The cedar essential oil and the compound containing the cedar essential oil can maintain the integrity and stability of a dermis connecting area, remarkably improve the expression of a key protein Lamin 5 or COL VII of an epidermis connecting area, achieve the anti-aging effect, can be used as an active ingredient, and can be used for preparing cosmetics with the effects of delaying aging, resisting aging and keeping skin functions.
Description
Technical Field
The invention relates to the field of cosmetics, in particular to the use of cedar essential oil and a composite containing cedar essential oil as a efficacy additive in external skin preparations, especially cosmetics.
Background
Cedar is a plant of genus cedar of family pinaceae, with a scientific name: G.Don, the name Himalayan cedar, the native Himalayan mountain, has a west altitude of 1000-4000 m. The area of origin is up to 75m, and the crown is conical. The cedar is a evergreen arbor, the crown of the cedar is in a pyramidal shape, the big branches are flat, and the small branches slightly droop; leaf needle shape, length 8-60 cm, hard, grey green or silver gray, scattering on long branch, clustering on short branch; flowering in 10-11 months, the cones mature next year, oval in shape, and reddish brown when mature. The cedar is one of famous garden ornamental trees in the world, has strong dust-proof, noise-reducing and sterilizing capabilities, and is also suitable for being used as greening trees for industrial and mining enterprises.
The medicinal use of cedar has long been known and can be traced back to the biblical age at the earliest. Cedar wood is rich in essential oils and can be extracted from wood chips or chips by distillation. The essential oil of cedar contains cedrol and wedeol as main components, has the medical effects of antisepsis, sterilization, diuresis, menstruation regulation, phlegm elimination, disinsection, sedation and the like, is also beneficial to treating arthritis, bronchitis, rheumatism and respiratory tract problems, and is applied to palace beauty treatment in ancient Egypt period.
The astringent and antibacterial properties of cedar essential oil can be used for improving acne, pore obstruction and dermatitis, treating dandruff and rash, and improving oily skin.
The skin is covered on the whole human body surface, is in direct contact with the external environment, protects the body from being damaged by various external factors, and is a mirror reflecting the health state of the whole body. The skin is composed of epidermis, dermis, and subcutaneous tissue, and contains skin appendages, abundant nerves, blood vessels, lymphatic vessels, and muscles. The junction area between the dermis and epidermis, also known as the basement membrane zone, is not only a structural model but also has versatile functions. The basement membrane is the supporting cushion for epithelial cells, and serves as a structural link between epithelial cell tissue and connective tissue, as well as other functions. For example, the basement membrane acts as a selective cellular barrier, generally preventing connective tissue cells from contacting epithelial cells on the basement membrane, but allowing lymphocytes, macrophages, and neuronal synapse to cross; in addition, many vital phenomena such as cell morphology, cell polarity, cell metabolism, protein distribution in plasma membranes, cell survival, cell proliferation and differentiation, and cell migration are very closely related to the basement membrane.
The genuine epidermal junction region is composed mainly of collagen, laminin, catenin, and diafiln. Collagen is a closely related, but genetically distinct macromolecule, and more than 20 kinds of vertebrate collagen have been identified at present. All collagen molecules are composed of three polypeptide subunits, called the alpha chain, and the human genome encodes these polypeptides in up to 25 distinct genes. Genetically distinct collagens have a characteristic tissue distribution. For example, the most abundant collagens, such as type I and type III, are widely distributed, and the less abundant collagen is type VII collagen (COL VII), at the dermal and epidermal junction. Collagen serves as an anchoring function, firm connection between the true epidermal cells is formed, and the corresponding collagen or related genes are lacked to generate mutation, so that various diseases such as bullous diseases and the like can be caused.
Laminin (lamin) is a non-collagen glycoprotein with multiple biological functions, and plays a vital role in the construction of a basement membrane and the adhesion, growth, migration and differentiation of epidermal basal layer cells. Laminin is assembled into a cross-shaped or T-shaped structure by three polypeptide chains of alpha, beta and gamma through disulfide bond connection, and a molecule has a long arm (the carboxyl end of the polypeptide chain is polymerized through disulfide bonds) and three short arms (the amino ends of the three polypeptide chains form). The molecule has functional sites for binding type IV collagen, cells and proteoglycans. At present, the laminin component polypeptide chains which are proved to have 5 alpha chains, 3 beta chains and 3 gamma chains, and 15 laminins are found so far, wherein laminin-5 plays a greater role in the construction of a basement membrane network structure and the mediation of adhesion and migration of basal layer cells on a basement membrane than other types of laminins. Laminin-5 (Laminin 5) is synthesized in the rough endoplasmic reticulum in the cytoplasm of epithelial cells, forms beta 3 gamma 2 dimer and then forms alpha 3 beta 3 gamma 2 trimer, is secreted to a basement membrane outside the cells after glycosylation reaction in a Golgi complex, and plays an important role in the processes of constructing and repairing the basement membrane, regulating adhesion and homing of basal layer cells and the like.
