Abstract
本发明涉及一种复方中药提取物及其在制备口腔护理产品中的应用,所述复方中药提取物的制备方法,包括如下步骤:(1)按权利要求1‑2任一项所述配方取干燥的苦参、陈皮、秦皮,将其粉碎后,加入水、适量的纤维素酶和木瓜蛋白酶混匀,调pH5.5‑6.5,于30‑40℃下,发酵2‑3天,过滤得发酵液,发酵液浓缩得发酵物;(2)步骤(1)得到的发酵物通过大孔树脂吸附,用体积分数为30‑40%的乙醇溶液洗脱,收集3‑5倍柱体积洗脱液,再经活性炭脱色、浓缩、干燥得所述复方中药提取物。
Description
技术领域
本发明属于中药日化品领域,具体涉及一种复方中药提取物及其在制备口腔护理产品中的应用。
背景技术
苦参为豆科槐属植物,其干燥根作为传统中药苦参入药,始载于汉代《神农本草经》,列为中品,味苦,性寒,具有清热燥湿,杀虫利尿的功效,对热痢、便血、尿闭、赤白带下、阴肿阴痒、湿疹、湿疮有良好作用,外用皮肤瘙痒、滴虫性阴道炎也有很好的疗效。陈皮(即橘皮),来源于芸香科植物橘及其栽培变种的成熟果皮,《神农本草经》曰:陈皮,味辛、温。主胸中瘾热,逆气,利水谷。秦皮为常用中药,始载于《神农本草经》,列为中品,《中国药典》2015年版规定秦皮为木犀科植物苦枥白蜡树Fraxinus rhynchophylla Hance、白蜡树Fraxinus chinensis Roxb.、尖叶白蜡树Fraxinusszaboana Lingelsh.、宿柱白蜡树Fraxinus stylosaLingelsh.的干燥枝皮或干皮,是临床常用中药。其主要功效为清热燥湿、平喘止咳、明目,临床上用于治疗肠炎、白带过多、慢性支气管炎、细菌性痢疾、目赤肿痛、迎风流泪、牛皮癣等。本发明提供一种苦参、陈皮、秦皮复方中药提取物、其在杀菌、除臭方面的活性,以及开发用于制备口腔护理产品(例如牙膏)的用途。
发明内容
本发明提供一种复方中药提取物,其特征在于按重量份计所述复方中药的配方为苦参3-5份、陈皮0.5-1份、秦皮2-3份。
上述复方中药提取物,其特征在于按重量份计所述复方中药的配方优选为苦参3份、陈皮0.5份、秦皮2份。
本发明的另一实施方案提供上述复方中药提取物,其特征在于所述复方中药提取物的制备方法,包括如下步骤:
(1)按配方量取干燥的苦参、陈皮、秦皮,将其粉碎后,加入水、适量的纤维素酶和木瓜蛋白酶混匀,调pH5.5-6.5,于30-40℃下,发酵2-3天,过滤得发酵液,发酵液浓缩得发酵物;
(2)步骤(1)得到的发酵物通过大孔树脂吸附,用体积分数为30-40%的乙醇溶液洗脱,收集3-5倍柱体积洗脱液,再经活性炭脱色、浓缩、干燥得所述复方中药提取物。
步骤(1)中水优选蒸馏水或去离子水,水的质量用量为苦参、陈皮、秦皮质量之和的5-8倍,纤维素酶的质量用量为苦参、陈皮、秦皮质量之和的0.5-1.0%,木瓜蛋白酶与纤维素酶的质量比为1:1;
步骤(2)所述大孔树脂的型号选自AB-8、D101、HZ818、HPD100中的一种。
本发明的另一实施方案提供上述复方中药提取物的制备方法,其特征在于包括如下步骤:
(1)按配方量取干燥的苦参、陈皮、秦皮,将其粉碎后,加入水、适量的纤维素酶和木瓜蛋白酶混匀,调pH5.5-6.5,于30-40℃下,发酵2-3天,过滤得发酵液,发酵液浓缩得发酵物;
(2)步骤(1)得到的发酵物通过大孔树脂吸附,用体积分数为30-40%的乙醇溶液洗脱,收集3-5倍柱体积洗脱液,再经活性炭脱色、浓缩、干燥得所述复方中药提取物。
