Abstract

本发明公开了一种用于尿道或肛门的丁香油栓剂、一种用于尿道或肛门的丁香酚栓剂以及用于尿道或肛门的栓剂制备方法。本发明公开的用于尿道或肛门的丁香酚/丁香油栓剂,能够有效抑制泌尿系统、消化系统中常见的金黄色葡萄球菌、大肠杆菌、痢疾杆菌。并且由于其单独以丁香酚/丁香油作为药效成分,使得该栓剂在使用过程中的药效评价甚至不良反应也易于收集统计。

用于尿道或肛门的丁香酚/丁香油栓剂及其制备方法
技术领域
本发明涉及医用制品技术领域,特别涉及一种用于尿道或肛门的丁香酚/丁香油栓剂,以及该栓剂的制备方法。
背景技术
丁香是桃金娘科植物丁香树的干燥花蕾,其中丁香油为丁香花蕾蒸汽蒸馏出的挥发油,为无色或淡如黄色的液体,具丁香的特异香气。丁香油的药物活性成分主要是丁香酚,药用丁香油含85%-90%丁香酚。据相关文献报道,丁香油中所含丁香酚具有抗菌、抗炎以及麻醉镇痛等多方面作用。现有技术中,丁香油作为助剂已经在不少制剂和组合物中得到应用,如中国专利CN 102793759A(一种用于妊娠期阴道炎症的药物组合物)、CN104623644A(一种妇女清洁凝胶栓剂) 等,但是上述专利技术均是将丁香油作为香料而入选配方内的。
发明内容
本发明提供一种用于尿道或肛门的丁香酚/丁香油栓剂,其单独以丁香酚/丁香油作为药效成分,能有效对泌尿系统、消化系统中常见的金黄色葡萄球菌、大肠杆菌、痢疾杆菌起到抑制作用。

本发明还提供了上述用于尿道或肛门的栓剂制备方法,包括以下步骤:
a.按重量计称取处方量的原料;
b.将硬脂酸甘油酯于40~85℃水浴加热使融化成为油相;
c.将PEG4000、PEG6000与相应量甘油和适量吐温-80混合同温水浴,搅匀使成为水相,再加入处方量蒸馏水于水相中,混合均匀;
d.将水相与油相混匀,取丁香油/丁香酚以1~20滴/分钟的速度加入熔融基质中,边加边搅拌;
e.将拌匀的混合物注入已涂好液体石蜡的栓剂模具中,降至室温,取出,削去溢出部分,脱模得到栓剂。
其中,步骤c所述水浴温度为62~64℃;步骤d所述水相与油相的混合方法是将水相于等温下逐渐加入油相并混匀;步骤e所述脱模后,泌尿系统栓剂还需要置于-26℃环境下冷冻0.5h。
本发明公开的用于尿道或肛门的丁香酚/丁香油栓剂,能够有效抑制泌尿系统、消化系统中常见的金黄色葡萄球菌、大肠杆菌、痢疾杆菌。并且由于其单独以丁香酚/丁香油作为药效成分,使得该栓剂在使用过程中的药效评价甚至不良反应也易于收集统计。
具体实施方式
下面通过具体实施方式对本发明作进一步详细说明。但本领域技术人员将会理解,下列实施例仅用于说明本发明,而不应视为限定本发明的范围。实施例中未注明具体技术或条件者,按照本领域内的文献所描述的技术或条件(例如参考J.萨姆布鲁克等著,黄培堂等译的《分子克隆实验指南》,第三版,科学出版社)或者按照产品说明书进行。所用试剂或仪器未注明生产厂商者,均为可以通过市购获得的常规产品。
实施例1:处方及成型温度筛选
甘油在处方中作为保湿剂,其用量的多少对栓剂成型影响不大,而其他则为亲水性和亲脂性基质,其用量变化对栓剂成型影响大,故而将其它基质作为变量,甘油作为固定量,对其余组分进行单因素筛选。
将甘油的用量固定为1.0g,调整其他基质的用量,于60-80℃间选择3个温度点进行水浴,结果如下:

由表一结果可知,处方4在65℃时所制得的栓剂从外观成型性及融变时限两个指标来判断均为良好,可推测出一定范围内温度越高,会导致栓剂质越硬,因此以下选择62-64℃温度范围对温度进行筛选,结果如下:

综上,筛选出最佳处方比例为甘油:蒸馏水:PEG4000:PEG6000:吐温-80:单硬脂酸甘油酯:丁香油=1:10.5:11:11:1.5:15:2;最佳成型(水浴)温度为64℃。
实施例2:冷却时间筛选结果
随机选取栓剂12粒(已冷却至室温后脱模制得,若先冷冻再脱模会导致脱模困难),于-26℃冰箱分别冷冻0.5h、1h、2h、4h,各时间段测定其60min内融变时限结果分别为溶解、少量未溶、约1/2未溶、基本不溶。可知,于-26℃冰箱冷冻0.5h时栓剂融变情况良好,而其余温度段溶解不符合要求。
实施例3:丁香油/用于尿道或肛门的丁香酚栓剂的制备

上述用于尿道或肛门的栓剂制备方法,包括以下步骤:
a.按重量计称取如上任一项所述的原料;
b.将硬脂酸甘油酯于40~85℃水浴加热使融化成为油相;
c.将PEG4000、PEG6000与相应量甘油和适量吐温-80混合同温水浴,搅匀使成为水相,再加入处方量蒸馏水于水相中,混合均匀;
d.将水相与油相混匀,取丁香油/丁香酚以1~20滴/分钟的速度加入熔融基质中,边加边搅拌;
e.将拌匀的混合物注入已涂好液体石蜡的栓剂模具中,降至室温,取出,削去溢出部分,脱模得到栓剂。
实施例4:抑菌效果考察结果
将丁香油栓剂用小刀切下适量,并手动粗略加工为大小相近的圆饼状,分别置于已经制备好并分别涂有金黄色葡萄球菌、大肠杆菌、痢疾杆菌及乙型伤寒杆菌的琼脂培养皿中,培养48h后观察抑菌情况,经测量,其各自抑菌圈大小分别为:金黄色葡萄球菌(1.60cm、 2.00cm、1.80cm)、大肠杆菌(1.50cm、1.40cm、1.40cm)、痢疾杆菌 (1.1cm、1.0cm、1.0cm)。即对金黄色葡萄球菌的抑菌效果在四种细菌中为最好,大肠杆菌次之,对痢疾杆菌有较弱抑菌效果。
以上对本发明的较佳实施方式进行了具体说明,但本发明创造并不限于所述实施例,熟悉本领域的技术人员在不违背本发明精神的前提下还可作出种种的等同变型或替换,这些等同的变型或替换均包含在本申请权利要求所限定的范围内。

a.按重量计称取如权利要求1~4任一项所述的原料;
b.将硬脂酸甘油酯于40~85℃水浴加热使融化成为油相;
c.将PEG4000、PEG6000与相应量甘油和适量吐温-80混合同温水浴,搅匀使成为水相,再加入处方量蒸馏水于水相中,混合均匀;
d.将水相与油相混匀,取丁香油/丁香酚以1~20滴/分钟的速度滴加入熔融基质中,边加边搅拌;
e.将拌匀的混合物注入已涂好液体石蜡的栓剂模具中,降至室温,取出,削去溢出部分,脱模得到栓剂。
6.根据权利要求5所述的用于尿道或肛门的栓剂制备方法,其特征在于:步骤c所述水浴温度为62~64℃。
7.根据权利要求5所述用于尿道或肛门的栓剂制备方法,其特征在于,步骤d所述水相与油相的混合方法为:将水相于等温下逐渐加入油相并混匀。
8.根据权利要求5所述用于尿道或肛门的栓剂制备方法,其特征在于:步骤e所述脱模后,栓剂置于-26℃环境下冷冻0.5h。
