Preferably, in the preparation method as described above, in step 5), the filtering of the glue juice specifically includes:
filtering the glue juice by a screen with 100-140 meshes, and then filtering by 3-5 layers of gauze;
more preferably, in step 5), the gum juice filtration specifically comprises:
filtering the glue juice by a 120-mesh screen and then filtering by 4 layers of gauze.
Preferably, in the preparation method as described above, in step 5), the specific parameters of oil-fat separation are as follows: passing the obtained glue juice through an oil-fat separator at the flow rate of 80-120L/h and the rotation speed of 18000-22000 r/min;
more preferably, in step 5), the specific parameters of the oil-fat separation are as follows: passing the obtained glue juice through an oil-fat separator at a flow rate of 100L/h and a rotation speed of 20000 r/min.
Preferably, in the preparation method, in the step 7), the low-temperature freezing is performed at-10 ℃ to-15 ℃ for 20h to 30 h;
more preferably, in the step 7), the low-temperature freezing condition is-12 ℃ to-13 ℃ for 25 h.
Preferably, in the preparation method as described above, in step 7), the conditions of the cooling glue are as follows:
cooling the glue in a glue cooling room at the temperature of 25-30 ℃ and the relative humidity of 40-60% for 6-8 days;
more preferably, in step 7), the conditions for cooling the gel are as follows:
the temperature is 27-28 ℃, and the relative humidity is 45-55 percent.

The examples, in which specific conditions are not specified, were conducted under conventional conditions or conditions recommended by the manufacturer. The reagents or instruments used are not indicated by the manufacturer, and are all conventional products available commercially.
Example 1
1. Taking 2000g of fresh deerskin, adding 3 times of water (the volume ratio of the added water is the volume ratio) and repeatedly cleaning for 2-3 times;
2. carrying out hair removal treatment on the deerskin obtained in the step 1, wherein the deerskin cannot be damaged in the process, and then removing residual meat and fat on the inner layer of the deerskin surface;
3. soaking the deerskin obtained in the step 2 in alkaline water (pH is 10-11) for 24 hours,
4. cleaning the deerskin obtained in the step 3 for 2-3 times, adding 2 times of water, and soaking for 24 hours until the deerskin is neutral;
5. cutting the deerskin obtained in the step 4 into small blocks of 15-20 cm;
6. putting the small deerskin blocks obtained in the step 5 into a steaming ball, adding 1.5 times of water, heating to 90-100 ℃ for scalding the deerskin, washing off part of grease, and repeatedly washing for 2-3 times with water;
7. adding 2 times of water into the small pieces of the clean deerskin obtained in the step 6, melting the deerskin for 6 hours under the pressure of 0.15MPa, discharging the glue juice after melting the deerskin, and repeating the melting for 4 times by the same method;
8. mixing the juices, filtering with 100 mesh screen, and filtering with 5 layers of gauze;
9. enabling the glue juice obtained in the step 8 to pass through a grease separator, wherein the flow rate is 80L/h, and the rotating speed is 18000 r/min;
10. heating the glue solution obtained in the step 9 to slightly boil, so that impurities in the glue solution can float on the water surface conveniently, beating out floating foam with foam, and repeating the beating for 10-15 times;
11. continuously concentrating the glue juice obtained in the step 10 until the water content reaches about 35%, adding 300g of fish oil, 8g of yellow wine, 5g of soybean oil and 10g of rock sugar, and uniformly mixing;
12. continuously concentrating the glue juice obtained in the step 11 until the water content reaches 26%, pouring the glue into a glue box, and solidifying for 24 hours at room temperature;
13. transferring the rubber blocks in the step 12 to-10 ℃ for freezing for 30 hours to form rubber lumps;
14. pouring out the glue lumps, cleaning four sides of the glue lumps, cutting the glue lumps into large strips of 4-5 cm, and cutting the large strips into small strips of 10 multiplied by 3 multiplied by 1 cm;
15. transferring the strips cut in the step 14 to a cold glue room with the temperature of 30 ℃ and the relative humidity of 60% for cooling glue for 8 days, and turning over the glue blocks 2 times every day;
16. putting the aired rubber blocks in the step 15 into a wooden box, sealing and sealing the wooden box, taking out the wooden box after 3 days, repeatedly cooling the rubber, and repeating the steps for three times until the rubber blocks are aired until the water content reaches 15%;
17. and (4) sterilizing and packaging the molded rubber block obtained in the step (16).
