Abstract

一种岩牡蛎人工催产的方法,其特征是首先筛选壳高15‑20 cm的岩牡蛎作为亲贝,进行亲贝蓄养,当水温上升至26‑27°C,岩牡蛎亲贝性腺指数发育至50‑60%时进行人工催产。岩牡蛎催产时,挑选性腺发育指数70%以上的长牡蛎雌性个体,制备不含卵子的长牡蛎卵巢组织滤出液,将长牡蛎卵巢组织滤出液均匀撒于岩牡蛎亲贝四周,30‑120 min后岩牡蛎亲贝即可产卵排精。使用本发明能有效诱导岩牡蛎产卵排精,不对岩牡蛎亲贝造成损伤,岩牡蛎催产率、受精率和孵化率均达90%以上,解决了岩牡蛎成熟精卵难以获取的育苗关键问题,能使岩牡蛎育苗达到稳产高产水平。

一种岩牡蛎人工催产的方法
技术领域
本发明涉及一种岩牡蛎人工催产的方法。
背景技术
岩牡蛎(Crassostrea nippona)隶属软体动物门、双壳纲、牡蛎科、巨蛎属,为广温性种类,自然分布于日本和韩国沿海,具有个体大、出肉率高以及对环境适应能力强等特点,是具有较高济价值的优良品种。岩牡蛎成体主要生活在潮下带至20 m深的浅海区,产卵期一般在8-9月,且有分批多次产卵排精的特点,因而使岩牡蛎在夏季体内糖原含量仍能维持较高水平,具有较好的口感风味,可弥补长牡蛎(Crassostrea gigas)夏季产卵后不能上市的市场空缺,因此又称“夏牡蛎”。在日本,从20世纪90年代初开始进行岩牡蛎人工育苗和养殖等相关研究,但并没有建立起规模化人工苗种生产技术,岩牡蛎产品仍以采捕野生群体为主,因而导致岩牡蛎自然资源量逐年下降,市场价格居高不下。
我国山东、辽宁等地浅海面积广阔,具备岩牡蛎生存的自然条件。引进岩牡蛎这一优良品种,在我国开展人工繁殖及增养殖,对于有效利用我国浅海资源,开发养殖新品种具有重要意义。
目前,岩牡蛎人工育苗技术还不成熟,关键问题是获取成熟精卵较为困难,人工授精无法开展。采用传统的解剖受精及物理化学催产方法不仅容易对亲贝造成损伤,并且受精率、孵化率以及幼虫存活率都较低,影响育苗成功率。
本发明通过从日本引进岩牡蛎成贝,通过研究岩牡蛎的繁殖习性,建立了一种岩牡蛎的人工催产方法,可以大规模获得岩牡蛎成熟的精子和卵子。
发明内容
本发明的目的是提供一种岩牡蛎人工催产的方法,可以大规模获得岩牡蛎成熟的精子和卵子。
一种岩牡蛎人工催产的方法,其特征是首先筛选壳高15-20 cm的岩牡蛎作为亲贝,在常温下进行亲贝蓄养,蓄养期间饵料投喂以小新月菱形藻(Nitzschia closterium f. minutissima)为主,辅助投喂亚心形扁藻(Platymonas subcordiformis)和小球藻(Chlorella vulgaris)。当水温上升至26-27°C,岩牡蛎亲贝性腺指数发育至50-60%时进行人工催产。岩牡蛎催产前,挑选性腺发育指数70%以上的长牡蛎雌性个体,解剖获取卵巢组织,制备不含卵子的长牡蛎卵巢组织滤出液。将长牡蛎卵巢组织滤出液均匀撒于岩牡蛎亲贝四周,30-120 min后岩牡蛎亲贝即可产卵排精。
本发明用于岩牡蛎催产的长牡蛎卵巢组织滤出液简单易得,不对岩牡蛎亲贝造成损伤,能有效诱导岩牡蛎产卵、排精,岩牡蛎催产率、受精率和孵化率均达90%以上,幼虫存活率高,解决了岩牡蛎成熟精卵难以获取的育苗关键问题,能使岩牡蛎育苗达到稳产高产水平。
具体实施方式
操作时,首先于5月筛选规格适宜的岩牡蛎亲贝,一般选择壳高为15-20 cm健康有活力的个体作为亲贝,在常温下按1个/m3的密度进行蓄养,亲贝蓄养期间饵料投喂以小新月菱形藻为主,辅助投喂亚心形扁藻和小球藻。7月下旬水温上升至26-27°C时,亲贝发育成熟,性腺发育指数达50-60%,准备人工催产。催产水温与亲贝蓄养水温保持一致。
选择性腺发育指数70%以上的长牡蛎个体,用解剖刀轻轻划破长牡蛎性腺组织,再用滴管吸取少量性腺组织成分置于滴有海水的载玻片上进行观察,若性腺组织呈颗粒状迅速散开则为雌性。将长牡蛎雌性个体卵巢置于孔径为25-30 μm的筛绢上,用拇指推压,使卵巢组织液流出。平均每100 g长牡蛎卵巢组织加500 mL海水进行过滤,获得不含卵子的长牡蛎卵巢组织滤出液。将长牡蛎卵巢组织滤出液均匀撒于岩牡蛎亲贝四周,30-120 min后岩牡蛎亲贝陆续在水泥池中产卵排精。经此法对岩牡蛎进行人工催产,可获得大量成熟精子和卵子,催产率、受精率和孵化率均达90%以上。岩牡蛎幼虫和稚贝的培育方法与长牡蛎相似。
本发明的方法以壳高、性腺发育指数为指标筛选亲贝,在水温26-27°C时采用长牡蛎卵巢组织滤出液进行诱导,可以有效地进行岩牡蛎的人工催产。
通过连续二年育苗试验,已经研究出岩牡蛎规模化人工育苗的技术方法,特别是岩牡蛎育苗中如何选择亲贝、亲贝培育的水温、诱导产卵的时机以及诱导产卵的方法,为岩牡蛎大规模苗种培育提供了保障。

1.一种岩牡蛎人工催产的方法,其特征是首先筛选壳高15-20 cm的岩牡蛎作为亲贝,进行亲贝蓄养,当水温上升至26-27°C,性腺指数发育至50-60%时,进行人工催产,使用性腺发育指数70%以上的长牡蛎雌性个体制备长牡蛎卵巢组织滤出液,将长牡蛎卵巢组织滤出液均匀撒于岩牡蛎亲贝四周,30-120 min后岩牡蛎亲贝陆续产卵排精。
