Abstract
本发明公开了一种玉米低聚肽保健品的制备方法。本发明通过物理方法对玉米蛋白粉进行预处理,然后通过酶解制备玉米低聚肽,有效提升了玉米低聚肽的生成效率以及品质,在此基础上,将制得玉米低聚肽,并配以其他保健成分协同作用,制得的产品具有增强免疫力的作用,能有效改善人体的健康状况。

技术领域
本发明涉及一种玉米低聚肽保健品的制备方法,医药保健领域。
背景技术
随着社会物质文明的进步,人们的生活方式和行为习惯发生了巨大变化,工作压力大、生活节奏快、缺乏体力锻炼等现象日益普遍,导致现代人出现失眠、乏力、无食欲、易疲劳、心悸,抵抗力差、易激怒、经常性感冒或口腔溃疡、便秘等“亚健康”状态。长期处于“亚健康”状态,会导致机体的免疫力下降。另一方面,我国已经进入老龄化社会,由于机体的衰老,老年人的免疫力大幅度降低,导致免疫相关疾病的发病率急剧增高,很大程度上影响了老年人的生活质量。对于免疫力低下人群,提高免疫力对维持机体健康具有重要意义。
玉米是我国重要的农作物之一,种植历史早,种植面积大,年产量超过2.2亿吨。玉米曾是人类最重要的一种食物来源,随着人们生活水平的不断提高,玉米的主食地位已经逐渐被其它种类繁多的食品所取代,致使玉米的主要用途从食用转向了工业利用。
玉米低聚肽粉是以玉米蛋白粉为原料,经调浆、蛋白酶酶解、分离、过滤、喷雾干燥等工艺生产而成的。玉米低聚肽粉特殊的氨基酸组成结构,决定了它除具有肽类物质的优良特性——优于氨基酸或蛋白质的直接吸收、溶解性强(在大范围的pH值下均能完全溶于水,无浑浊和沉淀物产生)、稳定性强(对热稳定,组分不改变,功能不丧失)、安全性高(天然食品蛋白,安全可靠,无毒副作用)等特性以外,还具有自己所独有的特殊功能。玉米低聚肽具有多种生物活性,如抗氧化、抗高血压、增强免疫力、抗疲劳、保护肝脏等。
当前玉米肽的制备大多采用酶解的方式,特定的酶来水解玉米蛋白粉中的抗原蛋白,其作用效果受到酶的种类、水解前处理方式、水解程度等诸多因素的影响,现有技术采用的方法均存在效率低下,制备的玉米肽口感差等问题。
发明内容
本发明的目的在于提供一种玉米低聚肽保健品的制备方法,以解决现有技术中存在的问题,本发明通过物理方法对玉米蛋白粉进行预处理,然后通过酶解制备玉米低聚肽,有效提升了玉米低聚肽的生成效率以及品质,在此基础上,将制得玉米低聚肽,并配以其他保健成分协同作用,制得的产品具有增强免疫力的作用,能有效改善人体的健康状况。
为实现上述目的,本发明提供的一种玉米低聚肽保健品的制备方法,包括如下方法,包括如下步骤:
(1)对玉米蛋白粉进行物理预处理
玉米蛋白粉与水的混合物在110℃以上的温度下加热,并将加热所得产物进行双螺杆挤压,得到预处理玉米蛋白粉;
(2)酶解反应得到玉米低聚肽
(21)使用一种或多种微生物蛋白酶酶解所述预处理玉米蛋白粉,灭酶后得到酶解液;
(22)将酶解液解液离心后,对离心上清液进行膜过滤,得到滤液;
(23)在膜过滤后,对滤液进行脱色和浓缩;
(24)在浓缩后可进行灭菌和干燥,从而制得玉米低聚肽粉;所述干燥为喷雾干燥;
(3)配制玉米低聚肽保健品原料
按照如下重量组分进行配料:
(4)将上述原料混合均匀,经过杀菌处理后,真空包装得到产品。
优选的,在所述步骤(1)中,玉米蛋白粉与水的质量比为80:20-60:40,加热温度为150-170℃。
