Abstract
本发明公开了一套评测杀菌剂对枝干表层腐烂病菌铲除效果的方法,涉及植物病害防治药剂评测方法。具体方法是用腐烂病菌的孢子悬浮液接种苹果的活体枝条,让接种枝条在自然条件下生长2~4周后,在接种枝条表面施用待测杀菌剂,施药1周后,从树体上剪取处理枝条,破坏芽部及周围皮层组织,在离体条件下诱导伤口发病,依据伤口发病的百分率,评测杀菌剂的铲除效果。本发明克服了现有技术难以评测杀菌剂对潜伏于枝条表层腐烂病菌铲除效果的问题,为准确评测各种杀菌剂对腐烂病的防治效果提供一套准确、可靠而又简便的方法。
Description
技术领域
本发明涉及植物病害防治药剂评测方法,尤其适用于评测杀菌剂对在枝条表皮内潜伏,且主要通过伤口侵染枝条病菌的铲除效果。
背景技术
腐烂病是苹果和梨树的重要病害,主要造成死枝、死树和毁园。在苹果树上腐烂病是第一大病害,而且难以防治,被认为是苹果树的“癌症”。自上世纪50年代,苹果树腐烂病在中国已大规模流行4次,几乎毁灭了建国以来新栽植的所有苹果树,目前正面临第五次大流行的威协。腐烂病也是梨树上的重要病害,主要危害西洋梨、新疆库尔勒梨等感病品种。
腐烂病菌具有潜伏侵染现象。目前研究认为,腐烂病菌能从剪锯口、伤口、皮孔、芽眼等部位侵染,病菌侵染后并不能马上致病,而是潜伏于伤口死组织内、枝干表层死组织内、木质部等部位。当树势衰弱,或者树体遭受冻害后,苹果枝条的木栓层遭受破坏,病菌随机侵入皮层内扩展致病。因此,铲除枝条表层潜伏的病原菌是防治腐烂病的主要措施。
腐烂病菌产生孢子数量很大,而且侵染条件非常容易满足。流行学研究结果表明,苹果树上腐烂病斑上的分生孢子器和子囊壳遇到能使枝条湿润的降雨(降雨量超过2mm),就能释放大量的分生孢子和子囊孢子,病菌孢子释放后主要随雨水水流,或在雨滴中随气流传播到苹果枝干上。当枝条表面湿润,且温度在5℃~37℃范围时,病菌孢子就能够萌发,并在枝条的表层死组织生长扩展,并且长期存活。因此,在已经开始发病的果园内,苹果枝干上都潜带有大量的腐烂病菌。
目前评估杀菌剂对腐烂病的防治效果,或筛选有效的杀菌剂主要有2种方法:室内试验和田间试验。室内主要通过培养基混药法评估杀菌剂对腐烂病菌生长的抑制效果,但通过室内评测出的有效药剂,多数在田间都达不到理想效果。田间试验主要是通过田间施药后,观测第二年春季腐烂病的发生情况来评测杀菌剂对腐烂病的防治效果。然而,腐烂病的侵染发病机制较为复杂,田间试验的成功率非常低,难以对杀菌剂的防治效果作准确的评价。
发明内容
针对现有技术的缺陷,本发明的目的在于提供一套评测杀菌剂对潜伏于枝条表层腐烂病菌铲除效果的技术与防治;为准确评价各种杀菌剂对腐烂病菌的铲除效果提供一套准确、可靠而又简便的方法。
本项发明采用技术原理为:当枝条受伤后,潜伏在苹果枝条表层的腐烂病菌能够生长扩展,侵入枝条皮层的活体细胞,导致皮层发病,根据枝条发病的轻重,就可以判断枝条带菌量的多少。苹果枝条叶腋处容易积水,腐烂病菌很容易在叶腋处萌发,形成菌丝后潜伏叶芽周围的死组织内。通过人为方法,使叶芽周围的皮层受伤,若叶芽周围有潜伏病菌,病菌会通过新形成的伤口侵入皮层,形成典型的腐烂病斑。
本项发明采用技术方案为:用腐烂病菌的孢子悬浮液接种1~2年生的活体苹果枝条,病菌接种后培养2~4周,然后在接种枝条上施用杀菌剂,施药1周后将处理枝条剪下,用小锤或木棒等敲击苹果枝条的叶芽处,使叶芽和叶芽周围皮层受伤,然后将受伤枝条转入适宜于腐烂病菌生长致病,而不利于伤口愈合的温度和湿度下培养,待清水处理的对照枝条发病后,观察记录枝条每个叶芽处的发病情况。将药剂处理枝条叶芽发病率与清水对照处理枝条叶芽发病率比较后,评测药剂的防治效果。
具体实施方案:
(1)孢子悬浮液配制:用纯净水将腐烂病菌新鲜的分生孢子或子囊孢子配制成孢子悬浮液,浓度控制在105~106个孢子/毫升;孢子悬浮液现配现用,放置时间不超过1小时;
