Abstract
本发明涉及药物制剂技术领域,尤其涉及一种治疗咽炎的老盐柠檬组合物及其制备方法。本发明老盐柠檬组合物以老盐、柠檬为主要成分,结合茶多酚、薄荷粉、荷叶碱、金银花、银杏叶、麦冬、白芍、黄芪、紫杉醇、葡萄糖酸钙、甘草、丁香、肉桂醛等成分,各组份之间实现协同增效的作用,提高本发明组合物的治疗咽炎的效果。本发明组合物具有价格低廉、疗效快、无毒副作用等优点。

技术领域
本发明涉及医药制剂技术领域,尤其涉及一种治疗咽炎的老盐柠檬组合物及其制备方法。
背景技术
咽炎是咽部粘膜,粘膜下组织的炎症,常为上呼吸道感染的一部分。引起咽炎的因素主要有病原微生物感染、物理或化学性刺激、邻近器官疾病、全身疾病等。细菌感染仍为咽炎的重要致病因素之一。致病菌主要为溶血性链球菌,其次为金黄色葡萄球菌、流感噬血杆菌、肺炎支原体等。依据病程的长短和病理改变性质的不同,可分为急性咽炎和慢性咽炎。急性咽炎常继发于急性鼻炎或急性扁桃体发炎之后或为上呼吸道感染之一部分。慢性咽炎主要为咽黏膜慢性炎症,弥漫性炎症常为上呼吸道慢性其他性炎症的一部分,局限性炎症则多伴有咽淋巴样组织的炎症。
治疗咽炎有多种方法,如茶疗、食疗、中药、西药等,其中茶疗、食疗、中药治疗等虽然副作用小,但普遍疗效较慢、效果不佳,而西药相对于中药具有较大的副作用,因此寻找一种副作用小、疗效快、治疗效果佳的咽炎治疗药物,一直以来都是本领域技术人员的一个重要研究方向。
发明内容
针对以上现有技术的不足之处,本发明提供一种治疗咽炎的老盐柠檬组合物,该组合物由多种成分组成,各组份之间能够发挥协同增效作用,对咽炎具有显著的治疗效果。
本发明采取的技术方案如下:
一种治疗咽炎的老盐柠檬组合物,包括以下组份:老盐、柠檬、茶多酚、荷叶碱、金银花、银杏叶、麦冬、白芍、黄芪、紫杉醇、葡萄糖酸钙、甘草、丁香、肉桂醛。
优选的,所述的一种治疗咽炎的老盐柠檬组合物,包括以下重量份数的组份:老盐9~13份、柠檬100~147份、茶多酚1~4份、荷叶碱10~20份、金银花3~5份、银杏叶25~30份、麦冬13~18份、白芍15~20份、黄芪20~25份、紫杉醇2~8份、葡萄糖酸钙10~12份、甘草10~12份、丁香1~10份、肉桂醛3~5份。
优选的,所述的一种治疗咽炎的老盐柠檬组合物,其特征在于,包括以下重量份数的组份:老盐10份、柠檬110份、茶多酚2份、荷叶碱10份、金银花4份、银杏叶26份、麦冬15份、白芍15份、黄芪21份、紫杉醇3份、葡萄糖酸钙10份、甘草10份、丁香3份、肉桂醛5份。
相应的,本发明还提供了所述的一种治疗咽炎的老盐柠檬组合物的制备步骤:
(1)取处方量的金银花、银杏叶、柠檬、麦冬、黄芪和丁香,粉碎,过筛,加入处方量肉桂醛,混匀,用60%~80%乙醇水溶液加热回流提取,重复4~6次,每次3h,得提取液,将提取液过大孔树脂柱,用蒸馏水洗脱,收集流出液,浓缩,干燥,得混合物A,备用;
(2)称取处方量的白芍、甘草、粉碎,过筛,混合,得混合粉末,用含有1~5%碳酸氢钠的55%~75%乙醇水溶液加热回流提取,重复4~5次,每次2h,过滤,得提取液和滤渣,将提取液上大孔树脂柱,用蒸馏水洗脱,合并洗脱液后加入盐酸溶液调节pH 1.0~4.5,浓缩,减压干燥,得粉末①;按质量比1:5~10,将滤渣与蒸馏水混合,于80~100℃条件下超声水浴3~5h,冷却,加入无水乙醇,过滤,浓缩,干燥,得粉末②;将粉末①与粉末②混匀,得混合物B;
