本发明涉及药品制作领域,具体是指一种枇杷止咳糖浆的制备方法,具体包括以下步骤:(1)将原料枇杷叶、罂粟壳、百部、白前、桑白皮、桔梗干燥后粉碎,混合,加水后冷藏;(2)冷藏后产物取出,加入表面活性剂,超声,得到提取物,过滤,将滤液经加热浓缩之后置于常温下保存,待用;(3)取蔗糖添加水,加热混合均匀后,加入枸橼酸、苯甲酸钠,得到蔗糖混合溶液;(4)将薄荷脑、香精溶解到乙醇中,得到乙醇溶液;(5)将提取物、蔗糖混合溶液、乙醇溶液混合,搅拌,静置,滤过,加水至全量,混匀,即得目标产物。本发明通过增加黄酮类物质的浓度以及控制表面活性剂的浓度达到抗菌,增溶作用,优化糖浆的质量和疗效;此外黄酮为糖浆中原有物质,而鼠李糖脂也被运用在食药行业,两者均不会对糖浆产生任何不良影响。

背景技术
糖浆剂系指药材提取物或芳香物质的浓蔗糖水溶液,是临床常用的剂型之一,因其制剂制备工艺简单,服用方便,显效迅速;另外,由于糖浆剂中所含的糖和芳香剂具有矫味的作用,能掩盖某些药物的苦咸等不适气味,改善口感,使药物容易服用,故深受广大医生患者欢迎,特别是儿童更容易接受。然而,在实际生产贮存过程中,由于原料提取物的溶解性问题以及蔗糖中引入的细菌问题,极易造成许多疗效确切的糖浆剂稳定性受到影响,进一步影响到糖浆剂的疗效和质量。
申请号为CN201610192731.X公开了一种止咳枇杷糖浆的制备方法,在此发明制备止咳枇杷糖浆的方法中,苦杏仁在第二次煎煮时才加入,避免其影响枇杷叶的药效发挥,同时添加绿茶提取物提高止咳枇杷糖浆的稳定性,延长保质期。虽然可以通过调整加入苦杏仁的时机,以及添加绿茶提取物来提高止咳枇杷糖浆的稳定性以及保质期,但是同时,苦杏仁以及绿茶提取物也会对糖浆的口感以及药效产生影响,另一方面绿茶提取物本身也有保质期限,作用是有限的。
我公司枇杷系列产品包括枇杷止咳胶囊、枇杷止咳胶囊以及强力枇杷露,其中强力枇杷露质量标准收载于,由枇杷叶69g、罂粟壳50g、百部15g、白前9g、桑白皮6g、桔梗6g、薄荷脑0.15g为原料,蔗糖600g、蜂蜜适量、枸橼酸适量、香精适量、防腐剂2.5g为辅料制成,制备工艺为:以上七味,除薄荷脑外,其余枇杷叶等六味加水煎煮二次,每次2小时,合并煎液,滤过,浓缩至约750ml,加苯甲酸钠2.5g,搅拌使溶解,加蔗糖600g,继续加热至沸,保持20分钟,静置,滤过,加入枸橼酸适量、用乙醇溶解的香精适量及薄荷脑,搅拌,混匀,静置,滤过,加水至1000ml,混匀,即得。
为进一步提高活性成分的含量,延长产品有效期和稳定性,技术人员进行相应的工艺和辅料研究,意外地发现,通过增加黄酮类物质的浓度以及控制表面活性剂的浓度可以达到抗菌、增溶作用,优化糖浆的质量和疗效。

