发明领域

发明背景
银和银盐用作抗微生物剂已经很多年。银的早期医学用途是应用硝酸银水溶液来预防新生婴儿的眼睛感染。银盐、胶体和复合物也已用于预防和控制感染。例如,胶体金属银已经局部用于皮炎、结膜炎和包括阴道感染的感染。
还发现其它金属比如金、锌、铜和铈无论单独还是与银组合也都具有抗微生物性质。这些和其它金属已经显示出提供抗微生物性质,即使在极其低浓度下,一种称为“微动(oligodynamic)”的性质。金属离子,特别是那些重金属,都显示出该作用。准确的作用机制仍然是未知的。来自银的数据表明这些离子通过结合反应基团使靶细胞或生物体的酶变性,导致其沉淀和失活。最新证据是改变细胞DNA结构本身。银也与氨基-、羧基-、磷酸酯(根)-和咪唑-基团反应,减弱了乳酸脱氢酶和谷胱甘肽过氧化物酶的活性。细菌通常受微动作用的影响,似乎该作用对于细菌菌株是不可知的。病毒通常对该作用不那么敏感。
银是一种天然存在的元素,其存在于我们的环境中,包括我们呼吸的空气、我们饮用的水和我们消耗的食物。然而,银并非天然存在于人和动物的组织中。银对于动物和人类是相对低毒性的。然而,其对于更简单的生命形式比如细菌毒性极大。银的抗菌性质是已知的,且至少怀疑了数千年。古代希腊人使用银锅和其它器具。近代医学之父希波克拉底,记录了银具有有益的愈合和抗-病性质。腓尼基人将水、葡萄酒和醋贮存在银瓶中以防止变质。在20世纪初期,人将银圆放置在奶瓶中以延长奶的新鲜度并不少见。银的可锻性和无毒性使其成为用于镶牙和补牙的牙用合金的一种有用材料。
银的广泛应用随着现代抗生素的发展而减少,许多抗生素用于杀死病原体,但是过度使用会导致细菌抗药性增加。因此,对于银作为广谱抗微生物剂再次产生了兴趣。当局部施用时,银表现出对抗有时显示出抗性特性的微生物的功效。存在许多处理或杀死细菌的市售产品。这些产品以多种形式存在,包括液体、泡沫、凝胶、洗剂和软膏剂。
另外,已知银用于牙科和医疗装置,比如管口、牙托(mouthguards)、牙用器械以及导管、套管和支架的抗微生物用途。另外,银化合物已经被用在用于消费的流体中,且被并入衣物和包装中以预防感染、细菌繁殖和变质。
每年,由于细菌引起的医院获得性感染使约超过100,000人死亡。该数字大于AIDS、乳腺癌和车祸总计导致的总死亡人数。据估计每年该经济负担大于52亿美元。这些感染是第四大主要死亡原因。手部清洁不足也导致与食品相关的疾病,包括沙门氏杆菌和大肠杆菌感染。根据疾病控制和预防中心(The Center for Disease Control andPrevention)(此后,“CDC”),每年多达7600万美国人感染食物传染疾病。其中,几乎5,000人由于这些疾病而死亡。其他人感受到恶心、呕吐和腹泻的令人烦恼的症状。
已公布的CDC指南增加了洗手液在美国的销量,根据A.C.Nielsen提供的销售信息,其在2004-2010期间经历了两位数的增长。估计,每年总的美国感染预防工业达94亿美元。
对于许多装置,优选地在装置上具有润滑涂层。润滑涂层有助于装置插入、减少对组织的损伤和减少细菌的粘附。将银和其他金属直接应用在也期望具有润滑涂层的装置表面上的常规方法的另一个缺点是,在装置的抗微生物涂层上必须应用第二次润滑涂层,增加了生产成本和时间。
这些涂层的一些将银离子以不同程度释放到基质周围的溶液或组织中。
微动金属可以以多种方式物理结合到凝胶中。例如,银盐的液体溶液可以浸渍、喷雾或刷涂在哺乳动物皮肤上和皮肤中。
微动试剂的颗粒沉降是由于颗粒的尺寸和密度引起的。从这种溶液的颗粒沉降会导致组合物中微动试剂浓度的不可预见的改变。对于生产市售产品,离子浓度的这些变化产生一些缺点。首先,微动试剂浓度的不可预见的变化使其难以生产具有特定、均质、精确浓度的抗微生物离子的组合物,且从而难以产生特定和特异性的效力。另外,这些变化使其难以生产多批具有相同抗菌浓度的组合物。而且,抗微生物离子浓度会影响组合物的其它性质,比如其粘合性质和润滑性质。抗微生物活性的一致性在生产抗微生物装置方面很重要。
与颗粒悬浮液有关的另一个问题是颗粒团聚。颗粒团聚产生更大粒径,增加了从溶液的颗粒沉降。
许多研究人员尝试克服这些问题。
Yamamoto的名称为“Method of Forming Chelated Collagen HavingBactericidal Properties”的USP 4,847,049描述了一种用于保护复原的胶原蛋白抗细菌和霉菌侵袭的方法。该方法包括使胶原蛋白接触pH范围4.0至9.0的含有银离子的溶液,并将银螯合的胶原蛋白暴露于紫外线照射。

因此,已经确定需要提供一种使凝胶抵抗感染的方法,通过减少或消除主要在皮肤表面上或对皮肤中伤口或疮的不期望的细菌生长。重要的是金属氧化物复合物是均质分散的,在凝胶的整个使用寿命期间是耐受的,并且该复合物保持稳定,以使其可以在贮存期限期间显示延长的活性。本领域还需要可以并入凝胶中以提供抗微生物活性的金属氧化物组合物。还需要克服常规微动组合物的溶解性、沉降和团聚问题,并且当这些试剂(对于本发明主要是氧化银,但是铜及其它金属氧化物也是有用的)分散在凝胶、液体和某些固体内时显示出增加的、缓释释放的微动试剂的组合物。以成本有效和可再现的方式将金属氧化物引入对于哺乳动物皮肤应用并且甚至摄入可接受的产品中是用于提供容易获得的制备物品所需要的,并且是本发明的一个主题。
发明简述

另一个实施方案包括凝胶组合物与下述物质混合和/或注入到其中或以其它方式直接用作下述物质:调理剂、发用摩丝、喷发胶、染发剂、隐形眼镜溶液、除臭剂、个人和工业润滑剂、食品级可食用物质、冰形式的水、皮肤洗剂、局部制剂、牙膏、口服凝胶剂、和食用油脂的替代物。凝胶组合物的凝胶是用任何阳离子体系形成的,所述阳离子体系可以为阳离子聚合物体系。而且,阳离子聚合物体系可以选自藻酸盐、壳聚糖、葡聚糖、角叉菜胶、果胶和黄原胶。
凝胶必须是与基于多元醇/甘油的螯合的金属氧化物悬浮液可溶混的和/或相容的,所以凝胶与所述悬浮液混合得到获得抗菌和抗微生物且在一些情况下抗病毒性质的最终化合物。在制备这些化合物或最终的制品中,可以包括加入其它添加剂比如着色剂(主要是偶氮化合物)、无机元素、无机和有机化合物(包括粘土、颜料),同样地所有的填充剂不干扰或显著地改变最后凝胶的最终抗菌或物理化学性质。

在仍然另一个实施方案中,抗菌、抗微生物和/或抗病毒制品包括凝胶组合物和均匀悬浮在多元醇中的螯合的金属氧化物颗粒,所述多元醇与聚合组合物的聚合物混合且注入其中,所述凝胶组合物包括注入多元醇内浓度为至少0.001重量%的金属氧化物的多元醇金属氧化物复合物,使得多元醇金属氧化物复合物的浓度不超过聚合组合物的25重量%。

螯合的氧化银溶液组合物
在第一个方面,本发明提供抗微生物、抗菌和在某些情况下抗-病毒金属氧化物复合物,其主要在基于水和/或多元醇的溶液中形成稳定均质的螯合的金属氧化物复合物凝胶,没有显示出沉淀。
在第二个方面,将复合微动试剂悬浮液直接注入、混合、掺合凝胶或与其复合。如本发明中使用的术语“微动试剂”在该情况下指任何螯合的金属氧化物化合物,其提供相关的抗微生物(或其它期望的)特征或活性,即使当以相对少量和低浓度存在时。