(3)取处方量的老盐、茶多酚、葡萄糖酸钙、紫杉醇,按质量比1:5加入蒸馏水,于80~100℃恒温水浴锅中加热,在100~200r/m的转速下搅拌2~3h,冷却,加入步骤(1)的混合物A、步骤(2)的混合物B以及处方量的荷叶碱,搅拌均匀,即得本发明组合物。
优选的,所述的一种治疗咽炎的老盐柠檬组合物,在制备治疗咽炎的药物中应用。
本发明一种治疗咽炎的老盐柠檬组合物主要采用了以下组份:
老盐:一般指存放已满三十年的粗盐,又称战备盐。
柠檬:柠檬含有丰富的柠檬酸、柠檬香精油、类黄酮、维生素C、维生素A、钙、铁等微量元素,是一种具有高营养与药用价值的水果。具有抗氧化、抗癌、抗肿瘤、抗过敏、抗病毒、消炎、杀菌、降血压和降血脂等生理活性功能。
茶多酚:为淡黄至茶褐色略带茶香的粉状固体或结晶,具涩味。茶多酚具有较强的抗氧化作用和抑菌作用,对葡萄球菌、大肠杆菌、枯草杆菌等有抑制作用。
荷叶碱:有降血脂作用、抗自由基、抑制高胆固醇血症和动脉硬化等药疗、食疗功效,而且还具有抗有丝分裂的作用,有较强的抑菌效果。
银杏叶:具有扩张血管、调血脂、拮抗血小板活化因子、保护缺血损伤、抗炎及抗肿瘤等多种药理作用,其主要药用成分为黄酮类和萜内酯类化合物。
金银花:其功效主要是清热解毒,主治温病发热、热毒血痢、痈疽疔毒等。现代研究证明,金银花含有绿原酸、木犀草素苷等药理活性成分,对溶血性链球菌、金黄葡萄球菌等多种致病菌及上呼吸道感染致病病毒等有较强的抑制力,另外还可增强免疫力、抗早孕、护肝、抗肿瘤、消炎、解热、止血(凝血)、抑制肠道吸收胆固醇等,可与其它药物配伍用于治疗呼吸道感染、菌痢、急性泌尿系统感染、高血压等40余种病症。
麦冬:(学名:Ophiopogon japonicus(Linn.f.)Ker-Gawl.)百合科沿阶草属,多年生常绿草本植物,现代药理研究也表明,麦冬中含有丰富的生物活性成分,如皂苷类,高异黄酮类和多糖类成分,具有抗疲劳、清除自由基、提高细胞免疫功能以及降血糖的作用。另外,麦冬有镇静、催眠、抗心肌缺血、抗心律失常、抗肿瘤等作用。
白芍:(学名:Paeonia lactiflora Pall.)也称白花芍药,是毛茛科芍药属植物。白芍的化学成分主要为单萜及其苷类、三萜类、黄酮、鞣质、多糖、挥发油等,白芍对抗炎、免疫系统、中枢神经系统、心血管系统、消化系统等方面具有一定的药理作用。
黄芪:黄芪为常用中药,以多糖、三萜、黄酮等成分为主;黄芪具有降压、补气、消炎、强心、提高免疫力、消肿等功效。
紫杉醇:(英文名:Paclitaxel)别名泰素、紫素、特素,化学名称5β,分子量853.92,分子式C47H51NO14,是细胞有丝分裂抑制剂,被用于癌症治疗。
葡萄糖酸钙:葡萄糖酸钙,分子式为Ca(C6H11O7)2,该品为白色结晶性或颗粒性粉末,葡萄糖酸钙主要用作食品的钙强化剂与营养剂、缓冲剂、固化剂、鳌合剂。
甘草:(学名:Glycyrrhiza uralensis),又名乌拉尔甘草,是一种补益中草药。甘草及其活性成分能提高吞噬细胞的吞噬功能、调节淋巴细胞数量与功能、抑制IgE抗体形成、抗炎症介质及前炎性细胞因子,具有抗炎、抗变态反应的药理活性。甘草黄酮类化合物(异甘草素、异甘草苷、甘草素、甘草查耳酮A、光甘草定、甘草醇等)、甘草多糖和甘草酸是其抗炎、抗变应性炎症的活性成分。
