Description

Sea grape extract with function of inhibiting allergy, preparation method and application
Technical Field
The invention relates to a sea grape extract, in particular to a sea grape extract capable of inhibiting allergy and not causing allergy, a preparation method and application thereof.
Background
Allergy is a phenomenon of excessive reaction to environmental substances (also called allergens) due to the abnormality of the immune system of the human body, and the allergens entering the body bind to IgE antibodies. At this time, the mast cells are stimulated to release inflammatory substances such as histamine, which causes inflammatory reaction of body tissues, and allergic symptoms vary depending on the body part releasing histamine, including symptoms of nasal discharge, lacrimation, itching, shortness of breath, dry skin, and the like, and serious patients may suffer from shock and thus be threatened. Allergy in people with allergic constitution is usually relieved by using anti-histamines and other drugs, and the allergic drugs generally have certain side effects, so that the inhibition of the allergic activity by using natural plants or extracting components with the function of inhibiting allergy from the plants is also an issue of public concern.
The ampelopsis belongs to green algae, is rich in algal polysaccharides, has zero fat and low calorie, has fresh and crisp mouthfeel, is the best natural health food in the minds of Japanese people and many experts like the caviar, and is also a high-grade food material in Japanese food shops. Since the sea grape contains a large amount of trace elements and has an anti-aging effect, the sea grape extract has been legally used as a cosmetic raw material by the world cosmetic society.
Most of the existing sea grape extraction technologies are extraction with alcohol or other organic solvents, taiwan patent I484966, which is an extraction method of sea grape extract, comprising the steps of performing crude extraction on sea grape powder with alcohol, and sequentially separating effective substances from the sea grape powder with n-hexane and ethyl acetate, wherein the effective substances have the effect of inhibiting cancers; korean patent KR101252149 uses water and C1-C4 alcohol to prepare a sea grape extract having antioxidant, anti-inflammatory, moisturizing, etc. effects. The extract of the sea grape in the prior art has no effect of inhibiting allergy, and the sea grape is extracted by an organic solvent, so that the sea grape can cause harm to human bodies even if a small amount of the organic solvent is remained.
The present invention relates to a method for extracting a vitis amurensis extract, and more particularly to a method for extracting a vitis amurensis extract from a vitis amurensis extract, which is an important subject to be solved by the present invention.

The following examples are provided by way of illustration only, and the present invention is not limited to the following examples.
Example 1
Preparation method of vitis amurensis extract (I)
Taking 80g of sea grape dry powder and 1600g of pure water, using sea grape as Caulerpa lentillifera, beating and mixing uniformly (the temperature is about 85 ℃, the time is about 90 minutes), and then filtering by using a filter screen with the size of 5 mu m and a filter screen with the size of 1 mu m to obtain liquid sea grape extract (I); and then performing membrane filtration, wherein the 1 st membrane is an EW membrane of GE (General Electric) company, the Microfiltration (Microfiltration) removes impurities with larger volume, the 2 nd membrane is a PW membrane of the GE company, the Ultrafiltration (Ultrafiltration) removes impurities with fine volume, the 3 rd membrane is a DK membrane of the GE company, the Nanofiltration (nanofiltation) removes salt, and the freeze drying is performed after the membrane filtration to obtain the sea grape refined extract (I). The above Caulerpa lentillifera can also be replaced by other strains of Styrax amurensis (Caulerpa taxifolia) such as Caulerpa yezoensis.
The extract (I) of Vitis amurensis does not cause allergy
Cells were observed for release of the allergic medium beta-hexosaminidase (hexosaminidase) without using a stimulant (A23187(-)) added only to the extract (I) of Vitis heyneana and the extract (I) of Vitis heyneana alone as shown in FIG. 1, and the release of beta-hexosaminidase was not increased by the extract (I) of Vitis heyneana and the extract (I) of Vitis heyneana alone as compared with the control group, indicating that the extract (I) of Vitis heyneana and the extract (I) of Vitis heyneana alone did not themselves provoke allergy.
Third, allergy Induction experiment
Calcium ionophore a23187 stimulates degranulation of cells to release the allergen mediator β -hexosaminidase (β hexosaminidase), and the β -hexosaminidase concentration was detected by enzyme-linked immunosorbent assay (ELISA), as shown in fig. 1, indicating that a23187 stimulation induces β -hexosaminidase release.
Sichuan grape extract (I) for suppressing allergy
According to the results of fig. 2, the vitis amurensis extract (I) inhibited the release of β -hexosaminidase induced by a23187 stimulation (group a23187(+), the release of β -hexosaminidase was reduced by 8% by 2 mg/ml for vitis amurensis extract (I), and the release of β -hexosaminidase was reduced by 25% by 4mg/ml for vitis amurensis extract (I); the inhibitory effect of the vitis amurensis extract (I) on the release of the beta-hexosaminidase is more obvious than that of the vitis amurensis extract, the release of the beta-hexosaminidase is inhibited by 45% at 1mg/ml of the vitis amurensis extract (I), the release of the beta-hexosaminidase is inhibited by 82% at 4mg/ml of the vitis amurensis extract (I), and the cell survival rate is not affected (as shown in fig. 1).
Application of Vitis amurensis extract (I) in cosmetics
Heating quantitative pure water to 50-60 ℃, sequentially adding the surfactant, and stirring and mixing uniformly. Then adding the emollient in sequence, stirring and mixing uniformly. Stopping heating, cooling to 30 deg.C, adding Vitis heyneana extract (I), fructus Citri sinensis essential oil, and sodium anisate, stirring, and mixing to obtain mousse shampoo containing Vitis heyneana extract (I), with the formula ratio shown in Table 1.
