Abstract
本发明公开了一种具有抗氧化作用的鹰嘴豆组合物及应用,该组合物由鹰嘴豆、红豆、蓝莓粉和维生素C组成。该组合物具有提高体内超氧化物歧化酶(SOD)活力、降低丙二醛(MDA)、蛋白质羰基和谷胱甘肽(GSH)含量等作用。本发明提供的组合物可以用于制备抗氧化作用的食品、保健食品或药品。
Description
技术领域
本发明涉及一种鹰嘴豆组合物,尤其涉及一种鹰嘴豆组合物的抗氧化作用及其在食品、保健食品或药品中的应用。
背景技术
抗氧化性是众多生理功能的基础,体内的氧化平衡被破坏时会造成氧化应激,引起脂质、蛋白质等结构和功能损伤,导致人类的许多疾病如癌症、动脉粥样硬化和衰老的发生,氧化损伤是衰老过程的直接原因。当前常用的食品抗氧化剂大多是合成品,存在着安全性等问题,单一天然成分的抗氧化剂,其抗氧化能力有限,不能全面起到抗氧化的效果。因此天然的抗氧化组合产品,越来越受到人们的亲睐,而且随着人口高龄化的进展,人们对抗氧化功能的产品需求将不断增长。
鹰嘴豆( Chickpea)属豆科野豌豆族鹰嘴豆属(Cicer arietinum L ) ,一年生或越年生草本植物,鹰嘴豆在《维吾尔族药志》、《维吾尔民族医常用药材》、《药物宝库》、《草本遗》、《医典》等传统典籍中均有记载。中医认为,鹰嘴豆味甘、性平、无毒,具有补中益气、温肾壮阳功效,现代研究表明,鹰嘴豆异黄酮和总皂苷可以提高抗氧自由基酶类的活性发挥抗氧化作用。鹰嘴豆蛋白质水解后,可以清除1,1-二苯基苦基苯肼(DPPH)、能增强捕获羟自由基(·OH)的能力,能与金属离子的螯合而降低自由基的发生等。但未见关于鹰嘴豆清除氮氧自由基作用的报道。
目前公开的鹰嘴豆食品药品组合主要有:专利《一种包含鹰嘴豆、山药的食品及其制作方法》(申请号:201310730140.X)公开的鹰嘴豆、山药和VC以及调味品等制成果蔬泥产品,专利《一种具有抗疲劳功效的鹰嘴豆保健营养粉》(申请号:201310420729.X )公开了鹰嘴豆、山药、红景天、人参叶、龙眼药、酸枣仁、芡实、枸杞和薄荷等中药制成的具有抗疲劳功效的营养粉,专利《一种助消化鹰嘴豆奶茶粉及其制备方法》(申请号:201410291927.5)公开了由鹰嘴豆、大腹皮、淡竹叶、黄花蒿、马鞭草、苦瓜根、野艾蒿、葫芦茶、羊栖菜、富国菜、云雾果、枇杷汁、燕麦片和杏仁粉等制成的营养粉,专利《一种具有调节血脂作用的鹰嘴豆保健营养粉》(申请号:201310417872.3)鹰嘴豆和银耳、荷叶、水蛭、人参、陈皮、干姜、百合等制成具有其降血脂效果的营养粉,鹰嘴豆和茯苓、芡实、山药、党参、枸杞、龙眼肉、百合、酸枣仁、红枣等制成的有改善营养性贫血作用营养粉。上述已有的鹰嘴豆与中药组方,虽然均具有一定的保健功效,但其组方大,药味多,药物作用成分复杂,未见抗氧化作用的配方。因此选择药味少,功效成分清晰,能发挥多成分多靶点抗氧化优势,疗效确切的组方,具有较大的意义。
红豆营养丰富,深受大众喜爱,堪称粮食佳品,中医认为,红豆有健脾止泻、利水消肿、解毒排脓、清热去湿、补脾补血、生津益气等功效,被李时珍称为“心之谷”。红豆多糖在生物体内可直接参与猝灭自由基的途径,还能参与调动或激活机体的内源性抗氧化剂,使其数量增大和活性增强到机体需要的水平,从而避免或减轻自由基对机体的损伤。红豆多糖能阻断亚硝胺的合成,对亚硝酸钠也有一定的清除能力,能有效降低体内氮氧自由基的含量。