丁香:富含黄酮类化合物,具有良好的治疗心血管疾病、抗肿瘤、抗氧化、抗病毒、抗菌、抗炎镇痛、降血脂等生物活性作用。
肉桂醛:(英文:Cinnamaldehyde)是一种醛类有机化合物,为黄色黏稠状液体,大量存在于肉桂等植物体内。
发明人研究发现,以上组份之间的组合使用能够起到协同增效作用。
与现有技术相比,本发明的有益效果是:
本发明组合物通过合理的配伍,保持各组分最大功效的基础上,使各组份发挥协同增效作用,具有清热解毒、抗菌消炎等作用,在治疗咽炎方面具有显著的治疗效果;本发明组合物的制备方法能够保存原料中的有效成分,生产工艺合理,通过本发明组合物的制备方法的工艺处理,各成分之间达到协同增效的作用,适合工业化生产。
具体实施方式
下面结合具体实施例来进一步描述本发明,本发明的优点和特点将会随着描述而更为清楚,但本发明的保护范围不限于以下实施例。
实施例1:
一种治疗咽炎的老盐柠檬组合物,包括以下重量份数的组份:老盐10份、柠檬110份、茶多酚2份、荷叶碱10份、金银花4份、银杏叶26份、麦冬15份、白芍15份、黄芪21份、紫杉醇3份、葡萄糖酸钙10份、甘草10份、丁香3份、肉桂醛5份。
一种治疗咽炎的老盐柠檬组合物,包括以下制备步骤:
(1)取处方量的金银花、银杏叶、柠檬、麦冬、黄芪和丁香,粉碎,过筛,加入处方量肉桂醛,混匀,用60%乙醇水溶液加热回流提取,重复6次,每次3h,得提取液,将提取液过大孔树脂柱,用蒸馏水洗脱,收集流出液,浓缩,干燥,得混合物A,备用;
(2)称取处方量的白芍、甘草、粉碎,过筛,混合,得混合粉末,用含有1%碳酸氢钠的55%乙醇水溶液加热回流提取,重复5次,每次2h,过滤,得提取液和滤渣,将提取液上大孔树脂柱,用蒸馏水洗脱,合并洗脱液后加入盐酸溶液调节pH 1.0,浓缩,减压干燥,得粉末①;按质量比1:5,将滤渣与蒸馏水混合,于100℃条件下超声水浴5h,冷却,加入无水乙醇,过滤,浓缩,干燥,得粉末②;将粉末①与粉末②混匀,得混合物B;
(3)取处方量的老盐、茶多酚、葡萄糖酸钙、紫杉醇,按质量比1:5加入蒸馏水,于100℃恒温水浴锅中加热,在200r/m的转速下搅拌3h,冷却,加入步骤(1)的混合物A、步骤(2)的混合物B以及处方量的荷叶碱,搅拌均匀,即得本发明组合物。
实施例2:
一种治疗咽炎的老盐柠檬组合物,包括以下重量份数的组份:老盐9份、柠檬100份、茶多酚1份、荷叶碱20份、金银花5份、银杏叶30份、麦冬13份、白芍15份、黄芪25份、紫杉醇8份、葡萄糖酸钙10份、甘草12份、丁香10份、肉桂醛5份。
一种治疗咽炎的老盐柠檬组合物,包括以下制备步骤:
(1)取处方量的金银花、银杏叶、柠檬、麦冬、黄芪和丁香,粉碎,过筛,加入处方量肉桂醛,混匀,用80%乙醇水溶液加热回流提取,重复4次,每次3h,得提取液,将提取液过大孔树脂柱,用蒸馏水洗脱,收集流出液,浓缩,干燥,得混合物A,备用;
(2)称取处方量的白芍、甘草、粉碎,过筛,混合,得混合粉末,用含有5%碳酸氢钠的75%乙醇水溶液加热回流提取,重复4次,每次2h,过滤,得提取液和滤渣,将提取液上大孔树脂柱,用蒸馏水洗脱,合并洗脱液后加入盐酸溶液调节pH 4.5,浓缩,减压干燥,得粉末①;按质量比1:10,将滤渣与蒸馏水混合,于80℃条件下超声水浴3h,冷却,加入无水乙醇,过滤,浓缩,干燥,得粉末②;将粉末①与粉末②混匀,得混合物B;
