Abstract
本发明涉及声调控型抗肿瘤药物递送系统的制备方法及应用,有效解决现有肿瘤治疗药物载药量低,递送过程会出现药物泄露、肿瘤响应灵敏度低、释放慢且易产生耐药性、无法进行体外调控的问题,以中空介孔二氧化钛为基体,内部荷载全氟己烷及药物博来霉素,通过内源性生物膜‑红细胞膜包裹,并以肿瘤靶向多肽进行修饰,构建超声调控型抗肿瘤药物递送系统;以中空介孔二氧化钛为基体,内部荷载全氟己烷及模型药物博来霉素,将RBC包裹在中空介孔二氧化钛的表面,通过内源性生物膜‑红细胞膜包裹,制备出纳米级粒径分布的递药系统,并以肿瘤靶向多肽NGR末端进行二酰基脂质修饰,构建具有远程超声调控功能的肿瘤递药体系。

技术领域
本发明属于医药化工领域,特别是一种声调控型抗肿瘤药物递送系统的制备方法及应用。
背景技术
目前,化疗药物通常难溶于水、副作用大、易产生耐药。开发靶向控释纳米递药系统可在一定程度解决上述问题。近年来,各国科学家在纳米控释递药体系领域进行了多方面的研究,如肿瘤响应型pH控释系统、近红外光控释系统等,虽然这些纳米递药系统可实现药物在肿瘤部位的响应释放,但亦存在一些问题:如载药量低,递送过程会出现药物泄露、肿瘤响应灵敏度低、释放慢且易产生耐药性、无法进行体外调控等。如何将药物高效转运至肿瘤组织、使药物在非肿瘤部位零释放,在肿瘤组织或细胞中快速释放,寻找可调控型智能递药系统是克服上述缺点重要途径之一。
超声波对生物组织有较强的穿透力,聚焦超声可无创伤地将声能聚焦于深层组织,无需通过内镜,操作简单等优点,因而超声激活敏感剂产生的声动力学疗法被认为是一种有潜力、极具推广前景的抗癌新疗法。自1987年Fechheimer等报到了超声通过增加细胞的通透性来增强基因的表达以来,超声在增强目的基因表达及药物递送领域取得了长足的发展。超声脂质微泡作为药物的载体,可以以多种方式携带药物或基因,将载药微泡经外周静脉注射后,可随血流到达靶组织,待其到达靶区域后,增强声压,引爆微泡,使微泡在靶区定位爆破并局部释放药物,从而增强药物递送或基因的转染。
有关研究发现,TiO2纳米结构是一种相当安全的材料,无明显的细胞毒作用,而且TiO2粒子能够被机体清除并被排除体外,对于作为药物转运载体而言具有重要的实际应用意义。中空介孔二氧化钛(HPTiO2)是一种新型的TiO2纳米结构,具有比表面积大、药物负载量高、毒性低、化学稳定性好等优点,Zhang等人将NH3▪H2O和TiCl4采用低温沉积法制备出内部中空,外层由小TiO2粒子堆积而成的HPTiO2。相比其他TiO2纳米结构,HPTiO2具有超大的比表面积,可高效装载多种抗肿瘤药物(紫杉醇、阿霉素、鱼藤酮等),中空和孔道中均可荷载药物,拥有高药物荷载量。
红细胞(Red Blood Cells,RBCs)是血液中数量最多且寿命最长的细胞。由于正常成熟红细胞无核、无细胞器,红细胞膜提取分离较简单,一般通过离心方法得到血液中的红细胞,采用低渗溶血方式获得红细胞膜。Luk等研究了纳米粒内核的表面电性、曲率半径等因素对红细胞膜包裹纳米粒的影响。结果显示,红细胞膜可包封粒径为65~340nm的纳米粒,其中静电作用是能包裹并保持稳定的主要原因。红细胞膜包裹的纳米粒(RBC-NP)作为药物载体具有以下优点:
(1)红细胞膜用于纳米粒修饰可保护并转运药物。RBC-NP可形成一个隔离空间以保护运载的物质,避免药物提前释放,使药物不受内源性因素的影响而过早失活和降解,提高药物在体内的稳定性;其具有的纳米级粒径可在循环中保持稳定,并可通过EPR效应,通过肿瘤血管渗漏到肿瘤部位;(2)与普通药物载体相比,免疫原性低,体内能够逃避免疫细胞的吞噬及识别,显著延长药物的体内循环时间;(3)它具有良好的形变、跨膜和融合能力,易于跨过生物屏障,有利于细胞摄取。作为内源性物质,具有高度的生物安全性;通过EPR效应到达肿瘤部位后,提高对肿瘤细胞的识别和亲和能力可显著延长递药系统在靶部位的滞留时间,促进肿瘤细胞对递药系统的内吞作用,实现药物精准、高效的转运至肿瘤细胞。CD13受体在乳腺癌MCF-7细胞膜过表达,使其成为MCF-7肿瘤细胞识别的有效靶点。NGR(可与CD13受体特异性结合)靶向是一种公认的、高效的肿瘤细胞靶向策略。基于此,对递药系统进行NGR修饰,将使该体系在体内具有高度的肿瘤细胞靶向性,可显著降低化疗药物的毒性,提高其疗效。
目前,博来霉素(BLM)是临床上使用较为广泛的抗肿瘤药物之一,它的药理作用主要是嵌入细胞核内DNA而抑制核酸合成。研究表明,肿瘤细胞内铜离子浓度较高,BLM可与肿瘤细胞内铜离子结合,进而降低超氧化物歧化酶活性(SOD),从而减少对ROS的清除,有利于声动力学产生的ROS在肿瘤细胞内的积聚,发挥协同抗肿瘤作用。
发明内容
针对上述问题,为解决现有技术之缺陷,本发明的目的就是提供一种声调控型抗肿瘤药物递送系统的制备方法及应用,可有效解决现有肿瘤治疗药物载药量低,递送过程会出现药物泄露、肿瘤响应灵敏度低、释放慢且易产生耐药性、无法进行体外调控的问题。
本发明的技术方案是:以中空介孔二氧化钛为基体,内部荷载全氟己烷(PFH)及药物博来霉素,通过内源性生物膜-红细胞膜包裹,并以肿瘤靶向多肽进行修饰,构建超声调控型抗肿瘤药物递送系统;
具体包括下列步骤:
(1)制备中空介孔二氧化钛(HPTiO2):
将0.1mol/LNH3▪H2O 5-100ml溶于350-450ml丙酮中,得NH3▪H2O丙酮溶液,再将5-11mLTiCl4的丙酮溶液滴加到上述溶液中,室温下反应25-35min,3000-4000rpm离心8-12min,35-45℃下干燥除去丙酮,再加入过量的甲醇溶液,使反应产物中的NH4Cl结晶析出分离,除去产物中的NH4Cl 晶体,35-45℃抽真空干燥24h除去甲醇,即得中空介孔二氧化钛;
(2)制备红细胞膜(RBC):
取2-5ml雌性大鼠血液加入40-100μL肝素,4℃下3500rpm离心8-12min,收集沉淀红细胞,用1mol/LpH7.4的PBS缓冲液5-30ml洗涤后,置于5-15ml 0.1-0.3mol/LpH7.4的PBS缓冲液中,冰浴2h,9000rpm离心10min,去除血红蛋白,重悬于0.1-0.3mol/LpH7.4的PBS中,过滤,得红细胞膜;
(3)制备红细胞膜包裹的二氧化钛/博来霉素/全氟己烷(RBC/HPTiO2/BLM/PFH):
称取10-20mg中空介孔二氧化钛,加入到2-40ml去离子水中,探头超声溶解,加入5-10mg博来霉素及20-40μl全氟己烷,室温搅拌12-24h,除去游离药物,得二氧化钛/博来霉素/全氟己烷;将步骤(2)所得的红细胞膜与二氧化钛/博来霉素/全氟己烷按体积比0.5-4:1混合均匀,过滤,得红细胞膜包裹的二氧化钛/博来霉素/全氟己烷;
(4)制备多肽修饰递药系统(NGR-RBC/HPTiO2/BLM/PFH):
取0.1-1.0mg的NGR-diacyllipid加入到2-10ml的红细胞膜包裹的二氧化钛/博来霉素/全氟己烷中,在37℃摇床中孵育4-8h后,8000rpm离心8-12min,得超声调控型抗肿瘤药物递送系统。
本发明所述方法制备的超声调控型抗肿瘤药物递送系统在制备肿瘤部位靶向给药、肿瘤部位的超声远程调控定位释药、超声响应自供氧声动力学治疗体系药物、肿瘤的多机制治疗药物以及肿瘤超声成像诊断药物中的应用。