(3)取蔗糖添加总量50%的水,加热混合均匀后,加入枸橼酸、苯甲酸钠,得到蔗糖混合溶液;
(4)将薄荷脑、香精溶解到乙醇中,得到乙醇溶液;
(5)将步骤(2)中提取物、步骤(3)中蔗糖混合溶液、步骤(4)中乙醇溶液混合,搅拌,静置,滤过,加水至1000ml,混匀,即得目标产物。
进一步的,所述枇杷止咳糖浆成份按照如下重量份:枇杷叶30-70份、罂粟壳40-60份、百部10-25份、白前5-12份、桑白皮3-9份、桔梗3-9份、薄荷脑0.1-0.5份、蔗糖400-600份、枸橼酸0.2-0.4份、苯甲酸钠1-5份、表面活性剂1.5-2.5份制成。
进一步的,所述冷藏室的温度为0-5℃,时间为12-24h。
进一步的,所述乙醇浓度不超过56%。
进一步的,所述表面活性剂为鼠李糖脂,浓度为1.5-2.5mg/ml糖浆。
进一步的,所述表面活性剂的制备方法为:(1)发酵液的制备:将菌体接种到含有酵母浸出膏的无机盐培养液中,1d之后,震荡培养作为富集培养基,再将菌种接到含有葡萄糖的无机盐培养液中,3d之后,再次震荡培养,得到含有鼠李糖脂的发酵液;(2)鼠李糖脂的分离:将发酵液中的pH调节至弱碱性,离心,收集上清液,将发酵液中的pH调至酸性,静置1d之后,再次离心,收集底物,用冲洗,冷冻干燥,得到粗鼠李糖脂混合物;(3)纯化:粗鼠李糖脂混合物溶于氯仿,经过真空抽虑除去蛋白质等不溶杂志之后,柱色谱方法进行纯化,即得。
其中,所述无机盐培养液的成份按照如下重量份:硝酸钠0.2-0.5份、磷酸二氢钾0.1-0.3份、十二水合磷酸氢二钠0.1-0.3份、硫酸镁0.01-0.02份,七水硫酸亚铁0.001-0.002份、酵母浸出膏0.05-0.07份。
综上所述,本发明的有益效果在于:第一、枇杷止咳糖浆主要化学成分为三萜酸类化合物,倍半萜类化合物,多酚类化合物,黄酮类化合物,糖苷类化合物以及挥发性化合物;鼠李糖脂作为一种阴离子表面活性剂被添加到中药中,在超声辅助下增加了中药中的有效化学成分的提取率,特别是黄酮类化合物,提取率达到19.2%。第二、黄酮类化合物是植物长期抵抗环境阻力而形成的一大类次级代谢产物是一种高效的生物抗氧化剂,在植物整个生长代谢过程中起重大作用,现代医学发现,黄酮类物质具有抗真菌,抗细菌作用,黄酮类物质的增加可以使糖浆中的细菌减少,优化糖浆的质量和疗效。第三、鼠李糖脂除了能增加止咳糖浆有效化学成分的提取率之外,当其在糖浆中浓度为1.5-2.5mg/ml时,还具有理想的抗微生物的性质,能抑制细菌、真菌的生长,使两者作用达到最佳,优化了糖浆的质量和疗效。第四、本发明采取冷提取的方式对原料的活性成分进行浸出、表面活性剂辅助超声提取,提高有效成分含量,且减少加热过程对药材活性物质的影响。
综上,本发明通过增加黄酮类物质的浓度以及控制表面活性剂的浓度达到抗菌,增溶作用,优化糖浆的质量和疗效;此外黄酮为糖浆中原有物质,而鼠李糖脂也被运用在食品行业,两者均不会对糖浆产生任何不良影响。
具体实施方式
下面对本发明的具体实施方式作进一步详细的说明,但本发明并不局限于这些实施方式,任何在本实施例基本精神上的改进或代替,仍属于本发明权利要求所要求保护的范围。
实施例1
材料:枇杷叶30g、罂粟壳60g、百部10g、白前12g、桑白皮3g、桔梗9g、薄荷脑0.1g、蔗糖600g、枸橼酸0.2g、苯甲酸钠5g、鼠李糖脂1.5g
制备方法:(1)将原料枇杷叶、罂粟壳、百部、白前、桑白皮、桔梗干燥后粉碎,混合,加入10倍水,然后放入冷藏室中0-5℃冷藏12h;
(2)将步骤(1)所得产物取出,加入表面活性剂,超声20min,得到提取物,过滤,将滤液经加热浓缩之后置于常温下保存,待用;
(3)取蔗糖添加总量50%的水,加热混合均匀后,加入枸橼酸、苯甲酸钠,得到蔗糖混合溶液;
(4)将薄荷脑、香精溶解到适量乙醇中,得到乙醇溶液;
(5)将步骤(2)中提取物、步骤(3)中蔗糖混合溶液、步骤(4)中乙醇溶液混合,搅拌,静置,滤过,加水至1000ml,混匀,即得目标产物。
实施例2
材料:枇杷叶70g、罂粟壳40g、百部25g、白前5g、桑白皮9g、桔梗3g、薄荷脑0.5g、蔗糖400g、枸橼酸0.4g、苯甲酸钠1g、鼠李糖脂2.5g
制备方法:(1)将原料枇杷叶、罂粟壳、百部、白前、桑白皮、桔梗干燥后粉碎,混合,加入15倍水,然后放入冷藏室中0-5℃冷藏20h;
(2)将步骤(1)所得产物取出,加入表面活性剂,超声20-45min,得到提取物,过滤,将滤液经加热浓缩之后置于常温下保存,待用;
(3)取蔗糖添加总量50%的水,加热混合均匀后,加入枸橼酸、苯甲酸钠,得到蔗糖混合溶液;
(4)将薄荷脑、香精溶解到适量乙醇中,得到乙醇溶液;
(5)将步骤(2)中提取物、步骤(3)中蔗糖混合溶液、步骤(4)中乙醇溶液混合,搅拌,静置,滤过,加水至1000ml,混匀,即得目标产物。
实施例3
材料:枇杷叶69g、罂粟壳50g、百部15g、白前9g、桑白皮6g、桔梗6g、薄荷脑0.15g、蔗糖600g、枸橼酸0.3g、苯甲酸钠2.5g、鼠李糖脂2.0g
制备方法:(1)将原料枇杷叶、罂粟壳、百部、白前、桑白皮、桔梗干燥后粉碎,混合,加入12倍水,然后放入冷藏室中0-5℃冷藏24h;
(2)将步骤(1)所得产物取出,加入表面活性剂,超声30min,得到提取物,过滤,将滤液经加热浓缩之后置于常温下保存,待用;
(3)取蔗糖添加总量50%的水,加热混合均匀后,加入枸橼酸、苯甲酸钠,得到蔗糖混合溶液;
(4)将薄荷脑、香精溶解到适量乙醇中,得到乙醇溶液;
(5)将步骤(2)中提取物、步骤(3)中蔗糖混合溶液、步骤(4)中乙醇溶液混合,搅拌,静置,滤过,加水至1000ml,混匀,即得目标产物。
实施例4
取0.9%氯化钠注射液,500ml,待用;
实施例5
取市场上出售的贵州神奇强力枇杷露,500ml,待用;

