Cutting potato into small pieces, cutting live silkworm pupa into pieces, adding corn flour, wheat bran and a proper amount of water, boiling for 30min, filtering, taking filtrate, adding other components, subpackaging, sterilizing, cooling, inoculating slant strain on a shaking table for culturing at 17-19 ℃ and 130rpm for 16 days.
Preparing seeds in a seeding tank: the culture medium comprises 1.5% of glucose, 1.0% of molasses, 2.0% of corn flour, 3.5% of silkworm chrysalis powder, 0.5% of soybean cake powder, 0.5% of yeast powder, 0.2% of peptone, 1.5% of bran, 0.01% of magnesium sulfate, 0.02% of potassium dihydrogen phosphate and the balance of water. Firstly, extracting silkworm chrysalis meal, corn flour, wheat bran, soybean sprout powder and a proper amount of water by slight boiling for 30min, filtering, taking filtrate, adding other components, sterilizing and cooling, inoculating shake flask seeds with 10% of inoculum size at the temperature of 17-19 ℃, stirring at the rotation speed of 100 plus 120rpm, and ventilating for 1: 0.2-0.4vvm, and culturing for 8 days.
Culturing in a fermentation tank: the culture medium comprises 1.5% of glucose, 1.0% of molasses, 2.0% of corn flour, 3.0% of silkworm chrysalis meal, 1.0% of soybean cake meal, 2.0% of selenium-rich yeast powder (total selenium 1000ug/g), 0.2% of peptone, 1.5% of bran, 0.01% of magnesium sulfate, 0.02% of potassium dihydrogen phosphate, 0.003% of sodium selenite and the balance of water. Firstly, carrying out micro-boiling extraction on silkworm chrysalis meal, corn flour, wheat bran, soybean sprout powder and a proper amount of water for 30min, filtering, taking filtrate, adding other components, sterilizing and cooling, inoculating seeds into a seeding tank with 15% of inoculum size to a fermentation tank, carrying out 17-19 ℃ of temperature, stirring at a rotation speed of 100-120rpm, and ventilating at a speed of 1: 0.2-0.4vvm, and the growth is thick after 8 days of culture.

The mycelium powder is gray yellow to yellow brown powder, has slight fishy smell and no peculiar smell, is taken a little, is dyed by methylene blue, and is placed under a microscope for oil microscope examination to show multinuclear hyphae.

Claims (5)

1. A fermented Chinese caterpillar fungus mycelium powder is characterized in that the Chinese caterpillar fungus mycelium powder takes Chinese caterpillar fungus as a strain, sodium selenite or/and selenium-enriched yeast liquid is added into a culture medium for submerged fermentation culture, fermentation liquor is subjected to solid-liquid separation and drying, the obtained dry mycelium contains 625-1350ug/g total selenium, wherein organic selenium is not less than 92%, the mycelium is crushed into 80-100 meshes to prepare the mycelium powder, the mycelium powder is gray yellow to yellow brown, has slight fishy smell and is free of peculiar smell, and after a small amount of mycelium is stained with methylene blue, the mycelium with separated polynuclear hyphae can be seen under a microscope by oil microscope examination.