蓝莓营养成分丰富,除含常规的富含有机酸、糖、多种维生素不饱和脂肪酸和矿物质等成分外,尚含有超氧化物歧化酶( SOD),被联合国粮农组织列为人类五大健康食品之一。蓝莓所含的主要抗氧化成分花青苷口服后,可以分布到不同器官,并且可穿越血脑屏障而分布到脑,而发挥抗衰老作用。
维生素C是高等灵长类动物与其他少数生物的必需营养素,抗坏血酸在大多的生物体可借由新陈代谢制造出来,但是人类是最显著的例外,在生物体内,维生素C是一种抗氧化剂,保护身体免于自由基的威胁。实验研究表明,VC与鹰嘴豆抗氧化多肽的具有较好的协同作用,能显著增强鹰嘴豆抗氧化多肽的还原能力。
发明内容
本发明的首要目的在于提供一种鹰嘴豆组合物,该化合物具有显著的抗氧化作用。
本发明的另一目的在于提供上述鹰嘴豆组合物在制备抗氧化食品、保健品或药品中的应用。
本发明通过以下技术手段实现:
一种具有抗氧化作用的鹰嘴豆组合物。
该组合物由如下重量份的原料制成:鹰嘴豆800-950重量份,红豆粉80-95重量份,蓝莓粉15-20重量份,维生素C 1-3重量份。
优选地,所述组合物由如下重量份的原料制成:鹰嘴豆粉892重量份,红豆粉88 重量份,蓝莓粉18重量份,维生素C 2重量份。
鹰嘴豆异黄酮和总皂苷可以提高抗氧自由基相关酶类的活性起到抗氧化作用,鹰嘴豆蛋白质水解后的肽则可以清除1,1-二苯基苦基苯肼(DPPH)、能增强捕获羟自由基(·OH)的能力,能与金属离子的螯合而降低自由基的发生等。研究表明,VC与鹰嘴豆抗氧化多肽具有较好的协同作用,能显著增强鹰嘴豆抗氧化多肽的还原能力。红豆多糖在生物体内可直接参与猝灭自由基的途径,还能参与调动或激活机体的内源性抗氧化剂,从而避免或减轻自由基对机体的损伤。红豆多糖能阻断亚硝胺的合成,同时对亚硝酸钠也有一定的清除能力,能有效降低体内氮氧自由基的含量。蓝莓本身营养成分丰富,含有抗氧化能力极强的花青素,还含有超氧化物歧化酶( SOD),可而发挥较强的抗衰老作用。
因此,本组合物通过将鹰嘴豆粉、红豆粉、蓝莓粉和维生素C粉混合,充分发挥各组分的优势,分别从清除自由基、提高抗氧化酶活性、直接猝灭自由基、阻断氮氧自由基生成并清除氮氧自由基、直接补充SOD多途径协同发挥抗氧化作用。
本发明上述具有抗氧化作用的鹰嘴豆组合物的制备方法,可以采用如下方法:
(1)粉碎过筛:取鹰嘴豆和红豆,粉碎过80-300目筛,蓝莓粉和维生素C过80目筛,备用。
(2)混合:将鹰嘴豆粉、红豆粉、蓝莓粉与维生素C粉混合均匀,即得。
本发明提供上述组合物在制备用于抗氧化的食品、保健品或药品的应用。
优选地,所述食品、保健品为固体饮料、饼干或糕点,所述药品为胶囊或片剂。
优选地, 所述食品、 保健品或药品还包括食品、保健品或药品可接受的辅助药物或辅料。
本发明具有以下优点:
1)本发明以对抗氧化作用机理各异的鹰嘴豆、红豆、蓝莓与维生素C,从清除自由基、提高抗氧化酶活性、直接猝灭自由基、阻断氮氧自由基生成并清除氮氧自由基、直接补充SOD等多徐径协同发挥作用,起到提高体内超氧化物歧化酶(SOD)活力、降低丙二醛(MDA)、蛋白质羰基和谷胱甘肽(GSH)含量等作用,组份取材均为普通食品,组方简单,安全性高。