The genuine epidermal junction separates the basal cells of the epidermis from the connective tissue underlying them, provides structural support to the cells, and maintains epidermal-dermal function through epidermal-dermal cytokine regulation. The overlap and rupture of the basement membrane of the skin can be caused by aging and ultraviolet irradiation. The connection between the skin and the epidermis and the dermis and the information transmission are blocked, which can cause various skin troubles and even skin pathological changes. Therefore, smooth circulation and communication among the epidermis, the basement membrane and the dermis are important factors for healthy skin, and can be called as 'golden circulation'. There are various factors causing changes of the base film, and photoaging is one of important factors. The photoaging mediated by UV mainly causes changes in the levels of major components in the basement membrane, resulting in changes in the function of the basement membrane, thereby affecting the functions of the epidermis and dermis, causing unsmooth "golden cycle" between the three, and causing various skin problems.
Therefore, the research on how to maintain the stability of the true epidermal junction area is of great significance to the health protection of the skin and the delay of skin aging.
There is no report that cedar essential oil or its complex is used for improving or delaying skin aging, maintaining stability of the true epidermal junction region and stabilizing the structure of the basement membrane of the skin, and there is no report that cedar essential oil or its complex increases the expression of laminin or promotes collagen synthesis.
Disclosure of Invention
The invention aims to apply cedar essential oil to a skin external preparation, in particular to a skin external preparation with an anti-aging function.
The invention also provides a compound containing cedar essential oil, namely the cedar essence compound, and the compound is used for a skin external agent, in particular to the skin external agent with the anti-aging function.
The cedar essential oil can be used as active ingredient for preparing skin external preparation, especially as anti-aging active ingredient for skin external preparation.
Cedar essential oil has functions of maintaining the integrity and stability of dermal junction region, promoting the production of laminin or collagen, and can be used as active ingredient for external preparation for skin.
Cedar essential oil increases epidermal layer thickness, and increases expression of lamin 5 or COL VII.
The skin external preparation containing Cedar essential oil as active ingredient has effects in maintaining integrity and stability of dermal junction region, and promoting generation of laminin or collagen. Such external skin preparations can increase the thickness of the epidermal layer, and increase the expression of Lamin 5 or COL VII.
The addition amount of the cedar essential oil is 0.00001 wt% -10 wt%, preferably 0.00005 wt% -5 wt%, more preferably 0.0001 wt% -3 wt%.
A preferred embodiment of the invention shows that when the content of the cedar essential oil is 0.0001 wt%, the thickness of an epidermal layer can be obviously increased, and the expressions of Lamin 5 and COL VII can be improved, so that the skin function can be improved, and the skin aging can be delayed.
The skin external preparation is a cosmetic, such as astringent, essence, lotion, cream, pack, eye cream, smoothing toner, cleansing cream, sunscreen cream/emulsion, foundation, lipstick, etc. The skin external preparation contains an acceptable carrier.
The skin external preparation contains other active ingredients with moisturizing, anti-aging, anti-allergic, sun-screening or whitening functions.
Preferably, the skin external preparation contains glacial water. The pH value of the glacier water is 7.1-7.9, the deuterium content is 130-140 ppm, and the glacier water further comprises the following components: sr 2+ 0.2~0.5mg/L、K + 0.5~10.0mg/L、Na + 1.0~5.0mg/L、Ca 2+ 14.0 to 30.0Mg/L and Mg 2+ 2.0-10.0 mg/L; and the total soluble solid content in glacier water is 60-150 mg/L, and bromate is not contained. Preferably, the pH value of the glacier water is 7.2-7.6, and more preferably 7.4. Preferably, the glacier water is himalaya glacier water.
Preferably, the skin preparation for external use further contains a dispersant. The type and amount of dispersant, such as caprylic/capric triglyceride, etc., can be selected by one skilled in the art as desired.
The cedar essential oil is prepared from cedar and/or bark, especially Himalayan cedar and/or bark, by steam distillation, cold soaking or supercritical carbon dioxide extraction.
