Abstract
本发明涉及饲料添加剂技术领域,尤其涉及一种缓解仔猪免疫应激的饲料添加剂及其制备方法和应用。所述缓解仔猪免疫应激的饲料添加剂,由3.0份复方中草药、2.0份谷氨酰胺和1.0份茶多酚组成;所述复方中草药,由以下中药材组成:知母20份、茅香12份、冰片12份、赤芍18份、龙骨14份、生地黄18份、金银花25份、龙胆草8份、鱼腥草20份、穿心莲12份、刺五加12份、川芎10份、党参8份、桔梗15份、三七8份。所述添加剂能够缓解仔猪正常疫苗免疫导致免疫应激,提高免疫应激期仔猪日采食量和日增量,降低料肉比。

技术领域
本发明涉及饲料添加剂技术领域,尤其涉及一种缓解仔猪免疫应激的饲料添加剂及其制备方法和应用。
背景技术
免疫应激又称免疫激发,在免疫学中指注入抗原以激发免疫应答。在动物生命过程中,有许多因素包括各种感染、创伤和内部肿瘤等都可引起免疫应激。猪的生长过程中常常遇到病原性或非病原性微生物、疫苗和异源蛋白等刺激而处于免疫应激状态,影响猪的生产性能和健康状况。许多研究证实,免疫应激使畜禽采食量减少,饲料转化率降低,上市时间延缓,蛋白质沉积减少,胴体瘦肉率降低,眼肌面积减少。如疫苗免疫是为了预防猪特定的疾病,给猪注射的低活性或灭活的病毒或细菌,但是疫苗免疫也会导致猪产生免疫应激,降低猪的生产性能,如何缓解猪的免疫应激,提高猪免疫应激期的生产性能,对养猪生产具有重大的意义。
中国专利CN106880733A中公开了一种降低断奶仔猪应激的添加剂(断奶保护散),CN103315184B公开了一种缓减运输应激对仔猪生长产生不利影响的饲料添加剂,本领域尚未有公开缓解仔猪免疫应激的饲料添加剂。
发明内容
针对现有技术中的不足之处,本发明所述一种缓解仔猪免疫应激的饲料添加剂及其制备方法和应用,所述添加剂能够缓解仔猪正常疫苗免疫导致免疫应激,提高免疫应激期仔猪日采食量和日增量,降低料肉比。
为了实现上述目的,本发明提供如下技术方案:
一种缓解仔猪免疫应激的饲料添加剂,按重量份计,所述饲料添加剂的原料包括:
2.5份~3.5份复方中草药;
0.6份~1.5份茶多酚;
1.0份~5.0份谷氨酰胺或谷氨酰胺衍生物;
所述复方中草药,按重量份计包括:知母15份~25份、冰片8份~15份、龙骨10份~18份、生地黄15份~20份、金银花20份~30份、龙胆草5份~10份、鱼腥草15份~25份、穿心莲10份~15份和刺五加10份~15份。
进一步地,所述复方中草药,按重量份计包括:3.0份复方中草药、0.6份~1.0份茶多酚和1.0份~5.0份谷氨酰胺或谷氨酰胺衍生物。
进一步地,所述饲料添加剂,按重量份计包括:3.0份复方中草药、1.0份茶多酚和2.0份谷氨酰胺。
进一步地,所述复方中草药,按重量份计还包括:茅香10份~15份、赤芍15份~20份、川芎5份~15份、党参5份~10份、桔梗10份~20份和三七5份~10份。
在本发明的一个优选实施方式中,所述复方中草药,按重量份计包括:知母20份、茅香12份、冰片12份、赤芍18份、龙骨14份、生地黄18份、金银花25份、龙胆草8份、鱼腥草20份、穿心莲12份、刺五加12份、川芎10份、党参8份、桔梗15份和三七8份。
进一步地,所述谷氨酰胺衍生物为丙氨酰谷氨酰胺或甘氨酰谷氨酰胺。
在本发明中,所述谷氨酰胺衍生物可以是能够在动物体内转化为谷氨酰胺的化合物,或在结构上与谷氨酰胺结构类似并与谷氨酰胺具有类似效果的化合物。
本发明还提供所述一种缓解仔猪免疫应激的饲料添加剂的制备方法,包括以下步骤:
(1)将复方中草药的各组分分别进行超微粉碎后过100目筛混匀得到复方中草药混合物;
(2)向复方中草药混合物中加入茶多酚,以及谷氨酰胺或谷氨酰胺衍生物,混匀即得。