2)本发明可将该组合物制成固体饮料、饼干和糕点等食品,或者加入辅助药物和药学上可接受的载体,制成制剂,适用于需延缓衰老,口感适宜,方便携带和食用。
具体实施方式:
下面通过具体实施方式来进一步说明本发明,以下实施例为本发明具体的实施方式,但本发明的实施方式并不受下述实施例的限制,其他的任何未背离本发明的精神实质与原理下所作的改变、修饰、替代、组合、简化,均应为等效的置换方式,都包含在本发明的保护范围之内。
实施例1
一种具有抗氧化作用的鹰嘴豆组合物,该组合物由如下重量份的原料制成:鹰嘴豆892重量份,红豆粉88重量份,蓝莓粉18重量份,维生素C 2重量份;
其制备工艺包括如下步骤:
(1)粉碎过筛:取鹰嘴豆和红豆,粉碎过300目筛,蓝莓粉和维生素C过80目筛,备用。
(2)混合:将鹰嘴豆粉、红豆粉、蓝莓粉与维生素C粉混合均匀,即得。
实施例2 抗氧化作用的药理实验
采用实施例1的组合物为受试样品。
(1)试验动物及管理:雌性KM小鼠,23~26g,动物饲养在广州中医药大学实验动物中心,饲养温度与湿度:20~25 ℃,40~70 %,采用12 h:12 h昼夜间断照明;自由进食饮水,饮用水为实验动物中心制备的蒸馏水。
(2)分组与剂量设置:适应性饲养一周后,将小鼠按照体重随机分为空白组、模型组、组合物高、中、低剂量组,每组各14只。将0.8 g/kg定为实验组的高剂量组。另外分别设定一个中剂量组(0.4 g/kg)和一个低剂量组(0.2 g/kg)。
(3)数据分析:本实验所有数据均采用均值±标准差()表示,组间差异性分析采用SPSS 17.0软件进行单因素方差分析,以P<0.05作为统计学差异参考标准。
(4)实验方法:给药同时模型组和各给药组腹腔注射D-半乳糖150mg/kg;正常组皮下注射等体积的生理盐水;连续8周。造模的同时供试品组灌胃给药,空白组给予等容量的蒸馏水。实验结束后,取血清检测丙二醛、蛋白质羰基、超氧化物歧化酶、还原性谷胱甘肽等指标含量。
(5)检测结果:由表1可知,给药8周后,取血清检测丙二醛、蛋白质羰基、超氧化物歧化酶、还原性谷胱甘肽等指标含量。与空白组比较,模型组小鼠血清MDA、蛋白质羰基含量明显升高(P<0.05),SOD活力、GSH含量降低(P<0.05);与模型组相比,给药组小鼠MDA含量下降明显,各剂量组均具有统计学差异(P<0.01),SOD活力升高,其中鹰嘴豆中、高剂量组有统计学差异(P<0.05),GSH含量虽有升高的趋势。这表明鹰嘴豆具有一定的抗氧化作用。

(6)讨论:随着年龄的增长,机体抗氧化防御物质的合成和活性下降,清除自由基能力降低,产生和清除自由基的平衡被破坏,自由基堆积。丙二醛(MDA)为多不饱和脂肪酸被自由基攻击后通过链式反应最终断裂形成的脂质过氧化产物,其含量可反映体内自由基多少,间接地反映出自由基对机体的损伤程度。1987年Oliver等人报道了蛋白质羰基含量与衰老的关系,指出蛋白质羰基含量随年龄的增长而增加,以后的大量研究结果对这一结论给予肯定。目前蛋白质羰基的形成已经成为判定蛋白质氧化损伤的重要标志。还愿型谷胱甘肽(GSH)是机体内最重要的非酶性抗氧化物,可清处O2、H2O2、LOOH,因而GSH量的多少是衡量机体抗氧化能力大小的重要因素。