一个优选的实施方案是悬浮在多元醇中螯合的氧化银。在水或多元醇(例如PEG–聚乙二醇或甘油)中或多元醇和水组合中,将均质螯合的金属(在一个实例中是银)氧化物悬浮液浓缩至4000ppm。较高初始浓度的例如PEG是可能的,但是主要焦点是提供不允许形成沉淀的螯合的金属氧化物(特别是氧化银)溶液。该高浓度的螯合的金属氧化物溶液是一种复合悬浮液,接着将其稀释,结果其被并入基本上任何凝胶或其它可接受的(与多元醇载体可溶混的)液体。该并入赋予进入基质底物中的螯合的金属氧化物期望的性质,而没有任何预期或已知不期望的物理化学性质变化。
本发明中可以应用任何凝胶,包括亲水性凝胶、疏水性凝胶和这两种类型的混合物。使用亲水性凝胶可能是优选的,因为这样的凝胶可以具有另外的益处。这些益处包括为了患者舒适的增加的润滑性,增加从身体吸收水性流体,其有助于从组合物释放微动离子,抑制细菌附着,和对于某些金属氧化物螯合物改善的溶解性。最适合于本发明的亲水性凝胶是溶于水或含水有机溶剂的那些。向凝胶中加入水而没有金属氧化物沉淀的能力是本发明的附加益处,而不是必要条件。水促进盐胶体的形成,其可以在加入凝胶之前或之后沉淀,得到最终组合物。为此,优选地以重量计凝胶基本上不含水,一定不超过5至30%的水。
然而,使用水不受限制,因为金属氧化物螯合物也可以形成或悬浮在乙醇、有机溶剂或含有很少或没有水的上述两者中。当本发明中使用疏水性凝胶时,使用含有0至1%水的乙醇和有机溶剂是优选的。螯合的金属氧化物的活性随它们应用在其中的载体变化。基于复合物悬浮液内初始浓度的金属氧化物的性质,沉淀也将变化。
亲水性食品级可食用凝胶的实例包括使用甘油和乳化剂制备的那些,比如Grune等人的美国专利号8557264B2中描述的,将其内容通过援引全部并入本文。矿脂基凝胶一般是疏水性的。在本发明中,混合疏水性和亲水性凝胶是可能的。最终凝胶组合物可以包括不会干扰螯合的银(和其它金属)氧化物的抗菌和抗微生物性质的基本上任何无机或有机填料,并且可以包括某些或所有下述有机聚合物;聚氨酯,包括聚醚聚氨酯、聚酯聚氨酯、聚氨酯脲、及其共聚物;聚乙烯吡咯烷酮;聚乙烯醇;聚乙二醇及其共聚物;聚丙二醇及其共聚物;聚氧乙烯及其共聚物;聚丙烯酸;聚丙烯酰胺;羧甲基纤维素;纤维素及其衍生物;葡聚糖及其它多糖;淀粉;瓜尔胶;黄原胶及其它树胶和增稠剂;胶原蛋白;明胶;及其它生物聚合物。亲水性凝胶中使用的优选的亲水性聚合物包括聚氨酯和聚氨酯共聚物,比如聚醚聚氨酯脲。
最适合用于疏水性凝胶(通常在矿脂基质中提供)的疏水性聚合物的实例包括,但不限于:聚四氟乙烯、聚氯乙烯(PVC)、聚乙酸乙烯酯、聚(对苯二甲酸乙二酯)、聚硅酮、聚酯、聚酰胺、聚脲、苯乙烯-嵌段共聚物、聚甲基丙烯酸甲酯、丙烯酸-丁二烯-苯乙烯共聚物、聚乙烯、聚苯乙烯、聚丙烯、天然和合成橡胶、丁腈橡胶、及任何上述的混合物和共聚物。
本发明的螯合物包含一种或多种微动金属氧化物。在如下方法的讨论中,金属氧化物为氧化银,但是铜、锡、铝、锌和钛都形成可以螯合的金属氧化物。用于本发明的微动金属氧化物包括,但不限于银、铂、金、锌、铜、铈、镓、锇等。对于本发明的目的而言,优选的微动金属为银。
可以使用其它金属的螯合作用形成螯合物。盐可以充当部分螯合剂,且含有阳离子,包括但不限于钙、钠、锂、铝、镁、钾、锰等,并且也可以包括微动金属阳离子比如铜、锌等。这些盐含有阴离子,其包括但不限于乙酸盐、抗坏血酸盐、苯甲酸盐、酒石酸氢盐、溴化物、碳酸盐、氯化物、枸橼酸盐、叶酸盐、葡糖酸盐、碘酸盐、碘化物、乳酸盐、月桂酸盐、草酸盐、棕榈酸盐、过硼酸盐、苯磺酸盐、磷酸盐、丙酸盐、水杨酸盐、硬脂酸盐、琥珀酸盐、磺胺嘧啶、硫酸盐、硫化物、磺酸盐、酒石酸盐、硫代氰酸盐、巯基乙酸盐、硫代硫酸盐等。本发明也可以采用用于形成螯合物的氧化物实施,所述氧化物包括但不限于钙、钠、锂、铝、镁、钾、锰等的氧化物,并且也可以包括微动金属比如银、铜、锌、钛等的氧化物。本发明的组合物也可含有另外的医用化合物的任何组合。这种医用化合物包括,但不限于抗微生物剂、抗生素、抗真菌剂、抗病毒剂、抗血栓形成剂、麻醉剂、抗炎剂、镇痛剂、抗癌剂、血管舒张物质、伤口愈合剂、血管生成剂、血管抑制剂、免疫促进剂、生长因子和其他生物试剂。合适的抗微生物剂包括,但不限于双胍化合物,比如氯己定及其盐;三氯生;青霉素;四环素;氨基糖苷类,比如庆大霉素和妥布霉素(一种源自暗黑链霉菌(Streptomyces tenebrarius)的氨基糖苷抗生素)、多粘菌素;利福平;杆菌肽;红霉素;万古霉素;新霉素;氯霉素;咪康唑;喹诺酮,比如奥索利酸(oxolinic acid)、诺氟沙星、萘啶酸、培氟沙星、依诺沙星和环丙沙星;磺酰胺;壬苯醇醚9;夫西地酸;头孢菌素;和这种化合物和类似化合物的组合。另外的抗微生物化合物提供增强的抗微生物活性。
凝胶组合物也可以含有辅助组分。这种辅助组分的实例包括,但不限于粘度和流量控制剂、抗氧剂、常规着色颜料、脱气剂或消泡剂、及脱色剂。凝胶组合物也可含有常规染料和颜料,以赋予颜色或射线不透性,从而增强凝胶的美学外观。特别地,偶氮染料已经与凝胶基质一起被加入到多元醇螯合的金属氧化物复合物载体中,以提供增强的颜色外观。
凝胶也可以含有增加患者舒适性和组织健康的另外的润滑剂和其他添加剂。如之前提及的,在加入螯合的金属氧化物复合物之前、期间或之后,向多元醇载体中加入任何调味料或调味剂也是可能的。这些调味剂可以是人工(合成的)或天然存在的调味料。
虽然不希望受到下述机制的束缚,但是据信本发明组合物的许多有利的性质是由于多元醇悬浮液中存在的不同螯合的金属氧化物的溶解性差异导致的。在多元醇中金属氧化物的这些不同的溶解性,是确定浓度水平和导致沉淀以及提供活性微动金属的可变释放动力学的关键。例如,使用由本发明的组合物组成的或涂布其的医疗装置,当将该装置插入患者中时,那些具有高水溶性的螯合物将相当快地释放,提供高初始剂量的抗微生物活性以杀死细菌。该初始剂量有时被称为“快速杀死”,并且通过将装置或组合物置于细菌培养物中时在装置或组合物周围产生无细菌繁殖区的能力鉴定该抗微生物活性。该测试被称为“抑制区”测定。那些具有较低多元醇溶解性的螯合物将从亲水性组合物中更快地释放出,产生随时间推移持续或连续的抗微生物活性。
在组合物中具有不同溶解程度的螯合金属氧化物的选择允许该组合物适合凝胶组合物的特定应用。特别地,在局部或内部应用于哺乳动物且更优选人类期间,本发明的组合物可以通过使用快速释放抗微生物金属氧化物包围液体和组织,适于杀死引入的细菌,接着长期抑制细菌迁移和生长。调节微动试剂释放的能力有利地高于常规抗微生物凝胶组合物,因为它提供立即和持续抗微生物活性。
本发明的组合物的另一个优点是在凝胶组合物内形成悬浮在多元醇中的螯合物,其产生具有用于金属氧化物的最小粒径的超细颗粒。按照粒径分布,本发明的氧化银颗粒的初始粒度约为4至40微米。该最小粒径分布阻止了沉降和团聚,但是足够大,因此颗粒不被认为是纳米尺寸的。在组合物中使用悬浮的螯合物也允许结合较高量的抗微生物金属氧化物,而没有与其他相关技术有关的困难。
