Abstract
本发明公开了一种前列腺神经内分泌癌基因工程动物模型的构建方法,包括如下步骤:将敲除PSGL‑1基因后的TRAMP小鼠饲养至24周龄以上即得前列腺神经内分泌癌基因工程动物模型。该模型在整体水平更自然地模拟临床上疾病的发生发展过程,可有效地将复杂的生物学问题与药物的作用结合起来考察药物的治疗效果。而且在我们建立的小鼠模型活体动物中进行研究,能兼顾类似于人体内的各种背景因素,研究结论具有很高的可靠性。因此,我们发现的本实验小鼠模型是一种很好的用来筛选抗前列腺神经内分泌癌药物的有效工具,对寻找有效治疗药物的关键靶点以及神经疾病的治疗方法具有重要的意义。
Description
技术领域
本发明涉及疾病动物模型制备技术领域,具体地,涉及一种前列腺神经内分泌癌基因工程动物模型的构建方法及应用。
背景技术
前列腺癌是男性中常见的泌尿生殖系统肿瘤,严重影响男性的身体健康,给患者造成心理上的巨大压力。前列腺癌发病率与死亡率占男性恶性肿瘤第一位,近年来我国前列腺癌的发病率呈明显上升趋势。临床上对于前列腺癌患者的主要的治疗方法有手术治疗、放射治疗、化学疗法、内分泌疗法等。手术治疗是目前治疗癌症的首选方法,但是前列腺癌患者的诊断多在晚期,常错过手术的最佳时机,并且前列腺根治性手术多损伤较大。放射治疗是目前对于前列腺癌的治疗十分有效的方法,主要用于无法采用手术治疗但尚无远处转移的患者。放射治疗常可以使前列腺肿瘤减小。化学疗法是作为晚期前列腺癌的辅助治疗方法,主要用于手术治疗或者放射治疗后局部肿瘤已消除的患者。化疗药物可以消除潜在的、尚无探测到的微小病灶。但是单独使用化学疗法不能治愈原发病灶,只有对于采用其他方法治疗的患者进行化学疗法辅助治疗,可以延长病人的生存时间。在临床上内分泌疗法是治疗前列腺癌晚期的重要手段。前列腺癌具有典型的激素依赖性,当体内雄激素水平下降时,癌细胞出现死亡,可以使前列腺病变和症状明显得到缓解。
临床上,绝大部分前列腺癌为起源于外分泌细胞的导管和腺泡腺癌(PCA),5%~10%患者伴有局灶性的神经内分泌分化(NED),而0.5~2%患者则出现广泛的NED,称为前列腺神经内分泌癌(NEPC),侵袭性更强。尽管目前对于前列腺癌的治疗方法有很多种,但是一旦患者肿瘤出现NED,则目前尚无有效的临床治疗手段。NED的发生与肿瘤的进展、雄激素非依赖转化和不良预后等有重要关系。临床上,NEPC对目前治疗前列腺癌的主要方法-雄激素阻断疗法无反应,且预后差,患者平均生存时间5~17.5个月。
尽管目前有TRAMP等自发前列腺癌基因工程小鼠用于前列腺癌治疗药物的开发,但是模型小鼠前列腺癌病理进程发展较慢,导致研究周期较长,这严重制约了抗前列腺神经内分泌癌药物的开发研制。所以,寻找新的、对于人类前列腺癌临床病理发展过程能够快速模拟的基因工程小鼠模型,将为临床前列腺神经内分泌癌肿瘤治疗提出了新的治疗途径和希望,并具有更加广阔的应用前景。
发明内容
本发明为了克服现有技术的上述不足,提供一种前列腺神经内分泌癌基因工程动物模型的构建方法。
本发明的另一个目的是提供一种前列腺神经内分泌癌基因工程动物模型在筛选治疗前列腺神经内分泌癌药物中的应用。
为了实现上述目的,本发明是通过以下技术方案予以实现的:
一种前列腺神经内分泌癌基因工程动物模型的构建方法,包括如下步骤:将敲除PSGL-1基因后的TRAMP小鼠饲养至24周龄以上即得前列腺神经内分泌癌基因工程动物模型。
本发明发现自发前列腺癌的TRAMP基因工程小鼠的前列腺肿瘤为腺癌。而在敲除PSGL-1后的TRAMP小鼠(TRAMP;PSGL-1-/-)饲养至24周龄以上时,前列腺肿瘤出现显著的神经内分泌癌特性。肿瘤组织病理学表现为由燕麦形的小细胞组成,细胞异型性明显,细胞小,胞浆少,核仁不明显,几乎不表达分泌前列腺特异性抗原(PSA),但表达神经内分泌细胞标志物,如嗜铬粒蛋白A(CgA),突触素(Syn)和神经元特异性烯醇化酶(NSE),且电镜下可见胞浆内含有很多神经内分泌颗粒,这些是临床病理诊断NEPC的依据,因此,这样的模型可以作为研究前列腺神经内分泌癌发病机理及筛选靶向药物和疗效评估的动物模型。