D-半乳糖致衰老模型是我国学者基于衰老的代谢学说而复制的衰老模型,在一定的时间内给动物连续注射D-半乳糖,使其细胞内半乳糖浓度增高,在醛糖还原酶的催化下,还原成半乳糖醇,后者不能被细胞进一步代谢而堆积在细胞内,影响正常的渗透压,导致机体衰老。本研究显示造模后动物SOD活力明显下降,MDA、蛋白质羰基含量升高,提示对动物造成氧化损伤;给药8周后,发现通过给予鹰嘴豆,动物氧化损伤指标SOD活力、MDA含量等均有改善,表明鹰嘴豆具有一定的抗氧化作用。
(7)结论:实验结果表明D-半乳糖所致过氧化损伤小鼠连续给予鹰嘴豆后,体内SOD活力上升、MDA含量下降,提示鹰嘴豆具有一定的抗氧化作用。
实施例3 抗氧化组合物饼干制备
一种具有抗氧化作用的鹰嘴豆组合物饼干,该组合物由如下重量份的原料制成:鹰嘴豆900重量份,红豆粉80重量份、蓝莓粉15重量份与维生素C粉1重量份和其它饼干用原料;
其制备工艺包括如下步骤:
(1)粉碎过筛:取鹰嘴豆和红豆,粉碎过120目筛,蓝莓粉和维生素C过80目筛,备用。
(2)混合:将鹰嘴豆粉、红豆粉、蓝莓粉与维生素C粉混合均匀,即得。
(3)制饼:取鹰嘴抗氧化组合物混合粉豆300g、玉米须粉70g、面粉153.8g、玉米淀粉23.5g、鸡蛋40g、麦芽糖醇200g、食用油200g、碳酸氢铵6.72g、泡打粉3.36g、小苏打1.34g、食盐0.84g、钙粉0.34g、甜蜜素0.10g,混合均匀,加水制成团,压成圆形生饼,以面温140℃,底温180℃进行烘烤,时间8min,得熟饼,包装即得抗氧化组合物饼干。
实施例4 抗氧化组合物胶囊制备
配方:鹰嘴豆800重量份,红豆95重量份、蓝莓20重量份、维生素C 3重量份,硬脂酸镁 2重量份。
其制备工艺包括如下步骤:
本实施例将具有抗氧化作用的鹰嘴豆组合物制备成胶囊剂,具体的配方及其制备方法如下:
(1)粉碎过筛:取鹰嘴豆和红豆,粉碎过80目筛,蓝莓粉和维生素C过80目筛,备用。
(2)混合:将鹰嘴豆粉、红豆粉、蓝莓粉与维生素C粉混合均匀,即得。
(3)取上述混合物,加入硬脂酸镁混匀, 填充胶囊,即得。
Claims (6)
1.一种具有抗氧化作用的鹰嘴豆组合物,其特征在于,该组合物由如下重量份的原料制成:鹰嘴豆800-950重量份,红豆粉80-95重量份,蓝莓粉15-20重量份,维生素C 1-3重量份 。
2.根据权利要求1所述的具有抗氧化作用的鹰嘴豆组合物,其特征在于,所述组合物由如下重量份的原料制成:鹰嘴豆892重量份,红豆粉88重量份,蓝莓粉18重量份,维生素C 2重量份。
3.根据权利要求1或2所述的具有抗氧化作用的鹰嘴豆组合物的制备方法,其特征在于,包括如下方法:
(1)粉碎过筛:取鹰嘴豆和红豆,粉碎过80-300目筛,蓝莓粉和维生素C过80目筛,备用;
(2)混合:将鹰嘴豆粉、红豆粉、蓝莓粉与维生素C粉混合均匀,即得。
4.根据权利要求1或2所述的具有抗氧化作用的鹰嘴豆组合物在制备具有抗氧化的食品、保健品或药品中的应用。
5.根据权利要求4 所述的具有抗氧化作用的鹰嘴豆组合物在制备抗氧化的食品、保健品或药品中的应用,其特征在于,所述食品、保健品为固体饮料、饼干或糕点,所述药品为胶囊或片剂。
6.根据权利要求5所述的具有抗氧化作用的鹰嘴豆组合物在制备抗氧化的食品、保健品或药品中的应用,其特征在于,所述食品、保健品或药品还包括食品、保健品或药品可接受的其它辅助药物或辅料。
