Abstract
本发明公开了一种缓解仔猪断奶应激的替抗饲料添加剂及其制备方法。仔猪人工断奶后易出现断奶应激,营养消化不良导致腹泻,饲料转化率和免疫力降低。该替抗饲料添加剂的成分肉桂醛、高良姜素、山柰酚和药根碱均是从天然植物中提取的,不仅能替代断奶仔猪饲料的部分抗生素,而且该仔猪断奶替抗饲料添加剂中的肉桂醛、高良姜素、山柰酚和药根碱成分进入仔猪肠道内有很好的抑菌效果,能维持肠道微生态平衡,促进肠道营养物质消化吸收,调节仔猪机体免疫性能,提高仔猪生长发育水平,减少断奶仔猪腹泻,最终有效缓解断奶仔猪的断奶后应激,提高养猪业的经济效益。
Description
技术领域
本发明涉及一种缓解仔猪断奶应激的替抗饲料添加剂及其制备方法,具体涉及一种用于提高断奶仔猪生长性能和免疫性能、减少断奶仔猪腹泻以及缓解断奶应激的替抗饲料添加剂及其制备方法。
背景技术
现代集约化养猪中,出生21-28天后的断奶仔猪普遍采用人工断奶的方式,促使母猪更早更快地进入下一个繁殖期,减少仔猪感染母体疾病的几率,促进仔猪生长,提高养殖经济效益。人工断奶后仔猪的饲粮突然从母乳转变成固体颗粒饲料,仔猪被迫与母猪分开,断奶仔猪消化系统和免疫系统的发育还未完全适应饲粮和环境的变化,外界营养、心理、环境的变化使断奶仔猪对外部环境中的病原微生物更加敏感,此时断奶仔猪很容易发生断奶应激。断奶应激主要表现为断奶仔猪采食量下降、生长停滞、饲料转化率降低、免疫性能降低、腹泻、精神状况差,严重时出现僵猪,甚至导致断奶仔猪死亡,大大降低养猪的经济效益。
自20世纪抗生素就被广泛用作饲料添加剂,促进动物生长,对养殖业的发展做出了很大的贡献。抗生素作为断奶仔猪饲料添加剂既大大提高了仔猪的生产性能,更关键的是抗生素在断奶应激时预防和治疗细菌疾病的重要作用,大大地提高了养猪业的利润。然而,仔猪长期使用抗生素,会造成抗生素在仔猪体内残留,导致抗生素耐药细菌的产生,不利于疾病的防控。早在2006年,欧盟就禁止抗生素作为生长促进剂添加在动物饲料中。我国也开始逐步限制抗生素的使用,如2016年7月我国农业部发布禁止硫酸黏菌素用于动物促生长的公告。
随着人们对抗生素潜在危害认识的不断深入,越来越多的抗生素替代品被用于预防断奶仔猪疾病和促进仔猪生长,其中,植物提取物替抗饲料添加剂在断奶仔猪已经得到广泛应用。已有大量试验表明,植物提取物替抗饲料添加剂在替代饲用抗生素、提高断奶仔猪生长性能和免疫性能、缓解断奶应激方面,有突出的贡献,近年来关于替抗在断奶仔猪的应用研究也越来越深入。研究和利用替抗饲料添加剂,不仅能解决抗生素的残留和耐药性问题,而且能提高断奶仔猪生长性能和免疫性能,缓解断奶仔猪的断奶应激,提高养猪业的竞争力和产品质量,促进养猪业的可持续健康发展。
发明内容
本发明的目的是提供了一种缓解仔猪断奶应激的替抗饲料添加剂及其制备方法。
上述替抗饲料添加剂的作用是提高断奶仔猪生长性能和免疫性能、减少断奶仔猪腹泻以及缓解断奶应激。
本发明采取如下技术方案是实现上述替抗饲料添加剂的作用:
一种缓解仔猪断奶应激的替抗饲料添加剂,制备中包括以下成分:肉桂醛、高良姜素、山柰酚和药根碱,这几种成分都是天然植物提取物成分。
一种缓解仔猪断奶应激的替抗饲料添加剂,其特征在于,包括以下成分:肉桂醛、高良姜素、山柰酚和药根碱,各个成分的质量份是:肉桂醛5~10质量份,高良姜素5~10质量份,山柰酚7~15质量份,药根碱8~15质量份;还含有载体50~75质量份,所述载体为沸石粉、微晶纤维素和白炭黑的混合物,其质量比为(9~10):1:2。
肉桂醛从桂皮中提取,高良姜素从高良姜中提取,山柰酚从金缕梅中提取,药根碱从黄柏中提取。
