众所周知,通常需要高温程序方法来测定各种样品基质中的半挥发性分析物,例如PAH和农药。然而,当使用例如图1中所示的环形束12的现有技术LTM柱技术时,在这些高温下获得较差的峰形状和较差的分辨率是相当典型的。这个问题主要是由GC柱10中的真实温度与方法中设定的值不匹配造成的,所述不匹配是因为GC柱上存在的冷点20,特别是在靠近导电箔14的区域。此外,在GC柱10上不靠近温度传感器18设置处的区域也可存在冷点20。这个现象在图1中示出,图1仅示出了环形束12内部分可能的冷点20的示例。
发明内容
本发明的第一实施例包括一种用于执行野外便携式GC/MS测量的系统和方法,野外便携式GC/MS测量用于对环境样品中的高温沸腾半挥发性有机化合物进行快速取样和测量,其中GC柱的毛细管中可防止高温沸腾半挥发性成分洗脱GC柱的冷点可通过低热质(LTM)GC模块的新设计来消除。
对于本领域的技术人员而言,通过考虑以下结合附图的详细描述,本发明的这些和其它目的、特征、优点和替代方面将变得显而易见。

在附图中,本发明的各个元件将给出数字标号并对本发明进行讨论,现在将参照附图,以便使得本领域的技术人员能够制造和使用本发明。但是应当理解的是,下面的描述仅是本发明的原理的示例,而不应当被视为缩小所附权利要求的范围。
本文献描述了野外便携式GC/MS(Torion T-9,PerkinElmer公司,Shelton,CT)对各种样品(气体、液体、固体)的分析结果,包括对高沸点、半挥发性有机化合物(SVOC)的分析,分析时间通常不超过10分钟。
为了使如图1所描述的LTM GC模块内的冷点减少,如图2所示,设计了新LTM GC柱束。图2示出了用薄铝覆盖物包裹单层有序排列的GC柱和绝缘的加热线的LTM GC柱30的第一实施例。这个柱在几乎消除或至少部分最小化沿着柱的冷点的同时,可提供均匀的热分布,从而改进高温沸点化合物所需的高温GC运行时用于SVOC的色谱分离。这个新LTM GC模块30的原理在从图2开始的第一实施例中示出。
图2示出了LTM GC模块30的第一实施例的透视图。LTM GC模块30可包括LTM加热器柱组装件、LTM毛细管柱组装件和传感器组装件。
图3是图2中所示的LTM GC模块30的截面图。截面图示出GC柱30设计的各个组件。LTM GC模块30可包括内柱环32、中柱环34和外柱环36。突出部(tab)42也可设置在外柱环36上,以使突出部42可折叠在内柱环32上方并以理想的形状将LTM GC模块固定在一起。内柱环32、中柱环34和外柱环36可包括例如铝的导热材料。
如图所示,加热线38可设置在内柱环32和中柱环34之间。此外,单层的毛细管40可设置在中柱环34和外柱环36之间。在图中未示出温度传感器但将在后面示出。
加热线38设置在内柱环32和中柱环34之间,以便加热中柱环34的内表面。如上所述,选择用于中柱环34的材料,使得热可通过中柱环34均匀地传递至中柱环34的外表面。
应当理解的是,加热线38围着内柱环32盘绕的具体次数不重要,并且该次数取决于使用的加热线38的规格。重要的是,对设置在加热线38上方的中柱环34的内表面的加热应该基本上是均匀的。因此,围绕内柱环32的加热线38的线圈的数量可与图3中所示出的线圈数有所不同。此外,只要不干扰中柱环34的内表面的均匀加热,加热线38可重叠。因此,加热线的任意重叠也应该是均匀的,以使中柱环34的加热是均匀的。
加热线可以是任意适当的厚度和材料。在图3中所示的示例中,加热线38是27AWG(American wire gauge),但是这仅是示例,而不应当被认为是对可使用的加热线厚度的限制。