本发明以中空介孔二氧化钛(HPTiO2)为基体,内部荷载可通过超声实现相变的全氟己烷(PFH)及模型药物博来霉素(BLM),采用挤压法,将RBC包裹在中空介孔二氧化钛的表面,通过内源性生物膜-红细胞膜包裹,制备出了具有较均一的纳米级粒径分布(180nm左右)的递药系统。并以肿瘤靶向多肽NGR末端进行二酰基脂质(diacyllipid)修饰,利用diacyllipid和RBC的强亲和力实现NGR的连接,构建具有远程超声调控功能的肿瘤递药体系(NGR-RBC/HPTiO2/BLM/PFH)。采用CD13受体高表达的乳腺癌MCF-7位细胞模型,该系统通过超声调控实现药物在靶部位的有效定位释药及肿瘤细胞的有效摄取;实现递药系统在到达肿瘤前药物零释放或少释放,显著降低对正常组织的副作用,到达肿瘤部位后,超声远程调控使得药物爆发性释放,实现对肿瘤的有效杀伤。其次,该系统具有自供氧(全氟己烷携氧功能)特征,可高效补充超声诱导二氧化钛产生活性氧所需的氧气,增强声动力学(SDT)效果;同时,实现了药物博来霉素和声动力学治疗在细胞内同一位点(细胞核)多机制协同治疗肿瘤的目的。并且制备工艺简单、节能环保、易于工业化生产,经济和社会效益显著。
具体实施方式
以下结合实施例对本发明的具体实施方式作详细说明。
本发明在具体实施中,是由以下实施例实现。
实施例1
本发明所述的超声调控型抗肿瘤药物递送系统,具体包括以下步骤:
(1)制备中空介孔二氧化钛
将0.1mol/LNH3▪H2O 5-100ml溶于400ml丙酮,将8mL TiCl4的丙酮溶液滴加到上述溶液中,室温下反应30min,4000rpm离心12min,40℃干燥除去丙酮,再加入过量的甲醇溶液,使反应产物中的NH4Cl结晶析出分离,除去产物中的NH4Cl晶体,40℃抽真空干燥24h除去甲醇,即得中空介孔二氧化钛;
(2)制备红细胞膜:
取5ml雌性大鼠血液加入100μL肝素,4℃下3500rpm离心10min,收集沉淀红细胞,用1mol/LpH7.4的PBS缓冲液10ml洗涤后,置于10ml 0.25mol/LpH7.4的PBS缓冲液中,冰浴2h,9000rpm离心10min,去除血红蛋白,重悬于0.25mol/LpH7.4的PBS中,过滤,得红细胞膜;
(3)制备红细胞膜包裹的二氧化钛/博来霉素/全氟己烷:
称取15mg中空介孔二氧化钛,加入到20ml去离子水中,探头超声溶解,加入8mg博来霉素及30μl全氟己烷,室温搅拌12h,除去游离药物,得二氧化钛/博来霉素/全氟己烷;将步骤(2)所得的红细胞膜与二氧化钛/博来霉素/全氟己烷按体积比2:1混合均匀,过滤,得红细胞膜包裹的二氧化钛/博来霉素/全氟己烷;
(4)制备多肽修饰递药系统:
取0.5mg的NGR-diacyllipid加入到8ml的红细胞膜包裹的二氧化钛/博来霉素/全氟己烷中,在37℃摇床中孵育6h后,8000rpm离心10min,得超声调控型抗肿瘤药物递送系统。
实施例2
本发明所述的超声调控型抗肿瘤药物递送系统,具体包括以下步骤:
(1)制备中空介孔二氧化钛:
将0.1mol/LNH3▪H2O 5ml溶于350ml丙酮中,得NH3▪H2O丙酮溶液,得NH3▪H2O丙酮溶液,再将5mLTiCl4的丙酮溶液滴加到上述溶液中,室温下反应25min,3000rpm离心8min,35℃下干燥除去丙酮,再加入过量的甲醇溶液,使反应产物中的NH4Cl结晶析出分离,除去产物中的NH4Cl 晶体,35℃抽真空干燥24h除去甲醇,即得中空介孔二氧化钛;
(2)制备红细胞膜:
取2ml雌性大鼠血液加入40μL肝素,4℃下3500rpm离心8min,收集沉淀红细胞,用1mol/LpH7.4的PBS缓冲液5ml洗涤后,置于5ml 0.1mol/LpH7.4的PBS缓冲液中,冰浴2h,9000rpm离心10min,去除血红蛋白,重悬于0.1mol/LpH7.4的PBS中,过滤,得红细胞膜;
(3)制备红细胞膜包裹的二氧化钛/博来霉素/全氟己烷:
称取10mg中空介孔二氧化钛,加入到2ml去离子水中,探头超声溶解,加入5mg博来霉素及20μl全氟己烷,室温搅拌24h,除去游离药物,得二氧化钛/博来霉素/全氟己烷;将步骤(2)所得的红细胞膜与二氧化钛/博来霉素/全氟己烷按体积比0.5:1混合均匀,过滤,得红细胞膜包裹的二氧化钛/博来霉素/全氟己烷;
(4)制备多肽修饰递药系统:
取0.1mg的NGR-diacyllipid加入到2ml的红细胞膜包裹的二氧化钛/博来霉素/全氟己烷中,在37℃摇床中孵育4h后,8000rpm离心8min,得超声调控型抗肿瘤药物递送系统。
实施例3
本发明所述的超声调控型抗肿瘤药物递送系统,具体包括以下步骤:
(1)制备中空介孔二氧化钛:
将0.1mol/LNH3▪H2O 100ml溶于450ml丙酮中,得NH3▪H2O丙酮溶液,得NH3▪H2O丙酮溶液,再将11mLTiCl4的丙酮溶液滴加到上述溶液中,室温下反应35min,4000rpm离心12min,45℃下干燥除去丙酮,再加入过量的甲醇溶液,使反应产物中的NH4Cl结晶析出分离,除去产物中的NH4Cl 晶体,45℃抽真空干燥24h除去甲醇,即得中空介孔二氧化钛;
(2)制备红细胞膜:
取5ml雌性大鼠血液加入100μL肝素,4℃下3500rpm离心12min,收集沉淀红细胞,用1mol/L pH7.4的PBS缓冲液30ml洗涤后,置于15ml 0.3mol/LpH7.4的PBS缓冲液中,冰浴2h,9000rpm离心10min,去除血红蛋白,重悬于0.3mol/LpH7.4的PBS中,过滤,得红细胞膜;
(3)制备红细胞膜包裹的二氧化钛/博来霉素/全氟己烷:
称取20mg中空介孔二氧化钛,加入到40ml去离子水中,探头超声溶解,加入10mg博来霉素及40μl全氟己烷,室温搅拌18h,除去游离药物,得二氧化钛/博来霉素/全氟己烷;将步骤(2)所得的红细胞膜与二氧化钛/博来霉素/全氟己烷按体积比4:1混合均匀,过滤,得红细胞膜包裹的二氧化钛/博来霉素/全氟己烷;
(4)制备多肽修饰递药系统:
取1.0mg的NGR-diacyllipid加入到10ml的红细胞膜包裹的二氧化钛/博来霉素/全氟己烷中,在37℃摇床中孵育8h后,8000rpm离心12min,得超声调控型抗肿瘤药物递送系统。
本发明一种超声调控型抗肿瘤药物递送系统,以中空介孔二氧化钛(HPTiO2)为基体,内部荷载可通过超声实现相变的全氟己烷(PFH)及模型药物博来霉素(BLM),通过内源性生物膜-红细胞膜包裹,并以肿瘤靶向多肽NGR进行修饰,构建具有远程超声诱导控释体系功能的肿瘤递药系统(NGR-RBC/HPTiO2/BLM/PFH),实现递药系统在递送过程中药物零释放,超声远程引爆药物在肿瘤部位的定点快速释放;构建超声响应自供氧声动力学治疗体系,实现肿瘤化疗与声动力学多机制协同治疗肿瘤的目的;构建具有超声分子成像功能的造影剂,提高肿瘤治疗的敏感性和特异性以及对早期癌症诊断的准确性。
本发明采用的技术方案是通过低温沉积法制备出外壳由小TiO2粒子堆积而成的多孔HPTiO2,它的外壳和其他多孔TiO2纳米结构的致密外壳不同,低温堆积作用形成的外壳,在正常情况下是稳定的,当有外界能量激发时,这些小粒子之间原有的作用力被打破,导致HPTiO2的结构坍塌,崩解为小TiO2粒子。将抗肿瘤药物及相变材料装载于其中空多孔结构中,再通过挤压法实现红细胞膜在HPTiO2表面的包裹,实现体内长循环和门控目的,最终将对肿瘤细胞具有特异识别能力的靶头(NGR)连接到红细胞膜上,提高载体对肿瘤细胞的靶向识别能力,采用超声远程调控药物释放,实现化疗和声动力学治疗在细胞内同一位点多机制协同治疗肿瘤的目的。
本发明所述的超声调控型抗肿瘤药物精准递送系统,利用低温沉积法制备出外壳由小TiO2粒子堆积而成的多孔HPTiO2,通过挤压法实现红细胞膜在HPTiO2表面的包裹。
