技术领域
本发明涉及中药技术领域,特别是涉及一种促进伤口愈合的组合物及其制备方法和应用。
背景技术
伤口愈合是一个高度动态的过程,涉及复杂的细胞和生化过程。在成年哺乳动物中,早期炎症阶段以嗜中性粒细胞和巨噬细胞浸润为特征,随后,富含未成熟胶原束和新形成的血管的纤维增生组织形成,并且发生迅速的再上皮化。最后,发生真皮胶原重塑和瘢痕出芽,至此常规的组织修复完成。
在这些过程中存在许多影响伤口愈合的因素:伤口炎性白细胞可能几乎干扰组织修复的所有阶段;中性粒细胞和炎性常规活化的巨噬细胞则可能损害伤口愈合。另一方面,巨噬细胞可以从促炎表型转化成抗炎/血管生成/愈合表型,从而发挥抗炎和促进愈合作用。有研究表明,在没有与微生物群接触的情况下,皮肤伤口愈合加速并且不产生疤痕。已经有大量研究表明外用中药可激活局部的巨噬细胞、对白细胞有化学趋化作用、能增加局部免疫力、调节基质代谢、促进局部的微循环,从而起到抗炎和抑菌的作用,最终可促进伤口愈合,减少疤痕组织生成。
因此,利用我国传统中医中药资源,开发作用明确传统药物产品符合大众需求并具有极为重要的现实意义和社会意义。
发明内容
本发明的目的,就是克服现有技术的不足,提供一种促进伤口愈合的组合物及其制备方法和应用,该组合物具有较好的促进伤口愈合作用。
为了达到上述目的,采用如下技术方案:
一种促进伤口愈合的组合物,所述组合物的有效成分由以下重量份的药材制备而成:
上述组合物,选用鹿花盘、蒲公英、乳香、没药和薄荷脑相互配合。其中,鹿花盘镇痛散瘀、软坚散结;蒲公英清热解毒,活血化瘀;乳香没药为一药对,活血止痛,消肿生肌;且利用薄荷脑经皮促进吸收的活性,可提高其他药物的经皮透过率,同时还能起到止痛的效果。多味药物共同作用,起到了非常好的促进皮肤伤口愈合的效果。

鹿花盘提取:将鹿花盘以水为溶剂,加热提取,即得鹿花盘提取物;
蒲公英提取:将蒲公英榨汁,即得蒲公英提取物;
乳香和没药提取:取乳香和没药混合,以水为溶剂,加热提取,即得乳香没药提取物;
混合:将上述鹿花盘提取物、蒲公英提取物、乳香没药提取物和薄荷脑混合均匀,即得。
在其中一个实施例中,所述鹿花盘提取步骤中,每次按照1g药材:15-25ml水的料液比加入水,在110-130℃,0.09-0.10MPa的条件下提取1-3次,每次0.2-1hr。
在其中一个实施例中,所述蒲公英提取步骤中,取蒲公英鲜品榨汁。
在其中一个实施例中,所述乳香和没药提取步骤中,每次按照1g药材:5-15ml水的料液比加入水,在80-100℃下回流提取1-3次,每次1-3hr。
在其中一个实施例中,所述鹿花盘提取物和乳香没药提取物均回收溶剂至浓度为1ml/g药材,所述蒲公英提取物加入水制成浓度为1ml/g蒲公英。