The preferred extraction method is steam distillation or supercritical carbon dioxide extraction. The steam distillation method comprises the following steps:
(1) immersing cedar wood and/or bark in water, distilling at 95-110 ℃, and collecting fractions;
(2) separating the distillate with water oil, collecting oil phase, drying, and filtering to obtain cedar essential oil.
In step (2), in order to accelerate the water-oil separation, salt can be added into the fraction to perform the water-oil separation, such as solid and/or solution of sodium chloride, sodium sulfate or sodium bicarbonate; the oil phase is dried over anhydrous sodium sulfate or anhydrous magnesium sulfate and filtered through a microfiltration membrane.
The cedar essential oil-containing compound, namely the cedar essence compound, comprises the components of cedar essential oil and glacier water, wherein the weight ratio of the cedar essential oil to the glacier water is 1: 10-40000, preferably 1: 100 to 30000, more preferably 1: 5000 to 25000.
The pH value of the glacier water is 7.1-7.9, the deuterium content is 130-140 ppm, the glacier water does not contain bromate, and the glacier water also contains the following components: sr 2+ 0.2~0.5mg/L、K + 0.5~10.0mg/L、Na + 1.0~5.0mg/L、Ca 2+ 14.0-30.0 Mg/L and Mg 2+ 2.0-10.0 mg/L; and the total soluble solid content in glacier water is 60-150 mg/L. Preferably, the pH value of the glacier water is 7.2-7.6, and more preferably 7.4. Preferably, the glacier water is himalaya glacier water.
The cedar essential oil-containing composition further comprises a dispersant for homogenization and stabilization. The type of dispersant, and the ratio of cedar essential oil to dispersant used, can be selected by one skilled in the art based on common general knowledge.
The dispersant can be caprylic/capric triglyceride.
Preferably, the dosage ratio of the cedar essential oil to the dispersing agent is 1: 1 to 500; more preferably 1: 10 to 200.
In a preferred embodiment of the present invention, the composition of the cedar essential oil-containing compound is: 0.5 wt% of himalaya cedar essential oil and 0.5 wt% of dispersant caprylic/capric triglyceride and 99.5 wt% of glacier water. Wherein, the weight ratio of the cedar essential oil to the dispersant caprylic acid/capric acid triglyceride is 1: 99.
the cedar essential oil-containing compound is prepared by the following method: mixing the cedar essential oil and the dispersant uniformly, adding glacier water, and mixing uniformly. The obtained complex can be added into skin external preparation.
The compound containing cedar essential oil as active ingredient can be used for preparing skin external preparation, especially skin external preparation with antiaging effect.
The composition containing cedar essential oil is used as active ingredient for preparing skin external preparation with effects of maintaining stability and integrity of dermal junction.
Alternatively, the composition containing cedar essential oil as an active ingredient can be used for preparing an external preparation for skin capable of promoting the formation of laminin or collagen.
Alternatively, the composition containing cedar essential oil can be used as active ingredient for preparing skin external preparation for increasing epidermal thickness and increasing Laminin5 or COL VII expression.
The composition containing cedar essential oil can also be added with other active ingredients with moisturizing, anti-aging, anti-allergic, sunscreen or whitening functions.
In the skin external preparation, the addition amount of the cedar extract compound as an active ingredient is 0.01 to 90 wt%, preferably 0.5 to 60 wt%; significant effects are obtained at a level of 0.0001 wt% based on the weight of cedar essential oil.
The skin external preparation is cosmetic, and comprises astringent, essence, lotion, cream, facial mask, eye cream, smoothing toner, astringent, cleaning cream, sunscreen cream, foundation solution, lipstick, etc.
The invention firstly proposes that the cedar essential oil or cedar essence compound can obviously increase the thickness of epidermis, obviously improve the expression of COL VII protein and Lamin 5 protein which are key proteins at the junction of the true epidermis of the skin, have the functions of maintaining the structural integrity of DEJ (the junction of the true epidermis) and protecting the long-term stability, and can be used for preparing the skin external agent, in particular the cosmetics, which has the effects of delaying senility and resisting aging, is beneficial to promoting the regeneration of the epidermis, and tightens the structure of the true epidermis to keep the skin function.

