Abstract
本发明公开了一种抗肿瘤的高价金属氮化锇配合物及其制备方法。该高价金属氮化锇配合物化学式为OsVI(N)(PhenOH)Cl3,具有良好的溶解性;对肿瘤细胞杀伤力大,对正常细胞无毒或低毒;所述高价金属氮化锇配合物能联合X‑ray产生活性氧ROS,对肿瘤细胞的杀伤力提高3倍以上。制备时,将[Bu4N][OsNCl4]与1,10‑菲咯啉‑2‑醇分别溶于甲醇溶液或者醇溶液中,混合后常温搅拌或加热搅拌,加入NH4PF6后继续搅拌,过滤,洗涤滤渣,干燥,溶于二氯甲烷,放进乙醚中,常温挥发,得高价金属氮化锇配合物晶体。本发明提供的高效低毒的联合放疗增敏抗肿瘤药物对正常细胞基本无毒,安全系数大。

技术领域
本发明涉及一种抗肿瘤药物,特别涉及提供一种抗肿瘤的联合用药物;属于生物治疗药物领域。
背景技术
癌症的发病率日益增高,据世界卫生组织报道,2012年全球约有1400万新发癌症病例和820万例癌症相关死亡。癌症成为全球发病和死亡的主要原因,并且预计今后二十年新发病例数将增加约70%(WTO,2014年世界癌症报告)。虽然当今人们对癌症的治疗手段越来越多,可使用药物也在增加,但目前大多数癌症仍无法治愈,病人生存期较短。因此,任何新的安全有效的癌症治疗方法或能提高现有治疗方法效果的手段都会对癌症患者的健康具有实际意义。
目前,抗肿瘤药物已广泛应用于癌症的治疗,但这些药物对肿瘤细胞和正常细胞选择性,在治疗癌细胞的同时,也影响了正常机体功能。因此,提高药物生物利用度,减少全身毒性反应是肿瘤化学治疗迫切解决的问题。开发安全低毒、具有肿瘤特异性且能高效负载抗肿瘤药物的肿瘤递药系统势在必行。
金属配合物是抗肿瘤药物重要的一员。顺铂是第一个临床应用抗肿瘤活性的,且铂类抗肿瘤药物得到广泛的应用。但临床上使用顺铂会产生严重的毒副作用,如肾毒性、神经毒性、恶心和呕吐等,因此寻找低毒高效的非铂类药物成为亟待解决的问题。铁、钌配合物抗肿瘤研究相对较多,锇、钌性能相似,锇配合物的相关研究很缺乏。
初步研究显示,高价的金属锇氮化物作为潜在的抗肿瘤药物具有广泛的应用前景。在国际专利申请利WO 2016/004156 Al中,合成了一系列结构相似的含不同取代基的邻菲罗啉配体组成的高价金属锇氮化物,邻菲罗啉配体所选用的取代基是甲基、苯环等弱的给电子基团,虽然该药物对于卵巢癌的毒性显示与顺铂相当,但对正常细胞也有一定的毒性,总体安全系数有待提高。
发明内容
本发明旨在克服现有肿瘤治疗中的缺点与不足,提供一种对正常细胞基本无毒,安全系数大的高效低毒的联合放疗增敏抗肿瘤药物的制备方法。
锇本身作为一种重金属元素,具有高X射线光子捕获界面和康普顿散射效应,能产生剂量增强效应,提高DNA等敏感靶分子的放射损伤,能对受照细胞产生放射增敏作用。因此设计合成新型的高效低毒锇金属配合物联合放疗进行抗肿瘤细胞是安全有效的癌症治疗方法。
本发明所合成的金属锇氮化物具有的配体虽与现有技术WO 2016/004156 Al同为邻菲罗啉配体,但是本发明所用的取代基不同于专利WO 2016/004156 Al中的任何一种,取代基是羟基,是强的给电子基团,对金属中心电子云密度影响较大,对整个配合物的活性影响也比较大;特别是本发明所合成的配合物对正常细胞基本无毒,安全系数大,对宫颈癌细胞有一定的抑制作用,在X射线作用下,能产生放疗增敏作用,对宫颈癌有显著抑制作用,证明该物质是潜在的高效低毒的联合放疗增敏药物。
本发明所合成出来的配合物是全新的配合物,经由剑桥晶体学数据中心(TheCambridge Crystallographic Data Centre,CCDC)的数据库(CSD)认证,取得了唯一识别号1843169。
本发明的目的通过下述技术方案实现:
