Abstract
本发明公开了一种双裂海木耳的乙酸乙酯提取物的制备方法,并公开了所制备的双裂海木耳的乙酸乙酯提取物在制备抗炎镇痛药物或抗炎镇痛食品中的用途。本发明制备的双裂海木耳的乙酸乙酯提取物具有很高的发展潜力,可用于制备抗炎镇痛的功能性食品,也可以用于制备抗炎止痛的药物。
Description
技术领域
本发明涉及医药技术领域,具体是双裂海木耳乙酸乙酯提取物在制备抗炎镇痛药物中 的用途。
背景技术
海洋天然产物的发展已经大幅增长,并已确认了多达4196个化合物的生物活性。藻 类占25%的海洋天然化合物,其余来自腔肠动物、棘皮动物门等。25%海洋天然产物领域的 是藻类产品,主要原因是藻类可再生、食用、环保。相对于绿藻和褐藻,红藻生物多样性 较高,这使得有更高机会可开发更多种活性物质。例如,先前研究报告也发现,细毛石花菜的半乳糖、从隅江篱的硫酸多糖具有止痛作用。因此,现有技术中已指出红藻具有巨大潜力,可作为新药开发的来源。
因为过度捕捞,近年来双裂海木耳的数量已经减少,并威胁到其生存。目前,对于养殖型双裂海木耳 生物活性的相关研究极少。现有技术中,只有两个非SCI期刊论文已经报导关于海木耳的生物活性。先前 大鼠的研究结果表明,双裂海木耳的水提取物可以消除自由基,具有抗糖尿病的特性,降低甘油三酸酯水 平。双裂海木耳之磷酸盐缓冲液(phosphate buffered saline,PBS)的提取物,也被发现有抗菌活性。活 体试验结果还表明,同属不同种的大型藻锡兰海木耳的水提取物有抗肿瘤活性,以及免疫调节作用。锡兰海木耳的水提取物,也被发现有抗氧化作用。现有技术中,相对于其它红藻,目前关于双裂海木耳之活性 物质成份的已知资料非常少,尚未公开关于双裂海木耳的乙酸乙酯提取物(ethyl acetate extract)在抗 炎、镇痛方面的研究。
发明内容
本发明的目的在于探讨养殖型大型红藻双裂海木耳之乙酸乙酯提取物的抗炎作用,以 解决上述背景技术中提出的问题。
为实现上述目的,本发明提供如下技术方案:
双裂海木耳的乙酸乙酯提取物的制备方法,步骤如下:
1)将双裂海木耳在50℃的条件下烘干24小时,获得干燥双裂海木耳;
2)将干燥双裂海木耳研末,浸泡在乙醇水溶液中,在室温下进行提取,获得浸泡液;
3)将浸泡液经过过滤、减压浓缩后,获得乙醇粗萃物;
4)将乙醇粗萃物悬浮在水中,并使用乙酸乙酯进行分配分离,且分配分离操作重复 进行三次,获得双裂海木耳的乙酸乙酯提取物。
作为本发明进一步的方案:步骤1)中,双裂海木耳为养殖型双裂海木耳。
作为本发明再进一步的方案:步骤2)中,所述乙醇水溶液的乙醇体积浓度为95%。
作为本发明再进一步的方案:步骤4)中,乙酸乙酯的体积用量与水的体积用量相同。
作为本发明再进一步的方案:步骤1)中,双裂海木耳双裂海木耳的用量为干重1kg。
作为本发明再进一步的方案:步骤3)中,所获得的乙醇粗萃物为34.3g。
作为本发明再进一步的方案:步骤4)中,水的加入量为1公升,乙酸乙酯的加入量为1公升。

与现有技术相比,本发明的有益效果是:
本发明制备的双裂海木耳的乙酸乙酯提取物,能抑制脂多糖(lipopolysaccharide, LPS)所诱导巨噬细胞所产生的炎性蛋白質诱导型一氧化氮合酶(inducible nitric oxide synthase,iNOS),口服双裂海木耳的乙酸乙酯提取物可以减少角叉菜胶(carrageenan)致 脚掌肿胀和炎性疼痛,且双裂海木耳的乙酸乙酯提取物可显著抑制角叉菜胶所致的白细胞 浸润(leukocyte infiltration),以及在发炎组织内炎症介质的蛋白质表达。本发明制备 的双裂海木耳的乙酸乙酯提取物具有很高的发展潜力,可用于制备抗炎镇痛的功能性食 品,也可以用于制备抗炎止痛的药物。