Detailed Description
Example 1 extraction of essential oil of Himalayan Cedrus deodara
The extraction method of the himalaya cedar essential oil by the distillation method in water comprises the following steps:
1. cutting collected Himalayan cedar raw materials into small sections of 1.5-5 cm, washing with clear water, draining, keeping the humidity of the wood, putting into a steamer, and adding distilled water until the wood is submerged to the height of the wood;
2. heating the steamer in a distillation device; distilling for 2-3 hours at 100-105 ℃, and collecting fractions to obtain an oil-water mixture; the steam with the temperature of 100-105 ℃ can be directly introduced into the steamer for heating or an electric heating sleeve is used for heating;
3. separating an oil-water mixture: taking an oil phase after the oil-water mixture is layered;
in order to accelerate the oil-water separation, solid or saturated solution of sodium chloride, sodium sulfate, sodium carbonate or sodium bicarbonate can be added into the oil-water mixture until the salt concentration in the oil-water mixture is 0.1-0.2 g/mL;
4. drying and filtering: adding anhydrous magnesium sulfate or anhydrous sodium sulfate into the oil phase to remove water, and filtering with 0.22 μm microporous membrane to obtain Himalayan cedar essential oil.
Or extracting Himalayan cedar essential oil from Himalayan cedar wood by carbon dioxide supercritical method.
Laminin (lamin) is the major protein in the junction region of true epidermis, with lamin 5 playing an important role in the construction of basement membrane and the adhesion, growth, migration, differentiation of epidermal basal lamina cells, and is more potent than other types of Laminin. The type VII collagen is collagen with a small content in the true epidermis, and plays an anchoring role at the junction of the dermis and the epidermis, so that firm connection is formed between the true epidermis cells.
The dermoepidermal junction is an important representative of the barrier function of the reactive skin, so to verify the efficacy of cedar extract in protecting the dermoepidermal junction, we analyzed both the growth status of the 3D skin model after sample treatment and the intensity of the fluorescent staining of both the Laminin5 and COL VII antibodies.
If cedar essential oil is added, the growth state of a skin model is good, the integrity of the true epidermis junction area is superior to that of a sample group without adding cedar essential oil, then the fluorescence expression quantity can be detected in a mode of Lamin 5 and COL VII immunofluorescence staining, and if the expression quantity is increased relative to a control group without adding cedar essential oil, the cedar essential oil can be reflected to have an effect of protecting the integrity of the true epidermis junction area, and further the anti-aging effect is achieved.
Example 2 preparation of cedar extract composites
1g of cedar essential oil is uniformly mixed with 99g of caprylic/capric triglyceride; and (3) uniformly mixing 0.5g of the mixture with 99.5g of glacier water to obtain the himalaya cedar essence compound with the initial concentration of 1. In cytological experiments, 2% himalayan cedar extract complex was used for subsequent efficacy assessment tests.
The glacier water is obtained from Himalaya North slope poly Gltungeny Ma spring, contains no bromate, has pH of 7.4 and deuterium content of 133-: sr 2+ 0.2~0.5mg/L、K + 0.5~10.0mg/L、Na + 1.0~5.0mg/L、Ca 2+ 14.0 to 30.0Mg/L and Mg 2+ 2.0-10.0 mg/L; and the total soluble solid content in glacier water is 60-150 mg/L.
Example 3Masson staining for epidermal thickness
The test method is as follows:
(I) culture of 3D skin model
Inoculating human fibroblast NHDF-P4 generation and human keratinocyte NHEK-P2 generation into a skin bracket, and sequentially performing dermis culture stage, epidermis immersion culture stage and gas-liquid phase culture stage to construct a 3D skin model; during the culture, the skin model was cultured in normal medium or medium containing 2% himalayacedar extract complex, respectively, from D3 after fibroblast seeding on the scaffold to the final D35.
The himalaya cedar essence compound is prepared according to the method of the embodiment 1, and mainly comprises cedar essential oil and himalaya glacier water, and the initial concentration is 100%.
The 3D skin model cultured with the normal medium was defined as an untreated group (NT), and the skin model cultured with the 2% himalayas cedar essence complex medium was defined as a sample group.
(II) analysis of epidermal thickness
The skin model was cut into sections by paraffin sectioning technique. Dyeing with hematoxylin working solution, soaking in distilled water for cleaning, and dyeing with alkaline fuchsin dyeing solution; washing with phosphotungstic acid solution, dyeing with brilliant green dye solution, and washing with deionized water. And finally, dehydrating and sealing.
The method for analyzing the thickness of the epidermis comprises the following steps: 3Masson staining pictures were taken for each of the sample group and the untreated group, 10 points were taken for each recording of the epidermis thickness value and 3 observation values were taken for the analysis after averaging, and the analysis results are shown in FIG. 1.
In the test of measuring the thickness of the epidermis of the 3D skin model, if the growth condition of the skin model added with the extract is better and the thickness of the epidermis is higher than that of the skin model not added with the extract, the cedar essence compound can be preliminarily judged to have certain good effect on the skin growth, and the skin function can be improved.