一种抗肿瘤的高价金属氮化锇配合物:所述的高价金属氮化锇配合物化学式为OsVI(N)(PhenOH)Cl3,具有良好的溶解性;对肿瘤细胞杀伤力大,对正常细胞无毒或低毒;所述高价金属氮化锇配合物能联合X-ray产生活性氧ROS,对肿瘤细胞的杀伤力提高3倍以上。
为进一步实现本发明目的,优选地,所述的肿瘤为人宫颈癌。
优选地,所述的对肿瘤细胞杀伤力大表现为高价金属氮化锇配合物对HeLa细胞的半致死浓度为42.7μM;
所述的对正常细胞无毒或低毒表现为对正常细胞H9C2和NCM460的半致死浓度分别为122μM和127.9μM;
所述的对肿瘤细胞的杀伤力提高3倍表现为高价金属氮化锇配合物联合X-ray使用后,HeLa细胞的半致死浓度为13.55μM,相对于高价金属氮化锇配合物对HeLa细胞的半致死浓度为42.7μM,杀伤力倍数为42.7μM/13.55μM。
所述的抗肿瘤的高价金属氮化锇配合物的制备方法,包括以下步骤:
1)将[Bu4N][OsNCl4]与1,10-菲咯啉-2-醇分别溶于甲醇溶液或者醇溶液中,混合后常温搅拌或加热搅拌0.5~48小时,加入NH4PF6后继续搅拌0.5~72小时,过滤,洗涤滤渣,干燥,溶于二氯甲烷,放进乙醚中,常温挥发,得高价金属氮化锇配合物OsVI(N)(PhenOH)Cl3晶体,晶体用二氯甲烷与乙醚混合液洗涤;
2)将步骤1)所得的产物晶体进行干燥,得抗肿瘤的高价金属氮化锇配合物。
优选地,步骤1)中所述[Bu4N][OsNCl4]和1,10-菲咯啉-2-醇的摩尔比是1:1~2:1。
优选地,溶解[Bu4N][OsNCl4]与1,10-菲咯啉-2-醇用的甲醇或者乙醇与1,10-菲咯啉-2-醇的摩尔比为80:1~250:1;加入NH4PF6的量为1,10-菲咯啉-2-醇的物质的量的0.1~0.3倍。
优选地,所述的加热搅拌的温度为40-120℃;所述的常温搅拌或加热搅拌的转速为100~1200转/分。
优选地,所述的洗涤滤渣是用甲醇/乙醚或者用以乙醇/乙醚洗涤;甲醇/乙醚中甲醇与乙醚的体积比为1:5~1:10;乙醇/乙醚中乙醇与乙醚的体积比为1:5~1:10。
优选地,所述的二氯甲烷的加入量为高价金属氮化锇配合物OsVI(N)(PhenOH)Cl3晶体质量的10~40倍。
优选地,步骤2)所述的产物晶体进行干燥的温度为20~50℃,真空度为-0.04MPa~-0.07MPa,干燥时间为5~12小时。
本发明以[Bu4N][OsNCl4]为锇氮化物合成平台,通过与1,10-菲咯啉-2-醇在甲醇溶液或乙醇溶液中的作用,常温搅拌或加热搅拌,加入NH4PF6析出沉淀,得粗产物。
本发明相对于现有技术具有如下的优点及效果:
1)当前使用的小分子抗肿瘤药物和诊断剂,对肿瘤细胞的选择性较低。本发明所合成药物基于肿瘤细胞和正常细胞的差异,具有一定的选择性。本发明所用的邻菲罗琳配体,取代基羟基不同于专利WO 2016/004156Al中的任何一种,是强的给电子基团,对金属中心电子云密度影响较大,对整个配合物的活性影响也比较大,可能由于配体的独特性质使其对肿瘤细胞杀伤力大,对正常细胞几乎无毒。对比于传统的铂类抗肿瘤药物,具有更低的毒性和更强的选择性,是一种新型的高效低毒锇金属配合物。
2)本发明所涉及的高价氮化锇配合物,其中心金属锇本身作为一种重金属元素,具有高X射线光子捕获界面和康普顿散射效应,能产生剂量增强效应,提高DNA等敏感靶分子的放射损伤,能对受照细胞产生放射增敏作用;本发明证实了高价氮化锇金属配合物能联合X-ray,产生活性氧ROS,增加对肿瘤细胞的杀伤力。