具体实施方式
下面结合具体实施方式对本发明的技术方案作进一步详细地说明。
实施例1
双裂海木耳的乙酸乙酯提取物的制备方法,步骤如下:
1)将双裂海木耳在50℃的条件下烘干24小时,获得干燥双裂海木耳,其中,双裂海木耳双裂海木 耳的用量为干重1kg,双裂海木耳为养殖型双裂海木耳,来自台湾地区水产试验所;
2)将干燥双裂海木耳研末,浸泡在乙醇水溶液中,在室温下进行提取,获得浸泡液, 其中,所述乙醇水溶液的乙醇体积浓度为95%;
3)将浸泡液经过过滤、减压浓缩后,获得乙醇粗萃物,所获得的乙醇粗萃物为34.3g;
4)将乙醇粗萃物悬浮在水中,并使用乙酸乙酯进行分配分离,且分配分离操作重复 进行三次,获得双裂海木耳的乙酸乙酯提取物,其中,水的加入量为1公升,乙酸乙酯的加入量为1公升,获得的乙酸乙酯提取物为7.7g。
采用苯酚-硫酸法测定双裂海木耳的乙酸乙酯提取物(PD1)的多糖含量。测量表明,双 裂海木耳的乙酸乙酯提取物(PD1)不含有多糖。
参与炎症反应的细胞,会释放各种促炎介质,包括诱导型一氧化氮合酶和白细胞介素 -1β(interleukin-1β,IL-1β)等。白细胞介素-1β激活疼痛感觉神经并传输到大脑,从而在病理条件下造成生物的疼痛反应。诱导型一氧化氮合酶主要被表达在巨噬细胞。在发炎反应中,当免疫系统的巨噬细胞被激活时,诱导型一氧化氮合酶的表现量会增加,从而加速一氧化氮的合成,而可能最终导致细胞毒性、感染性休克、癌症和其他严重的疾病。白细胞介素-1β是白细胞介素家族最强的促炎介质。先前的研究表明,注射白细胞介素 -1β抑制剂或诱导型一氧化氮合酶抑制剂可以减轻神经病变性大鼠的疼痛。现有技术中指 出,诱导型一氧化氮合酶和白细胞介素-1β在炎症反应中具有重要角色,相关抑制物很有 潜力可发展为新的抗炎药物。当细胞处于氧化压力高于抗氧化压力的状态时,将进一步促 进发炎因子的合成与释放。髓过氧物酶(myeloperoxidase,MPO)常作为氧化压力的标志物, 可用于评估物质对于氧化压力的抑制作用。在受损组织中,物质在降低髓过氧物酶之量的 效果,反映了它降低氧化压力的能力。同时,髓过氧物酶也是一种白细胞的标记物,因此, 髓过氧物酶(myeloperoxidase,MPO)的量可作为分析组织发炎程度的指标。
为了研究双裂海木耳之乙酸乙酯提取物的抗炎作用,使用脂多糖所导致小鼠巨噬细胞 发炎模式作为体外测试平台。在活体大鼠实验中,我们藉由角叉菜胶诱导脚掌肿胀所导致 发炎性疼痛,及诱导型一氧化氮合酶、白细胞介素-1β和髓过氧物酶等蛋白质的大量增加, 探讨双裂海木耳之乙酸乙酯提取物(PD1)潜在的抗炎作用与止痛效果。
实施例2
通过体外实验评估双裂海木耳之乙酸乙酯提取物(PD1)的抗炎活性。
从美国模式培养物保藏所中获得了小鼠RAW 264.7巨噬细胞(TIB-71;Manassas,VA, USA)。我们利用Dulbecco改良的Eagle培养基,添加10%热灭活胎牛血清、50U/ml青霉素、50μg/mL链霉素,2mM谷氨酰胺、1mM丙酮酸,和4.5g/L的葡萄糖,在37℃的 加湿培养箱孵化器(5%二氧化碳:95%空气)培养巨噬细胞。
细胞存活率:利用阿尔玛蓝处理,以测量细胞的存活率。阿尔玛蓝是一种四唑染剂, 可藉由活细胞减少的萤光产品,原理类似于细胞存活率实验所使用的噻唑蓝染剂。这种方 法已被证实能精确地测量RAW 264.7巨噬细胞的存活率。在细胞培养实验中,我们用100% 二甲基亚砜(dimethyl sulfoxide,DMSO)溶解双裂海木耳之乙酸乙酯提取物(PD1),二 甲基亚砜在培养基的最终使用浓度为1%。