各个成分的质量份是:肉桂醛5质量份,高良姜素5质量份,山柰酚7质量份,药根碱8质量份,载体75质量份。
其制备方法是:(1)稀释和吸附有效成分:先将肉桂醛、高良姜素、山柰酚和药根碱分别用载体进行搅拌混匀吸附;(2)混合混匀制得替抗饲料添加剂:将载体吸附过的肉桂醛、高良姜素、山柰酚和药根碱混合混匀搅拌制得所述替抗饲料添加剂。
替抗饲料添加剂应用于断奶仔猪饲料中,该替抗饲料添加剂在饲料的添加剂量为300~700g/t。
本发明中的替抗饲料添加剂应用到断奶仔猪饲料中,具体作用效果表现为:该替抗饲料添加剂中肉桂醛、高良姜素、山柰酚和药根碱皆从天然植物中提取,可以替代断奶仔猪饲料中的部分抗生素,减少抗生素的耐药性问题、残留问题和环境污染问题;该替抗饲料添加剂可以提高断奶仔猪生长性能和免疫性能,缓解断奶仔猪的断奶应激,促进断奶仔猪的健康养殖。
肉桂醛作为断奶仔猪抗应激添加剂的主要成分之一,有良好的抗菌杀菌效果。但是肉桂醛本身极易氧化和挥发,我们通过一些试验研究发现,将肉桂醛暴露在空气和光照下24小时,肉桂醛的损失率达到70%-80%,肉桂醛的不稳定性极大地影响了其断奶仔猪抗应激的效果。我们在本发明中通过添加高良姜素、山柰酚和药根碱来减少肉桂醛的氧化和挥发,提高肉桂醛的抑菌效果。高良姜素和山柰酚作为植物黄酮类化合物,具有抗氧化作用,在空气或饲料中与肉桂醛发生竞争性还原反应,减少肉桂醛的氧化挥发。添加肉桂醛与高良姜素、山柰酚和药根碱的混合物后,肉桂醛损失率减少了60%以上。
本发明通过试验表明,肉桂醛、高良姜素、山柰酚和药根碱具有协同增效作用,该替抗饲料添加剂在体外抑菌试验中能替代部分抗生素,有效抑制大肠杆菌和沙门氏菌等有害菌的生长,达到有效的最低抑菌浓度。肉桂醛、高良姜素、山柰酚和药根碱共同配合作用比肉桂醛单独作用的抗菌谱更广,抑菌效果更强。
肉桂醛作用于细菌的细胞膜和膜蛋白,破坏细胞膜的通透性,通过破坏细菌的细胞膜达到抑制和杀灭真菌的作用。肉桂醛穿透细胞膜进入细胞内,抑制细菌的DNA合成和细胞分裂,影响APT酶合成,从而抑制细菌生长和繁殖。高良姜素和山柰酚是植物黄酮类提取物,具有抗菌活性,进入断奶仔猪肠道,抑制大肠杆菌、沙门氏菌和金黄色葡糖球菌等有害微生物的繁殖,使仔猪肠道形成一个良好的微生态环境,提高肠道消化力和机体免疫力。高良姜素和山柰酚还能清除机体自由基,保护胃肠道黏膜层,提高仔猪机体抗氧化性能,调节仔猪机体免疫系统。药根碱具有抑菌活性,对多种细菌都有抑制作用,与肉桂醛、高良姜素、山柰酚配合还会对细菌的耐药性产生消除作用。另外,肉桂醛、高良姜素、山柰酚和药根碱的混合物添加到动物饲料中,由于具有抗氧化性,能减少饲料脂质氧化变质,延长饲料保质期。
将肉桂醛、高良姜素、山柰酚和药根碱的混合物添加到断奶仔猪饲料中,在仔猪肠道部位发挥作用,抑制肠道病原微生物的生长,维持肠道微生态稳定性,促进仔猪肠道的代谢吸收。单独添加肉桂醛到仔猪饲料中,肉桂醛只能在仔猪肠道前段发挥抑菌效果,本发明将高良姜素、山柰酚和药根碱与肉桂醛配合协同作用,替抗饲料添加剂能在仔猪肠道整段发挥抑菌效果。本发明通过试验表明,该替抗饲料添加剂能部分替代断奶仔猪中的抗生素,提高了断奶仔猪日采食量和日增重,降低饲料转化率,断奶仔猪腹泻降低36.2%-51.7%,提高体内代谢酶水平,提高机体补体和免疫球蛋白水平,总体表现为该替抗饲料添加剂提高生长性能和免疫性能,减少仔猪腹泻,从而抑制仔猪断奶后应激的发生。
具体实施方式:
本发明的实施方式不局限于以下实施例。
实施例1:体外抑菌试验。
(1)取肉桂醛5质量份,高良姜素5质量份,山柰酚7质量份,药根碱8质量份,载体75质量份,配制成替抗饲料添加剂产品。将试验分成3组,分组及抗生素或替抗饲料添加剂添加量见下表1,用20%的乙醇水溶液中配制成5120μg/ml母液。