3)现有的肿瘤治疗方法主要包括手术、化疗和放疗等,每种方法都各有优缺点,但相对癌症的高发病率与致死率,现有的治疗方法的效果仍不能令人满意。金属配合物高价性质与X-ray联合的出现给肿瘤治疗带来了新的希望,其安全性好,疗效显著。未来在临床应用时,可望降低金属配合物用量,同时提高治疗效果。在改善病人的生存期和生存质量的同时,大大降低治疗药用费用,具有较大的经济价值与社会意义。
4)本发明所涉及高价氮化锇配合物的配体属于邻菲罗琳类化合物,是常见的双齿配体,易跟金属配位,这使得其更利于合成;更重要的是本发明所涉及的高价氮化锇配合物的原料廉价易得,且合成、纯化步骤操作性强,制备的产物重复性和稳定性良好,可通过优化工艺,适当扩大合成规模,实现药物的商业化和应用。
5)本发明所制备的高价金属锇氮化物OsVI(N)(PhenOH)Cl3对宫颈癌细胞(包括Caski人宫颈鳞状细胞癌、HeLa人宫颈鳞状癌细胞、SiHa人宫颈鳞状癌细胞)有一定抑制作用,联合X-ray放疗能显著抑制宫颈癌细胞,同时对于正常细胞(H9C2鼠正常心肌细胞,NCM460人正常肠上皮细胞)没有显著抑制功效,是一种高效低毒的联合放疗宫颈癌药物。

具体实施方式
为更好地理解本发明,下面结合附图和实施例对本发明作进一步的描述,但本发明的实施方式不限于此。
实施例1、2、3所得高价金属氮化锇配合物OsVI(N)(PhenOH)Cl3的合成原理如图1所示。
实施例1:高价金属氮化锇配合物OsVI(N)(PhenOH)Cl3的制备
将58.8mg[Bu4N][OsNCl4]和19.6mg 1,10-菲咯啉-2-醇分别溶于3.1mL甲醇,然后混合,常温搅拌2小时,加入3mg NH4PF6继续搅拌3小时(搅拌的转速都为500转/分),过滤、干燥。用甲醇/乙醚(甲醇与乙醚的体积比为2:3)洗涤,在25℃的真空干燥箱中,干燥6小时。随后将产物溶于10mL二氯甲烷中,放进装有40mL乙醚的密闭蓝盖瓶中,静置挥发,即可得到高价金属氮化锇配合物OsVI(N)(PhenOH)Cl3晶体,晶体用二氯甲烷与乙醚混合液(二氯甲烷与乙醚比例为1:5)洗涤。
将所得的高价金属氮化锇配合物OsVI(N)(PhenOH)Cl3晶体晶体进行干燥,得抗肿瘤的高价金属氮化锇配合物。
0.5~48小时,加入NH4PF6后继续搅拌0.5~72小时
实施例2:高价金属氮化锇配合物OsVI(N)(PhenOH)Cl3的制备
将120mg[Bu4N][OsNCl4]和39mg 1,10-菲咯啉-2-醇分别溶于12.5mL甲醇,然后混合,在70℃油浴条件下搅拌48小时,加入5mg NH4PF6继续搅拌0.5小时(搅拌转速都为100转/分),过滤、干燥。用甲醇/乙醚(甲醇与乙醚的体积比为2:3)洗涤,在40℃的真空干燥箱中,干燥12小时。随后将产物溶于15mL二氯甲烷中,放进装有60mL乙醚的密闭蓝盖瓶中,静置挥发,即可得到高价金属氮化锇配合物OsVI(N)(PhenOH)Cl3晶体,晶体用二氯甲烷与乙醚混合液洗涤。
将所得的高价金属氮化锇配合物OsVI(N)(PhenOH)Cl3晶体进行干燥,得抗肿瘤的高价金属氮化锇配合物。
实施例3:高价金属氮化锇配合物OsVI(N)(PhenOH)Cl3的制备
将1.3g[Bu4N][OsNCl4]和390mg 1,10-菲咯啉-2-醇分别溶于120mL乙醇,混合后于80℃搅拌0.5小时,加入30mg NH4PF6继续搅拌72小时(搅拌转速都为1200转/分),过滤、干燥。用甲醇/乙醚(甲醇与乙醚的体积比为2:3)洗涤,在25℃的真空干燥箱中,干燥6小时。随后将产物溶于150mL二氯甲烷中,放进装有250mL乙醚的密闭蓝盖瓶中,静置挥发,即可得到高价金属氮化锇配合物OsVI(N)(PhenOH)Cl3晶体,晶体用二氯甲烷与乙醚混合液洗涤。
将所得的高价金属氮化锇配合物OsVI(N)(PhenOH)Cl3晶体晶体进行干燥,得抗肿瘤的高价金属氮化锇配合物。
