Background
Haze is a collective term for fog and haze, which is caused by the accumulation of a large number of artificially emitted pollutants under specific meteorological conditions. High concentrations of fine particulate matter (particulate matter having a particle size of 2.5 microns or less, abbreviated as PM2.5) or aerosol contamination are the root causes of haze.
The PM2.5 fine particles have small aerodynamic particle size and large specific surface area, are easy to carry a large amount of toxic and harmful substances such as industrial waste gas, volatile chemical substances, motor vehicle tail gas and the like, pass through the filtration effect of nose hair, enter the deep lung tissue of a human body through a respiratory tract and circulate blood, and cause serious harm to the health of the human body. Atmospheric particulates entering the respiratory tract damage the respiratory tract defense barrier by stimulating and corroding the alveolar wall, so that the pulmonary function is damaged, the incidence of pneumonia, chronic bronchitis, chronic obstructive pulmonary disease, bronchial asthma and the like is increased, and the respiratory system symptoms such as cough, expectoration, wheeze and the like appear in patients. Epidemiological statistics show that: with the increase of atmospheric particulate pollution concentration, the incidence of respiratory symptoms, pulmonary hypofunction, the morbidity and mortality of cardiopulmonary diseases are all obviously increased, and the two are directly related, and PM2.5 becomes an important influence factor of the continuous increase of the morbidity of a respiratory system.
In addition, many problems caused by smoking have become a problem of great social attention in recent years, and according to the investigation of the world health organization, one quarter of cancer patients account for 90% of smoking in industrially developed countries; dead from bronchitis, 75% of smoking; and most people are suffering from the harm of second-hand smoke, so that the method has great significance on reducing and lightening the harm to human beings caused by haze and improving the health of smokers.

2. Evaluation of rat respiratory function:
the lung function of a conscious rat is measured by adopting an EMKA animal lung function monitoring system, the EMKA animal lung function monitoring system is placed in a quiet laboratory, the system is corrected, calibrated and ventilated before formal measurement, the data acquisition frequency is automatically acquired once every 5 seconds, and the measurement result is displayed once every minute. The rats are placed in a whole body scanning meter box to adapt for 5-10min, the recording time is 3-8min, and the average value of the results of each animal is taken as the measurement result of the lung function of the animal. The measured indices include inspiration time (Ti), expiration time (Te), Tidal Volume (TV), Expiration Volume (EV), Relaxation Time (RT), and Minute Ventilation (MV).
The respiratory function of the animals was measured every 2 weeks during the exposure period of the rats, and the rats were placed in a whole plethysmograph box and connected to a respiratory function measuring instrument for measurement, and the results are shown in Table 2.
Table 2: continuous inhalation of infected SD rat respiratory function determination:
and (3) analyzing test results:
the Ti value of the inhaled toxicant of the model group rats is obviously higher than that of the control group (p <0.05), and the RT value of the toxicant SD rats is obviously higher than that of the control group (p <0.05), which indicates that when the lung function indexes related to the flow and the volume of the model group rats are obviously inhibited, the animals can maintain the relatively required lung function by prolonging the expiration time and the relaxation time. The rats TV, EV and MV in the model group all decreased to different degrees during the observation period, indicating that the volume of breath was significantly decreased and the lung ventilation function reserve was also decreased. Compared with the model group, the experimental group 1-6 can obviously improve and improve the lung function damage of the rat caused by continuous fine particulate matter inhalation, and has certain improvement effect. Second, the study of the technology
1. Screening an extraction mode: in the process of technological research, in order to utilize the active ingredients of each medicament to the maximum extent, modes such as water decoction extraction, ethanol reflux extraction, ethanol percolation extraction and the like are respectively investigated, and the results show that the high-temperature water extraction mode is adopted, so that the extraction efficiency of the active ingredients of the medicinal materials in the formula is high, the contents of houttuynin, amygdalin and the like are high, the dissolved substances are more, the water decoction mode is more suitable for the production of beverages, the influence of non-drinking solvents on the safety of the products is avoided, and the method is more economic. The factors such as production time, production cost and the like in the large-scale production are integrated, and the extraction temperature and the extraction time are further investigated.
The results of the examination by adopting 5 temperature gradients (70-80 ℃, 80-90 ℃, 90-95 ℃, 95-100 ℃ and 90-100 ℃) and 4 extraction time periods (30-60min, 60-120min, 90-180min and 30-120min) show that when the extraction is carried out at the constant temperature of 90-100 ℃ for 30-120min, the extract can enrich the active ingredients of the medicine, and the detection shows that the crude drug with the houttuynine sodium bisulfite content of more than or equal to 0.04 percent, the crude drug with the triolein content of more than or equal to 0.65 percent, the crude drug with the amygdalin content of more than or equal to 7.2 percent and the crude drug with the total extract of more than or equal to 50 percent can exert the maximum efficacy. Wherein, the effect is best when the temperature is kept for soaking for 60-120min and the constant temperature is 95-100 ℃ before extraction, the content of active ingredients is ideal, and the extraction time can be shortened to 50-70 min. Due to the dual functions of soaking in advance and high-temperature extraction, the extraction time is effectively shortened, and the method is very beneficial to mass production.
2. Subsequent process study: in the research process, the temperature in the process is continuously kept to be 70-90 ℃ in the subsequent auxiliary material adding, constant volume and filling stages, so that the inconvenience of prolonging the process time, increasing the production cost and the like caused by temperature rise in subsequent high-temperature sterilization is avoided, and more importantly, the operation of continuously keeping higher temperature in the whole production link can avoid introducing infection of foreign bacteria, so that the safety of the product is ensured.
3. Raw material crushing granularity screening: the crushing or crushing degree of raw medicinal materials has great influence on the dissolution of active ingredients during extraction, the study finds that the almond and the peach kernel contain oily ingredients, the dissolution of the active ingredients is not facilitated due to overlarge crushing particle size, the adhesion and influence of the almond and the peach kernel on medicinal materials such as reed rhizome, platycodon root and the like and the self are increased in the water extraction process due to partial exudation of the oily substances caused by crushing due to over fine crushing, and the crushing degree of the medicinal materials such as houttuynia cordata and the like also considers the crushing particle size of the almond and the like to balance the two so as to facilitate the extraction.
In conclusion, the preparation process obtained by optimization is very simple and easy to operate, and can save the production cost and improve the production efficiency. And the plant extraction efficiency is high, the active ingredients of the medicine can be enriched, and the maximum medicine effect can be exerted.
The specific implementation mode is as follows:
