Abstract

本发明公开了一种洁净且具有优良光热效果的复合光热剂的制备方法,包括以下步骤:以盐酸多巴胺(DA)为反应单体,以水溶性醇和去离子水为混合溶剂,加适量浓氨水调整溶液pH,然后通过氧化自聚合充分反应制备出粒径均一的聚多巴胺纳米粒子;将得到的PDA纳米粒子分散于适量去离子水中得到分散液,然后再与二价铜盐溶液混合,发生络合反应,制备出PDA‑Cu2+络合物;将所得到的溶液为种子液,加入硫化物作为硫原,一定条件下充分反应,制备出在PDA上负载有CuS的复合微球,再通过透析滤去未反应的小分子,得到复合微球的分散液。本发明不仅可提高合成后材料的光热效果,而且合成方法绿色,产物洁净,同时本发明实验可重复性高,易于后续的推广与应用。

Description

一种具有光热效果的复合光热剂的制备方法
技术领域
本发明涉及材料科学与生物医用领域,尤其涉及一种具有光热效果的复合光热剂的制备方法。
背景技术
据统计,目前,世界上因疾病死亡的总死亡率中,癌症高居第二位。癌症的治疗引起了科学家们的广泛关注。传统的癌症治疗手段包括手术治疗、药物治疗、基因治疗等等。这些治疗手段往往都存在一些缺陷,比如不可避免的对正常组织的损伤、抗药性、治疗的不确定性等等。
光热治疗(PTT),作为一种非侵入性的新型治疗手段吸引了科学界极大的关注。该技术利用具有较高光热转换效率的材料,将其注射入生物体内,通过EPR效应使其在肿瘤区域富集,然后在近红外光(NIR)的照射下将光能转换成热能来杀死癌细胞。由于NIR具有较强的组织穿透力,因此该手段对正常组织的损害很小,具有较大的潜力应用于肿瘤治疗中。
聚多巴胺(PDA)是一种新兴的光热材料,它具有生物相容性良好、合成条件温和等优点,目前得到了广泛的研究,但它的光热吸收强度较弱,阻碍了它进一步的生物应用。基于此,本技术引入光热效应优良的硫化铜(CuS)进行联合产热,可以在提高复合微球光热效果的同时避免引入合成CuS所需的稳定剂,使得产物洁净且达到更好的肿瘤治疗效果。因此,通过简易的方法制备出光热效应良好且产物洁净的光热试剂具有极大的现实意义。
为此,我们提出了一种具有光热效果的复合光热剂的制备方法来解决上述问题。
发明内容
基于背景技术存在的技术问题,本发明提出了一种具有光热效果的复合光热剂的制备方法。
本发明提出的一种具有光热效果的复合光热剂的制备方法,包括以下步骤:
(1)以盐酸多巴胺为反应单体,以水溶性醇和去离子水为混合溶剂,加适量浓氨水调整溶液pH,然后通过氧化自聚合充分反应制备出粒径均一聚多巴胺纳米粒子;
(2)将步骤(1)中所得到的PDA纳米粒子分散于适量去离子水中得到分散液,然后再与二价铜盐溶液混合,发生络合反应,制备出PDA-Cu2+络合物;
(3)以步骤(2)所得到的溶液为种子液,加入硫化物作为硫源,一定条件下充分反应,制备出在PDA上负载有CuS的复合微球,再通过透析滤去未反应的小分子,得到复合微球的分散液。
优选地,所述步骤(1)中,所述的盐酸多巴胺水溶液中盐酸多巴胺的质量分数为3.7%-5.4%;所述水溶性醇为甲醇、乙醇和丙醇中任意一种;所述乙醇和去离子水体积之比为3.5-4.5:9;所述溶液pH值为8-9;所述反应条件为温和磁力搅拌下在水浴30℃下反应20-24h。
优选地,所述步骤(2)中,所述PDA分散液的浓度为0.5-1mg/ml;所述二价铜盐是氯化铜和硫酸铜中的一种;所述二价铜盐和PDA的质量比为1:2.4-3.7;所述络合反应条件为室温条件下反应3-5小时。
优选地,所述步骤(3)中,所述硫化物为硫化钠和硫代硫酸钠中的一种;所述硫化物中S2-与(2)中Cu2+的摩尔比为1:1;所述反应条件为88-90℃下,在氮气氛和冷凝水条件下反应45-60min;所述透析为采用截留分子量为14000D的透析袋在去离子水中透析三天,每6-8h换一次水。
本发明的有益效果如下:
(1)采用生物相容性良好的PDA和CuS作为联合光热试剂,其在人体血液循环系统中可以稳定存在,在保证复合微球具有良好生物相容性的同时,进一步提高材料的光热效果,是一种具有极大应用潜力的光热复合材料。
(2)整个复合微球的合成过程未加入任何表面活性剂,合成方法绿色,产物洁净。
(3)本发明中采用的原材料来源广泛,成本低廉,且制备方法简便,实验可重复性高,易于后续的推广与应用。