测试结果:对实施例1、2、3所得的化合物进行质谱表征,结果一致。根据质谱图可知化合物的分子量与OsVI(N)(PhenOH)Cl3的分子量一致。化合物最高峰m/Z=453.9968为[OsVI(N)(PhenOH)OHCl]+,是目标产物OsVI(N)(PhenOH)Cl3水解失去两个氯离子上一个羟基所得,m/Z=520.33为[OsVI(N)(PhenOH)Cl3-H-]+。
实施例4:高价金属氮化锇配合物OsVI(N)(PhenOH)Cl3的表征
(1)采用安捷伦科技Super Nova单晶衍射仪和Bruker Smart Apex CCD单晶衍射仪上进行晶体结构测试,测试条件150K,铜靶。
(2)以溴化钾为背景,在玛瑙研砵中与1~2毫克高价金属氮化锇配合物OsVI(N)(PhenOH)Cl3研成粉末,测试400到4000cm-1的红外光谱。
(3)配制浓度为50μmol/L的高价金属氮化锇配合物OsVI(N)(PhenOH)Cl3溶液,扫描其200到800nm的紫外光谱。
(4)以最大的紫外吸收峰的波长为激发波长,测试380到600nm的荧光发射光谱图。
(5)测试结果:利用单晶衍射测定实施例1、2、3所得高价金属氮化锇配合物OsVI(N)(PhenOH)Cl3的结构,结果如图2所示,高价金属氮化锇配合物OsVI(N)(PhenOH)Cl3的结构如实施例1、2、3中目标产物一致。中心金属锇与配体上的两个氮、氮配体结合,形成带有正三价的高价锇配合物。三个氯的存在,是配合物显中性。利用红外光谱图3对高价金属氮化锇配合物OsVI(N)(PhenOH)Cl3的官能团进行进一步确认。在1000cm-1左右,是(OsN)键的特征波长。此外,紫外光谱如图4所示,高价金属氮化锇配合物OsVI(N)(PhenOH)Cl3溶液在380nm处有最大的吸收峰,应该是配体的Л→Л跃迁。还测量了高价金属氮化锇配合物OsVI(N)(PhenOH)Cl3荧光发射光谱图如图5,发现高价金属氮化锇配合物OsVI(N)(PhenOH)Cl3基本无荧光。
实施例5:高价金属氮化锇配合物OsVI(N)(PhenOH)Cl3体外抗肿瘤活性研究
采用MTT法测量实施例1中高价金属氮化锇配合物OsVI(N)(PhenOH)Cl3对恶性肿瘤细胞(Caski人宫颈鳞状细胞癌、HeLa人宫颈鳞状癌细胞、SiHa人宫颈鳞状癌细胞)以及正常细胞(H9C2鼠正常心肌细胞,NCM460人正常肠上皮细胞)的生长抑制的方法,主要包括如下步骤:
(1)将高价金属氮化锇配合物OsVI(N)(PhenOH)Cl3溶于DMSO溶剂中,置于超声仪上超声1~2min,制备5mmol/L溶液备用;
(2)取对数生长期的不同肿瘤细胞和正常细胞(美国模式培养物集存库)以密度为2×104cells/mL接种于96孔板中(100μL/孔),使其贴壁生长24小时。再分别加入步骤(1)中的不同浓度药物100μL,继续培养72h。细胞出现一定损伤程度后,每孔加入30μL的MTT溶液(5mg/mL,PBS溶液),孵育3小时。接着,抽走96孔板中的上层培养液(DMEM高糖培养基,Gibco),加入150μL DMSO(二甲基亚砜),于摇床上轻轻摇动15分钟,使96孔板中的紫色结晶物充分溶解。接着用多功能酶标仪测量570nm下各孔的吸光值,并计算细胞存活率,同时作图以求得半数抑制浓度(IC50)。细胞存活率(%)=(OD570实验组/OD570对照组)×100%。
测试结果:如下表1。

b:Safer Index=IC50(NCM460)/IC50(HeLa)=3.0。