优选的,在所述步骤(1)中,将加热所得产物进行双螺杆挤压的步骤,其中,挤压机末端机筒温度为150-170℃,挤压机前三段机筒的温度可分别为30-50℃,50-70℃,70-90℃。
优选的,在所述步骤(21)中,优选的,所用的微生物蛋白酶的量为预处理玉米蛋白粉质量的0.5-1.5%;所用的微生物蛋白酶为碱性蛋白酶、中性蛋白酶和酸性蛋白酶中的一种或多种;酶解温度为45-55℃。
优选的,在所述步骤(22)中,所述离心的转速为3000-5000r/min,离心可采用常规设备进行,例如卧螺离心机、管式离心机等。此外,可采用孔径为50-100nm的滤膜进行所述膜过滤;膜过滤时,可控制膜过滤的绝对压力为0.1-0.3MPa,温度为50-70℃。
优选的,在所述步骤(23)中,优选的,可以采用常规脱色剂进行脱色,脱色剂为活性碳粉,脱色剂与滤液的质量配比可以为(10-15):100,脱色的温度可控制在60-80℃,脱色时间为10-30min,脱色可在搅拌下进行,在脱色后,可通过过滤去除脱色剂。
优选的,采用双效降膜蒸发器进行蒸发浓缩,并且可控制蒸发时的蒸汽压为0.05-0.08MPa,蒸发温度为50-70℃,经浓缩后,浓缩液的体积可降至原体积的1/4-1/2。
优选的,所述杭白菊粉是以干燥杭白菊为原料,经粉碎、浸泡、水煮、过滤、浓缩、灭菌、干燥处理而制得,其总黄酮含量达5.0wt%以上;
优选的,所述粟米粉是以粟米为原料,经粉碎、酶解、过滤、浓缩、干燥处理而制得。
其中,余甘子为大戟科植物余甘子的干燥成熟果实,味甘、酸、涩,性凉,归肺、胃经,具有清热凉血,消食健胃,生津止咳功效,其维生素C、鞣酸等抗氧化物质含量丰富,尤其是富含的天然维生素C,能促进机体胶原蛋白的合成。
其中,杭白菊性味辛、甘、苦,微寒,具有疏散风热、平肝明目、清热解毒功效,其富含黄酮、酚酸等活性成分,具有抗氧化、抗炎、保护心脑血管等作用,其所含的黄酮类化合物与余甘子中的维生素C能发挥协同作用,增加活性物质的生物利用度。
本发明相对现有技术,具备以下特点和显著效果:
(1)物理方法对玉米蛋白粉进行预处理,可提高玉米蛋白粉的酶解效率、缩短酶解时间,并可提高谷酶解产物中分子量小于1000Da的组分;
(2)选用微生物蛋白酶,并配以优化的酶解工艺,有效提升了玉米低聚肽的生成效率以及品质;
(3)本法在制备玉米低聚肽粉时,取酶解液离心上清液进行膜过滤,可进一步地截留分子量较大的成分,从而最大限度地去除酶解液中的大分子蛋白组分,使得短肽含量高;
(4)本发明将玉米低聚肽粉与松花粉、余甘子粉、杭白菊粉、粟米粉、紫薯粉和葡萄糖成分组合,通过各组分间的协同作用,实验数据表明能增加小鼠抗体生成细胞数的能力,能提高小鼠巨噬细胞吞噬能力和NK细胞活性,具有增强免疫力的作用,可用于改善机体的健康状况。
具体实施方式
下面结合实施例对本发明的实施方案进行详细描述。
实施例1
玉米蛋白粉与水的混合物在110℃以上的温度下加热,并将加热所得产物进行双螺杆挤压,得到预处理玉米蛋白粉;玉米蛋白粉与水的质量比为80:20,加热温度为150℃。将加热所得产物进行双螺杆挤压的步骤,其中,挤压机末端机筒温度为150℃,挤压机前三段机筒的温度可分别为30℃,50℃,70℃。
使用微生物蛋白酶酶解所述预处理玉米蛋白粉,灭酶后得到酶解液;所用的微生物蛋白酶的量为预处理玉米蛋白粉质量的0.5%;所用的微生物蛋白酶为碱性蛋白酶;酶解温度为45℃。
