Background
Noni (Noni), also known as morinda citrifolia, FOLIUM JAPONICA, and the like, belongs to the genus Morinda of the family Rubiaceae. The plant is mainly distributed in islands of the south pacific, such as the philippines, australia, etc. At present, China is mainly distributed in heat zone areas such as Hainan area, Xisha islands and the like, and large-scale planting is carried out. Noni has strong functional characteristics, and roots, stems, leaves, flowers and fruits can be used as medicines. Yellow or red dye is obtained from root for dyeing clothes, and the fruit is edible and used as food-grade health product. Noni has been used as a food or pharmaceutical product for over 2000 years by the boysenia in the islands of the south pacific ocean. Noni has attracted the attention of food and drug researchers as a new medicinal plant in the world, and about 200 compounds mainly comprising anthraquinone, flavone, coumarin, glucoside, triterpenes and the like are separated and identified from noni, wherein the effective active ingredients of the noni are scopolia, rutin, ursolic acid and tannin. Furthermore, noni is rich in unsaturated fatty acids, vitamins, minerals, antioxidants, rare elements and proxeronine, Xeronine converting enzyme, etc. Noni has stronger antioxidant function, and the antioxidant capacity of noni is stronger than that of common antioxidants, including vitamin C, grape seed powder, pycnogenol and the like. Currently, the research on the biological activity of noni mainly focuses on the antibacterial, tumor prevention, free radical removal, oxidation resistance and other functional characteristics. The antioxidant effect of the noni is higher than that of a large number of antioxidants, the noni is even higher than that of grape seeds by 30 percent, the noni can effectively eliminate free radicals and enhance the function of the immune system, and amino acids, vitamins, trace elements and sugar esters in the noni can regulate the immune system of the organism, resist diseases and prevent infection. Because of its unique functional properties, noni is now one of the best-selling health products worldwide, the demand for noni in the market is increasing at a rate of 50% per year, and the government is also increasing the development and development of noni. Through retrieval, the application of the noni extract emulsion in whitening cosmetics or skin care products is not reported.

TABLE 1
Figure BDA0001106433840000051
The noni fruit extract emulsion components obtained in examples 1 to 4 show that the ethanol content in the emulsion is significantly increased when the stirring temperature is lower than the boiling point of ethanol (78.3 ℃), the ethanol content in the emulsion is significantly decreased when the stirring temperature is higher than the boiling point of ethanol, and the extraction rate of the noni fruit raw material in example 4 is higher. The emulsion components of the noni fruit extracts obtained in examples 4 to 6 and comparative example 1 show that the content of the noni fruit extract in the emulsion increases with the addition amount of the noni fruit raw material.
ABTS free radical scavenging Capacity assay
ABTS was used in this experiment to determine the free radical clearance. Preparing a mixed solution of ABTS with the concentration of 7mmol/L and potassium persulfate with the concentration of 2.45mmol/L, standing for 10-16 h at room temperature in a dark place, and diluting with water to ensure that the light absorption value of the mixed solution at 734nm is 0.7 +/-0.02 (marked as A)0) Adding 1mL of the samples of examples 1 to 8 and comparative example 1 and 1.5mL of ABTS working solution into an ABTS working solution test tube; the blank group uses distilled water to replace the sample solution; the control group used distilled water instead of ABTS working solution. The above groups were left at room temperature in the dark for 1h, and the absorbance (denoted as A) was measured at 734nm, with ascorbic acid as a positive control.
Clearance (%) ═ a0-A)/A0*100%。
The results of determination of the clearance in the ABTS radical scavenging ability assay of the emulsions obtained in examples 1 to 8 and comparative example 1 are shown in table 3:
TABLE 3
Figure BDA0001106433840000061
As can be seen from the graphs, the emulsions obtained in examples 1 to 8 have better radical scavenging ability, and particularly, the emulsion in example 4 has the best radical scavenging ability when the mass fraction of the morinda citrifolia extract in the emulsion is 10% and the mass fraction of the surfactant is 8%. It is shown that the emulsions obtained in examples 1 to 8 have a protective effect on whitening and skin-caring ingredients in noni fruits, so that they are not easily oxidized.
Whitening Activity test
The potato extract contains tyrosinase, wherein the tyrosinase is an oxidoreductase, the tyrosinase can promote L-tyrosine to generate melanin by oxidation, and in the process of forming the melanin by the tyrosine, the tyrosinase can promote L-tyrosine to generate L-dopa and further act to enable the L-dopa to form bakinone, and the dopinone finally generates the melanin.
The calculation formula of the inhibition rate is as follows: [1- (T2-T1)/(C2-C1) ] 100%
C1 is the absorbance value of the mixture of the buffer solution and the potato extract, C2 is the absorbance value of the mixture of the buffer solution, the potato extract and tyrosine, and C2, measured with C1 as a reference, is C2-C1, which represents the absorbance value of the melanin produced after subtracting the absorbance of the potato extract. T1 is the absorbance value of the mixture of the emulsion sample, the potato extract and the buffer solution, T2 is the absorbance value of the mixture of the emulsion sample, the buffer solution, the potato extract and tyrosine, and T2-T1 was obtained by measuring the absorbance value of T2 with reference to T1, which represents the absorbance value of melanin after the inhibition of the sample, with the influence of the absorbance of the sample and the potato extract being deducted. The results of the inhibition rate of the emulsions obtained in examples 1 to 8 and comparative example 1 in the whitening activity test are shown in table 2:
TABLE 2
Figure BDA0001106433840000062
The results of the above test on the inhibition rate show that the emulsions obtained in examples 1 to 8 all have a high inhibition rate and can be used for skin whitening, and particularly, in example 4, when the mass fraction of the noni fruit extract in the emulsion is 10% and the mass fraction of the surfactant is 8%, the whitening effect of the emulsion is the best. The inhibition ratios of example 4, example 7 and example 8 show that the compounding ratio of the surfactant is also important for the whitening effect of the emulsion, and when the content is 8%, the protective effect on the components having the whitening and skin-care effects in noni fruits is better. Meanwhile, the inhibition ratio of comparative example 1 was significantly reduced, indicating that the whitening effect was better not as the content of the morinda citrifolia extract was more.
In conclusion, the noni extract emulsion provided by the invention has remarkable free radical scavenging capacity and whitening and skin-care effects, the preparation method is simple and easy to operate, the requirements on equipment and instruments are not high, and the production cost is low. The noni extract emulsion provided by the invention can be directly applied to skin care and whitening, and can also be applied to preparation of other whitening cosmetics or skin care products.
The above description is only for the purpose of illustrating the preferred embodiments of the present invention and is not to be construed as limiting the invention, and any modifications, equivalents, improvements and the like that fall within the spirit and principle of the present invention are intended to be included therein.
