In one embodiment of the absorbable healing promoting hemostatic composition of the present invention, the absorbable healing promoting hemostatic composition of the present embodiment comprises the following components: 7 mu g of silver nitrate, 4g of vitamin K, 2g of vitamin E and 0.1g of epidermal growth factor.
An absorbable healing promoting hemostatic dressing comprising an absorbent substrate and the absorbable healing promoting hemostatic composition of the present embodiment; the absorbable heal-promoting hemostatic composition is attached to an adsorption substrate, the adsorption substrate is carboxymethyl cellulose potassium salt gauze, and the weight percentage of the absorbable heal-promoting hemostatic composition in the absorbable heal-promoting hemostatic dressing is 2%.
The preparation method of the absorbable wound dressing for promoting healing and hemostasis comprises the following steps:
(1) preparing an absorbable healing-promoting hemostatic composition solution: dissolving the absorbable healing promoting hemostatic composition in 30mL double distilled water;
(2) soaking carboxymethyl cellulose sylvite gauze in an absorbable healing-promoting hemostatic composition solution for 3 hours;
(3) taking out, repeatedly washing with methanol for many times, and freeze-drying to obtain the absorbable healing-promoting hemostatic dressing;
the preparation method of the carboxymethyl cellulose potassium salt gauze comprises the following steps:
(a) and (3) pretreatment: washing absorbent cotton gauze in double-distilled water at the temperature of 85 ℃ for 4 hours to obtain pretreated absorbent cotton gauze;
(b) and alkalization treatment: soaking the pretreated absorbent cotton gauze obtained in the step (a) in a KOH solution with the mass percentage of 20%, wherein the weight ratio of the absorbent cotton gauze to the KOH solution is as follows: absorbent cotton gauze: KOH solution ═ 1: 55, using acetone as a diluent to carry out an alkalization reaction, and reacting for 6 hours at the temperature of 37 ℃ under the stirring or ultrasonic vibration condition to obtain the alkalized absorbent cotton gauze;
(c) and (3) etherification treatment: adding chloroacetic acid into the alkalized absorbent cotton gauze obtained in the step (b) for etherification, wherein the weight ratio of the alkalized absorbent cotton gauze to the chloroacetic acid is as follows: and (3) alkalizing the degreased cotton gauze: 1:100 of chloroacetic acid, reacting for 3 hours at the temperature rising rate of 2 ℃ and the temperature rising temperature of 65 ℃, continuing to heat to 85 ℃, and keeping the temperature for 2 hours to obtain the cotton wool gauze subjected to etherification treatment;
(d) alcohol washing and neutralization: washing the etherified absorbent cotton gauze obtained in the step (c) for multiple times by using methanol, and simultaneously dropwise adding glacial acetic acid to adjust the pH value to 6.5-7.5;

In one embodiment of the absorbable healing promoting hemostatic composition of the present invention, the absorbable healing promoting hemostatic composition of the present embodiment comprises the following components: 4 mug of silver nitrate, 2g of vitamin K, 0.8g of vitamin E and 0.07g of epidermal growth factor.
An absorbable healing promoting hemostatic dressing comprising an absorbent substrate and the absorbable healing promoting hemostatic composition of the present embodiment; the absorbable heal-promoting hemostatic composition is attached to an adsorption substrate, the adsorption substrate is carboxymethyl cellulose potassium salt gauze, and the weight percentage of the absorbable heal-promoting hemostatic composition in the absorbable heal-promoting hemostatic dressing is 2%.
The absorbable wound dressing for promoting healing and hemostasis described in this example was prepared as described in example 1.
Example 5
In one embodiment of the absorbable healing promoting hemostatic composition of the present invention, the absorbable healing promoting hemostatic composition of the present embodiment comprises the following components: 6 mug of silver nitrate, 3g of vitamin K, 1.5g of vitamin E and 0.09g of epidermal growth factor.
An absorbable healing promoting hemostatic dressing comprising an absorbent substrate and the absorbable healing promoting hemostatic composition of the present embodiment; the absorbable heal-promoting hemostatic composition is attached to an adsorption substrate, the adsorption substrate is carboxymethyl cellulose potassium salt gauze, and the weight percentage of the absorbable heal-promoting hemostatic composition in the absorbable heal-promoting hemostatic dressing is 2%.
The absorbable wound dressing for promoting healing and hemostasis described in this example was prepared as described in example 1.
Example 6
The cytotoxicity detection of the absorbable healing-promoting hemostatic dressing of embodiments 1-5 comprises the following steps:
(1) preparing each gauze leaching liquor: leaching the absorbable healing-promoting hemostatic dressing and the instant yarn (American well-being) in the embodiments 1-5 according to 0.1g/mL after being saturated by a DMEM culture medium which is used for leaching and absorbing 10% fetal calf serum, and leaching for 24 hours at 37 ℃, wherein the leaching liquor is used as a test sample group;
(2) cell preparation: the frozen L929 cells were recovered and transferred to 2 generations for preparation of 1X 104Adding each/mL cell suspension into a 96-well plate, and culturing for 24h in a 5% carbon dioxide incubator at 37 ℃, wherein each well comprises 200 mu L of cell suspension;
(3) sample adding: discarding original culture solution from cultured cells, washing dead cells with PBS, adding 200 μ L of test sample into each well, and paralleling each sample;
(4) and (3) detection: and (3) placing the mixture in an incubator with 37 ℃ and 5% carbon dioxide, culturing for 24 hours, and detecting cytotoxicity by adopting an MTT method.
The test results are shown in table 1.

