本发明涉及一种植物多酚改性黄精膳食纤维的制备方法,属于本发明涉及膳食纤维技术领域。
技术背景
1976年,英国营养病学专家H.C.Trowell将“不被人体消化吸收的多糖类碳水化合物与木质素”定义为“膳食纤维”。膳食纤维在现代营养学中被称为“第七大营养素”。膳食纤维有预防肥胖症、防治糖尿病、调节血脂水平、预防结肠癌、抑制有毒发酵产物、预防肠憩室和乳腺癌等生理功能。膳食纤维中的酸性多糖类具有较强的阳离子交换功能力,能与肠道中的Na+、K+进行交换,对消化道的pH值、渗透压及氧化电位产生影响,使血液中的Na+、K+比值降低,产生降血压作用。
膳食纤维提取改性活化的方法有很多种,常用的膳食纤维改性处理有生物技术方法(酶法、发酵法)、化学法(主要有酸法、碱法)和物理法(机械降解处理法)等。其中,利用化学法如酸法或碱法通过反复的浸泡和频繁的热处理过程,会明显减少纤维素产品的持水力和膨胀性,而且所排放的酸性或碱性工业废液对环境造成严重污染;利用物理法如机械降解处理法膳食纤维的得率不高;利用生物技术方法如酶法或发酵法处理综合效果最佳。
黄精是一种传统的药食同源性植物原料,富含淀粉、维生素、矿物质元素和多种活性物质。黄精具有补脾润肺、养阴生津、补中益气等功效和抑菌、抗病毒、抗肿瘤、抗衰老以及降糖降脂等作用,黄精的主要活性成分有糖类、甾体皂苷、黄酮、蒽醌类化合物、氨基酸和微量元素。但现有文献报道中很少关于研究提取、改性黄精膳食纤维。
植物多酚是多羟基酚类化合物的总称,又称为单宁,广泛存在于蔬菜、水果、豆类、谷物类、茶等植物中。多酚的独特结构赋予了它一些列独特的化学性质,使其具有抗肿瘤、抗氧化、抗动脉硬化、防治冠心病与中风等心脑血管疾病以及抗菌等多种生理功能。本发明利用植物多酚与黄精膳食纤维之间的反应,将二者的功效结合起来。
发明内容
为克服现有技术存在不足,本发明旨在提供一种植物多酚改性黄精膳食纤维的制备方法,包括如下步骤:
(1)将100重量份洗净后的黄精切片、500~1000重量份的水加入到磨浆机中磨浆18~30min,然后50~70℃加热下搅拌0.5~1h,冷却得到浆液;将浆液打入高压均质机中,1000~2000bar压力下剪切均质3~5次,得到稳定浆液。
(2)向(1)中稳定浆液中加入0.01~0.03重量份酶活为1200~2400AU的蛋白酶,pH=5.0~7.0、搅拌下、40~60℃酶解1~3h,酶解充分后煮沸0.5~1h灭酶,然后冷却。
(3)向经(2)步骤处理后的浆液中加入0.05~0.3重量份30000~60000u/mlα-淀粉酶,pH=5.0~7.0、搅拌下、40~60℃酶解1~3h,酶解充分后煮沸0.5~1h灭酶;然后60~80℃下加入15~30份植物多酚反应3~6h,经冷却、离心过滤后,沉淀即为改性黄精膳食纤维粗产品。
(4)将步骤(3)中所得改性黄精膳食纤维粗产品加水水洗,过滤,50~65℃干燥沉淀,再经粉碎、过筛后即得植物多酚改性黄精膳食纤维。
所处的水为蒸馏水或去离子水。
所述的乙醇为分析级。
所述的植物多酚为茶多酚、苹果多酚、葡萄多酚、花青素、儿茶素、栎精、没食子酸、鞣花酸、熊果苷中的至少一种。
所述的过筛为过100~400μm筛。
相对于现有技术,本发明具有以下优点:(1)利用植物多酚与多糖的反应,首次制备了植物多酚改性黄精膳食纤维,所述的改性黄精膳食纤维兼有黄精膳食纤维、植物多酚二者的优点;(2)采用两步酶处理分离法制备黄精膳食纤维,减少传统的粗分离法、化学分离法对膳食纤维中有效成分以及品质的破坏和对环境的污染,同时利用植物多酚可将可溶性黄精膳食纤维沉降,提高黄精的综合利用价值;(3)所述的制备方法中的时间、温度、pH值、均质压力等协同作用,使得所述的植物多酚改性黄精膳食纤维性能最优,且适口性较好。

下面结合实施例对本发明的一种植物多酚改性黄精膳食纤维的制备方法做进一步的描述。可以理解的是,此处所描述的具体实施例仅仅用于解释相关发明,而非对该发明的限定。
实施例1
一种植物多酚改性黄精膳食纤维的制备方法,包括如下步骤:
(1)将100重量份洗净后的黄精切片、600重量份的水加入到磨浆机中磨浆20min,然后55℃加热下搅拌0.6h,冷却得到浆液;将浆液打入高压均质机中,1200bar压力下剪切均质4次,得到稳定浆液。
(2)向(1)中稳定浆液中加入0.02重量份酶活为1600AU的蛋白酶,pH=6.0、搅拌下、50℃酶解1.5h,酶解充分后煮沸0.7h灭酶,然后冷却。
(3)向经(2)步骤处理后的浆液中加入0.15重量份40000u/mlα-淀粉酶,pH=6.0、搅拌下、48℃酶解1.5h,酶解充分后煮沸0.7h灭酶;然后80℃下加入30份茶多酚反应3h,再经冷却、离心过滤后,沉淀即为改性黄精膳食纤维粗产品。
