背景技术
百香果含有17 种氨基酸,丰富的蛋白质、脂肪、糖、维生素、钙、磷、铁、钾、SOD 酶和超纤维等165 种对人体有益物质,更被称为水果中的VC 之王,口感独特。百香果有消炎止痛、活血强身、降脂降压、滋阴补肾、提神醒酒、消除疲劳、护肤养颜等神奇功效,制作成饮料,冷热饮皆宜,口味极佳,常饮还有消除青春痘、消除痔疮、消除肠胃炎。蚯蚓含有丰富的氨基酸、酶类、维生素和活性物质,不但是传统的药材,可用以开发出多种保健食品、化妆品;还是家禽养殖、淡水鱼养殖饲料的动物性蛋白质的重要来源。特别是由于蚯蚓能以工业、农业及城乡生活有机废弃物为食,并将其转化为优质的抗菌性有机肥料蚯蚓粪便,因此对于环保及发展有机农业具有重要的经济意义和现实意义,用蚯蚓来处理生活垃圾和秸杆等废弃物,减少焚烧产生的烟雾,也是发展低碳经济的有力措施。随着城市化进程的加快,农用耕地面积逐年减少,土地资源日渐紧缺,如何能在土地上套养套种,以提高土地的使用率和经济效益,也是一个急待解决的重要课题。
百香果,又名西番莲,因其果肉可散发出香蕉、菠萝、草莓、荔枝、柠檬、芒果、酸梅等十几种水果的浓郁香味而被誉为“百香果”,百香果富含人体所需的十七种氨基酸、多种维生素和类胡萝卜素以及各种微量元素,不仅营养丰富,而且味道鲜美。百香果适应性广泛,在中国的南方,一年四季均适宜种植,在北方,冬季适宜采用蔬菜大棚种植。

百香果属于雌雄异花,雌花结果,雄花用于授粉,其授粉媒介主要是蜜蜂和昆虫,也可通过人工授粉。春季种植的百香果,当年7 月即可开花,9 月可收果。第二年以后,每年4 月开始开花,一年可开5-6 批,每批花相隔20 天,从开花到果实成熟需60-70 天,果实收获4 个多月。
百香果的繁殖方法主要有两种,一种是用种子直接播种繁殖,另一种则是用蔓条扦插繁殖。每亩种植70-80 株主蔓,主蔓逐级分出侧蔓在棚架上攀延结果。百香果每年每亩产量达1-1.5 吨,高产的达2 吨以上。
百香果产于南美洲属多年生常绿攀缘性藤本热带水果,因其果汁具有番石榴、芒果、香蕉等多种水果的香气而得名。百香果适应性强、生长旺盛,抗病虫,投产早,产量高,品质好,耐贮运、销路广,经济效益高,在国外有“摇钱树”之美称。百香果具有多种水果的浓郁香味,和很高的营养价值,不仅可作鲜果吃和制果汁饮料,同时也是用加工果酱,果冻,冰淇淋,糕点,饼干最理想的加工用果。百香果果汁作为一种纯天然“绿色”果汁饮料以其色香味俱佳,以及良好的保健效果,老少皆宜,正风摩世界。
百香果(Passiflora edulis Sims),学名西番莲,西番莲科, 西番莲属,为热带、亚热带多年生常绿藤本浆果类果树。因其果汁散发出番桃、草莓、柠檬、芒果、菠萝、香蕉、石榴、酸梅等百余种水果的香味而得名百香果。当前种植百香果病虫害严重,即百香果种植后根部腐烂;成活的百香果植株病虫害爆发严重,导致大量使用农药进行防治;百香果植株果实落果严重,产量下降。百香果植株的幼苗自身携带病毒,导致百香果种植障碍产生的现象亟待解决。因此,建立一种有效的百香果脱毒苗培养方法对于百香果产业的发展具有重要的意义。
发明内容
本发明的目的在于提供一种百香果脱毒苗的培养方法,旨在解决上述的技术问题。
本发明是这样实现的,一种百香果脱毒苗的培养方法,所述培养方法包括以下步骤:
A、选取健康百香果植株茎尖,采用无菌水清洗植株茎尖表面4-6次,将植株茎尖置于消毒液中浸泡消毒12min~18min,置于40℃~47℃水中浸泡处理48min~70min后,再将植株茎尖置于消毒液中浸泡消15min~20min;
B、经前处理后的植株茎尖接种至基础培养基上进行常规培养18d-23d,将新生腋芽取出再次接种至基础培养基上进行常规培养15d-20d;
