白藜芦醇作为疫苗佐剂的应用
技术领域
本发明涉及白藜芦醇作为疫苗佐剂的应用。
背景技术
佐剂是一种能够增强对疫苗组分抗原特异性免疫应答或改变免疫反应的物质。在疫苗制剂中,佐剂的功能主要包括:增强疫苗抗原的免疫原性;促进细胞免疫和体液免疫,优化免疫应答,促进免疫能力较弱人群中的免疫应答;增进抗原与黏膜之间的传递以及免疫接触;减少疫苗成分中抗原的需求量以及在实施过程中的免疫接种次数;优化抗原结构,维持抗原构象等。佐剂的种类丰富,根据其功能,可分为运载传递系统和免疫刺激性佐剂。前者包括运载体即能与半抗原结合的一类具有免疫原性质的物质,还包括对整个疫苗系统起运输传递和衬托作用的一类物质。根据佐剂的来源,可分为细菌性、非细菌性、微生物性、细胞因子类以及合成体类等。根据理化性质,可分为颗粒性、凝胶性、乳胶佐剂等。跟据其性能,可分为治疗性佐剂、免疫刺激性佐剂、黏膜佐剂以及基因佐剂等。
白藜芦醇,又称为芪三酚,是一种生物性很强的天然多酚类物质,主要来源于花生、葡萄(红葡萄酒)、虎杖、桑椹等植物。目前发现该物质的功效主要有:降低血液粘稠度,抑制血小板凝结和血管舒张,保持血液畅通,预防癌症的发生及发展,抗动脉粥样硬化和冠心病,缺血性心脏病,防治高血脂等作用。
发明内容
本发明的目的一方面是拓展白藜芦醇的新的应用领域,另一方面是提供一种更高效的疫苗佐剂,增强对兔魏氏梭菌病疫苗抗原特异性免疫应答。
为了实现上述目的,本发明采用了以下技术方案:

与现有技术相比较,本发明具备的有益效果:
将白藜芦醇作为兔魏氏梭菌病疫苗佐剂能够显著提高免疫效率降低死亡率,并且安全、有效、不会引起毒副作用。
具体实施方式
下面通过实施例对本发明的技术方案作进一步阐述。
实施例1
(1)简介
魏氏梭菌病本病又称产气荚膜杆菌病。魏氏梭菌性肠炎主要由A型和E型菌及产生的α毒素所致。它广泛存在于土壤、饲料、蔬菜、污水、粪便中。本病的发生无明显季节性,除哺乳仔兔外,各种年龄、品种、性别的兔子均有易感性。传染途径主要是消化道或伤口,粪便污染的病原在传播方面起主要作用。本病的主要传染原是病兔和带菌兔及排泄物。病原菌自消化道或伤口侵入机体,在小肠和盲肠绒毛膜上大量繁殖并产生强烈的α毒素,改变毛细血管的通透性,使毒素大量进入血液,引起全身性毒血症。
正确的饲养可减少本病的发生;采用较低能量、较高纤维素的日粮可明显减少腹泻死亡率。一旦发现病兔或可疑为病兔,应立即隔离或淘汰。预防方法可用魏氏梭菌氢氧化铝灭活菌苗,每只兔皮下注射2毫升,7天后开始产生免疫力,免疫期4~6个月。本病的治疗,可内服土霉素、四环素,均有一定的疗效。另外用高免血清治疗本病效果也较好。其方法是发现患病兔泻痢后,视病兔大小,每只皮下注射高免血清5~10毫升,每天1次,连用2~3天即可康复。
(2)免疫试验
将36只生长健康状态良好的平均体重1.2千克的中国白兔随机分配成三组:对照组、疫苗组和疫苗+白藜芦醇组;对照组每只白兔皮下注射2毫升生理盐水;疫苗组每只白兔皮下注射2毫升市售兔用魏氏梭菌灭活菌苗;疫苗+白藜芦醇组每只白兔皮下注射2毫升市售兔用魏氏梭菌灭活菌苗和内含白藜芦醇230μmoL的混合物。第一次注射后第8天再次进行一次注射,第一次注射后第20天,采用A型魏氏梭菌攻毒,剂量为420cfu/只,观察20天,统计白兔的死亡情况,并计算相对免疫保护率,结果如表1所示。
表1试验结果
表1中相对免疫保护率一是与对照组相比的计算结果,二是与疫苗组相比的计算结果,从表1可以看出,试验白兔接受市售兔用魏氏梭菌灭活菌苗免疫后,与对照组相比,白兔死亡率有所下降。而试验白兔接受疫苗+白藜芦醇共同免疫后,比疫苗组的相对免疫保护率提高了57.1%,说明白藜芦醇能够提高市售兔用魏氏梭菌灭活菌苗的保护效率。