具体实施方式
下面结合具体实施例对本发明作进一步解说。
实施例1
(1)聚多巴胺纳米粒子的制备:量取18ml去离子水和8ml乙醇于100ml单口瓶中,在微弱的磁力搅拌条件下迅速加入0.6ml浓氨水(质量分数约为25%),30℃条件下搅拌30min,称取0.1g盐酸多巴胺溶解于2ml去离子水中,加入到溶液体系中。在30℃的条件下反应24h。
反应完毕后选用截留分子量为14000D的透析袋透析三天,得到聚多巴胺纳米微球的分散液。
(2)取(1)中分散液5ml与25ml去离子水加于100ml四口瓶中,磁力搅拌。称取0.0051gCuCl2.2H2O分散于1ml去离子水中,然后在磁力搅拌条件下逐滴加入四口瓶中,滴加完毕后在室温的条件下反应3h。反应完毕后,用去离子水离心洗涤,除去未络合的离子。
(3)称取0.0072gNa2S.9H2O分散于1ml去离子水中,然后逐滴加入到(2)中的反应液中,室温条件下磁力快速搅拌10min。然后在通入氮气和接入冷凝管的条件下水浴升温至90℃,恒温反应持续45min。反应结束后将反应液置于冷水浴中迅速降温,用去离子水洗涤数次,置于4℃冷藏储存。
图1为本实例中的纯PDA和后续复合微球的Uv-Vis吸收光谱图。从图中我们可以看到相较于纯的PDA微球,复合微球的在近红外区域处的吸光强度更大。
图2为配制复合微球浓度分别为200μg/ml、100μg/ml、50μg/ml的分散液,并以不含所述功能微球的去离子水为对照组,用波长为808nm,能量密度为2W/cm2激光照射各分散液和对照组,并测定不同时间段其温度的变化,测试结果如图2所示。可以看到该材料具有良好的光热效果,有应用于光热治疗的潜能。
实施例2
(1)聚多巴胺纳米粒子的制备:量取18ml去离子水和8ml甲醇于100ml单口瓶中,在微弱的磁力搅拌条件下迅速加入0.6ml浓氨水(质量分数约为25%),30℃条件下搅拌30min,称取0.1g盐酸多巴胺溶解于2ml去离子水中,加入到溶液体系中。在30℃的条件下反应24h。反应完毕后选用截留分子量为14000D的透析袋透析三天,得到聚多巴胺纳米微球的分散液。
(2)取(1)中分散液10ml与20ml去离子水加于100ml四口瓶中,磁力搅拌。称取0.0102gCuCl2.2H2O分散于1ml去离子水中,然后在磁力搅拌条件下逐滴加入四口瓶中,滴加完毕后在室温的条件下反应3h。反应完毕后,用去离子水离心洗涤,除去未络合的离子。
(3)称取0.0144gNa2S.9H2O分散于1ml去离子水中,然后逐滴加入到(2)中的反应液中,室温条件下磁力快速搅拌10min。然后在通入氮气和接入冷凝管的条件下水浴升温至90℃,恒温反应持续45min。反应结束后将反应液置于冷水浴中迅速降温,用去离子水洗涤数次,置于4℃冷藏储存。
实施例3
(1)聚多巴胺纳米粒子的制备:量取18ml去离子水和8ml丙醇于100ml单口瓶中,在微弱的磁力搅拌条件下迅速加入0.6ml浓氨水(质量分数约为25%),30℃条件下搅拌30min,称取0.1g盐酸多巴胺溶解于2ml去离子水中,加入到溶液体系中。在30℃的条件下反应24h。反应完毕后选用截留分子量为14000D的透析袋透析三天,得到聚多巴胺纳米微球的分散液。
(2)取(1)中分散液5ml与25ml去离子水加于100ml四口瓶中,磁力搅拌。称取0.0075gCuSO4.5H2O分散于1ml去离子水中,然后在磁力搅拌条件下逐滴加入四口瓶中,滴加完毕后在室温的条件下反应3h。反应完毕后,用去离子水离心洗涤,除去未络合的离子。
(3)称取0.0072gNa2S.9H2O分散于1ml去离子水中,然后逐滴加入到(2)中的反应液中,室温条件下磁力快速搅拌10min。然后再通入氮气和接入冷凝管的条件下水浴升温至90℃,恒温反应持续45min。反应结束后将反应液置于冷水浴中迅速降温,用去离子水洗涤数次,置于4℃冷藏储存。
实施例4

