Description

Composition containing catfish body surface antibacterial peptide and active polysaccharide for promoting wound healing
Technical Field
The invention relates to a composition for promoting wound healing, which contains catfish body surface antibacterial peptide and active polysaccharide, and application thereof.
Background
At present, some household pharmaceutical preparations for promoting wound healing mainly comprise antibiotics and other components, and do not contain nutrients necessary for wound healing. For example, the wound dressings which are relatively universal in the market are widely accepted by people because of convenient carrying, convenient use, free stretching and no damage to the skin, and more people like to treat wounds by using the wound dressings. However, the existing wound dressings usually have the antibacterial and anti-inflammatory components or mainly contain antibiotics, and the antibiotics have certain potential harm, so that the antibiotics are frequently used to cause bacterial tolerance and induce novel bacteria. However, the development of antibiotics is far less advanced than the rate of evolution of new bacteria, and bacterial infection is likely to be caused in the future without the availability of drugs. In addition, the existing wound dressings do not have the defect of providing necessary nutrients for wound healing.
Along with the improvement of living standard of people, the source of effective components of the medicine is more and more emphasized, if the effective components are derived from natural animal or plant components, the side effect is smaller, and the effective components are popular with wide users.
In view of the above, the present invention is intended to provide improvements to existing drugs for promoting wound healing.
Disclosure of Invention
The invention aims to provide a composition for promoting wound healing, which contains catfish body surface antibacterial peptide and active polysaccharide, wherein the active ingredients of the composition are the catfish body surface antibacterial peptide and the active polysaccharide, and antibiotics are not adopted, so that the composition can prevent the defects caused by abuse of the antibiotics and can provide necessary nutrient substances for wound healing.
In the invention, the catfish (Parasilurus asotus Linnaeus) belongs to the family Parasilurus of class Cyprinidae, has the advantages of sexual activity and strong adaptability to the survival environment, and can generate some antibacterial active substances for resisting the severe environment in body surface mucus and viscera of the catfish in comparison with other kinds of fish by stress, mainly comprising antibacterial peptide, active polysaccharide, C-reactive protein, catamaran, lysozyme, chitinase, interferon, immunoglobulin and the like, and particularly the active substances can generate more substances after being injured. Austin et al (Journal of Fish Diseases,1988,11, 275-277) first reported in 1988 that rainbow trout epidermal mucus had inhibitory effects on infectious pathogens. Hellio et al, International Journal of Antimicrobial Agents,2002,20,214-219, 2002, discovered that various fish epidermal mucus extracts have broad-spectrum Antimicrobial activity. Meanwhile, many scholars have conducted intensive studies on the antibacterial components of the mucus on the skin of fish, and Kitani et al [ Federation of European Biochemical Society,2007,274,125-136] separated the antibacterial protein SSAP on the body surface of the rockfish, studied the molecular structure and antibacterial mechanism, and found that SSAP antibacterial peptide has broad-spectrum antibacterial property and can inhibit G + and G-bacteria simultaneously. Zhao Zhifeng et al (Shanxi agricultural science, 2008,36, 76-80) found that catfish body surface mucus contained protein matter content about 40% of mucus dry weight, and polysaccharide matter about 4.2% of mucus dry weight. Litting and the like [ modern food technology, 2015,31,67-73] research on the bacteriostatic effect and mechanism of catfish body surface mucus extract on pseudomonas aeruginosa, and the research shows that the catfish body surface mucus extract can inhibit the synthesis of certain proteins of thalli, so that the intracellular protein content is reduced and deleted, and the inhibition effect on various bacteria is good.