同样地,LTM GC模块32可包括ID约0.10mm且长度约为5.5米的毛细管。应当理解的是,这些尺寸仅用于说明目的,而不是对可使用的尺寸的限制。在野外便携式气相色谱/质谱(GC/MS)中所使用的GC柱可仅由外柱环36的应用和尺寸限制。因此可能的是,为了改变分辨率和灵敏度,毛细管的ID和长度可以更大或更小。因此,毛细管的ID和长度可仅由内柱环32、中柱环34、外柱环36的尺寸限制,使毛细管能以单层的形式盘绕在中柱环上。此外,应当理解的是,在台式型GC柱中不存在这样的尺寸限制。
图4A是LTM加热器柱组装件50的透视分解图。在图4B中所示的组装的LTM加热器柱组装件50可包括内柱环32、中柱环34和如图4A中所示设置在之间的加热线38。加热线套管52可用于覆盖加热线38的暴露的引线。
图5A是LTM加热器柱组装件50、毛细管40、外柱环36、凸缘盖54和多个突出部42的透视分解图。将毛细管40围绕LTM加热器柱组装件50的外表面盘绕,然后将外柱环36围绕毛细管设置。
图5B示出了形成LTM GC模块30的组装后的组件以及独立的温度传感器60。完全组装的LTM GC模块30如图2中所示。温度传感器60能够感测例如从-70℃至500℃的大范围的温度。然而,这个温度范围应当仅被认为是示例,而不是对第一实施例的限制。
在图2至图5B中所示的第一实施例中存在对LTM GC模块30的按需要运行很重要的几个方面。
在图4B中所示的第一方面中,LTM加热器柱组装件50需要在中柱环34的外表面上提供连续的加热表面。中柱环34的外表面基本上是圆柱体的表面。
第一实施例的另一方面是,中柱环34的外表面需要基本上是平滑的,使得如果柔性物体围绕中柱环34缠绕,则该物体将始终与外表面接触,以在外表面上对物体提供均匀的加热。
第一实施例的另一方面是,围着内柱环32盘绕的加热线38需要以这样的方式盘绕,使得当中柱环34围着内柱环放置以将加热线封闭在内柱环32和中柱环34之间时,结果应当是中柱环34的外表面被均匀加热。这可以通过将加热线38围绕着内柱环32盘绕的任何允许出现均匀加热的方式来完成。例如,在内柱环32上可均匀地分布加热线38。均匀地分布可能需要相等的间隔或消除加热线38的绕组之间的所有间隔。
然而,内柱环34提供均匀加热的外表面可能是不够的。第一实施例的另一方面是,毛细管40必须围绕中柱环34缠绕,以这样的方式使围绕中柱环的所有毛细管被均匀加热。毛细管40的均匀加热可以通过缠绕毛细管来实现,使得毛细管的任意部分都不与任意其它部分重叠。因此,如果所有毛细管40都与中柱环34的外表面接触,那么冷点可以被最小化。因此,毛细管40很可能是单层布置。
第一实施例的另一方面是,温度传感器60可不必如图1中所示的现有技术一样沿着毛细管40的整个长度设置。因为LTM GC模块30的温度由于加热线38和毛细管40的布置可以是均匀的,所以温度传感器60可仅设置在LTM GC模块上的一个位置中。在这个示例中,温度传感器60可设置在内柱环32的内部。
图6A是本发明的第一实施例的温度传感器60的组件的透视分解图。温度传感器60可包括本领域的技术人员所熟知的电阻式温度检测器(RTD)传感器。然而,任意合适的温度传感器可在本发明的第一实施例中被使用。
温度传感器60可包括RTD传感器62、外壳64、卷边管(crimp tubing)66、两个连接线68和两个绝缘电线套管70。
图6B是在第一实施例中所使用的温度传感器60的组装后组件的透视图。
图7是已经设置在安装板70上的完全组装的LTM GC模块30的透视图。多个绝缘体柱72用于将LTM GC模块30固定在安装板70上。绝缘体柱72的数量可以变化,并且不应当被认为是对本发明的第一实施例的限制。连接器74联接至加热线38并联接至温度传感器60,并且可被用于向安装板70发送信号并从安装板70接收信号。