体外抗炎试验:将小鼠巨噬细胞RAW 264.7培养在仅含有脂多糖(0.01μg/ml;L2654;Sigma,St.Louis MO,USA)的培养基中16小时,以诱导发炎反应。对于抗炎活 性测定,在脂多糖处理的5分钟前,对三个实验组的巨噬细胞分别添加双裂海木耳之乙酸 乙酯提取物10μg/ml、20μg/ml和50μg/ml,对正控制组的巨噬细胞添加氟美松 (dexamethasone)10μM。然后,用冰冷的磷酸盐缓冲液冲洗巨噬细胞,并在冰冷的裂解缓 冲液中裂解巨噬细胞,然后在20000×g、4℃条件下离心巨噬细胞30分钟。用一种改良自 Lowry等人的方法,自沉淀物上移出并保留上清液,并以DC蛋白质分析试剂盒测量上清 液的蛋白质浓度。我们进行了西方点墨法。在样品中加入等体积的样品缓冲液。置入样品 到三甲基甘氨酸-十二烷基硫酸钠-聚丙烯酰胺凝胶,在150V、90分钟条件下,进行样品 的电泳。在125mA、4℃的过夜条件下,在转渍缓冲液中,将样品转渍至聚偏氟乙烯膜。 用5%脱脂奶粉在室温下封闭膜50分钟,然后将膜以一级抗体在室温下浸泡180分钟,包 括诱导型一氧化氮合酶抗体(1:1000稀释;目录编号6103322;多株抗体;BD Pharmingen, San Diego,CA,USA)和环氧化酶-2(cyclooxygenase-2,COX-2)抗体(1:1000稀释;目录编 号160106;多株抗体;CaymanChemical,USA)。利用化学冷光法侦测髓过氧化物酶结合的 二级抗体。使用UVP BioChemi成像系统与LabWorks 4.0软件得到图像,以进行相对光 密度定量分析。在同一张影像上计算了双裂海木耳之乙酸乙酯提取物合并组或化合物合并 组的条带与仅有脂多糖处理组之间的相对变化量。
实验结果分析:
附图1为不同浓度的双裂海木耳之乙酸乙酯提取物对RAW264.7巨噬细胞存活率影响 结果图。使用阿尔玛蓝减少试验,我们分析PD1处理24、48小时和72小时后对RAW264.7细胞存活率的影响。结果表明,双裂海木耳之乙酸乙酯提取物(1μg/ml、10μg/ml)并不 明显影响细胞存活率。在第24小时,相较于控制组,双裂海木耳之乙酸乙酯提取物(100 μg/ml)明显地增加细胞存活率。试验结果指出,除了双裂海木耳之乙酸乙酯提取物(100 μg/ml)在第24小时显著地提高RAW264.7细胞存活率,其它组别的细胞存活率并没有受 到显著的影响。其中,数据呈现:平均值±平均值标准误差,与控制组比较为P<0.05。
附图2为双裂海木耳之乙酸乙酯提取物对脂多糖所诱导RAW264.7之发炎性蛋白质诱 导型一氧化氮合酶的影响结果图。以单独脂多糖组之诱导型一氧化氮合酶的蛋白质表现量 视为100%。在10μg/ml、20μg/ml和50μg/ml的双裂海木耳之乙酸乙酯提取物处理后,诱导型一氧化氮合酶之蛋白质表现量则分别成为103.1±11.6%、79.1±3.4%和64.6±5.0%。统计分析显示,双裂海木耳之乙酸乙酯提取物在20μg/ml和50μg/ml浓度可明显 抑制脂多糖所诱导RAW264.7细胞的诱导型一氧化氮合酶蛋白质表现量。与脂多糖组相比, 在正控制组中,氟美松也会显着地减少诱导型一氧化氮合酶的表达。其中,数据呈现:平 均值(mean)±平均值标准误差(standard error of the mean,SEM),与单独脂多糖组比 较为P<0.05。
附图3为双裂海木耳之乙酸乙酯提取物对脂多糖所诱导RAW264.7之发炎性蛋白质诱 导型环氧化酶-2的影响结果图。以单独脂多糖组之环氧化酶-2的蛋白质表达量视为100%。 统计分析表明,在10μg/ml、20μg/ml和50μg/ml的双裂海木耳之乙酸乙酯提取物处理 后,脂多糖所诱导RAW264.7细胞之环氧化酶-2蛋白质的表达,并没有受到明显的抑制作用。我们还观察到双裂海木耳之乙酸乙酯提取物并未造成β-肌动蛋白(β-actin)在组间 的显著性差异。其中,数据呈现:平均值±平均值标准误差,与单独脂多糖组比较为P<0.05。