进一步地,所述方法还包括将复方中草药的各组分在超微粉碎之前进行第一次干燥,干燥温度是60℃~70℃。第一次干燥的目的在于,除去中药材浅表的水分。
进一步地,所述方法还包括将复方中草药混合物置于隧道式微波干燥箱进行第二次干燥;所述第二次干燥的工艺参数:微波频率为2400MHz~2500MHz,干燥温度为60℃~100℃,干燥时间为2min~5min。
本发明所述制备方法首先对各复方中草药进行超微粉碎得到较小粒径的中药材细粉颗粒,将混匀之后的复方中草药混合物置于隧道式微波干燥箱进行第二次干燥,由于微波加热时传热和传质方向一致,形成的内部压力梯度使水分能扩散到表面而蒸发,能进一步降低中药材原料的含水量,所制得饲料添加剂的保藏时间长;并且能够缓和所述中药材原料的药性,使得药性偏于缓和,适合仔猪食用;此外,中药材原料中的难闻气味主要由挥发油形成,由于水是强极性分子,微波干燥过程中首先受热汽化产生大量的水蒸气,产生巨大的能量,有利于挥发油的逸出,减轻中药材原料的难闻药味,提高所述饲料添加剂的适口性。不会因为药味影响仔猪的摄食量。
本发明还提供所述饲料添加剂用于制备缓解仔猪免疫应激的饲料中的应用,按重量计,所述饲料添加剂在饲料中的添加比例为0.5%~1.0%。本发明的饲料添加剂作为饲料添加剂可以添加到任何类型的仔猪饲料中。
本发明所述饲料添加剂中各组分的药理作用如下:
茶多酚:是茶叶中多酚类物质的总称,包括黄烷醇类、花色苷类、黄酮类、黄酮醇类和酚酸类等。主要为黄烷醇(儿茶素)类,儿茶素占60~80%。类物质茶多酚又称茶鞣或茶单宁,是形成茶叶色香味的主要成份之一,也是茶叶中有保健功能的主要成份之一。在体内具有抗氧化的作用,能够减少细胞因免疫应激引起的氧化对细胞的损伤,显著提升动物应激期的免疫能力。
谷氨酰胺:学名2-氨基-4-氨甲酰基丁酸,英文Glutamine(Gln)。是谷氨酸的酰胺,可用于治疗胃及十二指肠溃疡、胃炎及胃酸过多,也用于改善脑功能。还具有重要的免疫调节作用,它是淋巴细胞分泌、增殖及其功能维持所必需的。作为核酸生物合成的前体和主要能源,谷氨酰胺可促使淋巴细胞、巨噬细胞的有丝分裂和分化增殖,增加细胞因子TNF、IL-1等的产生和磷脂的mRNA合成;参与合成谷胱甘肽,提高机体抗氧化能力;肠道免疫重要营养物质,胃肠道管腔细胞的基本能量来源,维持肠道屏障的结构及功能。
知母:单子叶植物百合科知母的干燥根茎。具有清热泻火,滋阴润燥的功效,研究表明,知母使动物的心率减慢,尿中肾上腺素排出量减少,使参与合成儿茶酚胺的多巴胺p羟化酶(DBH)活性降低,并可使尿内分泌17OHCS排出量减少,氧消耗降低,使交感肾上腺神经的兴奋性降低。
冰片:又名片脑、桔片、艾片、龙脑香、梅花冰片、羯布罗香、梅花脑、冰片脑、梅冰等,是由菊科艾纳香茎叶或樟科植物龙脑樟枝叶经水蒸汽蒸馏并重结晶而得。为开窍药物,能够增强抑制状态。
龙骨:主含羟基磷酸钙(羟磷灰石);尚含少量碳酸钙及铁、铝、镁、锰、锶等元素,能调整中枢神经的兴奋状态,持重性降,具有镇定安神作用。
生地黄:生地黄为玄参科植物地黄的块根,现代药理证明生地黄能提高T淋巴细胞数,并增强其功能,清热药物,具有良好的抗感染作用。
金银花:为忍冬科忍冬属植物忍冬及同属植物干燥花蕾或带初开的花,能够促进淋巴细胞的转化。
龙胆草:为龙胆科多年生草本植物条叶龙胆等的根和茎,味苦,性寒,功效清热燥湿,能够增强吞噬细胞的功能。
鱼腥草:为三白草科植物蕺菜的干燥地上部分,具有促进白细胞的吞噬能力。
穿心莲:又名春莲秋柳,一见喜,榄核莲、苦胆草、金香草、金耳钩、印度草,苦草等。药理证明具有增强溶菌酶的活性的功效。
刺五加:别名坎拐棒子、老虎潦、一百针。药理证明具有促进网状内皮系统的吞噬功能,增强其吞噬异物和抗原的能力,并提高血清中抗体的浓度。
