本发明公开了一种黄精提取物的制备方法,包括以下步骤:取黄精干粉,加入用于细胞破碎的破碎物和蒸馏水,混合均匀,其中,黄精干粉与蒸馏水的料液比为1g:40‑50ml,黄精干粉与破碎物的质量比为1g:5‑8mg,破碎物包括纤维素酶、果胶酶、CTAB;将混合后的液体置于60‑70℃水浴中5‑6h,水浴结束后,对该混合后的液体加热至沸腾并持续10‑15min,而后加入活性炭,静置冷却后离心,收集上清液;在上清液中加入5‑6倍体积的无水乙醇,静置6‑8h,离心取沉淀,对该沉淀进行干燥,获得黄精提取物。本发明还公开一种上述制备方法制备的黄精提取物及提取物的应用。本发明的黄精提取物具有抗氧化能力。

技术领域
本发明涉及生物医药技术领域,特别是涉及一种黄精提取物及其制备方法和应用。
背景技术
黄精为百合科多年生草本植物,是一种药食同源植物,富含淀粉、维生素、矿物质和多种活性物质。黄精的主要活性成分有糖类、甾体皂苷、黄酮、氨基酸及微量元素等。研究表明,黄精中的物质具有抗氧化的作用,保护人体免受自由基的损伤。
氧化可造成机体自由基泛滥,自由基又称游离基,是具有非偶电子的基团或原子,由于自由基含未配对的电子,所以极不稳定,会从邻近的分子夺取电子,让自己处于稳定的状态,这样邻近的分子又变成一个新的自由基,然后再去夺取电子,该连锁反应会使细胞结构受到破坏,造成细胞功能丧失、基因突变、甚至死亡。抗氧化就是抑制自由基的氧化反应,其作用机理可以是直接作用在自由基,也可以是间接消耗掉容易生成自由基的物质。
如何提供一种可抗氧化的纯天然物质,是抗氧化领域亟待解决的问题之一。
发明内容
本发明主要解决的技术问题是提供一种黄精提取物及其制备方法和应用,可制备出纯天然的黄精提取物,该提取物具有抗氧化功能。
为解决上述技术问题,本发明提供一种黄精提取物的制备方法,包括以下步骤:将干燥的黄精粉碎至70-90目,获得黄精干粉;取黄精干粉,加入用于细胞破碎的破碎物和蒸馏水,混合均匀,其中,黄精干粉与蒸馏水的料液比为1g:40-50ml,黄精干粉与破碎物的质量比为1g:5-8mg,破碎物包括纤维素酶、果胶酶、CTAB;将混合后的液体置于60-70℃水浴中5-6h,水浴结束后,对该混合后的液体加热至沸腾并持续10-15min,而后加入活性炭,静置冷却后离心,收集上清液;在上清液中加入5-6倍体积的无水乙醇,静置6-8h,离心取沉淀,对该沉淀进行干燥,获得黄精提取物。

为解决上述技术问题,本发明提供一种黄精提取物作为抗氧化药物或保健品的应用。具体地,黄精提取物作为抗氧化药物或保健品的剂型为片剂、胶囊剂、颗粒剂或散剂。

实施例1
黄精提取物的制备方法包括以下步骤:将干燥的黄精粉碎至80目,获得黄精干粉,其中,粉碎的黄精为根茎部分;取黄精干粉,加入用于细胞破碎的破碎物和蒸馏水,混合均匀,其中,黄精干粉与蒸馏水的料液比为1g:45ml,黄精干粉与破碎物的质量比为1g:7mg,破碎物包括纤维素酶、果胶酶、CTAB(十六烷基三甲基溴化铵);将混合后的液体置于65℃水浴中5.5h,水浴结束后,对该混合后的液体加热至沸腾并持续13min,而后加入活性炭,静置冷却后离心,收集上清液;在上清液中加入5.5倍体积的无水乙醇,静置7h,离心取沉淀,对该沉淀进行干燥,获得黄精提取物。

