背景技术
肝脏是机体发挥代谢功能、储存肝糖、合成分泌性蛋白质和生物转化等多种生物学功能的重要器官,也是机体最大的解毒器官,在机体新陈代谢过程中所产生的一些氧自由基等有害废物及外来的毒物、毒素、药物的代谢和分解产物,均在肝脏中进行分解、氧化转变为无毒害物质后排出体外。另外肝脏还是机体最大的腺体,它在机体的胆汁生成、凝血、免疫、热量产生及水与电解质的调节中均起着非常重要的作用,可以说是机体内一个巨大的“化工厂”。
随着我国养殖业向集约化、规模化方向发展,畜禽的疾病发病率逐年上升,并且新的疾病不断增加,畜牧从业者被迫在畜禽养殖过程中长期大量应用抗生素。抗生素的不合理使用,易产生耐药性和药物残留,造成畜禽肝脏的不可逆损伤,并丧失部分解毒功能。同时,肝脏作为非常重要的屏障机构,易受饲料和饲料原料中的霉菌毒素、重金属、药物添加剂、细菌、病毒、毒素、营养代谢病等各种致病因素的侵害,引起肝细胞的代谢障碍,导致肝脏变性、坏死、出血等一系列的病理变化。上述各种因素导致的畜禽肝中毒,在临床中表现为:肝细胞损伤,消化酶分泌功能降低,消化功能下降,饲料转化率低;肝脏解毒功能低下,毒素中毒现象增多,体内毒素增加,加重肾脏负担,出现肾衰,生产性能下降,大量死亡;部分治疗性药物在肝脏内转化低下,药效下降;易出现脂肪肝、肝肿大、肝出血等现象。
畜禽肝中毒的病因多,发病率高且难治疗,严重损害畜禽的健康,不仅造成重大经济损失,而且威胁食品安全,危害人体健康。因此研制安全、毒副作用小,不易产生耐药性、疗效高且成本低的药物来治疗畜禽肝中毒,营养肝脏、减缓肝脏损伤、提高肝脏的解毒功能是当前养殖业急待解决的难题,也是畜牧水产业的研究热点领域。

申请公布号为CN108094757A的发明专利,公开了一种用于水产养殖的保肝产品及其制备工艺,该产品为5~10份葡萄糖,5~10胆汁酸,5~10份牛磺酸,其他辅料1~5份,70~80份膨润土,在V型混合机中混合制备而成。该产品能改善水产动物的肝功能,增强水产动物的抗病能力,促进水产动物的生长。
从上述公开的专利以及其他科技文献检索结果来看,动保专家在不断的研制有效的保肝药物,并获得了效果不错的药物制剂。然而目前市场上治疗畜禽肝病的药物大多以采用葡萄糖、牛磺酸、Vc、葡醛内酯等排毒解毒的化学成分进行辅助性的保健为主;也有部分以名贵中药材人参、水飞蓟、葛根等成分进行治疗为主;或者是采用大配方以十几种甚至几十种中草药相混合,这种重复多余的用药方法,既浪费中药资源,又导致使用成本高昂。尚未发现有对抗多种原因引起的肝脏损伤,显著减轻肝脏变性和坏死程度,恢复肝脏代谢功能,且疗效确切、经济实用,能供临床大面积应用的治疗畜禽肝中毒的中药制剂。
发明内容
本发明针对目前市场上治疗畜禽肝病的药物所存在的不足,提供一种治疗畜禽肝中毒的中药组合物及其制备方法和制剂。
本发明解决上述技术问题的技术方案如下:
一种治疗畜禽肝中毒的中药组合物,按重量份计,包括如下组分:骆驼刺10~45份,肉苁蓉5~25份,锁阳1~20份,甘草10~40份,蒙脱石40~80份。
进一步,按重量份数计,包括如下组分:骆驼刺25份,肉苁蓉15份,锁阳10份,甘草20份,蒙脱石60份。
本发明的中药组合物中使用的各个中药成分的特性如下:
骆驼刺:新疆地区特有的一种豆科落叶灌木,能分泌糖类物质,干燥后收集的刺糖可直接食用,也可煮水,是用于治疗肠胃不适、腹痛、腹胀、腹泻、咳嗽下血、口舌生疮及滋补强壮、平衡体液和异常胆液质的民间用药。从骆驼刺中分离出来的一些化合物,在抗氧化、抗过敏、抗肿瘤等方面具有良好的表现,其全新化合物的抗癌活性为首次发现。
肉苁蓉:肉苁蓉是一种寄生在梭梭、红柳根部的寄生植物,别称不老草。性味甘、咸,温。归肾、大肠经。补肾阳,益精血,润肠通便。主治滑精,阳痿,垂缕不收,宫寒不孕,腰胯疼痛,肠燥便秘。现代药理研究表明,它可以调整内分泌、促进代谢,对阳虚和阴虚的动物肝脾核酸含量下降和升高有调节作用,并可激活肾上腺释放皮质激素,辅助治疗肿瘤、抗辐射等,被誉为“沙漠人参”。
锁阳:多年生肉质寄生草本,性味甘,温,归肝、肾、大肠经。平肝补肾,益精养血,润肠通便。主治阳痿遗精、腰膝酸软、肠燥便秘,是补肾壮阳气药材中最常使用的一味药。现代研究表明,它可增强免疫功能,清除自由基,抗血小板聚集,具有糖皮质激素样作用,补充维生素和矿物质。
甘草:性味甘,平。入心、肺、脾、胃经。补脾益气,祛痰止咳,和中缓急,解毒,调和诸药,缓解药物毒性、烈性。主治脾胃虚弱,倦怠无力,咳喘,咽喉肿痛,中毒,疮疡。炙用于脾胃虚弱之腹胀纳呆,乏力,能缓解腹中拘挛性疼痛,生用缓和药物的烈性和毒性,治痈疮肿毒,药物、食物中毒。现代药理学证明甘草具有抗微生物、抗炎、镇咳祛痰、抗肿瘤、抗突变、抗氧化等功效,对消化系统、心血管系统、生殖系统及脑、肾功能等具有调节作用。
蒙脱石:含水硅铝酸盐组成的层状矿物,其结构层带负电荷,具有高比表面积和强吸附/脱附力,吸附量大、稳定性高、价格低廉、无毒副作用等特性。蒙脱石在医药中应用广泛,可以做医药载体,起控释剂功效;作为饲料辅助添加剂,凭借其自然性状和复合功能,已广泛应用于家畜家禽的饲养。其独特的离子交换性和抗毒性,在供给畜禽及水产动物生长发育所需的多种常量和微量元素的同时,还吸附霉菌毒素及内毒素、中和毒性基团。它不仅可补偿动物养分,提高畜禽生产性能,而且能够吸附消化道内有毒有害的代谢产物,阻止其在体内聚积,对某些重金属具有专门吸附作用,消除、减轻或者抑制病原微生物和重金属等有毒有害物质对动物的毒害作用。
本发明中药组合物的药理作用及配伍原理如下:
中药组合物的配伍原则是“增效减毒”,肉苁蓉和锁阳的功效相仿,同时配伍使用能够相互配合,双向调节,利用肉苁蓉补肾阳、益精血和锁阳平肝补肾、益精养血,从内部对肝肾进行调节,保证机体的气血充足,同时提高小肠推进度,缩短通便时间,并明显抑制大肠对水分的吸收,润肠通便,提高机体的排毒和营养的吸收利用。甘草调和诸药解百毒,对抗多种原因引起的肝脏损伤,减轻肝细胞坏死,促进肝细胞再生,恢复肝脏代谢功能。蒙脱石具有良好的胃肠道黏膜亲和性和控释性能,作为控释载体,更利于肉苁蓉、锁阳和甘草活性成分全肠道控制释放、吸收,有效发挥作用。同时,蒙脱石对各种有毒物及毒素有优良的吸附能力,在消化道内可形成胶体保护膜,修复消化道损伤,提高营养吸收能力。上述各药相互配伍,全方得以标本兼治、扶正祛邪,全面调整、平衡阴阳,共同达到清除体内毒素,解毒保肝的作用,且调整体内气血运行,有效对抗多种原因引起的肝脏损伤,显著减轻肝脏变性和坏死程度,快速缓解各种肝中毒症状,恢复食欲及生产性能,降低死淘率,提高养殖经济效益。

2)本发明的中药组合物使用方便,人畜无害,治愈率高,药物无残留,无毒副作用;
3)本发明的中药组合物采用西北地区资源丰富、品质优异的纯中药制成,来源广泛、成本低廉,经济效益高,适于大规模生产应用。
本发明还要求保护上述中药组合物的制备方法,包括如下步骤:
1)取骆驼刺、肉苁蓉和锁阳冲洗干净,浸泡润湿后切制饮片,烘干后研磨粉碎,向其中加入5~10倍重量的乙醇于70~80℃下回流提取2~3次,每次2~3小时,合并提取液后减压浓缩,喷雾干燥得到骆驼刺、肉苁蓉和锁阳的提取物;
2)取甘草冲洗干净,浸泡润湿后切制饮片,烘干后研磨粉碎,向其中加入4~8倍重量的水于100~120℃下煎煮提取3~4次,每次2~3小时,合并提取液后减压浓缩,喷雾干燥得到甘草提取物;
