浮液至均匀稠度。以不超过5 ml/200g的量进行引入到大鼠中。
通过二氧化碳吸入方法将动物处以安乐死。
在单次引入2000mg/kg剂量的式I化合物时,在雄性大鼠中,胃 毒性作用相当于平均0.6点,在雌性中相当于0.4点(表4)。

因此,对于式I的化合物,UD50值>2000mg/kg,其超过了大多 数NSAID的类似参数(表3)。例如,所要求的化合物的毒性比萘普生 低40倍。
式I的化合物的体外抗炎活性
结构酶环氧合酶1(COX 1)在各种细胞类型中表达,并且参与确 保其正常(生理学)功能活动。在重度炎症病症中,环氧合酶2(COX 2) 负责前列腺素的合成。
COX活性的分析包括测量环氧合酶的过氧化物酶活性。在PGG2(前 列腺素G2)与作为COX探针使用的试剂盒("环氧合酶(COX)活性分析试 剂盒(荧光测定)"(Biovision,目录号_K 549-100)中存在的ADHP (10-乙酰基-3,7-二羟基吩嗪反应期间,形成荧光组分试卤灵 (resorufin),其吸收波长为530-540nm,发射波长为(585-595)nm。 荧光强度与样品中COX的残余活性成正比。选择皮肤成纤维细胞(HSF) 作为环氧合酶的来源。成纤维细胞溶解产物显示出具有2.131μU/mg 的COX活性。
细胞溶解产物的制备用10ml的磷酸盐缓冲液洗涤皮肤成纤维 细胞的细胞(2-6×106)一次。将所述细胞再悬浮在5ml的缓冲液中, 并转移到15ml的管中。将其以1500g离心4分钟。然后,倾倒出上 清液,并将细胞沉淀物再悬浮在0.5-1ml的含有蛋白酶混合物的冷裂解缓冲液(约0.4ml的缓冲液/100μl的细胞沉淀物),并在4℃下以 10,000g离心15分钟。分离上清液,并用作COX的来源。
试剂的制备。在即将使用之前,通过将2μl的辅因子加入到 398μl的缓冲液中将COX辅因子溶解200倍。混合65μl的花生四烯酸 和65μl的NaOH,然后用1170μl的蒸馏水稀释10倍。该溶液稳定1 小时。
通过混合如下指出的下述试剂制备反应混合物(用于2个平行 孔)∶
-COX探针-2μl;
-溶解辅因子-4μl(稳定1小时);
-细胞溶解产物—20μl;
-借助于COX缓冲液,体积恢复到172μl。
试验物质的制备:
使用萘普生储备溶液和在二甲亚砜(DMSO)中浓度1000μM的式I 的物质以2个步骤递增量制备初步稀释液。分别获得从62.5至 1000μM的5个浓度。
使用多道移液管,将试剂加入到合适板的各孔中:首先是反应混 合物,然后是一系列浓度的试验物质在DMSO中的溶液和对照物质(抑 制剂COX 1 SC560和抑制剂COX 2塞来考昔),纯溶剂DMSO(用于具 有较高活性的各孔)。重复使用两份。并将它们培养5分钟。
引发反应。在反应开始之前,在板的所有孔中进行单个荧光测 量,以解释试验物质和对照物质的自身信号。然后,使用多道移液 管,将10μl的花生四烯酸溶液加入到板的所有孔中,以引发反应。 在加入花生四烯酸之后,每15秒立即以动力学模式测量荧光(Ex/Em= 535/587nm),共测量30分钟。
根据下式计算对照和试验物质图像的荧光参数∶

根据下式计算COX抑制的百分比:
进一步,对于试验物质和参照物质(萘普生)使用可用数据点,我 们构建了最合适的S型抑制曲线。基于该曲线,计算试验样品对于COX 的IC50。
根据使用的试剂盒的制造商的建议,将2μl体积(该量足以完全 抑制合适的同工型)的COX-1抑制剂SC560和COX-2抑制剂塞来考昔加 入到反应混合物中。在用SC560和塞来考昔处理之后,细胞溶解产物 中包含的COX的残余活性分别为72.8%和32.0%。
在抑制活性的研究中,孔中萘普生和试验物质的浓度为1.25至 20μM。
为了测定每种同工型制剂的抑制比例,在选择性抑制剂COX 1 SC560的存在下,以及在选择性抑制剂COX 2塞来考昔的存在下,以 浓度模式测试所研究的物质。加入足量的抑制剂以完全抑制所述酶的 相应同工型,而其它同工型保留活性。
通常,假定在仅试验制剂存在下的活性为100%,则计算COX 1和 COX 2的残余活性和所述制剂对每种同工型的抑制百分比。式I的化 合物显示出对于COX-2同工型优先的抑制活性.