在上述实验过程中,将数据以平均值±平均值标准误差的形式进行呈现,再以单向方 差分析对数据进行分析(one-way analysis of variance,ANOVA),然后采用邓肯方法 (Duncan’s method)进行多重比较,并将p<0.05视为有显著性差异。
实施例3
评估双裂海木耳之乙酸乙酯提取物(PD1)的活体抗炎或镇痛作用。
角叉菜胶致大鼠脚掌发炎的活体模式的制备:根据美国生理学会的动物照护和使用指 导原则,将饲养Wistar大鼠(体重:250-285g)在控制温度(22℃±1℃)和12h光暗 循环的房间。用4%异氟醚在塑料箱内麻醉大鼠后,经由面罩对大鼠给予2.5%异氟醚(在空气/氧气的混合气体)。在对大鼠右后脚进行脚掌内注射1.5%lambda型角叉菜胶的100μ L无菌生理食盐水的1个小时前,对大鼠口服2ml的空白溶剂(vehicle;2%二甲基亚砜)、 双裂海木耳之乙酸乙酯提取物(20或50mg/kg),或吲哚美辛(indomethacin;20mg/kg)。
大鼠疼痛行为分析:将大鼠放在高架金属丝网上,藉由测量后脚缩足反射阈值(paw withdrawal threshold;g)以分析机械痛敏(mechanical allodynia)的程度。采用一系列 的von Frey单丝纤维组(Stoelting,Wood Dale,IL,USA)对足底中部区域,使用Chaplan 的"向上-向下"方法确定最接近疼痛反应阈值的单丝纤维(缩足或舔脚)。我们将热源疼痛 测定仪(低强度热源(热光源激发强度为25),自动停止时间则为30秒;IITC,Woodland Hills,CA,USA)的热光源,置于脚掌底部以测量缩足反射潜伏期(pawwithdrawal latency; 秒),并进而评估热痛敏(thermal hyperalgesia)。
脚掌组织的组织病理学和免疫组织化学染色分析:心脏灌流含肝素(0.2U/mL)的冰冷 磷酸盐缓冲液,随后则改用4%多聚甲醛继续灌流,最后用4%多聚甲醛固定脚掌样品2小 时。将不同组大鼠的组织置于相同的冷冻包埋剂组织块中,然后用冷冻切片机(MicromHM 550;Thermo Scientific,Waltham,MA,USA)进行组织切片(20μm),并再使用苏木精 和伊红染色。关于免疫组织荧光染色分析,我们用0.1%的曲拉通X-100渗透切片20分钟, 再浸泡于4%正常马血清30分钟。然后再另浸泡于分别针对髓过氧物酶(1:200稀释;目录 编号ab9535;兔子多株抗体;Abcam,Cambridge,UK)、诱导型一氧化氮合酶(1:200稀释; 目录编号sc-7271;小鼠单株抗体;Santa Cruz Biotechnology Inc.,Santa Cruz,CA,USA) 或白细胞介素-1(1∶200稀释;目录编号AF-501-NA;山羊多株抗体;R&D systems,Minneapolis,MN,USA)的一级抗体溶液在4℃条件下过夜。我们在室温下将切片浸泡40 分钟的二级抗体溶液,包括绿色荧光兔子二级抗体(Alexa Fluor 488;1:400稀释)分别和 红色荧光小鼠二级抗体(rhodamine;1:400稀释)或红色荧光山羊二级抗体(rhodamine; 1:400稀释)的混合溶液。