Abstract
The invention discloses a whitening and skin-care composition containing an orange seed extract. The whitening skin care composition comprises the following components in percentage by weight: 0.1-8.0% of citrus seed extract, 1.0-5.0% of synergist, 2.0-5.0% of emulsifier, 0.3-0.8% of thickener, 5.0-35.0% of moistening agent, 0.5-3.0% of preservative, 0.01-0.5% of essence and the balance of deionized water. The citrus seed extract contains limonin substances, especially limonin, and has antioxidant, antiinflammatory, and skin whitening effects. The skin care composition containing citrus seed extract has the effects of alleviating skin pigmentation, relieving oxidative stress of skin cells, and reducing the generation of skin wrinkles, and thus has whitening and skin caring effects.
Description
Technical Field
The invention relates to a cosmetic composition containing plant extracts, belongs to the technical field of personal care products, and particularly relates to a whitening and skin-care composition containing citrus seed extracts.
Background
Citrus is a plant of the Citrus genus (Citrus) of the Rutaceae family (Rutaceae), a tropical, subtropical evergreen fruit tree, with 3 genera used for economic cultivation: poncirus, Citrus, and Poncirus. In recent years, as the living standard and aesthetic standard of people are improved, more and more people worried to whiten healthy skin. While anti-oxidation is the most important method for maintaining skin whitening, it is a well-known method for delaying skin aging, pigmentation, and wrinkle generation by acting on skin cells using an antioxidant. With the increasing awareness of the side effects of chemically synthesized antioxidants, plant antioxidants from natural sources are becoming more and more popular with people due to their advantages of being green, natural and free of side effects, and the research and development of plant antioxidant whitening skin care factors are of great significance.
In fact, as the waste of the processing of the citrus industry, namely the reprocessing and utilization of citrus seeds, the problem of environmental pollution caused by the waste can be effectively solved. Then, no research report exists that citrus seeds are reprocessed and applied to whitening and skin-care products.
Disclosure of Invention

The preparation process of the citrus seed extract of the whitening and skin-care composition comprises the following steps:
1) pretreatment: removing pulp from Mandarin orange, collecting Mandarin orange seed, sun drying, and pulverizing;
2) removing grease and protein: leaching oil in the citrus seed powder by using normal hexane, drying under reduced pressure, and volatilizing residual normal hexane to obtain citrus seed powder A; dissolving and leaching the protein in the citrus seed powder A by using a NaCl solution as a protein extraction solvent, centrifuging, collecting filter residue, and drying under reduced pressure to obtain citrus seed powder B;
3) primary extraction of limonin: adding the citrus seed powder B into an aqueous solution of sodium salicylate, extracting at 40-60 ℃ for 2-8h, removing residues, adding dichloromethane into the filtrate, fully mixing and extracting, and collecting a lower dichloromethane phase to obtain a solution containing limonin;
4) refining and crystallizing limonin: adding isopropanol into the dichloromethane dissolved phase, mixing uniformly, standing the solution at 10-10 deg.C for 3-48h, filtering to remove dichloromethane and isopropanol to obtain the final product.
Further, in the step 4), the prepared citrus seed extract is limonin in white crystal shape.
The whitening and skin-care composition provided by the invention has the beneficial effects that the synergist is selected from one or more of vitamin C, vitamin E and derivatives thereof.
The whitening and skin-care composition of the invention has the emulsifier which is nonionic, anionic, cationic or amphoteric, and the combination thereof, such as: one or more of higher fatty alcohol polyoxyethylene ether series, polyethylene glycol stearate, ethoxylated methyl glucoside stearate, polyoxyethylene sorbitan monostearate series, fatty glyceride, sorbitan monofatty ester, and fatty acid citric glyceride.
The whitening skin care composition provided by the invention has the advantages that the thickening agent is one or the combination of more than two of polyacrylate and polyacrylamide copolymer, polyacrylate and polyacrylamide dimethyl taurate copolymer, hydroxyethyl acrylate and acrylic acid dimethyl taurate copolymer, xanthan gum, cellulose and hyaluronic acid.
The whitening skin care composition of the invention is prepared by mixing more than two of natural grease, modified grease or polyalcohol, such as nut oil, shea butter, lanolin, white mineral oil, vaseline, glycerol, diglycerol and 1, 3-butanediol.
The skin whitening and caring composition comprises more than two of phenoxyethanol and ethylhexyl glycerin compound, benzyl alcohol, polyhydric alcohol, chlorphenesin, parabens, DMDM hydantoin and iodopropynyl butyl carbamate.
The whitening skin care composition containing the citrus seed extract has the following technical effects:

The invention is further illustrated by the following specific examples, which are intended to be illustrative in more detail only and are not intended to be limiting in any way.
1. The preparation method of the citrus seed extract comprises the following steps:
the citrus seed extract used in the invention is a white crystal citrus seed extract obtained by extracting and refining citrus seeds serving as raw materials. The method comprises the following specific steps:
1) pretreatment: removing pulp from Mandarin orange, collecting Mandarin orange seeds, sun drying, pulverizing, and sieving with 40 mesh standard sieve;
2) removing grease and protein: leaching oil in the citrus seed powder with n-hexane, filtering to obtain residue, drying at 50 deg.C under reduced pressure, and volatilizing residual n-hexane to obtain citrus seed powder A; dissolving and leaching the protein in the citrus seed powder A by using 6% NaCl aqueous solution as a protein extraction solvent, centrifuging, collecting filter residue, and drying under reduced pressure at 60 ℃ to obtain citrus seed powder B;
3) primary extraction of limonin: adding the citrus seed powder B into the aqueous solution of sodium salicylate, extracting at 55 deg.C for 4 hr, removing residue, adding dichloromethane into the filtrate, mixing and extracting, and collecting the lower dichloromethane phase to obtain solution containing limonin;
4) refining and crystallizing limonin: adding isopropanol (dichloromethane: isopropanol 1:3) into dichloromethane solution containing limonin, mixing, standing at 0 deg.C for 24 hr, and filtering to remove dichloromethane and isopropanol to obtain citrus seed extract (white crystal limonin).
FIG. 1 shows a high performance liquid chromatogram of the detection result of limonin standard. Fig. 2 shows the hplc detection results of limonin, which shows that the peak time of the citrus seed extract of the present invention is consistent with that of the hplc of the standard. The limonin can be extracted by the operation steps of the invention.
2. And (3) testing cytotoxicity and melanogenesis inhibition effects of the citrus seed extract:
the above citrus seed extract was used as a test sample to evaluate the cytotoxicity and melanogenesis inhibitory effect in the following manner (3 parallel experiments in total).
1) Cell inoculation, addition of test substances:
mouse B16 melanoma cells were seeded into 6-well plates at 100,000 cells/well. The next day, a test substance solution (DMSO as solvent) was added at a certain concentration.
2) Cell proliferation assay
After 3 days of addition of the test substance solution, the medium was removed by suction, and then 1ml of EMEM medium containing 10% of Amani blue solution was added and allowed to react at 37 ℃. After 30 minutes, 100. mu.l of the solution was transferred to a 96-well plate, and fluorescence was measured under excitation at 544nm and measurement at 590 nm. As can be seen from table 1, the citrus seed extract of the present invention has no significant cytotoxicity, and at the same time, has an excellent melanin inhibiting effect, and is effective in reducing skin cytochrome deposition.
The citrus seed extract of the present invention can be used in cosmetic compositions of various formulations such as lotions, milky lotions, creams, etc., and the formulation is not particularly limited.
Example 1
An essence or skin lotion added with an orange seed extract comprises the following raw materials in percentage by weight:

1) preparing a water phase: dissolving xanthan gum in propylene glycol, adding glycerol, hydroxyethyl urea, EDTA-2Na, allantoin and deionized water, stirring, heating to 85 deg.C, dissolving completely, and stirring for 30 min;
2) oil phase preparation: stirring and heating hexadecanol/octadecanol, high-carbon fatty alcohol dextran emulsifier, mixture of PEG-100 stearate and monoglyceride, 26# white mineral oil, caprylic/capric triglyceride, methyl hydroxybenzoate and propyl hydroxybenzoate to 85 ℃, dissolving completely, and stirring for 30min under heat preservation;
3) adding the oil phase into the water phase at 85 deg.C, stirring and emulsifying for 10min, homogenizing at 10000-20000rpm for 3-5min with a homogenizer, slowly adding polyacrylate and polyacrylamide copolymer, and vitamin E acetate, and further homogenizing for 2 min;
4) slowly cooling to below 40 deg.C, adding essence and citrus seed extract, and stirring for 30min to obtain whitening emulsion composition containing citrus seed extract.
Example 3
A cream added with an orange seed extract comprises the following raw materials in percentage by weight:
the preparation process comprises the following steps:
1) preparing a water phase: dissolving sodium hyaluronate and xanthan gum in propylene glycol, adding glycerol, diglycerol and deionized water, stirring and heating to 85 ℃, dissolving completely, and continuing to keep the temperature and stir for 25 min;
2) oil phase preparation: stirring and heating hexadecanol/octadecanol, high-carbon fatty alcohol dextran emulsifier, monoglyceride and isomeric hexadecane to 80-85 ℃, dissolving completely, and stirring for 30min under heat preservation;
3) adding the oil phase into the water phase at 80-85 deg.C, stirring and emulsifying for 10-15min, homogenizing for 7min at 10000-;

2. Efficacy testing

the antiseptic is a mixture of more than two of phenoxyethanol and ethylhexyl glycerin compound, benzyl alcohol, polyalcohol, chlorphenesin, parabens, DMDM hydantoin and iodopropynyl butyl carbamate.