As can be seen from the test results in Table 1, the absorbable accelerated healing hemostatic dressings and the instant yarns described in examples 1 to 5 are non-toxic, and the absorbable accelerated healing hemostatic dressings described in examples 1 to 5 can promote cell growth, wherein the effect of example 1 is the most significant.
Experimental example 7
The hemostatic time detection method of the absorbable healing-promoting hemostatic dressing comprises the following specific operation steps:
(1) selecting 12 healthy New Zealand white rabbits, disinfecting ear marginal veins, slowly injecting a 3% sodium pentobarbital solution, and carrying out an experiment after the rabbits are completely anesthetized;
(2) selecting the central aorta of the rabbit ears, disinfecting the artery of the rabbit ears by iodophor, and then using a scalpel as a wound surface of 1 multiplied by 1 cm;
(3) after the ear artery of the rabbit bleeds, blood is absorbed by using common sterile gauze, and the absorbable healing-promoting hemostatic dressing prepared in the embodiment 1-5 is cut into 2 x 2cm and applied to the bleeding wound surface;
(4) absorbing exuded blood of the wound surface by using common sterile gauze every 20s, and observing the hemostatic effect;
(5) three parallel experiments were performed with each dressing using speed-instant yarn (american hadamard) as a control, and the time to complete hemostasis was recorded separately and the average calculated.
The test results are shown in table 2.

As can be seen from the results in Table 2, the hemostatic time of the absorbable accelerated healing hemostatic dressings of examples 1-5 is less than that of the immediate healing hemostatic dressings and significantly different (P <0.05), and the hemostatic time of example 1 is the shortest.
Experimental example 8
The method for detecting the degradation speed of the absorbable healing-promoting hemostatic dressing comprises the following specific operation steps:
(1) selecting 12 healthy New Zealand white rabbits, disinfecting ear marginal veins, slowly injecting a 3% sodium pentobarbital solution, and carrying out an experiment after the rabbits are completely anesthetized;
(2) selecting the central aorta of the ear of the rabbit, shaving and disinfecting the thigh part of the rabbit by iodophor, and then using a scalpel as a wound surface of 1 cm;
(3) after bleeding, absorbing blood by using common sterile gauze, cutting the absorbable healing-promoting hemostatic dressing of the embodiment 1-5 into 1 x 2cm strips, placing the strips in the wound, and suturing the wound;
(4) observing the change of the abdominal wound every day, removing stitches after 8 days, and observing the degradation condition of each gauze.
(5) Three parallel experiments were performed for each dressing using speed, i.e. gauze (hadamard usa) as a control, and the remaining amount of each dressing was recorded separately and the average was calculated.
The test results are shown in table 3.

The results in table 3 show that the absorbable healing-promoting hemostatic dressings in examples 1 to 5 have a degradation rate greater than the speed, i.e., the difference between the degradation rates is significant (P <0.05), and the degradation rate of example 1 is the fastest.
Example 9
In order to explore the proportion of the absorbable healing promoting and hemostatic composition and the effects of promoting wound healing and hemostasis, 17 test groups and 4 control groups in table 4 are arranged, the absorbable healing promoting and hemostatic compositions of the test groups 1-17 and the control groups 1-4 are prepared into the absorbable healing promoting and hemostatic dressing according to the preparation method of the absorbable healing promoting and hemostatic dressing described in embodiment 1, the sum of the total amounts of the absorbable healing promoting and hemostatic compositions of the test groups 1-17 and the control groups 1-4 is the same as that of embodiment 1, the effects of promoting wound healing and hemostasis of the test groups 1-17 and the control groups 1-4 are tested, the method for testing the effect of promoting wound healing is the same as that of embodiment 6, and the method for testing the hemostatic effect is the same as that of embodiment 7.