高价金属氮化锇配合物OsVI(N)(PhenOH)Cl3呈现良好的抗肿瘤活性。其中,(Caski人宫颈鳞状细胞癌对高价金属氮化锇配合物OsVI(N)(PhenOH)Cl3最为敏感,IC50为32.97。值得一提的是,高价金属氮化锇配合物OsVI(N)(PhenOH)Cl3的相对选择性比较好,没有明显的毒性作用,其对正常细胞H9C2、NCM460)半数抑制浓度高达122μM(H9C2)和127.9μM(NCM460),安全系数分别为2.9和3.0。而现有技术WO 2016/004156Al中化合物1、2、3、4对正常细胞半数抑制浓度为21.57、16.59、17.19、8.45,安全系数除了化合物4(2.95)外,均小于本发明。以上结果表明,高价金属氮化锇配合物OsVI(N)(PhenOH)Cl3在体外能发挥优良的抗肿瘤活性,且对正常细胞无明显的毒性作用。
实施例6:高价金属氮化锇配合物OsVI(N)(PhenOH)Cl3联合X-ray的体外抗肿瘤活性研究
采用MTT法测量实施例1中高价金属氮化锇配合物OsVI(N)(PhenOH)Cl3联合X-ray对HeLa人宫颈鳞状癌细胞的生长抑制的方法,主要包括如下步骤:
(1)将高价金属氮化锇配合物OsVI(N)(PhenOH)Cl3溶于DMSO溶剂中,置于超声仪上超声1~2min,制备5mmol/L溶液备用;
(2)取对数生长期的不同肿瘤细胞以密度为2×104cells/mL接种于96孔板中(100μL/孔),使其贴壁生长24小时。再分别加入步骤(1)中的不同浓度药物100μL,六个小时后,在不同剂量的X-ray下照射,继续培养72h。细胞出现一定损伤程度后,每孔加入30μL的MTT溶液(5mg/mL,PBS溶液),孵育3小时。接着,抽走96孔板中的上层培养液(DMEM高糖培养基,Gibco),加入150μL DMSO(二甲基亚砜),于摇床上轻轻摇动15分钟,使96孔板中的紫色结晶物充分溶解。接着用多功能酶标仪测量570nm下各孔的吸光值,并计算细胞存活率,同时作图以求得半数抑制浓度(IC50)。细胞存活率(%)=(OD570实验组/OD570对照组)×100%。
测试结果:同一X-ray剂量,随着药物浓度增加,细胞存活率越低;同一药物浓度,随着X-ray剂量的增加,细胞存活率越来越低。生成曲线图如图6所示,放疗大大增加的药物的杀伤力。在图7中可以看到,药物在X-ray(0Gy、2Gy、4Gy)照射时,HeLa人宫颈鳞状癌细胞的半致死浓度分别是42.71μM、31.73μM、13.55μM。这些数据显示,跟X-ray作用后,高价金属氮化锇配合物OsVI(N)(PhenOH)Cl3对HeLa人宫颈鳞状癌细胞的杀伤力显著增强。
实施例7:等效线图分析高价金属氮化锇配合物OsVI(N)(PhenOH)Cl3具有放疗协同作用等效线图分析X射线和高价锇配合物之间的协同増敏作用。主要包括如下步骤:
(1)取对数生长期的不同肿瘤细胞)以密度为2×104cells/mL接种于96孔板中(100μL/孔),使其贴壁生长24小时。加入100μL/孔,放在在不同剂量(0Gy、2Gy、4Gy、8Gy、16Gy)的X-ray下照射。继续培养72h后,每孔加入30μL的MTT溶液(5mg/mL,PBS溶液),孵育3小时。接着,抽走96孔板中的上层培养液(DMEM高糖培养基,Gibco),加入150μL DMSO(二甲基亚砜),于摇床上轻轻摇动15分钟,使96孔板中的紫色结晶物充分溶解。接着用多功能酶标仪测量570nm下各孔的吸光值,计算细胞存活率,并以单独放疗对细胞的IC50值设为Y轴上的B点。
(2)根据实施例6中,在X-ray为0Gy时,单独高价金属氮化锇配合物OsVI(N)(PhenOH)Cl3对细胞的IC50值设为X轴上的A点,
(3)连接X轴上A点和Y轴上B点即得出一条等效线。并添加X-ray剂量为2Gy、4Gy时,细胞的IC50值的点。