C、在健壮无病害的植株上剪取5 ~ 8cm 小段外植体,在洗衣液中浸泡3 ~ 5min,再用自来水流水冲洗1h,在无菌条件下, 用酒精洗涤30s, 再使用0.1%的升汞溶液灭菌10~ 12min,用无菌水冲洗3 ~ 5 次,备用;
D、基础培养后的新生芽,进行脱毒培养,得到的组培苗在40℃~45℃下进行热处理3d-6d后,新生芽剥取0 .5cm的茎尖,茎尖置于消毒液中浸泡消毒15min~20min,用无菌水清洗茎尖3-5次,将茎尖接种至脱毒培养基上进行常规培养15d-20d;将得到的组培苗在40℃~45℃下进行热处理3d-6d后,新生腋芽剥取0 .2cm的茎尖,将茎尖置于消毒液中浸泡消毒7min~12min,用无菌水清洗茎尖5-7次,再将茎尖接种至脱毒培养基上进行常规培养20d-25d;
E、将脱毒培养后的百香果脱毒苗接种至增殖组培快繁培养基中,按照常规的组培方法进行增殖组培快繁培养25d-30d;
F、增殖组培快繁培养后的百香果脱毒苗接种至生根培养基中,按照常规的组培方法进行生根培养35d-45d。
本发明的进一步技术方案是:步骤A中所述的消毒液中各组分的比例为:按质量百分数计,抗生素0 .002%~0 .004%、恶霉灵0 .005%~0 .015%、乌洛托品0 .02%~0 .04%、霜霉威盐酸盐0 .06%~0 .08%、甲酚磺酸0 .01%~0 .02%,余量为水。
本发明的进一步技术方案是:步骤B所述的基础培养基配方为:按质量百分数计,蔗糖2%~3%、琼脂0 .8%~1 .0%、抗生素0 .002%~0 .004%、恶霉灵0 .005%~0 .015%、乌洛托品0 .02%~0 .04%、霜霉威盐酸盐0 .06%~0 .08%、甲酚磺酸0 .01%~0 .02%、氯化锌0 .01%~0 .02%、MS+6-BA 0 .3%~0 .4%、NAA 0 .06%~0 .08%、IAA 0 .05%~0 .07 %,余量为水。
本发明的有益效果是:操作简单,可用于大批量的百香果脱毒苗的工业化生产,同时脱毒苗种植后,有效降低百香果植株的病害率,与常规无脱毒的百香果苗种相比,本发明的百香果脱毒苗种植后发病率仅为2%,大大提高了百香果的经济效益。
具体实施方式
本发明提供的一种百香果脱毒苗的培养方法,其详述如下:
步骤A,选取健康百香果植株茎尖,采用无菌水清洗植株茎尖表面4-6次,将植株茎尖置于消毒液中浸泡消毒12min~18min,置于40℃~47℃水中浸泡处理48min~70min后,再将植株茎尖置于消毒液中浸泡消15min~20min。
所述的消毒液中各组分的比例为:按质量百分数计,抗生素0 .004%、恶霉灵0.015%、乌洛托品0 .04%、霜霉威盐酸盐0 .08%、甲酚磺酸0 .02%,余量为水。
步骤B,经前处理后的植株茎尖接种至基础培养基上进行常规培养18d-23d,将新生腋芽取出再次接种至基础培养基上进行常规培养15d-20d;
所述的基础培养基配方为:按质量百分数计,蔗糖2%、琼脂0 .8%、抗生素0 .004%、恶霉灵0 .015%、乌洛托品0 .04%、霜霉威盐酸盐0 .08%、甲酚磺酸0 .02%、氯化锌0 .02%、MS+6-BA0 .3%、NAA 0 .06%、IAA 0 .05%,余量为水。
步骤C,在健壮无病害的植株上剪取5 ~ 8cm 小段外植体,在洗衣液中浸泡3 ~5min,再用自来水流水冲洗1h,在无菌条件下, 用酒精洗涤30s, 再使用0.1%的升汞溶液灭菌10 ~ 12min,用无菌水冲洗3 ~ 5 次,备用;在晴好天气的中午或下午在健壮无病害的植株上剪取5cm 小段幼嫩的茎段,每个茎段含2 个节,去掉基端节上的叶片,留上节的1片叶,在搅匀的洗衣液中浸泡3min,再用自来水流水冲洗1h,在无菌条件下, 用75%的酒精洗涤30s, 再使用0.1%的升汞溶液灭菌10min,用无菌水冲洗3 次,备用。