所述的HPTiO2具有均一的粒径分布,带负电位(-18mv),在超声作用下结构坍塌,可分解为4-8nm的小纳米粒,同时在超声照射下能够激发ROS生成。
所述的RBC/HPTiO2/BLM/PFH粒径比较均一,在180nm左右,用低频超声治疗仪(UGT型,1.0W/cm2)作用10s后,粒径变为微米级(~2.2μm),可实现全氟己烷诱发的纳米粒向微粒的转变。
将NGR的末端进行二酰基脂质(diacyllipid)修饰(公司合成),得到NGR-diacyllipid衍生物,利用diacyllipid 能够高效插入磷脂双分子层的性质实现NGR和RBC/HPTiO2/BLM/PFH表面红细胞膜的高效连接。
相关实验资料如下:
实验一、对本发明所述方法制得的中空介孔二氧化钛进行透射电镜形态观察、Zetasizer Nano S90激光粒度分析仪测定其电位。合成的HPTiO2粒径约为80-100nm,电位为-18mv,可被红细胞膜包裹。
实验二、超声导致HPTiO2结构坍塌考察:取一定量HPTiO2分散于适量水中,置于六孔板内,将含有HPTiO2的小孔底部涂上耦合剂,用低频超声治疗仪(UGT 型),以1.0W/cm2的声强作用10s,透射电镜观察形态,超声作用后,HPTiO2结构坍塌,分解为4-8nm小纳米粒。
实验三、超声激发HPTiO2产生生ROS的能力:将HPTiO2与MCF-7细胞共孵育4h后,在低频超声治疗仪(UGT型,1.0W/cm2)照射30s后,使用活性氧试剂盒检测细胞内活性氧的生成量,用流式细胞仪检测,HPTiO2在超声照射下活性氧产生具有浓度依赖性,浓度为10和20μg/ml时,活性氧产生量分别26.8%和41.2%,与空白组相比具有显著性差异。
实验四、红细胞膜包封的全氟己烷(PFH)在超声作用下相变:将提取得到的红细胞膜,加入一定量的PBS缓冲液振荡,然后在冰浴中用高速分散均质机上分散(8000rpm,5min),同时按 PFH与溶液体积比1:5的比例逐滴加入 PFH,之后用声振仪细胞破碎仪乳化(80% power,60s),最后将其装入离心管,进行3次离心(2000rpm,5min)和PBS洗涤,以去除未包封的红细胞膜,最后加入PBS轻柔混匀,得到红细胞膜包封的全氟己烷。用低频超声治疗仪(UGT 型,1.0W/cm2)作用10s后,使用Zetasizer Nano S90激光粒度分析仪测定超声前后粒径变化。结果表明,超声可诱导PFH相变,实现纳米粒(~180nm)向微泡(~2.2μm)的转变。
实验五、NGR-RBC/HPTiO2/BLM/PFH递药系统载药量的测定:取一定量NGR-RBC/HPTiO2/BLM/PFH递药系统溶于适量甲醇中,使用细胞破碎仪对其探头超声处理一定时间,确保RBC破裂以及HPTiO2彻底崩散,高速离心,上清样品通过HPLC检测确定NGR-RBC/HPTiO2/BLM/PFH的装载量。结果显示药物负载率可达到68%。
实验六、超声作用下NGR-RBC/HPTiO2/BLM/PFH药物释放:将NGR-RBC/HPTiO2/BLM/PFH置于透析袋内(截留分子量MW=3500Da)中,浸入磷酸盐PBS缓冲液中,100r/min,37℃条件下震荡,每隔一定时间取出部分,采用HPLC法测定BLM,测定其浓度,并计算其释放速度。设置超声对照组。对释药数据进行拟合,探讨其释药机制。结果显示超声后药物累计释放百分率提高了60%。