(4)将步骤(3)中所得改性黄精膳食纤维粗产品加水水洗,过滤,62℃干燥沉淀,再经粉碎、过筛后即得植物多酚改性黄精膳食纤维。
实施例2
一种植物多酚改性黄精膳食纤维的制备方法,包括如下步骤:
(1)将100重量份洗净后的黄精切片、1000重量份的水加入到磨浆机中磨浆30min,然后70℃加热下搅拌0.5h,冷却得到浆液;将浆液打入高压均质机中,2000bar压力下剪切均质3次,得到稳定浆液。
(2)向(1)中稳定浆液中加入0.03重量份酶活为2400AU的蛋白酶,pH=7.0、搅拌下、60℃酶解1h,酶解充分后煮沸0.5h灭酶,然后冷却。
(3)向经(2)步骤处理后的浆液中加入0.3重量份30000u/mlα-淀粉酶,
pH=7.0、搅拌下、60℃酶解1h,酶解充分后煮沸0.5h灭酶;然后60℃下加入24份苹果多酚反应6h,再经冷却、离心过滤后,沉淀即为改性黄精膳食纤维粗产品。
(4)将步骤(3)中所得改性黄精膳食纤维粗产品加水水洗,过滤,55℃干燥沉淀,再经粉碎、过筛后即得植物多酚改性黄精膳食纤维。
实施例3
一种植物多酚改性黄精膳食纤维的制备方法,包括如下步骤:
(1)将100重量份洗净后的黄精切片、500重量份的水加入到磨浆机中磨浆18min,然后50℃加热下搅拌1h,冷却得到浆液;将浆液打入高压均质机中,1000bar压力下剪切均质5次,得到稳定浆液。
(2)向(1)中稳定浆液中加入0.01重量份酶活为1200AU的蛋白酶,pH=5.0、搅拌下、40℃酶解3h,酶解充分后煮沸1h灭酶,然后冷却。
(3)向经(2)步骤处理后的浆液中加入0.05重量份30000u/mlα-淀粉酶,pH=5.0、搅拌下、40℃酶解3h,酶解充分后煮沸1h灭酶;然后70℃下加入15份茶多酚反应4h,经冷却、离心过滤后,沉淀即为改性黄精膳食纤维粗产品。
(4)将步骤(3)中所得改性黄精膳食纤维粗产品加水水洗,过滤,65℃干燥沉淀,再经粉碎、过筛后即得植物多酚改性黄精膳食纤维。
实施例4
一种植物多酚改性黄精膳食纤维的制备方法,包括如下步骤:
(1)将100重量份洗净后的黄精切片、800重量份的水加入到磨浆机中磨浆26min,然后65℃加热下搅拌0.8h,冷却得到浆液;将浆液打入高压均质机中,1600bar压力下剪切均质4次,得到稳定浆液。
(2)向(1)中稳定浆液中加入0.028重量份酶活为2000AU的蛋白酶,pH=6.0、搅拌下、55℃酶解2.5h,酶解充分后煮沸0.6h灭酶,然后冷却。
(3)向经(2)步骤处理后的浆液中加入0.2重量份50000u/mlα-淀粉酶,pH=6.0、搅拌下、50℃酶解2.5h,酶解充分后煮沸0.8h灭酶;然后75℃下加入15~25份苹果多酚反应5h,经冷却、离心过滤后,沉淀即为改性黄精膳食纤维粗产品。
(4)将步骤(3)中所得改性黄精膳食纤维粗产品加水水洗,过滤,55℃干燥沉淀,再经粉碎、过筛后即得植物多酚改性黄精膳食纤维。
实施例1-4中所述的植物多酚改性黄精膳食纤维的制备方法检测数据如下表:
Claims (5)
1.一种植物多酚改性黄精膳食纤维的制备方法,其特征在于:包括如下步骤:
(1)将100重量份洗净后的黄精切片、500~1000重量份的水加入到磨浆机中磨浆18~30min,然后50~70℃加热下搅拌0.5~1h,冷却得到浆液;将浆液打入高压均质机中,1000~2000bar压力下剪切均质3~5次,得到稳定浆液;
(2)向(1)中稳定浆液中加入0.01~0.03重量份酶活为1200~2400AU的蛋白酶,pH=5.0~7.0、搅拌下、40~60℃酶解1~3h,酶解充分后煮沸0.5~1h灭酶,然后冷却;
(3)向经(2)步骤处理后的浆液中加入0.05~0.3重量份30000~60000u/mlα-淀粉酶,pH=5.0~7.0、搅拌下、40~60℃酶解1~3h,酶解充分后煮沸0.5~1h灭酶;然后60~80℃下加入15~30份植物多酚反应3~6h,经冷却、离心过滤后,沉淀即为改性黄精膳食纤维粗产品;
(4)将步骤(3)中所得改性黄精膳食纤维粗产品加水水洗,过滤,50~65℃干燥沉淀,再经粉碎、过筛后即得植物多酚改性黄精膳食纤维。
2.根据权利要求1所述的一种植物多酚改性黄精膳食纤维的制备方法,其特征在于:所处的水为蒸馏水或去离子水。
3.根据权利要求1所述的一种植物多酚改性黄精膳食纤维的制备方法,其特征在于:所述的乙醇为分析级。
4.根据权利要求1所述的一种植物多酚改性黄精膳食纤维的制备方法,其特征在于:所述的植物多酚为茶多酚、苹果多酚、葡萄多酚、花青素、儿茶素、栎精、没食子酸、鞣花酸、熊果苷中的至少一种。
5.根据权利要求1所述的一种植物多酚改性黄精膳食纤维的制备方法,其特征在于:所述的过筛为过100~400μm筛。