(3)取处方量的老盐、茶多酚、葡萄糖酸钙、紫杉醇,按质量比1:5加入蒸馏水,于80℃恒温水浴锅中加热,在100r/m的转速下搅拌2h,冷却,加入步骤(1)的混合物A、步骤(2)的混合物B以及处方量的荷叶碱,搅拌均匀,即得本发明组合物。
实施例3:
一种治疗咽炎的老盐柠檬组合物,包括以下重量份数的组份:老盐13份、柠檬147份、茶多酚4份、荷叶碱10份、金银花3份、银杏叶25份、麦冬18份、白芍20份、黄芪20份、紫杉醇2份、葡萄糖酸钙12份、甘草10份、丁香1份、肉桂醛3份。
一种治疗咽炎的老盐柠檬组合物,包括以下制备步骤:
(1)取处方量的金银花、银杏叶、柠檬、麦冬、黄芪和丁香,粉碎,过筛,加入处方量肉桂醛,混匀,用70%乙醇水溶液加热回流提取,重复5次,每次3h,得提取液,将提取液过大孔树脂柱,用蒸馏水洗脱,收集流出液,浓缩,干燥,得混合物A,备用;
(2)称取处方量的白芍、甘草、粉碎,过筛,混合,得混合粉末,用含有2%碳酸氢钠的60%乙醇水溶液加热回流提取,重复4次,每次2h,过滤,得提取液和滤渣,将提取液上大孔树脂柱,用蒸馏水洗脱,合并洗脱液后加入盐酸溶液调节pH 2.0,浓缩,减压干燥,得粉末①;按质量比1:8,将滤渣与蒸馏水混合,于90℃条件下超声水浴5h,冷却,加入无水乙醇,过滤,浓缩,干燥,得粉末②;将粉末①与粉末②混匀,得混合物B;
(3)取处方量的老盐、茶多酚、葡萄糖酸钙、紫杉醇,按质量比1:5加入蒸馏水,于90℃恒温水浴锅中加热,在120r/m的转速下搅拌3h,冷却,加入步骤(1)的混合物A、步骤(2)的混合物B以及处方量的荷叶碱,搅拌均匀,即得本发明组合物。
实施例4:
一种治疗咽炎的老盐柠檬组合物,包括以下重量份数的组份:老盐12份、柠檬120份、茶多酚2份、荷叶碱16份、金银花4份、银杏叶26份、麦冬15份、白芍16份、黄芪22份、紫杉醇4份、葡萄糖酸钙11份、甘草11份、丁香6份、肉桂醛4份。

(2)称取处方量的甘草、粉碎,过筛,混合,得混合粉末,用含有1%碳酸氢钠的55%乙醇水溶液加热回流提取,重复5次,每次2h,过滤,得提取液和滤渣,将提取液上大孔树脂柱,用蒸馏水洗脱,合并洗脱液后加入盐酸溶液调节pH 1.0,浓缩,减压干燥,得粉末①;按质量比1:5,将滤渣与蒸馏水混合,于100℃条件下超声水浴5h,冷却,加入无水乙醇,过滤,浓缩,干燥,得粉末②;将粉末①与粉末②混匀,得混合物B;
(3)将混合物A与混合物B混匀,即得。

(1)称取处方量的原料,混匀,得到物料a;
(2)在物料a中加入乙醇溶液浸泡7~10天,过滤得到滤液b和滤渣c;
(3)按质量比1:8,在滤渣中加入水煮提4次,每次0.5~1.5h,合并滤液,浓缩得到稠膏d;
(4)将滤液b浓缩得到稠膏e,将稠膏d和稠膏e混合均匀,干燥,粉碎,过筛,得到组合物。
对比例6:
一种治疗咽炎的老盐柠檬组合物,包括以下重量份数的组份:与实施例1相同。
一种治疗咽炎的老盐柠檬组合物,包括以下制备步骤:
(1)取处方量的金银花,粉碎,过筛,加入六分之一处方量肉桂醛,混匀,用60%%乙醇水溶液加热回流提取,重复6次,每次3h,得提取液,将提取液过大孔树脂柱,用蒸馏水洗脱,收集流出液,浓缩,干燥,得混合物A1,备用;
(2)取处方量的银杏叶、柠檬、麦冬、黄芪和丁香,分别按照步骤(1)的方法操作,得到混合物A2~混合物A6。