将酶解液解液离心后,对离心上清液进行膜过滤,得到滤液;其中,所述离心的转速为3000r/min,离心可采用常规设备进行,例如卧螺离心机、管式离心机等。此外,可采用孔径为50nm的滤膜进行所述膜过滤;膜过滤时,可控制膜过滤的绝对压力为0.1MPa,温度为50℃。
在膜过滤后,可对滤液进行脱色和浓缩;采用的脱色剂为活性碳粉,脱色剂与滤液的质量配比可以为10:100,脱色的温度可控制在60℃,脱色时间为10min,脱色可在搅拌下进行。在脱色后,可通过过滤去除脱色剂。优选的,采用双效降膜蒸发器进行蒸发浓缩,并且可控制蒸发时的蒸汽压为0.05MPa,蒸发温度为50℃,经浓缩后,浓缩液的体积可降至原体积的1/4。
在浓缩后可进行灭菌和干燥,从而制得玉米低聚肽粉;所述干燥为喷雾干燥。

其中,所述杭白菊粉是以干燥杭白菊为原料,经粉碎、浸泡、水煮、过滤、浓缩、灭菌、干燥处理而制得,其总黄酮含量达5.0wt%以上;其中,所述粟米粉是以粟米为原料,经粉碎、酶解、过滤、浓缩、干燥处理而制得。
将上述原料混合均匀,经过杀菌处理后,真空包装得到产品。
实施例2
玉米蛋白粉与水的混合物在110℃以上的温度下加热,并将加热所得产物进行双螺杆挤压,得到预处理玉米蛋白粉;玉米蛋白粉与水的质量比为70:30,加热温度为160℃。将加热所得产物进行双螺杆挤压的步骤,其中,挤压机末端机筒温度为160℃,挤压机前三段机筒的温度可分别为40℃,60℃,80℃。
使用微生物蛋白酶酶解所述预处理玉米蛋白粉,灭酶后得到酶解液;所用的微生物蛋白酶的量为预处理玉米蛋白粉质量的1%;所用的微生物蛋白酶为碱性蛋白酶、和酸性蛋白酶;酶解温度为40℃。
将酶解液解液离心后,对离心上清液进行膜过滤,得到滤液;其中,所述离心的转速为4000r/min,离心可采用常规设备进行,例如卧螺离心机、管式离心机等。此外,可采用孔径为100nm的滤膜进行所述膜过滤;膜过滤时,可控制膜过滤的绝对压力为0.2MPa,温度为60℃。
在膜过滤后,可对滤液进行脱色和浓缩;采用的脱色剂为活性碳粉,脱色剂与滤液的质量配比可以为12:100,脱色的温度可控制在70℃,脱色时间为20min,脱色可在搅拌下进行。在脱色后,可通过过滤去除脱色剂。优选的,采用双效降膜蒸发器进行蒸发浓缩,并且可控制蒸发时的蒸汽压为0.06MPa,蒸发温度为60℃,经浓缩后,浓缩液的体积可降至原体积的1/3。
在浓缩后可进行灭菌和干燥,从而制得玉米低聚肽粉;优选的,所述干燥为喷雾干燥。
按照如下重量组分配制玉米低聚肽保健品原料:
其中,所述杭白菊粉是以干燥杭白菊为原料,经粉碎、浸泡、水煮、过滤、浓缩、灭菌、干燥处理而制得,其总黄酮含量达5.0wt%以上;其中,所述粟米粉是以粟米为原料,经粉碎、酶解、过滤、浓缩、干燥处理而制得。
将上述原料混合均匀,经过杀菌处理后,真空包装得到产品。
实施例3
玉米蛋白粉与水的混合物在110℃以上的温度下加热,并将加热所得产物进行双螺杆挤压,得到预处理玉米蛋白粉;玉米蛋白粉与水的质量比为60:40,加热温度为170℃。将加热所得产物进行双螺杆挤压的步骤,其中,挤压机末端机筒温度为170℃,挤压机前三段机筒的温度可分别为50℃,70℃,90℃。