为了使LTM GC模块30可适用于野外便携式GC/MS单元,外柱环36的直径应保持为相对较小。使用示例中给出的尺寸的毛细管,外柱环36的直径可以是10cm或更小。
在第一实施例的概述中,教导了将加热系统用于最小化执行野外便携式气相色谱/质谱(GC/MS)测量所用的柱的毛细管中的冷点。这种加热系统的组件可包括内圆柱形环、围绕内圆柱形环设置的加热线、围绕加热线和内圆柱形环设置的中圆柱形环,其中加热线均匀地加热中圆柱形环,用作GC柱的毛细管围绕中圆柱形环的外表面设置,其中毛细管在中圆柱形环上自身不重叠,并且外圆柱形环围绕毛细管和中圆柱形环设置。
加热系统还可包括温度传感器,温度传感器设置在内圆柱形环上,从而能够读取内圆柱形环的温度,并且因此能够读取LTM GC柱30的温度。为了使系统能用于野外便携式GC/MS测量系统中,外圆柱形环的直径可小于10厘米。
本文献描述了野外便携式GC/MS(Torion T-9,PerkinElmer公司,Shelton,CT)对各种样品(气体、液体、固体)的分析结果,包括对高沸点、半挥发性的有机化合物(SVOC)的分析,分析时间通常不超过10分钟。研究的半挥发性化合物包括:分离沸点范围从萘(218℃)至苯并苝(550℃)的多环芳烃(PAH)的混合物;表征沸点范围从2,6-二氯-4-硝基苯胺(dichlroan)(130℃)至溴氰菊酯(572℃)的一组有机氯农药;量化沸点为155℃-177℃的萜烯混合物;检测例如沸点为270℃、为环境水样中细菌活动的副产物的土臭素的天然化合物;分析沥青和煤焦油基砾石样品中的PAH;并筛选水中的酚类化合物和邻苯二甲酸酯,酚类化合物和邻苯二甲酸酯用于制造许多塑料组件。
为了更好地理解第一实施例的实际功能,有必要简要概述其功能。尽管该技术是为便携性和速度而设计的,但气相色谱仪的设计旨在提供等同于台式系统的色谱分辨率和性能。通过将使用直接接触电阻加热的用低热质(LTM)GC柱束替换常规毛细管柱和对流炉实现了微型尺寸。在该设计中,小直径金属毛细管柱可以与电阻加热线和温度感测线捆在一起,该电阻加热线和该温度感测线可以与绝缘体线编织在一起。该方法可以提供更加可控的加热、更大的加热和冷却速度以及非常低的能耗。由于柱式加热比常规的GC所需的操作功率要小得多,因此可延长电池的寿命。第一实施例结合了直接电阻加热和快速温度升温速率,可以在几分钟内分离多组分的分析物。
质谱仪可使用环形离子阱配置,与例如常规圆柱形离子阱或线性四极阱的其它类型的质谱仪相比,这种配置非常适合于小型化。尽管新配置的尺寸小,但第一实施例的新配置可允许大的捕获量。结果可能是离子计数大、灵敏度提高、噪音水平低、光谱质量好。可以将离子阱质量分析器加热至约175℃并在真空中操作,这可导致电极长时间保持干净。这可减少频繁维护的需要,同时提高质谱质量和质谱再现性。在高温下运行也可导致长期的MS分辨率稳定性,提供超过45-500amu质量范围的单位质量分辨率。
样品制备站还可在野外提供不同模块的功能,包括样品解吸(SD)、加热顶空(HS)、吹扫和捕集(PT)以及内标(IS)添加模块。样品制备站可以很容易地配置用于在采样位置处样品制备和分析的具体应用要求。样品制备站允许将在常规尺寸的收集阱上收集的空气样品转移到微阱中,以便注入微孔毛细管GC。在该解吸过程中,分析物从常规的捕集阱转移到仪器的针头捕集阱中,以便注入GC-MS。完整的模块可以通过实验室气体供应(氦气或氮气)和线路电源来操作,也可以通过电池电源和内部加压气体罐来操作。
此外,使用板载库不仅可以识别未知的目标化合物,还可允许用户自定义地构建目标化合物库。该特性得到去卷积算法的支持,以确保在复杂混合物中可靠地识别均匀共洗脱化合物(even co-eluting compounds),并且与大量的NIST(National Institute ofStandards and Technology)数据库结合使用;可以很容易地辨识未知峰。
