Example 3 decoction
The embodiment of the invention provides a traditional Chinese medicine composition for treating diarrhea type IBS, which is prepared from the following raw material medicines: 15g of lotus seeds (containing lotus plumule), 12g of cortex albiziae, 2g of amber powder, 15g of spina date seeds, 6g of radix bupleuri, 15g of white paeony roots, 10g of radix curcumae, 15g of oldenlandia diffusa, 9g of rhizoma cimicifugae, 9g of liquorice, 12g of white hyacinth beans, 6g of fructus amomi, 3g of fructus evodiae, 10g of pomegranate rind, 9g of nutmeg and 10g of myrobalan.
The Chinese medicinal composition is in the form of decoction, and the preparation method comprises the following steps:
weighing the medicines according to the weight, processing the medicines according to the procedures in the preparation method, mixing the medicines except the amber powder according to the formula proportion, soaking, decocting according to a conventional method to obtain a traditional Chinese medicine decoction, filtering dregs of a decoction, adding the amber powder, and stirring uniformly to obtain the traditional Chinese medicine decoction for taking.
5. The taking dosage is as follows: the decoction is one dose per day. The tablet and capsule preparation is taken at a dose of 1 g/day/person per day, and is equivalent to 6.0g crude drug/day/person according to crude drug amount.
6. Convincing experimental evidence (i.e. pharmacological experiments: including clinical experiments, animal experiments or in vitro experiments (microorganisms), etc.), experimental evidence providing specific experimental compounds, dosage, administration method, experimental method and calculation method for judging conclusion, etc.
1) Acute toxicity test: an acute toxicity test is carried out by using 42 NIH mice, with SPF level, half male and female, and the weight of 18-20 g. Mice were randomly divided into two groups of 20 mice each, control and dosing groups, fasted for 12 hours prior to the experiment; the capsule prepared in example 1 of the present invention was dissolved in water (concentration of 2.58g crude drug/ml, maximum concentration) and gavage was performed with a gavage volume of 5ml/kg (i.e., single administration dose of 12.9 crude drug/kg), the control group was administered with an equal amount of physiological saline 2 times a day with an interval of 6 hours, and after administration, observation was continued for 14 days, and the toxic reaction and death number of mice were recorded. The experimental results show that: compared with a control group, the mice have no obvious difference after administration, and the mice have normal general conditions, diet, drinking water and weight increase after continuous observation for 14 days in the experiment. The capsule LD50 of the invention is orally administrated to mice with a gastric lavage of more than 12.9 crude drugs/kg, and the maximum daily dosage is 25.8 crude drugs/kg/day. The clinical dosage of the traditional Chinese medicine is equivalent to 6.0g crude drug/day/person, the weight of an adult is measured by 60KG, and the average dosage of the traditional Chinese medicine is 0.1g crude drug/KG/day. According to the weight: the tolerance of the traditional Chinese medicine for oral gavage of mice (the average body weight is calculated by 20g) is 258 times of the clinical dosage. Therefore, the traditional Chinese medicine composition disclosed by the invention is low in acute toxicity and safe in clinical medication.
2) Clinical data:
general data I: 184 patients with diarrhea-type IBS treated in a hospital during 6 months-2016 12 months in 2014 were selected, 96 men, 88 women, and 26-67 years old. The groups were randomly divided into 4 groups, 1-3 treatment groups and a control group, 46 per group. The comparison of the difference of the patients in age, sex and disease condition has no statistical significance and is comparable.

psychological test: the assessment of mental health by questionnaires to detect anxiety, depression or other psychological disorders in the patient helps support diagnosis of the disorder;
sixthly, organic lesions such as intestinal cancer, ovarian cancer and the like are eliminated.
III, the curative effect standard is as follows:

(2) Treatment group 2: the tablets of example 2 were administered to 46 patients 2 times a day, 0.5g once, and 20 days as a treatment course. After treatment, statistics shows that 16 patients are cured clinically, 23 patients are treated with obvious effect, and 7 patients are treated with effective effect. During the treatment period, no side effect caused by the medicine is found.
(3) Treatment group 3: the decoction of the embodiment 3 is used by 46 patients, one dose is taken by each person in 2 times, and 20 days are a treatment course. After treatment, statistics shows that 21 patients are cured clinically, 20 patients are treated with obvious effect, and 5 patients are treated with effective effect. During the treatment period, no side effect caused by the medicine is found.
(4) Control group: the patient in 46 cases takes trimebutine maleate (0.1 mg/tablet, manufactured by Hainanpril pharmaceutical Co., Ltd., national standard H20000390) 3 times a day, 0.1mg each time, and 20 days as a treatment course. After treatment, statistics shows that 11 patients are clinically cured, 19 patients are effectively treated, 12 patients are effectively treated, and 4 patients are not effective. During the treatment period, no side effect caused by the medicine is found.
And V, conclusion:
the clinical tests show that: compared with a control group, the clinical cure and significant efficiency of the traditional Chinese medicine composition of the embodiments 1 to 3 of the invention are obviously improved compared with the control group. The traditional Chinese medicine composition disclosed by the invention has a remarkable advantage for treating diarrhea type IBS.
It should be noted that the above-mentioned embodiments are only preferred embodiments of the present invention, and the present invention is not limited thereto, and although the present invention has been described in detail with reference to the foregoing embodiments, it will be apparent to those skilled in the art that modifications and equivalents can be made in the technical solutions described in the foregoing embodiments, or equivalents thereof. Any modification, equivalent replacement, or improvement made within the spirit and principle of the present invention should be included in the protection scope of the present invention. Although the present invention has been described with reference to the specific embodiments, it should be understood by those skilled in the art that various changes and modifications may be made without departing from the spirit and scope of the invention.

Claims (9)

1. A traditional Chinese medicine composition for treating diarrhea-predominant irritable bowel syndrome is characterized by being prepared from the following raw material medicines in parts by weight: 12-18 parts of lotus seeds; 10-15 parts of cortex albiziae; 1-4 parts of amber powder; 12-18 parts of spina date seeds; 4-8 parts of radix bupleuri; 12-18 parts of white peony root; 8-12 parts of curcuma aromatica; 12-18 parts of oldenlandia diffusa; 6-12 parts of cimicifugae foetidae, 6-12 parts of liquorice, 10-15 parts of white hyacinth beans, 4-8 parts of fructus amomi, 1-5 parts of fructus evodiae, 8-12 parts of pomegranate rind, 6-12 parts of nutmeg and 8-12 parts of myrobalan;
the lotus plumule is not removed from the lotus seed;
the processing method of the raw materials comprises the following steps:
1) honey-fried cimicifugae foetidae and licorice: diluting refined honey with boiled water at a ratio of 6:4, spraying into cimicifugae rhizoma or Glycyrrhrizae radix slice, stirring, placing into a stir-frying container, heating with an electric ceramic furnace at 800w power, taking out, cooling, and adding refined honey 30g per 100g cimicifugae rhizoma or Glycyrrhrizae radix slice;
2) parching semen Ziziphi Spinosae: parching semen Ziziphi Spinosae with slow fire until outer skin is swollen at 150 deg.C for 5 min;
3) stir-frying white hyacinth bean and pomegranate rind with sand: placing a proper amount of sand in a medicine frying pot, heating for 5 minutes under 2200w power by using an electric ceramic furnace, reducing the heating to 800w power, adding white hyacinth beans or pomegranate rinds, stir-frying, stopping fire when the medicinal materials become slightly yellow, continuously stir-frying by using the residual temperature of the sand until the surface is brown, screening out sandy soil, and mashing when in use;
4) processing bupleuri radix with vinegar; mixing the radix bupleuri green sheet with rice vinegar, mixing with 20g of rice vinegar per 100g of radix bupleuri green sheet, moistening thoroughly, placing in a pan, parching with slow fire, taking out, and cooling;
5) coating roasted nutmeg and myrobalan on the surface: moistening semen Myristicae or fructus Chebulae, wrapping with 3 layers of flour, adding into parched hot sand, parching at 180 deg.C for 20 min, taking out, sieving to remove sand, peeling, and cooling;
6) cortex Albizziae, Succinum powder, radix Paeoniae alba, radix Curcumae, herba Hedyotidis Diffusae, fructus evodiae, semen Nelumbinis and fructus Amomi, and semen Nelumbinis and fructus Amomi should be smashed.

The traditional Chinese medicine composition for treating diarrhea-predominant irritable bowel syndrome according to claim 1, which is prepared from the following raw material medicines in parts by weight: 12-15 parts of lotus seeds; 10-12 parts of cortex albiziae; 1-2 parts of amber powder; 12-15 parts of spina date seeds; 4-6 parts of radix bupleuri; 12-15 parts of white peony root; 8-10 parts of curcuma aromatica; 12-15 parts of oldenlandia diffusa; 6-9 parts of cimicifugae foetidae, 6-9 parts of liquorice, 10-12 parts of white hyacinth beans, 4-6 parts of fructus amomi, 1-3 parts of fructus evodiae, 8-10 parts of pomegranate rind, 6-9 parts of nutmeg and 8-10 parts of myrobalan;
the lotus seed does not have lotus plumule.

