Description

Method for increasing germination rate of Calligonum mongolicum seeds by more than 40%
Technical Field
The invention relates to a method for improving the germination rate of seeds of an psammophyte, in particular to a method for improving the germination rate of seeds of an psammophyte, namely, calligonum mongolicum.
Background
"Sha Hui Zao" (fructus Hoveniae)Calligonum mongolicum) Belonging to genus Polygonaceae (Polygonaceae) Calligonum (Polygonaceae) ((Polygonaceae))Calligonum) The plant is a drought or super-drought plant widely distributed in desert and gobi in northern arid regions (semiarid regions) in China, is also one of important vegetation for vegetation in desert in middle Asia and middle Asia, is distributed in Asia, southern Europe and northern Africa, can grow in extremely harsh arid desert regions, has the characteristics of wind erosion resistance, sand burying resistance, cutting resistance, drought resistance, barrenness resistance and the like, and is an excellent sand-fixing pioneer plant. Therefore, the Calligonum mongolicum has important research significance on the treatment and improvement of drought desertification. In addition, the tender branches can be eaten by livestock, and have higher economic value; the fruits are beautiful, are a beautiful landscape in desert, and have higher ornamental value. Seeds can be adopted for seeding and seedling raising in the breeding of the Calligonum mongolicum, but because the seed shells of the Calligonum mongolicum are hard, the germination rate of the seeds is extremely low by adopting the common soaking treatment with clear water, even the seeds hardly germinate at all, at present, methods such as seed coat damage pretreatment, 98% concentrated sulfuric acid soaking and the like are also adopted, but the germination rate does not reach 10%, and obviously how to greatly improve the germination rate of the Calligonum mongolicum seeds is an unsolved technical problem at present. Therefore, it is necessary to provide a method for improving the germination rate of the Calligonum mongolicum seeds.
Chinese patent 2014103746964 discloses a method for atriplex canescens seed germination, which comprises the following steps: soaking the atriplex canescens seeds by using concentrated sulfuric acid, cleaning the seeds after the seed soaking treatment, and placing the seeds on a vermiculite matrix for germination treatment. The technique can effectively improve the germination rate of the atriplex canescens seeds, and the data disclosed in the patent shows that the germination rate of the atriplex canescens seeds basically can reach more than 70%. Another Chinese patent 2014103784839 discloses a method for increasing germination rate of atriplex canescens seeds, which comprises soaking seeds in sodium hypochlorite solution and/or gibberellin solution, and germinating on vermiculite matrix after the seed soaking treatment. However, similar methods for seed soaking have not been satisfactory in these technical suggestions, and it is difficult to achieve satisfactory germination rates, as will be described later in the text.
Obviously, how to greatly improve the germination rate of the Calligonum mongolicum seeds is an unsolved technical problem at present.
Disclosure of Invention
The invention provides a method which can overcome the defects of the prior art and can greatly improve the germination rate of Calligonum mongolicum seeds, in particular to a method which can improve the germination rate of Calligonum mongolicum seeds to more than 40 percent.
The method for improving the germination rate of the Calligonum mongolicum seeds to be more than 40% comprises the steps of immersing the Calligonum mongolicum seeds into a concentrated sulfuric acid solution with the volume ratio of 70% -75% for full immersion, taking out the immersed seeds, kneading, cleaning, uniformly dibbling the seeds into a culture medium fully soaked by water, wherein the used culture medium is vermiculite with the particle size of 1-3 mm.
The preferable method for improving the germination rate of the Calligonum mongolicum seeds is to soak the seeds for 6-10 hours by using concentrated sulfuric acid, and tests show that the optimal time for soaking the seeds by the concentrated sulfuric acid is 6 hours.
If the seeds which are soaked in concentrated sulfuric acid, taken out and cleaned are soaked in water for 12-24 hours, the effect is better, and the seed germination time is obviously advanced by 2-3 days.
The method can greatly improve the germination rate of Calligonum mongolicum seeds, and has positive significance for large-area planting of Calligonum mongolicum and sand control and fixation.
