本发明公开了一种水飞蓟祛痘修痕凝胶,属于化妆品以及消毒产品技术领域;每100份成品由下述重量份数的原料制成:水飞蓟精华3~8份,植物提取液8~13份,丙二醇25~40份,卡波U20 0.5~1份,维生素0.5~1份,己脒定二羟乙基磺酸钾0.02~0.2份,三乙醇胺0.5~1份,防腐剂0.6~1份,余量为水。其制备方法为:1)将三乙醇胺用水稀释得备料A;2)将水飞蓟精华与丙二醇制得备料B;3)将卡波U20、水、己脒定二羟乙基磺酸钾,制得备料C;4)利用备料A、备料C中、备料B,以及其他剩余的原料,制备得成品。通过水飞蓟精华与己脒定二羟乙基磺酸钾及植物混合提取液的合理复配,具有良好的祛痘修痕作用,而且安全不刺激,无肌肤依赖性和副作用,消费者可以长期使用。

背景技术
水飞蓟(Silybum marianum)作为药用植物具有近2000年的历史。水飞蓟精华(silymarin),是从水飞蓟的种子的得到的水飞蓟提取物,其CAS号为:65666-07-1,分子式:C25H22O10,分子量:482.44,分子结构式为:
在古代用于治疗包括肝炎、肝硬化在内的肝脏和胆囊的一系列病症的草药,作为抗野生蘑菇、酒精、化学品和环境毒素的肝保护剂。水飞蓟宾是水飞蓟精华(silymarin)的主要成分(为水飞蓟素A和水飞蓟素B的50∶50的混合物的形式)。其它的组分包括异水飞蓟宾(异水飞蓟素A和异水飞蓟素B),水飞蓟宁,水飞蓟亭,异水飞蓟亭,黄杉素及其他。
水飞蓟精华(silymarin)为少数几个在临床上广泛使用的药物之一,并且经过国际医学界近60年的临床检验,证明该药的确切疗效且无任何毒副作用,是目前发现唯一没有任何毒副作用的药物成分,美国食品药品管理局FDA收录成分。
1、抗自由基活性
水飞蓟精华对于由CCl4、半乳糖胺、醇类和其它毒素造成的细胞损害有保护作用。实验表明水飞蓟精华对烷氧基自由基有非常强的清除作用;水飞蓟精华抑制luminol-过氧化酶产生的化学发光,则表示其抗超氧阴离子的作用。

通过灭活自由基,水飞蓟精华阻碍自由基与不饱和脂肪酸的作用。实验表明·OH自由基所致的脂质过氧化被水飞蓟精华减少72%。·OH自由基可攻击脂肪酸、蛋白质核酸,导致组织损伤。在所有活性氧自由基中,·OH引起的损害最大。在等摩尔浓度时,水飞蓟精华在线粒体中抗脂质过氧化作用为α-生育酚的10倍。
3、抗脂氧酶作用
水飞蓟精华在实验模型中,其抗溃疡作用优于雷尼替丁,溃疡指数(ulcerindex)为(0.58士0.34)%。在体外实验也证明水飞蓟精华为脂氧酶抑制剂和前列腺素合成酶抑制剂。研究水飞蓟精华在各种炎症模型上和白细胞迁移的药理作用,表明水飞蓟精华对由角叉菜胶引起急性炎症的抑制作用,其ED50为62.42mg/kg。水飞蓟精华对由甲苯引起的小鼠耳水肿型作用,依据临床给药不同而产生不同药效。局部给药时的抑制率为44.52%,而吲哚美辛为35.96%。水飞蓟精华在高剂量时能抑制由角叉菜胶所致的白细胞浸润,ED50为73.34mg/kg,而且中性白细胞迁移也明显减少。这表明水飞蓟精华有较强的抗炎作用,可作为抗炎药物使用。
4、抗GSH徘空作用
GSH被认为是对抗化学诱导细胞毒最重要的生物分子。GSH能参与活性异生物质的清除、氢过氧化物的还原或直接清除自由基。GSH在异生物质诱导细胞损伤中的保护作用已经为实验所证实。水飞蓟精华具有对异生物质,包括毒伞素(phalloidin)、鹅膏蕈碱(α-amanitin)、镨(praseodymium)、CCl4、乙硫酰胺(ethione)引起细胞毒性具有保护作用。水飞蓟精华不同于其它抗氧化的黄酮类似物,它可以增加组织中GSH水平,增加氨基酸的通透性,可以保护上述组织免受异生物质的损害。
5、抗皮肤肿瘤作用
研究显示水飞蓟精华对引起小鼠皮肤肿瘤增长的若干个皮肤肿瘤起动因子(EGFR)的防护作用,实验证明这些皮肤肿瘤增长因子的作用完全可以被水飞蓟精华抑制。用水飞蓟精华处理过的细胞,完全抑制了配基诱导EGFR激活,并且不影响蛋白质水平。结果表示水飞蓟精华对皮肤病的化学防护作用是由EGFR信息修补体系调节所致。对LPS诱导的痤疮丙酸杆菌致敏的小鼠损伤有保护作用。
发明内容
基于现有技术的不足,本发明所要解决的技术问题是:提供了一种以水飞蓟精华(silymarin)为主要功效成分的祛痘修痕凝胶。
为了解决上述技术问题,本发明采用的技术方案为:一种水飞蓟祛痘修痕凝胶,每100份水飞蓟祛痘修痕凝胶由下述重量份数的原料制成:水飞蓟精华3~8份,植物提取液8~13份,丙二醇25~40份,卡波U200.5~1份,维生素0.5~1份,己脒定二羟乙基磺酸钾0.02~0.2份,三乙醇胺0.5~1份,防腐剂0.6~1份,余量为水。
水飞蓟精华含量低于3%时效果不明显,超过8%时溶解度不佳,皮肤吸收不理想,对中药的浪费比较大。
所述的一种水飞蓟祛痘修痕凝胶,所述的植物提取液为苦参提取物,野菊花提取物,积雪草提取物和忍冬提取物;
所述的维生素为D-泛醇;
所述的防腐剂为苯氧乙醇和乙基己基甘油。
优选地,所述的水飞蓟祛痘修痕凝胶,每100份水飞蓟祛痘修痕凝胶由下述重量份数的原料制成:水飞蓟精华(silymarin)3~8份,苦参提取物2~4份,野菊花提取物2~3份,积雪草提取物2~3份,忍冬提取物2~3份,丙二醇25~40份,卡波U200.5~1份,D-泛醇0.5~1份,己脒定二羟乙基磺酸钾0.02~0.2份,三乙醇胺0.5~1份,苯氧乙醇0.3~0.5份,乙基己基甘油0.3~0.5份,余量为水。

