Abstract
本发明公开了一种木糖试纸及其制备方法。所述的制备方法包括以下步骤:配制好显色剂及缓冲溶液,并在此基础上配制不同梯度的木糖标准溶液;将基纸浸渍在配制好的显色剂中,浸匀晾干,上下表层涂覆一层氧化钙;将不同浓度梯度的木糖溶液分别滴加在木糖试上反应显色,拍照制备比色卡,通过对比比色卡能够得知所测样品木糖浓度。本发明的制备方法制备工艺简单,易于操作,适于工业化生产;所得产物能够快速有效对木糖样品进行检测,具有良好的应用前景。
Description
技术领域
本发明涉及一种木糖试纸及其制备方法。
背景技术
木糖是一种戊糖,天然D-木糖是以多糖形态存在于植物中。在农产品废弃物中,如玉米穗轴,秸秆,棉花壳中有较多含量。然而木糖是以木聚糖大分子形式广泛存在于植物半纤维素中,用酸或酶降解木聚糖得到,自然界中并无游离木糖,因此其利用也是人们所广泛关注的。木糖现已在食品领域有广泛应用,如制造木糖醇,在食品、饮料中作为无热量甜味剂,高档酱油色等。不仅如此,在轻工、化工领域也有一定用途。所以高效简便的监测手段,对于相关的生产工艺过程中木糖的含量及变化,都是十分必要的。
试纸,指用化学药品浸渍过的、可通过其颜色变化检验液体或气体中某些物质存在的一类纸。各种试纸已经深入到各个领域中,并因便利性及廉价性被广泛认同和接受。但是木糖试纸的制备工艺仍在探索中。
发明内容
为解决现有技术的缺点和不足之处,本发明的首要目的在于提供一种木糖试纸的制备方法。
本发明的另一目的在于提供上述制备方法制得的木糖试纸。
本发明目的通过以下技术方案实现:
一种木糖试纸的制备方法,包括以下制备步骤:
(1)配制DNS显色剂及pH=5.50的乙酸-乙酸钠缓冲溶液,并在此基础上配制木糖标准溶液;
(2)制作木糖吸光度-木糖浓度的标准曲线,其拟合线斜率为K,截距为C0,并判断R2是否大于0.99:如果R2>0.99,则进行下一步,否则重新进行步骤(1);
(3)选取中速定性滤纸为基纸,将其浸泡配制好的DNS显色剂中,浸润均匀后取出,在室温下避光晾干,得到木糖试纸基纸;
(4)将木糖试纸基纸上下表面都涂覆一定量的氧化钙,制备得到木糖试纸;
(5)将不同浓度梯度的木糖溶液(优选0.02mL)分别滴加在木糖试纸不同位置,反应10s,得到木糖试纸比色卡;
(6)根据对比比色卡来确定木糖试纸的测定范围。
优选地,步骤(1)所述DNS显色剂配制方法如下:称取3,5-二硝基水杨酸3.15g,加水500mL,搅拌5s,水浴至45℃。然后逐步加入100mL 0.2g/mL的氢氧化钠溶液,同时不断搅拌,保证溶液温度不要超过48℃,直到溶液清澈透明。再逐步加入四水酒石酸钾钠91.0g、苯酚2.50g和无水亚硫酸钠2.50g。继续45℃水浴加热,同时补加水300mL,不断搅拌至完全溶解。停止加热,冷却至室温后,用超纯水定容至1000mL。用烧结玻璃过滤器过滤。取滤液,储存在棕色瓶中,避光保存。室温下存放7d后可以使用,有效期为6个月。
优选地,步骤(1)所述pH=5.50的乙酸-乙酸钠缓冲溶液的配制方法如下:称取三水乙酸钠23.14g,加入冰乙酸1.70mL。再加水溶解,定容至2000mL。测定溶液的pH,如果pH偏离5.50,再用0.1mol/L的乙酸溶液或者0.1mol/L的乙酸钠溶液调节至5.50。乙酸-乙酸钠缓冲溶液浓度优选为0.1mol/L。
步骤(1)所述木糖标准溶液的配制方法如下:称取10.000g无水木糖,加pH=5.50的乙酸-乙酸钠缓冲溶液溶解,定容至100mL。
优选地,所述中速定性滤纸直径φ=7cm;所述氧化钙为粉末状,涂覆量为0.10g/面。
优选地,步骤(5)所述不同浓度梯度的木糖溶液配制方法如下:分别吸取木糖标准溶液1.00mL、2.00mL、3.00mL、4.00mL、5.00mL、6.00mL和7.00mL,分别用pH=5.50的乙酸-乙酸钠缓冲液定容至10mL,配制成浓度为10.0mg/mL、20.0mg/mL、30.0mg/mL、40.0mg/mL、50.0mg/mL、60.0mg/mL和70.0mg/mL木糖溶液。
优选地,步骤(5)所述木糖试纸比色卡通过纸数码相机进行拍摄显色反应后的木糖试纸来制备。
一种木糖试纸,通过以上方法制备得到。将待测木糖溶液滴加到木糖试纸上,反应10s后,将试纸与制好的比色卡进行比对,即可确定待测木糖溶液的浓度范围。
与现有技术相比,本发明具有以下优点及有益效果:
(1)本发明的木糖试纸的制备方法制备工艺简单,易于操作,适于工业化生产;
(2)本发明制得的木糖试纸能够有效检测样品中木糖含量,具有良好的应用前景。

具体实施方式
下面结合实施例和附图对本发明作进一步详细的描述,但本发明的实施方式不限于此。
实施例1
一种木糖试纸的制备方法,步骤如下:
(1)配制DNS显色剂及pH=5.50的乙酸-乙酸钠缓冲溶液,并在此基础上配制木糖标准溶液,各自配制方法如下:
DNS显色剂配制方法:称取3,5-二硝基水杨酸3.15g,加水500mL,搅拌5s,水浴至45℃。然后逐步加入100mL 0.2g/mL的氢氧化钠溶液,同时不断搅拌,保证溶液温度不要超过48℃,直到溶液清澈透明。再逐步加入四水酒石酸钾钠91.0g、苯酚2.50g和无水亚硫酸钠2.50g。继续45℃水浴加热,同时补加水300mL,不断搅拌至完全溶解。停止加热,冷却至室温后,用超纯水定容至1000mL。用烧结玻璃过滤器过滤。取滤液,储存在棕色瓶中,避光保存。室温下存放7d后可以使用,有效期为6个月。
pH=5.50的乙酸-乙酸钠缓冲溶液的配制方法:称取三水乙酸钠23.14g,加入冰乙酸1.70mL。再加水溶解,定容至2000mL。测定溶液的pH,如果pH偏离5.50,再用0.1mol/L的乙酸溶液或者0.1mol/L的乙酸钠溶液调节至5.50。
木糖标准溶液的配制方法:称取10.000g无水木糖,加pH=5.50的乙酸-乙酸钠缓冲溶液溶解,定容至100mL。
(2)制作木糖吸光度-木糖浓度的标准曲线,如图1所示,其斜率K=0.783,截距为C0=0.0208,R2=0.9997>0.99;
(3)选取φ=7cm的中速定性滤纸为基纸,将其浸泡在配制好的DNS显色剂中,浸润均匀后取出,在室温下避光晾干,得到木糖试纸基纸;
(4)将木糖试纸基纸上下表层各涂覆0.10g的氧化钙,制备得到木糖试纸;
(5)分别吸取木糖标准溶液1.00mL、2.00mL、3.00mL、4.00mL、5.00mL、6.00mL和7.00mL,分别用pH=5.50的乙酸-乙酸钠缓冲液定容至10mL,配制成浓度为10.0mg/mL、20.0mg/mL、30.0mg/mL、40.0mg/mL、50.0mg/mL、60.0mg/mL和70.0mg/mL木糖溶液。
将0.02mL上述不同浓度梯度的木糖溶液分别滴加在制备好的木糖试纸不同位置上,反应10s,通过数码相机拍照得到木糖试纸比色卡;
(6)依据比色卡,可以通过比色卡较易辨别出棕色、橙色、黄色三种明显的不同梯度,因此此工艺制备得到的木糖试纸可以区分的木糖浓度范围是<30.0mg/mL,30.0mg/mL~40mg/mL,>40mg/mL。将待测木糖溶液滴加到木糖试纸上,反应10s后,将试纸与制好的比色卡进行比对,即可确定待测木糖溶液的浓度范围。
上述实施例为本发明较佳的实施方式,但本发明的实施方式并不受上述实施例的限制,其他的任何未背离本发明的精神实质与原理下所作的改变、修饰、替代、组合、简化,均应为等效的置换方式,都包含在本发明的保护范围之内。
Claims (5)
1.一种木糖试纸的制备方法,其特征在于,包括以下制备步骤:
(1)配制DNS显色剂及pH=5.50的乙酸-乙酸钠缓冲溶液,并在此基础上配制木糖标准溶液;
(2)制作木糖吸光度-木糖浓度的标准曲线,其拟合线斜率为K,截距为C0,并判断R2是否大于0.99:如果R2>0.99,则进行下一步,否则重新进行步骤(1);
(3)选取中速定性滤纸为基纸,将其浸泡配制好的DNS显色剂中,浸润均匀后取出,在室温下避光晾干,得到木糖试纸基纸;
(4)将木糖试纸基纸上下表层各涂覆一定量的氧化钙,制备得到木糖试纸;
(5)将不同浓度梯度的木糖溶液分别滴加在木糖试纸不同位置上,反应10s,用数码相机拍照得到木糖试纸比色卡;
(6)根据对比比色卡来确定木糖试纸的测定范围。
2.根据权利要求1所述的木糖试纸的制备方法,其特征在于,所述的pH=5.50的乙酸-乙酸钠缓冲溶液浓度是0.1mol/L。
3.根据权利要求1所述的木糖试纸的制备方法,其特征在于,所述的木糖标准溶液是指100.0mg/mL的木糖标准溶液。
4.根据权利要求1所述的木糖试纸的制备方法,其特征在于,所述中速定性滤纸直径φ=7cm;所述氧化钙为粉末状,涂覆量为0.10g/面。
5.一种木糖试纸,其特征在于,其由权利要求1至4任一项所述的木糖试纸的制备方法制备得到。