Example 2
1. Taking 2500g of fresh deerskin, adding 5 times of water (the volume ratio of the added water is the volume ratio) and repeatedly cleaning for 2-3 times;
2. carrying out hair removal treatment on the deerskin obtained in the step 1, wherein the deerskin cannot be damaged in the process, and then removing residual meat and fat on the inner layer of the deerskin surface;
3. soaking the deerskin obtained in the step 2 in alkaline water (pH is 10-11) for 48 hours,
4. cleaning the deerskin obtained in the step 3 for 2-3 times, adding 2 times of water, and soaking for 24 hours until the deerskin is neutral;
5. cutting the deerskin obtained in the step 4 into small blocks of 15-20 cm;
6. putting the small deerskin blocks obtained in the step 5 into a steaming ball, adding 2 times of water, heating to 90-100 ℃ for scalding the deerskin, washing off part of grease, and repeatedly washing for 2-3 times with water;
7. adding 5 times of water into the small pieces of the clean deerskin obtained in the step 6, melting the deerskin for 4 hours under the pressure of 0.2MPa, discharging the glue juice after melting the deerskin, and repeating the melting for 2 times by the same method;
8. mixing the juices, filtering with 140 mesh screen, and filtering with 3 layers of gauze;
9. enabling the glue juice obtained in the step 8 to pass through a grease separator, wherein the flow rate is 120L/h, and the rotating speed is 22000 r/min;
10. heating the glue solution obtained in the step 9 to slightly boil, so that impurities in the glue solution can float on the water surface conveniently, beating out floating foam with foam, and repeating the beating for 10-15 times;
11. continuously concentrating the glue juice obtained in the step 10 until the water content reaches about 45%, adding 100g of fish oil, 5g of yellow wine, 10g of soybean oil and 15g of rock sugar, and uniformly mixing;
12. continuously concentrating the glue juice obtained in the step 11 until the water content reaches 28%, pouring the glue into a glue box, and solidifying for 24 hours at room temperature;
13. transferring the rubber blocks in the step 12 to-15 ℃ for freezing for 20 hours to form rubber lumps;
14. pouring out the glue lumps, cleaning four sides of the glue lumps, cutting the glue lumps into large strips of 4-5 cm, and cutting the large strips into small strips of 10 multiplied by 3 multiplied by 1 cm;
15. transferring the small strips cut in the step 14 to a cold glue room with the temperature of 25 ℃ and the relative humidity of 40% for cooling glue for 6 days, and turning over the glue blocks 2 times every day;
16. putting the aired rubber blocks in the step 15 into a wooden box, sealing and sealing the wooden box, taking out the wooden box after 3 days, repeatedly cooling the rubber, and repeating the steps for three times until the rubber blocks are aired until the water content reaches 14%;
17. and (4) sterilizing and packaging the molded rubber block obtained in the step (16).