通过减少或消除与常规抗微生物凝胶组合物有关的问题,本发明提供了具有特定抗微生物螯合的金属氧化物浓度与特定抗微生物离子释放曲线的可再生组合物,其可以通过所选择的特定组合处理以在延长的时间段提供最佳抗菌活性。例如,本发明的组合物可以调节在1小时或至多5天之内释放大部分其微动试剂。通过改变螯合的金属氧化物在多元醇中相对高的初始浓度,物质的最终聚合组合物可适合于特定浓度的金属氧化物。微动试剂从组合物中的初始释放和释放持续时间取决于几个因素。这些因素包括在悬浮液中形成的具体螯合物的相对溶解度、在悬浮液(载体)中金属氧化物的浓度、和凝胶基质材料的至少相位(液体样或固体样)、密度和化学性质。该释放可以根据所制备凝胶的预期目的,通过在组合物中形成的金属氧化物螯合物的选择和数量调节。
为了更完整地定义多元醇的含义,在整个说明书和权利要求书中应用下述说明。即,糖醇,一类多元醇,因为其比糖的热值含量更低而通常被加入到食品中;然而,它们也通常更不甜,且通常与高强度甜味剂混合。因为它们不会被口中的细菌分解或代谢成酸,且因而不会导致蛀牙,它们也被加入到口香糖中。麦芽糖醇、山梨醇、木糖醇和异麦芽糖醇是一些更常见的类型。糖醇可以在温和的还原条件下由它们的类似物糖形成。这些多元醇可用于提供本发明的螯合金属盐的分散的悬浮液。
在高分子化学中,多元醇也是已知的,且对于具有多羟基官能团的化合物(其通常可用于有机反应)有特异性。具有两个羟基的分子为二醇,具有三个羟基的分子为三醇,具有四个羟基的分子为四醇,依次类推。
单体多元醇比如甘油、季戊四醇、乙二醇和蔗糖通常充当聚合多元醇的起始点。这些物质通常被称为"引发剂",可以与环氧丙烷或环氧乙烷反应,生成聚合多元醇,包括聚乙二醇。然而,不应当将它们与常用于促进其它聚合反应的自由基“引发剂”相混淆。用作聚合多元醇的起始点的官能团不必是羟基;存在大量适于提供胺官能性的重要的多元醇。伯氨基(-NH2)通常起两个聚合链的起始点的作用,特别是在聚醚多醇的情况下。
聚合多元醇通常用于生产其它聚合物。它们可以与异氰酸酯反应以制备常用于制备床垫、用于仪器(冰箱和冷冻机)的泡沫绝缘材料、家用和汽车座椅、橡胶鞋底、纤维(例如Spandex)和粘合剂的聚氨酯。
聚合多元醇通常是聚醚或聚酯。聚醚多醇是通过在催化剂,通常是强碱比如氢氧化钾或双金属氰化物催化剂比如六氰基钴酸锌-叔丁醇复合物的存在下,使环氧化物如环氧乙烷或环氧丙烷与多官能引发剂反应制备的。常见的聚醚二醇为聚乙二醇、聚丙二醇和聚(四亚甲基醚)二醇。如下显示的实例为基于甘油(三醇)的相当低分子量的三醇。聚醚多醇占工业使用的聚合多元醇的约90%;余量是聚酯多元醇。
另一类聚合多元醇为聚酯。聚酯是由二醇和二羧酸(或它们的衍生物)的缩合或阶梯式生长(step-growth)聚合化形成的,例如二甘醇与邻苯二甲酸反应。可选地,羟基和羧酸(或其衍生物)可以在同一分子内,如在己内酯的情况下。下述实例为可以通过季戊四醇(四醇)与γ-丁酸内酯反应获得的理想化的结构。
羟基-末端接的聚丁二烯为用于生产聚氨酯的多元醇。来自植物油的聚酯多元醇,称为天然油多元醇或NOP,可代替一些基于环氧化物的多元醇,也可以用于制备本发明的分散的金属氧化物悬浮液“载体”。
上列所有各种形式和类型的多元醇都可以用于分散螯合的金属氧化物和/或有助于在最终胶凝的基质中形成含有均匀分散的螯合金属氧化物物质(载体)的凝胶组合物。在将氧化银引入多元醇的情况下,为了提供更高浓度的氧化银,可能必须首先将螯合的氧化银悬浮在水或甘油或聚乙二醇(PEG)中,然后将注入的甘油并入到凝胶中。
在一个公开的实施方案中,本发明包括作为微动试剂的一种或多种螯合的氧化银形式,其与液相多元醇或甘油混合,得到螯合的氧化银在凝胶中的均质分散的悬浮液。在另一个实施方案中,所述组合物任选地含有其他微动金属比如锌、金、铜、铈等的另外螯合的金属盐。在仍然另一个实施方案中,所述组合物任选地包含铂族金属比如铂、钯、铑、铱、钌、锇等的另外螯合的盐。所述组合物任选地含有任何其它组分,其提供给组合物有益的性质或改善组合物的抗微生物功效。
在第二个方面,本发明涉及制备这些抗微生物凝胶组合物的方法。所述方法包括在溶液、分散液或凝胶和分散液的组合中形成微动试剂的螯合物。首先,形成螯合的金属氧化物悬浮液(在某些情况下,其为载体),然后可以将其加入到胶凝的基质中形成另一种(新)组合物,或者可以如同形成凝胶那样原位形成螯合的金属氧化物复合物。
一个优选的实施方案是用于螯合的金属氧化物氧化银,在将其加入到凝胶之前或期间,将其任选地与着色剂一起分散在甘油和/或水中,已知该着色剂提供颜色而不会干扰最终化合物的加工性、期望的颜色或物理化学性质。
因此,最终凝胶组合物可以为一个、两个或至少三个组分体系;螯合的金属氧化物悬浮液,加入到凝胶中的金属氧化物悬浮液,以及金属氧化物,凝胶组合物,和任选地加入的着色剂。四个或更多组分体系应当包括另外加入的有机和无机填料,比如抗氧剂、其它的抗菌组合物、调味剂、阻燃剂等,其会赋予最终凝胶组合物特定的附加性能和益处。
还发现在聚合体系中加入金属氧化物导致凝胶组合物具有变成导电的或至少半导电的能力,特别地用于膜和电线和电缆应用。另外,加入到基质凝胶中的螯合的金属氧化物将提供增强的物理化学性质。螯合的金属氧化物具有极大益处的应用是UV抗性。尽管通常与氧化锌有关,但是螯合的氧化银也显示出UV抗性。对于可以由这些化合物形成的一些产品,使用螯合的氧化铜、氧化镍、氧化锡和氧化铝可以提供更好的经济益处。对于两组分或更多组分凝胶系统的这些相关的性质也是本发明的一部分。
在大多数情况下,即使不是所有情况,与没有把螯合的金属氧化物复合入来自金属氧化物复合物的系统(载体系统)中的相同凝胶组合物相比,没有螯合的金属氧化物的聚合物的物理化学、物理和/或机械性质没有受到损害或者基本上没有受到损害。这也是本发明的非常重要的且可区别的特征。在商业价值方面,基于非常低浓度(通常小于1000ppm)的金属氧化物,独特的性质可以以相对较小增加的成本并入凝胶组合物中的事实,提供了相对于任何已知的凝胶添加剂技术(gel additive technology)非常显著的优点,特别是引入氧化银。
本发明凝胶(在聚合物内形成的螯合的金属氧化物)的用途包括几乎难以列举量的用途。如下列出的不旨在包括全部,但是已经基于期望的参数特征进行区分。
对于抗菌、抗微生物、抗病毒、抗-真菌和抗-霉菌凝胶,期望的制品包括至少下述的:
凝胶的医学和医院用途,其与下述的联合或与其分离:管、口罩、垫子、塑料杯、手术器械等,其还包括但不限于导管、套管、支架、导线、植入装置、隐形眼镜、IUD、蠕动泵室、气管导管、胃肠饲管、动静脉分流、避孕套、氧合器和肾膜、手套、起搏器导线、伤口敷料以及呼吸面罩和接触到皮肤或提供过滤人类吸入或摄取的液体的气体的机制的基本上任何产品。在医学界,需要减少或消除“葡萄球菌感染”是熟知的,本发明的凝胶已经证实提供了这些性质,而没有牺牲其它物理化学特性。这部分是由于加入相对低(ppm)浓度的螯合金属氧化物的性质引起的。

还包括那些引入食品可食用的甘油凝胶的凝胶,所述食品可食用的甘油凝胶含有可以注入到任何液体或固体食品物质的螯合的氧化银。螯合的氧化银不仅是GRAS(公认为是安全的),而且也是无毒的,且满足任何FDA要求,所以其可以与甘油用于构建用于动物和人类摄入的许多新的和独特的物品(固体食物、流体食物和包括水和冰的饮料)。
其中凝胶可存在有用性的其它家用产品包括浴室配件,比如浴室装置、地板、清洁产品、空气过滤物品、食品储藏物品HVAC、皮箱和办公室和学校产品。马桶座圈和移动厕所(包括用于飞机和汽车用途的那些)是与高水平的细菌有关的区域,其也得益于本文所述的技术。