进一步优选地,所述的水飞蓟祛痘修痕凝胶,每100份水飞蓟祛痘修痕凝胶由下述重量份数的原料制成:水飞蓟精华(silymarin)5份,苦参提取物2.8份,野菊花提取物2.8份,积雪草提取物2.6份,忍冬提取物2.5份,丙二醇32份,卡波U200.7份,D-泛醇0.8份,己脒定二羟乙基磺酸钾0.08份,三乙醇胺0.68份,苯氧乙醇0.35份,乙基己基甘油0.35份,余量为水。

为解决第二个技术问题,采用的技术方案是:一种水飞蓟祛痘修痕凝胶的制备方法,包括以下步骤:
按重量份数称取各原料;
1)将三乙醇胺用水稀释9~11倍,得到备料A;
2)将水飞蓟精华(silymarin)和丙二醇预混合,通过分散机均匀分散,得到备料B;
3)将卡波U20润湿分散于余量的水中,然后加入己脒定二羟乙基磺酸钾,搅拌混匀,然后加热至50~100℃,在真空度0.05~0.09MPa的条件下恒温搅拌10~60分钟,得到备料C;
4)将备料A加入备料C中,继续于温度50~100℃、真空度0.05~0.09MPa的条件下,恒温搅拌至呈均匀胶体后,冷却至40~50℃,加入备料B,以及其他剩余的原料,继续于温度40~50℃、真空度0.05~0.09MPa的条件下,恒温搅拌至呈均匀胶体后,冷却至室温,即得水飞蓟祛痘修痕凝胶。
优选地,所述的水飞蓟祛痘修痕凝胶的制备方法,包括以下步骤:
1)按重量份数称取各原料;将三乙醇胺用水稀释10倍,得到备料A;
2)将水飞蓟精华(silymarin)和丙二醇预混合,通过分散机均匀分散,得到备料B;
3)将卡波U20润湿分散于余量的水中,然后加入己脒定二羟乙基磺酸钾,搅拌混匀,然后加热至80℃,在真空度0.08MPa的条件下恒温搅拌30分钟,得到备料C;
4)将备料A加入备料C中,继续于温度80℃、真空度0.08MPa的条件下,恒温搅拌至呈均匀胶体后,冷却至45℃,加入备料B,以及其他剩余的原料,继续于温度45℃、真空度0.09MPa的条件下,恒温搅拌至呈均匀胶体后,冷却至室温,即得水飞蓟祛痘修痕凝胶。
上述原料除水飞蓟精华(silymarin)外均为普通市售产品。
所述的水飞蓟精华(silymarin)是镇江壹心生物工程技术有限公司自主研发,主要是以水飞蓟粕为原料,将水飞蓟粕按1:3~1:4比例加入丙酮,用提取罐和浓缩罐进行物料回流,提取8~10小时,物料温度控制在25~30℃,同时用冷凝器回收丙酮溶剂,将溶剂过滤,进行浓缩2~4小时,温度60~65℃,蒸气压力1.5兆帕,同时用冷凝器回收丙酮溶剂,得到糊状浸膏;将浸膏放到夹套锅内,加入1:5的反渗透水,水温加热到80~90℃,搅拌1~2小时,放掉水,这样反复重复5~6次,丙酮溶于水,水飞蓟素不溶于水,经过反复5~6次后,丙酮的溶剂可去掉90%以上,同时可将剩余的水飞蓟残油去除;将水飞蓟浸膏放到真空干燥罐内进行真空干燥,干燥温度90~120℃,负压0.8兆帕,连续进行4~6小时,目的是把丙酮的气体及水份用真空泵吸出,得到含水量在5%左右的水飞蓟素干粉块,将其粉碎成80~90目,再放入真空罐中进行二次真空,干燥2~4小时,温度80~110度,负压0.8兆帕以上,丙酮溶剂可去掉95%,即得到水飞蓟素的干粉,便于保存。
所述的己脒定二羟乙基磺酸钾(HDI)是一种水溶性的具有广谱抗菌和杀菌性能的阳离子物质,对各种革兰氏阳性菌及阴性菌以及各种霉菌和酵母菌都有很高的杀菌和抑菌性能,特别是对引起头屑的卵状糠秕孢子菌,引起痘痘粉刺的痤疮丙酸杆菌有很好的的抑制和杀灭效果。配合水飞蓟精华(silymarin),达到协同增效去屑、祛痘的功效,而且安全不刺激,无肌肤依赖性和副作用,消费者可以长期使用。
所述的苦参提取物、野菊花提取物、积雪草提取物、忍冬提取物由湖州佳美公司提供。苦参(SOPHORA FLAVESCENS)提取物,野菊(CHRYSANTHEMUM INDICUM)花提取物,积雪草(CENTELLAASIATICA)提取物,忍冬(LONICERAJAPONICA)花提取物,从苦参、野菊花、积雪草、忍冬等多味名贵中药中提取分离得到的活性物质,能减少皮脂分泌,增强皮肤抵抗力,有效预防和治疗不同类型的痤疮,赋予皮肤新的生机与活力。