测试结果:联合放化疗实验后,理论IC50add和实验IC50mix没有显著性差异时,则表明两者相互作用的类型为相加;如果IC50mix显著低于IC50add(在相加线下方)时,则说明两者具有协同作用;反之则为拮抗作用。如图8所示,无论X-ray剂量是2Gy,4Gy,都在等效线下方。即高价金属氮化锇配合物OsVI(N)(PhenOH)Cl3与X-ray都属于协同作用,这大大降低了单独放疗的剂量和单独化疗的用量,进而提高了高价锇配合物作为放疗增敏剂的生物安全性。
实施例8:DCF探针检测细胞内活性氧(ROS)水平
ROS主要包括超氧阴离子(O2·-)、过氧化氢(H2O2)、羟基自由基(HO·),采用DCF探针检测高价金属氮化锇配合物OsVI(N)(PhenOH)Cl3对HeLa人宫颈鳞状癌细胞细胞内活性氧(ROS)水平影响的方法,主要包括如下步骤:
(1)将实施例1高价金属氮化锇配合物OsVI(N)(PhenOH)Cl3溶于DMSO溶液中,置于超声仪上超声1~2min,制备5mmol/L溶液备用;
(2)取对数生长期的HeLa人宫颈鳞状癌细胞接种于96孔板(密度为2×104/mL,100μL/孔),待细胞24h贴壁后,加入浓度为15μmol/L的高价金属氮化锇配合物OsVI(N)(PhenOH)Cl3孵育6h,接受4Gy X-ray射线照射,再抽掉每孔中的培养基,加入DCF探针,继续培养30min。于多功能荧光酶标仪上检测吸光度(激发和发射波长分别为488nm和525nm)。同时用荧光显微镜实时监测细胞中DCF荧光强度。
测试结果:理想的放疗增敏剂能提高乏氧肿瘤细胞对X射线的敏感性,在X射线的照射下产生大量自由基,从而增强细胞内活性氧(Radical oxygen species,ROS)含量,最终导致肿瘤细胞死亡。因此,本发明用DCF探针检测经高价金属氮化锇配合物OsVI(N)(PhenOH)Cl3和X射线联合作用后Hela细胞内的活性氧水平变化。如图9所示。单独X射线或高价金属氮化锇配合物OsVI(N)(PhenOH)Cl3处理Hela细胞后胞内ROS水平有小幅度上升,当X射线与高价金属氮化锇配合物OsVI(N)(PhenOH)Cl3联合作用后Hela细胞内ROS水平明显增加,这说明了高价金属氮化锇配合物OsVI(N)(PhenOH)Cl3的确能提高Hela细胞响应X射线的能力,从而提高胞内活性氧含量。以上数据表明高价金属氮化锇配合物OsVI(N)(PhenOH)Cl3的重金属效应能提高对X射线的吸收能力,使其产生大量活性氧。
实施例9:克隆形成实验
克隆形成实验(Colony formation experiment)是反映肿瘤细胞对杀伤因素敏感性的重要方法。主要包括如下步骤:
(1)Hela细胞以2×103cells/mL的密度接种于6孔板皿中,待细胞贴壁24小时后加入不同浓度实施例1所得的高价金属氮化锇配合物OsVI(N)(PhenOH)Cl3。孵育6小时后接受4Gy X-ray射线照射,然后继续孵育8天。
(2)克隆群落形成后,抽掉上清的培养基,用PBS洗去培养基,加入1mL 5mg/mL甲基紫溶液染色3小时,抽掉甲基紫溶液,风干、拍照。
测试结果:实施例1所得的高价金属氮化锇配合物OsVI(N)(PhenOH)Cl3克隆形成实验如图10细胞克隆形成的照片所示,单独X射线或单独高价金属氮化锇配合物OsVI(N)(PhenOH)Cl3(分别为实施例7和实施例5)对Hela增殖的抑制作用不明显,当联合X射线与高价金属氮化锇配合物OsVI(N)(PhenOH)Cl3(药物浓度为8μM)一起作用时,Hela细胞的生存率明显降低。说明高价金属氮化锇配合物OsVI(N)(PhenOH)Cl3是有效的放疗增敏剂。综上所述,这些数据证明了氮化锇配合物联合X射线能明显抑制Hela细胞克隆群落的形成。
