本发明公开了一种均匀性及重复性更好的免疫质控品,包括由稀释液配制的质控物,所述稀释液包括PH6.9‑7.9的PBS缓冲液,在所述PBS缓冲液中添加有体积百分比30%‑70%的小牛血清和浓度小于30%的甘油。所述稀释液中添加有蛋白保护剂和防腐剂。所述PBS缓冲液的体积百分比为30%‑70%。本发明的优点在于:在稀释液中添加了甘油,提高了质控品的均匀性,在一定程度可以降低因操作等原因带来的人为误差,更好的保证了实验结果的可靠性。

Description

一种均匀性及重复性更好的免疫质控品
技术领域
本发明涉及实验室检测用质控品,尤其是涉及一种均匀性及重复性更好的免疫质控品。
背景技术
质控品是使用给定值来鉴定检测仪器的测量结果是否准确的一种材料或物质。
临床实验室的检测质量关系着病人的诊断及病情检测。实验室检测结果的可靠性、真实性,不仅需要对试剂、校准品、仪器、人员操作、方法学、环境进行考核,而且应对实验室进行室内质控与室间质量评估,进而保证实验室检测质量。由于目前国产质控品的制备工艺不够完善,所参与的室内质控与室间质评的质控品样本的稳定性不高或均匀性不好,不仅不能保证检测结果的真实性,而且会因错误结论的下达造成医疗浪费,严重时甚至会危及患者安全。
重复性是一个好的质控品优先考虑的指标。当前国内常用的传染病项目的质控品的重复性均不太高。不同的使用对象在对质控品的使用过程中也会有所不同,比如混匀方式、混匀状态、混匀时间等,如果质控样本未充分混匀,有时会失去质控的作用,不能保证检测质量。
研究表明,稀释液的配方对质控样本的均匀性可起到关键性的作用。现有的质控品稀释液是由PBS缓冲液、小牛血清、蛋白保护剂及防腐剂配制而成,重复性及均匀性均不高,满足不了对质控品的检测质量要求。
发明内容
本发明的目的在于提供一种均匀性及重复性更好的免疫质控品,制备的质控品均匀性和重复性均得以大大提高,提高了质控产品的性能。
为实现上述目的,本发明可采取下述技术方案:
本发明所述的均匀性及重复性更好的免疫质控品,包括由稀释液配制的质控物,所述稀释液包括PH6.9-7.9的PBS缓冲液,在所述PBS缓冲液中添加有体积百分比30%-70%的小牛血清和浓度小于30%的甘油。
所述稀释液中添加有蛋白保护剂和防腐剂。
所述PBS缓冲液的体积百分比为30%-70%。
本发明的优点在于:在稀释液中添加了甘油,提高了质控品的均匀性,在一定程度可以降低因操作等原因带来的人为误差,更好的保证了实验结果的可靠性。
具体实施方式
本发明所述的均匀性及重复性更好的免疫质控品,包括由稀释液配制的质控物,为提高质控品的均匀性,本发明采用体积百分比为30%-70%、PH6.9-7.9的PBS缓冲液作为稀释液溶剂,在PBS缓冲液中添加体积百分比30%-70%的小牛血清和浓度小于30%的甘油;必要时稀释液中还可以添加蛋白保护剂(氨基比林、海藻糖、蔗糖、聚乙二醇、明胶等)和防腐剂(Proclin300、氨苄西林钠、硫柳汞钠盐、叠氮化钠、氯化锌、硫酸庆大霉素等)。
下面结合具体实施例对本发明做更加详细的介绍,以方便本领域技术人员的理解。
实施例1 制备低、中、高三个水平的抗-HIV(抗-HCV、抗-TP)质控品
其具体步骤为:
(1)对搜集来的抗-HIV(或抗-HCV、抗-TP)的阳性血浆进行复检、确证,并进行灭活处理。
(2)用磁微粒化学发光法对抗-HIV(或抗-HCV、抗-TP)的阳性血浆效价验证。
(3)取体积百分比为50%、PH为7.4的PBS缓冲液作为溶剂,加入体积百分比50%的小牛血清,配制成体积1000ml的混合物,然后加入5%浓度的甘油;
(4)在磁微粒化学发光检测试剂盒上,向加入5%甘油浓度的稀释液中,加入抗-HIV(或抗-HCV、抗-TP)阳性血浆,进行调试,确认其低值检测值S/CO在2-4范围内,即得到低、中、高三个水平的抗-HIV(抗-HCV、抗-TP)质控品。
实施例2 检测实施例1制备的质控品的变异性
将实施例1配制成的质控品加入样本杯中,放于-20℃冰箱中冷冻,第二日解冻,在混匀与未混匀两种状态下,对每个样本连续测定10次,判定各自的变异。
抗-HIV质控品加甘油与未加甘油,在混匀与未混匀状态下变异结果如下表1:

从表1、表2、表3的测定数据中可以看出:实施例1制备的质控品在加入5%甘油后未混匀状态下的变异(或均匀性)相比未加入甘油混匀状态下的变异(或均匀性)相近,即在使用过程中可一定程度上降低因操作原因带来的人为误差,更好保证实验结果的可靠性。

1.一种均匀性及重复性更好的免疫质控品,包括由稀释液配制的质控物,其特征在于:所述稀释液包括PH6.9-7.9的PBS缓冲液,在所述PBS缓冲液中添加有体积百分比30%-70%的小牛血清和浓度小于30%的甘油。
2.根据权利要求1所述的均匀性及重复性更好的免疫质控品,其特征在于:所述稀释液中添加有蛋白保护剂和防腐剂。
3.根据权利要求1或2所述的均匀性及重复性更好的免疫质控品,其特征在于:所述PBS缓冲液的体积百分比为30%-70%。