(2)以该溶液为母液,以20%乙醇水溶液为稀释液分别得到以下不同浓度药液。稀释母液混合均匀,不要离心,取浑浊液进行稀释,得到稀释后浓度分别为512、256、128、64、32、16、4、2、1μg/ml。
(3)吸取6ml的各浓度的稀释液加入到冷却至50℃的有54ml的无菌MH琼脂液的100ml装锥形瓶中,充分摇晃混合均匀后按照20 ml/平皿的量倾注成不同浓度的药板。
(4)从已经分纯并过夜培养的细菌平板上挑取单菌落接种于0.85%生理盐水中培养18-20 h,再用生理盐水制成浊度为0.5麦氏标准的菌悬液(浓度约为×109cfu/ml)。准确吸取10μg上述菌悬液加入1 ml无菌生理盐水,使其稀释100倍,浓度达到约×107cfu/ml。分别将100μl稀释后的沙门氏菌菌悬液加入多点接种器贮液板上对应的孔槽之内,并用多点接种器将其接种至不同浓度和不同种类的抗生素平板上,每次接种1-2ul即浓度约为×104cfu/ml,37℃培养16-20 h。
(5)观察平板中细菌生长情况,得出各个试验组抗生素或替抗饲料添加剂的最低抑菌浓度。试验结果如下表2。

体外抑菌试验表明,试验组B组与对照组A组对比,试验中添加抗敌素比不添加抗敌素对大肠杆菌和沙门氏菌的最低抑菌浓度更低,表明抗敌素能有效抑制大肠杆菌和沙门氏菌。比较试验组B组和试验组C组,表明替抗饲料添加剂能替代抗敌素抑制大肠杆菌和沙门氏菌,达到有效的最低抑菌浓度。
实施例2:替抗饲料添加剂提高断奶仔猪生长性能和减少腹泻试验。
1、试验设计
试验选取120头杜长大三元杂交健康断奶仔猪(23±2日龄,初始体重7.07±0.34 kg),根据初始体重按照完全随机区组法,分为5组,每组6个重复,每个重复4头仔猪,公母各半,共用30个圈。其中A组为对照组,饲喂基础饲粮,B组在基础饲粮的基础上添加了抗敌素20g/t,C、D、E组分别在基础饲粮的基础上添加肉桂醛、高良姜素、山柰酚和药根碱的混合饲料添加剂300g/t、500g/t和700g/t,该混合替抗饲料添加剂的添加比例为肉桂醛5质量份,高良姜素5质量份,山柰酚7质量份,药根碱8质量份,载体75质量份。试验中每组各添加金霉素75g/t、喹乙醇100g/t和恩拉霉素10g/t,试验期为4周。

2、饲养管理
试验在某试验猪场进行,该场猪群健康、生产成绩稳定,饲养管理规范。每天饲喂仔猪4次,平均2小时巡圈观察仔猪采食状况,保证仔猪料槽内余料量不低于料槽容积的1/4,饲槽为矩形四口长槽,饮水器为乳头式。所有仔猪自由采食和饮水;试验过程中猪只发生死亡或因疾病淘汰时,称取个体重并结算饲料消耗量。试验期间保持猪舍通风、清洁,圈舍内温度控制在23~25℃,每天做圈舍清扫,试验期间消毒和免疫程序按照猪场规定进行。
试验第1、8、15、22、29日早晨仔猪空腹称重,并记录每周各圈加料量,每天结算各圈余料和损失料,以测定仔猪各阶段和全期生产性能,包括平均日增重(ADG)、平均日采食量(ADFI)和料肉比(FCR)。试验期间每天观察仔猪排粪情况并评分。
3、试验结果:本试验的试验结果如下表4。

由表4可以看出,与对照组相比,在断奶仔猪中添加300g/t、500g/t和700g/t替抗饲料添加剂均能提高断奶仔猪末重、采食量和日增重,降低饲粮转化率,明显地降低了断奶仔猪的腹泻,其中3组试验组腹泻评分分别减少了36.2%、48.3%和51.7%。与添加20g/t抗敌素组相比,在断奶仔猪中添加300g/t和700g/t替抗饲料添加剂能提高断奶仔猪末重、采食量和日增重,降低了断奶仔猪的腹泻率,说明断奶仔猪饲粮中替抗饲料添加剂能替代抗敌素的使用,提高了断奶仔猪生长性能和减少断奶仔猪腹泻。