多元醇中螯合的金属氧化物用于涂料、个人护理产品、水池(pool)和水疗(spa)产品、浆液和密封剂以及粘合剂是合适的(与所述凝胶复合和没有复合),并且通常用于其中霉菌和/或霉是关注源的家庭或工业/商业建筑的内部或外部。

育婴室、清洁用品、市售儿童高脚椅、给料器、门用五金(包括旋钮)、电子仪器、升降机、存储器、表面涂层和所有类型的运输产品也都得益于本文所述的技术。瑜珈垫和摔跤垫也是本发明涵盖的制品。
对于汽车内部,存在大量凝胶的应用,其包括用于方向盘和盖子、窗口把手和按钮、仪表板、无线电和导航控制等。
包括手机的所有形式的通讯装置和对于如个人计算机、移动装置、平板装置(iPad等)的所有外部设备,全部都是将得益于通过使用如本文所述的螯合的氧化银强化凝胶体系确保抗菌性质用途的制品。
在另一个方面,本发明涉及其包括凝胶的底物和抗微生物组合物的制品。在一个公开的实施方案中,凝胶组合物用于制备制品本身。因此,最终制品由单独或混合或应用在其它制品上或其中的一种或多种本发明的凝胶组合物组成。
任选地,可以向本发明的抗微生物凝胶组合物中加入另外的组分。这些组分包括,但不限于另外的微动试剂、另外的可溶性盐和提供给凝胶组合物有益的性质或增强凝胶组合物的抗微生物活性而没有干扰基本凝胶性质的任何其它组分。这样的组分包括,但不限于抗微生物剂、抗生素和其它医学试剂。

描述一种具有至少抗微生物、抗菌、和/或抗-病毒性质的初始液体组合物,其包含悬浮在多元醇中的螯合的金属氧化物颗粒,使得金属氧化物颗粒均匀分散在主要基于液体的多元醇载体中,并且使得所述螯合的金属氧化物颗粒形成稳定的复合物悬浮液,其可以最终变成碱性基水性氧化银分散液。所述悬浮液可以任选地进一步与壳聚糖的组合复合,其中得到的金属氧化物复合物以在0.1至25重量%之间的浓度提供。接着,液体组合物可以被加入到基本上任何凝胶或凝胶组合物/体系中。金属氧化物复合物也可以赋予胶凝的基质有益的半导电性或导电性以及渗透性和可燃性的性质变化。
在多元醇(“载体”)中形成的且最终俘获在形成本发明的抗菌制剂(基质)的凝胶内的螯合的氧化银复合物悬浮液的总量可以在宽参数内变化,但是对于组合物而言应当是起病菌-样(germ-like)屏障作用和提供有效地抑制、减少和基本上消除细菌繁殖的凝胶足量的。这也适用于赋予其它性质,包括降低渗透性(特别是对于氧)、可燃性和增加UV抗性。
通常,在一个实施方案中,多元醇中螯合的金属氧化物的抗菌、抗微生物、病菌屏障有效量通常在基于多元醇制剂总重量的0.001至10重量%范围内,因此人们可以将更少量和浓度的多元醇载体加入到分开或同时配制的凝胶中,以形成具有抗微生物有效量的氧化银复合物的最终凝胶组合物。
在另一个实施方案中,抗微生物组合物包括至少一种水相或甘油相配制的螯合的金属氧化物溶液,例如以选自油包水型乳剂、水包油型乳剂和复合型乳剂例如油包水包油型和水包油包水型三重乳剂的形式。
在另一个实施方案中,至少一个相包括多元醇和通常其它多元醇可溶性或可溶混性溶剂。溶剂可以选自短链一元醇,例如C1-C4的一元醇,比如乙醇和异丙醇;以及二元醇和多元醇自身,例如乙二醇、1,2-丙二醇、1,3-丁二醇、己二醇、二甘醇、二丙二醇、2-乙氧基乙醇、二甘醇一甲基醚、三甘醇一甲基醚和山梨醇。
在仍然另一个实施方案中,载体溶媒包括丙烯基乙二醇和/或甘油。用于制备制剂的抗菌部分的另一种有用组分是使用丙二醇。
在一个进一步的实施方案中,组合物包括至少一种与水不混溶的有机液相。所述至少一种与水不混溶的有机相通常包括至少一种导致该相与水不混溶的疏水性化合物。所述至少一种与水不混溶的有机相在室温(20至25摄氏度)下为液体(不存在结构化剂)。
在另一实施方案,所述至少一种与水不混溶的有机液相选择为用于螯合的氧化银的载体,并且可以来自油和/或油类的混合物,包括至少80%在25摄氏度下具有不超过4kPa(30mmHg)的蒸气压的化合物。所述至少一种与水不混溶的有机液相例如包括至少一种软化剂油,选自挥发性和非挥发性、聚硅酮基和烃-基软化剂油。这些软化剂油为例如在美国专利4,822,596和4,904,463中描述的,通过援引并入本文。
如本文使用的挥发性聚硅酮以已知的方式定义如在室温下具有挥发性的化合物。在这些化合物中,可以提及例如二甲基硅氧烷类型的环状和直链挥发性聚硅酮,其链包括3至9个聚硅酮-基残基。如本文使用的非挥发性聚硅酮以已知的方式定义为在室温下具有低蒸气压的化合物,比如聚烷基硅氧烷,比如直链聚烷基硅氧烷,包括直链聚二甲基硅氧烷或二甲硅油;聚烷基芳基硅氧烷,例如聚甲基苯基硅氧烷;以及聚醚和硅氧烷的共聚物,例如二甲硅油共聚物。在可以使用的非挥发性软化剂油中,实例包括烃基衍生物、矿物油、脂肪醇、C3-C18醇与C3-C18酸的酯、苯甲酸与C12-C18醇的酯及其混合物、C2-C2-C6多元醇例如选自甘油、丙二醇或山梨醇、聚亚烷基二醇聚合物。
载体物质和/或表面活性剂的混合物也用作螯合的金属氧化物的载体。以组合物总重量的重量百分数表示,使用的载体物质总量在一些实施方案中为1%至99%,在其它实施方案中为60%至98%。对于大多数凝胶体系,螯合的金属氧化物复合物通常的加入量为至多250至1000ppm重量。对于食品和饮料,该百分比通常更小。
在一个单独的实施方案中,抗微生物组合物进一步包括至少一种赋予凝胶组合物颜色或其它美学的其它试剂,包括非聚合的或聚合的有机结构化剂。非聚合的结构化剂的实例包括,但不限于着色剂或染料。
抗微生物螯合的金属氧化物组分分散的悬浮液可以分散在一种或多种选自在大多数情况下含有更低重量百分浓度的任一种下述物质的凝胶中:聚乙烯醇、聚乙烯吡咯烷酮、聚环氧烷、纤维素、纤维素衍生物、多糖、多糖衍生物、聚羧酸、聚羧酸的盐、聚氨基酸、肽、聚酰胺、聚丙烯酰胺、聚酯、聚(乙烯基甲基醚-共-马来酸酐)、藻酸盐、藻酸盐衍生物、果胶、聚环氧乙烷、明胶、角叉菜胶、壳聚糖、淀粉、淀粉衍生物、及其组合。这些“添加剂”在室温或环境条件下可以在液相或固相中,且能够保持螯合的金属溶液悬浮液是稳定的状态,使得将分散的螯合金属悬浮液掺合、混合和/或复合到凝胶基质中不会减少或消除用于制备最终凝胶组合物的最终凝胶组合物的抗菌作用。
本发明的凝胶组合物也可以包括其它组分,其可以根据胶凝的基质中制剂的载体和/或预期用途进行选择。任选的组分以不会实质上或不利地影响期望的抗菌作用的量使用。
除了螯合的金属氧化物复合物的防腐性质之外,另外的实施方案包括使用至少一种防腐化合物与局部抗擦伤螯合的氧化银凝胶复合物质的组合。防腐化合物的存在量可以为制剂的0.5%至约3%重量。期望地,防腐化合物有效地抗击酵母菌,特别是白色念珠菌(Candida albicans);霉菌,特别是黑曲霉(Aspergillus niger);和细菌,特别是金黄色葡萄球菌(S.aureus)、大肠杆菌(E.coli)和阴沟杆菌(E cloacae)。实例包括乙二胺四乙酸二钠、尼泊金甲酯和二偶氮烷基脲。螯合的氧化银复合物也充当螯合剂,用于阻止肺炎克雷白杆菌(Klebsiella pneumoniae)、奇异变形杆菌(Proteus mirabilus)和大肠杆菌产生的细菌尿素酶、脂肪酶、蛋白酶和脱羧酶的活性。
另外,螯合的氧化银可以充当药物、抗生素和其它药物的防腐剂。