实验七、NGR-RBC/HPTiO2/BLM/PFH递药系统体外抗肿瘤效果:时间效应(有/无超声照射):用NGR-RBC/HPTiO2/BLM/PFH对人乳腺癌MCF-7肿瘤细胞进行一次处理,并同时用超声照射一定时间后,在不同时间点考察其对人乳腺癌MCF-7肿瘤细胞生长的抑制作用(SRB法或其他方法测定);并考察超声照射前后细胞内活性氧水平的变化,彗星实验观察其对细胞DNA的损伤;剂量效应(有/无超声):用不同剂量NGR-RBC/HPTiO2/BLM/PFH处理细胞,超声照射一定时间,考察其对肿瘤细胞生长的抑制作用(SRB法或其他方法测定);并考察超声照射前后细胞内活性氧水平的变化,彗星实验观察其对细胞DNA的损伤;
以上实验均设不同实验组:RBC/HPTiO2、NGR-RBC/HPTiO2、BLM、RBC/HPTiO2/BLM/PFH、NGR-RBC/HPTiO2/BLM/PFH等。
结果证明NGR-RBC/HPTiO2/BLM/PFH递药系统在超声照射下能更好更快地发挥抗肿瘤效果,更明显地抑制人乳腺癌MCF-7肿瘤细胞的增殖。
实验八、NGR-RBC/HPTiO2/BLM/PFH递药系统体内分布研究:将肿瘤细胞接种到裸鼠肋腹的皮下。待肿瘤长到一定大小时,每只小鼠静脉注射NGR-RBC/HPTiO2/BLM/PFH。取不同时间小鼠的组织,称重匀浆后提取各组织中的药物,HPLC测定各个组织中的药物浓度,定量考察NGR-RBC/HPTiO2/BLM/PFH在荷瘤小鼠组织内的分布。设不同实验组:BLM、RBC/HPTiO2/BLM/PFH、NGR-RBC/HPTiO2/BLM/PFH。结果证明NGR-RBC/HPTiO2/BLM/PFH组药物在肿瘤部位分布的最多,说明NGR可介导递药系统实现肿瘤靶向递送。
实验九、NGR-RBC/HPTiO2/BLM/PFH递药系统体内抗肿瘤效果:将符合要求的荷瘤裸鼠随机分为以下五组并开始处理(静脉注射):BLM、RBC/HPTiO2、NGR-RBC/HPTiO2、RBC/HPTiO2/BLM/PFH、NGR-RBC/HPTiO2/BLM/PFH,同时设生理盐水对照组和阳性对照组。给药期间连续测定肿瘤体积,并使用PHILIPIU 22多功能彩色多普勒超声诊断仪监测肿瘤情况,直到动物处死为止。出死后取出肿瘤,称重。按照相对肿瘤增值率T/C评价效果。设超声实验组。
结果证明NGR-RBC/HPTiO2/BLM/PFH递药系统在超声照射下,小鼠的肿瘤体积的增加得到了明显的抑制。
实验十、NGR-RBC/HPTiO2/BLM/PFH递药系统超声成像:将肿瘤细胞接种到裸鼠肋腹的皮下。待肿瘤长到一定大小时,每只小鼠静脉注射NGR-RBC/HPTiO2/BLM/PFH。采用PHILIPIU 22多功能彩色多普勒超声诊断仪,12L-5线阵探头,MI=0.6,Depth3.5cm,在灰阶产生模式下扫查肿瘤部位并截图。用DFY软件分析视频强度,记录递药体系到达肿瘤部位的时间。分组为:RBC/HPTiO2、NGR-RBC/HPTiO2、RBC/HPTiO2/BLM/PFH、NGR-RBC/HPTiO2/BLM/PFH。结果证明NGR-RBC/HPTiO2/BLM/PFH在肿瘤部位的超声信号最强。
本发明与现有技术相比具有以下突出的有益技术效果:
本发明制备的超声调控型抗肿瘤药物精准递送系统具有远程调控药物释放特征,超声可远程引爆NGR-RBC/HPTiO2/BLM/PFH,实现药物的有效定位释放及肿瘤细胞对药物的高效摄取,具有远程调控、深部治疗、无损伤性特征。
超声相变功能,全氟己烷可在超声远程调控下实现递药系统从纳米粒向微泡的相变,一方面实现纳米粒的功能-药物的有效递送,另一方面实现超声诱导微泡破裂-定点快速释放药物;
所构建的递药系统为自供氧、超声响应型动力学治疗体系,当TiO2纳米粒在超声作用下产生活性氧,消耗氧气而使氧气不足时,能自供给弥补之,从而高效产生活性氧,与BLM在作用机制上协同治疗肿瘤。