实施例10:高价金属氮化锇配合物OsVI(N)(PhenOH)Cl3对肿瘤细胞周期分布的影响
采用流式细胞分析术分析高价金属氮化锇配合物OsVI(N)(PhenOH)Cl3联合X-ray对HeLa人宫颈鳞状癌细胞的周期分布影响,主要包括如下步骤:
(1)将实施例1所得的高价金属氮化锇配合物OsVI(N)(PhenOH)Cl3溶于DMSO溶液中,置于超声仪上超声1~2min,制备5mmol/L溶液备用;
(2)将HeLa人宫颈鳞状癌细胞接种于6mL培养皿中(细胞密度为2×104/mL),待细胞完全贴壁后,加入不同浓度的高价金属氮化锇配合物OsVI(N)(PhenOH)Cl3,孵育6小时后,在X-ray(4Gy)照射,放在培养箱孵育48小时,收集细胞,用预冷的70%乙醇置于-20℃固定过夜。去掉上清离心,倒立扣干,加入500μL PI避光染色30min,用Beckman流式细胞仪检测细胞周期,确保细胞数目不少于10000个。用Multi Cycle软件分析细胞周期分布。
测试结果:为探究高价金属氮化锇配合物OsVI(N)(PhenOH)Cl3增敏放疗作用,采用流式细胞术对单独放疗,单独加药以及放疗加药联合处理的细胞组进行周期分析,单独加药以及放疗加药联合处理的细胞组的只要表现都是是G2/M期阻滞。并随配合物的剂量增加而增加。如单独加药组G2/M由11.97%增加至14.41%(30μM)、20.65%(60μM),放疗加药物联合处理组的细胞则由9.54%增加至25.11%(30μM)、48.38%(60μM)。测试结果显示氮化锇配合物OsVI(N)(PhenOH)Cl3可以使Hela细胞停滞于G2/M期。

2.权利要求1所述的抗肿瘤的高价金属氮化锇配合物的制备方法,包括以下步骤:
1)将[Bu4N][OsNCl4]与1,10-菲咯啉-2-醇分别溶于甲醇溶液或者乙醇溶液中,混合后常温搅拌或加热搅拌0.5~48小时,加入NH4PF6后继续搅拌0.5~72小时,过滤,洗涤滤渣,干燥,溶于二氯甲烷,放进乙醚中,常温挥发,得高价金属氮化锇配合物OsVI(N)(PhenOH)Cl3晶体,高价金属氮化锇配合物OsVI(N)(PhenOH)Cl3晶体用二氯甲烷与乙醚混合液洗涤;
2)将步骤1)所得的产物晶体进行干燥,得抗肿瘤的高价金属氮化锇配合物。
3.根据权利要求2所述的抗肿瘤的高价金属氮化锇配合物的制备方法,其特征在于:步骤1)中所述[Bu4N][OsNCl4]和1,10-菲咯啉-2-醇的摩尔比是1:1~2:1。
4.根据权利要求2所述的抗肿瘤的高价金属氮化锇配合物的制备方法,其特征在于:溶解[Bu4N][OsNCl4]与1,10-菲咯啉-2-醇用的甲醇或者乙醇与1,10-菲咯啉-2-醇的摩尔比为80:1~250:1;加入NH4PF6的量为1,10-菲咯啉-2-醇的物质的量的0.1~0.3倍。

6.根据权利要求2所述的抗肿瘤的高价金属氮化锇配合物的制备方法,其特征在于:所述的洗涤滤渣是用甲醇/乙醚或者用以乙醇/乙醚洗涤;甲醇/乙醚中甲醇与乙醚的体积比为1:5~1:10;乙醇/乙醚中乙醇与乙醚的体积比为1:5~1:10。
7.根据权利要求2所述的抗肿瘤的高价金属氮化锇配合物的制备方法,其特征在于:所述的二氯甲烷的加入量为高价金属氮化锇配合物OsVI(N)(PhenOH)Cl3晶体质量的10~40倍。
8.根据权利要求2所述的抗肿瘤的高价金属氮化锇配合物的制备方法,其特征在于:步骤2)所述的产物晶体进行干燥的温度为20~50℃,真空度为-0.04MPa~-0.07MPa,干燥时间为5~12小时。