From the test results in table 5, it can be seen that when the absorbable composition for promoting healing and stopping bleeding adopts the mixture ratio in test group 1, the cell growth rate and the stopping bleeding rate are the best, and the cell growth rate and the stopping bleeding rate of test groups 3, 4, 7, 8, 11, 12, 15, 16 are superior to those of test groups 2, 5, 6, 9, 10, 14, 17, and are all superior to those of controls 1-4, which indicates that the absorbable composition for promoting healing and stopping bleeding can better exert the effects of promoting healing and stopping bleeding and resisting bacteria only under a certain mixture ratio.
Example 10
In order to explore the effect of the content of the absorbable healing promoting and hemostatic composition in the dressing on promoting wound healing and hemostasis, 5 test groups and 2 control groups in table 6 are set, the absorbable healing promoting and hemostatic compositions of the test groups 1-5 and the control groups 1 and 2 are prepared into the absorbable healing promoting and hemostatic dressing according to the composition and the preparation method of the embodiment 1, the effect of promoting wound healing and hemostasis of the test groups 1-5 and the control groups 1 and 2 is tested, the test method of the effect of promoting wound healing is the same as that of the embodiment 6, and the test method of the hemostatic effect is the same as that of the embodiment 7.

From the results in table 6, it can be seen that the absorbable pro-healing hemostatic composition is effective in shortening the hemostatic time and promoting wound healing when the content of the absorbable pro-healing hemostatic composition in the dressing is 0.1% by weight, and the hemostatic speed and the healing promoting effect are increased with the increase of the added amount of the absorbable pro-healing hemostatic composition, but the content of the absorbable pro-healing hemostatic composition is increased more than 2% in the dressing, and the improvement of the hemostatic speed and the wound healing promoting effect is not significant.
Finally, it should be noted that the above embodiments are only used for illustrating the technical solutions of the present invention and not for limiting the protection scope of the present invention, and although the present invention is described in detail with reference to the preferred embodiments, it should be understood by those skilled in the art that modifications or equivalent substitutions can be made on the technical solutions of the present invention without departing from the spirit and scope of the technical solutions of the present invention.
Claims (1)
1. An absorbable healing promoting hemostatic dressing, comprising an absorbent substrate and an absorbable healing promoting hemostatic composition; the absorbable healing promoting hemostatic composition is attached to an absorbent substrate;
the absorbable healing promoting hemostatic composition comprises the following components in parts by weight: silver nitrate 5X 10-6Parts of vitamin K2.5 parts, vitamin E1 part and epidermal growth factor 0.08 part;
the absorbable healing promoting hemostatic composition accounts for 2 percent by weight of the absorbable healing promoting hemostatic dressing; the adsorption base material is carboxymethyl cellulose sylvite gauze;
the preparation method of the absorbable wound dressing capable of promoting healing and hemostasis comprises the following steps:
(1) preparing an absorbable healing-promoting hemostatic composition solution: adding the absorbable healing promoting hemostatic composition into double distilled water;
(2) soaking the adsorption substrate in the absorbable healing-promoting and hemostatic composition solution for 1-3 hours;
(3) taking out, cleaning and drying to obtain the absorbable healing-promoting hemostatic dressing;
the preparation method of the carboxymethyl cellulose potassium salt gauze comprises the following steps:
(a) and (3) pretreatment: washing absorbent cotton gauze in double-distilled water at the temperature of 55-85 ℃ for 1-4 h to obtain pretreated absorbent cotton gauze;
(b) and alkalization treatment: performing alkalization treatment on the pretreated absorbent cotton gauze obtained in the step (a) to obtain alkalized absorbent cotton gauze;
(c) and (3) etherification treatment: adding chloroacetic acid into the alkalized absorbent cotton gauze obtained in the step (b) for etherification to obtain the etherified absorbent cotton gauze;
(d) alcohol washing and neutralization: washing the etherified absorbent cotton gauze obtained in the step (c) for multiple times by using methanol, and simultaneously dropwise adding glacial acetic acid to adjust the pH value to 6.5-7.5;
(f) freezing and drying the gauze treated in the step (d) to obtain carboxymethyl cellulose sylvite gauze;
in the step (b), the alkalization process comprises the following steps: soaking the pretreated absorbent cotton gauze in a KOH solution with the mass percentage of 10-20%, wherein the weight ratio of the absorbent cotton gauze to the KOH solution is as follows: absorbent cotton gauze: KOH solution =1: 40-55, using acetone as a diluent to carry out an alkalization reaction, and reacting for 3-6 hours at the temperature of-20-37 ℃ under the stirring or ultrasonic vibration condition to obtain the alkalized absorbent cotton gauze;
in the step (c), the weight ratio of the degreased cotton gauze subjected to the alkalization treatment to the chloroacetic acid is as follows: and (3) alkalizing the degreased cotton gauze: chloroacetic acid =1: 60-100, and the reaction conditions of the etherification treatment are as follows: reacting for 2-3 h when the temperature rise rate is 1-2 ℃ and the temperature rises to 65 ℃, continuously raising the temperature to 85 ℃, and keeping the temperature constant for 1-2 h.