Example 3
1. Taking 2300g of fresh deerskin, adding 4 times of water (the volume ratio of the added water is the volume ratio) and repeatedly cleaning for 2-3 times;
2. carrying out hair removal treatment on the deerskin obtained in the step 1, wherein the deerskin cannot be damaged in the process, and then removing residual meat and fat on the inner layer of the deerskin surface;
3. soaking the deerskin obtained in the step 2 in alkaline water (pH is 10-11) for 36 hours,
4. cleaning the deerskin obtained in the step 3 for 2-3 times, adding 2 times of water, and soaking for 24 hours until the deerskin is neutral;
5. cutting the deerskin obtained in the step 4 into small blocks of 15-20 cm;
6. putting the small deerskin blocks obtained in the step 5 into a steaming ball, adding 1.8 times of water, heating to 90-100 ℃ for scalding the deerskin, washing off part of grease, and repeatedly washing for 2-3 times with water;
7. adding 3 times of water into the small pieces of the clean deerskin obtained in the step 6, melting the deerskin for 5 hours under the pressure of 0.17MPa, discharging the glue juice after melting the deerskin, and repeating the melting for 3 times by the same method;
8. mixing the juices, filtering with 120 mesh sieve, and filtering with 4 layers of gauze;
9. passing the glue juice obtained in the step 8 through an oil-fat separator at the flow rate of 100L/h and the rotation speed of 20000 r/min;
10. heating the glue solution obtained in the step 9 to slightly boil, so that impurities in the glue solution can float on the water surface conveniently, beating out floating foam with foam, and repeating the beating for 10-15 times;
11. continuously concentrating the glue juice obtained in the step 10 until the water content reaches about 40%, adding 200g of fish oil, 6g of yellow wine, 7g of soybean oil and 13g of rock sugar, and uniformly mixing;
12. continuously concentrating the glue juice obtained in the step 11 until the water content reaches 27%, pouring the glue into a glue box, and solidifying for 24 hours at room temperature;
13. transferring the rubber blocks in the step 12 to-13 ℃ for freezing for 25 hours to form rubber lumps;
14. pouring out the glue lumps, cleaning four sides of the glue lumps, cutting the glue lumps into large strips of 4-5 cm, and cutting the large strips into small strips of 10 multiplied by 3 multiplied by 1 cm;
15. transferring the small strips cut in the step 14 to a cold glue room with the temperature of 27 ℃ and the relative humidity of 50% for cooling glue for 7 days, and turning over the glue blocks 2 times every day;
16. putting the aired rubber blocks in the step 15 into a wooden box, sealing and sealing the wooden box, taking out the wooden box after 3 days, repeatedly cooling the rubber, and repeating the steps for three times until the rubber blocks are aired until the water content reaches 14.5%;
17. and (4) sterilizing and packaging the molded rubber block obtained in the step (16).
Experimental example 1 deer skin glue functional animal experimental verification
1.1 Experimental animals and groups
1.1.1 animal selection
Rats were used as experimental animals.
1.1.2 animal groups
64 male SPF-grade Wistar rats (purchased from Weitonglihua, Beijing) were used and weighed 270. + -.50 g. Rats were fed and drunk freely, and were randomly divided into 8 groups of 8 rats, i.e., a blank control group (C1), an example 1 group (L1), an example 2 group (L2), an example 3 group (L3), a positive control group (a), a fish oil-free control group (F1), a fish oil control group (F2), a castration control group (C2), and a castrated deer skin gelatin dose group (L0). After adaptive feeding for one week, castration treatment is carried out on the castration control group and the castration deer skin gelatin dosage group to make an animal model. The specific operation is as follows.
Wherein the deer glue prepared in the examples 1-3 is fed in the examples 1-3 respectively, and the preparation method of the fish oil-free control group is the same as that of the example 3, except that fish oil is not added; the fish oil control group was administered only with fish oil, and the deer skin gelatin dose group was fed with deer gelatin prepared in example 3 to castrated rats.
1.2 animal treatment
The castration groups were castrated to create castration animal models. First, a rat was anesthetized with 4% chloral hydrate (0.7ml/100g) by intraperitoneal injection, then, the rat was fixed on an operating table, the abdomen was cut with hair, after conventional sterilization, both testicles were excised through an abdominal incision, and the muscle and skin incisions were sutured in order. After surgery, each rat was given an intramuscular penicillin injection (18000U/(kg. d)) for 7 consecutive days to prevent infection.
1.3 treatment with drug
Seven days after operation, the medicine feeding treatment is carried out for ten days according to the medicine feeding mode shown in the following table.

1.4 hormone assay
After the treatment was completed, the rats were sacrificed by cervical dislocation. Blood is taken out through heart puncture and placed in a sterilized disposable centrifuge tube, after standing for 30min at room temperature, the centrifuge tube is centrifuged for 10min at 4000r/min in a low-temperature centrifuge, serum is separated, and the content of testosterone and luteinizing hormone is detected by using a full-automatic micro-particle chemiluminescence immunoassay analyzer (Beckman Coulter Access).