本发明的有益效果为:相对于现有技术,本发明所述的水飞蓟精华(silymarin)是经过国际医学界近60年的临床检验,具有确切的疗效和无任何毒副作用,同时也提供了一种新的祛痘原料和方法,通过水飞蓟精华与己脒定二羟乙基磺酸钾及植物混合提取液的合理复配,具有良好的祛痘修痕作用,而且安全不刺激,无肌肤依赖性和副作用,消费者可以长期使用。

下述实施例中己脒定二羟乙基磺酸钾购自THOR(托尔国际贸易(上海)有限公司)公司的HDI(CAS:659-40-5),苦参提取物、野菊花提取物、积雪草提取物、忍冬提取物购自湖州佳美公司,卡波U20购自路博润公司,D-泛醇为DSM公司提供,苯氧乙醇和乙基己基甘油是购自THOR公司的PEHG。
实施例1
一种水飞蓟祛痘修痕凝胶,由下述重量份数的原料制成:水飞蓟精华(silymarin)5份,苦参提取物3份,野菊花提取物3份,积雪草提取物3份,忍冬提取物3份,丙二醇32份,卡波U200.8份,D-泛醇0.8份,己脒定二羟乙基磺酸钾0.1份,三乙醇胺0.8份,苯氧乙醇0.35份,乙基己基甘油0.35份,水47.8份。
水飞蓟祛痘修痕凝胶的制备方法,包括以下步骤:
1)按重量份数称取各原料;将三乙醇胺用水稀释10倍,得到备料A;
2)将水飞蓟精华(silymarin)和丙二醇预混合,通过分散机均匀分散,得到备料B;
3)将卡波U20润湿分散于余量的水中,然后加入己脒定二羟乙基磺酸钾,搅拌混匀,然后加热至80℃,在真空度0.08MPa的条件下恒温搅拌30分钟,得到备料C;

一种水飞蓟祛痘修痕凝胶,由下述重量份数的原料制成:水飞蓟精华(silymarin)6份,苦参提取物2.5份,野菊花提取物2.5份,积雪草提取物2.5份,忍冬提取物2.5份,丙二醇34份,卡波U200.7份,D-泛醇0.8份,己脒定二羟乙基磺酸钾0.1份,三乙醇胺0.7份,苯氧乙醇0.35份,乙基己基甘油0.35份,水47份。
水飞蓟祛痘修痕凝胶的制备方法,包括以下步骤:
1)按重量份数称取各原料;将三乙醇胺用水稀释10倍,得到备料A;
2)将水飞蓟精华(silymarin)和丙二醇预混合,通过分散机均匀分散,得到备料B;
3)将卡波U20润湿分散于余量的水中,然后加入己脒定二羟乙基磺酸钾,搅拌混匀,然后加热至85℃,在真空度0.09MPa的条件下恒温搅拌30分钟,得到备料C;

