Abstract
本发明属于医用生物材料技术领域,公开了一种用于腹壁缺损修复的复合生物补片,该种复合生物补片包含生物补片层以及覆膜于该生物补片层一侧表面的白芨纤维网层;且该白芨纤维网层为静电纺丝纳米级白芨纤维网。本发明通过将纳米白芨纤维网层与传统生物补片覆膜结合,使得到的复合生物补片的抗感染能力、创面修复能力得到有效提高;同时,该复合生物补片植入机体后的力学强度变化始终高于所需的力学强度,可加速组织重建,有助于解决生物补片现存技术瓶颈,有效改善腹壁缺损修复的治疗效果。
Description
技术领域
本发明涉及一种医用生物材料,尤其是一种医用生物补片。
背景技术
生物补片的研发和应用是当前疝和腹壁外科研究的一个重要方向。生物补片是指取自同种、异种的组织,经脱细胞处理去除组织中含有的各种细胞而完整保留细胞外基质的三维框架结构并能用于修复人体软组织的材料。其根据组织来源可分为同种异体材料如真皮脱细胞基质、羊膜、硬脑膜等,异种异体材料如猪小肠黏膜下层、牛马的心包、牛腹膜等。
生物补片植入后,随着宿主组织的长入.生物补片逐步降解,最终生物补片完全被宿主组织代替。因此,生物补片修复机制是“内源性组织再生”,其诱导干细胞进入生物补片,分泌细胞外基质替代降解的植入物。生物补片具有一定的耐受感染能力,这得益于其能实现早期局部快速再血管化、吞噬细胞早期进入,使细菌生物膜难以形成,故生物补片可用于伴有污染或潜在感染的创面。
但目前的生物补片尚存在一些亟待解决的难题。生物补片植入机体后补片本身降解的同时又有组织重建,如果这两者的合成力学曲线在植入体内的力学强度变化始终高于植入体的力学强度,那么该补片修补能达到预期目的;反之,修补就会失败。究其原因,无外乎以下两个原因:1、生物补片降解过快,而组织重建相对缓慢,导致这两者的合成力学曲线在某个阶段低于腹壁力学强度。2、生物补片降解虽慢,而组织重建相对不足,最终仍导致修补失败。
发明内容
本发明的目的在于提供一种适用于疝和腹壁外科修复的复合生物补片,有效改善临床腹壁缺损修复的治疗效果。
为解决上述技术问题,本发明提供了一种用于腹壁缺损修复的复合生物补片,该复合生物补片包含生物补片层以及覆膜于生物补片层一侧表面的白芨纤维网层。我国传统中药白芨在临床上对促进伤口愈合疗效独特,能加速组织重建。近年来,通过现代医学研究进一步确切证实,白芨的有效成分具有提高创面组织中巨噬细胞、促进血管内皮细胞粘附生长的功能,可显著提高细胞的增殖能力,同时还有抑菌止血的功能。本发明通过将白芨纤维网层与传统生物补片覆膜结合,使得到的复合生物补片在植入体内后的抗感染能力、创面修复能力得到有效提高。此外,本发明所提供的复合生物补片植入机体后的力学强度变化始终高于所需的力学强度,可加速组织重建,有助于解决生物补片现存瓶颈,进一步改善腹壁缺损修复的治疗效果。
优选地,本发明所提供的用于腹壁缺损修复的复合生物补片中,覆膜于传统生物补片表面的白芨纤维网层为纳米级白芨纤维网;进一步地,纳米级白芨纤维网的纤维直径为100~500纳米,该纳米级白芨纤维网的厚度为0.2~1毫米。纳米级的白芨纤维网层由于材料的微小直径使其具有较大的比表面积,在药物缓释应用中有极大地优势,有利于更好地发挥白芨纤维层对传统生物补片在修复效果上的改善作用。
优选地,本发明所提供的用于腹壁缺损修复的复合生物补片中,纳米级白芨纤维网为静电纺丝纳米级白芨纤维网。静电纺丝,也称电纺丝,是一种利用聚合物溶液或熔体在强电场作用下形成喷射流进行纺丝加工的工艺。制得的纤维直径可以达到纳米量级。还可在其中引入无机粒子、活性成分、药物等来改善材料的性能,通过结构、成分的控制,达到促进引导组织再生功能的作用,电纺技术制作的功能化纳米电纺丝材料,在组织工程、创伤敷料、酶的固定化和药物(基因)载体等生物医学方面得到了广泛的应用。本发明所提供的复合生物补片,采用以静电纺丝技术制备的纳米级白芨纤维网与传统生物补片覆膜结合,进一步优化了复合生物补片的医用性能。
进一步地,本发明所提供的用于腹壁缺损修复的复合生物补片中,生物补片层为市场购买的常规生物补片。具体可以是同种异体材料型补片或异种异体材料型补片。其中,同种异体材料型补片可以为真皮脱细胞基质来源补片、羊膜来源补片或硬脑膜来源补片等;异种异体材料型补片为猪小肠黏膜下层来源补片、牛心包来源补片、马心包来源补片或牛腹膜来源补片等。例如可以是购买自库克(Cook)、清源伟业、冠昊等公司的生物补片。

本发明的第一实施方式所涉及的用于腹壁缺损修复的复合生物补片包含生物补片层1以及覆膜于生物补片层1一侧表面的白芨纤维网层2。
上述的白芨纤维网层2的厚度为0.2毫米,纳米级白芨纤维的纤维直径为100纳米,且该纳米级白芨纤维网采用静电纺丝制备而得。上述的生物补片层1购自库克(Cook)公司,商品名为百得塞,为猪小肠粘膜下层脱细胞基质生物补片。
本发明的第二实施方式所涉及的用于腹壁缺损修复的复合生物补片也包含生物补片层以及覆膜于生物补片层一侧表面的纳米级白芨纤维网层。
本实施方式中的纳米级白芨纤维网层的厚度为1毫米,纳米级白芨纤维的纤维直径为300纳米,且该纳米级白芨纤维网采用静电纺丝制备而得。本实施方式中的生物补片层购自清源伟业公司。
本发明的第三实施方式所涉及的用于腹壁缺损修复的复合生物补片也包含生物补片层以及覆膜于生物补片层一侧表面的纳米级白芨纤维网层。
本实施方式中的纳米级白芨纤维网层的厚度为0.6毫米,纳米级白芨纤维的纤维直径为500纳米,且该纳米级白芨纤维网采用静电纺丝制备而得。本实施方式中的生物补片层购自冠昊公司。
本领域的普通技术人员可以理解,上述各实施方式是实现本发明的具体实施例,而在实际应用中,可以在形式上和细节上对其作各种改变,而不偏离本发明的精神和范围。

3.根据权利要求2所述的用于腹壁缺损修复的复合生物补片,其特征在于,所述纳米级白芨纤维网的纤维直径为100~500纳米。
4.根据权利要求2所述的用于腹壁缺损修复的复合生物补片,其特征在于,所述纳米级白芨纤维网的厚度为0.2~1毫米。