3)将步骤1)所得的骆驼刺、肉苁蓉和锁阳的提取物与步骤2)所得的甘草提取物合并混合均匀,然后向其中加入蒙脱石粉,搅拌混合均匀后即得中药组合物。
进一步,所述乙醇的浓度为60~70wt%。
本发明提供的中药组合物的制备方法的有益效果是:
采用本发明的提取技术可充分析出中草药中的活性成分,解决了传统中药散剂中粗纤维含量高、有效成分难以溶出的不足,更易于畜禽吸收,生物利用度高。
本发明的中药组合物可与药学上可接受的载体制成中药制剂,中药制剂可制成粉剂、散剂、颗粒剂、片剂、胶囊剂等剂型。
具体实施方式
以下结合实例对本发明的原理和特征进行描述,所举实例只用于解释本发明,并非用于限定本发明的范围。
实施例1:
一种治疗畜禽肝中毒的中药组合物,按重量份计,包括如下组分:骆驼刺15份,肉苁蓉10份,锁阳5份,甘草15份,蒙脱石55份。
实施例2:
一种治疗畜禽肝中毒的中药组合物,按重量份计,包括如下组分:骆驼刺25份,肉苁蓉15份,锁阳10份,甘草20份,蒙脱石60份。
实施例3:
一种治疗畜禽肝中毒的中药组合物,按重量份计,包括如下组分:骆驼刺35份,肉苁蓉20份,锁阳15份,甘草30份,蒙脱石75份。
实施例4:
一种治疗畜禽肝中毒的中药组合物,按重量份计,包括如下组分:骆驼刺45份,肉苁蓉25份,锁阳20份,甘草40份,蒙脱石80份。
实施例5:
一种治疗畜禽肝中毒的中药组合物,按重量份计,包括如下组分:骆驼刺10份,肉苁蓉5份,锁阳20份,甘草10份,蒙脱石40份。
实施例6:
一种治疗畜禽肝中毒的中药组合物,按重量份计,包括如下组分:骆驼刺10份,肉苁蓉25份,锁阳1份,甘草40份,蒙脱石80份。
实施例7:
一种治疗畜禽肝中毒的中药组合物,按重量份计,包括如下组分:骆驼刺45份,肉苁蓉25份,锁阳1份,甘草10份,蒙脱石40份。
实施例8:
一种治疗畜禽肝中毒的中药组合物,按重量份计,包括如下组分:骆驼刺45份,肉苁蓉5份,锁阳20份,甘草40份,蒙脱石80份。
实施例1~8的中药组合物的制备方法如下:
1)取骆驼刺、肉苁蓉和锁阳冲洗干净,浸泡润湿后切制饮片,烘干后研磨粉碎成20目的细粉,向其中加入5~10倍重量的乙醇于70~80℃下回流提取2~3次,每次2~3小时,合并提取液后减压浓缩,喷雾干燥成粒径100~200目的粉末,得到骆驼刺、肉苁蓉和锁阳的提取物;
2)取甘草冲洗干净,浸泡润湿后切制饮片,烘干后研磨粉碎成20目的细粉,向其中加入4~8倍重量的水于100~120℃下煎煮提取3~4次,每次2~3小时,合并提取液后减压浓缩,喷雾干燥成粒径100~200目的粉末,得到甘草提取物;
3)将步骤1)所得的骆驼刺、肉苁蓉和锁阳的提取物与步骤2)所得的甘草提取物合并混合均匀,然后向其中加入蒙脱石粉,搅拌混合均匀后即得中药组合物。
将本发明的中药组合物进行了实验一~实验四的效果测试实验,测试过程及测试结果如下。
实验一
实验动物:
山东潍坊某养殖场,27日龄艾维茵肉鸡,根据临床症状、病理剖检综合诊断为化学药物性肝中毒,随机选取3500只,分为7组,每组500只,分别采用实施例1~3制备的组合物及对照药物(市售肝泰乐,主要成分为葡醛内酯,批号20180316,湖北隆飞生物科技有限公司生产)进行治疗,并设置空白对照组,观察疗效并统计治愈、有效及无效数,计算有效率,结果详见表1。
实验方法:
实施例1、3制备的组合物及对照药物均采用1%量拌料混饲,连用7日;实施例2制备的组合物分别以0.5%(低剂量组)、1%(中剂量组)、1.5%(高剂量组)拌料混饲,连用7日;空白对照组不添加任何药物。