步骤(1)中水优选蒸馏水或去离子水,水的质量用量为苦参、陈皮、秦皮质量之和的5-8倍,纤维素酶的质量用量为苦参、陈皮、秦皮质量之和的0.5-1.0%,木瓜蛋白酶与纤维素酶的质量比为1:1;
步骤(2)所述大孔树脂的型号选自AB-8、D101、HZ818、HPD100中的一种。
本发明的另一实施方案提供上述复方中药提取物作为抗菌剂的应用。
本发明的另一实施方案提供上述复方中药提取物在制备预防和/或治疗细菌感染引起疾病的药物中的应用。所述细菌优选大肠杆菌或金黄色葡萄球菌。
本发明的另一实施方案提供上述复方中药提取物在制备口腔护理产品中的应用。所述口腔护理产品优选口气清新剂、牙膏、唇膏、漱口水等。
本发明的另一实施方案提供上述复方中药提取物在清除挥发性硫化物中的应用。所述挥发性硫化物优选二甲基硫醚、硫化氢、甲硫醇中的一种或几种。
本发明的另一实施方案提供一种口腔护理产品,其特征在于该口腔护理产品包含上述复方中药提取物。所述口腔护理产品优选口气清新剂、牙膏、唇膏、漱口水等。
本发明的另一实施方案提供一种杀菌除臭中药牙膏,其特征在于所述中药牙膏包含如下成分:
上述复方中药提取物1-2份、活性添加物0.02-0.05份、摩擦剂20-30份、湿润剂15-20份、表面活性剂7-10份、粘合剂2-3份、香料3-5份。
所述活性添加物选自叶绿素或氟化钠,摩擦剂选自碳酸钙、磷酸氢钙、二氧化硅、三水合氧化铝中的一种或几种,湿润剂选自水、甘油、山梨醇中的一种或几种,表面活性剂选自十二烷基硫酸钠、月桂酰肌氨酸钠、酚基谷氨酸钠、椰油酰胺丙基甜菜碱中的一种或几种,粘合剂选自黄原胶、海澡酸钠、羟乙基纤维素、瓜尔胶、羧甲基纤维素钠、卡拉胶中的一种或几种,香料选自薄荷油、薄荷醇、艾叶香精中的一种或几种。
本发明的另一实施方案提供上述杀菌除臭中药牙膏的制备方法,其特征在于包括如下步骤:
按照重量份称取上述复方中药提取物1-2份、活性添加物0.02-0.05份、表面活性剂7-10份、湿润剂15-20份搅拌形成混合溶液,再按照重量份加入摩擦剂20-30份、粘合剂2-3份、香料3-5份,搅拌研磨0.5-1小时,真空脱气制膏、包装即得所述杀菌除臭中药牙膏。
所述活性添加物选自叶绿素或氟化钠,摩擦剂选自碳酸钙、磷酸氢钙、二氧化硅、三水合氧化铝中的一种或几种,湿润剂选自水、甘油、山梨醇中的一种或几种,表面活性剂选自十二烷基硫酸钠、月桂酰肌氨酸钠、酚基谷氨酸钠、椰油酰胺丙基甜菜碱中的一种或几种,粘合剂选自黄原胶、海澡酸钠、羟乙基纤维素、瓜尔胶、羧甲基纤维素钠、卡拉胶中的一种或几种,香料选自薄荷油、薄荷醇、艾叶香精中的一种或几种。
所述复方中药提取物的制备方法,包括如下步骤:
(1)按配方量取干燥的苦参、陈皮、秦皮,将其粉碎后,加入水、适量的纤维素酶和木瓜蛋白酶混匀,调pH5.5-6.5,于30-40℃下,发酵2-3天,过滤得发酵液,发酵液浓缩得发酵物;
(2)步骤(1)得到的发酵物通过大孔树脂吸附,用体积分数为30-40%的乙醇溶液洗脱,收集3-5倍柱体积洗脱液,再经活性炭脱色、浓缩、干燥得所述复方中药提取物。
步骤(1)中水优选蒸馏水或去离子水,水的质量用量为苦参、陈皮、秦皮质量之和的5-8倍,纤维素酶的质量用量为苦参、陈皮、秦皮质量之和的0.5-1.0%,木瓜蛋白酶与纤维素酶的质量比为1:1;
步骤(2)所述大孔树脂的型号选自AB-8、D101、HZ818、HPD100中的一种。
所述杀菌除臭中药牙膏在清除挥发性硫化物中的应用。所述挥发性硫化物优选二甲基硫醚、硫化氢、甲硫醇中的一种或几种。