使用微生物蛋白酶酶解所述预处理玉米蛋白粉,灭酶后得到酶解液;所用的微生物蛋白酶的量为预处理玉米蛋白粉质量的0.5-1.5%;所用的微生物蛋白酶中性蛋白酶;酶解温度为55℃。
将酶解液解液离心后,对离心上清液进行膜过滤,得到滤液;其中,所述离心的转速为5000r/min,离心可采用常规设备进行,例如卧螺离心机、管式离心机等。此外,可采用孔径为100nm的滤膜进行所述膜过滤;膜过滤时,可控制膜过滤的绝对压力为0.3MPa,温度为70℃。
在膜过滤后,可对滤液进行脱色和浓缩;采用的脱色剂为活性碳粉,脱色剂与滤液的质量配比可以为15:100,脱色的温度可控制在60-80℃,脱色时间为30min,脱色可在搅拌下进行。在脱色后,可通过过滤去除脱色剂。优选的,采用双效降膜蒸发器进行蒸发浓缩,并且可控制蒸发时的蒸汽压为0.08MPa,蒸发温度为70℃,经浓缩后,浓缩液的体积可降至原体积的1/2。
在浓缩后可进行灭菌和干燥,从而制得玉米低聚肽粉;优选的,所述干燥为喷雾干燥。
按照如下重量组分配制玉米低聚肽保健品原料:
其中,所述杭白菊粉是以干燥杭白菊为原料,经粉碎、浸泡、水煮、过滤、浓缩、灭菌、干燥处理而制得,其总黄酮含量达5.0wt%以上;其中,所述粟米粉是以粟米为原料,经粉碎、酶解、过滤、浓缩、干燥处理而制得。
将上述原料混合均匀,经过杀菌处理后,真空包装得到产品。
实施例产品性能测试:
将实施例1-3的产品为研究对象,对小鼠进行为期20天的喂养试验。参照卫生部《保健食品检验技术与评价规范》中“增强免疫力”功能评价方法,评价各组合物的增强免疫力功能。
1试验材料与主要仪器
1.1试验动物
SPF级昆明种雌性小鼠,6-8周龄,体重18-22g。
1.2主要仪器与试剂
Olympus光学显微镜,紫外分光光度计,Thermo酶联免疫检测仪,二氧化碳培养箱等。小牛血清,绵羊红细胞(SRBC)、氧化型辅酶I、琼脂糖。
2试验方法
2.1剂量选择与受试物给予方式
实验样品的推荐用量为每人每日5g,每人按65kg体重计,相当于0.08g/日/kg·BW。将小鼠随机分组,每组20只,组间体重无显著差异。将实施例1-3的组合物样品各设低、高两个剂量组,即各组小鼠每日剂量分别为0.4g/kg·BW、1.2g/kg·BW,另设一个阴性对照组(纯净水),各试验组均按上述剂量每天经口给予受试物1次(20mL/(kg·bw)灌胃容量),阴性对照组给予等体积蒸馏水。所有动物按剂量设计连续灌胃20天后,测定各项免疫指标。
2.2测定方法
小鼠抗体生成细胞测定、血清溶血素测定、腹腔巨噬细胞吞噬鸡红细胞实验等按照《保健食品检验与评价技术规范》(2003版)规定的方法进行。
2.3数据分析
用SPSS软件对实验数据进行统计学分析,组间比较采用t检验,p<0.05表示有显著性差异,p<0.01表示有极显著性差异。
3.结果与分析
表1实施例样品对小鼠抗体生成细胞数及血清溶血素水平的影响
由表1可知,实施例1-3各剂量组小鼠的抗体生成细胞数均高于阴性对照组,并且中、高剂量组与阴性对照组比较具有显著性差异(P<0.05)或极显著性差异(P<0.01),表明受试样品具有增加小鼠抗体生成细胞数的能力。小鼠血清溶血素实验中,实施例1-3高剂量组的抗体积数与对照组相比具有显著性差异(P<0.05),表明受试样品具有提高小鼠血清溶血素水平的作用。