在一个其它实施方案中,所述抗菌基于多元醇的金属氧化物液体复合物悬浮液和最终凝胶组合物也可包括香料。实例包括,但不限于柑橘(citrus)、花(floral)、辛辣(spicy)、熏衣草(lavender)、木香(woody)、生苔(mossy)、东方香(oriental)、草药(herbal)、皮革-烟草(leather-tobacco)和醛香(aldehydic)组。通常,香料呈浓缩物提供,其通常含有至多约3%重量的芳香剂。实例包括天然产物,比如精油、花油、来自树脂的天然提取物、树胶、香胶、豆类、苔藓和其它植物,和动物产品比如龙涎香和麝香,以及合成的芳香物质。
用于制备组合物的方法和记录结果
在第一个方面,本发明涉及用于制备本发明的组合物的方法。一般来说,所述方法包括形成在多元醇中首先制备的微动试剂的悬浮的螯合物,同时或接着在存在或不存在使用压力和/或加热下,经由混合/掺合/复合将其加入到已经完成形成的凝胶组合物中。通常,首先形成螯合的金属氧化物悬浮液,接着,加入到凝胶组合物中,或者可以在胶凝期间原位形成。
这得到基于多元醇水或甘油或PEG或所有三种的螯合的氧化银复合物悬浮液,其在室温下为液体。研究液体悬浮液的方法需要下述步骤,比如如下实施例1中所述。
实施例1∶用于制备250ppm均质螯合的氧化银凝胶的方法
形成用于基于多元醇的液体复合物悬浮液的螯合的氧化银的方法最好为如下描述的:
以500克基础制备在水/甘油/PEG基质中悬浮的4000ppm浓度螯合的氧化银溶液,所有物质都在混合前称重出;
1.在单独的容器中,制备蒸馏(DI或其它合适的过滤水,其不包含离子)水,并加入1-5重量%(优选地2%)浓度的壳聚糖。接着,加入α-羟基酸,在该情况下为羟乙酸,其起完全分散和溶解壳聚糖的作用。将该浆液溶液置于一边。
2.在单独的容器中,如下制备酸溶液:通过取5.65wt.%(28.25克)的枸橼酸,将该酸加入5.65%(28.25)克的蒸馏水,加热至82摄氏度并在快速搅拌下混合3-4分钟,在浆液中以高速(再次1000rpm或更大)继续混合3-4分钟。该溶液的pH将保持稳定在1.83。也将该溶液置于一边。
3.如下制备于水中AgO的第三个溶液:取0.47wt.%(2.35)克的氧化银粉末(正态粒径分布在4-40微米之间)-其在该情况下是来自Colonial Metals of Elkton,MD的氧化银(I),并在180华氏度(82摄氏度)下,以高速(至少1000rpm)掺合到31.54wt.%(157.7)克蒸馏水中(在该情况下使用水,但是此处也可以使用甘油和/丙二醇和/或PEG)至多3分钟。该浆液的pH应当升高达至少8.0,且优选地在8.3至8.4之间(推测由于产生了AgOH)。超过3分钟将引起氧化过多。将该深色浆液溶液置于一边。
4.将上述溶液(1)和(2)混合到单个容器中。然后,用滴定管(滴定类方法)将氧化银溶液(3)快速加入到在单个容器内慢慢搅拌的(1)和(2)的溶液中。(1)和(2)的总重量占比将为最终溶液的56.18重量%,43.82重量%将为AgO溶液的占比,最初将其在82摄氏度下全部混合在一起。该方法将确保在螯合过程期间不形成枸橼酸银,并且这点对该方法很重要。螯合反应非常快(在将AgO溶液(3)完全加入到混合溶液(1)和(2)中之后的小于3分钟之内)。
5.温度处于82摄氏度,反应似乎在几分钟内完成(没有银粉末保留在溶液中的证据),且溶液变澄清。在该阶段,优选地使用碳酸钠和/或精氨酸或少量NaOH有可能中和pH至7或更高,以确保不出现沉淀。精氨酸和其它氨基酸似乎结合溶液中螯合的氧化物复合物。
此时,500gm.螯合的氧化银复合物为悬浮液(充当载体)溶液,其为均质的、稳定的且随时可使用的。使用这5个步骤的方法,使氧化银复合物的浓度稳定(没有沉淀)至至少4000ppm。形成沉淀会导致形成想要避免的氧化银悬浮液的其它复合物,因为沉淀不能有效地提供稳定性,该稳定性产生不含沉淀的悬浮液的有效抗菌、抗病毒和/或抗微生物活性。同样,据信在没有形成沉淀下,UV、导电性和/或渗透性/可燃性性质是唯一恒定且可再现的。
在该特定实施例中,加入悬浮在水溶液中的4000ppm螯合的氧化银。如上所述,还成功地施用使用其它多元醇,比如甘油、丙二醇、PEG或甚至任意多元醇与水的组合。一旦形成上述步骤1-5的基于溶液的复合物悬浮液,并且在该情况下用于水中的4000ppm浓度的螯合的氧化银稳定,就有可能将该液体悬浮液成功地结合到凝胶组合物中完成制成品。
根据下述步骤完成得到1,000克250ppm的凝胶组合物的方法:
1.在室温下,取736.8克的蒸馏水(83.98%),并在中度搅拌下,慢慢地加入44克的聚季铵盐(Celquat H-100)-4.415%。混合5分钟。
2.在室温下,取115.5克的USP甘油99%,并加入到H2O/聚季铵盐混合物中-1.155%。继续中度搅拌。
3.立即加入24.5克的聚季铵盐/透明质酸混合物(BHA-10),并继续中度搅拌-2.45%。
4.立即加入17.5克的25%精氨酸水溶液-1.75%。
5.立即加入61.5克含有壳聚糖的4,000ppm的螯合的氧化银液体悬浮液–6.25%。
在上述步骤1中第一次加入聚季铵盐之后约10分钟,将形成凝胶组合物。
为了制备60千克的凝胶,必须使用与制备500克所需要的相同方法制备3.69千克螯合的氧化银液体悬浮液。然后,如下完成凝胶:
1.在第一个容器中装满44.211kg的蒸馏水和2.649kg的聚季铵盐,其为阳离子的(比如Akzo Nobel生产的Celquat Akzo Nobel的美国分部设在Chicago Ill.),以及6.930kg.的USP级有机犹太洁食(kosher)认证的甘油,在室温下,通过慢慢地混合(避免形成气泡)将其全部分散在容器中。这引起凝胶化的立即开始。接着,向同一容器中加入1.470kg.的透明质酸与聚季铵盐聚合物(比如Ontario,Canada的Koda Corporation生产的)的组合。在搅拌2-4分钟之后,该溶解的胶凝溶液的pH小于7.0。
2.为了提高容器中该溶液的pH,在单独的容器中,将75wt.%蒸馏水和25wt.%精氨酸的溶液混合在一起。然后,将1.040kg的该溶液加入到上述步骤(1)的第一个容器中。这应当提高第一个容器中溶液的pH至至少7.4或更高。
3.然后,第三个步骤是在该情况下,在完成步骤(2)的10分钟之内,加入3.69kg的4,000ppm螯合的氧化银复合物悬浮液溶液,并用行星式(或其它合适的)搅拌器开始混合,以确保均匀性。
如此,使用阳离子的(带正电荷的)离子物质进行整个胶凝过程,以便得到触变凝胶而没有沉淀。如果在制备部分中存在阴离子,则有可能形成银盐的沉淀—其是不期望的。实际上,如果形成这些盐,则其将导致凝胶丧失一定比例的其触变性质。对于本发明,术语“触变”指凝胶的时间依赖性剪切稀化性质。当振摇、搅拌或以其它方式加压时,随时间推移,在静态条件下变稠(粘稠)的一些凝胶或液体将流动(变稀,粘性更低),因此关于粘度其是时间依赖性的。然后,它们需要一个固定时间恢复更粘的状态。