我们使用显微镜(Leica DM-6000CS;Leica Instruments Inc.,Wetzlar,Germany),配合光学影像撷取系统(Idea 5MP CMOS;SPOT Imaging Solutions, adivision of Diagnostic Instruments,Inc.,Sterling Heights,MI,USA)或荧光影 像撷取系统(Xplorer;SPOT Imaging Solutions),分析了每个组织病理学和免疫组织化 学染色的脚掌切片。我们利用影像分析系统软件(MetaMorph;Molecular Devices,Downington,PA,USA),定量分析免疫活性阳性反应面积(immunoreactive-positive area)的像素值,以求得免疫组织荧光染色分析数据。
实验结果分析:
一、口服PD1对角叉菜胶所致大鼠脚掌肿胀的影响
图4为双裂海木耳之乙酸乙酯提取物对角叉菜胶所致大鼠脚掌肿胀的影响结果图。图 5为随着角叉菜胶注射后的时间点,合并给予PD1大鼠之脚掌体积增加的百分比变化结果 图。图6为将合并给予不同剂量PD1所造成大鼠脚掌肿胀程度对时间点的变化转换为曲线 下面积值的示意图。相较于角叉菜胶合并空白溶剂组,PD1组的值较小,则反映出PD1明 显有效地抑制角叉菜胶所致大鼠脚掌肿胀。我们选择合并给予吲哚美辛(20mg/kg)作为正 控制组。数据呈现:每组6只大鼠的平均值±平均值标准误差。*,与角叉菜胶合并空 白溶剂组比较为P<0.05。
在活体抗发炎试验上,我们探讨PD1对角叉菜胶所致大鼠脚掌肿胀的影响。对大鼠脚 掌的外观病理现象进行影像纪录(图4),而图片记录结果表明,注射角叉菜胶导致大鼠脚 掌产生了明显的肿胀。合并口服PD1(20或50mg/kg),则减少大鼠脚掌肿胀。同时,我 们使用脚掌容积测量仪测量大鼠脚掌肿胀程度的变化(图5),并发现角叉菜胶所导致大鼠 脚掌肿胀在第9小时达到最明显的程度,随后肿胀情况逐渐消退。PD1(20或50mg/kg) 的合并给予,显著抑制角叉菜胶所诱导大鼠脚掌肿胀。与角叉菜胶合并空白溶剂组相比, 在正控制组中,吲哚美辛(20mg/kg)的合并给予,也可明显减少大鼠脚掌水肿。将大鼠脚 掌水肿程度对时间的变化曲线,换算成曲线下面积值(图6)。结果表明,相较于角叉菜胶 合并空白溶剂组,合并给予PD1(20或50mg/kg)可明显减少脚掌肿胀的程度。
二、PD1对角叉菜胶所诱导发炎性疼痛的影响
图7-8为合并口服PD1或吲哚美辛,对角叉菜胶所诱发大鼠疼痛行为的止痛效果,包 括热痛敏(图7)和机械痛敏(图8)。选择将合并给予吲哚美辛(20mg/kg)作为正控制组。数据呈现:每组6只大鼠的平均值±平均值标准误差。*,与角叉菜胶合并空白溶剂组 比较为P<0.05。
利用角叉菜胶所诱发大鼠的热痛敏和机械痛敏模式,分析PD1对发炎性疼痛的镇痛作 用。研究结果显示,角叉菜胶所引起热痛敏和机械痛敏至少会持续12小时以上。合并给 予PD1(20或50mg/kg)可以明显地改善角叉菜胶所诱导热痛敏(图7)和机械痛敏(图8)的情况,而且该止痛效果大约可持续12个小时。选择将合并给予吲哚美辛(20mg/kg)作 为正控制组。与角叉菜胶合并空白溶剂组相比,证明PD1能改善大鼠的热痛敏和机械痛敏 现象。
三、利用组织病理学分析PD1对角叉菜胶所注射脚掌内白细胞浸润的影响
图9-10为合并注射PD1对角叉菜胶所致大鼠脚掌组织内白细胞浸润的影响。图片显 示各组大鼠脚掌组织内白细胞(箭头)的情况,包括控制组(图9A)、角叉菜胶组(图9B)、角叉菜胶合并PD1组(图9C)。并针对各组白细胞数量(图10)进行定量分析。比例尺:(图9A)、(图9A)、(图9A)为150μm;(图9A)插图、(图9A)插图、(图9A)插图为35μm。 数据呈现:每组6只大鼠的平均值±平均值标准误差。*,与控制组比较为P<0.05;#, 与角叉菜胶合并空白溶剂组比较为P<0.05。
