Abstract
本发明公开了一种茶粕用作miR‑376b‑5p抑制剂及在高血压治疗方面的应用。本发明发现,小果油茶粕为miR‑376b‑5p的有效抑制剂。研究显示,miR‑376b‑5p对丹参酮改善老年SHR血压及血流动力学的作用有逆转效果,由于较高的基础血压和异常血流动力学状况可影响心室重构的进程,miR‑376b‑5p可能通过影响血压及血流动力学参与对丹参酮作用的调控。本发明中,小果油茶粕通过抑制miR‑376b‑5p的表达显著降低高血压模型大鼠的血压水平,可以制备成防治高血压的食品、药品或保健品。
Description
技术领域
本发明涉及小果油茶粕用作miR-376b-5p抑制剂及在高血压治疗方面的应用。
背景技术
小果油茶(Camellia meiocarpa Hu.)又名江西子、小茶、鸡心子、小叶油茶、羊屎子等。果小、叶小、芽小,芽、苞片没有毛是其显著特征。小果油茶耐荫、果小、皮薄、产量高,其栽培面积和年产量仅次于普通油茶,分布于福建、江西、广西、湖南、贵州、江浙等省。
小果油茶所含化学成分主要有皂苷、黄酮、多酚、多糖和蛋白类化合物等。药理研究表明,油茶植物不仅具有抗菌、消炎、清热解毒等功效,而且在抗氧化、抗肿瘤等方面也显示出较好的生物活性,其中油茶皂苷对心脑血管的药理活性显著。
发明内容
本发明目的在于提供小果油茶粕用作miR-376b-5p抑制剂及治疗高血压的应用。
本发明的上述目的是通过下面的技术方案得以实现的:
小果油茶粕用作miR-376b-5p抑制剂的用途。
小果油茶粕用于制备防治高血压的食品、药品或保健品的用途。
一种用于抑制miR-376b-5p表达的食品、药品或保健品,含有小果油茶粕,还含有食品、药品或保健品中可以接受的载体或赋形剂。
本发明发现,小果油茶粕为miR-376b-5p的有效抑制剂。研究显示,miR-376b-5p对丹参酮改善老年SHR血压及血流动力学的作用有逆转效果,由于较高的基础血压和异常血流动力学状况可影响心室重构的进程,miR-376b-5p可能通过影响血压及血流动力学参与对丹参酮作用的调控(参考文献:丹参酮通过抑制miR-376b-5p逆转自发性高血压大鼠左室重构,中国老年学杂志2016年11月第36卷)。本发明中,小果油茶粕通过抑制miR-376b-5p的表达显著降低高血压模型大鼠的血压水平,可以制备成防治高血压的食品、药品或保健品。
具体实施方式
下面结合实施例具体介绍本发明实质性内容,但并不以此限定本发明的保护范围。
一、实验材料
取小果油茶的茶果洗净、风干后压榨除油制备得到小果油茶粕。
SPF级9周龄SHR,,体质量180~200g,由北京维通利华实验动物技术有限公司提供。
二、实验方法
1、分组和给药
所有大鼠自由饮食饲养3d后,均适应性测量血压5次。平均血压≥160mmHg(1mmHg=0.133kPa)的SHR进入实验分组,称体质量后按随机数字表法分为模型组(蒸馏水,1mL/100g)和茶粕低、中、高剂量组(小果油茶粕50、100、200mg/kg),每组8只。每7天称量一次体重调整给药量,各组大鼠每天灌胃给予相应药物1次,连续给药28d。
2、血压测定
最后一次给药6h后,将各组大鼠置于配套固定器中,温水浸泡软化尾巴,采用PowerLab ML125/R无创尾动脉血压测定分析系统测定大鼠血压。重复3次,取均值。
3、miR-376b-5p表达水平测定
最后一次给药后将各组大鼠禁食、不禁水12h,按照其体质量不同分别采用3%水合氯醛腹腔注射,麻醉后颈椎脱臼处死,开胸取出心脏。
取各组大鼠心肌组织50mg,DEPC冲洗后,按Trizol一步法提取总RNA,紫外分光光度计测其纯度与浓度,其中OD260/OD280在1.8~2.0代表其纯度较好。提取总RNA后,采用颈环结构的MegaPlexTM RNA反转录引物混合物进行反转录,反转录结束后按照miRNA RT-PCR反应体系进行扩增,以U6作为内参照,进行PCR产物的半定量分析。引物序列为:
U6上游引物:5'-GCCCGATCTTGTCTGATCTC-3';
U6下游引物:5'-CCTGACCCTGCTTAGCTTCC-3';
miR-376b-5p上游引物:5'-GTAATAGATACTGATCGTCTAGTC-3';
miR-376b-5p下游引物:5'-CTCAGTCATAACGTCTGATATTGT-3'。
PCR反应条件:95℃预变性10min,95℃变性15s,60℃退火30s,75℃延伸30s,75℃终极延伸1min,共40个循环。采用2-ΔΔCt法计算miR-376b-5p的相对表达水平。
4、数据处理及分析
采用SPSS19.0软件,多组间比较采用单因素方差分析检验,组间两两比较采用q检验。以P<0.05为差异有统计学意义。
三、实验结果
1、各组大鼠血压水平比较
与模型组相比,茶粕低、中、高剂量组大鼠收缩压(SBP)和舒张压(DBP)均显著降低(P<0.05),且SBP、DBP降低程度与茶粕给药剂量呈现一定依赖性。结果见表1。

与模型组相比,茶粕低、中、高剂量组大鼠心肌miR-376b-5p表达水平显著降低(P<0.05),且降低程度与茶粕给药剂量呈现一定依赖性。结果见表2。

上述结果表明,小果油茶粕通过抑制miR-376b-5p的表达显著降低高血压模型大鼠的血压水平,可以制备成防治高血压的食品、药品或保健品。另外,小果油茶粕还可以用作筛选其他miR-376b-5p抑制剂的阳性药。
上述实施例的作用在于具体介绍本发明的实质性内容,但本领域技术人员应当知道,不应将本发明的保护范围局限于该具体实施例。