在该具体实例中,本发明的最终凝胶含有250ppm螯合的氧化银复合物。如下表1所示,已经证实所述凝胶完全消除了大肠杆菌(Escherichi coli)(e-coli)、奇异变形杆菌(Proteus mirabilis)、肺炎杆菌(Klebsiella pneumoniae)、粪肠球菌(Enterococcusfaecalis)、绿脓杆菌(Pseudenomas aeruginosa)ATCC 27853、蜡状芽胞杆菌(Bacilluscereus)35和蜡状芽胞杆菌(Bacillus cereus)ATCC 13061。通过外部实验室(Schulke,Inc.of 30Two Bridges Rd.,Fairfield,NJ 07004),使用TS Agar/TTC培养基作为用于最初允许这些细菌初始生长的培养物,进行该测试。使用相同的培养基制备第二培养物,但是加入了本发明的最终凝胶组合物。

表2中的结果来自加入在PEG(聚乙二醇)基质中2000ppm的螯合的氧化银复合物,接着,以20:1的降低(let-down)比加入到内部丙烯酸半光泽涂料(产品码240100)中。在PEG中氧化银螯合物的最终浓度仅为100ppm。然后,测试该涂料,如下确定抗微生物和抗菌性质:
使用涂有三层涂料的9×13玻璃皿;用每种105生物体悬浮液擦洗1/5区域。使该生物体悬浮液在所述皿表面上干燥约三十分钟。给所述皿表面接种绿脓杆菌(P.aeruginosa)(ATCC 9027)、金黄色葡萄球菌(S.aureus)(ATCC 6538)、大肠杆菌(E.coli)(ATCC 8739)、念珠球菌(C.albicans)(ATCC 10231)和黑曲霉菌(A.niger)(ATCC16404)。将接触载玻片置于接种的区域。在30-35摄氏度下,培养绿脓杆菌(P.aeruginosa)、金黄色葡萄球菌(S.aureus)和大肠杆菌(E.coli)72小时,并计数。在室温下培养念珠球菌(C.albicans)和黑曲霉菌(A.niger)5天。
从如下制备的溶液中收集到上述表2中的数据;
工作实施例2∶
按下述方式制备500ml量的基于多元醇的复合物悬浮液与100ppm的氧化银复合物:
溶液A:
265gms.枸橼酸,加入到299.735gms.含有2%壳聚糖溶液的蒸馏水中。混合5–10分钟。
浆液1
11gms.氧化银,加入到99.89gms.蒸馏水中,分散5-10分钟。
收集制备溶液A,并置于中度搅拌下规定的时间。慢慢地加入之前在水性浆液1中分散的氧化银,并以高速混合30-45分钟。得到的氧化银是一种螯合物,无色无味。移除搅拌,经由定性分析滤纸比如VWR 20415过滤,并储存在不透光的容器中。该氧化银复合物现在准备用于制备用于各种目的的甘油组合物。取30ml的2%壳聚糖溶液,向其中加入70ml的蒸馏水。混合1分钟,然后加入400ml之前制备的100ppm螯合的氧化银复合物。混合所有物质3分钟,然后加入到有或没有水的多元醇比如丙二醇和/或甘油中,作为稳定该悬浮液溶液的非沉淀的复合物悬浮液。
申请人已经发现当螯合的氧化银和多元醇和/或甘油复合成凝胶组合物时,其显示出组合了润滑性、加工性、抗微生物(以及半导电性/导电性、UV抗性、渗透性和可燃性改变)性质的所需独特的特性,满足了多用途标准。螯合的银或其它金属氧化物复合物,其也都是生物相容的,给凝胶增加了抗微生物性质,以提供减少或消除细菌、真菌、霉菌、病菌和微生物且在某些情况下病毒的污染。一系列证据表明相同的金属氧化物复合物将提供给凝胶和利用这些凝胶的后续制品导电性、UV抗性、渗透性和可燃性改变的性质。
在本发明中,预期使用不同于枸橼酸的酸类(比如乙醇酸、硝酸等)的其它金属氧化物螯合物。使用枸橼酸和氨基酸产生能被人体和其它哺乳动物摄入(无毒)的螯合的氧化银。对于外部应用于皮肤,上述凝胶组合物形式的螯合的氧化银非常安全有效。也可能提供无毒可摄入的以及“绿色”且可耐受的凝胶组合物-其也与前述研究者开发的其它金属氧化物组合物不同。
另外,可以向组合物中加入不会在溶液中反应但提供一些有益作用的其它盐,所述有益作用比如稳定悬浮液、改进螯合的金属氧化物的离子释放速率、促进电流作用、增加抗微生物功效或增强生物相容性。进一步,可以向组合物中加入其它化合物,包括,但不限于医学试剂、润滑剂、营养剂、抗氧剂、染料和颜料和其它添加剂。
如上所述,任何凝胶都可以用于形成本发明的组合物。当使用亲水性凝胶时,优选地该凝胶溶于水或含有一些水的有机溶剂中。向凝胶组合物中加入水而不使聚合物沉淀的能力允许直接加入水溶性盐。在凝胶组合物中使用水提高了所述盐的溶解性,导致形成更精细更稳定的胶体。
相反,当单独使用疏水性凝胶或与亲水性凝胶组合使用时,期望限制组合物中存在的水的含量,以避免凝胶的疏水性组分沉淀。在这样的情况下,凝胶组合物中水的存在量优选地为10%或更少。因此,当本发明中使用疏水性凝胶时,凝胶组合物的优选的水含量在约1至20%重量之间。当使用疏水性凝胶时,使用可溶于乙醇或有机溶剂的盐是有利的。
作为用于螯合氧化银的溶质样悬浮剂有用的多元醇包括,但不限于聚乙二醇、聚酯多元醇、聚醚多元醇、蓖麻油多元醇和聚丙烯酸酯多元醇,包括Desmophen A450、Desmophen A365和Desmophen A160(从Mobay Corporation可获得)、聚(己二酸乙烯酯)、聚(己二酸二甘醇酯)、聚己酸内酯二醇、聚己酸内酯-聚己二酸酯共聚物二醇、聚(乙烯基-对苯二甲酸酯)二醇、聚碳酸酯二醇、聚四亚甲基醚二醇、聚氧丙烯二醇的环氧乙烷加成物和聚氧丙烯三醇的氧化乙烯加成物。
本发明的螯合的氧化银溶液可以进一步被加工用于多种分散方法,但是对于本发明的目的,提供于甘油、PEG和/或水基质中的分散液。
本发明还涉及一种抗微生物局部组合物,其包含螯合的金属氧化物的复合物,且更特别地利用壳聚糖-氧化银的变体,特别地涉及包含复合的壳聚糖-氧化银生物膜的溶液,以及制备其形成螯合的氧化银悬浮液的基于多元醇的分散液的方法,所述分散液可以复合或掺合胶凝物质,得到具有当加工时基本上无法感知的物理化学性质变化的抗菌化合物。使用壳聚糖对于制备螯合的金属氧化物复合物悬浮液有帮助,因为其赋予所述复合物生物膜性质,对于应用于哺乳动物皮肤有用。
对于该实施方案,通过首先提供壳多糖形成壳聚糖溶液,壳多糖为β(1-4)-连接N-乙酰基-D葡糖胺的均聚物。然后,经洗涤、干燥且研磨的壳多糖可以进行脱乙酰过程,以使一些N-乙酰基-葡糖胺转化为壳聚糖的主要成分葡糖胺。接着,可以通过混合壳聚糖与α-羟基酸比如乙醇酸且使其变稠来制备壳聚糖溶液。可以通过混合氧化银与羧酸和壳聚糖溶液(比如枸橼酸)的组合形成氧化银螯合物来制备银溶液。因为壳聚糖溶液是阳离子的,所述银溶液通常可能是中性的,因此得到的银-氧化物壳聚糖复合物主要是阳离子的。本发明的阳离子溶液将友好地粘合通常带负电荷的人类皮肤。在使用时,枸橼酸盐有助于促进细菌摄入银。
根据本发明的一个优点,提供具有高的即时和短期效力的抗微生物溶液。这是由于细菌被吸引到溶液中的枸橼酸盐而实现。枸橼酸盐促进细菌摄入氧化银离子,产生杀死细菌的功效。实际上,如上表1和表2以及从独立试验实验室获得的其它数据所示,已经评价了包含壳聚糖-氧化银复合物的本发明针对沙门氏杆菌(salmonella)、大肠杆菌(e-coli)、MRSA(staph)、绿脓杆菌(pseudomonas aeroginosa)、粘质沙雷氏菌(serratiamarcescens)和肺炎杆菌(klebsiella pnuemoniae)的效力。