采用苏木精-伊红染色法对大鼠脚掌组织进行组织病理学分析。苏木精-伊红染色结果 表明,与对照组(图9A)相比,角叉菜胶会导致大鼠脚掌组织内的白细胞浸润(图9B)。合 并注射PD1,可以减少角叉菜胶所致白细胞浸润的程度(图9C)。统计结果(图10)也证明合并口服PD1可以减少角叉菜胶所诱导白细胞数目增加的程度。
四、PD1对角叉菜胶所致大鼠脚掌组织内发炎相关蛋白质表达的影响
图11-14分析PD1对角叉菜胶所诱发大鼠脚掌组织内髓过氧物酶(绿色)、白细胞介素 -1β(红色)、诱导型一氧化氮合酶(红色)之免疫活性增加的影响。组别设计包括控制组(图11A、图11D、图11G)、角叉菜胶组(图11B、图11E、图11H)、角叉菜胶合并给予PD1 组(图11C、图11F、图11I)。并呈现髓过氧物酶(图12)、白细胞介素-1β(图13)、诱导 型一氧化氮合酶(图14)之免疫活性的定量结果。数据呈现:每组6只大鼠的平均值±平 均值标准误差。*,与控制组比较为P<0.05;#,与角叉菜胶合并空白溶剂组比较为P<0.05。 比例尺:50μm。
用免疫组织化学方法,分析PD1对角叉菜胶所诱发大鼠脚掌组织内髓过氧物酶、白细 胞介素-1β、诱导型一氧化氮合酶等蛋白质表达的影响。结果显示,与控制组相比(图11A、 图11D、图11G),角叉菜胶所诱导大鼠脚掌组织中髓过氧物酶、白细胞介素-1β和诱导型 一氧化氮合酶的免疫活性明显地增加(图11B、图11E、图11H)。在合并给予PD1组中,髓过氧物酶、白细胞介素-1β和诱导型一氧化氮合酶的免疫活性有显著地降低(图11C、图11F、图11I)。荧光定量结果证明,合并口服PD1显著地抑制角叉菜胶所注射脚掌组织内 髓过氧物酶、白细胞介素-1β、诱导型一氧化氮合酶的免疫活性(图12、图13、图14)。 以上显示,PD1能抑制角叉菜胶所诱导大鼠脚掌组织的发炎反应。
五、PD1对角叉菜胶所致大鼠脚掌组织内白细胞介素-1β和诱导型一氧化氮合酶在髓 过氧物酶阳性反应细胞上表达的影响
图15为PD1对角叉菜胶所诱导大鼠脚掌组织中白细胞介素-1β(红色)和诱导型一氧 化氮合酶(红色)在髓过氧物酶阳性反应细胞(绿色)上表达的影响。组别设计包括控制组 (图15A、图15D)、角叉菜胶组(图15B、图15E)、角叉菜胶合并给予PD1组(图15C、图 15F)。双重免疫荧光染色结果显示,合并给予PD1能明显地抑制角叉菜胶所诱导白细胞介 素-1β和诱导型一氧化氮合酶分别与髓过氧物酶的共同表现(co-localization)(箭头)情 形。比例尺:50μm。
采用双重免疫组织化学染色,观察PD1对角叉菜胶所诱导大鼠脚掌组织中白细胞介素 -1β和诱导型一氧化氮合酶在髓过氧物酶阳性反应细胞上表达的影响。结果显示,控制组 之髓过氧物酶和白细胞介素-1β的免疫活性极低图15A)。在角叉菜胶组(图15B)中,我们 可以观察到很高量的白细胞介素-1β被表现在髓过氧物酶阳性反应细胞上,而这样的情况 则会受到合并口服PD1所抑制(图15C)。在控制组中,诱导型一氧化氮合酶与髓过氧物酶的免疫活性也极低(图15D)。在角叉菜胶组(图15E)中,我们观察到有高量的诱导型一氧 化氮合酶被表达在髓过氧物酶阳性反应细胞上,但这种现象则可以受到合并口服PD1所抑 制(图15F)。
在上述实验过程中,将数据以平均值±平均值标准误差的形式进行呈现,再以单向方 差分析对数据进行分析,然后采用邓肯方法进行多重比较,并将p<0.05视为有显著性差 异。
实施例2的体外实验显示,分离自双裂海木耳的PD1在100μg/ml浓度无细胞毒性。在20μg/ml,PD1显著地抑制促炎蛋白质诱导型一氧化氮合酶的表达。实施例3在角叉 菜胶所诱导脚掌发炎的结果显示,合并口服PD1能减轻疼痛行为和脚掌肿胀。合并给予PD1 可抑制角叉菜胶所致脚掌组织内白细胞浸润等发炎反应,还能减少髓过氧物酶、诱导型一 氧化氮合酶和白细胞介素-1β等发炎介质的表达。口服PD1大鼠并未产生明显的外观或行 为异常。综上,研究结果证明,PD1具有抗发炎和止痛的效果。