2 results of the experiment
2.1 hormone content
Compared with the group C1, the rats in the group C2 were castrated, and the serum testosterone and luteinizing hormone contents were remarkably reduced. After the deer skin glue is taken, compared with the C2 group, the contents of testosterone and luteinizing hormone in serum of the L0 group are obviously improved (p is less than 0.05). The contents of serum testosterone and luteinizing hormone in the L1, L2 and L3 groups are remarkably improved compared with the C1 group (p is less than 0.01). Compared with the group C1, the contents of serum testosterone and luteinizing hormone are also obviously improved in the groups F1 and F2 (p is less than 0.05).

Claims (9)
1. The deer skin glue is characterized by being prepared from the following raw materials in parts by weight:
2000-2500 parts of deerskin, 100-300 parts of fish oil, 5-8 parts of yellow wine, 5-10 parts of soybean oil and 10-15 parts of rock candy;
the preparation method of the deer skin glue comprises the following steps:
1) cleaning deer skin for depilation, removing residual meat and fat on the inner layer of the skin surface, and soaking in alkaline water;
2) soaking again in clear water and washing the deer skin to remove alkaline water;
3) cutting the deer skin obtained in the step 2), blanching the skin through steam, and cleaning again;
4) adding the deer skin obtained in the step 3) into water, pressurizing and melting the skin to discharge the glue juice;
5) filtering the glue juice obtained in the step 4), separating oil and fat, heating to slightly boil, and removing floating foam floating on the surface layer of the water surface;
6) concentrating the glue juice obtained in the step 5) until the water content reaches 35-45%, adding fish oil, yellow wine, soybean oil and rock candy, mixing uniformly, continuing to concentrate until the water content reaches 26-28%, discharging glue, pouring the glue into a glue box, and solidifying into glue blocks;
7) freezing the rubber block at low temperature to form rubber lumps, cutting the rubber lumps, and drying the rubber lumps;
8) sealing and sealing the aired rubber blocks for sealing, taking out after 2-4 days, and repeatedly cooling the rubber blocks according to the method in the step 7); repeating the operation until the water content of the rubber block reaches 14-15%; sterilizing, and packaging.
2. The deerskin glue according to claim 1, characterized by being prepared from the following raw materials in parts by weight:
2200-2300 parts of deerskin, 150-250 parts of fish oil, 6-7 parts of yellow wine, 7-8 parts of soybean oil and 10-15 parts of rock candy.
3. Deerskin glue according to claim 1 or 2, characterized in that said fish oil is shark oil.
4. The deer skin glue according to claim 1, wherein in step 1), the alkali water soaking is to be specifically soaking in alkali water with a pH of 10-11 for 24-48 hours.

The deer-hide glue of claim 1, wherein step 4) specifically comprises: adding the deerskin obtained in the step 3) into water with the volume 2-5 times of that of the deerskin, melting the deerskin for 4-6 hours under the pressure of 0.15-0.2 MPa, and repeating melting for 2-4 times by the same method.
6. The deer skin glue according to claim 1, wherein in step 5), the glue juice filtration specifically comprises:
filtering the glue juice by a screen with 100-140 meshes, and then filtering by 3-5 layers of gauze.
7. The deerskin glue according to claim 1, wherein in step 5), the specific parameters of oil separation are as follows: the obtained glue juice passes through an oil-fat separator, the flow rate is 80L/h-120L/h, and the rotating speed is 18000 r/min-22000 r/min.
8. The deer skin glue according to claim 1, wherein in the step 7), the low-temperature freezing condition is-10 ℃ to-15 ℃ for 20h to 30 h.
9. The deer-hide glue of claim 1, wherein in step 7), the glue cooling conditions are as follows:
and cooling the glue in the glue cooling room for 6-8 days at the temperature of 25-30 ℃ and the relative humidity of 40-60%.