因此,萘普生IC50(原型)为9.56μM,其为式I的化合物的IC50的1.25倍,等于7.7μM。根据上述,可以得出结论所要求的式I的 化合物作为环氧合酶抑制剂更有效,因为其在较低浓度下起作用。
式I的化合物的体内抗炎活性
使用6-7周龄雌性和雄性Wistar大鼠进行实验,体重180-220g。 每组动物数量为10。
通过足下(脚底或跖腱膜下)给药0.1ml的1%角叉菜胶溶液(来自 爱尔兰海苔的硫酸多糖)产生急性炎症反应(水肿)。在诱导炎症之后 3小时,通过爪体积的变化(器官体积测量的)评价炎症反应的严重程 度。在引入1%的角叉菜胶溶液之前1小时,用探针将式I的物质给药 到胃中。使用0.5%的吐温80溶液作为阴性对照。将通过水肿减少评 价的抗炎作用表示为对照的百分比。根据四个剂量分析物的作用结 果,进行ED50的计算。
为了制备50mg/kg的剂量,将1000mg的式I化合物用天平Vibra (Shinko Denshi,Japan,cat.№AF225DRCE)称重于聚苯乙烯皿中, 转移到精确等级A的200ml容量瓶中,用0.5%的吐温80水溶液调节 至200ml,并使用分散器SilentCrusher(Heidolph,Germany,P/N 595-06000-00-3)搅拌制剂悬浮液至均匀稠度。以不超过1ml/100g 的量进行引入到大鼠中。得到的溶液的浓度为5mg/mL。
为了制备20mg/kg的剂量,采用40ml的浓度5mg/mL(50mg/kg) 的溶液,转移到精确等级A的100ml容量瓶中,用0.5%的吐温80水 溶液调节至100ml,并使用分散器搅拌制剂悬浮液至均匀稠度。
为了制备10mg/kg的剂量,采用20ml的浓度5mg/mL(50mg/kg) 的溶液,转移到精确等级A的100ml容量瓶中,用0.5%的吐温80水 溶液调节至100ml,并用磁力搅拌器MR Hei-Standard(Heidolph, Germany)搅拌制剂悬浮液至均匀稠度。
为了制备5mg/kg的剂量,采用4ml的浓度5mg/mL(50mg/kg) 的溶液,转移到精确等级A的100ml容量瓶中,用0.5%的吐温80水 溶液调节至100ml,并使用磁力搅拌器搅拌制剂悬浮液至均匀稠度。
在给药30分钟之后,然后每小时至少一次,共4小时,个体地观 察动物。在即将给药制剂之前记录体重,以计算给药体积。
通过吸入二氧化碳将动物处以安乐死。
当向雄性大鼠给药50、20、10和5mg/kg剂量的式I的化合物时, 与对照组相比,炎性水肿分别减少62.34、51.25、23.57和12.39%。 在雌性大鼠中,当给药50、20、10和5mg/kg剂量的式I的化合物时, 水肿分别减少65.47、67.89、38.64和13.37%。结果列在表5中。

在胃内给药之后式I的化合物对于急性渗出性炎症的作用
根据统计处理所得数据的结果,式I的化合物对于雄性的ED50为 23mg/kg,对雌性为14mg/kg。在没有性别差别下ED50的平均值为18.5 (~19)mg/kg。
根据文献,萘普生的ED50为15mg/kg,布洛芬-48mg/kg,双氯 酚酸钠-8mg/kg,吡罗西康-20mg/kg,保泰松-56mg/kg,吲哚美辛-10mg/kg,阿司匹林-98mg/kg[Guidelines forPre-clinical Drug Research.Part one/edited by A.N.Mironov.—M.:Grif and KPubl.,2012.p.944]。因此,本申请人通过实验证实了式I的化合 物存在高的抗炎活性,其抗炎活性值(七种的四种情形)优于目前 NSAID的抗炎活性或与其相当。
式I化合物的镇痛作用
在式I化合物的镇痛性质的研究中,使用6-7周龄的雌性和雄性 wistar大鼠,体重质量180-220g。每组的动物数为10。