步骤D,基础培养后的新生芽,进行脱毒培养,得到的组培苗在40℃~45℃下进行热处理3d-6d后,新生芽剥取0 .5cm的茎尖,茎尖置于消毒液中浸泡消毒15min~20min,用无菌水清洗茎尖3-5次,将茎尖接种至脱毒培养基上进行常规培养15d-20d;将得到的组培苗在40℃~45℃下进行热处理3d-6d后,新生腋芽剥取0 .2cm的茎尖,将茎尖置于消毒液中浸泡消毒7min~12min,用无菌水清洗茎尖5-7次,再将茎尖接种至脱毒培养基上进行常规培养20d-25d。
所述的脱毒培养基配方为:按质量百分数计,蔗糖3%、琼脂0 .8%、抗生素0 .001%、恶霉灵0 .002%、乌洛托品0 .005%、霜霉威盐酸盐0 .004%、MS+6-BA 0 .06%、NAA 0 .06%、IAA 0 .02%、维生素B1 0 .02%、维生素B2 0 .02%、维生素B6 0 .03%、维生素B9 0 .02%,余量为水。
步骤E,将脱毒培养后的百香果脱毒苗接种至增殖组培快繁培养基中,按照常规的组培方法进行增殖组培快繁培养25d-30d。
所述的增殖组培快繁培养基配方为:按质量百分数计,蔗糖2%、琼脂0 .8%、MS+6-BA0 .08%、NAA 0 .06%、IAA 0 .05%、维生素B1 0 .04%、维生素B2 0 .03%、维生素B6 0.03%、维生素B9 0 .02%,余量为水。
步骤F,增殖组培快繁培养后的百香果脱毒苗接种至生根培养基中,按照常规的组培方法进行生根培养35d-45d。
所述的生根培养基配方为:按质量百分数计,蔗糖3%、琼脂1 .0%、MS+6-BA 0.12%、NAA0 .08%、IAA 0 .07%、CCC 0 .4%、维生素B1 0 .06%、维生素B2 0 .05%、维生素B60 .05%、维生素B90 .04%,余量为水。
所述的生根培养基配方为:按质量百分数计,蔗糖 2 .5%、琼脂0 .9%、MS+6-BA 0.10%、NAA 0 .08%、IAA 0 .06%、CCC 0 .35%、维生素B1 0 .05%、维生素B2 0 .04%、维生素B6 0 .04%、维生素B9 0 .03%,余量为水。

Claims (3)
1.一种百香果脱毒苗的培养方法,其特征在于,所述培养方法包括以下步骤:
A、选取健康百香果植株茎尖,采用无菌水清洗植株茎尖表面4-6次,将植株茎尖置于消毒液中浸泡消毒12min~18min,置于40℃~47℃水中浸泡处理48min~70min后,再将植株茎尖置于消毒液中浸泡消15min~20min;
B、经前处理后的植株茎尖接种至基础培养基上进行常规培养18d-23d,将新生腋芽取出再次接种至基础培养基上进行常规培养15d-20d;
C、在健壮无病害的植株上剪取5 ~ 8cm 小段外植体,在洗衣液中浸泡3 ~ 5min,再用自来水流水冲洗1h,在无菌条件下, 用酒精洗涤30s, 再使用0.1%的升汞溶液灭菌10~ 12min,用无菌水冲洗3 ~ 5 次,备用;

E、将脱毒培养后的百香果脱毒苗接种至增殖组培快繁培养基中,按照常规的组培方法进行增殖组培快繁培养25d-30d;
F、增殖组培快繁培养后的百香果脱毒苗接种至生根培养基中,按照常规的组培方法进行生根培养35d-45d。
2.根据权利要求1所述的培养方法,其特征在于,步骤A中所述的消毒液中各组分的比例为:按质量百分数计,抗生素0 .002%~0 .004%、恶霉灵0 .005%~0 .015%、乌洛托品0.02%~0 .04%、霜霉威盐酸盐0 .06%~0 .08%、甲酚磺酸0 .01%~0 .02%,余量为水。