根据本发明的实施方案的另一个优点,提供具有高残余效力和稳定性的抗微生物溶液。这是因为抗微生物溶液不仅可以制备为相对稳定而且显示出低挥发性且不容易蒸发。在某种程度上,这应归于在任何螯合之前氧化银粉末在去离子水(蒸馏水或其它合适过滤的无离子水)中水合,以及如下事实:这导致氧化银粉末再次在任何螯合之前和加入壳聚糖和羧酸之前形成稀且最终碱性的浆液。使用将增大螯合的金属氧化物颗粒的某些碱性物质是不合适的,因为这会引起过早沉淀。
相关的且根据该实施方案的进一步的优点,提供容易粘合使用者皮肤使得其留在原地的抗微生物氧化银溶液。这是因为在该氧化银复合物悬浮液中,氧化银进一步键合壳聚糖以进一步形成形成生物膜的复合物,且因此形成带正电荷的或阳离子的分子而实现的。人体皮肤通常显示出负电荷,因此是阴离子的。壳聚糖-氧化银复合物对皮肤表面的天然静电引力允许该复合物粘合皮肤。实际上,实验室结果显示在实验室的条件下,100ppm壳聚糖-氧化银复合物具有大于24小时的残余功效。这是在没有使用合成物且没有利用乙醇、苯扎氯铵或三氯生下获得的。螯合的氧化银组分是活性成分,其使细菌快速接触预期表面以及提供其它螯合的银复合物观察不到的非常卓越的生物相容性性质。这主要是由于氧化银在螯合和复合之前在碱性介质中是稳定的事实。
根据又进一步的优点,提供抗微生物氧化银基于碱性的溶液,其是稳定的、可携带的且容易分配的。氧化银在水性碱性介质悬浮液中的稳定化很复杂,其进一步提供瞬时生物相容性和在细胞水平与大多数不期望的细菌、霉菌和/或真菌生物体的抗微生物作用。最新证据表明氧化银实际地在经由DNA和通过螯合作用提供细胞变化方面是有效的,据信当与其它已知的液体或固体形式的氧化银(或任何金属)-特别是胶态金属氧化物相比时,本发明的氧化银具有更高的细胞内活性。

虽然结合一个或多个优选的实施方案描述了本发明,但是应当理解不打算将本发明限制于那些实施方案。相反,其预期涵盖所有替代、修饰和等价物,如同可以包括在如所附权利要求书定义的本发明的精神和范围内一样。
提供形成壳聚糖的方法。也提供原材料。原材料为壳多糖。壳多糖是β(1-4)-连接的N-乙酰基-D-葡糖胺的均聚物。壳多糖是大量天然存在且可再生来源的生物聚合物。壳多糖存在于无脊椎动物的外骨骼中。在一个优选的实施方案中,壳多糖来源于十足甲壳类家族比如虾和对虾。如此获得的壳多糖通常具有约在500至900k道尔顿的分子量。这些步骤是本发明所必需的,因为壳多糖是不溶性的。
通过除去非壳多糖组分处理壳多糖。在一个实施方案中,通过使用盐酸(HCL)完成该步骤。HCL除去或剥离附着于壳的任何残留肉组织。应当理解,在没有背离本发明的广泛方面下,可以结合剥离残留肉组织的其它酸或方法,在剥离残留肉组织之后,使用氢氧化钠(NaOH)溶液洗涤并中和外骨骼。在优选的实施方案中,使用约20%的NaOH溶液。
将壳多糖干燥,并将壳多糖处理成具有期望的尺寸。优选地,研磨壳多糖使得其具有约24目的平均粒径(平均颗粒尺寸0.0278英寸)。
接着,在优选的实施方案中,脱乙酰化包括混合1份壳多糖与4份50%NaOH,其为向其中加入1份纯水的碱。得到的混合物总共包括5份,其中所述溶液具有40%NaOH/1份壳多糖。将该混合物加热至约70摄氏温度约72小时,以进行脱乙酰化过程。脱乙酰化过程将一些N–乙酰基葡糖胺转化成葡糖胺。脱乙酰化的结果是壳聚糖分子聚集和沉淀。
移除壳聚糖以除去剩余的NaOH和任何其它杂质。在该优选的实施方案中,洗涤壳聚糖的步骤包括三次洗涤。还应当理解在没有背离本发明的广泛的方面下,可以可选地使用其它大量的洗涤。然后,优选地使壳聚糖干燥。
现在转向图1,提供制备2%壳聚糖溶液的步骤10。
下述优选的实施方案得到约1升或1000ml螯合的氧化银复合物。该方法(10)的第一步(11)是提供去离子水。测量182ml的去离子水,并放置于中度至高度搅拌下。然后,在步骤(12)中提供20克的壳聚糖(洗涤并干燥),并测量。在中度至高度搅拌下,将壳聚糖粉末分散在去离子水中并与其混合。接着,在步骤(13)中,提供α-羟基酸比如乙醇酸。在优选的实施方案中,使用没有强烈气味且具有约70%纯度的乙醇酸。应当理解,在没有背离本发明的广泛的方面下,可以使用其它α-羟基酸。可以加入约45ml的乙醇酸,并且可以慢慢地混合该混合物约另外45至60分钟。在该时间段之后,该混合物优选地为粘性的。当获得期望的粘度时,该壳聚糖溶液优选地备用。
现在转向图2,提供制备氧化银溶液的方法(20)。首先,在步骤(21)中,可以提供银盐。主要地,提供使用粉末形式的氧化银。接着,在步骤(22)中提供水和氧化银粉末的碱性水溶液。在步骤(23)中,结果是形成碱性水溶液形式的氧化银。一旦产生且完全形成氧化银稀浆液,则可以一起提供枸橼酸与较稠的壳聚糖(在步骤24中),并且可以在步骤25中提供去离子水。
使用下述步骤得到构成的1000ppm螯合的氧化银溶液的1升或1000ml批次。通过将约1.10克螯合的氧化银加入到约198.9克蒸馏水中并分散约5-10分钟制备浆液1。可以将在水浆液1中分散的螯合的氧化银慢慢地加入到壳聚糖和枸橼酸的溶液中,以形成第二溶液(2),以高速混合约30-45分钟。得到的氧化银螯合物无色无味。移除搅拌,经由定性分析滤纸比如VWR 415过滤,并储存在不透光的容器中(在步骤26中)。
首先,将银分散在蒸馏水中形成稀浆液,使得存在银表面积的更大暴露,形成可以与枸橼酸结合的氧化银分子。Ag2O分子在溶液中仅微溶;因此,向该混合物中加入枸橼酸也增加了与壳聚糖的溶解度,产生了由于路易斯酸-碱反应形成配位化合物的氧化银化合物的银离子部分。此处,银离子为酸(受体),壳聚糖/枸橼酸溶液起配体基质(供体)的作用。
特别地,所形成的溶液为形成生物膜的消毒剂,所述消毒剂为阳离子和生物粘附性的,且含有浓度足以产生剩余抗菌活性数小时的螯合的氧化银。一旦加入到如上所述的凝胶组合物中,则该溶液通过将螯合的氧化银注入到最终凝胶组合物中赋予相同的性质。
图3提供将抗微生物复合物注入到凝胶中以制备本发明的螯合的金属氧化物凝胶组合物的方法(30)。
为了获得1升批次或1000ml批次,使用下述比例。首先,在多个步骤中,提供包括枸橼酸的约500ml的2%壳聚糖溶液和约400ml的去离子水。优选地,将壳聚糖、枸橼酸和去离子水慢慢地混合约3分钟(步骤31&32)。接着,提供约100ml的1000ppm氧化银溶液(步骤33)。将氧化银溶液加入到壳聚糖溶液中,并优选地慢慢地混合该溶液另外3分钟(步骤34)。得到的溶液为含有100ppm的氧化银和生物结合的壳聚糖的制剂。应当理解,在该方法中的所有容器和搅拌器都由高密度塑料或玻璃制成,且必须不含金属表面。将得到的溶液加入到凝胶组合物(步骤35)中,按照上述提供1,000克250ppm的上述凝胶组合物的方法讨论中所提供。
应当理解,在没有背离本发明的广泛的方面下,可以使用其它比例的壳聚糖溶液与氧化银溶液。
根据所述本发明,枸橼酸盐与银离子和氧化银以及壳聚糖复合,因此细菌接受该氧化银枸橼酸盐壳聚糖复合物。在接受银之后,不想要的细菌、病毒、霉菌和真菌快速死亡,因为银立即使至关重要的蛋白质以及细菌的代谢和再生功能丧失作用,该生物体往往在几分钟内死亡。然而,该螯合的氧化银化合物提供给期望的细胞和生物体生物相容性,使得当使用如上详述处理的螯合的氧化银复合物时没有任何已知的毒性副作用。