所述递药系统主要具有以下特点:1) 该系统可实现递药系统在递送过程中药物零释放,超声远程引爆药物在肿瘤部位的定点快速释放;2) 超声响应自供氧声动力学治疗体系,实现肿瘤化疗与声动力学多机制协同治疗肿瘤的目的;3) 具有超声分子成像功能,可提高肿瘤治疗的敏感性和特异性以及对早期癌症诊断的准确性。
Claims (5)
1.一种超声调控型抗肿瘤药物递送系统的制备方法,其特征在于,以中空介孔二氧化钛为基体,内部荷载全氟己烷及药物博来霉素,通过内源性生物膜-红细胞膜包裹,并以肿瘤靶向多肽进行修饰,构建超声调控型抗肿瘤药物递送系统;
具体包括下列步骤:
(1)制备中空介孔二氧化钛:
将0.1mol/LNH3▪H2O 5-100ml溶于350-450ml丙酮中,得NH3▪H2O丙酮溶液,再将5-11mLTiCl4的丙酮溶液滴加到上述溶液中,室温下反应25-35min,3000-4000rpm离心8-12min,35-45℃下干燥除去丙酮,再加入过量的甲醇溶液,使反应产物中的NH4Cl结晶析出分离,除去产物中的NH4Cl 晶体,35-45℃抽真空干燥24h除去甲醇,即得中空介孔二氧化钛;
(2)制备红细胞膜:
取2-5ml雌性大鼠血液加入40-100μl肝素,4℃下3500rpm离心8-12min,收集沉淀红细胞,
用1mol/LpH7.4的PBS缓冲液5-30ml洗涤后,置于5-15ml 0.1-0.3mol/LpH7.4的PBS缓冲液中,冰浴2h,9000rpm离心10min,去除血红蛋白,重悬于0.1-0.3mol/LpH7.4的PBS中,过滤,得红细胞膜;
(3)制备红细胞膜包裹的二氧化钛/博来霉素/全氟己烷:
称取10-20mg中空介孔二氧化钛,加入到2-40ml去离子水中,探头超声溶解,加入5-10mg博来霉素及20-40μl全氟己烷,室温搅拌12-24h,除去游离药物,得二氧化钛/博来霉素/全氟己烷;将步骤(2)所得的红细胞膜与二氧化钛/博来霉素/全氟己烷按体积比0.5-4:1混合均匀,过滤,得红细胞膜包裹的二氧化钛/博来霉素/全氟己烷;
(4)制备多肽修饰递药系统:
取0.1-1.0mg的NGR-diacyllipid加入到2-10ml的红细胞膜包裹的二氧化钛/博来霉素/全氟己烷中,在37℃摇床中孵育4-8h后,8000rpm离心8-12min,得超声调控型抗肿瘤药物递送系统。
2.根据权利要求1所述的超声调控型抗肿瘤药物递送系统,其特征在于,具体包括以下步骤:
(1)制备中空介孔二氧化钛
将0.1mol/LNH3▪H2O 5-100ml溶于400ml丙酮,将8mL TiCl4的丙酮溶液滴加到上述溶液中,室温下反应30min,4000rpm离心12min,40℃干燥除去丙酮,再加入过量的甲醇溶液,使反应产物中的NH4Cl结晶析出分离,除去产物中的NH4Cl晶体,40℃抽真空干燥24h除去甲醇,即得中空介孔二氧化钛;
(2)制备红细胞膜:
取5ml雌性大鼠血液加入100μl肝素,4℃下3500rpm离心10min,收集沉淀红细胞,用1mol/LpH7.4的PBS缓冲液10ml洗涤后,置于10ml 0.25mol/LpH7.4的PBS缓冲液中,冰浴2h,9000rpm离心10min,去除血红蛋白,重悬于0.25mol/LpH7.4的PBS中,过滤,得红细胞膜;
(3)制备红细胞膜包裹的二氧化钛/博来霉素/全氟己烷:
称取15mg中空介孔二氧化钛,加入到20ml去离子水中,探头超声溶解,加入8mg博来霉素及30μl全氟己烷,室温搅拌12h,除去游离药物,得二氧化钛/博来霉素/全氟己烷;将步骤(2)所得的红细胞膜与二氧化钛/博来霉素/全氟己烷按体积比2:1混合均匀,过滤,得红细胞膜包裹的二氧化钛/博来霉素/全氟己烷;
(4)制备多肽修饰递药系统:
取0.5mg的NGR-diacyllipid加入到8ml的红细胞膜包裹的二氧化钛/博来霉素/全氟己烷中,在37℃摇床中孵育6h后,8000rpm离心10min,得超声调控型抗肿瘤药物递送系统。
3.根据权利要求1所述的超声调控型抗肿瘤药物递送系统,其特征在于,具体包括以下步骤:
(1)制备中空介孔二氧化钛:
将0.1mol/LNH3▪H2O 5ml溶于350ml丙酮中,得NH3▪H2O丙酮溶液,得NH3▪H2O丙酮溶液,再将5mLTiCl4的丙酮溶液滴加到上述溶液中,室温下反应25min,3000rpm离心8min,35℃下干燥除去丙酮,再加入过量的甲醇溶液,使反应产物中的NH4Cl结晶析出分离,除去产物中的NH4Cl 晶体,35℃抽真空干燥24h除去甲醇,即得中空介孔二氧化钛;
(2)制备红细胞膜:
取2ml雌性大鼠血液加入40μl肝素,4℃下3500rpm离心8min,收集沉淀红细胞,用1mol/LpH7.4的PBS缓冲液5ml洗涤后,置于5ml 0.1mol/LpH7.4的PBS缓冲液中,冰浴2h,9000rpm离心10min,去除血红蛋白,重悬于0.1mol/LpH7.4的PBS中,过滤,得红细胞膜;
(3)制备红细胞膜包裹的二氧化钛/博来霉素/全氟己烷:
称取10mg中空介孔二氧化钛,加入到2ml去离子水中,探头超声溶解,加入5mg博来霉素及20μl全氟己烷,室温搅拌24h,除去游离药物,得二氧化钛/博来霉素/全氟己烷;将步骤(2)所得的红细胞膜与二氧化钛/博来霉素/全氟己烷按体积比0.5:1混合均匀,过滤,得红细胞膜包裹的二氧化钛/博来霉素/全氟己烷;
(4)制备多肽修饰递药系统:
取0.1mg的NGR-diacyllipid加入到2ml的红细胞膜包裹的二氧化钛/博来霉素/全氟己烷中,在37℃摇床中孵育4h后,8000rpm离心8min,得超声调控型抗肿瘤药物递送系统。
4.根据权利要求1所述的超声调控型抗肿瘤药物递送系统,其特征在于,具体包括以下步骤:
(1)制备中空介孔二氧化钛:
将0.1mol/LNH3▪H2O 100ml溶于450ml丙酮中,得NH3▪H2O丙酮溶液,得NH3▪H2O丙酮溶液,再将11mLTiCl4的丙酮溶液滴加到上述溶液中,室温下反应35min,4000rpm离心12min,45℃下干燥除去丙酮,再加入过量的甲醇溶液,使反应产物中的NH4Cl结晶析出分离,除去产物中的NH4Cl 晶体,45℃抽真空干燥24h除去甲醇,即得中空介孔二氧化钛;
(2)制备红细胞膜:
取5ml雌性大鼠血液加入100μl肝素,4℃下3500rpm离心12min,收集沉淀红细胞,用1mol/L pH7.4的PBS缓冲液30ml洗涤后,置于15ml 0.3mol/LpH7.4的PBS缓冲液中,冰浴2h,9000rpm离心10min,去除血红蛋白,重悬于0.3mol/LpH7.4的PBS中,过滤,得红细胞膜;
(3)制备红细胞膜包裹的二氧化钛/博来霉素/全氟己烷:
称取20mg中空介孔二氧化钛,加入到40ml去离子水中,探头超声溶解,加入10mg博来霉素及40μl全氟己烷,室温搅拌18h,除去游离药物,得二氧化钛/博来霉素/全氟己烷;将步骤(2)所得的红细胞膜与二氧化钛/博来霉素/全氟己烷按体积比4:1混合均匀,过滤,得红细胞膜包裹的二氧化钛/博来霉素/全氟己烷;
(4)制备多肽修饰递药系统:
取1.0mg的NGR-diacyllipid加入到10ml的红细胞膜包裹的二氧化钛/博来霉素/全氟己烷中,在37℃摇床中孵育8h后,8000rpm离心12min,得超声调控型抗肿瘤药物递送系统。
5.权利要求1或2-4任一项制备的超声调控型抗肿瘤药物递送系统在制备肿瘤部位靶向给药、肿瘤部位的超声远程调控定位释药、超声响应自供氧声动力学治疗体系药物、肿瘤的多机制治疗药物以及肿瘤超声成像诊断药物中的应用。