由表2可知,与对照组比较,实施例1-3产品各剂量组对小鼠腹腔巨噬细胞吞噬鸡红细胞的能力均具有明显增加的趋势。
根据《保健食品检验与评价技术规范》中增强免疫力保健功能的判定标准:“细胞免疫功能、体液免疫功能、单核-巨噬细胞功能、NK细胞活性四方面任两个方面结果阳性,可判定该受试样品具有增强免疫力功能。”按照此判定规则,实施例1-3产品的体液免疫功能、单核-巨噬细胞功能两项试验结果为阳性,可以判定实施例1-3的产品具有增强免疫力的作用。

Claims (9)
1.一种玉米低聚肽保健品的制备方法,包括如下方法,包括如下步骤:
(1)对玉米蛋白粉进行物理预处理
玉米蛋白粉与水的混合物在110℃以上的温度下加热,并将加热所得产物进行双螺杆挤压,得到预处理玉米蛋白粉;
(2)酶解反应得到玉米低聚肽
(21)使用一种或多种微生物蛋白酶酶解所述预处理玉米蛋白粉,灭酶后得到酶解液;
(22)将酶解液解液离心后,对离心上清液进行膜过滤,得到滤液;
(23)在膜过滤后,对滤液进行脱色和浓缩;
(24)在浓缩后可进行灭菌和干燥,从而制得玉米低聚肽粉;所述干燥为喷雾干燥;
(3)配制玉米低聚肽保健品原料
按照如下重量组分进行配料:
上述玉米低聚肽粉 57-65份
余甘子粉 6-8份
杭白菊粉 3-7份
粟米粉 10-15份
紫薯粉 2-5份
葡萄糖 2-5份;
(4)将上述原料混合均匀,经过杀菌处理后,真空包装得到产品。
2.如权利要求1所述的方法,其特征在于,在所述步骤(1)中,玉米蛋白粉与水的质量比为80:20-60:40,加热温度为150-170℃。
3.如权利要求1或2所述的方法,其特征在于,在所述步骤(1)中,将加热所得产物进行双螺杆挤压的步骤,其中,挤压机末端机筒温度为150-170℃,挤压机前三段机筒的温度可分别为30-50℃,50-70℃,70-90℃。
4.如权利要求1-3任一所述的方法,其特征在于,在所述步骤(21)中,所用的微生物蛋白酶的量为预处理玉米蛋白粉质量的0.5-1.5%;所用的微生物蛋白酶为碱性蛋白酶、中性蛋白酶和酸性蛋白酶中的一种或多种;酶解温度为45-55℃。
5.如权利要求1-3任一所述的方法,其特征在于,在所述步骤(22)中,所述离心的转速为3000-5000r/min,离心可采用卧螺离心机、管式离心机,此外,可采用孔径为50-100nm的滤膜进行所述膜过滤;膜过滤时,可控制膜过滤的绝对压力为0.1-0.3MPa,温度为50-70℃。
6.如权利要求1-3任一所述的方法,其特征在于,在所述步骤(23)中,优选的,可以采用常规脱色剂进行脱色,脱色剂为活性碳粉,脱色剂与滤液的质量配比可以为(10-15):100,脱色的温度可控制在60-80℃,脱色时间为10-30min,脱色可在搅拌下进行,在脱色后,可通过过滤去除脱色剂。
7.如权利要求5所述的方法,其特征在于,采用双效降膜蒸发器进行蒸发浓缩,并且可控制蒸发时的蒸汽压为0.05-0.08MPa,蒸发温度为50-70℃,经浓缩后,浓缩液的体积可降至原体积的1/4-1/2。
8.如权利要求1-6任一所述的方法,其特征在于,所述杭白菊粉是以干燥杭白菊为原料,经粉碎、浸泡、水煮、过滤、浓缩、灭菌、干燥处理而制得,其总黄酮含量达5.0wt%以上。
9.如权利要求1-6任一所述的方法,其特征在于,所述粟米粉是以粟米为原料,经粉碎、酶解、过滤、浓缩、干燥处理而制得。