茅香:禾本科茅香属植物茅香的根状茎入药,为寒性药,具有平肝息风的功效。
赤芍:为毛茛科植物赤芍或川赤芍的干燥根,具有清热凉血,散瘀止痛的功效,能增强体液免疫功能。
川芎:为活血化瘀药,可改善微循环,增加血液流速,改善微循环血管形态,解除微血管痉挛,减轻循环内红细胞的瘀滞和汇集,使微血管通透性降低,微血管周围渗出减少。
党参:为桔梗科植物党参、素花党参、或川党参等的干燥根,具有补中,益气,生津的功效,可起到迅速保护粘膜的作用,使刺激原无法刺激粘膜。
桔梗:为桔梗科植物桔梗的干燥根,具有宣肺,利咽,祛痰,排脓的功效,现代药理认为具有抗炎的作用能够降低毛细血管的通透性,抑制浆液渗出,促进组织的再生和伤口的愈合。
三七:为五加科植物三七的干燥根和根茎。具有散瘀止血,消肿定痛的功效,从药理上看,能够减少渗出和白细胞向炎灶聚集,抑制水肿,抑制透明脂酸酶的活性,从而起到抗炎作用。
本发明与现有技术相比,具有的有益效果如下:本发明所述缓解仔猪免疫应激的饲料添加剂,采用茶多酚、谷氨酰胺与传统中药材配合使用起到协同增效的作用。其中,茶多酚能够减少免疫应激引起的氧化物质对细胞的损害,谷氨酰胺的配合使用能够提高茶多酚的抗氧化性能;复方中草药中知母能够使动物的心率减慢,使尿中肾上腺素排出量减少,配合冰片、龙骨使用可以增强这种抑制状态,减少仔猪对免疫应激的反应;生地黄能提高T淋巴细胞数,并增强其功能,清热药物,具有良好的抗感染作用,配合金银花、龙胆草、鱼腥草、穿心莲和刺五加使用能够显著提高仔猪的免疫能力,有利于降低免疫导致的不良反应;川芎、党参、桔梗和三七的配合可以起到保护胃肠道粘膜和抗炎的作用,所述茶多酚、谷氨酰胺和复方中草药的配伍能非常有效地缓解仔猪正常疫苗免疫导致的免疫应激,提高免疫应激期仔猪日采食量和日增量,降低料肉比。
具体实施方式
本发明公开了一种缓解仔猪免疫应激的饲料添加剂及其制备方法和应用,本领域技术人员可以借鉴本文内容,适当改进工艺参数实现。特别需要指出的是,所有类似的替换和改动对本领域技术人员来说是显而易见的,它们都被视为包括在本发明。本发明的方法及应用已经通过较佳实施例进行了描述,相关人员明显能在不脱离本发明内容、精神和范围内对本文所述的方法和应用进行改动或适当变更与组合,来实现和应用本发明技术。
下述实施例中的实验方法,如无特别说明,均为常规方法。下述实施例涉及的原料若无特别说明,均为普通市售品,皆可通过市场购买获得。
实施例1:一种缓解仔猪免疫应激的饲料添加剂,以重量份计,由3.0份复方中草药、2.0份谷氨酰胺和1.0份茶多酚组成;其中复方中草药,由以下重量份的各组分组成:知母20份、茅香12份、冰片12份、赤芍18份、龙骨14份、生地黄18份、金银花25份、龙胆草8份、鱼腥草20份、穿心莲12份、刺五加12份、川芎10份、党参8份、桔梗15份、三七8份。
制备方法如下:将上述复方中草药的各组分分别置于65℃烘箱中进行干燥,然后再分别进行超微粉碎后过100目筛混匀得到复方中草药混合物,将该复方中草药混合物置于隧道式微波干燥箱进行干燥,所述微波频率为2400MHz,干燥温度为60℃~100℃,干燥时间为4min。
实施例2:一种缓解仔猪免疫应激的饲料添加剂,以重量份计,由2.5份复方中草药、2.0份谷氨酰胺和1.5份茶多酚组成;其中复方中草药,由以下重量份的各组分组成:知母15份、茅香10份、冰片12份、赤芍20份、龙骨10份、生地黄15份、金银花20份、龙胆草5份、鱼腥草20份、穿心莲10份、刺五加10份、川芎5份、党参5份、桔梗20份、三七10份。
制备方法如下:将上述复方中草药的各组分分别置于60℃烘箱中进行干燥,然后再分别进行超微粉碎后过100目筛混匀得到复方中草药混合物,将该复方中草药混合物置于隧道式微波干燥箱进行干燥,所述微波频率为2500MHz,干燥温度为60℃,干燥时间为2min。