本发明所述的苦参、陈皮、秦皮可以是中药材,也可以是中药饮片。
与现有技术相比,本发明的优点在于:(1)本发明提供一种苦参、陈皮、秦皮三味复方中药发酵提取物,该提取物在抗菌,清除挥发性硫化物方面表现出优异的特性;(2)本发明利用复方中药发酵提取物制备一种杀菌除臭牙膏。
具体实施方式
为了便于对本发明的进一步理解,下面提供的实施例对其做了更详细的说明。但是这些实施例仅供更好的理解发明而并非用来限定本发明的范围或实施原则,本发明的实施方式不限于以下内容。
实施例1
(1)取干燥的苦参300g、陈皮50g、秦皮200g,将其粉碎后,加入蒸馏水(2.75kg)、纤维素酶2.75g和木瓜蛋白酶2.75g混匀,调pH5.5-6.0,于30℃下,发酵3天,过滤得发酵液,发酵液浓缩得发酵物;
(2)步骤(1)得到的发酵物通过AB-8型大孔树脂吸附,用体积分数为40%的乙醇溶液洗脱,收集3倍柱体积洗脱液,再经活性炭脱色、浓缩、干燥得所述复方中药提取物(7.6g,以下简称产品A)。
实施例2
(1)取干燥的苦参500g、陈皮100g、秦皮300g,将其粉碎后,加入去离子水(7.2kg)、纤维素酶9g和木瓜蛋白酶9g混匀,调pH6.0-6.5,于40℃下,发酵2天,过滤得发酵液,发酵液浓缩得发酵物;
(2)步骤(1)得到的发酵物通过D101型大孔树脂吸附,用体积分数为30%的乙醇溶液洗脱,收集5倍柱体积洗脱液,再经活性炭脱色、浓缩、干燥得所述复方中药提取物(12.8g,以下简称产品B)。
实施例3
(1)取干燥的苦参500g、陈皮50g、秦皮200g,将其粉碎后,加入蒸馏水(4.5kg)、纤维素酶4.5g和木瓜蛋白酶4.5g混匀,调pH5.5-6.0,于35℃下,发酵2.5天,过滤得发酵液,发酵液浓缩得发酵物;
(2)步骤(1)得到的发酵物通过HPD100型大孔树脂吸附,用体积分数为35%的乙醇溶液洗脱,收集4倍柱体积洗脱液,再经活性炭脱色、浓缩、干燥得所述复方中药提取物(10.4g,以下简称产品C)。
实施例4
(1)取干燥的苦参300g、陈皮50g,将其粉碎后,加入蒸馏水(2.75kg)、纤维素酶2.75g和木瓜蛋白酶2.75g混匀,调pH5.5-6.0,于30℃下,发酵3天,过滤得发酵液,发酵液浓缩得发酵物;
(2)步骤(1)得到的发酵物通过AB-8型大孔树脂吸附,用体积分数为40%的乙醇溶液洗脱,收集3倍柱体积洗脱液,再经活性炭脱色、浓缩、干燥得所述复方中药提取物(3.2g,以下简称产品D)。
实施例5
(1)取干燥的陈皮50g、秦皮200g,将其粉碎后,加入蒸馏水(2.75kg)、纤维素酶2.75g和木瓜蛋白酶2.75g混匀,调pH5.5-6.0,于30℃下,发酵3天,过滤得发酵液,发酵液浓缩得发酵物;
(2)步骤(1)得到的发酵物通过AB-8型大孔树脂吸附,用体积分数为40%的乙醇溶液洗脱,收集3倍柱体积洗脱液,再经活性炭脱色、浓缩、干燥得所述复方中药提取物(2.1g,以下简称产品E)。
实施例6
(1)取干燥的苦参300g、秦皮200g,将其粉碎后,加入蒸馏水(2.75kg)、纤维素酶2.75g和木瓜蛋白酶2.75g混匀,调pH5.5-6.0,于30℃下,发酵3天,过滤得发酵液,发酵液浓缩得发酵物;
(2)步骤(1)得到的发酵物通过AB-8型大孔树脂吸附,用体积分数为40%的乙醇溶液洗脱,收集3倍柱体积洗脱液,再经活性炭脱色、浓缩、干燥得所述复方中药提取物(5.4g,以下简称产品F)。