氧化银复合物是阳离子性的,容易结合带负电荷的人或动物皮肤,而没有任何毒性作用。壳聚糖-氧化银复合物不会以比如对于许多胶体(离子)银化合物已知的方式引起银中毒。这是由于分子本身的机制产生的。当吸收到皮肤中时,所述复合物立即变成惰性,因为其结合我们身体中和我们的皮肤上天然存在的游离钠离子,并且氧种类主要起促进稳定和健康细胞生长的作用。根据个体的活动水平,螯合的氧化银分子最终经由肾脏或从皮肤的毛孔排出。
虽然已经结合其特定实施方案描述了本发明,但是很显然根据前述说明书,许多替代、修饰和变化对本领域技术人员来说将是显而易见的。因此,本发明意图涵盖如落入所附权利要求书的精神和宽范围内的所有这些替代、修饰和变化。
Claims (30)
1.抗微生物、抗菌和/或抗-病毒的均质凝胶组合物,其包含:不含沉淀的螯合金属氧化物液体复合物悬浮液和不含螯合的金属氧化物的凝胶组合物的组合,所述螯合金属氧化物液体复合物悬浮液是基于稳定的均质水溶液的,所述凝胶组合物包含水、聚季铵盐、甘油和透明质酸,其中所述均质凝胶组合物含有均匀悬浮在所述均质凝胶组合物中的螯合的金属氧化物颗粒,使得所述均质凝胶组合物内含有的所述螯合的金属氧化物颗粒的浓度为至少0.001重量%的浓度。
2.权利要求1的均质凝胶组合物,其中所述均质凝胶组合物不形成沉淀。
3.权利要求1的均质凝胶组合物,其中所述螯合的金属氧化物颗粒初始是氧化银颗粒,其与水一起构成碱性氧化银水分散液,其中初始加入氧化银颗粒到去离子水和/或蒸馏水中使所述水分散液的pH从5.5至7.0之间上升至7.7至8.5之间。
4.权利要求3的均质凝胶组合物,其中所述初始水分散液与壳聚糖和羧酸的组合进行复合,从而得到所述螯合的金属氧化物,其中所述螯合的金属氧化物颗粒的所述金属氧化物是螯合的氧化银颗粒,且其中所述氧化银颗粒的粒径分布在4至40微米之间。
5.权利要求1的均质凝胶组合物,其中所述金属氧化物颗粒的所述金属为下述的至少一种:银、铜、锌、钛、金、镍和锡氧化物。
6.权利要求1的均质凝胶组合物,其中所述组合物包括L-精氨酸和二糖聚合物,且其中所述二糖包括与透明质酸的聚合物。
7.权利要求1的均质凝胶组合物,其中所述螯合的氧化银颗粒在所述凝胶组合物中的浓度为在10至4000ppm之间,在所述凝胶组合物中的浓度优选地为在100至250ppm之间,在所述凝胶组合物中的浓度最优选地为250ppm。
8.权利要求1的均质凝胶组合物,其中将所述均质凝胶组合物混合到胶凝的基质中,其中所述胶凝的基质也可以是聚合物基质,且所述聚合物包括生物聚合物。
9.权利要求1的均质凝胶组合物,其中所述凝胶组合物与下述物质混合和/或注入到其中或以其它方式直接用作下述物质:调理剂、发用摩丝、喷发胶、染发剂、隐形眼镜溶液、除臭剂、个人和工业润滑剂、食品级可食用物质、冰形式的水、皮肤洗剂、局部制剂、牙膏、口服凝胶剂、和食用油脂的替代物。
10.权利要求1的均质凝胶组合物,其中所述凝胶组合物的所述凝胶是用阳离子体系形成,其中所述阳离子体系可以是阳离子聚合物体系,且其中所述阳离子聚合物体系选自藻酸盐、壳聚糖、葡聚糖、角叉菜胶、果胶和黄原胶。
11.抗微生物、抗菌和/或抗-病毒的均质凝胶组合物,其包含:螯合金属氧化物液体复合物悬浮液和不含螯合的金属氧化物的凝胶组合物的组合,所述螯合金属氧化物液体复合物悬浮液是基于稳定的均质多元醇溶液的,所述凝胶组合物包含水、聚季铵盐、甘油和透明质酸,其中所述均质凝胶组合物含有螯合的金属氧化物颗粒,其均匀悬浮在所述均质凝胶组合物中,使得所述均质凝胶组合物内含有的所述螯合的金属氧化物颗粒的浓度为至少0.001重量%的浓度。
12.权利要求11的均质凝胶组合物,其中所述多元醇为选自甘油、丙二醇和聚乙二醇中的一种或多种。
13.权利要求11的均质凝胶组合物,其中所述均质凝胶组合物不形成沉淀。
14.权利要求11的均质凝胶组合物,其中所述螯合的金属氧化物颗粒初始是氧化银颗粒,其与水一起构成碱性氧化银水分散液,其中初始加入氧化银颗粒到去离子水和/或蒸馏水中使所述水分散液的pH从5.5至7.0之间上升至7.7至8.5之间。
15.权利要求14的均质凝胶组合物,其中所述初始水分散液与壳聚糖和羧酸的组合进行复合,从而得到所述螯合的金属氧化物颗粒,且其中所述氧化银颗粒的粒径分布在4至40微米之间。
16.权利要求11的均质凝胶组合物,其中所述金属氧化物颗粒的所述金属为下述的一种或多种:铜、锌、钛、金、镍和锡氧化物。
17.权利要求11的均质凝胶组合物,其中所述组合物包括L-精氨酸和二糖聚合物,且其中所述二糖包括与透明质酸的聚合物。
18.权利要求11的均质凝胶组合物,其中所述螯合的氧化银颗粒在所述凝胶组合物中的浓度为在10至4000ppm之间,且其中所述螯合的氧化银颗粒在所述凝胶组合物中的浓度更优选地为在100至250ppm之间,在所述凝胶组合物中的浓度最优选地为250ppm。
19.权利要求11的均质凝胶组合物,其中将所述凝胶组合物混合到胶凝的基质中,其中所述胶凝的基质也可以是聚合物基质,且所述聚合物包括生物聚合物。
20.权利要求16的均质凝胶组合物,其中所述凝胶组合物与下述物质混合和/或注入到其中或以其它方式直接用作下述物质:调理剂、发用摩丝、喷发胶、染发剂、隐形眼镜溶液、除臭剂、个人和工业润滑剂、食品级可食用物质、冰形式的水、皮肤洗剂、局部制剂、牙膏、口服凝胶剂、和食用油脂的替代物。
21.权利要求11的均质凝胶组合物,其中所述凝胶组合物的所述凝胶是用阳离子体系形成,其中所述阳离子体系可以是阳离子聚合物体系,且其中所述阳离子聚合物体系选自藻酸盐、壳聚糖、葡聚糖、角叉菜胶、果胶和黄原胶。
22.权利要求11的均质凝胶组合物,其中所述凝胶被混入或注入到颜料以及个人护理产品比如洗发剂、发胶、唇膏和洗手液、以及水池和水疗产品、浆液和密封剂中,以及粘合剂中。

24.权利要求11的均质凝胶组合物,其中所述凝胶被注入到水中,其中所述水在液相、固相或汽相中。
25.权利要求11的均质凝胶组合物,其中所述凝胶被直接注入到热塑性加工设备中以将所述螯合的金属氧化物的性质直接注入到任何制品中。
26.权利要求11的凝胶均质组合物,其中所述凝胶被注入到任何海鲜产品中或与冰组合使用以确保在海鲜储存中减少或消除细菌生长。
27.制备均质凝胶组合物的方法,所述均质凝胶组合物包含:氧化银和水和/或多元醇螯合氧化银的混合物,所述混合物也含有共复合的生物膜形成剂,其中所述凝胶组合物静电结合带负电荷的表面,其中所述凝胶组合物包含:悬浮在所述混合物中的螯合的氧化银颗粒和包含水、聚季铵盐、甘油和透明质酸的凝胶,其中将所述螯合的氧化银颗粒均匀地分散和注入到所述凝胶中产生所述均质凝胶组合物,其中螯合的氧化银颗粒在所述均质凝胶组合物内以稳定复合物悬浮液存在,使所述螯合的氧化银颗粒的浓度为至少0.001重量%且保持悬浮在所述均质凝胶组合物中。
28.权利要求27的均质凝胶组合物,其中所述凝胶组合物不形成沉淀。
29.权利要求27的均质凝胶组合物,其中所述共复合生物膜形成剂为选择量的羧酸和壳聚糖。
30.权利要求27的均质凝胶组合物,其中所述凝胶组合物包含250ppm的所述螯合的氧化银。