为了更好地理解用于分离和分析半挥发性化合物的第一实施例的实际功能,对沸点范围为200℃-570℃的一组半挥发性有机化合物进行了现有技术的LTM GC柱和第一实施例的LTM GC柱之间的比较。
对半挥发性化合物的三种混合物进行评估,包括18种PAH化合物、17种有机氯农药化合物和10种拟除虫菊酯农药化合物。通过无溶剂线圈丝(coil wire filament,CWF)注射系统将相同量的每个样品导入三个不同的系统,并使用与表1中所示完全相同的GC色谱分离条件和质谱仪操作条件来运行。

每种混合物的比较色谱图可见于图8中(18种PAH化合物)、图9(17种有机氯农药)和图10(10种拟除虫菊酯农药)。标准LTM柱色谱图以红色显示,而新LTM色谱柱的版本是黑色的。
从图8、图9和图10中可以得出一些观察结果。首先,在现有技术的LTM GC柱中,峰形状和分辨率显得更差,因为现有技术的LTM GC柱产生的峰更宽,背景更嘈杂。其次,现有技术的LTM GC柱中的峰的保持时间基本上更长。而且,第一实施例的LTM GC柱中的峰强度明显高于常规LTM GC柱中的峰强度。最后,标准LTM GC柱无法分离并洗脱PAH混合物中的组分16、组分17和组分18,因为它们的挥发性非常低(沸点524℃-550℃)。
现在提供沸点范围大的一组不同半挥发性有机化合物的分析。
萜烯是由包括针叶树、啤酒花和大麻的多种植物生成的一大类有机化合物,典型的沸点范围为150℃-180℃。它们是许多种类的植物和花的精油的主要成分,广泛用作香水中的香料以及用于医药目的。天然萜烯的合成变体和衍生物也用于作为食品添加剂而使用的各种香味和香料。因此,为了举例说明该技术的可行性,将四种萜烯化合物掺入200mL的0.6%NaCl的水溶液中。然后,使用一半/一半固相微萃取(SPME)聚二甲基硅氧烷/二乙烯基苯(PDMS/DVB)65pm纤维在室温(22℃)下提取分析物15分钟,不进行摇动或振动。用这种取样方法,纤维的一半放置在液面上空间,而一半浸入样品的液相中,如图11所示。
在注入GC/MS之前,将四种萜烯分析物在室温(22℃)下用一半/一半SPME(PDMS/DVB 65μm纤维)提取15分钟。
然后使用早前所述的相似条件将该样品注入GC/MS系统,不同的是,280℃的最终温度保持了50秒,使总分析时间为175秒。四种萜烯(α+蒎烯、月桂烯、α+柠檬烯和异长叶烯)的总离子色谱图(TIC)如图12所示。
为四种萜烯化合物生成了四点校准图。标准品的浓度和使用相关系数(R2)的相应校准曲线在图13中示出。应该注意的是,这四种化合物的估计检出限为20ppt,这基于最低标准(样本1)的多次重复实验的统计分析。
土臭素是由多种微生物和细菌产生的有机化合物。土臭素具有独特的泥土味道和芳香,是甜的泥土味道和在天气干燥一段时间后雨水下落时出现在空气中的强烈气味的原因。土臭素是由包括蓝细菌和放线菌的几种微生物产生的,并且在这些微生物死亡时释放出来。当这些细菌的数量急剧下降而将土臭素释放到当地的供水(9)中时,用水依赖于地表水的社区可能会定期经历水难喝的时期。在化学上,土臭素是一种双环醇,分子式为C12H22O,并且是通常称为十氢化萘的十氢萘的衍生物。土臭素的沸点约为270℃。