实施例3:一种缓解仔猪免疫应激的饲料添加剂,以重量份计,由3.0份复方中草药、5.0份谷氨酰胺和0.6份茶多酚组成;其中复方中草药,由以下重量份的各组分组成:知母25份、茅香15份、冰片15份、赤芍15份、龙骨18份、生地黄18份、金银花25份、龙胆草8份、鱼腥草20份、穿心莲15份、刺五加12份、川芎15份、党参10份、桔梗15份、三七10份。
制备方法如下:将上述复方中草药的各组分分别置于70℃烘箱中进行干燥,然后再分别进行超微粉碎后过100目筛混匀得到复方中草药混合物,将该复方中草药混合物置于隧道式微波干燥箱进行干燥,所述微波频率为2400MHz,干燥温度为80℃,干燥时间为5min。
实施例4:一种缓解仔猪免疫应激的饲料添加剂,以重量份计,由3.5份复方中草药、3.0份谷氨酰胺和1.0份茶多酚组成;其中复方中草药,由以下重量份的各组分组成:知母20份、茅香12份、冰片10份、赤芍17份、龙骨10份、生地黄20份、金银花30份、龙胆草10份、鱼腥草15份、穿心莲13份、刺五加10份、川芎14份、党参5份、桔梗20份、三七7份。制备方法同实施例1。
实施例5:一种缓解仔猪免疫应激的饲料添加剂,以重量份计,由3.0份复方中草药、4.0份谷氨酰胺和0.8份茶多酚组成;其中复方中草药,由以下重量份的各组分组成:知母18份、茅香12份、冰片8份、赤芍15份、龙骨18份、生地黄20份、金银花24份、龙胆草7份、鱼腥草18份、穿心莲10份、刺五加13份、川芎7份、党参5份、桔梗13份、三七8份。制备方法同实施例1。
实施例6:一种缓解仔猪免疫应激的饲料添加剂,以重量份计,由2.5份复方中草药、1.0份谷氨酰胺和1.5份茶多酚组成;其中复方中草药,由以下重量份的各组分组成:知母15份、茅香10份、冰片10份、赤芍20份、龙骨15份、生地黄15份、金银花20份、龙胆草5份、鱼腥草25份、穿心莲12份、刺五加15份、川芎5份、党参8份、桔梗10份、三七5份。制备方法同实施例1。
实施例7:通过以下实验例来验证本发明所述饲料添加剂用于缓解仔猪免疫应激的作用:
1.试验设计
选择24日龄断奶的外三元“杜长大”仔猪300头,按照体重相近,性别一致原则,随机分为5个处理组,分别为空白组(处理1),模型组(处理2)、实验组(处理3)、对照组1(处理4)和对照组2(处理5),每个处理组6个重复,每个重复10头猪,试验期30天。本实验采用猪场经常使用、免疫应激反映较明显的口蹄疫和猪用三联苗(猪瘟、猪丹毒、猪肺疫三联苗)作为免疫应激源:模型组和实验组、对照组1和对照组2,于试验第10天免疫口蹄疫疫苗2头份/头,试验第20天免疫三联苗2头份/头,空白组不注射疫苗。试验期间饲养管理程序参照常规的商品猪饲养管理手册进行。各处理组的日粮如下:
空白组:以普通饲料作为日粮。
模型组:以普通饲料作为日粮。
实验组:在普通饲料中添加0.6%本发明实施例1所述饲料添加剂作为日粮。
对照组1:在普通饲料中添加0.3%复方中草药+0.3%玉米(替代茶多酚和谷氨酰胺)作为日粮,该复方中草药,按重量份计,由知母20份、茅香12份、冰片12份、赤芍18份、龙骨14份、生地黄18份、金银花25份、龙胆草8份、鱼腥草20份、穿心莲12份、刺五加12份、川芎10份、党参8份、桔梗15份、三七8份组成。
对照组2:按照每吨普通饲料添加0.2%谷氨酰胺+0.1%茶多酚+0.3%玉米(替代复方中草药)作为日粮。
2.检测指标
生产性能:分别于试验0天,30天各组进行称重,计算各组猪平均日增重(ADG);试验结束时统计各组仔猪耗料量,计算平均日采食量(ADFI)和料肉比。
3.数据处理
采用SAS 9.0(2001)中ANOVA分析试验数据,并用DUNCAN氏方法进行多重比较,显著水平P=0.05。
4.试验结果
不同处理对仔猪生产性能的影响见表1

注:不同小写字母表示差异显著,不同大写字母表示差异极显著。
5.结果分析
由表1中数据可知,处理2日增重和日采食量极显著低于处理1,料肉比极显著高于处理1,说明疫苗免疫产生免疫应激,极显著降低了仔猪的生产性能。
从处理3日增重和日采食量极显著高于处理2,料肉比极显著低于处理1,同时处理3日增重极显著高于处理1,料肉比显著低于处理1,说明本发明所述饲料添加剂极显著提高了疫苗免疫应激期仔猪的生产性能。从处理3的日增重和日采食量看,分别显著高于处理4和处理5,料肉比显著低于处理4和处理5;说明处理3的使用效果优于处理4和处理5;也就是说采用本发明的方案将复方中草药、茶多酚和谷氨酰胺联合使用与单独使用复方中草药或者茶多酚和谷氨酰胺的组合相比具有更优的效果,复方中草药、茶多酚和谷氨酰胺联合使用对于缓解仔猪免疫应激能够发挥协同增效的作用。
6.结论
疫苗免疫,极显著降低了仔猪的生产性能;而在免疫应激期仔猪饲料中加入0.6%本发明的添加剂,极显著提高了免疫应激期仔猪的生产性能。
经实验验证,本发明实施例2-6所述饲料添加剂均能达到上述效果。
Claims (10)
1.一种缓解仔猪免疫应激的饲料添加剂,其特征在于:按重量份计,所述饲料添加剂的原料包括:
2.5份~3.5份复方中草药;
0.6份~1.5份茶多酚;
1.0份~5.0份谷氨酰胺或谷氨酰胺衍生物;
所述复方中草药,按重量份计,包括:知母15份~25份、冰片8份~15份、龙骨10份~18份、生地黄15份~20份、金银花20份~30份、龙胆草5份~10份、鱼腥草15份~25份、穿心莲10份~15份和刺五加10份~15份。
2.如权利要求1所述的饲料添加剂,其特征在于,按重量份计,包括:3.0份复方中草药、0.6份~1.0份茶多酚和1.0份~5.0份谷氨酰胺或谷氨酰胺衍生物。
3.如权利要求1所述的饲料添加剂,其特征在于,按重量份计,包括:3.0份复方中草药、1.0份茶多酚和2.0份谷氨酰胺。
4.如权利要求1所述一种缓解仔猪免疫应激的饲料添加剂,其特征在于,按重量份计,所述复方中草药还包括:茅香10份~15份、赤芍15份~20份、川芎5份~15份、党参5份~10份、桔梗10份~20份和三七5份~10份。
5.如权利要求4所述的饲料添加剂,其特征在于,按重量份计,所述复方中草药包括:知母20份、茅香12份、冰片12份、赤芍18份、龙骨14份、生地黄18份、金银花25份、龙胆草8份、鱼腥草20份、穿心莲12份、刺五加12份、川芎10份、党参8份、桔梗15份和三七8份。
6.如权利要求1-5任一项所述的饲料添加剂,其特征在于,所述谷氨酰胺衍生物为丙氨酰谷氨酰胺或甘氨酰谷氨酰胺。
7.权利要求1-6任一项所述一种缓解仔猪免疫应激的饲料添加剂的制备方法,其特征在于,包括以下步骤:
(1)将复方中草药的各组分分别进行超微粉碎后过100目筛混匀得到复方中草药混合物;
(2)向复方中草药混合物中加入茶多酚,以及谷氨酰胺或谷氨酰胺衍生物,混匀即得。
8.如权利要求7所述的制备方法,其特征在于,所述方法还包括将复方中草药的各组分在超微粉碎之前进行第一次干燥,干燥温度是60℃~70℃。
9.如权利要求7或8所述的制备方法,其特征在于,所述方法还包括将中草药混合物置于隧道式微波干燥箱进行第二次干燥;所述第二次干燥的工艺参数:微波频率为2400MHz~2500MHz,干燥温度为60℃~100℃,干燥时间为2min~5min。
10.权利要求1~6任一项所述饲料添加剂用于制备缓解仔猪免疫应激的饲料中的应用,其特征在于,按重量计,所述饲料添加剂在饲料中的添加比例为0.5%~1.0%。