过度表现的诱导型一氧化氮合酶会导致大量一氧化氮的产生,而这将进一步加剧组织 的发炎反应。甚至可能进一步导致一系列慢性疾病,如癌症、糖尿病和心血管疾病。为了 筛选具有抗发炎活性的生物活性物质,许多研究报告使用了脂多糖所诱导小鼠巨噬细胞株 (RAW 264.7)的体外发炎模式。体外实验结果显示,PD1能显著地抑制脂多糖所诱导小鼠巨 噬细胞(RAW 264.7细胞)之促炎蛋白质诱导型一氧化氮合酶的大量表现,PD1也被证明对 这些细胞发炎反应具有剂量依赖的抑制效果(图2)。但PD1对环氧化酶-2则无明显影响(图 3)。已有先前研究指出,诱导型一氧化氮合酶藉由参与调控环氧化酶-2,而在发炎因子中 具有关键角色。根据我们的实验结果,我们认为PD1的抗发炎作用,是透过抑制诱导型一 氧化氮合酶的蛋白质表达,而不是经由调控环氧化酶-2的蛋白质表达。
在现有天然产物的活体消炎止痛相关研究中,角叉菜胶所致大鼠脚掌肿胀是最被广泛 使用于发炎相关研究的实验模式。我们用角叉菜胶诱发大鼠脚掌产生发炎现象,以评估PD1 在活体的抗发炎和止痛作用。结果显示,合并口服PD1可以降低角叉菜胶所诱导的脚掌肿 胀(图5),及热痛敏(图7)和机械痛敏(图8)等疼痛行为。因此,基于上述结果,可以证明 PD1可以有效地降低角叉菜胶注射大鼠的发炎性疼痛。
在大鼠脚掌注射角叉菜胶后,受损组织能刺激肥大细胞释放缓激肽、5-羟色胺和组织 胺,而提高血管通透性。这会导致大鼠脚掌逐渐肿胀,并引起胞噬性发炎反应。此外,受 损组织将释放细胞激素,促使白细胞参与胞噬反应,并促进诱导型一氧化氮合酶的表达, 从而导致产生大量的促炎介质(如前列腺素),最终导致发炎性疼痛。先前研究已经证明, 对大鼠脚掌注射角叉菜胶会导致脚掌组织的发炎,白细胞聚集在发炎区域而造成细胞浸 润。我们在先前研究中也发现,角叉菜胶能诱导大鼠脚掌组织内发生高度的白细胞浸润。 在本研究中,我们用苏木精和伊红染色来分析组织切片,观察发炎组织的白细胞浸润情况, 而这现象则是可以受到合并口服PD1所抑制(图9)。通过免疫荧光染色分析,我们发现角 叉菜胶能诱导大鼠脚掌组织中,诱导型一氧化氮合酶、白细胞介素-1β和髓过氧物酶等促 炎性蛋白质的大量表达,而这种现象可以受到合并口服PD1所抑制(图11)。现有研究已经 表明,诱导型一氧化氮合酶是发炎反应中最重要的调节酶。当诱导型一氧化氮合酶过度表 达,会导致大量一氧化氮产生,而发生严重的发炎和疾病。我们也发现角叉菜胶组大鼠脚 掌中有大量的诱导型一氧化氮合酶蛋白质表达。先前研究表明,诱导型一氧化氮合酶在角 叉菜胶所诱导发炎反应中具有重要的病理作用。周边给予专一性诱导型一氧化氮合酶抑制 剂,可抑制角叉菜胶所诱导的脚掌肿胀和疼痛行为。除了作为促炎介质外,白细胞介素-1β 还可以调节免疫功能。对大鼠注射白细胞介素-1受体拮抗剂,能减少神经病变性疼痛的产 生。髓过氧物酶也具有促炎作用。综上,诱导型一氧化氮合酶、白细胞介素-1β和髓过氧 物酶都在发炎反应中扮演着重要的角色,而我们发现PD1能抑制这三个促炎性蛋白质的表 达。