The components of embodiments of the present invention generally described and illustrated in the figures herein may be arranged and designed in a wide variety of different configurations.
The first embodiment is as follows:
extracting a mixture of catfish body surface antibacterial peptide and active polysaccharide:
5 fresh silurus meridionalis strips are purchased in a market, the silurus meridionalis strips are fed in a water tank for 7 days by using clean water, the clean water for feeding is filtered by using a plurality of layers of gauzes every 12-24 hours, filtrate is collected in a test tube, and the filtrate is stored at the temperature of 0-4 ℃.
Extracting components with molecular weight of 2000-30000 from the filtrate by gel chromatography, freeze drying at-20-50 deg.C to obtain powder or spray drying to obtain block, grinding the block to powder of about 80 meshes, storing in clean brown bottle at-80 deg.C. The optimal separation condition of gel chromatography is 0.02mol/L Tris-HCl buffer solution with pH8.0, and the elution speed is 0.3 ml/min.
In this example, the temperature conditions used for freeze drying and the temperature conditions for storage were optimum conditions, but not exclusive conditions. The temperature control during freeze drying and low temperature storage is performed at-4 deg.C, which is helpful for maintaining antibacterial activity, but antibacterial activity gradually decreases with time.
Example two:
as shown in fig. 1, a wound plaster 100 comprises a substrate layer 20, wherein the lower surfaces of two ends of the substrate layer are provided with adhesive parts 21, the lower surface of the whole substrate layer 20 is provided with adhesive, the middle part of the substrate layer is adhered with a medicine layer 30, and the medicine layer 30 contains a composition taking a mixture of catfish body surface antibacterial peptide and active polysaccharide as effective components; and the release layer 10 is covered on the surfaces of the base material layer 20 and the medicine layer 30. Thus, the wound dressing 100 added with the catfish body surface antibacterial peptide and the active polysaccharide is prepared.

The solution was diluted with a small amount of tannic acid, and then the medicated cotton was immersed in the solution, soaked for 24 hours, and then dried in the shade. The medicated cotton is cut into pieces of 1.5cm by 1.5cm to obtain medicated layer 30.
Example three: wound repair experiment
Removing 1 small tuft of hair from the left leg and the right leg of the rabbit, exposing the skin, and slightly scratching a small opening with the depth of less than 2mm by using a sterilized small blade, wherein the small opening is 1 cm. On the left leg, erythromycin was applied to the open mouth and lightly wrapped with gauze. On the right leg, the drug layer 30 of example 2 was covered and lightly wrapped with gauze. Keeping the living environment of the rabbit clean, sanitary and dry. After 6 days, the wound is checked, the left leg wound is basically healed, scabs are scratched off, and a raised scar is formed; the wound of the right leg is healed, scabs are scratched off, and scars are not obvious.
Through verification, the natural active peptides and active polysaccharide substances from animals and plants have the effects of supplementing skin nutrition and resisting skin aging [ the research progress of the anti-skin aging effect of active peptides such as Qin Zhang Xin, Li Wen Zhi and Tang Jian, etc. ], the research progress of the anti-skin aging effect of active peptides in food science and technology, 2013,31(5):63-68 ]. Peptides are fragments of proteins, which are substances that must be synthesized in the repair of skin cells of animals. The pharmacological and pharmacodynamic studies of the national authoritative institution prove that: the active polysaccharide has the magical effects of quickly and effectively activating, enhancing and regulating the immunity of a human body, resisting tumors, resisting inflammation and viruses, regulating blood sugar and blood fat, repairing autoimmune diseases and the like. Polysaccharides are also important biopolymer compounds, and are present in the bodies of higher animals, plants, fungi, algae, and animals in the natural world. Thus, active polysaccharides are also essential nutrients in the synthesis and proliferation of animal skin cells. The antibacterial peptide and polysaccharide used in the invention are from natural animals, so that the antibacterial peptide and polysaccharide have better biocompatibility and biotransformation rate.
According to the invention, the antibacterial peptide and active polysaccharide mixture with the molecular weight of 2000-30000 is separated by skillfully filtering the waste water for feeding the catfish, and the two substances are mixed without re-separation and used for preparing a medicament for promoting wound healing or preparing a wound plaster, so that under the condition of preventing the wound from being infected by bacteria, necessary cell animal source nutrient substances are provided for repairing skin and subcutaneous tissues, the wound healing is promoted, bacterial tolerance side effects are reduced compared with antibiotics and the like, and the abuse of antibiotics is avoided.
The above detailed description of embodiments of the invention presented in the drawings is not intended to limit the scope of the invention as claimed, but is merely representative of selected embodiments of the invention. All other embodiments, which can be derived by a person skilled in the art from the embodiments given herein without making any creative effort, shall fall within the protection scope of the present invention.
