Abstract
本发明公开一种猪精液的处置方法,属猪的人工受精技术领域,包括以下操作:A、对精液进行品质检查,即容量、温度、密度、活力、形态、颜色检查;B、稀释精液;C、分装精液;3、在精液瓶上做标记;D、保存精液。本发明通过对猪精液的品质检查、稀释、分装、保存及运输全过程的规范操作和控制,最大限度地保持精子活力,并使不同批次的精液保持了基本稳定的质量,延长保存期限。
Description
技术领域
本发明涉及猪的人工受精技术领域,特别是指一种猪精液的处置方法。
背景技术
目前,我国是生猪生产和消费大国,规模以上的生猪养殖场,出于避免疾病和提高受孕率等考虑,均采用人工受精。配种公猪的精液的处置方法成为生猪生产中的重要环节。现有技术中多为的猪精液稀释液的配方及制备方法,缺乏对猪精液的综合处置方法。现有的采精技术主要缺点为不注意消毒,超作规范,导致大量的细菌微生物在采精过程中流入精液中,致使公猪精子质量下量,母猪受胎率下量,增加养殖成本。
发明内容
本发明的目的是提供一种猪精液的处置方法,通过对猪精液的品质检查、稀释、分装、保存及运输全过程的规范操作和控制,最大限度地保持精子活力,并使不同批次的精液保持了基本稳定的质量,延长保存期限。
本发明提出一种猪精液的处置方法,包括以下操作:
A、对精液进行品质检查:
(1)容量检查:称量精液重量并做好记录。
(2)温度检查:测量精液温度,确保其温度在34℃以上,如果温度低于34℃则丢弃;
(3)密度检查:用玻璃棒滴出一点精液作为样品,滴到载玻片上,然后放到显微镜下观察密度,密度标准是二个精子之间的间隔为能容纳一个精子头部的距离;其密度没达到标准则丢弃处理,其密度如超过此标准,应进行稀释直到达到标准密度;
(4)活力检测:用玻璃管取少量原精放在载玻片上,盖上盖玻片,在37℃的加热器上放置15秒后放在显微镜下评估精子活力;合格精液的精子活力最低标准为70%,低于此标准应丢弃;
(5)形态检测:定期畸形率超过30%,则丢弃这头公猪的精液,并每周对这头公猪进行一次检测,以找出问题所在,改善它的状况;
(6)颜色检查:检查精液,其色泽应为乳白色、气味正常;
B、稀释精液:
(1)配制精液稀释液:将精液稀释剂粉末按额定剂量加入预热到37℃的蒸馏水中,混匀后的稀释液装入自封袋中,外部套上塑料稀释杯,在37℃的恒温箱中静置1小时备用;成品的稀释液直接在恒温箱中预热到37℃备用;
(2)稀释:先测量精液的温度,确保稀释液与精液的温度差在1℃范围之内,如果精液温度较低,则将备用的稀释液温度降低后再稀释,轻轻地将稀释液沿着采精袋的边缘缓慢地加入精液里,以减少对精子细胞的冲击;
C、分装精液:
(1)精液稀释完成后,把稀释后的精液轻轻混匀,然后将精液分装到输精瓶中,装盖子时排空气,每瓶精子含量控制在20-80亿;
(2)每分装6-8瓶精液就将余下的精液混匀一次,精液瓶放置于37℃±1℃的柔性织物上,需要储存的需放置1小时;
3、在精液瓶上做标记;
D、保存精液
(1)将精液瓶放入恒温17℃±1℃的精液贮藏箱,避免紫外光或阳光照射,在精液贮藏箱内要用柔性织物将精液瓶覆盖或包裹;
(2)每天翻转精液瓶两次,并做好记录;
优选地,所有操作中,不要让手指碰到采精袋里边;精液到达实验室后,要在3-5分钟之内完成精液的品质检查,检查完后立即稀释精液。
其还包括精液的运输,当传送精液到猪舍或办公室时,将精液装在便捷式恒温箱里,并使该精液运输箱里内的温度在17℃±1℃。
本发明的有益效果是:对精液进行容量、温度、密度、活力检查,目的是对公猪的精液要做到保质保量,宁缺毋滥,避免未达标精液使用后,造成母猪受孕率低或母猪产仔数少,甚至母猪不受孕,母猪的生产潜力不能充分发挥,增加养殖成本。提供了对猪精液的品质检查、稀释、分装、保存及运输全过程的规范操作的处置方法,每精液稀释温度、保存温度、运输温度精确控制,精细化操作流程,有效地延长精液保存的时间,以扩大良种公猪的使用效率,并使不同批次的精液保持了基本稳定的质量。经本方法处置后的精液,在保存5天后精子活力仍为70%以上,以满足养猪生产的需要。3-5分钟之内完成精液的品质检查、精液稀释温度定为35-37℃等都是为了取精后尽快地将精液与稀释液稀释,因为如果精液放置时间长,难以避免细菌、病原微生物等进入精液,破坏精子质量,而将稀释剂加温到37℃,而不是等精子冷却至常温后再与稀释剂稀释,也是为了缩短稀释前放置时间,稀释剂中有抗菌成份,所以尽早稀释可以保护精子免受感染;稀释液与精液的温度差在1℃范围之内是因为精子是很娇嫩的,大的温差会引起精子活力下降甚至死亡,精液瓶放置于37℃±1℃的柔性织物上放置1小时,37℃±1℃柔性织物能避免精液瓶突然接触外界冷物,引起精子活力下降甚至死亡,有毛巾等柔性物间隔保护,能够减少温差刺激,放置1小时是为了让精子缓慢降温至室温,以便放入恒温17℃±1℃的精液贮藏箱;在精液贮藏箱内要用柔性织物将精液瓶覆盖或包裹,以避免精液瓶直接接触箱壁等引起大温差致精子活力下降甚至死亡;17℃±1℃是最适合精子保存的温度,温度过高会引起精子活动加速,致精子疲劳引起精子活力下降甚至死亡,温度过低会使精子受冻致死。
具体实施方式
实施例一本实施例提出一种猪精液的处置方法,包括以下操作:
A、对精液进行品质检查:
(1)容量检查:称量精液重量并做好记录。
(2)温度检查:测量精液温度,确保其温度在34℃以上,如果温度低于34℃则丢弃;

Claims (2)
1.一种猪精液的处置方法,其特征在于,包括以下操作:
A、对精液进行品质检查:
(1)容量检查:称量精液重量并做好记录;
(2)温度检查:测量精液温度,确保其温度在34℃以上,如果温度低于34℃则丢弃;
(3)密度检查:用玻璃棒滴出一点精液作为样品,滴到载玻片上,然后放到显微镜下观察密度,密度标准是二个精子之间的间隔为能容纳一个精子头部的距离;其密度没达到标准则丢弃处理,其密度如超过此标准,应进行稀释直到达到标准密度;