髓过氧物酶主要存在于白细胞的初级颗粒中,并负责初级免疫。当细菌侵入生物体时, 白细胞会吞通过吞噬作用吞噬这些细菌,并形成吞噬体,而发生脱颗粒作用。同时,髓过 氧物酶将从初级颗粒释放到吞噬体。然后,催化生成次氯酸、酪氨酸自由基和活性氧,这 些都是能有效杀死入侵细菌的氧化剂。所以通过测量髓过氧物酶的表达,我们可以评估氧 化损伤的程度。此外,目前已经知道髓过氧物酶也是白细胞标记物。我们实验结果发现角 叉菜胶组大鼠脚掌组织内,髓过氧物酶之免疫活性有明显增加(图12),证明确实发生白细胞浸润(图9)。在角叉菜胶大鼠脚掌中,我们观察到诱导型一氧化氮合酶和白细胞介素 -1β的大量表达,这两个蛋白质并分别皆与髓过氧物酶有共同表现(图15B与15E)。同时, 这些现象也会受到合并口服PD1所抑制(图15C和15F)。本发明确认PD1的抗发炎作用跟 下列两个现象有关,包括抑制白细胞的浸润,减少促炎性蛋白质诱导型一氧化氮合酶、白 细胞介素-1β在髓过氧物酶阳性反应细胞上的表达。
能否以持续和稳定的方式供应海洋天然产物,往往是能否进行临床前和临床试验的关 键因素之一。由于双裂海木耳可以被养殖,所以可以稳定地供应PD1。基于本发明的实验 结果,PD1的优点在于,可以口服、低细胞毒性,并具有消炎止痛作用。同时,口服PD1 的大鼠,并没有在本实验过程中表现出运动行为或外观上的明显异常。本发明所采用的提 取方法,不同于其他大多数多糖的提取方法。一种常用的多糖提取方法是,在0.15N盐 酸溶液中浸泡和搅拌海藻粉72小时。从离心机中取出澄清液后,加入乙醇并进行24小时 的搅拌,使多糖沉淀,最后通过真空冷却而干燥得到多糖。藉由本发明所使用大型藻类的 乙醇提取法,可以确保不会溶出多糖,而使乙醇提取液不含任何多糖。同时,苯酚-硫酸 法的试验结果也表明PD1不含多糖。PD1的未来发展方向有三个。首先,在PD1所含可能 化合物的结构鉴定,以识别活性成分并促进相关药物的未来发展。患有慢性疼痛的病人不 得不忍受长期使用止痛药而导致的严重副作用,或担心成瘾问题。而具有高安全性、可长 期服用的抗炎膳食补充剂,已成为缓解疼痛的新方法。一些先前研究已经成功从食物中鉴 定生物活性成分,包括抗发炎和止痛成分。双裂海木耳原先只是一般的食品产品,透过我 们的研究发现其提取物PD1具有抗炎和止痛作用,而符合抗炎膳食补充剂的特性。第二个 方向,为了将PD1开发为功能性食品原料,可继续研究PD1之抗发炎和止痛作用的细胞学 机制,并探索PD1对于其他炎性疼痛性疾病的疗效。最后的发展方向,则可以继续探索PD1 其他可能的生物活性。预期本发明结果,将有助于其他科学家继续开发双裂海木耳的其它 生物医学应用。
上面对本发明的较佳实施方式作了详细说明,但是本发明并不限于上述实施方式,在 本领域的普通技术人员所具备的知识范围内,还可以在不脱离本发明宗旨的前提下作出各 种变化。
Claims (9)
1.双裂海木耳乙酸乙酯提取物在制备抗炎镇痛药物中的用途。
2.双裂海木耳乙酸乙酯提取物在制备抗炎镇痛食品中的用途。
3.根据权利要求1或2所述的用途,其特征在于,所述双裂海木耳的乙酸乙酯提取物的制备方法,步骤如下:
1)将双裂海木耳在50℃的条件下烘干24小时,获得干燥双裂海木耳;
2)将干燥双裂海木耳研末,浸泡在乙醇水溶液中,在室温下进行提取,获得浸泡液;
3)将浸泡液经过过滤、减压浓缩后,获得乙醇粗萃物;
4)将乙醇粗萃物悬浮在水中,并使用乙酸乙酯进行分配分离,且分配分离操作重复进行三次,获得双裂海木耳的乙酸乙酯提取物。
4.根据权利要求3所述的用途,其特征在于,步骤1)中,双裂海木耳为养殖型双裂海木耳。
5.根据权利要求3所述的用途,其特征在于,步骤2)中,所述乙醇水溶液的乙醇体积浓度为95%。
6.根据权利要求3所述的用途,其特征在于,步骤4)中,乙酸乙酯的体积用量与水的体积用量相同。
7.根据权利要求3所述的用途,其特征在于,步骤1)中,双裂海木耳双裂海木耳的用量为干重1kg。
8.根据权利要求7所述的用途,其特征在于,步骤3)中,所获得的乙醇粗萃物为34.3g。
9.根据权利要求8所述的用途,其特征在于,步骤4)中,水的加入量为1公升,乙酸乙酯的加入量为1公升。