实施例5
(1)聚多巴胺纳米粒子的制备:量取18ml去离子水和8ml乙醇于100ml单口瓶中,在微弱的磁力搅拌条件下迅速加入0.5ml浓氨水(质量分数约为25%),30℃条件下搅拌30min,称取0.1g盐酸多巴胺溶解于2ml去离子水中,加入到溶液体系中。在30℃的条件下反应24h。反应完毕后选用截留分子量为14000D的透析袋透析三天,得到聚多巴胺纳米微球的分散液。
(2)取(1)中分散液5ml与25ml去离子水加于100ml四口瓶中,磁力搅拌。称取0.0051gCuCl2.2H2O分散于1ml去离子水中,然后在磁力搅拌条件下逐滴加入四口瓶中,滴加完毕后在室温的条件下反应3h。反应完毕后,用去离子水离心洗涤,除去未络合的离子。
称取0.0037gNa2S2O3.5H2O分散于1ml去离子水中,然后逐滴加入到(2)中的反应液中,室温条件下磁力快速搅拌10min。然后在通入氮气和接入冷凝管的条件下水浴升温至90℃,恒温反应持续45min。反应结束后将反应液置于冷水浴中迅速降温,用去离子水洗涤数次,置于4℃冷藏储存。
以上所述,仅为本发明较佳的具体实施方式,但本发明的保护范围并不局限于此,任何熟悉本技术领域的技术人员在本发明揭露的技术范围内,根据本发明的技术方案及其发明构思加以等同替换或改变,都应涵盖在本发明的保护范围之内。

Claims (4)

1.一种具有光热效果的复合光热剂的制备方法,其特征在于,包括以下步骤:
(1)以盐酸多巴胺为反应单体,以水溶性醇和去离子水为混合溶剂,加适量浓氨水调整溶液pH,然后通过氧化自聚合充分反应制备出粒径均一聚多巴胺纳米粒子;
(2)将步骤(1)中所得到的PDA纳米粒子分散于适量去离子水中得到分散液,然后再与二价铜盐溶液混合,发生络合反应,制备出PDA-Cu2+络合物;
(3)以步骤(2)所得到的溶液为种子液,加入硫化物作为硫源,一定条件下充分反应,制备出在PDA上负载有CuS的复合微球,再通过透析滤去未反应的小分子,得到复合微球的分散液。
2.根据权利要求1所述的一种具有光热效果的复合光热剂的制备方法,其特征在于,所述步骤(1)中,所述的盐酸多巴胺水溶液中盐酸多巴胺的质量分数为3.7%-5.4%;所述水溶性醇为甲醇、乙醇和丙醇中任意一种;所述乙醇和去离子水体积之比为3.5-4.5:9;所述溶液pH值为8-9;所述反应条件为温和磁力搅拌下在水浴30℃下反应20-24h。
3.根据权利要求1所述的一种具有光热效果的复合光热剂的制备方法,其特征在于,所述步骤(2)中,所述PDA分散液的浓度为0.5-1mg/ml;所述二价铜盐是氯化铜和硫酸铜中的一种;所述二价铜盐和PDA的质量比为1:2.4-3.7;所述络合反应条件为室温条件下反应3-5小时。