该方法包括将20ppt的土臭素掺入500mL水样品中。在没有任何预处理步骤的情况下,然后由真空泵以25-35mL/min的流速输送,在常温下将其捕获在填充于去活的不锈钢固相萃取(SPE)解吸管中的聚二甲基硅氧烷(PDMS)颗粒(125μm-180μm大小)上。然后使用仪器的热解吸系统将目标分析物转移到PDMS针头捕集阱中。解吸步骤使用He(氦)载气在200℃下以6mL/min进行10分钟。使用针头捕集阱将样品导入GC-TMS的过程在270℃下进行60秒。样品输送方法的示意图在图14中示出。
色谱分离条件在表3中示出。在图15中示出分离的总离子色谱图(TIC)和提取离子色谱图(RIC),这两幅图示出了土臭素的母分子离子和相关的片段,由在其下面的NIST参考质谱图确定。
图16示出了证明使用仪器的去卷积算法很好地分离20ppt土臭素的去卷积色谱图和质谱图。基于土臭素校准的统计分析,估计检出限为个位数ppt水平。

道路和停车场表面通常由含有大量碳质化合物的沥青和/或煤焦油产品形成。因此,了解路面制备过程中所使用的砾石样品中PAH含量的成分非常重要。为了该研究,将40g砾石样品掺入储备标准溶液,以制备0.05ppm、0.25ppm、0.5ppm和1.0ppm PAH分析物的校准标准品。然后,通过手摇动约2-3min,用二氯甲烷(5mL)和水(约15mL)的混合物萃取样品。然后,将液相转移到另一个小瓶中以使两相分离开。对于一些样品,需要预浓缩以改善检测。这是通过将1mL有机相置于2mL小瓶中并使溶剂蒸发以获得适合测量的体积来实现的。然后,使用注射器将有机相的中的20μL等份试样样品导入玻璃管中,并使用真空泵或空气压缩机去除溶剂。然后,使用样品置换法将目标分析物转移到PDMS针头捕集阱中,在300℃下进行5分钟,吹扫流速为30ml/min。用于分离的GC的条件在表4中示出,而分离的总离子色谱图见图17,图17清楚地示出了诸如苯并苝和苯并荧蒽的高分子量、高沸点PAH已被分离并检测到。

第一实施例的便携式GC/MS技术还可以使用微液体萃取(MLE)和线圈线(CWF)用作水中SVOC的一般筛选工具。实验是在掺入有低ppb-sub ppm水平浓度的SVOC的自来水中进行的。使用少量(0.2mL-0.5mL)的例如二氯甲烷、己烷、戊烷或丙酮等合适的溶剂进行萃取。在使用0.5%-3%的NaCl时可施加手动摇动和盐析以加速萃取过程。萃取过程进行几分钟,然后,将含有分析物的溶剂施加到线圈上,或者如果需要,在转移到小瓶中后通过使溶剂蒸发来浓缩。在线圈上的溶剂蒸发后,使用线圈进行样品导入。筛选试验使用PAH、酚类化合物、邻苯二甲酸酯、有机氯化物、有机磷和拟除虫菊酯类农药和除草剂的混合物进行。然而,如前所示出的,PAH和各种农药的分离仅显示酚类化合物和邻苯二甲酸酯的代表性数据。酚类和邻苯二甲酸酯的色谱分离条件如表5所示。表5示出了用于筛选9种酚类化合物、6种邻苯二甲酸酯的色谱分离条件。使用类似条件筛选其它14种一般农药、6种除草剂和10种杀虫剂。

这些苯酚和邻苯二甲酸酯的筛选试验的总运行时间小于3分钟。在两种类型的样品上都发生了一定程度的离子分子化学反应,因此通过使用NIST库搜索功能证实了绝对识别。虽然这些分离的峰容量相对较低,但是去卷积算法有助于更准确地分离和识别分析物。实际样品中的动态范围和检出限将在未来的研究中确定并呈现。
在偏远的野外场所的恶劣条件下,对空气、水和固体基质样品中痕量的挥发性有机化合物和半挥发性有机化合物的分析的需求越来越大。该研究证明的是,利用便携式GC-MS结合快速样品制备/导入技术,现在可以实现实验室级的性能。这种结合能够进行用于定量和定性筛选目的的各种基于环境的分析,可以为现场非技术操作人员和缺乏经验的操作人员提供快速、可操作的数据。

Claims (16)
1.一种加热系统,用于使执行野外便携式气相色谱/质谱(GC/MS)测量所使用的柱的毛细管中的冷点最小化,所述加热系统包括:
内圆柱形环;
加热线,围绕所述内圆柱形环设置;