Masson staining picture results of the untreated group and the sample group show that both the two groups of 3D skin models have complete true epidermis junctions, and the epidermis is normally differentiated to have a basal layer, a spinous layer, a granular layer and a keratinized layer, which indicates that the 3D skin model is successfully constructed and has skin-like functions for analysis; as can be seen from fig. 1, the thickness of the 3D skin model of the sample group to which the 2% himalaya cedar extract complex was added was increased by 20% as compared with the untreated group, and the epidermis thickness was significantly increased, which indicates that the addition of the 2% himalaya cedar extract complex could improve the skin function and have the effect of delaying skin aging.
Example 4 fluorescent expression of Laminin5 and COL VII
The 3D skin model was cultured in the same manner as in example 2.
After 32 days of culture, frozen sections of the 3D skin models of the untreated group and the sample group were fixed with cold acetone; after the PBS was rehydrated, a serum-containing PBS solution was added and allowed to stand at room temperature for 1 hour. The primary anti-Laminin 5 antibody and COL VII antibody were added overnight at 4 ℃ and the next day the corresponding secondary antibodies Alexa Fluro 568anti-mouse and DAPI were incubated at room temperature in the dark. Dyeing, finally adding a sealing tablet, sealing, observing under a fluorescent microscope, and taking a picture.
In an immunofluorescence staining test, if the fluorescence expression quantity of the Lamin 5 and COLVII is obviously increased compared with that of a cell group without an added extract, the true epidermis junction is more complete, and the cedar essence compound can be inferred to induce the expression of the Lamin 5 and COL VII in the true epidermis junction area, so that the stability of a DEJ basement membrane structure is facilitated, and the effect of delaying skin aging is finally achieved.
The results are shown in FIGS. 2 and 3. From immunofluorescence analysis, it can be seen that the Laminin5 protein expression increased by 20% and the COL VII expression increased by 47% in the sample group to which 2% Himalayan cedar extract complex was added, as compared to the untreated group. Laminin, Laminin5, and collagen type VII, are closely related as key proteins in DEJ, helping maintain DEJ structure, stabilize the junction of the true epidermis, and aging. The results in fig. 2 and 3 demonstrate that himalayas cedar essence complex significantly increases the expression of lamin 5 and COL VII in the dermoepidermal junction region, activates key proteins, and thus plays a role in helping to delay skin aging.
The himalayas cedar essential oil extracted by supercritical carbon dioxide can also obviously increase the thickness of the epidermis and obviously improve the expression of Laminin5 and COL VII in the true epidermis connecting area.
Conclusion
The 3D skin model cultured by the construction of examples 2 and 3 found that the culture medium containing himalayan cedar essential oil and himalayan cedar extract complex enhanced skin function and increased epidermis thickness by 20% compared to the untreated group; and the expression of key proteins at the junction of the epidermis of the skin can be obviously improved, the expression of COL VII proteins can be increased by 47%, and the expression of Laminin5 proteins can be increased by 20%. The himalaya cedar essential oil and the refined compound can improve the expression of key proteins Lamin 5 and COL VII in the true epidermis connecting area through activation, so that the effect of improving the skin function at the true epidermis connecting area is promoted, the stability of a basement membrane structure is facilitated, the effect of improving the skin function is improved, and the effect of delaying the skin aging is finally realized.
Claims (5)
1. Use of cedar essential oil in preparing skin external preparation for maintaining integrity and stability of dermal junction region, increasing epidermal layer thickness, promoting Laminin or collagen generation, and improving Lamin 5 or COL VII expression is provided, wherein the cedar essential oil is prepared from cedar wood and/or bark as raw material by steam distillation, cold soaking or supercritical carbon dioxide extraction.
2. The use according to claim 1, wherein the external preparation for skin contains an active ingredient having moisturizing, anti-aging, anti-allergic, sunscreen or whitening functions.
3. The use according to claim 1, wherein the external preparation for skin comprises glacier water.
4. Use of a cedar essential oil-containing complex for the preparation of a skin preparation for external use having the effects of maintaining the integrity and stability of the dermal junction region, promoting the production of Laminin or collagen, increasing the thickness of the epidermal layer, or increasing the expression of Laminin5 or COL VII, wherein the cedar essential oil-containing complex comprises cedar essential oil and glacier water in a weight ratio of 1: 10-40000; the cedar essential oil is prepared from cedar wood and/or bark by steam distillation, cold soaking or supercritical carbon dioxide extraction.
5. The use according to claim 4, wherein the cedar essential oil-containing compound further comprises a dispersing agent.