(3)称取处方量的白芍、粉碎,过筛,混合,得混合粉末,用含有1%碳酸氢钠的55%乙醇水溶液加热回流提取,重复4次,每次2h,过滤,得提取液和滤渣,将提取液上大孔树脂柱,用蒸馏水洗脱,合并洗脱液后加入盐酸溶液调节pH 1.0,浓缩,减压干燥,得粉末①;按质量比1:5,将滤渣与蒸馏水混合,于100℃条件下超声水浴5h,冷却,加入无水乙醇,过滤,浓缩,干燥,得粉末②;将粉末①与粉末②混匀,得混合物B1;
(4)称取处方量的甘草,按照步骤(3)的方法操作,得到混合物B2;
(5)取处方量的老盐、茶多酚、葡萄糖酸钙、紫杉醇,按质量比1:5加入蒸馏水,于100℃恒温水浴锅中加热,在200r/m的转速下搅拌3h,冷却,加入步骤(1)~步骤(2)的混合物A1~混合物A6、步骤(3)~步骤(4)的混合物B1~混合物B2以及处方量的荷叶碱,搅拌均匀,即得老盐柠檬组合物。
实验例1:对冰醋酸致小鼠腹腔毛细血管通透性的影响
实验方法:取小鼠110只,雌雄各半,体重25~30克,随机分为11组,每组10只,分别为对照组、实施例样品组1~实施例样品组4、对比例样品组1~对比例样品组6。①正常对照组(纯净水);②实施例样品组0.4g/kg;③对比例样品组0.4g/kg。按以上各组剂量灌胃给药,连续给药7天末次给药60min后,小鼠均经尾静脉注射0.5%的伊文思兰生理盐水溶液10mL/kg体重,随即腹腔注射0.6%的醋酸溶液0.2ml/只,20min后拉椎处死,剪开腹腔,用6ml生理盐水分数次洗涤腹腔吸出洗涤液。合并洗涤液后加入生理盐水至10ml,3000r/m离心15min取上清液于波长590nm测定光密度值。实验结果见表1。

实验结果表明,0.6%的醋酸溶液0.2ml/只小鼠腹腔注射可制备小鼠腹腔毛细血管通透性增加的模型,实施例样品组可明显的抑制醋酸所致的小鼠腹腔毛细学通透性增加,与对照组比较有显著性差异,表明本发明组合物对醋酸所致的小鼠腹腔毛细血管通透性增加有明显的抑制作用。
实验例2:对大鼠棉球肉芽肿形成的影响
取大鼠,体重190±10g,对大鼠腹腔麻醉,在下腹部做切口,从切口处将棉球植入腹股沟皮下,去除手术失败的大鼠,将大鼠随即分成11组,每组10只,分别为对照组、实施例1样品组~实施例4样品组、对比例1样品组~对比例6样品组。①对照组(纯净水);②实施例样品组0.4g/kg;③对比例样品组0.4g/kg。手术第二天开始给药,10mL/kg,每天一次,连续7天,第8天称重后处死,剥离棉球肉芽组织,干燥12h后称重,减去棉球重量,即为肉芽重量。实验结果见表2。

实验结果表明,实施例样品组可明显抑制大鼠棉球肉芽的形成,与对比例样品组相比,具有明显的抑制作用。
实验例3:体外抑菌实验
MIC测定:分别取实施例1至实施例4、对比例1至对比例6的样品以营养肉汤稀释(二倍稀释法)配置成各浓度的实验液,分别置于试管中(1.0ml/管),每管加入1.0ml的菌液(1.5×106CFU/ml),混匀,同时设计空白对照(只含肉汤)、阳性对照(不加实验液)、阴性对照(不加菌液),置于37±1℃的生化培养箱中培养24h,肉眼观察细菌生长情况。上层液澄清为无菌生长,上层液浑浊为有菌生长,以试管中无细菌生长的最小药物浓度为MIC。实验结果见表3。