材料和制备方法同实施例3,其中鼠李糖脂用量为1.0g;

材料和制备方法同实施例3,其中原药材用量分别为:枇杷叶20g、百部5g、罂粟壳20g、8g、白前4g份、桑白皮2g、桔梗2g、薄荷脑0.1g。
试验1
(1)大鼠80只,体重为(200±20)g,雌雄各半,按体重、性别随机分8组,每组10只,将实施例1-9止咳糖浆以及氯化钠对大鼠灌胃,15ml/kg,连续5d,第五天灌胃5min之后,恒压条件下用浓氨水溶液喷雾引咳,将各组动物出现咳嗽的潜伏期以及咳嗽次数记录在表1;

(2)观察并记录实施例1-8中产品在0个月和放置6个月后的澄清度、口感、有效成分含量变化情况;按照实施例1-3、5-8中所使用的表面活性剂的用量,添加到培养有相同细菌数量的培养皿中,3d之后观察并记录培养皿中细菌数量;结果见表2;

1.一种枇杷止咳糖浆剂的制备方法,其特征在于,具体包括以下步骤:
(1)将原料枇杷叶、罂粟壳、百部、白前、桑白皮、桔梗干燥后粉碎,混合,加入10-15倍水,然后放入冷藏室中冷藏;
(2)将步骤(1)所得产物取出,加入表面活性剂,超声20-45min,得到提取物,过滤,将滤液经加热浓缩之后置于常温下保存,待用;
(3)取蔗糖添加总量50%的水,加热混合均匀后,加入枸橼酸、苯甲酸钠,得到蔗糖混合溶液;
(4)将薄荷脑、香精溶解到乙醇中,得到乙醇溶液;
(5)将步骤(2)中提取物、步骤(3)中蔗糖混合溶液、步骤(4)中乙醇溶液混合,搅拌,静置,滤过,加水至1000ml,混匀,即得目标产物;