A fermented Cordyceps sinensis mycelium powder is characterized in that Cordyceps sinensis is used as a strain, sodium selenite or/and selenium-enriched yeast liquid is added into a culture medium for submerged fermentation culture, a fermentation liquid is subjected to solid-liquid separation and drying, the obtained dry mycelium contains 1000 plus selenium of 1350ug/g, wherein the organic selenium is not less than 92%, the mycelium is crushed into 80-100 meshes to prepare the mycelium powder, the mycelium powder is gray yellow to yellow brown, has slight fishy smell and is free of peculiar smell, and after a small amount of mycelium is stained with methylene blue, the mycelium is placed under a microscope for oil microscopic examination, and the existence of isolated polynuclear mycelium can be seen.
3. A fermented Cordyceps sinensis mycelium powder is characterized in that Cordyceps sinensis is used as a strain, sodium selenite or/and selenium-rich yeast liquid is added into a culture medium for submerged fermentation culture, a fermentation broth is subjected to solid-liquid separation and drying, the obtained dried mycelium contains 625-1350ug/g of total selenium, wherein organic selenium is not less than 92%, 7-8% of cordycepic acid, 5-8% of cordyceps polysaccharide and 0.03-0.17% of adenosine are also contained in the dried mycelium, the mycelium is crushed into 80-100 meshes to prepare the mycelium powder, the mycelium powder is gray yellow to yellow brown, has slight fishy smell and is free of peculiar smell, a small amount of mycelium powder is taken, and after methylene blue is added for dyeing, and then the mycelium powder is placed under a microscope to be detected by an oil microscope to have isolated multinuclear mycelium.
4. A fermented Cordyceps sinensis mycelium powder is characterized in that Cordyceps sinensis is used as a strain, sodium selenite or/and selenium-enriched yeast liquid is added into a culture medium for submerged fermentation culture, fermentation liquor is subjected to solid-liquid separation and drying, the obtained dried mycelium contains 1000-cake total selenium 1350ug/g, wherein organic selenium is not less than 92%, 7-8% of cordycepic acid, 5-8% of cordyceps polysaccharide and 0.03-0.17% of adenosine are also contained in the dried mycelium, the mycelium is crushed into 80-100 meshes to prepare the mycelium powder, the mycelium powder is gray yellow to yellow brown, has slight fishy smell and is free of peculiar smell, a small amount of the product is taken, and after methylene blue is added for dyeing, and then the mycelium powder is placed under a microscope to be detected by an oil microscope to have multinuclear mycelium.
5. The method for producing the fermented cordyceps sinensis mycelium powder according to any one of claims 1 to 4, comprising: slant strain culture, shake flask seed culture, first-stage seeding tank culture, second-stage seeding tank culture, fermentation tank culture, solid-liquid separation, drying, detection and crushing; the method comprises the following specific steps:
(1) slant culture of bacterial
The slant culture of the strain selects DCH culture medium of China industrial microorganism strain preservation management center, and the weight volume ratio is as follows: 5.0% of sucrose, 0.3% of yeast powder, 1.0% of tryptone, 1.0% of agar powder and inorganic ion solution; the inorganic ion solution is 0.5 g of magnesium sulfate, 1 g of monopotassium phosphate, 1 g of sodium chloride, 0.3 g of calcium chloride, 0.01 g of manganese sulfate, 0.01 g of zinc sulfate, 0.01 g of ferrous sulfate and 0.01 g of copper sulfate, and the volume of distilled water is up to 1000 ml, and the pH value is 5.6-6.0; subpackaging, sterilizing at 121 deg.C, preparing slant, inoculating activated strain to slant culture medium, and culturing at 15-18 deg.C for 30-40 days;
(2) seed culture in shake flasks
Shake flask culture medium: the weight volume ratio is as follows: 1.0-2.0% of glucose, 1.0-2.5% of corn flour, 0.5-1.5% of molasses and 2.0-6.0% of potato; silkworm chrysalis powder 2.5 wt% or live silkworm chrysalis 4.0-6.0 wt%, soybean bud powder 0.5-1.0 wt%, yeast powder 0.2-1.0 wt%, and bran 1.0-1.5 wt%; 0.01 to 0.02 percent of magnesium sulfate, 0.02 to 0.1 percent of monopotassium phosphate and the balance of water; after the culture medium is subpackaged, sterilizing at 121 ℃, cooling, inoculating the cultured slant seeds on a shaking culture medium shaker, culturing at 17-19 ℃ and at the rotation speed of 100-130rpm for 12-18 days;
(3) first-class seed tank seed culture
First-level seeding tank culture medium: the weight volume ratio is as follows: 1.0-1.5% of glucose, 1.0-2.5% of corn flour or 1.5% of starch, 1.0-2.0% of molasses, 0-2.0% of potato, 1.5-3.5% of silkworm chrysalis meal, 1.5% of soybean cake powder or 0.5-1.0% of soybean sprout powder, 0.5-1.0% of yeast powder, 0.5-1.5% of bran and 0.2-1.0% of peptone; 0.01-0.05% of magnesium sulfate, 0.02-0.1% of monopotassium phosphate, 0-0.001% of sodium selenite and the balance of water; after the first-stage seeding tank culture medium is disinfected and cooled, inoculating the shake-flask seeds into the first-stage seeding tank in an inoculation amount of 5-10%, controlling the temperature to be 17-19 ℃, and ventilating for 1: 0.2-0.4vvm, culturing for 6-8 days;
(4) second-level seed tank seed culture
Secondary seed tank culture medium: the weight volume ratio is as follows: 1.0-1.5% of glucose, 1.0-2.5% of corn flour or 1.5% of starch, 1.0-2.0% of molasses, 0-2.0% of potato, 1.5-3.5% of silkworm chrysalis meal, 1.5% of soybean cake powder or 0.5-1.0% of soybean sprout powder, 0.5-1.0% of yeast powder, 0.5-1.5% of bran and 0.2-1.0% of peptone; 0.01 to 0.05 percent of magnesium sulfate, 0.02 to 0.1 percent of monopotassium phosphate, 0.001 to 0.002 percent of sodium selenite and the balance of water; after the culture medium of the secondary seed tank is disinfected and cooled, inoculating seeds of the primary seed tank into the secondary seed tank in an inoculation amount of 10-20%, controlling the temperature to be 17-19 ℃, and ventilating for 1: 0.2-0.4vvm, culturing for 6-8 days;
(5) fermentation tank culture medium: the weight volume ratio is as follows: 1.0-1.5 percent of glucose, 2.0-2.5 percent of corn flour or 1.5 percent of starch, 1.0-1.5 percent of molasses, 0-2.0 percent of potato, 1.5-3.5 percent of silkworm chrysalis meal, 1.5 percent of soybean cake meal or 0.5-1.0 percent of soybean sprout powder, 0.5-1.0 percent of yeast powder or 0.5-2.5 percent of selenium-enriched yeast powder, 0.5-1.5 percent of bran and 0.2-1.0 percent of peptone; 0.01-0.05% of magnesium sulfate, 0.02-0.1% of monopotassium phosphate, 0.0045-0.01% of sodium selenite and the balance of water; after the culture medium of the fermentation tank is disinfected and cooled, inoculating the seeds of the secondary seed tank with the inoculation amount of 10-30%, controlling the temperature to be 17-19 ℃, and ventilating for 1: 0.2-0.4vvm, and culturing for 8-10 days;
(6) solid-liquid separation, drying, detection and crushing
Performing solid-liquid separation on fermentation liquor placed in a tank, drying the solid, detecting that the obtained dried mycelium contains 625-1350ug/g of total selenium, the organic selenium accounts for more than or equal to 92 percent of the total selenium by mass, and also contains 4-8 percent of cordycepic acid, 5-8 percent of cordyceps polysaccharide and 0.03-0.17 percent of adenosine, crushing the mycelium into 80-100 meshes to prepare mycelium powder, wherein the mycelium powder is gray yellow to yellow brown powder, has slight fishy smell and no peculiar smell, taking a small amount of the product, adding methylene blue for dyeing, and placing the product under a microscope for oil microscopic examination, and the separated multinuclear mycelium can be seen.