优选地,所述前列腺神经内分泌癌基因工程动物模型的构建方法如下:将雄性PSGL-1-/-小鼠和雌性TRAMP小鼠杂交得F1代,F1代分笼鉴定;将F1代中的TRAMP阳性; PSGL-1+/-雌性小鼠与雄性PSGL-1-/-小鼠杂交得F2代,F2代分笼鉴定;将F2代中的TRAMP阳性;PSGL-1-/-雌性小鼠与PSGL-1-/-雄性小鼠杂交获得F3代,F3代分笼鉴定获得TRAMP;PSGL-1-/-小鼠;TRAMP;PSGL-1-/-小鼠饲养至24周龄以上即得前列腺神经内分泌癌基因工程动物模型。
作为优选实施方式,将TRAMP;PSGL-1-/-小鼠饲养至24周龄即得前列腺神经内分泌癌基因工程动物模型。
如上所述构建方法得到的前列腺神经内分泌癌基因工程动物模型在筛选治疗前列腺神经内分泌癌药物中的应用。
与现有技术相比,本发明具有如下有益效果:
本发明提供的前列腺神经内分泌癌基因工程动物模型可以用来研究前列腺神经内分泌癌发病机理及筛选靶向药物和疗效评估。由于该模型在整体水平更自然地模拟临床上疾病的发生发展过程,可有效地将复杂的生物学问题与药物的作用结合起来考察药物的治疗效果。而且在我们建立的小鼠模型活体动物中进行研究,能兼顾类似于人体内的各种背景因素,研究结论具有很高的可靠性。因此,我们发现的本实验小鼠模型是一种很好的用来筛选抗前列腺神经内分泌癌药物的有效工具,对寻找有效治疗药物的关键靶点以及神经疾病的治疗方法具有重要的意义。

具体实施方式
下面结合说明书附图和具体实施例对本发明作出进一步地详细阐述,所述实施例只用于解释本发明,并非用于限定本发明的范围。下述实施例中所使用的试验方法如无特殊说明,均为常规方法;所使用的材料、试剂等,如无特殊说明,为可从商业途径得到的试剂和材料。
TRAMP小鼠品系名称为C57BL/6-Tg(TRAMP)8247Ng/J,PSGL-1基因敲除小鼠(PSGL-1 Knock out;PSGL-1-/-)品系名称为B6.Cg-Selplgtm1Fur/J,TRAMP小鼠和PSGL-1基因敲除小鼠均购自美国Jackson实验室,C57小鼠购自广东省动物中心。实验动物均饲养于SPF级的环境中。
实施例1
一、TRAMP;PSGL-1-/-杂交小鼠的建系(构建流程见图1):将雄性PSGL-1-/-小鼠和雌性TRAMP小鼠合笼饲养,每笼以雄:雌=1:3的最佳比例配种,得F1代后分笼鉴定;将F1代中的TRAMP阳性; PSGL-1+/-雌性小鼠与雄性PSGL-1-/-小鼠杂交得F2代小鼠,F2代分笼鉴定;将F2代中的TRAMP阳性; PSGL-1-/-雌性小鼠与PSGL-1-/-雄性小鼠杂交扩群,获得F3代小鼠,F3代分笼鉴定将获得的TRAMP;PSGL-1-/-小鼠用于实验。
其中TRAMP及PSGL-1-/-小鼠的鉴定方法如下:
小鼠基因组DNA提取:组织放入已标记好的EP管中;将组织浸没于500ul裂解缓冲液A(25mM NaOH 0.5g;0.2mM Na2EDTA.2H2O0.037g加H2O至500ml)中;金属浴中100℃,1h;加入500ul裂解缓冲液B(40mM Tris Acid 6.3g加H2O至500ml),旋涡混合,离心,5000rpm,1min,4℃保存;
TRAMP基因片段的扩增:TRAMP小鼠PCR反应引物序列如下所示。
Mouse β Casein正向引物:5′-GAT GTG CTC CAG GCT AAA GTT-3′;
Mouse β Casein 反向引物:5′-AGA AAC GGA ATG TTG TGG AGT-3′;
Probasin-1 正向引物:5′-CCG GTC GAC CGG AAG CTT CCA CAA GTG CAT TTA-3′;
