Description
技术领域
本发明涉及包含橡木提取物、葡萄籽提取物和绿茶提取物的化妆品组合物,其可用于防御空气污染物。
现有技术
人们早已知道,空气污染对人类健康有重大影响,当局最近对此发布了一些具体的警告。例如,2014年美国环境保护署报告说,超过1.42亿美国人生活在空气质量不符合国家环境空气质量标准(https://www.epa.gov/air-trends)的地区。
同年,世界卫生组织(WHO)将700万人过早死亡归咎于空气污染暴露(http://who.int/mediacentre/news/releases/2014/air-pollution/en/),并且越来越多的证据表明空气污染物对内脏器官有破坏作用(Curr.Vasc.Pharmacol.,2006,4,199-203)。
除内脏外(其中呼吸道明显是空气污染物的主要目标)(Annu.Rev.PublicHealth,1994,15,107-132),由于表面大和持续暴露,皮肤是暴露最多的目标器官之一。
迄今为止,对皮肤健康的环境影响主要根据来自阳光的紫外线辐射对皮肤的影响来评估(J.Invest.Dermatol.,2003,120,1087-1093;Prog.Biophys.Mol.Biol.,2006,92,119-131);然而,除紫外线辐射外,污染还包括有机和无机物质,这些物质可能会对皮肤健康造成进一步的风险。
在环境污染物中,已经确定了各种化学类别,从NO、CO和SO2等气态污染物和挥发性有机化合物(VOC)到持久性有机污染物(包括杀虫剂和二恶英类化合物)。
此外,颗粒物质(气体中悬浮的液滴和/或固体的复杂混合物,而镉、铅、铬和汞等重金属是常见的空气污染物)也是生物累积造成的健康风险的原因,因为它通常被悬浮在空气中的碳颗粒吸附并且可以穿透皮肤并累积(Environ.Pollut.2008 151,362-367)。
在没有关于环境污染物如何损害皮肤的整体机制的解释的情况下,根据目前可获得的数据,一些机制已经被确定为皮肤损伤的主要原因。所述机制主要与重金属有关,包括:
-产生自由基(J.Toxicol.Cut&Ocular Toxicol.,1987,6(3),183-191);
-诱导炎症级联(已发现含有重金属的粉末增加促炎细胞因子的基因表达)(Toxicol.Lett.,1999,105,92-99);
-削弱自然防御机制。
皮肤的外部组织是最容易暴露于空气污染物的组织,空气污染物(特别是重金属)可以削弱细胞对自由基的防御。因此,重金属会加剧所有其他气态污染物的毒性作用,因为它们会削弱天然防御机制的功效并加速皮肤老化。
已经证明,这些事件尤其导致加速的外在皮肤老化(皱纹和暗斑形成的增加)(Biol.Chem.,2010,391,1235-1248;J.Investig.Dermatol.2010,130,2719-2726)和特应性皮炎的增加(Allergy,1996,51,532-539)。
虽然需要进一步深入研究以充分了解空气污染物对皮肤产生有害影响的机制,但应注意的是,鉴于有限的科学证据,除了限制暴露外,没有旨在保护头皮和毛发免受空气污染的指南(JEADV,2015,29,2326-2332)。
鉴于上述因素,仍然需要发现有效保护皮肤、头皮、毛发和外部粘膜,尤其是防御空气污染物的化妆品组合物。

发明详述
本发明涉及包含橡木提取物、葡萄籽提取物和绿茶提取物的化妆品组合物的用途,所述化妆品组合物可用于保护皮肤、头皮、毛发和外部粘膜防御空气污染物。
根据优选的实施方案,本发明的组合物可用于保护外部粘膜,尤其是鼻和口的粘膜(例如嘴唇)。
橡木提取物(Q)优选是总多酚含量为30%至60%w/w,更优选等于或大于45%w/w的提取物。橡木提取物(Q)优选为水性干提取物。
根据优选的实施方案,橡木提取物可以从树皮获得。
橡木提取物在组合物中的含量可以为0.01%至5%w/w,优选0.05%至1%w/w,更优选0.25%w/w,甚至更优选0.1%w/w。
葡萄籽(GS)提取物优选是总原花青素含量(通过Folin方法计算并表示为儿茶素类)等于或大于95%w/w并且通过HPLC方法评价的单体含量(由儿茶素和表示为儿茶素的表儿茶素的总和得到)在5-15%w/w之间的提取物,更优选为水性干提取物。
葡萄籽提取物在组合物中的含量可以为0.01%至5%w/w,优选0.05%至1%w/w,更优选0.25%w/w,甚至更优选0.1%w/w。
绿茶(GT)提取物优选是多酚含量(通过Folin方法计算并表示为儿茶素类)等于或大于40%w/w并且通过HPLC方法评价的儿茶素含量(表示为表儿茶素-3-O-没食子酸酯)等于或大于15%w/w的提取物,更优选水性干提取物。
根据优选的实施方案,绿茶提取物可以从叶子中获得。
绿茶提取物在组合物中的含量可以为0.01%至5%w/w,优选0.05%至1%w/w,更优选0.25%w/w,甚至更优选0.1%w/w。
所有提取物都可购买到,或者可以由技术人员使用已知技术容易地制备。
组合物可优选地局部施用。
包含本发明组合物的制剂可以通过例如“Remington’s PharmaceuticalHandbook”,Mack Publishing Co.,N.Y.,USA中记载的常规技术制得。
局部制剂的实例是乳液、凝胶、粉底、唇膏和软膏。
现已令人惊讶地发现,含有橡木提取物、葡萄籽提取物和绿茶提取物的组合物对于皮肤、头皮、毛发和外部粘膜防御空气污染物的保护活性大于单独使用这些提取物时可获得的保护活性,从而证明了协同活性。这得到了所进行的实验研究结果的支持。
具体地讲,已经证明本发明的组合物可用于减少重金属作为空气污染物所引起的有害影响。
以下实施例进一步说明了本发明。