MBC测定:在测定的MIC的基础上,将各管内容物用0.01ml的接种环接种于培养皿中,置于37±1℃的生化培养箱中培养24h,观察细菌生长情况,能将99.9%加入原始的细菌杀死的最低药物浓度为MBC。实验结果见表4。

实验结果表明:实施例组与对比例组相比具有明显的抑菌效果。实施例1至实施例4样品对金黄色葡萄球菌、肺炎链球菌、铜绿假单胞菌、α-溶血性链球菌、β-溶血性链球菌、白色念球菌、大肠杆菌具有很好的抑菌和杀菌效果,最低抑菌浓度(MIC)在0.313-0.125mg/ml之间,最低杀菌浓度(MBC)在0.0625-0.25mg/ml之间,说明本发明产品具有很好的抑菌效果。
其中,对比例6是采用实施例1的处方和制备方法,区别在于对比例6将各中药组份进行分开加工后再混合,结果导致对比例6样品与实施例样品相比抑菌效果非常差,由此说明,本发明组合物的制备方法能够使处方中各组份达到协同增效的结果,极大提高了本发明组合物治疗咽炎的效果。
以上所述仅为本发明的较佳实施例而已,并不用以限制本发明,凡在本发明的精神和原则之内,所做的任何修改、等同替换、改进等,均应包含在本发明保护的范围之内。
Claims (5)
1.一种治疗咽炎的老盐柠檬组合物,其特征在于,包括以下组份:老盐、柠檬、茶多酚、荷叶碱、金银花、银杏叶、麦冬、白芍、黄芪、紫杉醇、葡萄糖酸钙、甘草、丁香、肉桂醛。
2.根据权利要求1所述的一种治疗咽炎的老盐柠檬组合物,其特征在于,包括以下重量份数的组份:老盐9~13份、柠檬100~147份、茶多酚1~4份、荷叶碱10~20份、金银花3~5份、银杏叶25~30份、麦冬13~18份、白芍15~20份、黄芪20~25份、紫杉醇2~8份、葡萄糖酸钙10~12份、甘草10~12份、丁香1~10份、肉桂醛3~5份。
3.根据权利要求1所述的一种治疗咽炎的老盐柠檬组合物,其特征在于,包括以下重量份数的组份:老盐10份、柠檬110份、茶多酚2份、荷叶碱10份、金银花4份、银杏叶26份、麦冬15份、白芍15份、黄芪21份、紫杉醇3份、葡萄糖酸钙10份、甘草10份、丁香3份、肉桂醛5份。
4.根据权利要求1-3任一项所述的一种治疗咽炎的老盐柠檬组合物,其特征在于,其制备方法包括以下步骤:
(1)取处方量的金银花、银杏叶、柠檬、麦冬、黄芪和丁香,粉碎,过筛,加入处方量肉桂醛,混匀,用60%-80%乙醇水溶液加热回流提取,重复4~6次,每次3h,得提取液,将提取液过大孔树脂柱,用蒸馏水洗脱,收集流出液,浓缩,干燥,得混合物A,备用;
(2)称取处方量的白芍、甘草、粉碎,过筛,混合,得混合粉末,用含有1~5%碳酸氢钠的55%-75%乙醇水溶液加热回流提取,重复4~5次,每次2h,过滤,得提取液和滤渣,将提取液上大孔树脂柱,用蒸馏水洗脱,合并洗脱液后加入盐酸溶液调节pH 1.0~4.5,浓缩,减压干燥,得粉末①;按质量比1:5~10,将滤渣与蒸馏水混合,于80~100℃条件下超声水浴3~5h,冷却,加入无水乙醇,过滤,浓缩,干燥,得粉末②;将粉末①与粉末②混匀,得混合物B;
(3)取处方量的老盐、茶多酚、葡萄糖酸钙、紫杉醇,按质量比1:5加入蒸馏水,于80~100℃恒温水浴锅中加热,在100~200r/m的转速下搅拌2~3h,冷却,加入步骤(1)的混合物A、步骤(2)的混合物B以及处方量的荷叶碱,搅拌均匀,即得本发明组合物。
5.根据权利要求1-3任一项所述的一种治疗咽炎的老盐柠檬组合物,其特征在于,该组合物在制备治疗咽炎的药物中应用。