T-antigen reverse primer:5′-CTC CTT TCA AGA CCT AGA AGG TCC A-3。
配置25ul PCR反应体系:四个引物各1ul,2×PCR Master Mix 12.5ul,DNA模板3ul,加水补足25ul。
TRAMP小鼠PCR反应条件:94℃ 1min 预变性;(94℃ 1min 变性,60℃ 2min 退火,72℃ 3min 延伸)30个循环;72℃ 5min 充分延伸;4℃冷却扩增产物;取PCR反应产物10ul,加样于1.2%琼脂糖凝胶孔,于1×TAE缓冲液中电泳170V,30分钟。在凝胶成像系统观察电泳结果,如图2:TRAMP基因阳性小鼠有500 bp(野生型)和600 bp (Tag片段)两条带,阴性小鼠只有500 bp一条带。
PSGL-1基因片段的检测:检测引物如下所示。
Wild type 正向引物:5'-AGC TTC CTT GTG CTG CTG AC-3';
Common:5'-TCA AAA TCG TCA TCC CCA AC-3';
Mutant 1:5'-TCA AAA TCG TCA TCC CCA AC-3';
Mutant 2:5-'CCT TCT ATC GCC TTC TTG ACG-3'。
配置25ul PCR反应体系:四个引物各1ul,2×PCR Master Mix 12.5ul,DNA模板3ul,加水补足25ul。
PCR反应条件为:94℃ 5min 预变性;(94℃ 30s 变性,65℃ 1min 退火,72℃1min 延伸)35个循环;72℃ 2min 充分延伸;4℃冷却扩增产物;取PCR反应产物10ul,加样于1.2%琼脂糖凝胶孔,于1×TAE电泳缓冲液中电泳160V,25min。用凝胶成像系统观察电泳结果。如图3:PSGL-1杂合子为500 bp和140 bp;PSGL-1全敲为500 bp;野生型小鼠为140bp。
二、按照常规操作饲养TRAMP;PSGL-1-/-小鼠,定期检测不同周龄小鼠的肿瘤组织病理学特征,结果发现TRAMP;PSGL-1-/-小鼠饲养至24周龄时,前列腺肿瘤出现显著的神经内分泌癌特性。
TRAMP;PSGL-1-/-小鼠饲养至24周龄时的肿瘤组织病理学特征检测方法和结果如下:
一、H&E染色,取24周,55周TRAMP小鼠前列腺组织和24周TRAMP;PSGL-1-/-小鼠前列腺组织得到的石蜡切片,厚度为3~4μm。组织石蜡切片脱蜡、苏木素染色4 分钟,返蓝,伊红染色3 秒,37℃过夜、封片。正置显微镜下观察各个组织结构变化并拍照,结果见图4。
二、免疫组化染色
1、随机选取24周,55周TRAMP小鼠前列腺组织和24周TRAMP;PSGL-1-/-小鼠,每组小鼠各3只。
2、用脱臼法处死小鼠,取没有坏死区域的前列腺组织,将前列腺组织后叶制成石蜡组织切片。
① 固定:4%多聚甲醛固定液中固定24小时;
② 脱水:固定后的组织经过蒸馏水及70%乙醇清洗后进入脱水步骤,脱水过程使用梯度浓度酒精:70%—80%—90%—95%—无水乙醇1—无水乙醇2,其中70%至90%乙醇中放置30分钟,其余步骤均放置1小时。
③透明:二甲苯1放置1小时,二甲苯2放置1小时。
④浸蜡:准备两个蜡缸分别盛放低熔点石蜡及高熔点石蜡,预先放入65℃烘箱熔化。透明后的组织分别放入低熔点石蜡1小时,高熔点石蜡1小时。
⑤包埋:预先在自动包埋机熔化高熔点蜡,并保持其熔化状态,将浸蜡后的组织调整到切片需要的状态放入包埋盒,倒入高熔点蜡并置于冰上或室温等待其冷却凝固。
⑥切片:先将展片池的水温调节为42℃并在冰箱预冷蜡块,修片至需要的截面后将切片厚度调节为4μm开始切片。得到到切片放入展片池以除去皱褶,展片完成后用预先涂有APES的载玻片捞取组织,放置于60℃烘箱4小时或者37℃烘箱过夜烤干后4℃保存。
3、每个标本随机提取三张组织切片进行PSA、CgA、Syn和NSE特异性免疫染色:
①组织切片脱蜡及重新水化:将载玻片按次序放置于二甲苯1-二甲苯2-1/2二甲苯:乙醇-100%乙醇-95%乙醇-85%乙醇-75%乙醇中浸泡各10min。最后放置于双蒸水中振荡30s。
②将组织切片放入3%H2O2-甲醇溶液(提前预热30min)中,37℃,30min。
③从3%H2O2-甲醇溶液中取出切片,PBS洗三次,每次5min。
④胃蛋白酶修复:将玻片上的溶液擦干,于组织上加上0.4%胃蛋白酶,37℃,30min。
⑤PBS洗三次,每次5min。将玻片上的溶液擦干,于组织上加上10%BSA,以盖住组织为准,放置于封闭盒中,盒中放入适量双蒸水,37℃,1h。
⑥用微量移液器将10%BSA直接吸掉,分别加上一抗PSA、CgA、Syn和NSE,置于4℃,过夜。
⑦将组织切片从封闭盒中拿出,PBS 洗三次,每次5min。擦干,于组织上加上二抗goat-anti-mouse IgG(中衫金桥),置于封闭盒中,37℃,40min。
⑧从封闭盒中拿出玻片,PBS洗三次,每次5min。
⑨DAB显色,苏木素复染:擦干,于组织上加上DAB Substrate Kit溶液(现用现配),室温静置约5min,放置于水中停止反应。苏木素染色40s,用流水洗去染色剂,小心不要冲掉组织。
⑩脱水、封片。结果见图5。
三、透射电镜检测:小鼠用6%的水合氯醛麻醉,24周TRAMP小鼠和24周TRAMP;PSGL-1-/-小鼠前列腺组织,每组小鼠各3只,然后用含2%戊二醛和1%蔗糖的0.1 摩尔浓度的二钾砷酸盐溶液固定3 小时。样品再用0.1 摩尔浓度的二钾砷酸盐溶液清洗,然后用含有1%四氧化锇的0.1 摩尔浓度的二钾砷酸盐溶液固定2 小时,所有的固定条件都是25℃和pH7.0。组织用梯度乙醇溶液脱水后,用包埋剂EPON 包埋,包埋剂EPON 需要在60℃的条件下聚合48小时。聚合完全后,使用钻石刀头超薄切片机将组织切成80纳米的连续切片,用铜网收集切片。切片用醋酸双氧铀反透,再用1%的甲苯氨蓝染色。最后样品铜网用电子透射显微镜(Hitachi 600 TEM)拍照检测,结果见图6。
由上述检测结果可以看出:敲除PSGL-1后的TRAMP小鼠(TRAMP;PSGL-1-/-)饲养至24周龄时,前列腺肿瘤出现显著的神经内分泌癌特性,肿瘤组织病理学表现为由燕麦形的小细胞组成,细胞异型性明显,细胞小,胞浆少,核仁不明显,几乎不表达分泌前列腺特异性抗原(PSA),但表达神经内分泌细胞标志物,如嗜铬粒蛋白A(CgA),突触素(Syn)和神经元特异性烯醇化酶(NSE),且电镜下可见胞浆内含有很多神经内分泌颗粒,这些是临床病理诊断NEPC的依据,因此,这样的模型可以作为研究前列腺神经内分泌癌发病机理及筛选靶向药物和疗效评估的动物模型。
Claims (3)
1.一种前列腺神经内分泌癌基因工程动物模型的构建方法,其特征在于,包括如下步骤:将敲除PSGL-1基因后的TRAMP小鼠饲养至24周龄以上即得前列腺神经内分泌癌基因工程动物模型;
所述前列腺神经内分泌癌基因工程动物模型的构建方法具体如下:将雄性PSGL-1-/-小鼠和雌性TRAMP小鼠杂交得F1代,F1代分笼鉴定;将F1代中的TRAMP阳性; PSGL-1+/-雌性小鼠与雄性PSGL-1-/-小鼠杂交得F2代,F2代分笼鉴定;将F2代中的TRAMP阳性; PSGL-1-/-雌性小鼠与PSGL-1-/-雄性小鼠杂交获得F3代,F3代分笼鉴定获得TRAMP;PSGL-1-/-小鼠;TRAMP;PSGL-1-/-小鼠饲养至24周龄以上即得前列腺神经内分泌癌基因工程动物模型。
2.根据权利要求1所述的构建方法,其特征在于,TRAMP;PSGL-1-/-小鼠饲养至24周龄以上即得前列腺神经内分泌癌基因工程动物模型。
3.权利要求1或2所述构建方法得到的前列腺神经内分泌癌基因工程动物模型在筛选治疗前列腺神经内分泌癌药物中的应用。