通过向右后爪足下给药0.1ml的1%角叉菜胶溶液(一种来自爱尔 兰海苔的硫酸多糖)在大鼠中产生慢性免疫炎症模型。使用胃探针胃 内给药所述物质。使用萘普生作为比较药物,使用0.5%的吐温80溶 液作为阴性对照。将通过水肿减少评价的抗炎作用表示为相对于对照 的百分比;根据四个剂量试验物质的作用结果,进行ED50的计算。
由角叉菜胶引起炎性痛觉过敏(大鼠中发炎组织的疼痛敏感性提 高),并在引入角叉菜胶之前和引起其之后3小时,通过动物爪组织的 疼痛灵敏度阈值-PST(PST差异)降低为机械性刺激(挤压)评价炎性痛 觉过敏。对发炎的爪进行测量。使用的痛觉测量器UgoBasile S.R.L. (Italy)提供大鼠发炎爪上的后负荷逐渐升高直到疼痛反应出现(通过 动物的吱吱声或抽回爪评价)。在给药角叉菜胶之后2小时给药分析 物。在胃内给药分析物之后1小时,通过痛觉过敏降低评价镇痛作用, 用ED50评价。在研究中物质的影响下疼痛反应阈值的增大用肢体压力 的增加表示,并且表征制剂镇痛作用的强度。在试验之后,通过二氧 化碳吸入方法将动物处以安乐死。
为了制备50mg/kg的剂量,将1000mg的式I化合物用天平Vibra (Shinko Denshi,Japan,cat.№AF225DRCE)称重于聚苯乙烯皿中, 转移到精确等级A的200ml容量瓶中,用0.5%的吐温80水溶液调节 至200ml,并使用分散器SilentCrusher(Heidolph,Germany,P/N 595-06000-00-3)将悬浮液分散至均匀稠度。以不超过1ml/100g的 量进行引入到大鼠中。得到的溶液的浓度为5mg/mL。
为了制备20mg/kg的剂量,采用40ml的浓度5mg/mL(50mg/kg) 的溶液,转移到精确等级A的100ml容量瓶中,用0.5%的吐温80水 溶液调节至100ml,并使用分散器将悬浮液分散至均匀稠度。
为了制备10mg/kg的剂量,采用20ml的浓度5mg/mL(50mg/kg) 的溶液,转移到精确等级A的100ml容量瓶中,用0.5%的吐温80水 溶液调节至100ml,并用磁力搅拌器MR Hei-Standard(Heidolph, Germany)搅拌悬浮液至均匀稠度。
为了制备5mg/kg的剂量,采用4ml的浓度5mg/mL(50mg/kg) 的溶液,转移到精确等级A的100ml容量瓶中,用0.5%的吐温80水 溶液调节至100ml,并使用磁力搅拌器搅拌悬浮液至均匀稠度。
实验结果列在表6中。

式I的化合物对大鼠的镇痛作用
根据所给出的结果,可见所要求的式I的化合物在5mg/kg的剂 量下就已经显示出显著的麻醉作用,而萘普生(原型)只有在15mg/kg 的剂量下具有类似的作用,其为其具有较高效率的证明性事实。因此, 所要求化合物在其镇痛活性方面的治疗指数(LD50/ED50)大于1000,相 对于萘普生为42,安全指数(UD50/ED50)大于400,相对于萘普生为3.2。
式I的化合物对大鼠的解热作用
在式I化合物的解热性质的研究中,使用6-7周龄的雌性和雄性 Wistar大鼠,体重质量180-220g。每组的动物数为10。
通过皮下给药20%的面包酵母悬浮液产生发热反应。在引入酵母 菌之前和引入其之后18小时,用热电偶温度计测量直肠温度(这些测 量的差异为估计的高热反应)。在高热峰值(在18小时之后)向动物给 药制剂一次。通过在注射试验物质之后2小时高热降低评价解热作用, 然后以每小时间隔记录作用动态,共记录7小时。
为了制备75mg/kg的剂量,将1500mg的式I化合物用天平Vibra (Shinko Denshi,Japan,cat.№AF225DRCE)称重于聚苯乙烯皿中, 转移到精确等级A的200ml容量瓶中,用0.5%的吐温80水溶液调节 至200ml,并使用分散器SilentCrusher M(Heidolph,Germany)搅 拌制剂悬浮液至均匀稠度。以不超过1ml/100g的量进行引入到大鼠 中。得到的溶液的浓度为7.5mg/mL。