实施例7
(1)取干燥的苦参300g、陈皮10g、秦皮200g,将其粉碎后,加入蒸馏水(2.75kg)、纤维素酶2.75g和木瓜蛋白酶2.75g混匀,调pH5.5-6.0,于30℃下,发酵3天,过滤得发酵液,发酵液浓缩得发酵物;
(2)步骤(1)得到的发酵物通过AB-8型大孔树脂吸附,用体积分数为40%的乙醇溶液洗脱,收集3倍柱体积洗脱液,再经活性炭脱色、浓缩、干燥得所述复方中药提取物(6.2g,以下简称产品G)。
实施例8
取干燥的苦参300g、陈皮50g、秦皮200g,将其粉碎后,加入蒸馏水(2.75kg)混匀,调pH5.5-6.0,于30℃下,浸泡3天,过滤得滤液,滤液浓缩后通过AB-8型大孔树脂吸附,用体积分数为40%的乙醇溶液洗脱,收集3倍柱体积洗脱液,再经活性炭脱色、浓缩、干燥得所述复方中药提取物(4.6g,以下简称产品H)。
实施例9抗菌活性测试
分别将产品A-H配置成50μg/mL的溶液,用抑菌圈法测试其对大肠杆菌E.coliATCC25922、金黄色葡萄球菌S.aureus ATCC25923的抑菌活性。结果见表1。

实施例10产品A-H对二甲基硫醚(Me2S)的清除实验
用无水乙醇配置450mL质量分数为1%的二甲基硫醚溶液,均分成9份(每份50mL),分别向其中的8份加入产品A-H各3mg,第9份二甲基硫醚溶液不加任何样品作为对照,含A-H的8份溶液于37℃下,反应1小时,以气相色谱-质谱联用(GC-MS)检测二甲基硫醚的含量,结果见表2。

实施例11
按照重量份称取产品A 1份、氟化钠0.02份、十二烷基硫酸钠7份、山梨醇5份、水10份搅拌形成混合溶液,再按照重量份加入碳酸钙20份、羧甲基纤维素钠2份、薄荷油3份,搅拌研磨0.5小时,真空脱气制膏、(存膏、灌装)包装即得所述杀菌除臭中药牙膏。
实施例12
按照重量份称取产品B 2份、叶绿素0.05份、月桂酰肌氨酸钠10份、山梨醇5份、水15份搅拌形成混合溶液,再按照重量份加入碳酸钙20份、二氧化硅10份、黄原胶1份、海澡酸钠1份、薄荷醇2份、艾叶香精3份,搅拌研磨1.0小时,真空脱气制膏、(存膏、灌装)包装即得所述杀菌除臭中药牙膏。
实施例13
按照重量份称取产品C 1份、氟化钠0.03份、酚基谷氨酸钠8份、甘油8份、水10份搅拌形成混合溶液,再按照重量份加入碳酸钙20份、三水合氧化铝2份、卡拉胶2.5份、薄荷油2份、薄荷醇2份,搅拌研磨45分钟,真空脱气制膏、(存膏、灌装)包装即得所述杀菌除臭中药牙膏。
实施例11-13中的重量份,每份表示10g。
Claims (1)
1.一种复方中药提取物在清除挥发性硫化物中的应用,其特征在于所述挥发性硫化物选自二甲基硫醚;所述复方中药提取物,按重量份计复方中药的配方为苦参3-5份、陈皮0.5-1份、秦皮2-3份;
所述复方中药提取物的制备方法包括如下步骤:
(1)按配方量取干燥的苦参、陈皮、秦皮,将其粉碎后,加入水、适量的纤维素酶和木瓜蛋白酶混匀,调pH5.5-6.5,于30-40℃下,发酵2-3天,过滤得发酵液,发酵液浓缩得发酵物;
(2)步骤(1)得到的发酵物通过大孔树脂吸附,用体积分数为30-40%的乙醇溶液洗脱,收集3-5倍柱体积洗脱液,再经活性炭脱色、浓缩、干燥得所述复方中药提取物;
步骤(1)中水选自蒸馏水或去离子水,水的质量用量为苦参、陈皮、秦皮质量之和的5-8倍,纤维素酶的质量用量为苦参、陈皮、秦皮质量之和的0.5-1.0%,木瓜蛋白酶与纤维素酶的质量比为1:1。