The traditional Chinese medicine composition for treating diarrhea-predominant irritable bowel syndrome according to claim 1, which is prepared from the following raw material medicines in parts by weight: 15-18 parts of lotus seeds; 12-15 parts of cortex albiziae; 2-4 parts of amber powder; 15-18 parts of spina date seeds; 6-8 parts of radix bupleuri; 15-18 parts of white peony root; 10-12 parts of curcuma aromatica; 15-18 parts of oldenlandia; 9-12 parts of cimicifugae foetidae, 9-12 parts of liquorice, 12-15 parts of white hyacinth beans, 6-8 parts of fructus amomi, 3-5 parts of fructus evodiae, 10-12 parts of pomegranate rind, 9-12 parts of nutmeg and 10-12 parts of myrobalan;
the lotus seed does not have lotus plumule.
4. The traditional Chinese medicine composition for treating irritable bowel syndrome with diarrhea according to any one of claims 1 to 3, wherein the traditional Chinese medicine composition is prepared into decoction, tablets, granules, pills or oral liquid.
5. A Chinese medicinal preparation for treating diarrhea-predominant irritable bowel syndrome, which comprises the Chinese medicinal composition for treating diarrhea-predominant irritable bowel syndrome according to any one of claims 1 to 3.

The method for preparing a Chinese medicinal composition for treating irritable bowel syndrome with diarrhea according to claim 1, wherein except amber powder, the rest materials are mixed according to a formula ratio, soaked, decocted conventionally to obtain a Chinese medicinal decoction, and after the residue is filtered, amber powder is added, and the mixture is stirred uniformly to obtain the composition for administration.
7. The preparation method of the capsule of the traditional Chinese medicine composition for treating diarrhea-predominant irritable bowel syndrome according to claim 1, comprising:
a. soaking the Chinese medicinal composition in 7 times of water for 2 hr, extracting volatile oil for 5 hr, filtering the medicinal liquid, and collecting the residue; wherein the volatile oil is extracted by supercritical extraction method;
b. b, adding ethanol with the alcohol degree of 75% into the medicine dregs obtained in the step a, soaking for 1 hour, extracting for 3 times, 2 hours each time, adding 10 times of the ethanol of the mixture in the first time, adding 10 times of the ethanol of the mixture in the second time, combining the extracting solutions, and filtering; mixing the above two filtrates, filtering, concentrating the filtrate under reduced pressure to relative density of 1.30 at 80 deg.C, precipitating with ethanol twice, adjusting ethanol concentration to 75%, collecting supernatant, and concentrating to relative density of 1.40 at 80 deg.C to obtain extract; wherein the extraction adopts membrane separation technology;
c. and c, mixing the extract obtained in the step b with volatile oil, grinding for 10 minutes by using a colloid mill, continuously stirring, cooling to room temperature to obtain a traditional Chinese medicine composition, and filling the traditional Chinese medicine composition into a capsule shell to obtain a capsule finished product.
8. The preparation method of the traditional Chinese medicine composition tablet for treating diarrhea-predominant irritable bowel syndrome according to claim 1, comprising:
putting the traditional Chinese medicine composition into 82% ethanol which is 10 times the amount of the traditional Chinese medicine composition, heating and refluxing for two times, extracting active ingredients for 45 minutes each time, combining the two extracting solutions, standing to obtain an extracting solution I, and keeping dregs for later use; adding 10 times of water into the medicine residues, heating, refluxing and extracting for three times, wherein each time is 1 hour, and combining the three extracting solutions to obtain an extracting solution II; mixing the first extractive solution and the second extractive solution, recovering ethanol under reduced pressure, concentrating to obtain medicinal liquid with concentration of 0.5g crude drug/mL, and vacuum filtering to obtain filtrate with relative density of 1.08 at 20 deg.C; eluting the filtrate twice through a macroporous adsorption resin column with the volume of 10L, eluting with deionized water or distilled water with the volume of 10 times that of the resin column, eluting with 95% ethanol with the volume of 5 times that of the resin column, collecting ethanol eluate, and removing ethanol solvent to obtain raw material liquid medicine; adding starch, and making into tablet.
9. Use of the Chinese medicinal composition for treating diarrhea-predominant irritable bowel syndrome according to any one of claims 1 to 3 for the preparation of a medicament having efficacy of treating patients with coexisting anxiety and IBS.