实施例3:替抗饲料添加剂提高断奶仔猪免疫性能试验。
1、试验设计
选用175头杜长大三元杂交健康断奶仔猪(22±3日龄,初始体重7.89±0.23 kg),随机分为5组,每组7个重复,每个重复5头仔猪,公母各半。其中A组为空白对照组,饲喂基础饲粮,B组在基础饲粮的基础上添加了抗敌素20g/t,C、D、E组分别在基础饲粮的基础上添加肉桂醛、高良姜素、山柰酚和药根碱的混合饲料添加剂300g/t、500g/t和700g/t,该混合替抗饲料添加剂的添加比例为肉桂醛6质量份,高良姜素6质量份,山柰酚7质量份,药根碱8质量份,载体73质量份。试验中每组各添加金霉素75g/t、喹乙醇100g/t和恩拉霉素10g/t,试验期为4周。
2、饲养管理
试验在某试验猪场进行,该场猪群健康、生产成绩稳定,饲养管理规范。每天饲喂仔猪4次,平均2小时巡圈观察仔猪采食状况,保证仔猪料槽内余料量不低于料槽容积的1/4,饲槽为矩形四口长槽,饮水器为乳头式。所有仔猪自由采食和饮水;试验过程中猪只发生死亡或因疾病淘汰时,称取个体重并结算饲料消耗量。试验期间保持猪舍通风、清洁,圈舍内温度控制在23~25℃,每天做圈舍清扫,试验期间消毒和免疫程序按照猪场规定进行。
试验第29天早上8点,收集仔猪血清,检测血清补体C3、IgG(免疫球蛋白G)和IgM(免疫球蛋白M)、AST(天冬氨酸氨基转移酶)、ALT(丙氨酸氨基转移酶),使用全自动生化分析仪日立7020型仪器进行检测。
3、试验结果:本试验的试验结果如下表5。

由表5可以看出,与对照组相比,在断奶仔猪中添加300g/t、500g/t和700g/t替抗饲料添加剂均能提高断奶仔猪血清免疫球蛋白G、免疫球蛋白M、丙氨酸氨基转移酶水平,其中3组试验组腹泻评分分别减少了36.2%、48.3%和51.7%。在断奶仔猪中添加300g/t和500g/t替抗饲料添加剂均能提高断奶仔猪血清中血清补体C3和天冬氨酸氨基转移酶水平,说明替抗饲料添加剂能提高断奶仔猪免疫性能和代谢水平。与添加20g/t抗敌素组相比,在断奶仔猪中添加300g/t、500g/t和700g/t替抗饲料添加剂能提高断奶能提高断奶仔猪血清免疫球蛋白M,而血清补体C3、免疫球蛋白G、天冬氨酸氨基转移酶、丙氨酸氨基转移酶指标基本没有变化,说明断奶仔猪饲粮中替抗饲料添加剂能替代抗敌素的使用,维持断奶仔猪免疫水平和代谢水平。

1.一种缓解仔猪断奶应激的替抗饲料添加剂,其特征在于,包括以下成分:肉桂醛、高良姜素、山柰酚和药根碱,各个成分的质量份是:肉桂醛5~10质量份,高良姜素5~10质量份,山柰酚7~15质量份,药根碱8~15质量份;还含有载体50~75质量份,所述载体为沸石粉、微晶纤维素和白炭黑的混合物,其质量比为(9~10):1:2。
2.根据权利要求书1所述的替抗饲料添加剂,其特征在于:肉桂醛从桂皮中提取,高良姜素从高良姜中提取,山柰酚从金缕梅中提取,药根碱从黄柏中提取。
3.根据权利要求书1所述的替抗饲料添加剂,其特征在于:各个成分的质量份是:肉桂醛5质量份,高良姜素5质量份,山柰酚7质量份,药根碱8质量份,载体75质量份。
4.根据权利要求书1所述的替抗饲料添加剂,其应用于断奶仔猪饲料中,该替抗饲料添加剂在饲料的添加剂量为300~700g/t。
5.一种根据权利要求书1所述的替抗饲料添加剂的制备方法:(1)稀释和吸附有效成分:先将肉桂醛、高良姜素、山柰酚和药根碱分别用载体进行搅拌混匀吸附;(2)混合混匀制得替抗饲料添加剂:将载体吸附过的肉桂醛、高良姜素、山柰酚和药根碱混合混匀搅拌制得所述替抗饲料添加剂。