(2)病菌接种:在苹果的生长季节,选取当年生和1年生的苹果枝条,长度不短于30厘米,用手持喷雾器将孢子悬浮液均匀的喷施到苹果枝条上,接种后将整个枝条套塑料袋保湿48小时,解袋时标记接种部位;病菌接种在阴雨天或在湿度较大的傍晚进行,以保证病菌孢子能萌发,并在枝条表面生长;解袋后,让枝条在自然条件下生长2~4周,使接种病菌能在枝条表层定殖;
(3)药剂处理:将待测杀菌剂用蒸馏水配成相应浓度的药液,以蒸馏水为空白对照,用手持喷雾器均匀的喷撒到接种枝条上,直到枝条上有药液流下为止;药剂处理后,让处理枝条在自然条件下再生长1周,使处理药剂充分发挥其作用;
(4)诱导发病:将处理枝条剪下,摘除叶片,并剪成15~20厘米长的枝段,用木棒或小锤从每个芽的芽部敲击枝条,破坏芽部和芽部周围的皮层组织,使其形成明显的机械伤,标记后插入广口组培瓶中,下端1~2厘米浸入清水中,将组培瓶转入保湿缸或其他密闭的容器内,容器底部加入200~500毫升饱和的食盐水,使容器内相对湿度保持在70%~75%;将保湿缸转入20~30℃之间的恒温度环境中培养20~30天;
(5)发病调查:待对照处理的枝条上大部分芽眼发病后,把全部的枝条取出,检查记录每个受伤芽眼的是否发生腐烂病,计算芽眼的发病率,根椐芽眼的发病率评测药剂的防治效果。
应用实例:五种杀菌剂对枝干表层腐烂病菌的铲除效果:
(1)配制孢子悬浮液:用马铃薯葡萄糖琼脂(PDA)培养基上培养的苹果树腐烂病菌的新鲜菌饼,接种于大麦粒培养基(大麦粒70克,蛋白胨1%,6%蜂蜜水,121℃灭菌60分钟),25℃恒温箱中培养15天,取产生分生孢子器的大麦粒,转入直径为9厘米含有1%水琼脂培养皿中,25℃下保湿培养3天,待分生孢子器溢出橘黄色分生孢子角后,挑取新鲜分生孢子角置于纯净水中,配成106个/毫升孢子悬浮液;
(2)病菌接种:在3年生富士苹果树上,选择长度60厘米当年生健康无伤枝条,用于病菌接种;接种时,用手持喷雾器将孢子悬浮液均匀地喷撒到枝条上,直到孢子悬浮液流下为止。接种后立即用塑料袋把整个枝条全部密封保湿,48小时后解袋;
(3)药剂处理:在病菌接种3周后,将待测杀菌剂配制相应浓度的药液,用手持喷雾器将药液均匀的喷洒在接种枝条上,直到药液流下为止;每种杀菌剂药剂处理6个接种枝条,以清水为对照,共处理36个枝条;
(4)诱导发病:药剂处理1周后,将全部处理枝条自基部剪下,并剪成20cm长的枝段,用小锤轻轻敲打芽部,破坏芽部和芽部周围皮层结构;枝段上端用石蜡密封,置于组培瓶中,枝条下端1~2厘米浸入清水中,进行水培;将组织瓶转入保湿缸中,用饱和的食盐水控制相对湿度,然后将保湿缸转入25℃的恒温箱内培养;每周观察记录枝条的发病情况,当清水处理枝条的大部分芽眼发病后,将枝条取出,检查芽眼的发病情况;当受伤芽部出现典型的腐烂病斑时,检查所有受伤芽部的发病情况;
(5)试验结果:受伤枝体离体培养30天后,对照枝条60%以上的受伤部位都出现明显的腐烂病斑,取出所有枝条,检查记录枝条的发病情况;结果表明,在所测试的五种药剂中,吡唑醚菌酯处理的枝条中受伤部位发病率最低,为6%,病菌产除效果达94%;其次为波尔多液,受伤部位发病率为11%,病菌铲除效果为89%;甲基硫菌灵和代森锰锌的铲除效果为44%;苯醚甲环唑的铲除效果最差,为17%;具体数据见表1。

Claims (1)
1.一套评测杀菌剂对枝干表层腐烂病菌铲除效果的方法;其特征在于:包括孢子悬浮液配制、病菌接种、药剂处理、诱导发病和发病调查五个步聚,最终依椐芽眼的发病率评测药剂对枝干表层腐烂病菌铲除效果;
所述孢子悬浮液配制是指用纯净水将腐烂病菌新鲜的分生孢子或子囊孢子配制成孢子悬浮液,浓度控制在105~106个孢子/毫升;孢子悬浮液现配现用,放置时间不超过1小时;
所述病菌接种是指在苹果的生长季节,选取当年生和1年生的苹果枝条,长度不短于30厘米,用手持喷雾器将孢子悬浮液均匀的喷施到苹果枝条上,接种后将整个枝条套塑料袋保湿48小时,解袋时标记接种部位;病菌接种在阴雨天或在湿度较大的傍晚进行,以保证病菌孢子能萌发,并在枝条表面生长;解袋后,让枝条在自然条件下生长2~4周,使接种病菌能在枝条表层定殖;