疗效判定标准:痊愈:用药5~7日后,精神食欲恢复正常;有效:用药5~7日后,精神好转,食欲增加;无效:用药5~7日后,临床病症同治疗前,死亡数甚至增加。

结论:考察本发明的中药组合物对鸡肝中毒的治疗效果。统计分析表明,实施例1~3制备的组合物以1%拌料混饲及对照组肝泰乐对鸡肝中毒的作用,与空白对照组比较,均具有显著治疗作用(P<0.01),且有临床使用意义。实施例1~3与对照组肝泰乐比较,均具有显著治疗作用(P<0.05)。另外,实施例1~3以1%拌料时,实施例2的治愈率和总有效率显著高于实施例1和实施例3(P<0.01),而实施例2的中剂量组和高剂量组的治愈率和总有效率则无显著差异。本发明优选配比量为:骆驼刺25份、肉苁蓉15份、锁阳10份、甘草20份、蒙脱石60份。
实验二
实验材料:
本发明实施例2制备的中药组合物。
实验动物:
AB品系斑马鱼,由烟台大学生命科学院惠赠。
E3培养液(mmol/L):KCl 0.17,NaCl 5.0,MgSO40.33,CaCl20.33,pH 7.4。黄曲霉毒素B1(AFB1)购自上海阿拉丁生化科技股份有限公司。其他试剂均为国产分析纯。
实验方法:
将实施例2制备的中药组合物按低、中、高剂量10、20和40g/L的浓度,AFB1按0.625μmol/L的浓度,用E3培养液稀释备用。收集受精后6h的斑马鱼胚胎置于6孔细胞培养板中,30枚/孔,每孔加入5ml含有不同浓度受试物的E3溶液,另设不含受试物的E3溶液作为空白对照组,每个处理组设3个平行复孔。随后置于28.5℃恒温光照培养箱内,让胚胎继续发育。每24h更换含受试物E3培养液,观察胚胎发育状况,以卵凝结或心脏停跳作为胚胎死亡终点,移除并记数死亡胚胎数量至96h。计算胚胎死亡率:胚胎死亡率(%)=(死亡胚胎数量/总受试胚胎数量)×100。
采用SPSS17.0软件以One-wayANOVA方法进行组间均值差异显著性分析,若P<0.01表示差异水平极显著,若P<0.05表明差异水平显著。采用Pearson相关系数分析实验结果的相关性。实验数据均以表示。
实验结果:
直接将斑马鱼胚胎暴露于含不同浓度组合物,及0.625μmol/L浓度AFB1的E3培养液,观察至96h,结果如表2所示。结果表明,选用低、中、高剂量组合物与AFB1,24h-48h各实验组斑马鱼胚胎发育正常,72h时AFB1单独处理组斑马鱼胚胎出现死亡现象,死亡率42.39%,极显著高于其他四组(P<0.01)。96h时AFB1单独处理组斑马鱼胚胎全部死亡,低、中、高剂量组合物联合AFB1处理组斑马鱼胚胎出现死亡现象,死亡率分别为74.45%、28.31%、23.33%,极显著低于单独处理组(P<0.01)。用双变量的比较方法对实验数据进行Pearson相关系数分析比较,结果发现组合物浓度与斑马鱼胚胎死亡率Pearson相关系数r=-0.881,呈显著负相关(P<0.05)。结论:采用新兴模式生物斑马鱼,在动物发育水平考察了本发明组合物对黄曲霉素所致急性毒性的干预作用。证实本发明组合物对黄曲霉毒素B1所致的胚胎死亡有明显抑制作用。提示本发明组合物能明显降低黄曲霉素发育毒性,缓解或解除动物中毒症状,提高畜禽经济性能。

实验三
实验材料:
本发明实施例2制备的中药组合物。阳性对照药物为谷胱甘肽片,批号20171023,重庆药友制药有限责任公司生产。其他试剂均为国产分析纯。
实验动物:
昆明种小鼠,体重20±2g,雌雄各半,由山东省实验动物中心提供,合格证号SYXK(鲁)20140010。
实验方法:
取雌雄各半小鼠60只,随机均分为4组,即正常对照组,CCl4中毒组(0.1%CCl410ml/kg),阳性对照组(100mg谷胱甘肽/kg),实验药物组(100mg本发明组合物/kg),每日分别灌胃给药1次,正常对照组、中毒组给予等体积蒸馏水,连续7日,末次给药后除正常对照组外均腹腔注射0.1%CCl410ml/kg,18h后采血,分离血清用赖氏法测定谷丙转氨酶(SGPT)、谷草转氨酶(SGOT)值。采血后处死小鼠剖取肝脏,用15%福尔马林固定后,石蜡包埋,常规切片,HE染色,光镜观察脏器病理损害。
实验结果:
结果(见表3)表明,本发明组合物对CCl4引起的SGPT和SGOT增高有明显的抑制作用。

光镜下观察发现,CCl4中毒组小鼠肝组织显示从中央静脉为中心的肝细胞浑浊变性和坏死,并有明显的炎性细胞浸润,而阳性对照组(谷胱甘肽)和实验药物组(本发明组合物)病理改变明显轻于中毒组,结果见表4。
结论:CCl4是研究肝功能损害的常用病理模型,肝功能受损时,许多酶系统都会发生改变,重要的有血清谷丙转氨酶、谷草转氨酶等。本实验结果表明,本发明的中药组合物能明显抑制CCl4所致的小鼠肝损伤的病理改变,减轻肝细胞变性和坏死,同时明显地抑制CCl4引起的SGPT和SGOT增高,提示本发明组合物可对抗有毒物质引起的畜禽肝脏损伤,显著减轻肝脏变性和坏死程度,恢复肝脏代谢功能,具有显著的养肝护肝,分解肝毒素,恢复肝功能,提高生产性能的作用。

实验四
实验材料:
本发明实施例2制备的中药组合物,对照组采用市售Vc+葡萄糖;
实验动物:
河南洛阳某养猪场,2018年7月,三元保育仔猪发病,患猪主要症状为食欲降低甚至废绝,先便秘后腹泻下痢,泪斑严重,皮肤充血,外阴或肛门红肿。部分个体体重较大的患猪在出现症状后3~5d内死亡。综合剖检病理变化及饲养情况等,判断为霉菌毒素性肝中毒。
实验方法:
将保育仔猪随机分为2组,每组169头,A组使用本发明实施例2的中药组合物,B组使用市售Vc+葡萄糖,各组均全天按照1%比例拌料混饲,连续用药10天,记录仔猪日采食量、日增重、料肉比、腹泻率。结果见表5。

同列数据肩标不同字母表示差异显著(P<0.05);肩标相同字母表示差异不显著(P>0.05)。结论:表5中的实验结果表明,本发明的中药组合物对仔猪的霉菌毒素肝中毒具有显著治疗作用,对提高仔猪采食量、日增重,降低料肉比、腹泻率的效果均优于同类产品组。
以上所述仅为本发明的较佳实施例,并不用以限制本发明,凡在本发明的精神和原则之内,所作的任何修改、等同替换、改进等,均应包含在本发明的保护范围之内。
Claims (3)
1.一种治疗畜禽肝中毒的中药组合物,其特征在于,按重量份数计,由如下组分组成:骆驼刺25份,肉苁蓉15份,锁阳10份,甘草20份,蒙脱石60份;
所述的中药组合物的制备方法,包括如下步骤:
1)取骆驼刺、肉苁蓉和锁阳冲洗干净,浸泡润湿后切制饮片,烘干后研磨粉碎,向其中加入5~10倍重量的乙醇于70~80℃下回流提取2~3次,每次2~3小时,合并提取液后减压浓缩,喷雾干燥得到骆驼刺、肉苁蓉和锁阳的提取物;所述乙醇的浓度为60~70wt%;
2)取甘草冲洗干净,浸泡润湿后切制饮片,烘干后研磨粉碎,向其中加入4~8倍重量的水于100~120℃下煎煮提取3~4次,每次2~3小时,合并提取液后减压浓缩,喷雾干燥得到甘草提取物;
3)将步骤1)所得的骆驼刺、肉苁蓉和锁阳的提取物与步骤2)所得的甘草提取物合并混合均匀,然后向其中加入蒙脱石粉,搅拌混合均匀后即得中药组合物。
2.一种治疗畜禽肝中毒的中药制剂,其特征在于,包括权利要求1所述的中药组合物及药学上可接受的载体。
3.根据权利要求2所述的中药制剂,其特征在于,所述中药制剂的剂型为粉剂、散剂、颗粒剂、片剂或胶囊剂。