一种水飞蓟祛痘修痕凝胶,由下述重量份数的原料制成:水飞蓟精华(silymarin)4份,苦参提取物2.0份,野菊花提取物2.0份,积雪草提取物2.0份,忍冬提取物2.0份,丙二醇25份,卡波U200.6份,D-泛醇1份,己脒定二羟乙基磺酸钾0.08份,三乙醇胺0.6份,苯氧乙醇0.35份,乙基己基甘油0.35份,水60.02份。
水飞蓟祛痘修痕凝胶的制备方法,包括以下步骤:
1)按重量份数称取各原料;将三乙醇胺用水稀释10倍,得到备料A;
2)将水飞蓟精华(silymarin)和丙二醇预混合,通过分散机均匀分散,得到备料B;
3)将卡波U20润湿分散于余量的水中,然后加入己脒定二羟乙基磺酸钾,搅拌混匀,然后加热至75℃,在真空度0.08MPa的条件下恒温搅拌30分钟,得到备料C;
4)将备料A加入备料C中,继续于温度75℃、真空度0.08MPa的条件下,恒温搅拌至呈均匀胶体后,冷却至45℃,加入备料B,以及苦参提取物,野菊花提取物,积雪草提取物,忍冬提取物,D-泛醇,苯氧乙醇和乙基己基甘油,继续于温度45℃、真空度0.09MPa的条件下,恒温搅拌至呈均匀胶体后,冷却至室温,即得水飞蓟祛痘修痕凝胶。
实施例1所制得的水飞蓟祛痘修痕凝胶制品经THOR实验室按照《消毒技术规范》要求的方法检测,抗菌效果如表1。

从表1中可以明显看出,实施例1所制得的水飞蓟祛痘修痕凝胶具有良好的抗、抑菌效果。
实施例1所制得的水飞蓟祛痘修痕凝胶,经过800名痘痘自愿者(男女各400人,年龄在18~48周岁)的跟踪使用效果调查,分类及结果如下:
经检测,实施例2~3所制得的水飞蓟祛痘修痕凝胶均具有良好的祛痘修护,安全无副作用。

1.一种水飞蓟祛痘修痕凝胶,每100份水飞蓟祛痘修痕凝胶由下述重量份数的原料制成:水飞蓟精华3~8份,植物提取液8~13份,丙二醇25~40份,卡波U20 0.5~1份,D-泛醇0.5~1份,己脒定二羟乙基磺酸钾0.02~0.2 份,三乙醇胺0.5~1份,防腐剂0.6~1份,余量为水;
所述的水飞蓟精华,是以水飞蓟粕为原料,将水飞蓟粕按1:3~1:4比例加入丙酮,用提取罐和浓缩罐进行物料回流,提取8~10小时,物料温度控制在25~30℃,同时用冷凝器回收丙酮溶剂,将溶剂过滤,进行浓缩2~4小时,温度60~65℃,蒸气压力1. 5兆帕,同时用冷凝器回收丙酮溶剂,得到糊状浸膏;将浸膏放到夹套锅内,加入1:5的反渗透水,水温加热到80~90℃,搅拌1~2小时,放掉水,重复5~6次;将水飞蓟浸膏放到真空干燥罐内进行真空干燥,干燥温度90~120℃,负压0. 8兆帕,连续进行4~6小时,得到含水量在5%的水飞蓟素干粉块,将其粉碎成80~90目,再放入真空罐中进行二次真空,干燥2~4小时,温度80~110度,负压0. 8兆帕以上,丙酮溶剂去掉95%,得到水飞蓟素的干粉;

6.权利要求1-5任一项所述的一种水飞蓟祛痘修痕凝胶的制备方法,包括以下步骤:
按重量份数称取各原料;
1)将三乙醇胺用水稀释9~11倍,得到备料A;
2)将水飞蓟精华和丙二醇预混合,均匀分散,得到备料B;
3)将卡波U20润湿分散于余量的水中,然后加入己脒定二羟乙基磺酸钾,搅拌混匀,然后加热至50~100℃,在真空度0.05~0.09 MPa的条件下恒温搅拌10~60分钟,得到备料C;
4)将备料A加入备料C中,继续于温度50~100℃、真空度0.05~0.09 MPa的条件下,恒温搅拌至呈均匀胶体后,冷却至40~50℃,加入备料B,以及其他剩余的原料,继续于温度40~50℃、真空度0.05~0.09 MPa的条件下,恒温搅拌至呈均匀胶体后,冷却至室温,即得水飞蓟祛痘修痕凝胶。