以下实施例所用原料,均为市售购得。
实施例1
一种促进伤口愈合的组合物,通过以下方法制备得到:
1、原料。
取鹿花盘362g(1份)、蒲公英362g(1份)、乳香181g(0.5份)、没药181g(0.5份)和薄荷脑36g(0.1份),备用。
2、制备。
2.1、鹿花盘提取:将鹿花盘加入蒸馏水(1g:20ml水),121℃、0.098MPa高压提取半小时,提取2次,收集滤液,减压浓缩至1ml/g鹿花盘,即得鹿花盘提取物。
2.2、蒲公英提取:取蒲公英鲜品榨汁,加蒸馏水制成1ml/g蒲公英,即得蒲公英提取物。
2.3、乳香和没药提取:取乳香和没药混合,加入蒸馏水(1g:10ml水),浸泡过夜,80-100℃回流提取3次,每次2小时,收集滤液,减压浓缩至1ml/g药材。
2.4、混合:将上述浓度均为1ml/g的鹿花盘提取物、蒲公英提取物、乳香没药提取物和36g薄荷脑混合均匀,即得促进伤口愈合的组合物。
实施例2
一种促进伤口愈合的组合物,与实施例1的组合物基本相同,区别仅在于该组合物的药材组成如下:鹿花盘112g(0.5份)、蒲公英336g(1.5份)、乳香66g(0.3份)、没药154g(0.7份)和薄荷脑22g(0.1份)。
对比例1
一种中药组合物,其原料为:黄芪30克、当归12克、白芷10克、鹿角20克、蒲公英15克和甘草4克。
制法:将上述药材按照药液比为1g药材20ml水加入适量水,浸泡30分钟,煮沸后再用温火煎20分钟,滤液浓缩至1ml/g药材。

一种中药组合物,与实施例1的组合物基本相同,区别仅在于处方中缺少薄荷脑。
实验例
取上述实施例和对比例制备得到的组合物进行伤口愈合实验。
1、方法。
选取体重为20.0±2.0g的SPF级C57BL/6雄性小鼠日常饲养,观察一周,无死亡和精神异常出现。
将小鼠随机分为空白组和5组给药组,每组6只。背部皮肤脱毛后,用0.5mg/g戊巴比妥钠麻醉,用打孔器在鼠背部打一个圆孔,切取直径为10.0mm的全层皮肤,止血后以无菌纱布包扎,单笼喂养。
空白组给予凡士林:生理盐水:吐温80(1g:1g:0.02g)的混合物涂敷伤口;给药组给予凡士林:药液:吐温80(1g:1g:0.02g)的混合药物涂敷伤口。每天给药1次,连续给药4周,计算愈合率。
愈合率=[(原始创面面积-未愈合创面面积)÷原始创面面积]×100%。

注:*表示经统计学分析,与同时间空白组比P<0.05。
通过上述实验可以看出,实施例1和2的组合物具有较好的促进伤口愈合效果,且实施例1的组合物具有最佳的促进愈合效果。而对比例1-3的组合物,虽也有一定效果,但其愈合率不如实施例1-2。

2.权利要求1所述的促进伤口愈合的组合物的制备方法,其特征在于,包括以下步骤:
鹿花盘提取:将鹿花盘以水为溶剂,加热提取,即得鹿花盘提取物;
蒲公英提取:将蒲公英榨汁,即得蒲公英提取物;
乳香和没药提取:取乳香和没药混合,以水为溶剂,加热提取,即得乳香没药提取物;
混合:将上述鹿花盘提取物、蒲公英提取物、乳香没药提取物和薄荷脑混合均匀,即得。
3.根据权利要求2所述的促进伤口愈合的组合物的制备方法,其特征在于:所述鹿花盘提取步骤中,每次按照1g药材:15-25ml水的料液比加入水,在110-130℃,0.09-0.10MPa的条件下提取1-3次,每次0.2-1hr。
4.根据权利要求2所述的促进伤口愈合的组合物的制备方法,其特征在于:所述蒲公英提取步骤中,取蒲公英鲜品榨汁。
5.根据权利要求3所述的促进伤口愈合的组合物的制备方法,其特征在于:所述乳香和没药提取步骤中,每次按照1g药材:5-15ml水的料液比加入水,在80-100℃下回流提取1-3次,每次1-3hr。
6.根据权利要求1所述的促进伤口愈合的组合物的制备方法,其特征在于:所述鹿花盘提取物和乳香没药提取物均回收溶剂至浓度为1ml/g药材,所述蒲公英提取物加入水制成浓度为1ml/g蒲公英。
7.权利要求1所述的组合物在制备促进伤口愈合的药物中的应用。
8.根据权利要求7所述的应用,其特征在于,所述药物为外用。
9.根据权利要求7所述的应用,其特征在于,所述药物的剂型为酊剂、巴布剂、凝胶剂、膏剂或喷剂。