中圆柱形环,围绕所述加热线和所述内圆柱形环设置,其中所述加热线均匀地加热所述中圆柱形环;
毛细管,用作GC柱并围绕所述中圆柱形环的外表面设置,其中所述毛细管在所述中圆柱形环上自身不重叠;以及
外圆柱形环,围绕所述毛细管和所述中圆柱形环设置。
2.根据权利要求1所述的系统,其中所述系统进一步包括温度传感器,所述温度传感器设置在所述内圆柱形环上,从而能够读取所述内圆柱形环的温度。
3.根据权利要求2所述的系统,其中所述外圆柱形环的直径小于10cm以使所述加热系统能用于野外便携式GC/MS测量系统中。
4.根据权利要求3所述的系统,其中所述系统进一步包括电源,所述电源联接到所述加热线用于提供电力以能够加热所述加热线。
5.根据权利要求4所述的系统,其中所述系统进一步包括联接到所述毛细管的野外便携式GC/MS单元。
6.一种用于使执行野外便携式气相色谱/质谱(GC/MS)测量所使用的毛细管柱中的冷点最小化的方法,所述方法包括:
提供内圆柱形环、围绕所述内圆柱形环设置的加热线、围绕所述加热线和所述内圆柱形环设置的中圆柱形环、用作GC柱并围绕所述中圆柱形环的外表面设置的毛细管、以及围绕所述毛细管和所述中圆柱形环设置的外圆柱形环;并且
通过1)将所述加热线围绕所述内圆柱形环均匀分布来均匀地加热所述中圆柱形环、以及2)在将所述毛细管围绕所述中圆柱形环盘绕时不使所述毛细管的任意部分重叠来防止所述毛细管中的冷点,并且因此使所有的毛细管暴露于来自所述中圆柱形环的相同量的热量。
7.根据权利要求6所述的方法,其中所述方法进一步包括将温度传感器设置在所述内圆柱形环上,从而能够读取所述内圆柱形环的温度。
8.根据权利要求7所述的方法,其中所述方法进一步包括使所述外圆柱形环的直径小于10cm以使得所述毛细管能用于野外便携式GC/MS测量系统中。
9.根据权利要求8所述的方法,其中所述方法进一步包括将电源联接到所述加热线从而向所述加热线提供电力。
10.根据权利要求9所述的方法,其中所述方法进一步包括将野外便携式GC/MS单元联接至所述毛细管。
11.根据权利要求10所述的方法,其中所述方法进一步包括执行高温沸腾半挥发性有机化合物的取样和测量。
12.一种用于在野外便携式GC/MS仪器中执行高温沸腾半挥发性有机化合物的取样和测量的方法,所述方法包括:
提供内圆柱形环、围绕所述内圆柱形环设置的加热线、围绕所述加热线和所述内圆柱形环设置的中圆柱形环、用作GC柱并围绕所述中圆柱形环的外表面设置的毛细管、以及围绕所述毛细管和所述中圆柱形环设置的外圆柱形环,其中,所述毛细管被盘绕成单层;
通过将所述加热线围绕所述内圆柱形环均匀分布来均匀地加热所述中圆柱形环;并且
执行高温沸腾半挥发性有机化合物的取样和测量。
13.一种系统,用于在野外便携式GC/MS仪器中执行高温沸腾半挥发性有机化合物的取样和测量,所述系统包括:
内圆柱形环;
加热线,围绕所述内圆柱形环设置;
中圆柱形环,围绕所述加热线和所述内圆柱形环设置,其中所述加热线均匀地加热所述中圆柱形环;
毛细管,用作GC柱并围绕所述中圆柱形环的外表面设置,其中所述毛细管在所述中圆柱形环上自身不重叠;和
外圆柱形环,围绕所述毛细管和所述中圆柱形环设置;以及
野外便携式GC/MS仪器,联接到所述毛细管用于执行高温沸腾半挥发性有机化合物的取样和测量。
14.根据权利要求13所述的系统,其中所述系统进一步包括温度传感器,所述温度传感器设置在所述内圆柱形环上,从而能够读取所述内圆柱形环的温度。
15.根据权利要求14所述的系统,其中所述外圆柱形环的直径小于10cm以使所述加热系统能用于野外便携式GC/MS测量系统中。
16.根据权利要求15所述的系统,其中所述系统进一步电源,所述电源联接到所述加热线用于提供电力以能够加热所述加热线。