为了制备50mg/kg的剂量,采用66.7ml的浓度7.5mg/mL(75 mg/kg)的溶液,转移到精确等级A的100ml容量瓶中,用0.5%的吐 温80水溶液调节至100ml,并使用分散器搅拌制剂悬浮液至均匀稠 度。
为了制备20mg/kg的剂量,采用26.7ml的浓度7.5mg/mL(75 mg/kg)的溶液,转移到精确等级A的100ml容量瓶中,用0.5%的吐 温80水溶液调节至100ml,并用磁力搅拌器MRHei-Standard (Heidolph,Germany)搅拌制剂悬浮液至均匀稠度。将在制剂给药之 后和引入酵母菌18小时之后的温度差作为高热降低的参数。
在整个研究中,在给药之后分别观察各个动物。在即将给药所述 制剂之前记录体重,以计算给药体积。对于所有数据使用描述统计学∶ 计算平均值和平均标准误差,最终结果列在表7中。使用Wilcoxon 非参数标准进行实验组的统计比较。确定0.05显著性水平的差异。使 用R-studio程序进行统计分析。各种剂量的式I化合物的解热作用的 研究结果列在表7中。

在皮下引入面包酵母悬浮液之后18小时,在动物组中观察到直肠 温度正向升高平均1.5℃。在整个观察期间(在给药制剂之后直到7小 时),观察到对照组的动物温度没有降低。在向雄性和雌性都引入20 mg/kg剂量的式I化合物时,没有观察到任何解热行为。当剂量升高 至50mg/kg时,观察到雌性组的温度显著降低-因此,在引入物质之 后2和3小时,温度比引入之前降低0.9℃,在引入之后4、5、6和7 小时,降低(0.5-0.7)℃。
当引入50mg/kg剂量的式I化合物时,在给药之后3-4小时,观 察到温度显著降低。降低水平等于(0.34-0.97)℃。
在给药75mg/kg剂量的式I化合物的动物组中,观察到最大作用。 因此,在整个试验期间引入75mg/kg剂量的物质时,温度显著降低, 比引入之前低(0.76-1.5)℃。
当引入15mg/kg(或65μmol/kg)剂量的萘普生时,温度也显著地 降低。温度降低水平为(0.74-1.6)℃。
归纳所得到的结果,应当得出结论,在75mg/kg的剂量时,所要 求的化合物具有在15mg/kg的萘普生剂量水平下的解热性质。应当特 别地注意到,在摩尔方面,这些剂量事实上没有差异,因为萘普生的 摩尔质量显著低于式I的化合物的摩尔质量,等于230g/mol,相对 于所要求的化合物的摩尔质量为1036g/mol。所要求化合物在其解热 活性方面的治疗指数(LD50/ED50)大于67,相对于萘普生为42,安全 指数(UD50/ED50)大于27,相对于萘普生为3.2。
因此,根据上述得出:申请人获得了基于吡哆醇和萘普生的新化 合物-3-((S)-2-(6-甲氧基萘-2-基)丙酰氧基)-4,5-双(((S)-2-(6- 甲氧基萘-2-基)-丙酰氧基)-甲基)-2-甲基吡啶鎓(S)-2-(6-甲氧基 萘-2-基)-丙酸盐,其同时具有高的抗炎、镇痛和解热性质,优于萘普 生(原型)的类似性质或与其相当,同时保持极低毒性(包括胃毒性)。
通常,与萘普生(原型)相比,所要求的化合物的毒性低8倍且胃 毒性低40倍。
所要求的技术方案满足了应用于发明的“新颖性”的标准,因为 根据所研究的技术水平,没有发现在产生实现所述技术成果的基本特 征方面与所要求的所述方案一致的技术方案,所要求的技术方案为具 有低毒性(包括胃毒性)的新的吡哆醇和萘普生衍生物。所要求的化合 物扩展了用于治疗风湿性疾病以及伴随炎症、疼痛和发热的疾病的产 品范围,并且是全新的,在其有效性和使用安全性方面是世界上无与 伦比的。
所要求的技术方案满足应用于发明的“创造性”的标准,因为对 于本技术领域的技术人员而言是非显而易见,即,根据研究的技术水 平没有鉴定出所要求结构的任何抗炎、镇痛和解热药物。
所要求的技术方案满足“工业实用性”的标准,因为其可以使用 标准装置、熟知的国产材料和技术由任何专业化企业实施。本申请人 在实验室中获得了目标产品—基于萘普生的非甾体抗炎药,具有所声 称的技术成果且获得了所有所声称的目的。