为评价实施例1的化妆品组合物和单独使用的提取物(其用量等于组合物中的含量)的保护效力进行实验,实验的描述如下。
实施例2–对细胞活力的效力评价
在该测试中使用的实验模型由人皮肤衍生的成纤维细胞(ATCC-CRL-2703)代表。将细胞在含有10%胎牛血清的完全DMEM培养基中培养并保持在37℃,5%CO2的培养箱中。使细胞生长至汇合以进行测试。为了进行测试,用重金属混合物(Pb-Ef-Cr,每种金属90μM)处理人成纤维细胞培养物24小时,这些重金属是空气污染条件下典型存在的已知污染物。在环境损害的同时,用3种浓度的试验产物处理细胞培养物,这些浓度是从那些在初步细胞毒性试验中未证明具有细胞毒性的浓度中选择的。
将细胞培养物暴露于样品48小时。在测试期结束时,通过MTT测定评估细胞活力。MTT测定(3-(4,5-二甲基噻唑-2-基)-2,5-二苯基四唑溴化物)是用于测量细胞活力的简单、准确的标准比色方法。该测定基于黄色的四唑盐在细胞内被线粒体酶琥珀酸脱氢酶还原为蓝色/紫色的甲晶体。因此,该反应仅可以在代谢活性细胞中发生,并且从分光光度读数获得的光密度值与存在的活细胞的量相关。在每次处理结束时,用PBS洗涤所有的孔,用0.5mg/mL MTT溶液染色,并在36.5℃/5%CO2气氛中温育3小时。然后用异丙醇处理孔并在室温下温育2小时。温育后,在酶标仪中在570nm处获取吸光度读数(异丙醇溶液用作空白)。对于每个实验条件,处理的培养物的平均光密度与阴性对照的平均光密度之间的比率确定了细胞存活率。
将结果与阴性对照培养物(未处理的,CTR-)和阳性对照培养物(仅用重金属处理,CTR+)进行比较。处理一式三份地进行。
下表(表1)显示了与阳性对照相比以蛋白质含量和保护%表示的结果。统计学上显著的t检验值用(*)表示p<0.05,(**)表示p<0.95。

实施例3–对细胞代谢(蛋白质含量)的效力评价

所述评价通过Lowry比色法在细胞培养基上进行。Lowry测定采用与缩二脲方法相同的原理,即在碱性介质中Cu+离子与蛋白质复合并催化酪氨酸和色氨酸残基的氧化过程的反应。所述氧化导致Folin-Ciocalteu试剂的还原,其从特征性的黄色变成蓝色;蓝色越深,生物基质中存在的蛋白质越多。定量测定使用通过增加已知浓度的标准白蛋白构建的校准曲线。
下表显示了与阳性对照相比以蛋白质含量和保护%表示的结果。统计学上显著的t检验值用(*)表示p<0.05,(**)表示p<0.95。

如通过上述实施例清楚地证明的,通过施用本发明的组合物可获得的效果大于通过分别施用橡木提取物、葡萄籽提取物和任选的绿茶提取物可获得的个体效果的总和。换句话说,各个活性成分之间的相互作用产生了明显的协同作用。
Claims (11)
1.包含橡木提取物、葡萄籽提取物和绿茶提取物的化妆品组合物在保护皮肤、头皮、毛发和外部粘膜防御空气污染物中的用途,其中的空气污染物是重金属。
2.根据权利要求1所述的用途,其中外部粘膜是鼻粘膜或口腔粘膜。
3.根据权利要求2所述的用途,其中外部粘膜是嘴唇。
4.根据权利要求1-3所述的用途,用于局部施用。
5.根据权利要求1-4所述的用途,其中所述橡木提取物的特征在于总多酚含量为30%至60%w/w,更优选等于或大于45%w/w。
6.根据权利要求1-5所述的用途,其中所述橡木提取物的含量为0.01%至5%w/w,优选0.05%至1%w/w,更优选等于0.25%w/w,最优选等于0.1%w/w。
7.根据权利要求1-6所述的用途,其中葡萄籽提取物的特征在于总原花青素含量(通过Folin方法计算并表示为儿茶素类)等于或大于95%w/w,并且单体含量(由儿茶素和表示为儿茶素的表儿茶素的总和得到)范围为5%至15%w/w。
8.根据权利要求1-7所述的用途,其中所述葡萄籽提取物的含量为0.01%至5%w/w,优选0.05%至1%w/w,更优选等于0.25%w/w,最优选等于0.1%w/w。
9.根据权利要求1-8所述的用途,其中绿茶提取物的特征在于多酚含量(通过Folin方法计算并表示为儿茶素类)等于或大于40%w/w并且儿茶素含量(表示为表儿茶素-3-O-没食子酸酯)等于或大于15%w/w。
10.根据权利要求1-9所述的用途,其中所述绿茶提取物的含量为0.01%至5%w/w,优选0.05%至1%w/w,更优选等于0.25%w/w,最优选等于0.1%w/w。
11.根据权利要求1-10所述的用途,其中所述提取物是水性干提取物。