实施例4
在实施例2的基础上,利用蒸馏水对获得的黄精提取物进行充分溶解,而后进行Sephadex G-200凝胶柱层析,利用蒸馏水进行洗脱,蒸馏水的洗脱量为2BV,收集洗脱液,在洗脱液中加入5倍体积的无水乙醇进行醇析,静置5h后,离心取沉淀,获得纯化的黄精提取物。
实施例5
黄精提取物片剂的制备:将实施例1或实施例2制备的黄精提取物用单冲压片机打成直径为7mm,重量为350mg的片,即得黄精提取物片剂。
实施例6
黄精提取物胶囊剂的制备:将实施例1或实施例2制备的黄精提取物以350mg填充入1号肠溶胶囊,即得黄精提取物胶囊剂。
实施例7
黄精提取物颗粒剂的制备:将实施例1或实施例2制备的黄精提取物加水制成软材,过15目筛进行造粒,干燥后即得黄精提取物颗粒剂。
实施例8
黄精提取物散剂的制备:将实施例1或实施例2制备的黄精提取物过250目筛,即得黄精提取物散剂。
下面通过实验说明本发明黄精提取物的抗氧化能力。
取实施例1制备的黄精提取物放入锥形瓶中,设置6个重复,在每个锥形瓶中加入新鲜猪油,其中,黄精提取物与猪油的质量比为1:20;取实施例2制备的黄精提取物放入锥形瓶中,设置6个重复,在每个锥形瓶中加入新鲜猪油,其中,黄精提取物与猪油的质量比为1:20;各锥形瓶在70℃下搅拌30min,使加入物充分混合,塞好瓶塞,置于65℃恒温箱中,每隔12h搅拌5min,每3天取样测定油脂过氧化值;其中,油脂过氧化值的测定按照GB/T5009.37-2003食用植物油卫生标准的分析方法进行。
经实施例1制备的黄精提取物处理的猪油的过氧化值为:3天,4.02meq/kg;6天,4.46meq/kg;9天,5.15meq/kg。其中,各过氧化值为各重复的平均值。
经实施例2制备的黄精提取物处理的猪油的过氧化值为:3天,4.09meq/kg;6天,4.51meq/kg;9天,5.17meq/kg。其中,各过氧化值为各重复的平均值。
根据上述实验结果,可以看出实施例1或实施例2制备的黄精提取物表现出强的抗氧化能力,抗氧化效果好,且抗氧化能力持久。
利用上述实验方法计算黄精多糖的抗氧化能力,经黄精多糖处理的猪油的过氧化值为:3天,4.98meq/kg;6天,7.21meq/kg;9天,9.35meq/kg。其中,各过氧化值为各重复的平均值。根据计算结果,可以看出实施例1或实施例2制备的黄精提取物较黄精多糖具有更好的抗氧化能力。
利用上述实验方法测定实施例3和实施例4制备的黄精提取物的抗氧化能力,经实施例3制备的黄精提取物处理的猪油的过氧化值为:3天,4.00meq/kg;6天,4.38meq/kg;9天,5.06meq/kg。经实施例4制备的黄精提取物处理的猪油的过氧化值为:3天,4.01meq/kg;6天,4.40meq/kg;9天,5.09meq/kg。其中,各过氧化值为各重复的平均值。根据测定结果,可以看出实施例3、实施例4制备的黄精提取物较实施例1、实施例2制备的黄精提取物具有更好的抗氧化能力。
综上所述,本发明的黄精提取物具有强的抗氧化能力,且其为纯天然物质,对人体无副作用。

取所述黄精干粉,加入用于细胞破碎的破碎物和蒸馏水,混合均匀,其中,所述黄精干粉与蒸馏水的料液比为1g:40-50ml,所述黄精干粉与破碎物的质量比为1g:5-8mg,所述破碎物包括纤维素酶、果胶酶、CTAB;
将混合后的液体置于60-70℃水浴中5-6h,水浴结束后,对该混合后的液体加热至沸腾并持续10-15min,而后加入活性炭,静置冷却后离心,收集上清液;
在所述上清液中加入5-6倍体积的无水乙醇,静置6-8h,离心取沉淀,对该沉淀进行干燥,获得黄精提取物。
2.根据权利要求1所述的黄精提取物的制备方法,其特征在于,所述破碎物按质量百分比计,包括组分:65%-80%纤维素酶、20%-35%果胶酶、5%-10%CTAB。

6.根据权利要求5所述的黄精提取物的制备方法,其特征在于,所述上清液与加入其中的无水乙醇的体积比为1:5.5。
7.根据权利要求2所述的黄精提取物的制备方法,其特征在于,利用蒸馏水对所述黄精提取物进行溶解,而后进行凝胶柱层析,利用蒸馏水进行洗脱,蒸馏水的洗脱量为1.5-2.5BV,收集洗脱液,在洗脱液中加入4.5-5.5倍体积的无水乙醇进行醇析,静置4-6h后,离心取沉淀,获得纯化的黄精提取物。
8.权利要求1-7任意一项所述的制备方法制备得到的黄精提取物。
9.权利要求8所述的黄精提取物作为抗氧化药物或保健品的应用。
10.根据权利要求9所述的应用,其特征在于,所述黄精提取物作为抗氧化药物或保健品的剂型为片剂、胶囊剂、颗粒剂或散剂。
